Background: Vaginal childbirth is one of the main risk factors for pelvic floor dysfunction. Magnetic resonance imaging (MRI) can facilitate quantitative evaluation of the morphology and function of the pelvic floor in static and dynamic environments. The objective of this study was to investigate the changes in pelvic floor morphology and function in primigravida women before pregnancy (BP) and after vaginal delivery. Methods: Primigravida women underwent pelvic floor MRI scans BP, at 3 months postpartum (3mPP), and at 6 months postpartum (6mPP). Various pelvic floor MRI data were measured, including the obturator internus muscle (OIM) area, anterior pelvic area, puborectalis muscle thickness (PRT), levator plate angle (LPA), iliococcygeal angle, bladder–pubococcygeal line (B-PCL), uterus–pubococcygeal line (U-PCL), puborectal hiatus line, muscular pelvic floor relaxation line, levator hiatus area, urethral mobility, bladder neck descent, and cervix descent. Results: In the resting state, the OIM area and PRT decreased whereas the anterior pelvic area increased from BP to 3mPP. During the straining maneuver, all parameters except U-PCL and urethral mobility showed statistically significant differences (P < 0.05). The OIM area and PRT increased whereas the anterior pelvic area decreased from 3mPP to 6mPP. During the straining maneuver, B-PCL, bladder neck descent, levator hiatus area, and LPA showed statistically significant differences (P < 0.05). In the resting state, the OIM area and PRT decreased whereas the anterior pelvic area increased from BP to 6mPP. During the straining maneuver, B-PCL, muscular pelvic floor relaxation line, and bladder neck descent showed statistically significant differences (P < 0.05). Conclusion: Vaginal delivery can cause pelvic floor injury that may gradually recover over time. However, the injury does not fully recover to the pre-pregnancy level within 6mPP. Purpose: To perform risk assessment and analysis of potential infection during stomatology workflow in a hospital in the context of a major infectious disease outbreak, and to determine the key failure modes and measures to prevent and control infection. Method: Following the Failure Modes and Effects Analysis (FMEA) method based on the stomatology workflow, the opinions of 30 domain-experts in related fields were collected through questionnaires to determine all potential failure modes in the severity (S), occurrence (O), and detectability (D) dimensions. The group score was then integrated through the median method and the risk priority number (RPN) was obtained. Finally, combined with expert experience, a score above 100 was considered to define a key potential failure mode. The data collection period for this study was from August to September 2023. Results: The key failure modes identified were "Patient concealed epidemiological history (A110) (RPN: 149.6; Rank: 1)," "At the pre-examination door, safe distancing cannot be practiced due to the large number of visitors (A120) (RPN: 147; Rank: 2)," "The patient does not cooperate in checking the health code or itinerary code (A18) (RPN: 128, Rank: 3)," "The patient provides a non-personal health code or itinerary code (A19) (RPN:121.5; Rank: 4)," "Pre-examination personnel did not strictly implement the inquiry of epidemiological history (A12) (RPN: 120; Rank: 5)," and "The patient did not wear a mask according to the specification (A111) (RPN:108.0; Rank: 6)". The key risk area was "Before diagnosis and treatment (A)". Conclusion: Insufficient records of patient epidemiological history may increase the spread of COVID-19 in the oral diagnosis and treatment environment. High-density areas where patients gather are prone to become sources of infection. Finally, improper use of personal protective equipment increases the risk of cross-infection with COVID-19. However, in the face of the potential spread of major infectious diseases in the future, the government and hospitals need to build a more comprehensive epidemiological notification system to provide the population with early action trajectories and warning reports. Rationale: Colorectal cancer (CRC) is a leading cause of cancer-related mortality. Epigenetic modifications play a significant role in the progression of CRC. KAT7, a histone acetyltransferase, has an unclear role in CRC. Methods: In this research, we analyzed the expression of KAT7 in CRC patients and its correlation with prognosis using the GEO database, western blot, and immunohistochemistry. We assessed the impact of KAT7 on CRC cell functions through cell viability, colony formation, flow cytometry, scratch, and transwell assays. Mechanistic insights were obtained via RNA sequencing and ChIP-qPCR. Additionally, we evaluated the effects of KAT7 on CRC growth and metastasis in vivo using mouse subcutaneous tumor and lung metastasis models. Results: In this study, we discovered an upregulated KAT7 signaling pathway in CRC and its association with poor patient survival. Knockdown of KAT7 promotes apoptosis and inhibits proliferation, migration, and invasion of CRC cells. Conversely, KAT7 overexpression enhanced these cellular processes. In vivo assays confirmed that knockdown of KAT7 can inhibit CRC proliferation and lung metastasis. Mechanistically, KAT7 acetylated histone H3 at lysine 14 (H3K14) to enhance MRAS transcription, which activated the MAPK/ERK pathway and promoted tumorigenesis. The enzymatic function of KAT7 as an acetyltransferase is crucial for the advancement of colorectal cancer. In KAT7 knockdown CRC cells, re-expression of KAT7, but not an acetyltransferase-deficient mutant, rescued MRAS expression, ERK phosphorylation, and CRC tumorigenesis. Conclusion: We found that KAT7 is highly expressed in CRC patients, and those with high KAT7 expression have a worse prognosis. KAT7 enhances MRAS gene transcription by promoting H3K14 acetylation, thereby activating the MAPK/ERK pathway and promoting malignant phenotypes of CRC.
