Background: Febrile infection-related epilepsy syndrome is a refractory epilepsy syndrome that predominantly affects children, with a higher incidence in school-age children and is associated with infectious factors. Case report: A 6-year-old boy developed sudden symptoms characterized by fever and a disorder of consciousness, rapidly progressing into drug-refractory status epilepticus (SE). The electroencephalogram revealed diffuse slow-wave activity in the background, with partial seizures originating from the left temporal and occipital regions that gradually generalized. Brain MRI showed no abnormalities in the signal. Due to the failure of multi-drug anti-epileptic therapy, a ketogenic diet was administered and the frequency of seizure was reduced, however, a ketogenic diet was administered, which reduced the frequency of seizures; however, the ketogenic diet treatment was discontinued due to intolerance. After discharge, both the ketogenic diet and anti-seizure medications (ASMs) were administered to the patient. Upon follow-up, occasional seizures were reported, accompanied by unconsciousness and a significant decline in cognitive function compared to the pre-onset period. Conclusions: No specific treatment modality is available for febrile infection-related epilepsy syndrome; however, a ketogenic diet and vagus nerve stimulation (VNS) may offer potential efficacy in controlling acute seizures and improving prognosis.

3. A qualitative study on the caregiver burden experience in home reflux enema management of infants with congenital megacolon

Objective This study aims to explore the caregiver burden experience in the care of infants with congenital megacolon undergoing home reflux enema. The findings will provide a basis for developing targeted and effective nursing interventions. Methods A phenomenological research approach was employed. From October 2022 to January 2023, semi-structured in-depth interviews were conducted with 12 caregivers of infants with congenital megacolon undergoing home reflux enema in a tertiary paediatric hospital in Fujian Province. The collected data were analysed and organized using the Colaizzi’s 7-step analysis method, leading to the identification of key themes. Results The analysis yielded three major themes concerning the caregiver burden experience in the care of infants with congenital megacolon undergoing home reflux enema: inadequate disease-related knowledge, presence of multiple pressures during the caregiving process, and a desire for greater support. Conclusions This study employed qualitative interviews with the caregivers of 12 children with congenital Hirschsprung’s disease undergoing home reflux enema, and the feelings of caregivers of children with reflux enema at home after discharge were deeply understood. It is recommended to implement positive psychological interventions based on the PERMA model and incorporate “Internet + collaborative nursing” to provide caregivers with professional knowledge, address their pressures and needs, and promote their well-being while enhancing nursing abilities.

4. Phylogeographical and population genetics of Polyspora sweet in China provides insights into its phylogenetic evolution and subtropical dispersal

Abstract Background Geological movements and climatic fluctuations stand as pivotal catalysts driving speciation and phylogenetic evolution. The genus Polyspora Sweet (Theaceae), prominently found across the Malay Archipelagos and Indochina Peninsula in tropical Asia, exhibits its northernmost distribution in China. In this study, we investigated the evolutionary and biogeographical history of the genus Polyspora in China, shedding light on the mechanisms by which these species respond to ancient geological and climatic fluctuations. Methods Phylogenetic relationships of 32 representative species of Theaceae were reconstructed based on the chloroplast genome and ribosome 18-26 S rRNA datasets. Species divergence time was estimated using molecular clock and five fossil calibration. The phylogeography and population genetics in 379 individuals from 32 populations of eight species were analyzed using chloroplast gene sequences (trnH-psbA, rpoB-trnC and petN-psbM), revealing the glacial refugia of each species, and exploring the causes of the phylogeographic patterns. Results We found that Chinese Polyspora species diverged in the middle Miocene, showing a tropical-subtropical divergence order. A total of 52 haplotypes were identified by the combined chloroplast sequences. Chinese Polyspora exhibited a distinct phylogeographical structure, which could be divided into two clades and eight genealogical subdivisions. The divergence between the two clades occurred approximately 20.67 Ma. Analysis of molecular variance revealed that the genetic variation mainly occurred between species (77.91%). At the species level, Polyspora axillaris consists of three lineages, while P. speciosa had two lineages. The major lineages of Chinese Polyspora diverged between 12 and 15 Ma during the middle to late Miocene. The peak period of haplotype differentiation in each species occurred around the transition from the last interglacial to the last glacial period, approximately 6 Ma ago. Conclusion The primary geographical distribution pattern of Chinese Polyspora was established prior to the last glacial maximum, and the population historical dynamics were relatively stable. The geological and climatic turbulence during the Quaternary glacial period had minimal impact on the distribution pattern of the genus. The genus coped with Quaternary climate turbulence by glacial in situ survival in multiple refuges. The Sino-Vietnam border and Nanling corridor might be the genetic mixing center of Polyspora.

5. Study on Driver Gaze Characteristics in Sight Distance Limited Section of Mountain Highway Based on Visual Information

In order to study the gaze behavior characteristics of drivers in mountainous road sections with limited sight distance, the real vehicle test is carried out by using Smart Eye Pro 5.7 noninvasive eye tracker. Combined with the sight distance change rate theory, 6 typical test representative mountainous sections are selected to study the gaze distribution law and gaze duration of drivers in different mountainous sections. The research shows that when the driver drives on the test section with the most unfavorable sight distance of 44 m, 50 m, and 56 m, the fixation characteristics of “from far to near” are significant, and the long fixation duration accounts for a large proportion of the driver. When the driver drives on the section with the most unfavorable sight distance of more than 70 m, i.e., the sight distance change rate of less than 1.33, the fixation characteristics of “from far to near” disappear. The driver’s fixation stability increases, the fixation freedom increases, and the proportion of medium and long fixation duration decreases. The data analysis provides a theoretical basis for drivers to pass safely in mountainous sections.

6. The role of MTHFR C677T and ALDH2 Glu504Lys polymorphism in acute coronary syndrome in a Hakka population in southern China

Abstract Background Acute coronary syndrome (ACS) is the most serious type of coronary heart disease and is a global medical burden. The pathogenesis of ACS is very complex and still poorly understood. Epidemiologic studies have revealed that the manifestation of ACS are the results of the interactions between multiple environmental and genetic factors. The present study aimed to investigate the role of polymorphisms of MTHFR C677T and ALDH2 Glu504Lys as risk factors for ACS in a Hakka population in southern China. Methods Between September 1, 2015 and October 31, 2017, a total of 1957 individuals, including 860 ACS patients and 1097 controls were recruited. Blood samples were collected and genotypes were determined by DNA microarray chip method and direct sequencing method. Results For the MTHFR C677T polymorphism, frequencies of CC, CT, and TT genotypes were 53.60% versus 55.33, 39.53% versus 38.65 and 6.86% versus 6.02% in patients with ACS versus controls, respectively(p > 0.05). The differences in genotype frequencies between the ACS patients and controls in the three genetic model were not statistically significant. For the ALDH2 Glu504Lys polymorphism, the frequencies of ALDH2*1*1, ALDH2*1*2, and ALDH2*2*2 genotypes were 48.72, 42.67 and 8.6% in the ACS patients, respectively, while these were 53.33, 39.11 and 7.57% in the controls, respectively, showing no significant difference in the distribution of the ALDH2 genotype between the groups. Using the wild genotype ALDH2*1*1 as reference, relative risk analysis revealed a slightly increased risk for ACS in individuals with the ALDH2*1*2 plus ALDH2*2*2 genotypes (odds ratio (OR) = 1.203, 95% confidence interval (CI) = 1.006–1.438, p = 0.043). In a multivariate logistic regression model, even after adjusting for potential covariates, the association between ALDH2 *2 allele and ACS remained significant (OR = 1.242, 95% CI = 1.045–1.561, p = 0.038). Conclusions We present findings regarding the possible clinical impact of the ALDH2*2 variant on ACS patients in a Hakka population in southern China and our findings might help to stratify the high-risk ACS patients and implement appropriate strategies for this genetic subpopulation to ultimately guide the precision preventive procedures in the future.

Abstract Background The naked mole-rat (NMR, Heterocephalus glaber) is being bred as a novel laboratory animal due to its unique biological characteristics, including longevity, cancer resistance, hypoxia tolerance, and pain insensitivity. It is expected that differences exist between the microbiota of wild NMRs and that of NMRs in an artificial environment. Overall, the effect of environment on changes in the NMR microbiota remains unknown. In an attempt to understand the microbiota composition of NMRs in captivity, variability in the microbiota of the intestinal and respiratory tracts of two groups of NMRs was assessed under two conditions. Results The results obtained by high-throughput sequencing revealed significant differences at the phylum, class, order, family and genus levels in the microbiota between the two groups of NMRs examined (first group in conventional environment, second group in barrier environment). For the trachea, 24 phyla and 533 genera and 26 phyla and 733 genera were identified for the first and second groups of animals. Regarding the cecum, 23 phyla and 385 genera and 25 phyla and 110 genera were identified in the microbiota of first and second groups of animals. There were no obvious differences between females and males or young and adult animals. Conclusions Our results suggest that the intestinal and respiratory tract NMR microbiota changed during captivity, which may be related to the transition to the breeding environment. Such changes in the microbiota of NMRs may have an effect on the original characteristics, which may be the direction of further research studies.

Abstract Background Human papillomavirus (HPV) DNA testing is an important method in cervical cancer screening. However, the studies on prevalence and genotype distribution of HPV among women in northeastern Guangdong Province of China are very limited. Methods A total of 28,730 women attending the Department of Gynecology of Meizhou People’s Hospital (Huangtang Hospital), Meizhou Hospital Affiliated to Sun Yat-sen University between January 1st, 2013 and June 1st, 2015 were enrolled in this study. HPV type-specific distribution was tested using flow-through hybridization and gene chip. Results The overall prevalence of HPV infection was 19.81%, among which 79.09% were infected with high-risk HPV subtypes in the subjects. The 5 most predominant genotypes were HPV16, 52, 58, 18 and 81. Most HPV infections were observed in women aged 41–50 and women aged 30–59 accounted for a proportion of over 80%. Conclusions Our findings suggested a high burden of HPV infection among women in northeastern Guangdong Province of China. We identified the top 5 HPV genotypes as well as the age-specific distribution of HPV infections in this area.

The naked mole rat (NMR; Heterocephalus glaber) is a small rodent species found in regions of Kenya, Ethiopia and Somalia. It has a high tolerance for hypoxia and is thus considered one of the most important natural models for studying hypoxia tolerance mechanisms. The various mechanisms underlying the NMR's hypoxia tolerance are beginning to be understood at different levels of organization, and next-generation sequencing methods promise to expand this understanding to the level of gene expression. In this study, we examined the sequence and transcript abundance data of the muscle transcriptome of NMRs exposed to hypoxia using the Illumina HiSeq 2500 system to clarify the possible genomic adaptive responses to the hypoxic underground surroundings. The RNA-seq raw FastQ data were mapped against the NMR genome. We identified 2337 differentially expressed genes (DEGs) by comparison of the hypoxic and control groups. Functional annotation of the DEGs by gene ontology (GO) analysis revealed enrichment of hypoxia stress-related GO categories, including ‘biological regulation’, ‘cellular process’, ‘ion transport’ and ‘cell-cell signaling’. Enrichment of DEGs in signaling pathways was analyzed against the Kyoto Encyclopedia of Genes and Genomes (KEGG) database to identify possible interactions between DEGs. The results revealed significant enrichment of DEGs in focal adhesion, the mitogen-activated protein kinase (MAPK) signaling pathway and the glycine, serine and threonine metabolism pathway. Furthermore, inhibition of DEGs (STMN1, MAPK8IP1 and MAPK10) expression induced apoptosis and arrested cell growth in NMR fibroblasts following hypoxia. Thus, this global transcriptome analysis of NMRs can provide an important genetic resource for the study of hypoxia tolerance in mammals. Furthermore, the identified DEGs may provide important molecular targets for biomedical research into therapeutic strategies for stroke and cardiovascular diseases.

Polycomb group (PcG) proteins regulate the epigenetic status of transcription regulatory states during development. Progression from pluripotency to differentiation requires the sequential activation and repression of different PcG target genes, however, the relationship between early patterning signals, PcG expression, and the development of the central nervous system is still unclear. Using various models of neuronal differentiation, we provide evidence that CBX2 is a negative regulator of neuronal differentiation. Knock-down of CBX2 expression promotes neurite development, while overexpression of CBX2 inhibits neurite development. Further, we found that CBX2 is a direct target gene of miR-124. During neuronal differentiation, CBX2 was decreased while miR-124 was increased. Mechanistically, CBX2 directly interacts with the promoter region of several neuro-associated genes and regulates their expression. We found that the neuron-specific GAP-43 gene could contribute to the stimulating effect on neurite development associated with inhibition of CBX2.

11. The Protective Role of Autophagy in Heterocephalus glaber Hepatic Stellate Cells Exposed to H2O2 or Nutritional Stress

Background/Aims: Naked mole rats (NMRs) survive and thrive in dark, dank environments with low levels of oxygen and poor quality nutrition. Their long lifespan is attributed to sustained good health and pronounced resistance to cancer. Physiological and biochemical processes, such as autophagy, may contribute to the successful aging of this exceptionally long-lived species. We demonstrated that NMRs have higher levels of autophagy than short-lived C57BL/6 mice, and this may play an important role in the maintenance of cellular protein quality and the defense of cells against intracellular and extracellular aggressors in NMRs. The present study assesses autophagy as a means for cells to flexibly respond to environmental changes (H2O2 treatment and a shortage of nutrients). Methods: Primary NMR HSCs were isolated from liver and treated with serum-free medium. Cells in the experimental group were incubated with different concentrations of hydrogen peroxide (H2O2) in the presence and / or absence of 3-MA (5 mM).The LC3-II/LC3-I ratio was determined by western blot analysis. Western blotting was performed to analyze the expression level of Beclin 1 protein. Apoptosis and cell-cycle progression were analyzed by flow cytometry. Results: Our data reveal that both poor quality nutrition and H2O2 treatment induces apoptosis and autophagy in NMR hepatic stellate cells(HSCs). Conclusion: NMR cells have the capacity to induce cell death through apoptosis and downregulate the energy consuming processes through inhibition of proliferation when they become superfluous or irreversibly damaged.

12. High Autophagy in the Naked Mole Rat may Play a Significant Role in Maintaining Good Health

Background/Aims: The maximum lifespan of the naked mole rat is over 28.3 years, which exceeds that of any other rodent species, suggesting that age-related changes in its body composition and functionality are either attenuated or delayed in this extraordinarily long-lived species. However, the mechanisms underlying the aging process in this species are poorly understood. In this study, we investigated whether long-lived naked mole rats display more autophagic activity than short-lived mice. Methods: Hepatic stellate cells isolated from naked mole rats were treated with 50 nM rapamycin or 20 mM 3-methyladenine (3-MA) for 12 or 24 h. Expression of the autophagy marker proteins LC3-II and beclin 1 was measured with western blotting and immunohistochemistry. The induction of apoptosis was analyzed by flow cytometry. Results: Our results demonstrate that one-day-old naked mole rats have higher levels of autophagy than one-day-old short-lived C57BL/6 mice, and that both adult naked mole rats (eight months old) and adult C57BL/6 mice (eight weeks old) have high basal levels of autophagy, which may be an important mechanism inhibiting aging and reducing the risk of age-related diseases. Conclusion: Here, we report that autophagy facilitated the survival of hepatic stellate cells from the naked mole rat, and that treatment with 3-MA or rapamycin increased the ratio of apoptotic cells to normal hepatic stellate cells.

Background and objective Livin is a novel inhibitor of apoptosis protein (IAP), recent studies showed it overexpresses in a variety of carcinomas including lung cancer and contributes much to the cancerous development. The objective of this study is to explore the expression of livin in tissues of lung cancer and its relationshipwith histological types, chemotherapy, Lymph node metastasis and to study its correlation with the expression of pro-caspase3 as well. Methods Expressions of Livin and caspase3 were detected by Western blot assay in lung cancer tissues as well as in controls. Results Livin was expressed in 15 of 27 lung cancer, significantly more than those in lung para-cancerous (1/5) or benign disease lung tissues (2/12) (P 0.05). Conclusion Livin are differently expressed in different histological types of lung cancer; High levels of livin expression do not relate to chemotherapy, lymph node metastasis (P >0.05). The levels of livin tends to be positively associated with those of accordingly pro-caspase3, it is presumed that livin could bind pro-caspase3 and suppress its activation.

DNA fragmentation is a well-recognized hallmark of apoptosis. However, the precise DNA sequences cleaved during apoptosis triggered by distinct mechanisms remain unclear. We used next-generation sequencing of DNA fragments generated in Actinomycin D-treated human HL-60 leukemic cells to generate a high-throughput, global map of apoptotic DNA breakpoints. These data highlighted that DNA breaks are non-random and show a significant association with active genes and open chromatin regions. We noted that transcription factor binding sites were also enriched within a fraction of the apoptotic breakpoints. Interestingly, extensive apoptotic cleavage was noted within genes that are frequently translocated in human cancers. We speculate that the non-random fragmentation of DNA during apoptosis may contribute to gene translocations and the development of human cancers.

22. Prevalence of hepatitis B virus infection among pregnant women in the mountainous regions of southern China: A retrospective single‐center study

Hepatitis B virus (HBV) infection remains a major public health issue worldwide. Moreover, its prevalence varies significantly in different geographic areas of China. The current study aimed to assess the prevalence of HBV infection among Hakka pregnant women in Meizhou, a remote mountainous region in southern China.This research was performed between January 2015 and December 2020. In total, 16,727 pregnant women receiving antenatal care at Meizhou People's Hospital were included in the analysis. All pregnant women were screened for serum HBV markers.The prevalence rates of hepatitis B surface antigen (HBsAg) and hepatitis B surface antibody positivity among the participants were 11.74% (n = 1964) and 48.00% (n = 8029), respectively. The overall prevalence rates of susceptibility to infection, HBV immunity, previous/occult infection, inactive HBsAg carrier, and active infection were 36.16%, 33.61%, 16.94%, 8.11%, and 2.30%, respectively. According to age distribution, the prevalence rate of HBsAg positivity elevated concomitantly with increasing age (p 0.001). From 2015 to 2020, the prevalence rate of HBsAg positivity decreased from 14.50% to 8.19% and that of hepatitis B pre-core antigen positivity from 4.42% to 2.31%. In addition, pregnant women with HBsAg-positive status were more likely to present with gestational diabetes, thrombocytopenia, and anemia than those with HBsAg-negative status.The HBV infection rate remains high among pregnant women in the indigenous Hakka population in southern China. To prevent vertical transmission, cautious surveillance of maternal HBV infection status should be considered in Hakka pregnant women in Meizhou.

Orange peel waste (OPW) and sewage sludge (SS) valorization for volatile fatty acids (VFAs) production from anaerobic co-fermentation are attractive and feasible. The highest VFAs reached 11996.3 mg COD/L within 10 d at the mass ratio (TS/TS) of 1:1, which was approximately 30-fold of that in sole SS fermentation. The OPW provided plenty of organic substrates and facilitated the fermentation processes by disintegrating SS structure and inhibiting methanogenesis due to the abundant limonene. Also, the OPW feeds reshaped the microbial community and enriched fermentative bacteria, especially those saccharolytic ones (i.e. Prevotella-7). The key genes involved in membrane transport (i.e. ptsG), glycolysis (i.e. pgk), pyruvate metabolism (i.e. ace), and fatty acid biosynthesis (i.e. accA), which are associated with VFAs biosynthesis, were up-regulated in OPW/SS reactors. Overall, it was the increase in bioavailable organic matter and functional microorganisms, and the simultaneous enhancement of metabolic activity that improved the efficient VFAs production.

The reactions of K2ZnSb (0.234 mol) and Ni(CO)2(PPh3)2 (x mol) in ethylenediamine (en) solutions of 18-crown-6 at 60 °C yielded half-centered [Ni0.5@{Sb6Ni6(CO)8}]4− (x = 0.023, 1) and fully embedded [Ni@{Sb7Ni5(CO)6}]3− (x = 0.047, 2), wherein the chair-like Sb6 subunit is bicapped by two Ni3 triangles to form the single Sb6Ni6 icosahedron of 1 and by the triangular Ni2Sb and Ni3 to form the single Sb7Ni5 icosahedron of 2. Surprisingly, a smaller amount of Ni(CO)2(PPh3)2 (x = 0.016) led to the formation of the unprecedented dimer [(Sb8){Sb7Ni5(CO)4}2]6− (4), wherein the Sb7Ni5 icosahedral unit built of boat-like Sb6, Ni2Sb and Ni3 is linked together by the realgar-like Sb8 to form the icosahedral dimer 4, providing a new way of thinking for the preparation of icosahedral oligomers/polymers. A similar hexa-anionic dimer (3) can be solely obtained using PBu4+ as the countercation under the same reaction conditions. The structure and bonding of this novel series of icosahedral clusters containing polyantimony were discussed for the first time.

In this research, we present the first examples of MoIV3–WVI hybrid polyoxometalates (MoIV3-POMs) with empty Keggin structures: α-[MoIV3MoVIWVI8O31(OH)6py3]·2Hpy·4H2O (1·2Hpy·4H2O), γ-[MoIV6WVI6O28(OH)6py6]·2[H2N(CH3)2]·6.5H2O (2·2[H2N(CH3)2]·6.5H2O), γ-[MoIV6WVI6O29(OH)5py6Na(H2O)5]·2[NH2(CH3)2]·2H2O (3·2[NH2(CH3)2]·2H2O) and γ-[{MoIV6MoVIWVI5O28(OH)6py6}2Znpy4]·2Hpy·8H2O (4·2Hpy·8H2O). They were prepared through the solvothermal partial oxidation of a mixture of triangular bioxo-capped hexacarboxylate clusters [MoIVxWIV3−xO2(O2CCH3)6L3]q (x = 0–3, L = H2O, q = 2+; L = CH3CO2−, q = −1), providing a feasible route to the designed synthesis of mixed MoIV3–W POMs. The much lower yield and poor repeatability of 1 compared to those of 2, 3, and 4 are ascribed to the fact that the triangularly MoIV–MoIV bonded MoIV3 Lewis acid cluster unit of the former results from the breakage of MIV–WIV bonds (M = Mo, W) and recombination of MoIV in the Na[MoIVWIV2O2(O2CCH3)9] precursor because of the extremely easy oxidation of WIV to WVI as compared to MoIV. 2, 3, and 4 exhibit highly LCF-dependent (LCF = Lewis catalysis field) hydrogen transfer catalysis performance in the hydrazine reduction of nitrobenzene to aniline.

In the presence of the roughly flat sequestering agent, [K(18-crown-6)]+, the reaction of Rh2(CO)4Cl2 with K5Bi4 in ethylenediamine (en) solution at room temperature yielded the heterometallic cluster anion [Rh@Bi10(RhCO)6]3- (1), in which two hitherto unknown Binm- building blocks (i.e., Bi6 crown and Bi4 pyramid) were stabilized by six +Rh-CO units. When the reaction was carried out at 60 °C using Rh(acac)(CO)2 (acac = acetylacetonate) as the source of +Rh-CO units, one obtained the anion [Rh@Bi9(RhCO)5]3- (2) in which two different Binn- units (n = 2, 3) and two weakly bonded Bi atoms were stabilized by five +Rh-CO units. The structures and bonding of the novel heterometallic cluster anions 1 and 2 were discussed.

Wilms tumor is the most common childhood kidney malignant tumor. However, the genes and signaling pathways associated with the disease remain incompletely understood.GSE66405, GSE73209, and GSE11151 were collected from the Gene Expression Omnibus (GEO) database, and differentially expressed genes (DEGs) were detected using R software. A protein-protein interaction (PPI) network was constructed using the STRING database, and the clustering modules and hub genes were analyzed with the Cytoscape software. Genes functional enrichment analyses were performed using the package "clusterProfiler" in R software, and the gene set enrichment analysis (GSEA) analysis was performed using GSEA v4.1.0 software.Respectively, 3,092, 620, and 3,567 DEGs were screened in GSE66405, GSE73209, and GSE11151, with a total of 474 common DEGs detected in three expression profiles. For the common DEGs, the top 30 significant results of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways enrichment analyses were presented. Furthermore, five modules were found as the most related modules to Wilms tumor. GO term and KEGG pathway enrichment analyses of the genes in all the modules identified 10 GO terms and 5 KEGG pathways as significantly enriched. The top 10 hub DEGs of the PPI network wereOur study revealed robust gene signatures in Wilms tumor. Dysregulations of the signaling pathways were associated with the development and progression of the Wilms tumor, and 10 hub genes may play important roles in its diagnosis and therapy.

This work explored the feasibility of paper waste (PW)/sewage sludge (SS) co-fermentation for volatile fatty acids (VFAs) production, and disclosed its correlation with the key operational parameters (i.e., pH and PW/SS ratio). The results indicated that the maximal VFAs was 251.55 mg COD/g TSS at optimal conditions, which was approximately 10-folds of sole SS fermentation. PW feeding contributed to the bioavailable substrates and C/N balance during co-fermentation process. The pH exhibited evident impacts on organics solubilization/hydrolysis, in which acidic pH was more beneficial for carbohydrates metabolism while alkaline pH was better for proteins. Under optimal operational conditions, the metabolic functions associated with VFAs production (i.e., substrate membrane transport, intracellular metabolism and VFAs biosynthesis) were up-regulated. Moreover, functional microorganisms (i.e., Saccharofermentans and Bacteroides) responsible for VFAs generation were enriched. This work provided an innovative approach to recovery valuable products from biowastes, and in-depth understandings of microbial features in PW/SS co-fermentation systems.

The aim of this study is to clarify the possible association between methylenetetrahydrofolate reductase (MTHFR) C677T polymorphism and pre-eclampsia in Hakka pregnant women in southern China. Pre-eclampsia and normal pregnant women were consecutively collected and MTHFR C677T genotypes were determined by the DNA sequencing method. One hundred and thirteen pre-eclampsia patients were CC homozygote (113 of 191, 59.2%), 68 of 191 (35.6%) were CT heterozygote, and 10 of 191 (5.2%) were TT homozygote, with the frequency of the T allele equal to 0.77. This is in comparison with the normal control group where 106 of 202 (52.5%) were CC homozygote, 83 of 202 (41.1%) were CT heterozygote, and 13 of 202 (6.4%) were TT homozygote, with the frequency of the T allele equal to 0.27. No statistically significant differences were observed in genotype or allele frequencies between the pre-eclampsia and normal control for the C677T polymorphism of MTHFR gene (

Engineering vehicles on construction sites mainly include excavators, wheeled cranes and so on. If the engineering vehicle is working under or near the high-voltage power line, its bucket or boom probably enter the high-voltage breakdown range when they are lifted, which is very easy to result in accidents such as short circuit breakdown. So, it is necessary to find out the engineering vehicles working near high-voltage power line during inspection. Unmanned aerial vehicle (UAV) inspection is one of the main methods of electric power inspection at present. Lots of images are produced by UAV during the power line inspection. It will save a lot of inspection work if the engineering vehicles working near high-voltage power line can be recognized from these images. First, this paper analyzes the specific requirements of engineering vehicle recognition from aerial images of UAV power line inspection. Then, based on the research status of vehicle recognition in aerial images and other related fields at domestic and abroad, this paper comprehensively analyzes the research status of classical pattern recognition method and deep neural network method to recognize engineering vehicles in aerial images of UAV. Third, in view of the practical problems such as the low aerial image data of engineering vehicles, the roadmap of recognizing the engineering vehicles in the aerial image of UAV using the capsule network method is designed.

Bifunctional catalysis of frustrated Lewis pairs (FLPs) is one of the most active frontier areas in catalytic hydrogenation. The first MoIV-γ-Keggin-like cluster H2[(py3MOIV3)2MOVI7O32(OH)4] (γ-1, py = pyridine), which contains unprecedented MoIV–O–MoVIO FLPs (POM-FLPs), was prepared in a high yield. The synergistic effect of the POM-FLPs accounts for the high efficiency in catalytic hydrogenation as exemplified by the excellent performance (yield > 99%) of γ-1 as the heterogeneous catalyst in the hydrogenation reduction of nitroarenes to anilines with hydrazine hydrate under mild conditions, revealing the promising potential of such unique MOIV3–POMoVI FLP systems in catalytic hydrogenation.

In multicellular organisms, receptor tyrosine kinases (RTKs) control a variety of cellular processes, including cell proliferation, differentiation, migration, and survival. Sprouty (SPRY) proteins represent an important class of ligand-inducible inhibitors of RTK-dependent signaling pathways. Here, we investigated the role of SPRY1 in cells of the central nervous system (CNS). Expression of SPRY1 was substantially higher in neural stem cells than in cortical neurons and was increased during neuronal differentiation of cortical neurons. We found that SPRY1 was a direct target gene of the CNS-specific microRNA, miR-124 and miR-132. In primary cultures of cortical neurons, the neurotrophic factors brain-derived neurotrophic factor (BDNF) and Basic fibroblast growth factor (FGF2) downregulated SPRY1 expression to positively regulate their own functions. In immature cortical neurons and mouse N2 A cells, we found that overexpression of SPRY1 inhibited neurite development, whereas knockdown of SPRY1 expression promoted neurite development. In mature neurons, overexpression of SPRY1 inhibited the prosurvival effects of both BDNF and FGF2 on glutamate-mediated neuronal cell death. SPRY1 was also upregulated upon glutamate treatment in mature neurons and partially contributed to the cytotoxic effect of glutamate. Together, our results indicate that SPRY1 contributes to the regulation of CNS functions by influencing both neuronal differentiation under normal physiological processes and neuronal survival under pathological conditions.

Background Paclitaxel (PTX) has remarkable anti-tumor activity, but it causes severe toxicities. There is an urgent need to seek an appropriate pharmacokinetic parameter of PTX to improve treatment efficacy and reduce adverse effects. Objective To evaluate the association of pharmacokinetic parameter TC > 0.05 of paclitaxel (PTX) and its therapeutic efficacy and toxicity in patients with solid tumors. Methods A total of 295 patients with ovarian cancer, esophageal cancer, breast cancer, and non-small cell lung cancer (NSCLC), who were admitted to the Tumor Hospital of Shantou University Medical College, China, were recruited for this study. Patients received 3 weeks of PTX chemotherapy. The plasma concentrations of PTX were examined using the MyPaclitaxel™ kit. The patients' PTX TC > 0.05 (the time during which PTX plasma concentration exceed 0.05µmol/L) were calculated based on pharmacokinetic analysis. Results The results showed that: (1) the concentrations of PTX in these 295 patients ranged from 0.0358-0.127 µmol/L; (2) the PTX TC > 0.05 ranged from 14 to 38h with a median time of 27h; (3) among all treatment cycles, there was a statistically significant difference in the PTX TC > 0.05 between CR+PR and SD+PD; (4) with the increasing value of TC > 0.05, level of leukopenia and leukopenic fever increased; (5) high PTX TC > 0.05 led to the occurrence of neutropenia, neutropenic fever, severe anemia, and severe peripheral neurotoxicity. Taken together, our results indicated that the pharmacokinetic parameter PTX TC > 0.05 was an effective measure of treatment efficacy and toxicity in patients with solid tumors. Maintaining PTX TC > 0.05 at 26 to 30h could improve its efficacy and reduce the incidence of leukopenia, neutropenia, anemia, and peripheral neurotoxicity in these patients. Conclusion PTX TC > 0.05 is a key pharmacokinetic parameter of PTX which should be monitored to optimize individual treatment in patients with solid tumors.

IntroductionAltrenogest (ALT) is widely used to regulate the estrous cycle of sows and mares; however, currently used oral solutions of ALT are deficient in terms of dose accuracy and stability during use. To resolve these problems, we aimed to prepare softgel capsules of ALT with a unit dose equal to the clinically administered dose.MethodThe shell of the softgel capsule was mainly composed of gelatin and glycerol, with titanium dioxide and red iron oxide as masking agents. Using the shake flask method, the contents were screened for ethyl acetate as a lipophilic solubilizing carrier based on soya bean oil. The contents were automatically filled and compressed into softgel capsules by a softgel capsule machine. The quality of the prepared softgel capsules was evaluated through a drug content test, an in vitro release test, and stability studies. Furthermore, bioequivalence studies were conducted with atrenogest oral solution.ResultsThe content is an ALT concentration of 2% oil solution and the specification of the softgel is 20 mg/capsule. In in vitro dissolution experiments, the softgel capsules were rapidly disintegrated and released in three different pH buffers, with a cumulative release rate of nearly 100% at 1 h. The softgel capsules were stable at high temperature and under strong light for 10 days, and the concentration of ALT was >99% in the 6-month accelerated and long-term tests. In the bioequivalence study, Tmax of the softgel capsules was 2.20 ± 0.77 h, t1/2 was 6.36 ± 1.74 h, and Cmax was 64.65 ± 20.69 ng/ml. The main pharmacokinetic parameters Tmax, Cmax, AUC0−t, and AUC0−∞, did not differ significantly between the softgel capsules and the commercially available ALT oral solution (P > 0.05), and bioequivalence was demonstrated within the 90% confidence interval.ConclusionThe prepared softgel capsules have the advantages of higher content, ease of use with accurate dosing, good stability, and equivalence to ALT oral solution, implying that our softgels are ready for clinical use.

Microencapsulated essential oils (MEO)have been used as antibiotic alternatives that can be applied to alleviate diarrhea in weaning piglet. We examined a large group of weaned piglets and incorporated essential oil containing thymol (2%), carvacrol (5%) and cinnamaldehyde (3%) in the feed of weaned piglets on an intensive production farm. The piglets were divided into four groups; Control (no additions) and chlortetracycline (Chl), essential oil (EO) and microencapsulated essential oil (MEO) were fed ad libitum over a 28-day trial period. We found MEO significantly reduced the incidence of diarrhea in the piglets that was also accompanied by increased average daily weight gains from days 14–28 (p 0.05). Metagenomic analysis indicated that MEO impacted DNA expression, virulence factors, antioxidant activity and antimicrobial activity. Metabolomic analysis of the intestinal content also indicated that MEO impacted tyrosine metabolism and primary bile acid biosynthesis suggesting improved intestinal health and nutrient absorption. This study paves the way for further research into the development and optimization of MEO-based interventions aimed at improving piglet health and performance while also providing a reference for reducing reliance on antibiotics in animal agriculture.

Fluorouracil (5-FU) has been wildly used as a primary medication in the treatment of solid tumors including colorectal cancer. The treatment efficacy and toxicity of 5-FU varies greatly among individuals, suggesting a need for individualized regimen for cancer patients. The present study analyzed the blood concentration of 5-FU and its therapeutic efficacy and toxicity, evaluated the relationship of AUC (area under the plasma concentration-time curve), and the protein expression of DPD (dihydropyrimidine dehydrogenase) and TS (thymidylate synthetase), and therapeutic efficacy and toxicity. It was found that the AUC of 5-FU was 34.16±14.83mgmg·h/L in this cohort of study. The immunohistochemical analysis revealed 38.96% and 81.82% positive staining for DPD and TS in colorectal cancer tissues, respectively. We demonstrated that the expression of TS is positively correlated with the expression of DPD. There was a positive correlation between AUC and therapeutic efficacy, and gastrointestinal tract and neural toxicity. The expression of neither DPD nor TS had significant correlations with therapeutic efficacy and toxicity. Based on the blood 5-FU concentration and its relationship with treatment efficacy and toxicity, we determined an optimal therapeutic dosage of 5-FU to be equivalent to an AUC=28.03-38.94mgmg·h/L. Our study will be helpful in providing an individualized medical regimen for the treatment of colorectal cancer patients.

Hearing loss is a common neurosensory disorder, approximately half of the cases are caused by genetic factors, and approximately 70% of hereditary hearing impairments are nonsyndromic hearing loss (NSHL). The mutations of GJB2 (gap junction beta-2 protein), GJB3 (gap junction beta-3 protein), SLC26A4 (solute carrier family 26 member 4), and MT-RNR1 (mitochondrially encoded 12S RNA) are the most common inherited causes of NSHL. Because of different genetic backgrounds, the mutation spectrum of these common deafness-causing genes varies among different regions in China. Because no data are known on these mutations among the Hakka population of Southern China, we aim to investigate the mutation spectrum to add these to neonatal screening and genetic counseling. A total of 1252 blood samples from newborns have been detected by semiconductor sequencing for 100 mutations loci of 18 deafness-causing genes. Of the participants, 95 subjects carried deafness-causing genes mutations with the carrier rate of 7.59%. The mutation frequencies of GJB2, SLC26A4, GJB3, and mitochondrial genes were 3.04%, 3.51%, 0.16%, and 0.88%, respectively. We followed up subjects with single-gene homozygous or compound heterozygous mutations. Our study firstly analyzed deafness-causing genes mutation spectrum in Hakka population, providing evidence for future neonatal screening and genetic counseling in this area.

MicroRNA-124 (miR-124), a brain-specific miRNA molecule, has been implicated in stimulating neurite outgrowth and elongation during neuronal differentiation. However, the direct target genes and the mechanisms of miR-124-induced neurite outgrowth are poorly understood. In this study, we demonstrated that miR-124 directly targeted and downregulated the endogenous expression of oxysterol-binding protein (OSBP). A previous study found that the expression of miR-124 increased during brain development. In the present study, we demonstrated that the expression of OSBP decreased during the development of the C57BL/6 mouse cortex, which was negatively correlated with miR-124 expression. OSBP knockdown using specific shRNAs promoted neurite outgrowth and elongation in both Neuro-2a cells and primary cultured mouse cortical neurons. Conversely, OSBP overexpression strongly repressed the neurite elongation-enhancing effect of miR-124 in Neuro-2a cells. Our results suggested that OSBP may be a target and downstream effector of miR-124 for regulating neurite outgrowth and elongation.

Statins are potent cholesterol-lowering drugs and are generally well tolerated. Hepatotoxicity is a rare but serious adverse effect of statins; however, its mechanisms are not clear. Coenzyme Q10 deficiency has been suggested, and supplementation of reduced coenzyme Q10 (ubiquinol) has been shown to have hepatoprotective effects. MicroRNAs (miRNAs) are small nucleotides that have been shown to be up-regulated in drug-induced liver injury. We hypothesized that circulating miRNAs may be differentially regulated after simvastatin treatment and by comparing with that of simvastatin and ubiquinol supplementation could potentially uncover signatory miRNA profile for simvastatin-induced liver injury. In this double-blind, prospective, randomized-controlled trial, miRNA profiles and liver enzymes were compared between simvastatin-treated patients, with and without ubiquinol supplementation, over 12 weeks compared to baseline. miRNA expression was further validated in HepG2 liver cell lines by real-time PCR. Polycomb group (PcG) proteins regulate the epigenetic status of transcription regulatory states during development. Progression from pluripotency to differentiation requires the sequential activation and repression of different PcG target genes, however, the relationship between early patterning signals, PcG expression, and the development of the central nervous system is still unclear. Using various models of neuronal differentiation, we provide evidence that CBX2 is a negative regulator of neuronal differentiation. Knock-down of CBX2 expression promotes neurite development, while overexpression of CBX2 inhibits neurite development. Further, we found that CBX2 is a direct target gene of miR-124. During neuronal differentiation, CBX2 was decreased while miR-124 was increased. Mechanistically, CBX2 directly interacts with the promoter region of several neuro-associated genes and regulates their expression. We found that the neuron-specific GAP-43 gene could contribute to the stimulating effect on neurite development associated with inhibition of CBX2.

High-temperature superconductivity is closely adjacent to a long-range antiferromagnet, which is called a parent compound. In cuprates, all parent compounds are alike and carrier doping leads to superconductivity, so a unified phase diagram can be drawn. However, the properties of parent compounds for iron-based superconductors show significant diversity and both carrier and isovalent dopings can cause superconductivity, which casts doubt on the idea that there exists a unified phase diagram for them. Here we show that the ordered moments in a variety of iron pnictides are inversely proportional to the effective Curie constants of their nematic susceptibility. This unexpected scaling behavior suggests that the magnetic ground states of iron pnictides can be achieved by tuning the strength of nematic fluctuations. Therefore, a unified phase diagram can be established where superconductivity emerges from a hypothetical parent compound with a large ordered moment but weak nematic fluctuations, which suggests that iron-based superconductors are strongly correlated electron systems., Comment: 5 pages, 2 figures

Soluble epoxide hydrolase (sEH) inhibition has been demonstrated to have beneficial effects on various diseases, such as hypertension, diabetes, and brain ischemia. However, whether sEH inhibition has therapeutic potential in Parkinson's disease is still unknown. In this paper, we found that sEH expression is increased in 1-methyl-4-phenyl-1,2,3,6-tetrahydro pyridine (MPTP)-treated mice, and sEH deficiency and inhibition significantly attenuated tyrosine hydroxylase (TH)-positive cell loss and improved rotarod performance. The substrate of sEH, 14,15-epoxyeicosatrienoic acid (14,15-EET), protected TH-positive cells and alleviated the rotarod performance deficits of wild-type mice but not sEH-knockout mice. Moreover, the 14,15-EET antagonist 14,15-epoxyeicosa-5(Z)-enoic acid (14,15-EEZE) abolished the neuronal protective effects of sEH deficiency. In primary cultured cortical neurons, MPP(+) induced significant Akt inactivation in neurons from sEH wild-type mice, and this effect was not observed in neurons from knockout mice. Our data indicate that sEH deficiency and inhibition increased 14,15-EET in MPTP-treated mice, which activated the Akt-mediated protection of TH-positive neurons and behavioral functioning. We also found that sEH deficiency attenuated TH-positive cell loss in a paraquat-induced mouse model of Parkinson's. Our data suggest that sEH inhibition might be a powerful tool to protect dopaminergic neurons in Parkinson's disease.

The consumption of a high-fat diet (HFD) and obesity have been associated not only with metabolic diseases but also with neuropsychiatric diseases, such as depression and anxiety. However, results on the effects of an HFD on anxiety are controversial, since both anxiogenic and anxiolytic effects have been reported. In this study, we evaluated the effects of both short- and long-term intake of an HFD on anxiety-like behaviors. To explore the impact of time on the association between an HFD and anxiety, mice were fed with an HFD for 4 weeks or 12 weeks. Compared with control-diet mice, mice given an HFD for 4 weeks displayed anxiolytic-like behaviors. At the same time, we observed decreased SIRT1 expression in the mPFC and the amygdala of HFD-fed mice. Moreover, resveratrol, an activator of SIRT1, reversed the anxiolytic-like behaviors in HFD-fed mice. However, after 12 weeks of consuming a high-fat diet, mice did not exhibit any anti-anxiety behavior or further decreases in SIRT1 expression in the aforementioned brain regions compared with CD-fed mice. When EX-527, a SIRT1 inhibitor, was intraperitoneally injected, we observed anxiolytic effects in the CD-fed mice but not in the 12-week HFD-fed mice. Collectively, our data demonstrate that exposure to a short-term HFD can induce anxiolytic behaviors, which may be associated with decreased SIRT1 in the mPFC and the amygdala. However, this effect is abolished when the high-fat diet is extended to 12 weeks. Together, these results demonstrate that SIRT1 plays an essential role in regulating mood-related behaviors in HFD-fed mice.

We use transport and neutron scattering to study the electronic phase diagram and spin excitations of NaFe$_{1-x}$Cu$_x$As single crystals. Similar to Co- and Ni-doped NaFeAs, a bulk superconducting phase appears near $x\approx2\%$ with the suppression of stripe-type magnetic order in NaFeAs. Upon further increasing Cu concentration the system becomes insulating, culminating in an antiferromagnetically ordered insulating phase near $x\approx 50\%$. Using transport measurements, we demonstrate that the resistivity in NaFe$_{1-x}$Cu$_x$As exhibits non-Fermi-liquid behavior near $x\approx1.8\%$. Our inelastic neutron scattering experiments reveal a single neutron spin resonance mode exhibiting weak dispersion along $c$-axis in NaFe$_{0.98}$Cu$_{0.02}$As. The resonance is high in energy relative to the superconducting transition temperature $T_{\rm c}$ but weak in intensity, likely resulting from impurity effects. These results are similar to other iron pnictides superconductors despite the superconducting phase in NaFe$_{1-x}$Cu$_x$As is continuously connected to an antiferromagnetically ordered insulating phase near $x\approx 50\%$ with significant electronic correlations. Therefore, electron correlations is an important ingredient of superconductivity in NaFe$_{1-x}$Cu$_x$As and other iron pnictides., Comment: accepted for publication in PRB

The class IIa histone deacetylases (HDACs) play important roles in the central nervous system during diverse biological processes such as synaptic plasticity, axon regeneration, cell apoptosis, and neural differentiation. Although it is known that HDAC5 regulates neuronal differentiation, neither the physiological function nor the regulation of HDAC5 in neuronal differentiation is clear. Here, we identify HDAC5 as an inhibitor of neurite elongation and show that HDAC5 is regulated by the brain enriched microRNA miR-124 and miR-9. We discover that HDAC5 inhibits neurite extension both in differentiated P19 cells and primary neurons. We also show that the neuronal membrane glycoprotein GPM6A (M6a) is a direct target gene of HDAC5 regulated transcriptional factor MEF2C. HDAC5 inhibits neurite elongation, acting at least partially via a MEF2C/M6a signaling pathway. We also confirmed the miR-124/miR-9 regulated HDAC5-MEF2C-M6a pathway regulates neurite development in primary neurons. Thus, HDAC5 emerges as a cellular conductor of MEF2C and M6a activity and is regulated by miR-124 and miR-9 to control neurite development.

Short multi-walled carbon nanotubes (S-MWCNTs) were obtained by ball milling method and employed as a supporting matrix to explore a novel immobilisation and biosensing platform of redox proteins such as haemoglobin (Hb). Compared to long MWCNTs, the shortened MWCNTs-based biosensor did not show better electrocatalytic activity, which was contrary to previous theoretical expectations. The electron transfer rate constants of Hb on long and short MWCNTs modified electrodes in phosphate buffer solution were 1.08 and 0.95 s−1, respectively. The lower electron transfer rate between haemoglobin and the electrode can be attributed to their larger resistive properties, which were confirmed by cyclic voltammetry and AC impedance.
