Abstract Octacosanol is a plant natural product with the potential to ameliorate alcoholic liver injury. However, its poor solubility limits its application in the food industry. This study first verified that octacosanol has a favorable effect on acute and subchronic alcoholic liver injury in mice by protecting the gastric mucosa and ameliorating disease levels. Additionally, octacosanol microcapsules were prepared under the optimal drying conditions of the feed flow rate of 400 mL/h and drying temperature of 180°C to improve the ameliorative effects of octacosanol, with moisture content, water activity, dispersibility, and solubility of 1.53%, 0.16%, 89.84%, and 95.30%, respectively. The microcapsules had a spherical structure, and the droplet sizes after redissolution ranged from 0.061 to 4.63 µm. Instrumental measurements and simulated digestion experiments have shown that microencapsulation enhanced the solubility and thermal stability of octacosanol, with 90.88% of the octacosanol being continuously released. As expected, microencapsulation significantly enhanced the effect of octacosanol in ameliorating alcoholic liver injury by determining the disease index in the mice model. This study demonstrated the beneficial effects of octacosanol on alcoholic liver injury and explored a viable method to improve its solubility and bioaccessibility, thereby enhancing its therapeutic efficacy for the treatment of alcoholic liver injury.

Study region: Loess Plateau, China Study focus: The potential soil erosion has commonly been estimated using the revised universal soil loss equation (RUSLE) under bare land conditions. In few studies, biogeochemical models have been employed to simulate natural vegetation dynamics and assess the effects of regional vegetation restoration on soil loss control. In this study, a biogeochemical Biome-BGC (BBGC) model was developed to simulate the potential vegetation dynamics. The soil erosion rate and erosion control indices under both the current land use/cover (A, ERI) and the potential vegetation conditions (A’, ERI’) were estimated using the RUSLE model and an index-based ecosystem service approach, respectively. The effects of vegetation restoration on erosion control services were ultimately evaluated. New hydrologic insights for the region: The BBGC model performed well in simulating the potential vegetation dynamics. The average annual soil retention increased by 84% after vegetation restoration since 1999. The increased erosion control services accounted for 62% of the Loess Plateau. Erosion control services increased to a high degree (ERI-ERI’ >0.4) in the southeast. Our results demonstrated that vegetation restoration effectively improved the erosion control services on the Loess Plateau. The results could provide a guidance in quantifying the regional effects of vegetation restoration on ecohydrological services and protecting the soil conservation function of watershed ecosystems.

Morchella sextelata and Morchella importuna are the main cultivars of morel. However, the key compounds affecting their flavors (taste and odor) are currently unknown. Here, an ultra performance tandem mass spectrometry combined with two-dimensional gas chromatography-time-of-flight mass spectrometry method was used to detect and relatively quantify the metabolites in both morel cultivars. A total of 631 non-volatile compounds and 242 volatile compounds were identified. The odor activity value was calculated to assess the contribution of key odor volatile. The results indicated that M. importuna had a sweeter flavor than M. sextelata. The former posed more prominent mushroom flavor than the latter based on the correlation analysis of the metabolites. The flavor differences of the two morel cultivars are highly relevant with the content of lipids, carbohydrates, amino acids and derivatives, alcohols and ketones. This study provides new insights into the theoretical basis for the flavor differences in both morel cultivars.

Chinese plum (Prunus salicina Lindl.) is a nutritionally and economically important stone fruit widely grown around the world. Anthracnose, caused by Collectotrichum spp., is one of the primary biotic stress factors limiting plum production. Medicinal plants may harbor rhizospheric or endophytic microorganisms that produce bioactive metabolites that can be used as anthracnose biocontrol agents. Here, 27 bacterial isolates from the medicinal plant A. conyzoides with diverse antagonistic activities against C. fructicola were screened. Based on morphological, physiological, biochemical, and molecular characterization, 25 of these isolates belong to different species of genus Bacillus, one to Pseudomonas monsensis, and one more to Microbacterium phyllosphaerae. Eight representative strains showed high biocontrol efficacy against plum anthracnose in a pot experiment. In addition, several Bacillus isolates showed a broad spectrum of inhibitory activity against a variety of fungal phytopathogens. Analysis of the volatile organic compound profile of these eight representative strains revealed a total of 47 compounds, most of which were ketones, while the others included alkanes, alkenes, alcohols, pyrazines, and phenols. Overall, this study confirmed the potential value of eight bacterial isolates for development as anthracnose biocontrol agents.

The project studied the differences of dried and fresh wolfberry fruit in Ningxia region. Using non-targeted metabolomics analysed the differential metabolites of wolfberry fruit. The observed variables were determined using ultra-high performance liquid chromatography-quadrupole-electrostatic field orbitrap high-resolution mass spectrometry (UPLC-QE-Orbitrap-MS). Differential metabolites were screened out by mathematical statistical methods (principal component analysis (PCA), partial least squares analysis (PLS-DA), univariate statistical analysis, cluster analysis, volcano plot) to identify and analyse all metabolites. The results showed that 35 different metabolites were identified in the fresh and dried wolfberry fruit. The expression of organic acids, amino acids and flavonoids was up-regulated, while the expression of nucleotides, phenolic acids and coumarin compounds was down-regulated. This study revealed the characteristic compounds of fresh and dried wolfberry fruit in Ningxia. The research would provide a theoretical basis for the material basis of different wolfberry fruits.

Abstract The deprotonation mechanism for the phenolic hydroxyl and the complexing of metal ions with a commonly used food additive, propyl gallate (PG) were studied theoretically and experimentally. The interaction of procyanidins [PC, epicatechin16 (4 → 8) catechin], and its basic monomeric unit catechin (CA) with metal ions was studied by the fluorescence quenching spectra. The results showed that the 9-OH quinoid PG was formed at higher pH (10.9) by the oxidization of phenolic hydroxyl. The binding affinities (K a) and stoichiometry of these metal ions with PG were determined. The Al3+ in PG-Al complex [Al(PG)(H2O)2Cl2]- was coordinated at the 8,9-OH doubly deprotonated catechol site with double chloride ions (Cl-) and double water molecules (H2O). The fluorescence quenching titration with Sn2+, Zn2+, Cu2+, Al3+ and Fe3+ revealed that the stoichiometries of metal-bound PC were 1:1, 2:3, 2:3, 2:3 and 1:1, respectively. The presence of bovine serum albumin (BSA) could enhance the complexing strength of PC with metal ions.

Abstract Background Newcastle disease virus (NDV) is a highly infectious viral disease, which can affect chickens and many other kinds of birds. The main virulence factor of NDV, the fusion (F) protein, is located on the viral envelope and plays a major role in the virus’ ability to penetrate cells and cause host cell fusion during infection. Multiple highly conserved tyrosine and di-leucine (LL) motifs in the cytoplasmic tail (CT) of the virus may contribute to F protein functionality in the viral life cycle. Methods To examine the contribution of the LL motif in the biosynthesis, transport, and function of the F protein, we constructed and rescued a NDV mutant strain, rSG10*-F/L537A, with an L537A mutation using a reverse genetic system. Subsequently, we compared the differences in the syncytium formation ability, pathogenicity, and replication levels of wild-type rSG10* and the mutated strain. Results Compared with rSG10*, rSG10*-F/L537A had attenuated syncytial formation and pathogenicity, caused by a viral budding defect. Further studies showed that the LL-motif mutation did not affect the replication, transcription, or translation of the virus genome but affected the expression of the F protein at the cell surface. Conclusions We concluded that the LL motif in the NDV F CT affected the regulation of F protein expression at the cell surface, thus modulating the viral fusion ability and pathogenic phenotype.

Abstract Gallbladder carcinoma (GBC), an aggressive malignant tumor of the biliary system, is characterized by high cellular heterogeneity and poor prognosis. Fewer data have been reported in GBC than other common cancer types. Multi-omics data will contribute to the understanding of the molecular mechanisms of cancer, cancer diagnosis and prognosis. Herein, to provide better understanding of the molecular events in GBC pathogenesis, we developed GBCdb ( http://tmliang.cn/gbc/ ), a user-friendly interface for the query and browsing of GBC-associated genes and RNA interaction networks using published multi-omics data, which also included experimentally supported data from different molecular levels. GBCdb will help to elucidate the potential biological roles of different RNAs and allow for the exploration of RNA interactions in GBC. These resources will provide an opportunity for unraveling the potential molecular features of Gallbladder carcinoma.

ABSTRACT Anthracnose diseases caused by Colletotrichum species are among the most common fungal diseases. These symptoms typically manifest as dark, sunken lesions on leaves, stems, and fruit. In China, mango anthracnose seriously affects fruit yield and quality. Genome sequencing of several species shows the presence of mini-chromosomes. These are thought to contribute to virulence, but their formation and activity remain to be fully elucidated. Here, we assembled 17 Colletotrichum genomes (16 isolated from mango plus one from persimmon) through PacBio long-read sequencing. Half of the assembled scaffolds had telomeric repeats at both ends indicating full-length chromosomes. Based on comparative genomics analysis at interspecies and intraspecies levels, we identified extensive chromosomal rearrangements events. We analyzed mini-chromosomes of Colletotrichum spp. and found large variation among close relatives. In C. fructicola, homology between core chromosomes and mini-chromosomes suggested that some mini-chromosomes were generated by recombination of core chromosomes. In C. musae GZ23-3, we found 26 horizontally transferred genes arranged in clusters on mini-chromosomes. In C. asianum FJ11-1, several potential pathogenesis-related genes on mini-chromosomes were upregulated, especially in strains with highly pathogenic phenotypes. Mutants of these upregulated genes showed obvious defects in virulence. Our findings provide insights into the evolution and potential relationships to virulence associated with mini-chromosomes. IMPORTANCE Colletotrichum is a cosmopolitan fungal genus that seriously affects fruit yield and quality of many plant species. Mini-chromosomes have been found to be related to virulence in Colletotrichum. Further examination of mini-chromosomes can help us elucidate some pathogenic mechanisms of Colletotrichum. In this study, we generated novel assemblies of several Colletotrichum strains. Comparative genomic analyses within and between Colletotrichum species were conducted. We then identified mini-chromosomes in our sequenced strains systematically. The characteristics and generation of mini-chromosomes were investigated. Transcriptome analysis and gene knockout revealed pathogenesis-related genes located on mini-chromosomes of C. asianum FJ11-1. This study represents the most comprehensive investigation of chromosome evolution and potential pathogenicity of mini-chromosomes in the Colletotrichum genus.

ABSTRACT Lentinula edodes is one of the most widely cultivated edible mushrooms in the world. When cultivated in sawdust, the surface mycelium of L. edodes needs a long postripening stage wherein it forms a brown film (BF) by secreting and accumulating pigments. BF formation is critical for the high quality and yield of fruiting bodies. Protein lysine acetylation (KAC) is an important post-translational modification that regulates growth and development. Previous studies have shown that deacetylase levels are significantly increased during BF formation in the postripening stage of L. edodes. The aim of this study was to assess the role of protein acetylation during BF formation. To this end, we compared the acetylome of L. edodes mycelia before and after BF formation using anti-acetyl antibody-based label-free quantitative proteomics. We identified 5,613 acetylation sites in 1,991 proteins, and quantitative information was available for 4,848 of these sites in 1,815 proteins. Comparative acetylome analysis showed that the modification of 699 sites increased and that of 562 sites decreased during BF formation. Bioinformatics analysis of the differentially acetylated proteins showed significant enrichment in the tricarboxylic acid (TCA) cycle and proteasome pathways. Furthermore, functional assays showed that BF formation is associated with significant changes in the activities of proteasome, citrate synthase, and isocitrate dehydrogenase. Consistent with this hypothesis, the lysine deacetylase inhibitor trichostatin (TSA) delayed autophagy and BF formation in L. edodes. Taken together, KAC and autophagy play important roles in the mycelial BF formation and postripening stage of L. edodes. IMPORTANCE Mycelial BF formation and postripening of L. edodes affects the quality and quantity of its edible fruiting bodies. In this study, we explored the role of protein KAC in this biological process, with the aim of optimizing the cultivation and yield of L. edodes.

The single-cell triboelectric nanogenerator (TENG) often produces insufficient energy, leading to the use of a multicellular TENG structure. This work experimented with and simulated a dual-cell TENG with various configurations in parallel and series arrangements. The working principle of charge generation during each phase of a contact–separation cycle was explained through the analysis and comparison of five electrical configurations of a dual-cell TENG. Our observations indicate that measuring the output charge of a TENG provides a more reliable performance comparison. Finally, multicellular TENG with four cells arranged in an X-shape (X-TENG), self-supporting structure is fabricated and further experimented with, validating our conjectures derived from a dual-cell TENG.

Accurately quantifying the spatiotemporal nutrient loads from nonpoint sources (NPSs) is still difficult due to the high spatial heterogeneity and temporal variability in rainfall-runoff processes. The export coefficient model (ECM) is a widely used tool to estimate the NPS nutrient load on an annual time scale, but it hardly considers the spatiotemporal distribution of the export coefficient. To address this issue, the ECM is improved by incorporating a grid-based rainfall-runoff module together with land-use data and analysing the spatiotemporal characteristics of the NPS nutrient loads in Yancheng city located in eastern China. Moreover, the effect of NPS nutrient loads on river water quality is assessed using a conceptual model. The results indicate that the improved ECM can simulate reasonable spatiotemporal variation in NPS nutrient loads, and the relative error in the total phosphorus (TP) load is less than 17.14 %. Precipitation has a considerable influence on both the total nitrogen (TN) and TP loads, which varied greatly from 1990 to 2018, with average annual values of 30.57 × 103 tons and 2.44 × 103 tons, respectively. The spatial distributions of the TN and TP loads were greatly affected by land-use patterns, and bodies of water and rural land had relatively high load intensities. Both TN and TP concentrations in the river exceeded the national standard, but TP was significantly reduced in the past 10 years, while TN was still at high level. This study provides a reference for reducing the river pollution in Yancheng city, especially by identifying pollution sources requiring priority control.

Plant parasitic nematodes (PPNs) are highly destructive and difficult to control, while conventional chemical nematicides are highly toxic and cause serious environmental pollution. Additionally, resistance to existing pesticides is becoming increasingly common. Biological control is the most promising method for the controlling of PPNs. Therefore, the screening of nematicidal microbial resources and the identification of natural products are of great significance and urgency for the environmentally friendly control of PPNs. In this study, the DT10 strain was isolated from wild moss samples and identified as Streptomyces sp. by morphological and molecular analysis. Using Caenorhabditis elegans as a model, the extract of DT10 was screened for nematicidal activity, which elicited 100% lethality. The active compound was isolated from the extracts of strain DT10 using silica gel column chromatography and semipreparative high-performance liquid chromatography (HPLC). The compound was identified as spectinabilin (chemical formula C28H31O6N) using liquid chromatography mass spectrometry (LC-MS) and nuclear magnetic resonance (NMR). Spectinabilin exhibited a good nematicidal activity on C. elegans L1 worms, with a half-maximal inhibitory concentration (IC50) of 2.948 μg/mL at 24 h. The locomotive ability of C. elegans L4 worms was significantly reduced when treated with 40 μg/mL spectinabilin. Further analysis of spectinabilin against known nematicidal drug target genes in C. elegans showed that it acts via target(s) different from those of some currently used nematicidal drugs such as avermectin and phosphine thiazole. This is the first report on the nematicidal activity of spectinabilin on C. elegans and the southern root-knot nematode Meloidogyne incognita. These findings may pave the way for further research and application of spectinabilin as a potential biological nematicide.

Natural ecosystems of the West Liao River basin (WLRB) in northeast China have been damaged by both natural and human factors from the 1990s. Since 2000, China’s Grain-for-Green Program (GFGP) has been widely adopted with the aim of improving ecosystem services. An accurate evaluation of the eco-hydrological effects for policy implementation is essential to provide references for further restoration of ecosystem services. This study quantified and characterized the ecosystem services and their trade-offs/synergies using models and statistical methods in the WLRB from 1990 to 2020. Moreover, the impact of key drivers on ecosystem services was evaluated by the difference-in-differences model. Among them, the study mainly investigated how GFGP affects ecosystem services. The results confirmed that the water yield, carbon sequestration, habitat quality, and total ecosystem service of the WLRB decreased in the pre-GFGP period (1990–2000). However, this tendency was reversed in the regions where the GFGP was implemented during the period of 2001–2020. Furthermore, a synergistic relationship was shown among carbon sequestration, soil conservation, and habitat quality. Additionally, there were tradeoffs between water yield and the other three ecosystem services, especially in mountain areas. The GFGP could restore carbon sequestration, habitat quality, and total ecosystem services by 1.3%, 2.1%, and 0.6%, respectively. Nevertheless, GFCP may enlarge the tradeoff and imbalance between water yield and habitat quality. Results highlight the need for the governance of ecosystem protection and suggest natural restoration in the mountain area for maintaining water yield and helping ecosystem restoration. Timely adjustment of the policy implementation areas is the key to improving and balancing multiple ecosystem services in the future.

Abstract Flexible, compact, lightweight and sustainable power sources are indispensable for modern wearable and personal electronics and small-unmanned aerial vehicles (UAVs). Hierarchical honeycomb has the unique merits of compact mesostructures, excellent energy absorption properties and considerable weight to strength ratios. Herein, a honeycomb-inspired triboelectric nanogenerator (h-TENG) is proposed for biomechanical and UAV morphing wing energy harvesting based on contact triboelectrification wavy surface of cellular honeycomb structure. The wavy surface comprises a multilayered thin film structure (combining polyethylene terephthalate, silver nanowires and fluorinated ethylene propylene) fabricated through high-temperature thermoplastic molding and wafer-level bonding process. With superior synchronization of large amounts of energy generation units with honeycomb cells, the manufactured h-TENG prototype produces the maximum instantaneous open-circuit voltage, short-circuit current and output power of 1207 V, 68.5 μA and 12.4 mW, respectively, corresponding to a remarkable peak power density of 0.275 mW cm−3 (or 2.48 mW g−1) under hand pressing excitations. Attributed to the excellent elastic property of self-rebounding honeycomb structure, the flexible and transparent h-TENG can be easily pressed, bent and integrated into shoes for real-time insole plantar pressure mapping. The lightweight and compact h-TENG is further installed into a morphing wing of small UAVs for efficiently converting the flapping energy of ailerons into electricity for the first time. This research demonstrates this new conceptualizing single h-TENG device's versatility and viability for broad-range real-world application scenarios.

The duration of earthquake ground motion is acted as one of the three elements indicating of ground motion. Its influence on earthquake damage is gradually recognized by experts and scholars all over the world. However, in defining of duration, there is currently no uniform and clear concept. In a generalized definition, the paper gives the definition about the total duration of earthquake ground motion and the duration of strong ground motion. The researchers select more suitable duration according to the characteristics of their respective fields. The duration can be roughly divided into two broad categories: absolute duration and relative duration. The absolute duration is bracket duration based on absolute acceleration threshold, while the relative duration of energy control reflects the trend of intensity or energy change in the seismic process. There are many definitions of the duration, yet this paper simply reviews five definitions of duration of earthquake ground motion. The characteristics of the five kinds of duration definitions are analyzed meticulously. Moreover, the paper introduces the research results in prediction model of duration of earthquake ground motion. At last, the paper puts forward some insights of current application in duration of earthquake ground motion.

Cholangiocarcinomas (CCAs) are tumors that arise from the cholangiocytes. Although some genes have been shown with important roles in pathological process, interactions or cross-talks among different RNAs are important to understand the detailed molecular mechanisms in cancer development, especially discussing cross-talks among isomiRs and other RNAs. Herein, to characterize crucial genes in CCA, the protein expression profile was performed to survey potential crucial mRNAs and related non-coding RNAs (ncRNAs) in mRNA-ncRNA network, mainly including miRNAs/isomiRs and lncRNAs. Deregulated mRNAs were firstly obtained if consistent expression patterns were found at protein and mRNA levels, and related miRNAs/isomiRs were screened according to regulatory relationships. Diverse isomiRs from a given miRNA locus also contributed to interactions between the small RNAs and target mRNAs, and miRNAs were further used to survey related lncRNAs to expand the interactions. Thus, several groups of RNAs were constructed as candidate competitive endogenous RNA (ceRNA) networks. Finally, we found that RAB11FIP1:miR-101-3p:MIR3142HG may be a potential ceRNA network, and the interactions among them may be more complex due to variety of isomiRs. Simultaneously, RAB11FIP1 and miR-194-5p were also detected other related lncRNAs (FBXL19-AS1, SNHG1 and PVT1) that may be crucial in coding-non-coding RNA regulatory network. Our results show that diverse isomiRs with sequence and expression heterogeneities contribute to ceRNA regulatory network that may have crucial roles in CCA, which will expand our understanding of interactions among diverse RNAs and their contributions in cancer development.

A quantitative analysis of the precipitate effects of metal ions (Al3+, Fe2+, Cu2+, Zn2+) by bovine serum albumin (BSA) on two condensed tannins (CT) from sorghum and plum was presented in this study. The results showed that adding metal ions enhanced the precipitation of proteins by CT, depending on the type and concentration of the metal ions used in the reaction system. The presence of metal ions and precipitation results on the CT–protein complex showed that Al3+ and Fe2+ had a higher binding ability with CT and a weaker influence on the precipitation of the CT–protein complex than Cu2+ and Zn2+. However, when the initial reaction solution contained excessive amounts of BSA, the extra addition of metal ions had no significant effect on the amount of BSA precipitation. Reversely, adding Cu2+ or Zn2+ into the reaction solution increased the amount of precipitated BSA when the amount of CT was excessive. In addition, the amounts of CT from plum, rather than sorghum, generated more protein precipitate in the presence of Cu2+ or Zn2+, which may be due to the different binding modes between the metal ion and the CT–BSA complex. This study also proposed a model of the interaction between the metal ion and the CT–protein precipitate.

Lentinula edodes (Berk.) Pegler, the shiitake mushroom, is one of the most important mushrooms in the global mushroom industry. Although mycelium post ripeness and brown film (BF) formation are crucial for fruiting body initiation, the underlying molecular mechanisms of BF formation are largely unknown. In this study, proteomic quantification (relative and absolute) and metabolomic profiling of L. edodes were performed using isobaric tags and gas chromatography-mass spectroscopy, respectively. A total of 2,474 proteins were identified, which included 239 differentially expressed proteins. Notably, several proteins associated with autophagy were upregulated, including RPD3, TOR1, VAC8, VPS1, and VPS27. Transmission electron microscopy also indicated that autophagy occurred in post ripeness and BF formation. In time-dependent analysis of the metabolome, metabolites associated with oxidative stress and autophagy changed significantly, including mannitol, trehalose, myo-inositol, glucose, leucine, valine, glutamine, and 4-aminobutyric acid. Thus, oxidative stress and autophagy were important processes in post ripeness and BF formation in L. edodes, and new insights were gained into molecular mechanisms at proteome and metabolome levels.

Human papillomavirus (HPV) is a causal agent for most cervical cancers. The physical status of the HPV genome in these cancers could be episomal, integrated, or both. HPV integration could serve as a biomarker for clinical diagnosis, treatment, and prognosis. Although whole-genome sequencing by next-generation sequencing (NGS) technologies, such as the Illumina sequencing platform, have been used for detecting integrated HPV genome in cervical cancer, it faces challenges of analyzing long repeats and translocated sequences. In contrast, Oxford nanopore sequencing technology can generate ultra-long reads, which could be a very useful tool for determining HPV genome sequence and its physical status in cervical cancer. As a proof of concept, in this study, we completed whole genome sequencing from a cervical cancer tissue and a CaSki cell line with Oxford Nanopore Technologies. From the cervical cancer tissue, a 7,894 bp-long HPV35 genomic sequence was assembled from 678 reads at 97-fold coverage of HPV genome, sharing 99.96% identity with the HPV sequence obtained by Sanger sequencing. A 7904 bp-long HPV16 genomic sequence was assembled from data generated from the CaSki cell line at 3857-fold coverage, sharing 99.99% identity with the reference genome (NCBI: U89348). Intriguingly, long reads generated by nanopore sequencing directly revealed chimeric cellular–viral sequences and concatemeric genomic sequences, leading to the discovery of 448 unique integration breakpoints in the CaSki cell line and 60 breakpoints in the cervical cancer sample. Taken together, nanopore sequencing is a unique tool to identify HPV sequences and would shed light on the physical status of HPV genome in its associated cancers.

The 2020 Jiashi Mw 6.0 earthquake occurred at the Kepingtage fold-and-thrust belt in the South Tianshan front, Northwestern China. The ground shaking caused extensive co-seismic deformation of the Xiker dam in the meizoseismal area. We obtained strata distribution characteristics of the dam foundation through drilling. Using laboratory and in situ tests, the particle size distribution, standard penetration, and shear wave velocity of each layer were obtained. Along with peak ground acceleration, we evaluated the potential of sand liquefaction in various layers and proposed a relationship between dam fissures and sand liquefaction. Our results suggest that sand liquefaction occurred in the silty sand layer 0–3 m beneath the dam foundation. Sand liquefaction occurs behind the dam, resulting in uneven settlement of the dam foundation, making the horizontal deformation of the backslope of the dam significantly larger than the foreslope of the dam. Using numerical simulations, we found that sand liquefaction behind the dam can cause different horizontal deformation vectors (maximum deformation is ∼7.45 cm) in the dam foreslope and backslope, which cause the dam to rotate in the downstream direction. Large fissures also formed on the dam crest.

Hypsizygus marmoreus is a representative edible mushroom with low-temperature fruiting after a long postripening (LFLP). Clarifying the mechanism of LFLP and applying a rigorous low-temperature-limited process will optimize the mushroom cultivation process. This study performed an integrative multi-omics analysis of the molecular mechanism of LFLP in combination with genetic, physiological, and cultivation confirmation. The results showed that the amino acid content was increased during LFLP, mainly arginine. pH analysis showed acidification in the postripening stage and alkalization in the substrates of the reproductive growth stage. An enzyme activity test confirmed the increased enzyme activity of arginase and citrate synthase in the postripening stage. Weighted gene coexpression network analysis of the transcriptome and metabolomics indicated that pH variation is correlated mainly with changes in citrate and arginine. Multi-omics reveals a straightforward way of providing enriched materials for amino acid biosynthesis, namely, synergistically elevating citric acid and arginine through enhanced activity of the arginine synthesis branch pathway in the citrate cycle. Our study confirmed that GCN2 mediated metabolic adaptation by enhancing protein translation, highlighting its regulatory role during LFLP. Exogenously added citric acid and arginine shortened the postripening period by 10 days and increased the fruiting body yield by 10.2~15.5%. This research sheds light on the molecular mechanism of LFLP in H. marmoreus and highlights the promising application of nutrient accumulation in high-efficiency cultivation.

Prostate adenocarcinoma (PRAD), also named prostate cancer, the most common visceral malignancy, is diagnosed in male individuals. Herein, in order to obtain immune-based subtypes, we performed an integrative analysis to characterize molecular subtypes based on immune-related genes, and further discuss the potential features and differences between identified subtypes. Simultaneously, we also construct an immune-based risk model to assess cancer prognosis. Our findings showed that the two subtypes, C1 and C2, could be characterized, and the two subtypes showed different characteristics that could clearly describe the heterogeneity of immune microenvironments. The C2 subtype presented a better survival rate than that in the C1 subtype. Further, we constructed an immune-based prognostic model based on four screened abnormally expressed genes, and they were selected as predictors of the robust prognostic model (AUC = 0.968). Our studies provide reference for characterization of molecular subtypes and immunotherapeutic agents against prostate cancer, and the developed robust and useful immune-based prognostic model can contribute to cancer prognosis and provide reference for the individualized treatment plan and health resource utilization. These findings further promote the development and application of precision medicine in prostate cancer.

Robot-assisted rehabilitation devices can provide intensive and precise task-based training that differs from clinician-facilitated manual therapy. However, industrial robots are still rarely used in rehabilitation, especially in bilateral exercises. The main purpose of this research is to develop and evaluate the functionality of a bilateral upper-limb rehabilitation system based on two modern industrial robots. A `patient-cooperative' control strategy is developed based on an adaptive admittance controller, which can take into account patients' voluntary efforts. Three bilateral training protocols (passive, active, and self) are also proposed based on the system and the control strategy. Experimental results from 10 healthy subjects show that the proposed system can provide reliable bilateral exercises: the mean RMS values for the master error and the master-slave error are all less than 1.00 mm and 1.15 mm respectively, and the mean max absolute values for the master error and the master-slave error are no greater than 6.11 mm and 6.73 mm respectively. Meanwhile, the experimental results also confirm that the recalculated desired trajectory can present the voluntary efforts of subjects. These experimental findings suggest that industrial robots can be used in bilateral rehabilitation training, and also highlight the potential applications of the proposed system in further clinical practices.

The type of polymeric condensed tannins from plum fruit (Prunus salicina) (PCT), the degree of polymerization and the distribution of polymers were characterized by MALDI-TOF MS and NMR spectroscopy. The metal-binding capacity of PCT with five metal ions (Cu2+, Zn2+, Al3+, Fe2+, and Fe3+) was characterized by a fluorescence quenching method. The results demonstrated the following: epicatechin was the basic unit occurring in PCT, and A-type and B-type linkages were the most common between the structural units of the polymers. The PCT have a strong antioxidant activity, which is comparable with that of the synthetic antioxidant BHA. The quenching mechanism of the PCT’s fluorescence intensity by Zn2+, Cu2+, and Al3+ was different from that of Fe3+ and Fe2+. Fe3+, Al3+ and Fe2+ had much higher affinities for the PCT than Zn2+ and Cu2+. A simple UV-Vis spectra method was developed to determine the protein-precipitating capacity of tannins. Bovine serum albumin (BSA) was effectively precipitated by tannins isolated from plum fruits, Chinese gallnut, sorghum grain, and Platycarya strobilacea at pH values between 4.5 and 5.0. A statistically significant linear relationship (p < 0.0001 or p < 0.0003) existed between the amount of tannin–protein complex formed and the amount of tannins added to the reaction mixture. The slopes of these lines indicated the protein-precipitating capacity of tannins.

In coastal areas of southeastern China, multiple flood drivers such as river flow, precipitation and coastal water level can lead to compound flooding which is often much greater than flooding simulated by one flood driver in isolation. Bivariate probability distributions accounting for compound flooding from river discharge and sea level were constructed based on MvCAT (Multivariate Copula Analysis Toolbox) combined with goodness of fit tests in 15 coastal-estuarine regions of Southeastern China. Flood typing-based bivariate probability distributions considering multiple flood-generating mechanisms were also built. Our results indicated that the performance of flood typing-based bivariate distribution was not significantly better than the bivariate probability distribution in coastal-estuarine regions based on the Mann–Whitney U test; the compounding effects of river discharge and sea level had limited impact on bivariate return periods, but had greater impact on coastal flooding risk in terms of design values. Ignoring compounding effects of river discharge and sea level leads to significant underestimation of design values. The results suggest that the compounding effect of river discharge and sea level should be considered when calculating design values in coastal flood risk assessment.

MicroRNAs (miRNAs) are known to be critical regulators in cancer progression. MiR-451a is reported to be involved in the progression of many different forms of cancers, including osteosarcoma, colorectal cancer, and breast carcinoma. In this study, we illuminated the possible roles of miR-451a in the development of papillary thyroid carcinoma (PTC) cells in vitro and in vivo. MiR-451a was markedly down-expressed in PTC sample compared with paratumor tissue. Upregulation of miR-451a repressed PTC cells proliferation, migration ability and inhibited the invasiveness of PTC cells in vitro. Additional, miR-451a suppressed PTC cells growth and the lung metastasis of PTC cells in vivo, whereas downregulation of miR-451a caused opposite outcomes. Importantly, miR-451a inversely modulated the expression of Zinc Finger E-Box Binding Homeobox 1 (ZEB1) by directly binding to the 3′ untranslated region (UTR) of ZEB1 in PTC cells. The level of ZEB1 was negatively associated with miR-451a level in PTC tissues, and ZEB1 silencing mimicked the suppressive impacts of miR-451a on the proliferation, mobility, and invasive phenotypes of PTC cells. ZEB1 overexpression abrogated the inhibitory impacts of miR-451a on PTC cells. Together, this study revealed that miR-451a restrained the growth and metastatic phenotypes of PTC cells through targeting ZEB1.

Sugarcane yellow leaf virus (SCYLV), the causal agent of yellow leaf disease, naturally infects at least three plant species in Florida: sugarcane (Saccharum spp.), the weed Columbus grass (Sorghum almum) and cultivated sorghum (S. bicolor). All three hosts are also colonized by the sugarcane aphid (Melanaphis sacchari), the main vector of SCYLV worldwide. To understand the high incidence of SCYLV observed in sugarcane commercial fields and in germplasm collections, we investigated the transmission efficiency of SCYLV from sugarcane and Columbus grass to sugarcane using the sugarcane aphid and a spider mite (Oligonychus grypus) that also tested positive for SCYLV in Florida. Healthy and SCYLV-infected leaf pieces of sugarcane and Columbus grass carrying viruliferous aphids or spider mites were transferred to virus-free plants of the yellow leaf susceptible sugarcane cultivar CP96-1252. Three- and 6-months post inoculation, the 108 aphid-inoculated plants of Columbus grass and the 90 mite-inoculated plants of sugarcane tested negative for SCYLV by tissue blot immunoassay (TBIA) or reverse transcription polymerase chain reaction (RT-PCR). Similar results were obtained for 162 aphid-inoculated plants of sugarcane, except for two plants that tested positive for SCYLV by TBIA and RT-PCR. In two field experiments planted with SCYLV-free and virus-infected sugarcane (cultivar CP96-1252), only 18-28% of healthy plants became infected during a 24- to 28-month period. SCYLV prevalence in these field experiments did not differ between aphicide treated and untreated plots. Incidence of M. sacchari haplotypes in the Everglades agricultural area also indicated that the predominant haplotype that is currently colonizing sugarcane was not a vector of SCYLV in Florida. Lack of virus transmission by the spider mite suggested that this arthropod only acquired the virus when feeding on infected plants but was unable to transmit SCYLV. The current vector of SCYLV in Florida remains to be identified.

The straw-rotting edible fungus Volvariella volvacea is a widely cultivated edible fungus across China and Southeast Asian countries. Three complete mitochondrial genomes of V. volvacea from China, Thailand, and India were determined using the next-generation sequencing technology. The genome sizes of the three strains (China, Thailand, and India) were 62,541 bp, 64,531 bp, and 65,668 bp with GC contents of 38.46%, 38.56%, and 38.52%, respectively. All the genomes encoded 14 conserved protein-coding genes, the small ribosomal RNA subunits (rns), large ribosomal RNA subunits (rnl), and 23 tRNAs were located on the same strand. In the putative protein-coding genes, four introns were distributed in cox1 in the genomes of V23-1 and V8. 5 introns (four introns invaded into cox1and one intron invaded into cob) were detected in Tai8. The phylogenetic analysis confirmed that V. volvacea was a number of Agaricales. This mitochondrial genome may open new avenues for understanding the phylogeny and evolution of Pluteaceae and Agaricales.

The Tibetan Plateau (TP) is sensitive to climate change in the land–atmosphere coupling mechanism due to its complex topographic features and unique geographic location. This study explored the teleconnection between pre-summer thawing of frozen soil over the TP and summer precipitation in East Asia in the Meiyu–Baiu rainy season (June, July) from 1981 to 2019 using maximum covariance analysis (MCA). The precipitation fields forced by thawing of frozen soil were calculated by the coupled manifold technique. The variations in East Asian precipitation are significantly impacted by thawing of frozen soil over the TP, with a variance explained ratio in the surrounding middle and lower reaches of Yangtze River (MLYR) of around 10%–20%. The MCA analysis also revealed that the thickness of pre-summer frozen soil thawing had a positive relationship with summer precipitation in the MLYR and southern Japan (fraction = 0.59, correlation ≈ 0.99). To find out the possible mechanism, composite analyses were conducted on atmospheric and surface components with reanalysis products. The analysis results suggested that more (less) frozen soil thawing would increase (decrease) the sensible heat and land surface temperature with enhanced (weakened) surface diabatic heating over the TP. Then, the positive (negative) surface diabatic heating would result in an enhanced (weakened) South Asia High extending eastward followed by stronger (weaker) upper troposphere (200 hPa) westerlies, as well as the West North Pacific Subtropical High extending westward. As a result, the northeastward movement of the integrated vapor transport intensified (weakened) with the westward extension (eastward retreat) of the Meiyu–Baiu rain belt, leading to more (less) precipitation in the MLYR and southern Japan. It is thus suggested that pre-summer thawing of frozen soil over the TP could play a vital role in regulating East Asian summer precipitation and movement of the Meiyu–Baiu rain belt.

Water pollution poses threats to urban environments and subsequently impacts the ecological health and sustainable development of urban areas. Identifying the spatiotemporal variation in non-point sources (NPS) pollution is a prerequisite for improving water quality. This paper aimed to assess the NPS pollution load and then recognized the spatiotemporal characteristics of the pollution sources in a typical urbanized area. A combination model based on land use type was used to simulate the NPS pollution load. The results showed the following: (1) ponds and farmlands had higher pollution production intensities than other land use types, but the intensity and magnitude of pollution emissions were generally greater in urban areas; (2) monthly and annual total nitrogen (TN) and total phosphorus (TP) emissions had the same pattern as rainfall, and TN and TP emissions accounted for 56.2% and 58.0%, respectively, of the total in summer; (3) TN pollution was more serious than TP pollution in the study area, especially in farmlands; (4) urban runoff (UR) and livestock and poultry breeding (LPB) were the main sources of NPS, TN and TP emissions in the study area. If these NPS pollutants cannot be removed from this area, a large amount of freshwater is needed to dilute the current rivers to meet the requirement of the fourth category of China national environmental quality standards for surface water. This problem is serious in the control of polluted rivers in many cities throughout China.

This paper investigates a modified acoustic metamaterial system with local resonators coupled through linear springs. The proposed acoustic metamaterial system can provide three band gaps for broadband vibration suppression. First, the band structure of the modified acoustic metamaterial is calculated by using Bloch’s theorem under the assumption of infinite lattice. The existence of three band gaps is confirmed in the band structure. Effects of mass and spring parameters on the band gap behaviour of the modified metamaterial are investigated through a dimensionless parametric study. Based on the parametric study, optimal dimensionless parameters are proposed to achieve maximal total band gap width in the low frequency range. Subsequently, a more realistic finite lattice model is established. The transmittances of the conventional and modified metamaterial systems are compared. The three band gaps predicted from transmittances and broadband vibration suppression behaviour are consistent with the predictions from infinite lattice model using Bloch’s theorem. Finally, the time-domain responses are simulated and the superiority of the modified acoustic metamaterial over the conventional one is demonstrated.

Alcoholic liver disease (ALD) is a type of chronic liver disease caused by long-term heavy ethanol consumption. Danshen is one of the most commonly used substances in traditional Chinese medicine and has been widely used for the treatment of various diseases, and most frequently, the ALD. The current study aims to determine the potential beneficial effect of Danshen administration on ALD and to clarify the underlying molecular mechanisms. Danshen administration improved liver pathologies of ALD, attenuated alcohol-induced increment of hepatic 4-Hydroxynonenal (4-HNE) formation, and prevented hepatic Peroxisome proliferators activated receptor alpha (PPARα) suppression in response to chronic alcohol consumption. Cell culture studies revealed that both hepatoprotective effect and increased intracellular 4-HNE clearance instigated by Danshen supplementation are PPARα-dependent. In conclusion, Danshen administration can protect against ALD via inducing PPARα activation and subsequent 4-HNE degradation.

In this paper, the behavior of a piezoelectric stack generator subject to a direct mechanical force, such as that experienced by pedestrian loads, is characterized and modelled. A lumped element model is developed that describes the electrical output of piezoelectric stack generators for quasi-static, off-resonance and random input conditions which are typical of underfloor or supporting structure energy harvesters. This research demonstrates the ability for accurate predictions of output voltage and current based on the material properties and geometry of the piezoelectric stack. The behavior of real-world random loading patterns is incorporated in the model, pairing mechanical input with interfacing circuitry requirements in order to maximize energy transfer. The developed model facilitates time-efficient design by reducing the simulation time to the order of seconds.

The complete mitochondrial genome of the widely cultivated edible fungus Lentinula edodes was determined using the next-generation sequencing technology. The circular molecule is 116,819 bp in length with a GC content of 30.75%. Conserved genes including 13 putative protein-coding genes and 24 tRNAs were located on the same strand. We detected 14 introns invading 4 genes, including cob, cox1, nad1, and nad5. The phylogenetic analysis confirmed that L. edodes was a number of Agaricales. This mitochondrial genome may open new avenues for understanding the phylogeny and evolution of Omphalotaceae and Agaricales.

Distorted sex ratios occur in hematologic disorders. For example, chronic lymphocytic leukemia (CLL) displays disproportionate sex ratios with a large male excess. However, the underlying genetics for these disparities are poorly understood, and gender differences for specific cytogenetic abnormalities have not been carefully investigated. We sought to provide an initial characterization of gender representation in genetic abnormalities in CLL by using fluorescence in situ hybridization (FISH). We confirm the well known skewed male-to-female (M/F sex ratio) of ~1.5 in our CLL study population, but also determine the genotypic M/F sex ratio values corresponding to specific FISH DNA probes. Genetic changes in CLL detectable by four FISH probes were statistically compared with respect to gender. Initial FISH evaluations of 4698 CLL patients were retrospectively examined and new findings of the genotypic M/F sex ratios for these probes are reported. This study represents the largest CLL survey conducted in the United States using FISH probes. The CLL database demonstrated that FISH abnormalities (trisomy 12, 13q14.3 deletion and 17p13.1 deletion) probes had skewed M/F ratios of ~1.5. Also, by statistical analysis it was shown that ATM gene loss (11q22.3q23.1 deletion) solely or with other abnormalities was considerably higher in males with an M/F ratio of 2.5 and significantly different from M/F ratios of 1.0 or 1.5. We hypothesize that interactions involving these autosomal abnormalities (trisomy 12, and deletions of 11q22.3, 13q14.3, and 17p13.1), and the sex chromosomes may provide the genetic basis for the altered phenotypic M/F ratio in CLL.

OBJECTIVE: Gray matter loss in the limbic structures was found in recent onset post traumatic stress disorder (PTSD) patients. In the present study, we measured regional gray matter volume in trauma survivors to verify the hypothesis that stress may cause different regional gray matter loss in trauma survivors with and without recent onset PTSD. METHOD: High resolution T1-weighted magnetic resonance imaging (MRI) were obtained from coal mine flood disaster survivors with (n = 10) and without (n = 10) recent onset PTSD and 20 no trauma exposed normal controls. The voxel-based morphometry (VBM) method was used to measure the regional gray matter volume in three groups, the correlations of PTSD symptom severities with the gray matter volume in trauma survivors were also analyzed by multiple regression. RESULTS: Compared with normal controls, recent onset PTSD patients had smaller gray matter volume in left dorsal anterior cingulate cortex (ACC), and non PTSD subjects had smaller gray matter volume in the right pulvinar and left pallidum. The gray matter volume of the trauma survivors correlated negatively with CAPS scores in the right frontal lobe, left anterior and middle cingulate cortex, bilateral cuneus cortex, right middle occipital lobe, while in the recent onset PTSD, the gray matter volume correlated negatively with CAPS scores in bilateral superior medial frontal lobe and right ACC. CONCLUSION: The present study identified gray matter loss in different regions in recent onset PTSD and non PTSD after a single prolonged trauma exposure. The gray matter volume of left dorsal ACC associated with the development of PTSD, while the gray matter volume of right pulvinar and left pallidum associated with the response to the severe stress. The atrophy of the frontal and limbic cortices predicts the symptom severities of the PTSD.

Obesity is associated with inflammation of visceral adipose tissues, which increases the risk for insulin resistance. Animal models suggest that T-lymphocyte infiltration is an important early step, although it is unclear why these cells are attracted. We have recently demonstrated that dietary triglycerides, major components of high fat diets, promote intestinal absorption of a protein antigen (ovalbumin, "OVA"). The antigen was partly transported on chylomicrons, which are prominently cleared in adipose tissues. We hypothesized that intestinally absorbed gut antigens may cause T-lymphocyte associated inflammation in adipose tissue.Triglyceride absorption promoted intestinal absorption of OVA into adipose tissue, in a chylomicron-dependent manner. Absorption tended to be higher in mesenteric than subcutaneous adipose tissue, and was lowest in gonadal tissue. OVA immunoreactivity was detected in stromal vascular cells, including endothelial cells. In OVA-sensitized mice, OVA feeding caused marked accumulation of CD3+ and osteopontin+ cells in mesenteric adipose tissue. The accumulating T-lymphocytes were mainly CD4+. As expected, high-fat (60% kCal) diets promoted mesenteric adipose tissue inflammation compared to low-fat diets (10% Kcal), as reflected by increased expression of osteopontin and interferon-gamma. Immune responses to dietary OVA further increased diet-induced osteopontin and interferon-gamma expression in mesenteric adipose. Inflammatory gene expression in subcutaneous tissue did not respond significantly to OVA or dietary fat content. Lastly, whereas OVA responses did not significantly affect bodyweight or adiposity, they significantly impaired glucose tolerance.Our results suggest that loss or lack of immunological tolerance to intestinally absorbed T-lymphocyte antigens can contribute to mesenteric adipose tissue inflammation and defective glucose metabolism during high-fat dieting.
