Gully erosion can lead to the destruction of farmland and the reduction in crop yield. Gully mapping from remote sensing images is critical for quickly obtaining the distribution of gullies at regional scales and arranging corresponding prevention and control measures. The narrow and irregular shapes and similar colors to the surrounding farmland make mapping erosion gullies in sloping farmland from remote sensing images challenging. To implement gully erosion mapping, we developed a small training samples-oriented lightweight deep leaning model, called asymmetric non-local LinkNet (ASNL-LinkNet). The ASNL-LinkNet integrates global context information through an asymmetric non-local operation and conducts multilayer feature fusion to improve the robustness of the extracted features. Experiment results show that the proposed ASNL-LinkNet achieves the best performance when compared with other deep learning methods. The quantitative evaluation results in the three test areas show that the F1-score of erosion gully recognition varies from 0.62 to 0.72. This study provides theoretical reference and practical guidance for monitoring erosion gullies on slope farmland in the black soil region of Northeast China.

Sarcopenia, characterized by the loss of skeletal muscle mass and function, is a significant complication in patients with cirrhosis. This condition not only exacerbates the overall morbidity and mortality associated with liver disease but also complicates patient management, increasing the risk of hospitalization, infections, and hepatic encephalopathy. Despite its clinical significance, sarcopenia in cirrhotic patients remains underdiagnosed and undertreated. This review aims to summarize current knowledge on the pathophysiology of sarcopenia in cirrhosis, including mechanisms such as altered metabolism, hormonal imbalances, and inflammation. Additionally, we explore diagnostic challenges and discuss emerging therapeutic strategies, including nutritional support, exercise, and pharmacological interventions. By highlighting the gaps in existing research and proposing directions for future studies, this review seeks to improve the management and outcomes of cirrhotic patients affected by sarcopenia.

Abstract Background Sarcopenia, an age-related disorder characterized by loss of skeletal muscle mass and function, is recently recognized as a complication in elderly patients with type 2 diabetes mellitus (T2DM). Skeletal muscles play a crucial role in glycemic metabolism, utilizing around 80% of blood glucose. Accordingly, we aimed to explore the relationship between glucose metabolism and muscle mass in T2DM. Methods We employed the AWGS 2019 criteria for diagnosing low muscle mass and 1999 World Health Organization (WHO) diabetes diagnostic standards. This study included data of 191 individuals aged 60 and above with T2DM of Shanghai Pudong Hospital from November 2021 to November 2022. Fasting C-peptide (FPCP), fasting plasma glucose (FPG), 2-hour postprandial plasma glucose (PPG) and postprandial 2-hour C-peptide (PPCP), glycated hemoglobin A1c (HbA1c), glycated albumin (GA), serum lipids spectrum, renal and hepatic function, hemoglobin, and hormone were measured. Based on the findings of univariate analysis, logistic regression and receiver operating characteristic (ROC) curves were established. Measuring causal brain network is a significant topic for exploring complex brain functions. While various data-driven algorithms have been proposed, they still have some drawbacks such as ignoring time non-separability, cumbersome parameter settings, and poor robustness. To solve these deficiencies, we developed a novel framework: “time-shift permutation cross-mapping, TPCM,” integrating steps of: (1) delayed improved phase-space reconstruction (DIPSR), (2) rank transformation of embedding vectors’ distances, (3) cross-mapping with a fitting estimation, and (4) causality quantification using multi-delays. Based on the synthetic models and comparison with baseline methods, numerical validation results demonstrate that TPCM significantly improves the robustness for data length with or without noise interference, and achieves the best quantification accuracy in detecting time delay and coupling strength, with the highest determination coefficient ( ${R}^{{2}}={0}.{96}\text {)}$ of fitting verse coupling parameters. The developed TPCM was finally applied to ictal electrocorticogram (ECoG) analysis of patients with drug-resistant epilepsy (DRE). A total of 17 patients with DRE were included into the retrospective study. For 8 patients undergoing successful surgeries, the causal coupling strength ( $0.58~\pm ~0.20$ ) within epileptogenic zone network is significantly higher than those suffering failed surgeries ( $0.38~\pm ~0.16$ ) with ${P}\lt {0}. {001}$ through Mann-Whitney-U-test. Therefore, the epileptic brain network measured by TPCM is a credible biomarker for predicting surgical outcomes. These findings additionally confirm TPCM’s superior performance and promising potential to advance precision medicine for neurological disorders.

Fungi are rich sources of secondary metabolites of agrochemical, pharmaceutical, and food importance, such as mycotoxins, antibiotics, and antitumor agents. Secondary metabolites play vital roles in fungal pathogenesis, growth and development, oxidative status modulation, and adaptation/resistance to various environmental stresses. LaeA contains an S-adenosylmethionine binding site and displays methyltransferase activity. The members of velvet proteins include VeA, VelB, VelC, VelD and VosA for each member with a velvet domain. LaeA and velvet proteins can form multimeric complexes such as VosA-VelB and VelB-VeA-LaeA. They belong to global regulators and are mainly impacted by light. One of their most important functions is to regulate gene expressions that are responsible for secondary metabolite biosynthesis. The aim of this mini-review is to represent the newest cognition of the biosynthetic regulation of mycotoxins and other fungal secondary metabolites by LaeA and velvet proteins. In most cases, LaeA and velvet proteins positively regulate production of fungal secondary metabolites. The regulated fungal species mainly belong to the toxigenic fungi from the genera of Alternaria, Aspergillus, Botrytis, Fusarium, Magnaporthe, Monascus, and Penicillium for the production of mycotoxins. We can control secondary metabolite production to inhibit the production of harmful mycotoxins while promoting the production of useful metabolites by global regulation of LaeA and velvet proteins in fungi. Furthermore, the regulation by LaeA and velvet proteins should be a practical strategy in activating silent biosynthetic gene clusters (BGCs) in fungi to obtain previously undiscovered metabolites.

Chemical study of the nematicidal biocontrol fungus Pochonia chlamydosporia PC-170 led to discovery of six resorcylic acid lactones (RALs), including three nematicidal glycosylated RALs, monocillin VI glycoside (1), colletogloeolactone A (2) and monocillin II glycoside (3), and three antibacterial non-glycosylated RALs, monocillin VI (4), monocillin IV (5) and monocillin II (6). The planar structure of the new compound monocillin VI glycoside (1) was elucidated using HRESIMS and NMR data, and its monosaccharide configuration was further determined through sugar hydrolysis experiment and GC–MS analysis method. Furthermore, their two biosynthetic-related PKS genes, pchE and pchI, were identified through the gene knockout experiment. The glycosylated RALs 1–3 exhibited nematicidal activity against Meloidogyne incognita, with LC50 values of 94, 152 and 64 μg/mL, respectively, and thus had great potential in the development of new nematicidal natural products to control M. incognita in the future.

Ustilaginoidins are a class of bis-naphtho-γ-pyrone mycotoxins to threaten humans, animals and environment. Ustilaginoidins are produced by Villosiclava virens, the rice false smut pathogen. To prepare antibodies for quantitatively analyzing ustilaginoidins in rice samples, hemiustilaginoidins D and F from the laccase gene deficiency mutant of V. virens respectively reacted with diazonium 4-aminobenzoic acid to obtain haptens with a carboxyl group, which further reacted with bovine serum albumin or ovalbumin to get their complete antigens. Two monoclonal antibodies (mAbs) designated as 4A12C6 and 5F4F6 were developed by immunization. The relationships between mAb sensitivity and 20 ustilaginoidins were described. 4A12C6 was chosen for further analysis as it could recognize main ustilaginoidins and was more sensitive than 5F4F6. The achieved indirect competitive enzyme-linked immunosorbent assay (icELISA) based on 4A12C6 had a half maximal inhibitory concentration (IC50) of 0.76 ng/mL and working range of 0.2–2.8 ng/mL to ustilaginoidin A. The results of ustilaginoidins-contaminated rice samples by icELISA detection were consistent with those determined by HPLC‒DAD detection. Therefore, we developed a new strategy to get haptens from the biosynthetic precursors with half structures of ustilaginoidins. The achieved icELISA was demonstrated as a convenient method to monitor ustilaginoidin content in rice samples, and showed that the contents of total ustilaginoidins from the rice cultivars with low resistance to rice false smut were more than those of high resistance cultivars.

Obstructive Sleep Apnea (OSA), a sleep disorder with high prevalence, is normally accompanied by affective, autonomic, and cognitive abnormalities, and is deemed to be linked to functional brain alterations. To investigate alterations in brain functional connectivity properties in patients with OSA, a comparative analysis of global and local topological properties of brain networks was conducted between patients with OSA and healthy controls (HCs), utilizing functional near-infrared spectroscopy (fNIRS) imaging. A total of 148 patients with OSA and 150 healthy individuals were involved. Firstly, quantitative alterations in blood oxygen concentration, changes in functional connectivity, and variations in graph theory-based network topological characteristics were assessed. Then, with Mann–Whitney statistics, this study compared whether there are significant differences in the above characteristics between patients with OSA and HCs. Lastly, the study further examined the correlation between the altered characteristics and the apnea hypopnea index (AHI) using linear regression. Results revealed a higher mean and standard deviation of hemoglobin concentration in the superior temporal gyrus among patients with OSA compared to HCs. Resting-state functional connectivity (RSFC) exhibited a slight increase between the superior temporal gyrus and other specific areas in patients with OSA. Notably, neither patients with OSA nor HCs demonstrated significant small-world network properties. Patients with OSA displayed an elevated clustering coefficient (p < 0.05) and local efficiency (p < 0.05). Additionally, patients with OSA exhibited a tendency towards increased nodal betweenness centrality (p < 0.05) and degree centrality (p < 0.05) in the right supramarginal gyrus, as well as a trend towards higher betweenness centrality (p < 0.05) in the right precentral gyrus. The results of multiple linear regressions indicate that the influence of the AHI on RSFC between the right precentral gyrus and right superior temporal gyrus (p < 0.05), as well as between the right precentral gyrus and right supramarginal gyrus (p < 0.05), are statistically significant. These findings suggest that OSA may compromise functional brain connectivity and network topological properties in affected individuals, serving as a potential neurological mechanism underlying the observed abnormalities in brain function associated with OSA.

KCNQ2 epileptic encephalopathy is relatively common in early-onset neonatal epileptic encephalopathy and seizure severity varied widely, categorized as drug-sensitive epilepsy and drug-resistant epilepsy. However, in clinical practice, anti-seizure medicines need to be gradually adjusted based on seizure control which undoubtedly increases the economic burden of patients, so further positive anti-seizure regimens depend on whether seizure severity can be predicted in advance. In this paper, we proposed a reliable assessment to differentiate between drug-sensitive epilepsy and drug-resistant epilepsy caused by KCNQ2 pathogenic variants. Based on the electroencephalogram (EEG) and electrooculogram (EOG) signals, twenty-four classical temporal and spectral domain features were extracted and Gradient Boosting Decision Tree (GBDT) was employed to distinguish between patients with drug-sensitive epilepsy and drug-resistant epilepsy. In addition, we also systematically investigated the impact of channel combination and feature combination based on the forward stepwise selection strategy. By employing selected channels and features, the classification accuracy can reach 81.25% with a sensitivity of 57.14% and specificity of 100%. Compared with the state-of-the-art techniques, including the functional network, effective network, and common spatial patterns, the improvement of accuracy ranges from 37.5% to 56.25%, indicating the superiority of our proposed method. Overall, the proposed method may provide a promising tool to distinguish different seizure outcomes of KCNQ2 epileptic encephalopathy.

Abstract Background Non-alcoholic fatty liver (NAFLD) is a complex metabolic disease characterized by fatty degeneration of hepatocytes. Circular RNAs (circRNAs) have been reported to be essential for (NAFLD progression. The potential mechanism of circRNA low-density lipoprotein receptor (circLDLR) in the NAFLD was investigated in this study. Methods Hepatocyte (Hepa1-6) cells treated with oleic acid/palmitic acid (OA/PA) were used as the in vitro NAFLD model, and C57BL/6 mice fed with high-fat diet (HFD) were used as the in vivo NAFLD model. The circLDLR, LDLR, and miR-667-5p expression were measured by quantitative real-time polymerase chain reaction (qRT-PCR), while the protein levels of Light Chain Microtubule-Associated Protein 3 (LC3) and Sequestosome-1(p62) was examined by western blot. The circLDLR location was confirmed using RNA fluorescence in situ hybridization. Oil red O staining was carried out to measure lipid deposition in cells. The secreted levels of triglyceride (TG) and total cholesterol (TC) were detected through Enzymatic. The existence of the circLDLR/miR-667-5p/sirtuin 1 (SIRT1) regulatory axis was validated by applying the dual-luciferase reporter assay. Results The circLDLR expression showed a prominent down-regulation in OA/PA-treated Hepa1-6 cells, whereas the LDLR expression was up-regulated. Overexpression of circLDLR significantly attenuated lipid droplet accumulation in NAFLD models in vitro/vivo, reduced TG, TC, and p62 levels, and increased LC3-II levels and the amount of the green fluorescent protein (GFP)-LC3 puncta in cells. CircLDLR and SIRT1 are common targets of miR-667-5p to inhibit the TG and TC and promote the autophagy pathway. SIRT1 knockdown reversed the effects of circLDLR overexpression. Conclusions CircLDLR alleviated the development of NAFLD by inducing autophagic flux while modulating the miR-667-5p/SIRT1 axis reversed its effects, suggesting that targeting circLDLR/miR-667-5p/SIRT1 axis may be a promising therapeutic strategy for NAFLD.

Abstract Purpose The aim of this study is to explore the factors associated with the fall risk in type 2 diabetes (T2D) patients with a lacunar stroke. Materials and methods We compiled data of 146 T2D patients (mean age 68 years), including the Morse fall scale data (MFS), nutrition score, self-care scale, laboratory data, and data from continuous glucose monitoring system (CGMS) from 2019 to 2021 in Shanghai Pudong Hospital. Thereby, we evaluated the associations between MFS and other clinical parameters. Results The analyses showed that there were significantly increased size and numbers of lacunar infarction (p 0.05). The correlational study revealed the higher score of MFS was associated with the age (r = 0.41), number of lacunar infarction (r = 0.18), nutrition score (r = 0.20), self-care score (r = − 0.43), serum creatine level (r = 0.19), eGFR (r = − 0.26) (p

Villosiclava virens (anamorph: Ustilaginoidea virens) is the pathogen of rice false smut (RFS), which is a destructive rice fungal disease. The albino strain LN02 is a natural white-phenotype mutant of V. virens due to its incapability to produce toxic ustilaginoidins. In this study, three strains including the normal strain P1, albino strain LN02, and complemented strain uvpks1C-1 of the LN02 strain were employed to investigate the activation of the ustilaginoidin biosynthesis gene uvpks1 in the albino strain LN02 to influence sporulation, conidia germination, pigment production, stress responses, and the inhibition of rice seed germination. The activation of the ustilaginoidin biosynthesis gene uvpks1 increased fungal tolerances to NaCl-induced osmotic stress, Congo-red-induced cell wall stress, SDS-induced cell membrane stress, and H2O2-induced oxidative stress. The activation of uvpks1 also increased sporulation, conidia germination, pigment production, and the inhibition of rice seed germination. In addition, the activation of uvpks1 was able to increase the mycelial growth of the V. virens albino strain LN02 at 23 °C and a pH from 5.5 to 7.5. The findings help in understanding the effects of the activation of uvpks1 in albino strain LN02 on development, pigment production, stress responses, and the inhibition of rice seed germination by controlling ustilaginoidin biosynthesis.

Rice false smut (RFS) caused by Villosiclava virens (anamorph: Ustilaginoidea virens) has become one of the most destructive fungal diseases to decrease the yield and quality of rice grains. An albino strain LN02 was isolated from the white RFS balls collected in the Liaoning Province of China in 2019. The strain LN02 was considered as a natural albino mutant of V. virens by analyzing its phenotypes, internal transcribed spacer (ITS) conserved sequence, and biosynthesis gene clusters (BGCs) for secondary metabolites. The total assembled genome of strain LN02 was 38.81 Mb, which was comprised of seven nuclear chromosomes and one mitochondrial genome with an N50 value of 6,326,845 bp and 9339 protein-encoding genes. In addition, the genome of strain LN02 encoded 19 gene clusters for biosynthesis of secondary metabolites mainly including polyketides, terpenoids and non-ribosomal peptides (NRPs). Four sorbicillinoid metabolites were isolated from the cultures of strain LN02. It was found that the polyketide synthase (PKS)-encoding gene uspks1 for ustilaginoidin biosynthesis in strain LN02 was inactivated due to the deletion of four bases in the promoter sequence of uvpks1. The normal uvpks1 complementary mutant of strain LN02 could restore the ability to synthesize ustilaginoidins. It demonstrated that deficiency of ustilaginoidin biosynthesis is the cause of albinism for RFS albino strain LN02, and V. virens should be a non-melanin-producing fungus. This study further confirmed strain LN02 as a white phenotype mutant of V. virens. The albino strain LN02 will have a great potential in the development and application of secondary metabolites. The physiological and ecological functions of ustilaginoidins in RFS fungus are needed for further investigation.

A total of 99 endophytic fungal isolates were obtained from the roots of Glycyrrhiza inflata, which was a traditional medicinal plant mainly distributed in Xinjiang, China. Twenty-two distinct isolates were selected for further taxonomical identification by morphological traits and internal transcribed spacer (ITS) rRNA gene sequence analysis. Eleven genera were identified, among which Aspergillus, Alternaria and Fusarium were dominant. The crude extracts of 22 distinct identified fungi were successively evaluated for their antifungal activities on three rice fungal pathogens using the method of hyphal radial growth rate. Among them, the crude extract of Alternaria an-gustiovoidea Glinf007 showed the significantly mycelial growth inhibitory activity. The results demonstrated that G. inflata contained a diversity of culturable endophytic fungi, which could produce natural antimicrobial compounds that might be of great value to the agriculture and pharmaceutical industries.

Abstract Background Mirror syndrome (MS) is defined as maternal edema with fetal hydrops and placental edema with different etiologies, such as rhesus isoimmunization and twin-twin transfusion syndrome. Herein, we showcased a unique MS case secondary to fetomaternal hemorrhage (FMH). Case presentation A 32-year-old gravida 2 para 0 woman diagnosed with fetal hydrops was admitted to our hospital. Maternal laboratory tests revealed anemia, slightly increased creatinine and uric acid levels, hypoproteinemia, and significantly increased alpha-fetoprotein and hemoglobin-F levels. Therefore, FMH was diagnosed initially. Two days after admission, the woman had unexpectedly progressive anasarca and started to feel chest distress, palpitations, lethargy, and oliguria, and MS was suspected. An emergency cesarean section was performed to terminate the pregnancy. The maternal clinical symptoms and laboratory tests rapidly improved after delivery. A very preterm infant with a 2080-g birthweight at 31 weeks gestation survived after emergency cesarean section, active resuscitation, emergency blood transfusion, abdominocentesis, and advanced life support. Conclusions FMH could develop into MS, providing new insights into the etiology of MS. Once MS is diagnosed, emergency cesarean section might be an alternative treatment. The very preterm infant survived with a favorable long-term outcome, and a well-trained perinatal work team is needed for such cases.

The scientific location of earthquake emergency supply warehouses is conducive to the effective distribution of emergency relief resources and improved rescue efficiency in earthquake hazard. Comprehensively considering the regional population as well as coverage quality at the demand points, this paper aims to divide the coverage thresholds of earthquake emergency rescue and logistic supplies according to their time-series features, and to build a location model for supply warehouses according to the variety and amount of stored supplies considering their time-series features, in hope of optimizing the set covering issue of earthquake relief supply warehouses. The solution is approached with two methods: the target deviation rate minimization model and NSGA-II algorithm. The results obtained by solving the target deviation rate minimization model can balance every target. The branch and bound algorithm can find the global optimal solution at a certain calculation scale with high calculation efficiency, but its efficiency decreases significantly when the operation scale increases. The NSGA-II algorithm is more suitable for large-scale solution calculations with high calculation efficiency, and it can output a set of non-inferior solutions for decision makers to select from according to different preference. Taking Aba Prefecture in Sichuan Province as illustration, the feasibility of the model is validated; meanwhile, the effectiveness and benefits of the two approaches in solving the problem of multi-objective set covering of the warehouses are compared and analyzed.

Sorbicillinoids are a class of hexaketide metabolites produced by Ustilaginoidea virens (teleomorph: Villosiclava virens), an important fungal pathogen that causes a devastating rice disease. In this study, we investigated the effects of environmental factors, including carbon and nitrogen sources, ambient pH and light exposure, on mycelial growth, sporulation, as well as the accumulation of sorbicillinoids, and the expression of related genes involved in sorbicillinoid biosynthesis. It was found that the environmental factors had great influences on mycelial growth and sporulation of U. virens. Fructose and glucose, complex nitrogen sources, acidic conditions and light exposure were favorable for sorbicillinoid production. The relative transcript levels of sorbicillinoid biosynthesis genes were up-regulated when U. virens was separately treated with those environmental factors that favored sorbicillinoid production, indicating that sorbicillinoid biosynthesis was mainly regulated at the transcriptional level by different environmental factors. Two pathway-specific transcription factor genes, UvSorR1 and UvSorR2, were found to participate in the regulation of sorbicillinoid biosynthesis. These results will provide useful information to better understand the regulation mechanisms of sorbicillinoid biosynthesis, and be conducive to develop effective means for controlling sorbicillinoid production in U. virens.

Abstract Background Nonalcoholic fatty liver disease (NAFLD) is primarily characterized by the hepatic cholesterol accumulation. Circular RNA (circRNA), one of noncoding RNA, involves in many liver diseases progression. However, no recent studies on circRNA expression profiles in NAFLD have been reported previously. Methods A NAFLD mouse model was constructed by providing high-fat diet (HFD) for 32 weeks. The circRNAs expression profile in normal mice and NAFLD mice were determined using high-output RNA sequencing method and bioinformatics methods, while the differentially expressed circRNAs were confirmed using Sanger sequencing and qRT-PCR. The circRNA-miRNA network was also predicted. The biological functions of circRNAs were annotated by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). Results The results demonstrated the successful construction of NAFLD mice model by immunohistology and serology assay. In total, 93 dysregulated circRNAs were observed, including 57 upregulated circRNAs and 36 downregulated circRNAs, in the NAFLD group. The circRNA-miRNA network revealed the complex interaction between circRNAs and its potential miRNA targets in NAFLD. The characteristic of tissue-specific expression in circRNA was demonstrated. The differentially expressed circRNAs with important biological function were also annotated using GO and KEGG. Both DDAH1 and VAV3 genes were found to be associated with the NAFLD development. Conclusions Taken together, this study demonstrated the circRNAs expression profile and features in NAFLD, which may provide potential biological markers for the pathogenesis of NAFLD.

ObjectiveTo assess whether providing nasal continuous positive airway pressure (nCPAP) during delayed cord clamping is beneficial for preterm infants

Soil loss, both from surface soil loss and subsurface soil leakage, in the karst regions of southwestern China is a serious environmental problem that threatens sustainability in that region. The surface soil loss has been extensively studied, and many studies have been conducted to investigate the causes, impacts and mechanisms involved, but the study of subsurface soil leakage has received little attention due to the difficulties in studying the natural conditions. There is no consensus on the overall proportions between surface soil loss and subsurface soil leakage. To control soil loss, improve ecological restoration, and help locals out of poverty, the Chinese government carried out a series of ecological restoration projects in the karst regions of southwestern China starting in the 1980s. As a result, the intensity and areal extent of soil loss continues to decrease and the ecological situation is steadily improving. However, because of the fragile ecosystem in the karst regions, the soil loss control is a long-term task, and the soil loss in some karst regions continues to be a problem. Subsequently, we put forward some suggestions for the policy makers relative to conservation of soil loss and vegetation restoration. These suggestions include: (1) government, private organizations and individuals are encouraged to raise funds for soil loss control and vegetation restoration; (2) nature reserves should be established to increase biodiversity; (3) engineering projects such as small reservoirs, ponds, and flow diversion channels should be constructed in marginal karst regions. An interval-valued Pythagorean hesitant fuzzy set (IVPHFS) not only can be regarded as the union of some interval-valued Pythagorean fuzzy sets but also represent the Pythagorean hesitant fuzzy elements in the form of interval values. So IVPHFSs are extensions of Pythagorean hesitant fuzzy sets (PHFSs) and interval-valued Pythagorean fuzzy sets (IVPFSs), which are powerful tools to represent more complicated, uncertain and vague information. This paper focuses on the four kinds of correlation coefficients for PHFSs, and extends them to the correlation coefficients and the weighted correlation coefficients for IVPHFSs. In the processing, we develop the least common multiple expansion (LCME) method to solve the problem that the cardinalities of Pythagorean hesitant fuzzy elements (PHFEs) (or interval-valued Pythagorean hesitant fuzzy elements (IVPHFEs)) are different. In addition, we propose score functions and accuracy functions of Pythagorean fuzzy elements (PFEs) (or interval-valued Pythagorean fuzzy elements (IVPFEs)) to rank all the PFEs (or IVPFEs) in a PHFE (or an IVPHFE). Especially, score functions and accuracy functions of IVPFEs are both presented as interval numbers. Then use the comparison method of interval numbers to compare two revised IVPHFEs in order to keep the original fuzzy information as far as possible. What's more, we define the local correlations and local informational energies which can depict the similarity between two IVPHFEs more meticulously and completely. At last the numerical examples to show the feasibility and applicability of the proposed methods in multiple criteria decision making (MCDM) and clustering analysis.

The Pythagorean fuzzy set (PFS), introduced by Yager, is a generalization of the intuitionistic fuzzy set. The PFS is of great significance for the decision-making problem because it promotes the domain of intuitionistic fuzzy set. This paper aims to conduct the consistency analysis and priority weights for Pythagorean fuzzy preference relations (PFPRs). First, a consistency index to measure the multiplicative consistency of Pythagorean fuzzy preference relation (PFPR) is presented. Then, for the non-consistent PFPR, an iteration algorithm is put forward to adjust it to achieve acceptable consistency. Furthermore, an absolute deviation model is built to derive the priority weighting vector of PFPR. Finally, an illustrative example is provided to test and verify the effectiveness and applicability of the proposed method.

Pythagorean fuzzy set, characterized by membership function and non-membership function, has received increasing attention in recent years. In this paper, a new approach to decision making is proposed based on Pythagorean fuzzy preference relation and its additive consistency. Firstly, the concepts of Pythagorean fuzzy preference relations and its additive consistency are introduced, and followed by a discussion of their desirable properties. Then, a linear goal programming model is proposed to determine the consistency of PFPRs. For the PFPRs that does not satisfy the consistency, the consistency index is defined to measure the degree of consistency, and a consistency adjustment algorithm is proposed. Finally, based on the additive consistency, a new algorithm for decision making is presented. An example of CIM(Computing In Memory) is provided, and in comparison with other methods, the validity and rationality of the proposed method are verified.

Genomic analysis has demonstrated that many fungi possess essential gene clusters for the production of previously unobserved secondary metabolites; however, these genes are normally reduced or silenced under most conditions. These cryptic biosynthetic gene clusters have become treasures of new bioactive secondary metabolites. The induction of these biosynthetic gene clusters under stress or special conditions can improve the titers of known compounds or the production of novel compounds. Among the inducing strategies, chemical-epigenetic regulation is considered a powerful approach, and it uses small-molecule epigenetic modifiers, which mainly act as the inhibitors of DNA methyltransferase, histone deacetylase, and histone acetyltransferase, to promote changes in the structure of DNA, histones, and proteasomes and to further activate cryptic biosynthetic gene clusters for the production of a wide variety of bioactive secondary metabolites. These epigenetic modifiers mainly include 5-azacytidine, suberoylanilide hydroxamic acid, suberoyl bishydroxamic acid, sodium butyrate, and nicotinamide. This review gives an overview on the method of chemical epigenetic modifiers to trigger silent or low-expressed biosynthetic pathways to yield bioactive natural products through external cues of fungi, mainly based on the research progress in the period from 2007 to 2022. The production of about 540 fungal secondary metabolites was found to be induced or enhanced by chemical epigenetic modifiers. Some of them exhibited significant biological activities such as cytotoxic, antimicrobial, anti-inflammatory, and antioxidant activity.

Ustilaginoidins are a class of bis-naphtho-γ-pyrone mycotoxins produced by the pathogen Villosiclava virens of rice false smut, which has recently become one of the most devastating diseases in rice-growing regions worldwide. In this research, the nanobody phage display library was established after an alpaca was immunized with the hemiustilaginoidin F-hapten coupled with bovine serum albumin (BSA). Heterologous antigen selection and combing trypsin with competition alternant elution methods were performed for nanobody screening. Two nanobodies, namely, Nb-B15 and Nb–C21, were selected for the establishment of indirect competitive enzyme-linked immunosorbent assays (ic-ELISAs). For Nb–B15 and Nb-C21, their IC50 values were 11.86 μg/mL and 11.22 μg/mL, and the detection ranges were at 3.41–19.98 μg/mL and 1.17–32.13 μg/mL, respectively. Two nanobodies had a broad spectrum to quantify the contents of total ustilaginoidins in rice samples according to cross-reactivity. The recognition mechanisms of Nb-B15 and Nb-C21 against ustilaginoidin A were elucidated by molecular modeling and docking. The key amino acid sites for the binding of Nb–B15 or Nb–C21 to ustilaginoidin A were mainly located in the FR1 and CDR1 regions. As Nb-B15 was superior to Nb–C21 in the aspects of protein expression, ELISA titer, and tolerance to organic solvents, it was selected for application in the detection of actual contaminated rice samples. The total ustilaginoidin contents of rice samples were analyzed by Nb–B15-based ic–ELISA and HPLC-DAD, between which the results were found to be consistent. The developed immunoassay based on the nanobody from the alpaca can be employed as a rapid and effective method for detection of total utilaginoidins in contaminated rice samples.

Ustilaginoidea virens (teleomorph: Villosiclava virens) is an important fungal pathogen that causes a devastating rice disease. It can produce mycotoxins including sorbicillinoids. The biosynthesis and biological functions of sorbicillinoids have not been reported in U. virens. In this study, we identified a sorbicillinoid biosynthetic gene cluster in which two polyketide synthase genes UvSorA and UvSorB were responsible for sorbicillinoid biosynthesis in U. virens. In ∆UvSorA and ∆UvSorB mutants, the mycelial growth, sporulation and hyphal hydrophobicity were increased dramatically, while the resistances to osmotic pressure, metal cations, and fungicides were reduced. Both phytotoxic activity of rice germinated seeds and cell wall integrity were also reduced. Furthermore, mycelia and cell walls of ∆UvSorA and ∆UvSorB mutants showed alterations of microscopic and submicroscopic structures. In addition, feeding experiment showed that sorbicillinoids could restore mycelial growth, sporulation, and cell wall integrity in ∆UvSorA and ∆UvSorB mutants. The results demonstrated that both UvSorA and UvSorB were responsible for sorbicillinoid biosynthesis in U. virens, and contributed to development (mycelial growth, sporulation, and cell wall integrity), stress responses, and phytotoxicity through sorbicillinoid mediation. It provides an insight into further investigation of biological functions and biosynthesis of sorbicillinoids.

Multidrug-resistant (MDR) pathogens are responsible for a substantial burden of morbidity and mortality from neonatal sepsis; however, data on these sepsis-related pathogens among hospitalized neonates in China are not well characterized. In this study, a total of 240 strains were isolated from four Women and Children’s hospitals in Southwest China between 2014 and 2019. Of these included pathogens, 104 (43.33%) were gram-positive bacteria, 129 (53.75%) were gram-negative bacteria, and 7 (2.92%) were fungi. Escherichia coli (E. coli, 34.01%) and Klebsiella pneumoniae (K. pneumoniae, 15.35%) were the main pathogen of neonate bacteremia. ST167 were the most prevalent STs in E. coli and ST11 in K. pneumoniae. Our study found that E. coli (62.71%) was the predominate pathogen of early-onset sepsis, among which 64.86% were MDR. Late-onset sepsis was mainly caused by K. pneumoniae (28.31%) and E. coli (24.78%), with showing that 78.33% of these pathogens were MDR. Notably, the prevalence of EO/LO pathogens were quite different from Indian and south of China. Moreover, we found that blaCTX-M (42.06%) was most dominant resistant genes with about a third isolates (31.09%) were positive for blaCTX-M-15. All the carbapenem-resistant K. pneumoniae were positive for NDM-1. Moreover, late-onset sepsis and antibiotic exposure were significantly associated with MDR infection. Emerging multi-resistant pathogens of sepsis posts a serious threat to neonatal outcomes and emphasizes an urgent need to control their further spread.

Since some existing uncertainty measurement of covering-based rough intuitionistic fuzzy sets (CRIFSs) are unreasonable in some cases, an extended uncertainty measure criterion of CRIFSs is proposed. The discussion of the monotonicity of the criterion is further refined and more in line with the actual problem description. Taken into account this a modified roughness method is introduced, which is an uncertainty measure of CRIFSs satisfying the extended criterion. Furthermore, the criterion is extended to covering-based rough interval-valued intuitionistic fuzzy sets (CRIVIFSs), and the modified roughness method is proposed to measure the uncertainty of CRIVIFSs. Finally, the example is presented to illustrate the application of the modified roughness to attribute reductions. These conclusions provide a theoretical basis for the rationality (or irrationality) of existing uncertainty measures and also promote the application of CRIVIFSs.

The aim of this paper is to develop some novel operational laws for a hesitant fuzzy linguistic term set (HFLTS)-based on the improved supplementary regulation and Archimedean t-norms and s-norms. The improved supplementary regulation for HFLTSs can reserve the fidelity of original information commendably, and it brings a new conception to research on information measures of HFLTS. As a hot and key research topic for information fusion, some Archimedean t-norms and s-norms based hesitant fuzzy linguistic aggregation operators are proposed to aggregate HFLTSs. Some essential properties together with their special cases of such aggregation operators are discussed in detail. The entropy and cross-entropy of HFLTSs are proposed and applied to derive the attribute weights. An approach to multiple attributes group decision making with hesitant fuzzy linguistic information is developed. Finally, a numerical example related to the assessment of health-care waste disposal methods is provided to show the utility and effectiveness of our methods, which are then compared to the existing methods.

Complementing missing information and priority vector are of significance important aspects in group decision making (GDM) with incomplete hesitant fuzzy linguistic preference relations (HFLPRs). In this paper, an integer programming model is developed based on additive consistency to estimate missing values of incomplete HFLPRs by using additive consistency. Once the missing values are complemented, a mixed 0-1 programming model is established to derive the priority vectors from complete HFLPRs, in which the underlying idea of the mixed 0-1 programming model is the probability sampling in statistics and minimum deviation between the priority vector and HFLPR. In addition, we also propose a new GDM approach for incomplete HFLPRs by integrating the integer programming model and the mixed 0-1 programming model. Finally, two case studies and comparative analysis detail the application of the proposed models.

This paper provides a new multi-attributes decision making approach on the basis of the Gini OWA ( $\text{G}^{\mathrm {p,q}}$ -OWA) operator, in which the Gini operator is the combination of the Gini mean and the OWA operator. Desired properties and several generalized forms of the $\text{G}^{\mathrm {p,q}}$ -OWA operator are investigated. In order to determine the $\text{G}^{\mathrm {p,q}}$ -OWA operator weights, an orness measure is proposed, and a generalized least squares deviation model (GLSD) is put forward. Finally, a case study of low carbon supplier selection is shown to illustrate the effectiveness and practicality of the proposed method.

Fourteen new eremophilane-type sesquiterpenoids, named rhizoperemophilanes A~N (1~14), together with eight known congeners, were isolated from the culture of the endophytic fungus Rhizopycnis vagum. The structures of the new compounds were elucidated by extensive spectroscopic analyses, as well as ECD calculations and the modified Mosher's method for the assignment of the absolute configurations. Rhizoperemophilane J (10) contains an uncommon C-4/C-11 epoxy ring, while rhizoperemophilane N (14) features an unprecedented 3-nor-eremophilane lactone-lactam skeleton. These metabolites were evaluated for their antibacterial, cytotoxic, and phytotoxic activities. Among them, compounds 11, 16, and 20 displayed antibacterial activities, while 14 showed selective cytotoxicity against NCI-H1650 and BGC823 tumor cells. Moreover, compounds 5, 6, 12, 13, 16, and 19 exhibited strong phytotoxic activities against the radicle elongation of rice seedlings.

Pythagorean hesitant fuzzy sets are widely watched because of their excellent ability to deal with uncertainty, imprecise and vague information. This paper extends Pythagorean hesitant fuzzy environments to interval-valued Pythagorean hesitant fuzzy environments and proposes the concept of interval-valued Pythagorean hesitant fuzzy set (IVPHFS), which allows the membership of each object to be a set of several pairs of possible interval-valued Pythagorean fuzzy elements. Furthermore, we develop a series of aggregation operators for interval-valued Pythagorean hesitant fuzzy information and apply them to multiattribute group decision-making (MAGDM) problems. Then, some desired operational laws and properties of IVPHFSs are studied. Especially, considering an interval-valued Pythagorean fuzzy element (IVPHFE) is formed by several pairs of interval values, this paper proposes the concepts of score function and accuracy function in the form of two interval numbers which can retain interval-valued Pythagorean fuzzy information as much as possible. Then, the relationship among these operators is discussed by comparing the interval numbers. Eventually, an illustrative example fully shows the feasibility, practicality, and effectiveness of the proposed approach.

Incretin-based therapy is now a prevalent treatment option for patients with type 2 diabetes mellitus (T2DM). It has been associated with considerably good results in the management of hyperglycemia with cardiac or nephron-benefits. For this reason, it is recommended for individuals with cardiovascular diseases in many clinical guidelines. As an incretin hormone, glucagon-like peptide-1 (GLP-1) possesses multiple metabolic benefits such as optimizing energy usage, maintaining body weight, β cell preservation, and suppressing neurodegeneration. However, recent studies indicate that oral antidiabetic medications interact with endogenous or exogenous GLP-1. Since these drugs are transported to distal intestine portions, there are concerns whether these oral drugs directly stimulate intestinal L cells which release GLP-1, or whether they do so via indirect inhibition of the activity of dipeptidyl peptidase-IV (DPP-IV). In this review, we discuss the metabolic relationships between oral antihyperglycemic drugs from the aspect of gut, microbiota, hormones, β cell function, central nervous system, and other cellular mechanisms.

Diabetic retinopathy (DR) is one of the most serious complications of diabetic microangiopathy. DR has an early onset and is not easy to detect. When visual impairment occurs, the optimal period for therapy is often missed. Therefore, the prevention and treatment of DR should start from the early stage of diabetes. Sodium-dependent glucose transporter 2 inhibitor (SGLT2i) is a new antidiabetic drug which is mainly used in clinical practice to control blood glucose of patients with type 2 diabetes prone to develop chronic heart failure. Recent studies have found that SGLT2 is also expressed in the human retina. Now, the prevention and treatment of diabetic retinopathy with SGLT2i while reducing blood sugar has become a new research field. Hence, this article reviewed the recent therapeutic and research progress of SGLT2 in the treatment of diabetic retinopathy.

Sorbicillinoids are a family of hexaketide metabolites with a characteristic sorbyl side chain residue. Sixty-nine sorbicillinoids from fungi, newly identified from 2016 to 2021, are summarized in this review, including their structures and bioactivities. They are classified into monomeric, dimeric, trimeric, and hybrid sorbicillinoids according to their basic structural features, with the main groups comprising both monomeric and dimeric sorbicillinoids. Some of the identified sorbicillinoids have special structures such as ustilobisorbicillinol A, and sorbicillasins A and B. The majority of sorbicillinoids have been reported from fungi genera such as Acremonium, Penicillium, Trichoderma, and Ustilaginoidea, with some sorbicillinoids exhibiting cytotoxic, antimicrobial, anti-inflammatory, phytotoxic, and α-glucosidase inhibitory activities. In recent years, marine-derived, extremophilic, plant endophytic, and phytopathogenic fungi have emerged as important resources for diverse sorbicillinoids with unique skeletons. The recently revealed biological activities of sorbicillinoids discovered before 2016 are also described in this review.

Consistency and priority weights of preference relations are two important phases of decision-making process since the decision-making solutions are determined by them. Therefore, it is meaningful to investigate consistency and priority weights for preference relations. In this paper, consistency and uncertain 2-tuple linguistic priority weights of uncertain 2-tuple linguistic preference relations (U2TLPRs) are investigated. First, based on the additive consistency, an additive consistency index is developed to measure the additive consistency level of U2TLPRs. Second, a goal programming model is proposed to adjust the unacceptable additive consistent U2TLPR until it satisfies acceptable additive consistency. Furthermore, an optimization model is developed to derive the uncertain 2-tuple linguistic priority weights from an U2TLPR. Meanwhile, in group decision-making (GDM) problems, similarities and confidence degrees of decision makers (DMs) are defined to determine DMs' weights. Subsequently the properties of collective U2TLPR are discussed. Finally, the proposed methods are implemented in two examples including a GDM problem to verify the validity of the proposed methods.

The development of distribution linguistic provides a new research idea for linguistic information group decision-making (GDM) problems, which is more flexible and convenient for experts to express their opinions. However, in the process of using distribution linguistic fuzzy preference relations (DLFPRs) to solve linguistic information GDM problems, there are few studies that pay attention to both internal consistency adjustment and external consensus of experts. Therefore, this study proposes a fresh decision support model based on consistency adjustment algorithm and consensus adjustment algorithm to solve GDM problems with distribution linguistic data. Firstly, we review the concept of DLFPRs to describe the fuzzy linguistic evaluation information, and then we present the multiplicative consistency of DLFPRs and a new consistency measurement method based on the distance, and investigate the consistency adjustment algorithm to ameliorate the consistency level of DLFPRs. Subsequently, the consensus degree measurement is carried out, and a new consensus degree calculation method is put forward. At the same time, the consensus degree adjustment is taken the expert cost into account to make it reach the predetermined level. Finally, a distribution linguistic fuzzy group decision making (DLFGDM) method is designed to integrate the evaluation linguistic elements and obtain the final evaluation information. A case of the evaluation of China’s state-owned enterprise equity incentive model is provided, and the validity and superiority of the proposed method are performed by comparative analysis.

Background: Hyalodendriella sp. Ponipodef12, an endophytic fungus from a poplar hybrid, was a high producer of botrallin and TMC-264 with various bioactivities. In this study, the influences of eight metal ions (i.e., Mn2+, Na+, Mg2+, Zn2+, Cu2+, Fe2+, Fe3+ and Al3+) on botrallin and TMC-264 production in liquid culture of the endophytic fungus Hyalodendriella sp. Ponipodef12 were investigated. Results: Three most effective metal ions (Zn2+, Cu2+ and Mg2+) along with their optimum concentrations were screened. The optimum addition time and concentrations of Zn2+, Cu2+ and Mg2+ were further obtained respectively for improving botrallin and TMC-264 production. The combination effects of Zn2+, Cu2+ and Mg2+ on the production of botrallin and TMC-264 by employing statistical method based on the central composite design (CCD) and response surface methodology (RSM) were evaluated, and two quadratic predictive models were developed for botrallin and TMC-264 production. The yields of botrallin and TMC-264, which were predicted as 144.12 mg/L and 36.04 mg/L respectively, were validated to be 146.51 mg/L and 36.63 mg/L accordingly with the optimum concentrations of Zn2+ at 0.81 mmol/L, Cu2+ at 0.20 mmol/L, and Mg2+ at 0.13 mmol/L in medium. Conclusion: The results indicated that the enhancement of botrallin and TMC-264 accumulation in liquid culture of the endophytic fungus Hyalodendriella sp. Ponipodef12 by the metal ions and their combination should be an effective strategy.

Ustilaginoidea virens, the causal fungus of rice false smut, was found in previous studies to produce two types of metabolites, ustiloxins and ustilaginoidins. However, genome sequencing of U. virens revealed a plethora of secondary-metabolites-biosynthetic core genes that were capable to biosynthesize unreported metabolites. A large-scale fermentation of U. virens was thus performed, and the fungal extract was chemically re-investigated. After removing the known metabolites, we found a fraction containing unknown phytotoxic substances. Fractionation of this part has led to the isolation of six new sorbicillinoids, namely ustisorbicillinols A~F (1~6), and two new sorbicillinoid-related pyrones, named ustilopyrones A (7) and B (8), together with nine known cogeners (9~17). The structures of the new compounds were elucidated by analysis of their NMR, HRMS, and CD spectra, while ECD, 13C NMR and optical rotation calculations were additionally used for configurational assignments. Plausible biosynthetic pathways for the new compounds were proposed. Phytotoxicity assays revealed that the major sorbicillinoids (12~14, and 16) showed strong inhibition against the radicle and germ elongation of rice and lettuce seeds, with compound 12 displaying the strongest inhibition. The isolated compounds were also evaluated for their cytotoxic, antibacterial, and antifungal activities. Compounds 10, and 12~14 showed moderate cytotoxicities against the tested cell lines with IC50s of 8.83~74.7 μM. Compounds 2, and 10~13 were active against the tested bacteria (MICs 4~128 μg/mL), while compounds 11~13 displayed moderate antifungal activities.

Fungal phytotoxic secondary metabolites are poisonous substances to plants produced by fungi through naturally occurring biochemical reactions. These metabolites exhibit a high level of diversity in their properties, such as structures, phytotoxic activities, and modes of toxicity. They are mainly isolated from phytopathogenic fungal species in the genera of Alternaria, Botrytis, Colletotrichum, Fusarium, Helminthosporium, and Phoma. Phytotoxins are either host specific or non-host specific phytotoxins. Up to now, at least 545 fungal phytotoxic secondary metabolites, including 207 polyketides, 46 phenols and phenolic acids, 135 terpenoids, 146 nitrogen-containing metabolites, and 11 others, have been reported. Among them, aromatic polyketides and sesquiterpenoids are the main phytotoxic compounds. This review summarizes their chemical structures, sources, and phytotoxic activities. We also discuss their phytotoxic mechanisms and structure–activity relationships to lay the foundation for the future development and application of these promising metabolites as herbicides.

The endophytic fungus Berkleasmium sp. Dzf12, isolated from Dioscorea zingiberensis, was found to produce palmarumycins C12 and C13 which possess a great variety of biological activities. Seven biocompatible water-immiscible organic solvents including n-dodecane, n-hexadecane, 1-hexadecene, liquid paraffin, dibutyl phthalate, butyl oleate and oleic acid were evaluated to improve palmarumycins C12 and C13 production in suspension culture of Berkleasmium sp. Dzf12. Among the chosen solvents both butyl oleate and liquid paraffin were the most effective to improve palmarumycins C12 and C13 production. The addition of dibutyl phthalate, butyl oleate and oleic acid to the cultures of Berkleasmium sp. Dzf12 significantly enhanced palmarumycin C12 production by adsorbing palmarumycin C12 into the organic phase. When butyl oleate was fed at 5% (v/v) in medium at the beginning of fermentation (day 0), the highest palmarumycin C12 yield (191.6 mg/L) was achieved, about a 34.87-fold increase in comparison with the control (5.3 mg/L). n-Dodecane, 1-hexadecene and liquid paraffin had a great influence on the production of palmarumycin C13. When liquid paraffin was added at 10% (v/v) in medium on day 3 of fermentation, the palmarumycin C13 yield reached a maximum value (134.1 mg/L), which was 4.35-fold that of the control (30.8 mg/L). Application of the aqueous-organic solvent system should be a simple and efficient process strategy for enhancing palmarumycin C12 and C13 production in liquid cultures of the endophytic fungus Berkleasmium sp. Dzf12.

Clonostachys (teleomorph: Bionectria) fungi are well known to produce a variety of secondary metabolites with various biological activities to show their pharmaceutical and agrochemical applications. Up to now, at least 229 secondary metabolites, mainly including 84 nitrogen-containing metabolites, 85 polyketides, 40 terpenoids, and 20 other metabolites, have been reported. Many of these compounds exhibit biological activities, such as cytotoxic, antimicrobial, antileishmanial, antimalarial activities. This mini-review aims to summarize the diversity of the secondary metabolites as well as their occurrences in Clonostachys fungi and biological activities.

Nitrogen and phosphorus excessive enrichment are major causes of water eutrophication, and variations in nutrients enrichment are strongly influenced by human activities. In this study, annual average water quality from 2001 to 2018 was used to explore the spatiotemporal variations in total nitrogen (TN) and total phosphorus (TP) and their relationships with human activities. Spatially, TN and TP concentrations exhibited significant variations across the five sub-lake zones, and their values were relatively higher in the NW lake zone than the other sub-lake zones. Temporally, TN concentration exhibited weak correlations with years in the NW (R2 = 0.37, p < 0.05) and NE (R2 = 0.43, p < 0.05) lake zones and significant and positive correlations with years in the SW (R2 = 0.62, p < 0.05), SE (R2 = 0.79, p < 0.05), and C (R2 = 0.84, p < 0.05) lake zones. TP concentration exhibited decreasing trends in all lake zones except the NW lake zone (R2 = 0.37, p < 0.05), its value shows a relatively low level and is the restrictive factor to algal growth. The trophic state of the Lake Qiandaohu was determined as mesotrophic. Gross domestic product (GDP) and construction land exhibited strong correlations with TN and TP. Moreover, agriculture nonpoint source pollution was the largest contributor to the excessive enrichment of TN and TP, resulting in water eutrophication. In addition, aquaculture was another major source of nutrients starting in 1999. Although the managers of Lake Qiandaohu implemented a protection-oriented fishery policy, good results cannot be easily achieved with a unilateral policy concerning environmental protection. Thus, comprehensive policies may be more effective than unilateral policies.

Mycotoxins are toxic fungal secondary metabolites that pose a major threat to the safety of food and feed. Mycotoxins are usually converted into less toxic or non-toxic metabolites through biotransformation that are often made by living organisms as well as the isolated enzymes. The conversions mainly include hydroxylation, oxidation, hydrogenation, de-epoxidation, methylation, glycosylation and glucuronidation, esterification, hydrolysis, sulfation, demethylation and deamination. Biotransformations of some notorious mycotoxins such as alfatoxins, alternariol, citrinin, fomannoxin, ochratoxins, patulin, trichothecenes and zearalenone analogues are reviewed in detail. The recent development and applications of mycotoxins detoxification through biotransformation are also discussed.

Two new decalin/tetramic acid hybrid metabolites, hyalodendrins A (1) and B (2) were isolated from plant endophytic fungus Hyalodendriella sp. Ponipodef12. The structures of the new compounds were elucidated by analysis of the spectroscopic data, including NMR, HRMS and ECD, and by chemical conversion. Compounds 1 and 2 were phomasetin analogues, and both showed potent larvicidal activity against the fourth-instar larvae of Aedes aegypti with the median lethal dose (LC50) values of 10.31 and 5.93 μg/mL, respectively.

Rice false smut has become an increasingly serious fungal disease in rice (Oryza sativa L.) production worldwide. Ustilaginoidins are bis-naphtho-γ-pyrone mycotoxins previously isolated from the rice false smut balls (FSBs) infected by the pathogen Villosiclava virens in rice spikelets on panicles. To investigate the main ustilaginoidins and their distribution in rice FSBs, five main bis-naphtho-γ-pyrones, namely ustilaginoidins A (1), G (2), B (3), I (4) and C (5), were isolated and identified by NMR and high-resolution mass spectrometry as well as by comparison with the data in the literature. The rice FSBs at early, middle and late maturity stages were divided into their different parts and the contents of five main ustilaginoidins for each part were determined by HPLC analysis. The results revealed that the highest levels of ustilaginoidins were in late stage rice FSBs, followed by those at middle stage. Most ustilaginoidins, 96.4% of the total quantity, were distributed in the middle layer at early stage. However, ustilaginoidins were mainly distributed in the outer and middle layers at middle and late stages. Small amounts of ustilaginoidins A (1) and G (2) were found in the inner part of rice FSBs at each maturity stage. The contents of ustilaginoidins A (1) and G (2) without hydroxymethyl groups at C-2 and C-2’ of the γ-pyrone rings in rice FSBs were relatively high at early stage, while the contents of ustilaginoidins B (3), I (4), and C (5) with hydroxymethyl groups at C-2 or C-2’ were relatively high at late stage.

Rice false smut is an emerging and economically-important rice disease caused by infection by the fungal pathogen Villosiclava virens. Ustiloxin B is an antimitotic cyclopeptide mycotoxin isolated from the rice false smut balls that formed in the pathogen-infected rice spikelets. A monoclonal antibody (mAb) designated as mAb 1B5A10 was generated with ustiloxin B—ovalbumin conjugate. A highly-sensitive and specific indirect competitive enzyme-linked immunosorbent assay (icELISA) was then developed. The median inhibitory concentration (IC50) of the icELISA was 18.0 ng/mL for the detection of ustiloxin B; the limit of detection was 0.6 ng/mL, and the calibration range was from 2.5 to 107.4 ng/mL. The LOD/LOQ values of the developed ELISA used for the determination of ustiloxin B in rice false smut balls and rice grains were 12/50 μg/g and 30/125 ng/g, respectively. The mAb 1B5A10 cross-reacted with ustiloxin A at 13.9% relative to ustiloxin B. Average recoveries of ustiloxin B ranged from 91.3% to 105.1% for rice false smut balls at spiking levels of 0.2 to 3.2 mg/g and from 92.6% to 103.5% for rice grains at spiking levels of 100 to 5000 ng/g. Comparison of ustiloxin B content in rice false smut balls and rice grains detected by both icELISA and high performance liquid chromatography (HPLC) demonstrated that the developed icELISA can be employed as an effective and accurate method for the detection of ustiloxin B in rice false smut balls, as well as rice food and feed samples.

Background/Aims: Prevention of diabetes requires maintenance of a functional beta-cell mass, the postnatal growth of which depends on beta cell proliferation. Past studies have shown evidence of an effect of an incretin analogue, Exendin-4, in promoting beta cell proliferation, whereas the underlying molecular mechanisms are not completely understood. Methods: Here we studied the effects of Exendin-4 on beta cell proliferation in vitro and in vivo through analysing BrdU-incorporated beta cells. We also analysed the effects of Exendin-4 on beta cell mass in vivo, and on beta cell number in vitro. Then, we applied specific inhibitors of different signalling pathways and analysed their effects on Exendin-4-induced beta cell proliferation. Results: Exendin-4 increased beta cell proliferation in vitro and in vivo, resulting in significant increases in beta cell mass and beta cell number, respectively. Inhibition of PI3K/Akt signalling, but not inhibition of either ERK/MAPK pathway, or JNK pathway, significantly abolished the effects of Exendin-4 in promoting beta cell proliferation. Conclusion: Exendin-4 promotes beta cell proliferation via PI3k/Akt signaling pathway.

Hyalodendriella sp. Ponipodef12, an endophytic fungus from the hybrid “Neva” of Populus deltoides × P. nigra, is a high producer of the bioactive dibenzo-α-pyrones botrallin and TMC-264. However, both the botrallin and TMC-264 produced by Hyalodendriella sp. Ponipodef12 were retained as both intracellular and extracellular products. The aim of this study was to evaluate an in situ macroporous resin adsorption for enhancement of botrallin and TMC-264 production in mycelial liquid culture of Hyalodendriella sp. Ponipodef12. Production of botrallin and TMC-264 was most effectively enhanced by macroporous resin DM-301 among the thirteen nonionic macroporous resins tested. The highest botrallin yield (51.47 mg/L, which was 2.29-fold higher than the control at 22.49 mg/L) was obtained by adding resin DM-301 at 4.38% (g/mL) to the culture broth on day 24 and allowing a period of 4 days for adsorption. The highest TMC-264 yield reached 47.74 mg/L, which was 11.76-fold higher than that of the control (4.06 mg/L), and was achieved by adding DM-301 resin at 4.38% (w/v) in the culture broth on day 24 and allowing a period of 6 days for adsorption. The results show that in situ resin adsorption is an effective strategy for enhancing production of botrallin and TMC-264, and also for facilitating their recovery from mycelial liquid culture of Hyalodendriella sp.
