15. Multiplexed transcriptional profiling of Dermatophagoides house dust mites allergens in human epithelium cells.

Abstract

Allergic asthma, a chronic disease characterized by airway inflammation, poses a significant public health concern. It is well‐established that house dust mites (HDMs) are common inducers of allergic responses in individuals, particularly children. In central Taiwan, our research team observed that over 80% of allergic children exhibited sensitization to various HDMs species. This investigation aims to bridge the gap between these observations and a better understanding of the early fundamental mechanisms for preventing allergic diseases. Specifically, our study delves into the impact of crude extracts of HDMs on human epithelial BEAS‐2B cells. Our findings, based on RNA sequencing (RNA‐seq) analysis, shed light on how three major Dermatophagoides HDMs allergens activate a common Toll‐like receptor signaling pathway in human epithelial cells within a 4‐h treatment. During this process, the nuclear transcription factor NF‐κB translocated into the cell nucleus within 30 min of allergen stimulation, triggering the expression of pro‐inflammatory genes such as IL‐6 and IL‐8 over 4 h. Additionally, when the cells were treated with specific Dermatophagoides microceras (Der m) allergens, it resulted in the upregulation of genes that regulate type 1 diabetes mellitus (T1DM) signaling pathways. This led to the mediation of IL‐12A inflammation. Furthermore, there was an increase in gene sets associated with cilia function and the microtubule cytoskeleton in human epithelial cells after treatment with a combination of Der m allergens and Dexamethasone. Additionally, OMICs analysis was conducted to examine the effects of HDMs allergenic stimulation on human epidermal cells. We aimed to improve our understanding of the molecular mechanisms within cells and identify potential targets and natural products in the treatment of asthma caused by HDMs allergens. Abstract

Recently, purchasing intention towards green products has gained global attention due to their extensive use and high environmental issues. Thus, the current article investigates environmental concern, environmental knowledge, green product, and eco-innovation influence on the green purchase intention of green products in Taiwan industry. The research also examines the mediating role of consumers’ attention among environmental concerns environmental knowledge, green products eco-innovation, and green purchase intention. The article used questionnaires to collect the primary data from the sampled population. To examine the hypothesis, the PLS methodology was adopted. The results indicated that environmental concern, environmental knowledge, green product, and eco-innovation have a positive association with green purchase intention. The findings also exposed that consumer attention significantly and positively mediates environmental concern, environmental knowledge, green product, eco-innovation, and green purchase intention. The paper also proposed some guidelines to the practitioners which help them in the development of green-related policies to increase purchase intention.

Abstract

Economic growth is a global requirement that requires extensive energy consumption, and this phenomenon needs researchers' attention and regulators' focus. Thereby, the paper scrutinizes the determinants of energy consumption such as fossil fuel energy consumption (FFEC), energy use, nuclear energy consumption (NEC), energy import, and renewable energy consumption (REC) and sustainability-oriented eco-innovation and their effectiveness on the economic growth of Saudi Arabia. The study extracted data from the World Bank from 1989 to 2020. Stationarity was examined using augmented Dickey-Fuller (ADF) tests, and the associations among constructs were analyzed through QARDL model. The findings revealed that FFEC, EU, NEC, EI, REC, and sustainability-oriented eco-innovation are significantly correlated with the EG of Saudi Arabia. The study also provides insights to new researchers who will investigate this area in the future and guides regulators in developing regulations related to economic growth using an appropriate level of energy and adoption of sustainability-oriented eco-innovation. 35. AN INTERCULTURAL COMPARISON OF NEGOTIATION STYLES BETWEEN TAIWAN AND THE UNITED STATES

Abstract

Despite the implied enthusiasm for increasing global interaction and economic exchange, a lack of understanding of cultural differences has been found to hinder the ability of firms to conduct business or negotiations efficiently with different countries. By means of a thorough examination of the different styles of negotiation between Taiwan and the US, the research herein identifies the effects of culture on negotiations. The approach uses Casse and Deols’ model, which considers styles of negotiation and degrees of individualism to be the dependent and independent variables. Data was collected from sales and purchasing managers of public companies listed on the stock exchanges of Taipei and New York by means of an online survey. Structural equation modeling was used to test hypothesized models and the overall hypotheses of the research. A two-step approach was employed in the research that consisted of exploratory factor analysis and confirmatory factor analysis. The findings showed that an individualist attitude directly affects the style of negotiation, that nationality is a moderated variant of individualist attitude and style of negotiation, and that different styles of negotiation are preferred by Taiwanese and American negotiators. These findings could be useful in the application of a specific set of values and attitudes that directly relate to regional cultural attributes. The study may also assist prospective cross-cultural negotiators to develop better negotiation skills by providing insights into the nuances of international negotiations between businesses.

Abstract

This case is described of an eleven-month-old infant with lower limbs swelling and the left elbow skeletal malformation following a fall. The radionuclide bone scan was performed to exclude bone infection or congenital skeletal anomaly. The images unexpectedly showed multiple increased radioactive foci throughout the whole body. It was a strong probability of child abuse. All lesions are readily apparent on the following plain film radiographs and MRI.

Abstract

The study of protein function and dynamics in their native cellular environment is essential for progressing fundamental science. To overcome the requirement of genetic modification of the protein or the limitations of dissociable fluorescent ligands, ligand-directed (LD) chemistry has most recently emerged as a complementary, bioorthogonal approach for labeling native proteins. Here, we describe the rational design, development, and application of the first ligand-directed chemistry approach for labeling the A1AR in living cells. We pharmacologically demonstrate covalent labeling of A1AR expressed in living cells while the orthosteric binding site remains available. The probes were imaged using confocal microscopy and fluorescence correlation spectroscopy to study A1AR localization and dynamics in living cells. Additionally, the probes allowed visualization of the specific localization of A1ARs endogenously expressed in dorsal root ganglion (DRG) neurons. LD probes developed here hold promise for illuminating ligand-binding, receptor signaling, and trafficking of the A1AR in more physiologically relevant environments.
