To investigate the effect of mild cataract and its surgery on the ISCEV standard electroretinogram (ERG) by comparing pre- and postoperative ERGs elicited from fully dilated eyes. Twenty-two patients participated. Each eye had cataract of grade 2 according to Emery-Little classification. None had complications during and after phacoemulsification and intraocular lens implantation. According to the ISCEV standard, pre- and 1-week postoperative full-field ERGs, dark adapted (DA) 0.01, DA 3, DA 3 oscillatory potentials (OPs), DA 10, and light adapted (LA) 3, and LA 3 flicker, were elicited from fully dilated eyes using skin electrodes. Photopic negative response (PhNR) 1 amplitude was measured from the baseline to the trough before the i-wave in the LA 3 ERG waveform. Statistical analysis was performed using SigmaPlot version 11.0 (Systat Software, Inc., San Jose, CA, USA). Pre- and postoperative data were compared using a paired t-test. Non-normally distributed data were evaluated using the Wilcoxon signed rank test. All study participants were subjected to a standard protocol of examination and electrophysiological testing (single flash VEP response, light adapted, pupils not dilated), reporting on the P2 implicit time, N1-P1 and N2-P2 (amplitude).Results: The mean±SD of the age of the study children and controls was 43.22± 33.2 and 55.68± 35.2 months respectively (p=0.217). The mean±SD IOP and cup/disc ratio of the study children were 18.4± 5.2 and 5.2± 3.3mmHg and 0.7± 0.2 and 0.3± 0.3 at presentation and at testing, respectively. The VEP testing was conducted after 21.5± 21.3 months of surgery for PCG. There was no statistically significant differences in P2, N1-P1 and N2-P2 between patients and controls (p=0.941, 0.916,0.945, respectively). There was no statistically significant correlation between most of the clinical characteristics of the study eyes and any of the studied VEP parameters.Conclusion: Operated controlled PCG eyes have VEP parameters that match their normal fellow children.Keywords: primary congenital glaucoma, visual evoked potentials, optic nerve, electrophysiology

Dopamine modulation of retinal signaling has been shown to be an important part of retinal adaptation to increased background light levels, but the role of dopamine modulation of retinal inhibition is not clear. We previously showed that light adaptation causes a large reduction in inhibition to rod bipolar cells, potentially to match the decrease in excitation after rod saturation. In this study, we determined how dopamine D1 receptors in the inner retina contribute to this modulation. We found that D1 receptor activation significantly decreased the magnitude of inhibitory light responses from rod bipolar cells, whereas D1 receptor blockade during light adaptation partially prevented this decline. To determine what mechanisms were involved in the modulation of inhibitory light responses, we measured the effect of D1 receptor activation on spontaneous currents and currents evoked from electrically stimulating amacrine cell inputs to rod bipolar cells. D1 receptor activation decreased the frequency of spontaneous inhibition with no change in event amplitudes, suggesting a presynaptic change in amacrine cell activity in agreement with previous reports that rod bipolar cells lack D1 receptors. Additionally, we found that D1 receptor activation reduced the amplitude of electrically evoked responses, showing that D1 receptors can modulate amacrine cells directly. Our results suggest that D1 receptor activation can replicate a large portion but not all of the effects of light adaptation, likely by modulating release from amacrine cells onto rod bipolar cells. NEW & NOTEWORTHY We demonstrated a new aspect of dopaminergic signaling that is involved in mediating light adaptation of retinal inhibition. This D1 receptor-dependent mechanism likely acts through receptors located directly on amacrine cells, in addition to its potential role in modulating the strength of serial inhibition between amacrine cells. Our results also suggest that another D2/D4 receptor-dependent or dopamine-independent mechanism must also be involved in light adaptation of inhibition to rod bipolar cells.

The effects of dopamine D2-class receptor blockade by sulpiride on the electroretinographic (ERG) b-wave (ON response) and d-wave (OFF response) were investigated in light-adapted turtle (Trachemys scripta elegans) and frog (Rana ridibunda) eyecups. For turtle ERG, sulpiride (240 μM) produced an amplitude increase of the b- and d-waves, while the 40 μM and 120 μM of sulpiride were ineffective. Alternatively, for frog ERG, a well-developed and dose-dependent b- and d-wave amplitude decrease was obtained with 40 μM and 240 μM sulpiride. In both species, 240 μM sulpiride significantly affected the maximal voltage range of the ERG responses without altering their relative sensitivity (determined by the semi-saturation point). The absolute sensitivity of the ON and OFF responses (evaluated by threshold estimation) was not significantly altered for turtle ERG, but it was decreased for frog ERG. The time characteristics of the ERG responses were unchanged in both species. Our results show important differences between dopamine D2-class receptor-mediated pathways in turtle and frog retina (revealed by ERG).

Many demersal fish species undergo vertical shifts in habitats during ontogeny especially after larval metamorphosis. The visual spectral sensitivity shifts with the habitat, indicating a change in colour vision. Colour vision depends on sufficient ambient light and becomes ineffective at a particular low light intensity. It is not known how fishes see colour in dim light. By means of a behavioural experiment on larval African catfish Clarias gariepinus in the laboratory, we determined colour vision and colour discrimination in dim light. Light-adapted larvae were subjected to classical conditioning to associate a reward feed with a green or a red stimulus placed among 7 shades of grey. The larvae learned this visual task after 70 and 90 trials. A different batch of larvae were trained to discriminate between green and red and then tested for the ability to discriminate between these colours, as the light intensity was reduced. The larvae learned this visual task after 110 trials in bright light and were able to discriminate colours, as light was dimmed until 0.01 lx, the minimal illuminance measurable in this study, and similar to starlight. The retinae of the larvae were found to be light adapted at 0.01 lx; thus indicating cone-based colour vision at this illuminance. For comparison, three human subjects were tested under similar conditions and showed a colour vision threshold at between 1.5 and 0.1 lx. For the larvae of C. gariepinus, the ability of colour discrimination in dim light is probably due to its retinal tapetum, which could increase the sensitivity of cones.

The International Society for Clinical Electrophysiology of Vision (ISCEV) standard for full-field electroretinography (ERG) describes a minimum protocol for clinical testing but encourages additional ERG testing when appropriate. This ISCEV extended protocol describes methods to record and evaluate a light-adapted (LA) ERG stimulus–response series with increasing flash strengths. The LA ERG stimulus–response series (also referred to as the luminance–response or intensity–response series in the published literature) can characterise generalised cone system function more comprehensively than the ISCEV standard LA ERGs alone. The amplitude of LA ERG a-waves, arising from cones and cone off-bipolar cells, typically shows a saturating function. The LA ERG b-wave amplitudes, which arise primarily from activity of retinal bipolar cells, show an amplitude peak followed by a nonzero plateau (the “photopic hill” phenomenon). This ISCEV extended protocol specifies a stimulus–response series suitable to evaluate generalised dysfunction affecting the LA retina, to aid in distinguishing between the on- and off-responses of the cone system and to monitor ERG changes in these characteristics. The LA ERG stimulus–response series for a- and b-waves is recorded to a sequence of nine flash stimuli ranging from 0.03 to 300 cd s m−2, superimposed on a standard background of 30 cd m−2. A shorter protocol is also presented to measure the mid-range of the function (the “photopic hill”) using 5 flash stimuli.

During adaptation from dim to bright environments, changes in retinal signaling are mediated, in part, by dopamine. Dopamine is released with light and can modulate retinal receptive fields, neuronal coupling, inhibitory receptors, and rod pathway inhibition. However, it is unclear how dopamine affects inner retinal inhibition to cone bipolar cells, which relay visual information from photoreceptors to ganglion cells and are important signal processing sites. We tested the hypothesis that dopamine (D)1 receptor activation is sufficient to elicit light-adapted inhibitory changes. Local light-evoked inhibition and spontaneous activity were measured from OFF cone bipolar cells in dark-adapted mouse retinas while stimulating D1 receptors, which are located on bipolar, horizontal, and inhibitory amacrine cells. The D1 agonist SKF38393 reduced local inhibitory light-evoked response magnitude and increased response transience, which mimicked changes measured with light adaptation. D1-mediated reductions in local inhibition were more pronounced for glycinergic than GABAergic inputs, comparable with light adaptation. The effects of D1 receptors on light-evoked input were similar to the effects on spontaneous input. D1 receptor activation primarily decreased glycinergic spontaneous current frequency, similar to light adaptation, suggesting mainly a presynaptic amacrine cell site of action. These results expand the role of dopamine to include signal modulation of cone bipolar cell local inhibition. In this role, D1 receptor activation, acting primarily through glycinergic amacrine cells, may be an important mechanism for the light-adapted reduction in OFF bipolar cell inhibition since the actions are similar and dopamine is released during light adaptation. NEW & NOTEWORTHY Retinal adaptation to different luminance conditions requires the adjustment of local circuits for accurate signaling of visual scenes. Understanding mechanisms behind luminance adaptation at different retinal levels is important for understanding how the retina functions in a dynamic environment. In the mouse, we show that dopamine pathways reduce inner retinal inhibition similar to increased background luminance, suggesting the two are linked and highlighting a possible mechanism for light adaptation at an early retinal processing center.

To analyze the effects of different methods of measurement on the photopic negative response (PhNR), recorded as part of a standard ISCEV Photopic 3.0 ERG responses from patients with a variety of clinical diagnoses. ERGs were recorded from both eyes of 97 patients (187 eyes) as part of a standard clinical assessment. The average age was 56.4 ± 15.7 years, the gender balance was 35 M, 62F, and only recordable responses of acceptable quality were included. PhNR was measured at an identifiable trough before (PhNR1) and after the i-wave (PhNR2), and the amplitudes and peak times were compared with a-, b- and i-wave corresponding parameters. PhNR components were measured: from baseline and from b-wave peak. Correlation between PhNR troughs and with ERG parameters were tested for right eyes. The possibility to predict and substitute PhNR2 amplitude from PhNR1 amplitude was also tested. PhNR1 was recordable in 97.3% of eyes and PhNR2 in 85.6%. An identifiable PhNR2 peak was found to occur before 65 ms at ~ 50% of the records, while in ~ 38% of the cases was within 65–75 ms in ~ 12%—after 75 ms. The correlation between the PhNR1 and PhNR2 peaks was quite strong (with coefficients 0.81–0.98, depending on method of measurement, and slopes close to 1). Visual abilities of the honey bee have been studied for more than 100 years, recently revealing unexpectedly sophisticated cognitive skills rivalling those of vertebrates. However, the physiological limits of the honey bee eye have been largely unaddressed and only studied in an unnatural, dark state. Using a bright display and intracellular recordings, we here systematically investigated the angular sensitivity across the light adapted eye of honey bee foragers. Angular sensitivity is a measure of photoreceptor receptive field size and thus small values indicate higher visual acuity. Our recordings reveal a fronto-ventral acute zone in which angular sensitivity falls below 1.9°, some 30% smaller than previously reported. By measuring receptor noise and responses to moving dark objects, we also obtained direct measures of the smallest features detectable by the retina. In the frontal eye, single photoreceptors respond to objects as small as 0.6° × 0.6°, with >99% reliability. This indicates that honey bee foragers possess significantly better resolution than previously reported or estimated behaviourally, and commonly assumed in modelling of bee acuity.

Although many aspects of white-tailed deer (Odocoileus virginianus) biology and physiology have been studied thoroughly, few studies have examined deer perception. We obtained a behavioral measure of light-adapted visual sensitivity based on performance of 7 deer in forced-choice discrimination tests. We compared intensity thresholds at the short and long wavelengths of their purported visual spectrum. Utilizing an automated training device, we trained deer to discriminate between lit and unlit light-emitting diode (LED) lights expressing monochromatic wavelengths, and recorded performance across a series of decreasing intensities for each wavelength until deer could no longer discriminate the light stimulus. We regressed a best-fit logistic function to each deer's performance as intensity decreased at a single wavelength, demarcated the sensitivity threshold to that wavelength, and compared deer's sensitivity threshold across wavelengths. Our behavior-based sensitivity measurements agreed with previous estimates of white-tailed deer's cone photoreceptor sensitivity. We confirm that these cone photoreceptors contribute to deer behavior, and that deer have a greater perceptual sensitivity to shorter wavelengths and lower sensitivity to longer wavelengths. Our findings also suggest the deer have some sensitivity to ultraviolet light. The visual perception of the white-tailed deer is specialized for sensitivity during their crepuscular patterns, enables detection of predators along their horizon, and is well-adapted for a prey species. © 2014 The Wildlife Society.

To assess the results of perimetry recorded under dark- and light-adapted (DA and LA) conditions in patients with acute zonal occult outer retinopathy (AZOOR) and to compare the results of electroretinography (ERG) and spectral-domain optical coherence tomography (SD-OCT) in two groups of AZOOR patients.Twelve patients with AZOOR were studied. The diagnosis of AZOOR was based on the results of ophthalmoscopy, Goldmann kinetic perimetry, and multifocal ERGs. In addition, DA and LA perimetry, ERG, and SD-OCT were performed. The patients were followed for 1-9 years.The patients were classified into two types: type A patients (3) had a scotoma detected by both DA and LA perimetry, normal or equally abnormal cone and rod ERGs, atrophy of the outer nuclear layer (ONL), and disruption of the inner segment/outer segment (IS/OS) junction line in the OCT images. Type B patients (7) had a scotoma that was more prominent in LA than in DA perimetry and a continuous IS/OS junction line in the OCT images. Two patients had characteristics of both type A and type B AZOOR.Our findings suggest that eyes with type A AZOOR have focal and severe impairment of both the rods and cones, and eyes with type B AZOOR have focal and specific impairment of the cones.

This paper reports the first electrolysis-based oxygen generation and transportation device for the treatment of retinal ischemia; containing an isolated biocompatible electrolyte and inductive power transfer for on-demand oxygen. A finite element model of the retina along with oxygen delivery data demonstrates the feasibility of the device. The estimated therapeutic window for the device was found to be 210 minutes for the light adapted retina, and 55 minutes for the dark adapted retina.

The relatively large eye and pupil of the Piked Dogfish, Squalus acanthias, is visually arresting. However, knowledge of its basic visual characteristics lags far behind other areas in this generally well studied species. This study quantifies pupil dilation in a species that is naturally exposed to a broad range of light intensities and finds that the pupil area in the dark adapted state is 35.3% of the total eye area, an increase of 12.4% from the light adapted state. The anterior and posterior extents of the horizontal visual field are assessed and compared with both morphological and electrophysiological techniques and the results are integrated with the measured head yaw to derive the anterior convergence distance and blind area. The position of the eyes and the triangular, pointed snout of S. acanthias provides excellent anterior vision, which likely facilitates foraging upon its mobile prey.

We present a picosecond transient circular dichroism study of OBIP, the putative photoreceptor protein involved in the photophobic response of Blepharisma japonicum. The probe wavelength was chosen at 230 nm. The results are compared to those of the isolated chromophore, OxyBP, in solution. The CD changes in OBIP and OxyBP do not show the same dynamics: OBIP’s signal relaxes in a few ps whereas no such decay is obtained for OxyBP. This observation brings support to the formerly evoked existence of a fast photoinduced reaction in the chromoprotein, and demonstrates the implication of local geometrical changes that accompany this process. 2009 Published by Elsevier B.V.

The EOG is influenced by the corneo-retinal potential of the contralateral eye. This contralateral influence results in polarity reversal in severely destructed eyes which do not have their own corneo-retinal potential. In eyes with a decreased corneo-retinal potential the contralateral effect can cause polarity reversal only during the light adapted period. Contralateral influence on eyes with abolished light sensitive component but a relatively well preserved light-insensitive component results in a paradoxical curve. Deterioration of the light-sensitive part of the EOG concerns primarily the positive component. In severely impaired eyes the EOG value should be corrected to allow for the influence of the contralateral sound eye; a formula, devised for calculating the corrected values, is given in a second paper.

Retinomotor movements are morphological changes in the outer retina in response to changing light conditions. They can be separated into two components: Migration of pigment granules within the microvilli of the retinal pigment epithelium (RPE) and positional changes in photoreceptor cells. These positional changes optimize exposure of the cone and rod photoreceptors to light. The aim of this study was to analyze both the time course of retinomotor movements in the adult zebrafish and the maturation of these processes in the developing fish. We show that retinomotor movements are used as a dark/light adaptation mechanism in zebrafish. In adult zebrafish, melanin granules of the RPE migrate with constant speed and reach the fully light adapted (LA) state approximately after 1 h. In contrast, about two thirds of double cone outer segment movements are finished in 5 min, and are fully completed in 10 to 20 min. During development there are three crucial stages leading to mature retinomotor movements in response to light: at 5 dpf (days post fertilization) the migration of pigment granules begins, at 20 dpf the pigment granules condense in the apical part of the RPE microvilli, and at 28 dpf, concomitant with the functional maturation of rods, the double cones contract as in adult retinas.

We studied effects of dark adaptation on spatial and temporal tuning for motion coherence detection. We compared tuning for step size and delay for moving random pixel arrays (RPAs) at two adaptation levels, one light adapted (50 cd/m2) and the other relatively dark adapted (0.05 cd/m2). To study coherence detection rather than contrast detection, RPAs were scaled for equal contrast detection at each luminance level, and a signal-to-noise ratio paradigm was used in which the RPA is always at a fixed, supra-threshold contrast level. The noise consists of a spatio-temporally incoherent RPA added to the moving RPA on a pixel-by-pixel basis. Spatial and temporal limits for coherence detection were measured using a single step pattern lifetime stimulus, in which patterns on alternate frames make a coherent step and are being refreshed. Therefore, the stimulus contains coherent motion at a single combination of step size and delay only.The main effect of dark adaptation is a large shift in step size, slightly less than the adjustment of spatial scale required for maintaining equal contrast sensitivity. A similar change of preferred step size occurs also for scaled stimuli at a light-adapted level, indicating that the spatial effect is not directly linked to dark adaptation, but more generally related to changes in the available low-level spatial information. Dark-adaptation shifts temporal tuning by about a factor of 2. Long delays are more effective at low luminance levels, whereas short delays no longer support motion coherence detection. Luminance-invariant velocity tuning curves, as reported previously [Lankheet, M.J.M., van Doorn, A.J., Bouman, M.A., & van de Grind, W.A. (2000) Motion coherence detection as a function of luminance in human central vision. Vision Research, 40, 3599–3611], result from recruitment of different sets of motion detectors, and an adjustment of their temporal properties.

We evaluated the generality of two models of vertebrate phototransduction. The approach was to quantitatively optimize each model to the full waveform of high-quality, dark-adapted (DA), salamander rod flash responses. With the optimal parameters, each model was then used to account for signature, qualitative features of rod responses from three experimental paradigms (stimulus/response, “S/R suite”): (1) step responses; (2) the intensity dependence of the period of photocurrent saturation (Tsatvs. ln(I)); and (3) light-adapted (LA) incremental flash sensitivity as a function of background intensity. The first model was the recent successful model of Nikonov et al. (1998). The second model replaced the instantaneous Ca2+ buffering used in the Nikonov et al. model with a dynamic buffer. The results showed that, in the absence of the dynamic Ca2+ buffer, the Nikonov et al. model does not have sufficient flexibility to provide a good fit to the flash responses, and, using the same parameters, reproduce the salient features of the S/R suite—critical features at step onset and offset are absent; the Tsat function has too shallow a slope; and the model cannot generate the empirically observed I-range of Weber–Fechner LA behavior. Some features could be recovered by changing parameters, but only at the expense of the fit to the reference (Ref) data. When the dynamic buffer is added, the model is able to achieve an acceptable fit to the Ref data while reproducing several features of the S/R suite, including an empirically observed Tsat function, and an extended range of LA flash sensitivity adhering to Weber's law. The overall improved behavior of the model with a dynamic Ca2+ buffer indicates that it is an important mechanism to include in a working model of phototransduction, and that, despite the slow kinetics of amphibian rods, Ca2+ buffering should not be simulated as an instantaneous process. However, neither model was able to capture all the features with the same parameters yielding the optimal fit to the Ref data. In addition, neither model could maintain a good fit to the Ref data when five key biochemical parameters were held at their current known values. Moreover, even after optimization, a number of important parameters remained outside their empirical estimates. We conclude that other mechanisms will need to be added, including additional Ca2+-feedback mechanisms. The present research illustrates the importance of a hybrid qualitative/quantitative approach to model development, and the limitations of modeling restricted sets of data.

Normal subjects were first light adapted to a standard photopic background and a control photopic ERG was obtained. The subjects were then light adapted to a brighter background for 5 min at the end of which the luminance was returned to the control background and ERGs were taken at regular intervals. Most of the ERG/OP components were transiently enhanced following the above procedure. Given that the previously reported photopic light adaptation effect occurred following an increase in the luminance of the adapting field (from dark adaptation to light adaptation) while that reported in the present study is triggered following a decrease in the level of light adaptation, the opposite effects noted might suggest that the two retinal events result from the same, not yet identified, cone adaptation mechanisms which are solicited in an opposite way.

The light response of fly photoreceptor cells is modulated by changes in free Ca2+concentration. Fly phototransduction and most processes regulating it take place in or very close to the rhabdomere. We therefore measured the kinetics and the absolute values of the free Ca2+concentration in the rhabdomere of fly photoreceptor cellsin vivoby making use of the natural optics of the fly's eye. We show that Ca2+flowing into the rhabdomere after light stimulation of dark-adapted cells causes fast Ca2+transients that reach peak values higher than 200 μmin 2+concentration has declined again to ∼20 μm. The duration of the Ca2+transients becomes still shorter, and their size reduced, when the photoreceptor cell is light-adapted. This reduction in duration and size of the Ca2+transients is graded with the intensity of the adapting light. The kinetics and absolute values of the free calcium concentration found to occur in the rhabdomere are suitable to mediate the fast feedback signals known to act on the fly phototransduction cascade.

To assess the effects of sleep state on human retinal electric responses, full-field electroretinograms were obtained in a cross-sectional study from 123 preterm infants at 36, 40 and 56 weeks of post-gestational age. At each age, electroretinographic recordings were assigned to one of two groups according to whether the infants were in active sleep or quiet sleep. Both sleep states were determined behaviorally. Pure rod, maximal, 30 Hz flicker and light adapted single cone responses were evaluated when a sleep state was clearly established. Peak-to-peak amplitudes of most electroretinographic responses were significantly larger in active sleep relative to quiet sleep at 36 and 40 weeks of post-gestational age. We speculate that larger amplitudes during active sleep may play a role in the maturation of the visual system.

Exposure of algae or higher plants to bright light can result in a photoinhibitory reduction in the number of functional PS II reaction centers (n) and a consequential decrease in the maximum quantum yield of photosynthesis. However, we found that light-saturated photosynthetic rates (Pmax) in natural phytoplankton assemblages sampled from the south Pacific ocean were not reduced despite photoinhibitory decreases in n of up to 52%. This striking insensitivity of Pmax to photoinhibition resulted from reciprocal increases in electron turnover (\({1 \mathord{\left/ {\vphantom {1 {\tau _{PSII} }}} \right. \kern-\nulldelimiterspace} {\tau _{PSII} }}\))through the remaining functional PS II centers. Similar insensitivity of Pmax was also observed in low light adapted cultures of Thalassiosira weissflogii (a marine diatom), but not in high light adapted cells where Pmax decreased in proportion to n. This differential sensitivity to decreases in n occurred because \({1 \mathord{\left/ {\vphantom {1 {\tau _{PSII} }}} \right. \kern-\nulldelimiterspace} {\tau _{PSII} }}\) was close to the maximum achievable rate in the high light adapted cells, whereas \({1 \mathord{\left/ {\vphantom {1 {\tau _{PSII} }}} \right. \kern-\nulldelimiterspace} {\tau _{PSII} }}\) was initially low in the low light adapted cells and could thus increase in response to decreases in n. Our results indicate that decreases in plant productivity are not necessarily commensurate with photoinhibition, but rather will only occur if decreases in n are sufficient to maximize \({1 \mathord{\left/ {\vphantom {1 {\tau _{PSII} }}} \right. \kern-\nulldelimiterspace} {\tau _{PSII} }}\) or incident irradiance becomes subsaturating.

Fourier-transform Raman spectra are reported for bacteriorhodopsin (BR) in its light-adapted and dark-adapted states at room and elevated temperatures. Qualitative changes are observed in the FT-Raman spectrum above the midpoint temperature of the predenaturational transition indicating structural changes and inhomogeneous protein populations. Dark-adapted BR shows an increased all- trans component at 78°C, while light-adapted BR at 78°C is found to contain 13- cis BR, possibly as a result of a reduced extent of light adaptation or a change in protein—chromophore interactions.

In three separate experiments albino rabbits, pigmented rabbits, and humans were tested following administration of acetazolamide and without acetazolamide. In all three experiments, we recorded electroretinograms (ERGs) under dark adapted and light adapted conditions and measured the b -wave amplitudes. Dark adapted ERG b -wave amplitudes were increased following administration of acetazolamide as compared to control conditions, in albino rabbits, pigmented rabbits and humans. Light adapted b -wave amplitudes showed no statistically significant changes as a function of acetazolamide administration although in all three experiments there was a trend toward light adapted b -wave amplitude reduction following administration of acetazolamide. In the human experiments, ERG a -wave amplitudes were also measured. Light adapted a -wave amplitudes were reduced following administration of acetazolamide. In the human experiments, several behavioral tests were performed, including L' Anthony desaturated D-15, Farnsworth-Munseu 100 hue, Cogan-Gunkel chromatograph, Nagel anomaloscope, Goldmann-Weckers dark adaptometry. There were no consistent changes in the human dark adaptation thresholds or color discrimination, although several measures approached significance.

Electroretinography (ERG) is an important clinical tool that provides an objective quantitative measure of retinal function. Decreased a and b wave amplitudes and prolonged latencies correlate to reductions in retinal function that may be the result of toxicity, ischemic damage, or retinal dystrophy (Fishman et al. 2001, Ophthalmology monographs). Furthermore, since the different components of the ERG waveform correspond to the different layers of the retina, one is able to attribute changes in the ERG to damage to specific retinal layers. This data can be a useful surrogate for retinal health, for example establishing safety profiles for drugs under clinical development. Since 1989 the International Society for Clinical Electrophysiology of Vision (ISCEV) has provided standards for the recording of ERGs. These documents provide a framework for the clinical electrophyisologist to obtain “standard” ERG recordings (Marmor 1989). The variety of permissible ERG instruments and their individual calibration requirements contributes to significant inter-laboratory variability. This variability is recognized in the ISCEV standards and partly addressed by stating “it is incumbent on the manufactures and users to verify that full-field stimulation meets the requirements of this standard.” Placing the onus for compliance on the manufactures but leaving the clinical electrophyisologist to determine if the recording standards are indeed met. ERG standards have extended beyond the aand b-wave of the full field flash ERG. The pattern ERG (PERG) is the electroretinal response to a pattern reversing stimulus such as bar gratings or checkerboard pattern. The PERG primarily reflects ganglion cell function and since it is viewed on display monitors it largely represents ganglion cell function within the macula. The peak and trough components of the PERG have been formally defined as the N35, P50 and N95 which represent the polarity (Negativity or Positivity) and the mean latency of occurrence. The ISCEV has produced standards for the recording and reporting of the PERG (Holder et al. 2007). While the PERG provides a single waveform which represents the electroretinal response of the entire macular region, the clinical multifocal electroretinogram (mERG) provides information of local retinal function. The mERG is recorded typically displaying the local retinal response of 61 or 103 local regions within the central 45 of the posterior pole. The responses represent localized cone-driven ERGs obtained in the light adapted state. While the waveform morphology of the mERG is similar to the fullfield ERG the electroretinal

The present study examines the hypothesis that the electroretinogram (ERG) a-wave of patients with congenital stationary night blindness with myopia (CSNB) is solely cone mediated. The subjects include four patients with x-linked recessive inheritance, two patients with no family history, and thirteen age-matched normal subjects. Because normal subjects have relatively large b-waves (which can influence the measurement of the PIII component of the a-wave), the main response parameter studied was the slope of the a-wave. The results for the CSNB patients show that in the light adapted condition, the ratio of the responses to photopically balanced blue and red flashes was nearly unity; whereas, in the dark-adapted condition, the ratio was ~3 times greater. The ratios for the normal subjects were similar to those of the CSNB patients. These results lead to the conclusion that, in patients with CSNB, the ERG a-wave is not solely mediated by cones.

Loss in foveal sensitivity in retinitis pigmentosa (RP) has been attributed to a decrease in quantal catching ability. Using a psychophysical technique (the probe–flash paradigm), we previously found that the results obtained for six RP patients under light adapted conditions were not consistent with a quantal catch hypothesis. To test further this hypothesis twelve RP patients were examined in dark adapted conditions. Probe thresholds were normal for five patients and increased for seven patients. The decreased quantal catching hypothesis was rejected for six of the seven patients.

Summary. 1. As a rule the refractive index for dark adapted retinae of cows was 1.358. 2. In a material of 321 cases the light adapted retina's index was 1.3610 ± 0.0008. 3. The experiments carried out support the supposition expressed by Kuhne Aand Ewald that visual purple exudes water when it is bleached. For this reason the value of the refractive index in light adapted retinae is higher than that in dark adapted ones. 4. The retinal refractive index in newly born calves in a material of 104 samples was 1.3621 ± 0.0012. 5. The refractive index for the retinae of pigs was 1.35733 ± ± 0.00167. The low index value possibly depends on the fact that the retina is poor in rods. 6. The refractive index in horses' eyes was as a rule 1.361.

We recorded electroretinograms (ERG) with white and color stimuli in normal persons and four patients with cone dystrophies. Kodak Wratten-filters in blue, blue-green, green, yellow and red were used for the color flashes. ERGs to all color stimuli were recorded at dark and light adapted conditions with different stimulus intensities, to 30 Hz flicker stimulation and with special filtering for oscillatory potentials. Selective blue cone-responses were obtained using strong blue flashes at a yellow background. Patients with cone dystrophies showed slightly to moderately reduced responses at dark adaptation to blue, blue-green, green and yellow stimuli. Red stimuli elicited only small responses with a markedly delayed b-wave implicit time. The light adapted recordings, flicker responses and oscillatory potentials were reduced to all color stimuli. However, differences between patients with cone dystrophies could be detected concerning the responses to red and green. In two patients the responses were reduced to the same degree to all color stimuli. Another patient had very small responses and no oscillatory potentials to red, but his responses were only moderately reduced to green. A patient with combined red-green cone and rod dystrophy had a blue cone hypersensitivity. Responses to blue and blue-green were large at all stimulus conditions, but responses to all other stimuli were much smaller. The blue cone ERG showed a prominent blue cone response. ERG recording to colored stimuli allows a separation of retinal dysfunction in patients with cone dystrophies.

Utilizing a high intensity photographic flash unit, electroretinograms were recorded from normal adults under fully light adapted conditions over a 5 log unit range of stimulus luminance (-1.35 to 3.34 log cd-s/m2). At lower luminance levels b-wave amplitude increased with increased luminance until it reached a maximum (Vmax of the Naka-Rushton equation) in agreement with previous work. At higher luminance levels, the b-wave amplitude decreased to 33% of Vmax and then plateaued. This previously unreported phenomenon has been named the photopic hill. There was no appreciable change in b-wave amplitude with increased interstimulus intervals from 15 sec to 5 min and luminance-response functions serially recorded for increasing and for decreasing stimulus luminance were very similar. These latter results indicate that the photopic hill is not due to light adaptation. The reason for the photopic hill and possible clinical implications are discussed.

A standard methodology for assessing the impacts of simulated solar ultraviolet-B radiation (UV-B) on aquatic invertebrates was established. A solar simulator was used to expose a variety of aquatic invertebrates to different levels of UV-B. The simulator was calibrated as close as possible to match local ambient solar radiation measured in and out of water with a scanning spectroradiometer. A series of repeated exposures were conducted to determine the effects of UV-B on two species of Ceriodaphnia. Survivorship of C. reticulata declined with increasing UV-B with 100% mortality occurring after four daily 5 hr exposures to a UV-B irradiance that was 14% of ambient sunlight (40.8μW/cm 2 ) and 70% mortality for C. dubia after seven days of an exposure to 5% of ambient (14.5μW/cm 2 ). Significant reductions in fertility (#young/adult) was observed in both low and high light adapted individuals with low light individuals appearing to be more sensitive. This methodology allowed us to make comparisons to natural conditions in aquatic habitats and to make risk assessments for individual species.

The role of dopamine as a neurotransmitter in the retina of different species has been clearly established; however, there is still some controversy as to whether noradrenaline (NA) is present as a neurotransmitter in this tissue. In this study, we show that, under controlled conditions, NA is present in the retina of goldfish at a concentration of 0.15 +/- 0.03 ng/mg protein and its biosynthetic enzyme, dopamine beta-hydroxylase shows an activity of 2.5 +/- 0.2 pmol NA/hr/mg protein. The amount of NA increases to 1.88 +/- 0.24 ng/mg protein in light adapted animals and decreases to undetectable levels in dark adapted ones. By contrast, dopamine-beta-hydroxylase levels are not affected by changes in light conditions. This finding provides further evidence in favor of a neurotransmitter role for NA in vertebrate retina.

Escalator passengers sometimes experience a feeling of disorientation or dizziness while boarding or riding the machine. Prior work has shown that this perception may be due to a visual depth illusion caused by the appearance of the escalator tread. Measurements confirm that the escalator tread surface is a highly periodic visual stimulus exhibiting unusually high contrast as well as a spatial frequency matched to the preferences of the visual nervous system. Means of minimizing the depth illusion are explored. These center on disrupting the periodic structures of the tread surface. Expressions intended to serve as candidates for figure of merit for the tread appearance are developed. These take into account some properties of the visual nervous system including the decline with eccentricity of visual acuity and the bandpass character of spatial detail processing under light adapted conditions.

The psocopteran Psyllipsocus ramburi Selys-Longchamps can render food stuffs unpalatable and may serve as an intermediate host for cestodes. Its two circular compound eyes consist of about 26 facets, capped by strongly convexly curved corneae of 10-18 microm in diameter. Corneal nipples or interommatidial hairs are not developed. Beneath each corneal lens a cluster of four cone cells, enveloped by two primary pigment cells, separates an ommatidial group of eight retinula cells from the inner corneal surface. Membrane specializations of the retinula cells, known as the microvilli, measure 60 nm in diameter, and collectively make up the rhabdom, which is columnar in shape and has a distal diameter of 4 or 5 microm, depending on whether it is day- or night-adapted. Cone cell lengths measure 4.5 microm during the day and 8.5 microm at night and retinula cell screening pigments closely approach the edge of the rhabdom during the day. A 1-h exposure to UV-A (lambda(max)=351 nm) of ca. 1200 lx causes an almost total destruction of the photoreceptive membranes of the rhabdom and bleached all retinula cell screening pigments, but not the pigment grains of the primary pigment cells. Calculations, based on the anatomical data, suggest that the eyes are adapted to function under dim light levels, but cannot produce sharp images since their best possible acceptance angles are 22 degrees and 28 degrees in light- and dark-adapted states, respectively. Destruction of vision, likely affecting biorhythm and reproduction, by exposing the insects to UV-A may offer an alternative to the use of chemicals in controlling these insects.

Two mechanisms of screening pigment migration in superposition compound eyes are described. The first is the radial pigment migration mechanism in which screening pigment migrates proximally from the dark adapted position around the distal crystalline, cones into the clear zone of the eye when light adapted. This mechanism is able to substantially attenuate light intensity at the retina for small migrations of screening pigment away from the dark adapted position, and is found to be correlated with arthropods which can experience a wide range of light intensities in their habitats. The second mechanism is the cone pigment migration mechanism in which screening pigment migrates proximally from the dark adapted position around the distal crystalline cones to a position which occludes all but a small region at the tip of the crystalline cone when light adapted. This mechanism is not able to attenuate light intensity at the retina as substantially as the first mechanism (for a given pigment migration), but has a much finer control. Consequently, this mechanism is found to be correlated with arthropods which are active at one light intensity only.

The wavelength discrimination threshold of three goldfish was examined in a series of behavioral experiments. Using an auto-shaping technique, detection thresholds were established for 531 and 648 nm spectral increments presented on a 6.6 cd m(-2) white background. Next, discrimination between the wavelengths was established at equal, suprathreshold, intensities. Finally, the intensities of the two stimuli were reduced to establish the intensity threshold for the wavelength discrimination. The results indicate that goldfish, like several mammalian species, can discriminate wavelength at detection threshold intensity. This finding suggests that high color sensitivity is not confined to mammals or dependent upon a very high percentage of wavelength opponent ganglion cells. Rather, high color vision sensitivity may be based upon an inherent sensitivity advantage of wavelength opponent receptive fields compared to non-wavelength opponent receptive fields and be an important selective advantage of wavelength opponency and color vision.

The AII amacrine cell is a critical interneuron in the rod pathway of the mammalian retina. Rod signals pass into cone pathways by means of gap junctions between AII amacrine cells and ON cone bipolar cells. Filling AII amacrine cells with Neurobiotin produces labeling of cone bipolar cells by means of these gap junctions. However, tracer injections into bipolar cells do not produce labeling of the AII network (Vaney [1997] Invest Ophthalmol Vis Sci. 38:267–273), which suggests that the AII/bipolar gap junctions allow the passage of tracer in only one direction. This mechanism stands in contrast to physiological results, which indicate that light adapted signals can pass from ON cone bipolar cells into the AII network (Xin and Bloomfield [1999] Vis Neurosci. 16:653–665). Here, we report that a variety of ON and OFF bipolar cells are sometimes anomalously coupled to the A-type horizontal cell network. These relatively rare examples do not result from dye injection errors, but seem to represent minor developmental errors. However, this provides a method to obtain Neurobiotin-filled cone bipolar cells without the necessity of impaling them with a microelectrode. Under these conditions, Neurobiotin spreads from ON cone bipolar cells into neighboring AII amacrine cells. The dye-coupled AII amacrine cells, positively identified by double labeling with an antibody against calretinin, were centered around anomalously coupled ON bipolar cells. These results indicate that AII/bipolar cell gap junctions allow tracer coupling in both directions, consistent with previous physiological results. The previous failure to detect the passage of neuronal tracer from injected bipolar cells to AII amacrine cells may reflect electrode damage or perhaps the asymmetrical voltage sensitivity of a heterotypic gap junction. A physiological model of dark-adapted and light-adapted photoreceptors in the honeybee worker (Apis mellifera) has been developed. Almost all of the electrophysiological components of photoreceptors known up to date, e.g. the phototransduction cascade, the ion channels of the membrane, phototransduction gain and optical adaptation mechanisms, are adequately described by simple biophysical and biochemical models. The connections of these components were tried out in synthetic simulations for best fits of simulated to intracellularly recorded membrane potentials. Although the parameters of the best model were determined exclusively for the measured membrane potentials of dark-adapted photoreceptors, the model also accurately describes the light-adapted photoreceptor membrane potentials. Furthermore, the model correctly predicts the time-courses of measured photoreceptor responses with respect to squared-modulated flicker lights up to 200 Hz. This clearly demonstrates that the presented photoreceptor model is indeed a physiologically adequate description of the essential components of the phototransduction and the electrical membrane processes in the photoreceptors of the honeybee worker.
