2. Circular RNA circCNOT6L regulates cell development through modulating miR-384/FN1 axis in esophageal squamous cell carcinoma.

Abstract

OBJECTIVE: In recent years, circular RNAs (circRNAs) and microRNAs (miRNAs) have been shown to be related to the development of esophageal squamous cell carcinoma (ESCC). However, their functional mechanisms remain to be investigated. Herein, we focus our research on the functions and mechanisms of circCNOT6L and miR-384 in ESCC. MATERIALS AND METHODS: The levels of circCNOT6L, miR-384, and fibronectin 1 (FN1) were determined using quantitative Real Time-Polymerase Chain Reaction (qRT-PCR). RNase R was used to investigate circCNOT6L stabilization. Cell proliferation and apoptosis were assessed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and flow cytometry, respectively. Western blot assay was employed to analyze the protein levels of FN1, proliferation-related genes, and iron metabolism-related genes. In addition, the interaction between miR-384 and circCNOT6L or FN1 was predicted by starBase3.0 and confirmed by the Dual-Luciferase reporter assay. Mouse xenograft was carried out to measure the effect of circCNOT6L on tumor growth in vivo. RESULTS: CircCNOT6L and FN1 levels were upregulated, and miR-384 level was downregulated in ESCC tissues/cells. CircCNOT6L knockdown attenuated ESCC cell proliferation and iron metabolism disorder, as well as accelerated apoptosis. Notably, circCNOT6L targeted miR-384, and miR-384 targeted FN1. MiR-384 depletion and FN1 upregulation weakened the effects of circCNOT6L knockdown and miR-384 overexpression on ESCC cell progression, respectively. Besides, circCNOT6L knockdown inhibited tumor growth in vivo. CONCLUSIONS: Our results demonstrated that circCNOT6L positively regulated the development of ESCC cells via modulating miR-384/ FN1 axis. 3. Synthesis, structural characterizations and magnetic properties of a series of mono-, di- and polynuclear manganese pyridinecarboxylate compounds

Abstract

Seven new manganese(II, III, IV) pyridinecarboxylate compounds (Et4N)[MnCl2(pic)(2)] (1, Hpic = picolinic acid) [MnCl(Pic)(2)(H2O)].H2O (2), [MnCl(pic)(H2O)2](n) (3), [Mn(Pic)(2)](n) (4), [MnCl(pic)](n) (5), [MnCl2(4-C5H4NHCOO)](n) (6) and [Mn2O2(pic)(4)] (7) were synthesized and structurally characterized. The picolinate ligand coordinates to the Mn ion forming a chelating five-membered ring which constructs diverse architectures by various bridging modes, such as mu-Cl, mu(1,1)(-) and mu(1,3)-carboxylate bridges (syn-syn and syn-anti modes). The interaction between the pyridyl rings is discussed, displaying a face-to-face pi-pi stacking for complex 6 and a T-shaped C-H...pi attraction for complex 4. Complex 3 has an infinite zigzag chain structure in which two neighboring Mn ions are linked by a carboxylate bridge in a syn-anti mode. In complex 4, the Mn(pic)(2) fragments are joined to each other by double mu(1,1)-carboxylate bridges, forming a 2D layer structure. Complex 5 contains Mn4O4 square subunits which are connected by double mu-Cl bridges to form a grid-like 2D structure. The isonicotinate complex 6 has a zigzag chain structure containing mixed double mu-Cl and mu(1,3)-carboxylate bridges in a syn-syn mode. Protonation at the pyridyl-N position was found and identified by the IR and magnetic properties of 6. The participation of an oxidizing agent like MnO4- leads to complexes containing higher valent manganese (1, 2 and 7). The IR spectra of these complexes are discussed and found to be consistent with the structural features. The magnetic properties of complexes 4, 5 and 6 have been investigated. Antiferromagnetic coupling interactions were observed and satisfactory fitting results were obtained with J values ranging from -0.63 cm(-1) to -2.35 cm(-1). The magnetic parameters of these complexes, together with the parameters of other Mn complexes with comparable structures, are compared and discussed based on the bridging modes. Abstract

Three novel manganese(II) coordination polymers, [Mn (maleate)(phen)](n) (1; phen = 1,10-phenanthroline), [Mn-(maleate)(phen)](n).nH(2)O (2), and [Mn(maleate)(bpy)](n) (3; bpy = 2,2'-bipyridine), have been synthesized by treatment of Mn2+ with maleic acid with participation of chelate diimine ligands, and have been identified by single-crystal, X-ray diffraction to have either one-dimensional (1D) zigzag chain structures (1 and 2) or a two-dimensional (2D) sinuous layer structure (3). Each maleate dianion coordinates to three Mn centers, in different bridging modes (syn-anti in 1 and 2, syn-syn and anti-anti in 3). These compounds represent an interesting example of structural topology variation from 1D to 2D mediated by chemically similar auxiliary chelate ligands. Variable-temperature magnetic susceptibility measurements show weak anti-ferromagnetic exchange interactions between the adjacent Mn-II ions, with J = -0.06 cm(-1) (2) and J = -1.3 cm(-1), zJ' = -0.27 cm(-1) (3). Abstract

Two pyz complexes [Mn(pyz)(2)(H2O)(4)] (1) and [MnCl(pyz)(H2O)] (2) have been synthesized and structurally characterized. Complex 1 contains an eight-coordinate Mn(II) ion with square anti-prismatic geometry. The extensive intermolecular hydrogen-bonding interactions of O-carboxyl...H-O-aqua correlate the complex molecules, forming 2D polymeric layer structure. The structure of complex 2 contains 1D Mn/pyz chains, which link to each other by double mu(2)-Cl bridges to form 2D layer with Mn...Mn distance of 3.664 Angstrom. Hydrogen-bonding interactions of O-carboxyl...H-O-aqua are found between the layers to correlate the 2D layers to form a 3D framework. In addition, the variable temperature magnetic susceptibilities of complex 2 were measured and weak ferromagnetic exchange interactions between the neighboring magnetic species were found with J = 0.42 cm(-1)., QC 20100525

Abstract

A reaction system consisting of terephthalic acid, NaOH, inorganic Mn(II) or Mn(III) salt, and salicylidene alkylimine resulted in dinuclear manganese complexes (salpn)(2)Mn-2(mu-phth)(CH3OH)(2) (1, salpn = N,N'-1,3-propylene-bis(salicylideneiminato); phth = terephthalate dianion), (salen)(2)Mn-2(mu-phth)(CH3OH)(2) (2, salen = N,N'-ethylene-bis(salicylideneiminato)), (salen)(2)Mn-2(mu-phth)(CH3OH)(H2O) (3), and (salen)(2)Mn-2(mu-phth) (4), while the absence of NaOH in the reaction led to a mononuclear Mn complex (salph)Mn(CH3OH)(NO3) (5, salph = N,N'-1,2-phenylene-bis(salicylideneiminato)). In addition, a trinuclear mixed metal complex H{Mn2Na(salpn)(2)(mu-OAc)(2)(H2O)(2)}(OAc)(2) (6) was obtained from the reaction system by using maleic acid instead of terephthalic acid. Five-coordinate Mn ions were found in 4 giving rise to an intermolecular interaction and constructing a one-dimensional linear structure. Antiferromagnetic exchange interactions were observed for 1-3, and a total ferromagnetic exchange of 4 was considered to stem from intermolecular magnetic coupling. H-1 NMR signals of phenolate ring and alkylene (or phenylene) backbone of the diamine are similar to those reported in the literature, and the phth protons are at -2.3 to -10.1 ppm. Studies on structure, bond valence sum analysis, and magnetic properties indicate the oxidation states of the Mn ions in 6 to be +3, which are also indicated by ESR spectra in dual mode. Ferromagnetic exchange interaction between the Mn(III) sites was observed with J = 1.74 cm(-1). A quasireversible redox pair at -0.29V/0.12V has been assigned to the redox of Mn-2(III)/Mn(III)Mn(II), implying the intactness of the complex backbone in solution., QC 20100525

12. Induction of cathepsin D protein during estrogen carcinogenesis: possible role in estrogen-mediated kidney tubular cell damage.

Abstract

We have proposed that an early step in estrogen carcinogenesis in the hamster kidney is tubular damage followed by reparative cell proliferation. This tubular injury is progressive and increases in severity with continued estrogen treatment; one pertinent feature is a marked rise in the number of both secondary and tertiary lysosomes. Data presented herein indicate that cathepsin D, an estrogen-responsive lysosomal proteolytic enzyme, is increased in the kidney following estrogen treatment in the hamster. Three isoforms of cathepsin D were detected in estrogen-treated kidneys, 52, 31, and 27 kDa, the major being 52 kDa. At 1 and 3 months of estrogen treatment, 52-kDa cathepsin D content increased 1.4- to 1.6-fold. These changes coincided with a rise in renal estrogen receptor levels during the same estrogen treatment periods. More pronounced rises in cathepsin D levels, 2.7- and 3.5-fold, were seen after 4 and 5 months of estrogen treatment, respectively. A concomitant, 3.0- to 4.0-fold rise in estrogen receptor content was also observed. At 5 months of estradiol or DES treatment, both 27- and 31-kDa isoforms were present in hamster kidneys, in addition to the 52-kDa form. Neither progesterone nor DHT treatment affected the untreated levels of cathepsin D. Interestingly, either concomitant tamoxifen or DHT and estrogen treatment prevented the rise in cathepsin D and estrogen receptor content observed after estrogen treatment alone. Primary estrogen-induced renal tumors and their metastases exhibited markedly elevated levels of all three isoforms of cathepsin D. Immunohistochemical analysis of cathepsin D in kidney sections confirmed the Western blot findings. These data suggest a novel role for estrogen-induced cathepsin D in the hamster kidney during tumorigenesis; that is, mediating renal tubular damage as a prelude to reparative cell proliferation, thus initiating a multi-step estrogen-driven process which leads to renal tumor formation. 16. Microporous Metal−Organic Frameworks Built on a Ln<INF>3</INF> Cluster as a Six-Connecting Node

Abstract

Two novel microporous metal−organic frameworks, {[Pr3(ATPT)2(HATPT)4]·(NO3)·8H2O}n (1) and {[Nd3(ATPT)3(HATPT)3]·9H2O}n (2) (H2ATPT = 2-aminoterephthalic acid), were synthesized by slow diffusion of aqueous solution of H2ATPT and Pr(NO3)3·6H2O or Nd(NO3)3·6H2O in 2-propanol. X-ray diffraction analyses reveal that their three-dimensional structures are composed of ATPT spacers and trinulcear lanthanide nodes, in which there are two crystallographically independent lanthanide atoms, nine- and twelve-coordinated. Twelve ATPT anions, adopting an unprecedented pentadentate η1:η1:η1:η2 bridging coordination mode, ligate the trinuclear node. Each pair of ATPT anions acts as a double bridge and is considered as one connection. Thus each trinuclear core is connected to the six nearest Ln3 neighbors forming a NaCl-type crystal lattice with a three-dimensional intersecting channel system of ca. 1.3-nm spacing between the centers of the adjacent clusters. The guest water molecules located in the large cavities can be removed without ruining the microporous framework. Thermal gravimetric and powder X-ray diffraction analyses confirm that small molecules, four methanol molecules per Pr3 unit and six methanol molecules per Nd3 unit, respectively, can be absorbed into the dehydrated microporous frameworks.

31. Promotion of estrogen-induced mammary gland carcinogenesis by androgen in the male Noble rat: probable mediation by steroid receptors

Abstract

Both endogenous and exogenous estrogen exposure is associated with an increased breast cancer risk. In some studies, elevated serum testosterone levels have also been linked to an increased breast cancer risk. Estrogen alone or combined with progesterone induces high mammary tumor incidences in various strains of both male and female rats. Mammary gland ductal adenocarcinomas were induced after 17β-estradiol (E2) and testosterone propionate (TP) treatment in male Noble rats. Tumor incidence was 100% after 8-9 months of treatment. Such neoplasms were not detected after either estrogen or androgen exposure alone within this time period. TP alone caused disruption of mammary gland ducts and proliferation of stromal tissue, while E2 treatment alone induced both ductal epithelial growth and nodular atypical hyperplasia. To study the interaction of these hormones in mammary tumorigenesis, sex hormone receptors were characterized in mammary glands of Noble rats. Estrogen receptor-α (ER) was detected in age-matched, untreated mammary gland epithelium; in most early atypical hyperplastic lesions appearing after E2 and E2 + TP treatment and in E2 + TP-induced mammary tumors. Two major ER putative isoforms, 116 and 120 kDa, were detected in E2- and E2 + TP-treated mammary glands, and in the induced tumors. A 54 kDa ER protein was found in untreated and TP-treated mammary glands and in the induced tumors. Both progesterone receptor-B (PR-B) and PR-A2, as well as androgen receptor-B (AR-B) and AR-A isoforms were markedly elevated in all E2 + TP-induced mammary tumors. However, the levels of both PR and AR were very low in mammary glands of E2- and E2 + TP-treated male rats. Low and moderate levels of AR and PR, respectively, were detected in most atypical hyperplastic lesions induced by E2- and E2 + TP-treated mammary glands. These results suggest that androgens may interact with either AR or PR, and perhaps both receptors, in E2 + TP-induced mammary glands and the induced tumors to effect the reduction in latency period, enhance tumor size, and increase incidence to 100%.
