2. Diversity of Actinomycetes Isolated from Peat Soil of Undistrubed Forest and Pineapple Plantation in Sessang, Sarawak

Abstract

Peatland plays an important role not just as a carbon store but also in facilitating the flux of greenhouse gasses into the atmosphere. Apart from that, peatland is also home to a diverse population of microorganisms such as bacteria, fungi, and actinomycetes. Actinomycetes were known to be one of the most ubiquitous microbes that can be found in most of the soil types including peat soil. In this study, seventy isolates of actinomycetes were isolated from the peat soil using the soil dilution method. The 70 isolates of actinomycetes were later screened for their ability to produce secondary metabolites and antimicrobial activities using the agar diffusion method before the selected potential isolates were identified by targeting their 16S rRNA region. The results obtained showed 34.3% produce cellulase followed by, 12.8, 31.7, 80.0, and 51.4% for mannanase, xylanase, lipase, and protease respectively. The percentage of actinomycetes producing antimicrobial activity was 27.1 and 21.4% for Ralstonia solanacearum and Colletotrichum gleosporioides respectively. All the selected isolates of actinomycetes were identified as belonging to the genus of Streptomycetes spp. The potential actinomycetes were stored in freeze-dried form for future usage. This study showed that more diverse population of actinomycetes was obtained from the undisturbed forested peat soil area ecosystem compared to the agricultural peat soil area.

22. Comparative Studies of Region-Based Segmentation Algorithms on Natural and Remote Sensing Images

Abstract

Region-based segmentation algorithms are used as a preprocessing approach to generate over-segmented regions. Over-segmented regions refer to the creation of small regions in an image that represent no meaningful object regions. It has been observed that there are limited works on the performance comparison of the region-based segmentation algorithms on both natural and remote sensing (RS) images. Hence, the objective is to compare the performance of region-based segmentation algorithms on natural and RS images with different complexity of object regions of interest (ROIs). There are four algorithms (Felzenszwalb and Huttenlocher (FH), Quick Shift (QS), Compact Watershed (CW), and Simple Linear Iterative Clustering (SLIC)) being compared using two public datasets. The adapted rand error (ARE) and variation of information (VOI) are used for the segmentation evaluations. Generally, the experiments showed that the SLIC achieved better results as compared to the other algorithms for both images with different complexities of ROIs. This is mainly because the over-segmented regions produced by the SLIC adhered to the image object boundaries well than the over-segmented regions generated by other algorithms. However, CW achieved better average ARE than SLIC for RS images because CW has compactness and marker parameters which influence it to achieve better results.

Abstract

Non-alcoholic fatty liver disease (NAFLD) has been on a global rise. While animal models have rendered valuable insights to the pathogenesis of NAFLD, discrepancy with patient data still exists. Since non-alcoholic steatohepatitis (NASH) involves chronic inflammation, and CD4+ T cell infiltration of the liver is characteristic of NASH patients, we established and characterized a humanized mouse model to identify human-specific immune response(s) associated with NAFLD progression. Immunodeficient mice engrafted with human immune cells (HIL mice) were fed with high fat and high calorie (HFHC) or chow diet for 20 weeks. Liver histology and immune profile of HIL mice were analyzed and compared with patient data. HIL mice on HFHC diet developed steatosis, inflammation and fibrosis of the liver. Human CD4+ central and effector memory T cells increased within the liver and in the peripheral blood of our HIL mice, accompanied by marked up-regulation of pro-inflammatory cytokines (IL-17A and IFNγ). In vivo depletion of human CD4+ T cells in HIL mice reduced liver inflammation and fibrosis, but not steatosis. Our results highlight CD4+ memory T cell subsets as important drivers of NAFLD progression from steatosis to fibrosis and provides a humanized mouse model for pre-clinical evaluation of potential therapeutics.

Abstract

Solid‐state fermentation is one of the easiest and cheapest methods for producing microbial bioactive com‐ pounds. Trichoderma harzianum has long been recognised as one of the potential fungi for this purpose. Trichoderma sp. were isolated from banana rhizosphere using the soil dilution method and later screened for their ability to produce cellulases using filter paper activity (FPase) and the carboxylmethyl cellulase (CMCase) test. Trichoderma sp. were also subjected to one factor change at a time to determine the effects of different parameters on cellulase production. It was observed that T. harzianum TF2 showed the ability to produce higher cellulase activity when wheat bran was used as the substrate. The results showed that 38.5 U/g of cellulase was produced with the use of wheat bran coupled with an incubation temperature of 28 °C and moisture content of 60%. T. harzianum TF2 showed good potential for use as a culture for cellulase production in this study due to its higher cellulase production under solid‐state fermentation, with the possibility of its application to industry.

Abstract

Wood Identification has never been more important to serve the purpose of global forest species protection and timber regulation. Macroscopic level wood identification practiced by wood anatomists can identify wood up to genus level. This is sufficient to serve as a frontline identification to fight against illegal wood logging and timber trade for law enforcement authority. However, frontline enforcement official may lack of the accuracy and confidence of a well trained wood anatomist. Hence, computer assisted method such as machine vision methods are developed to do rapid field identification for law enforcement official. In this paper, we proposed a rapid and robust macroscopic wood identification system using machine vision method with off-the-shelf smartphone and retrofitted macro-lens. Our system is cost effective, easily accessible, fast and scalable at the same time provides human-level accuracy on identification. Camera-enabled smartphone with Internet connectivity coupled with a macro-lens provides a simple and effective digital acquisition of macroscopic wood images which are essential for macroscopic wood identification. Abstract

Peatland plays an important role not just as a carbon store but also in facilitating the flux of greenhouse gasses into the atmosphere. Apart from that, peatland is also home to a diverse population of microorganisms such as bacteria, fungi, and actinomycetes. Actinomycetes were known to be one of the most ubiquitous microbes that can be found in most of the soil types including peat soil. In this study, seventy isolates of actinomycetes were isolated from the peat soil using the soil dilution method. The 70 isolates of actinomycetes were later screened for their ability to produce secondary metabolites and antimicrobial activities using the agar diffusion method before the selected potential isolates were identified by targeting their 16S rRNA region. The results obtained showed 34.3% produce cellulase followed by, 12.8, 31.7, 80.0, and 51.4% for mannanase, xylanase, lipase, and protease respectively. The percentage of actinomycetes producing antimicrobial activity was 27.1 and 21.4% for Ralstonia solanacearum and Colletotrichum gleosporioides respectively. All the selected isolates of actinomycetes were identified as belonging to the genus of Streptomycetes spp. The potential actinomycetes were stored in freeze-dried form for future usage. This study showed that more diverse population of actinomycetes was obtained from the undisturbed forested peat soil area ecosystem compared to the agricultural peat soil area.
