Alkaline phosphatase (ALP) plays a vital role in clinical diagnoses and biomedical research. It is important to develop some convenient but sensitive methods for ALP activity detection. In this study, a multicolor biosensor for ALP activity has been developed based on the peroxidase activity of copper nanoclusters (CuNCs) and etching of gold nanorods (AuNRs). The presence of CuNCs can catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to produce blue TMB+. In acid solution, TMB+ can cause the etching of AuNRs accompanied by a significant color change of the system. The presence of sodium pyrophosphate (PPi) can inhibit the peroxidase activity of CuNCs, which can be recovered after the addition of ALP. Different ALP added results in the recovery of the catalytic activity of CuNCs to different degrees and generates different amounts of TMB+. This consequently affected the morphology of the AuNRs in the system and results in the output of a vivid color change, which can be recognized with the naked eyes easily without any complicated instruments. The biosensor has a linear relationship with ALP activity in the range of 10.0–80.0 U L−1, and the detection limit is 4.6 U L−1. Background We explored the effects of repeated plateletpheresis on the platelet P-selectin expression and soluble P-selectin (sP-selectin) concentrations in platelet donors. Methods Totally 289 platelet donors and 97 first-time whole blood (WB) donors were enrolled from the blood donor registry at the Fujian provincial blood center, China. The accumulative numbers of plateletpheresis in the last 2 y for participants were recorded, and the basal concentrations of platelet count, sP-selectin and total platelet P-selectin (pP-selectin) were determined. Results Platelet donors had significantly higher basal concentrations of sP-selectin compared to WB donors (24.12 ± 7.33 ng/mL vs. 20.74 ± 5.44 ng/mL, P < 0.0001), with no difference in platelet count and pP-selectin concentrations. Increased numbers of platelet donation were correlated with a steady increase of sP-selectin ( r = 0.18, P = 0.002). Multivariate regression analysis identified that the frequency of plateletpheresis is an independent factor for the rise of the sP-selectin concentration (t = 2.64, P = 0.009) while no association was found for pP-selectin and platelet count. Conclusions Repeated plateletpheresis could result in an increased basal concentration of sP-selectin in blood donors whereas not an alteration in the concentrations of total platelet P-selectin. • A multicolor sensor for H 2 S detection to evaluate meat freshness had been developed. • Inorganic mimetic enzyme (AuNPs/Cu2+-BNNS) was used to construct a multicolor sensor of H 2 S. • The aspect ratio of the gold nanorods (AuNRs) was changed to get visual colorimetric signals. • The multicolor sensor has the advantages of fastness, simplicity, visualization, etc. Hydrogen sulfide (H 2 S) has emerged as an important indicator in the spoilage process of meat. In this study, a mimetic enzyme based on Cu2+-modified boron nitride nanosheets-supported gold nanoparticles (AuNPs/Cu2+-BNNS) was synthesized, which can be used to catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB). The H 2 S gas can inhibit the activity of AuNPs/Cu2+-BNNS toward catalytic oxidation of TMB. Meanwhile, the usage of headspace method could avoid most interferences in the rotten sample. Various concentrations of TMB+ could change the aspect ratio of the gold nanoroads (AuNRs), which results in vivid color changing and UV–vis spectra shifting. The sensor had a good linear relationship with H 2 S concentration ranging from 10.0 to 90.0 μmol/L, and the detection limit is 7.8 μmol/L. Previous investigations demonstrated that Fusarium oxysporum (Fo), which is not pathogenic to cucumbers, could serve as a biological control agent for managing Fusarium wilt of cucumber caused by Fo f. sp. cucumerinum (Foc) in Taiwan. However, thus far it has not been possible to separate the populations of pathogenic Fo from the nonpathogenic isolates that have biological control potential through their morphological characteristics. Although these two populations can be distinguished from one another using a bioassay, the work is laborious and time-consuming. In this study, a fragment of the intergenic spacer (IGS) region of ribosomal DNA from an Fo biological control agent, Fo366, was PCR-amplified with published general primers, FIGS11/FIGS12 and sequenced. A new primer, NPIGS-R, which was designed based on the IGS sequence, was paired with the FIGS11 primer. These primers were then evaluated for their specificity to amplify DNA from nonpathogenic Fo isolates that have biological control potential. The results showed that the modified primer pair, FIGS11/NPIGS-R, amplified a 500-bp DNA fragment from five of seven nonpathogenic Fo isolates. These five Fo isolates delayed symptom development of cucumber Fusarium wilt in greenhouse bioassay tests. Seventy-seven Fo isolates were obtained from the soil and plant tissues and then subjected to amplification using the modified primer pair; six samples showed positive amplification. These six isolates did not cause symptoms on cucumber seedlings when grown in peat moss infested with the isolates and delayed disease development when the same plants were subsequently inoculated with a virulent isolate of Foc. Therefore, the modified primer pair may prove useful for the identification of Fo isolates that are nonpathogenic to cucumber which can potentially act as biocontrol agents for Fusarium wilt of cucumber. Hydrogen sulfide (H 2 S), as a hazardous gas, is often found around dump areas. Long term exposure can cause harm to health, it is highly necessary to develop some simple and sensitive methods for on-site H 2 S detection. Herein, a convenient photothermal assay has been designed for the quantitation of H 2 S using a handheld thermometer as readout. Au@Ag nanocubes (Au@Ag NCs), a core-shell nanocomposite with strong light absorption at ∼450 nm, was chosen as a novel photothermal agent in this study. Under the laser irradiation at 450 nm, the Au@Ag NCs show a strong photothermal effect, and a significant temperature enhancement can be measured by the thermometer easily. The presence of H 2 S can lead to the deposition of sulfur onto Au@Ag NCs, altering the localized surface plasmon resonance absorption, size, surface composition, and morphology of Au@Ag NCs and hence leading to the reduction of photothermal effect. The change of the temperature has a linear relationship with the H 2 S concentration in the range of 0.5–80.0 μM with a detection limit of 0.35 μM. By combining with simple sample purification procedures, the developed method has been applied to detect H 2 S in garbage odor gas with satisfactory results. Purpose: Transarterial chemoembolization (TACE) with tyrosine kinase inhibitors (TKIs) has been increasingly used to treat unresectable hepatocellular carcinoma (uHCC). However, the superiority of combination therapy to TACE monotherapy remains controversial. Therefore, here we performed a meta-analysis to evaluate the efficacy and safety of TACE plus TKIs in patients with uHCC. Methods: We searched four databases for eligible studies. The primary outcome was time to progression (TTP), while the secondary outcomes were overall survival (OS), tumor response rates, and adverse events (AEs). Pooled hazard ratios (HRs) with 95% confidence intervals (95% CIs) were collected for TTP and OS, and the data were analyzed using random-effects meta-analysis models in STATA software. OR and 95% CIs were used to estimate dichotomous variables (complete remission[CR], partial remission[PR], stable disease[SD], progressive disease[PD], objective response rate[ORR], disease control rate[DCR], and AEs) using RStudio’s random-effects model. Quality assessments were performed using the Newcastle–Ottawa scale (NOS) for observational studies and the Cochrane risk of bias tool for randomized controlled trials (RCTs). Results: The meta-analysis included 30 studies (9 RCTs, 21 observational studies) with 8246 patients. We judged the risk of bias as low in 44.4% (4/9) of the RCTs and high in 55.6% (5/9) of the RCTs. All observational studies were considered of high quality, with a NOS score of at least 6. Compared with TACE alone or TACE plus placebo, TACE combined with TKIs was superior in prolonging TTP (combined HR 0.72, 95% CI 0.65–0.80), OS (combined HR 0.57, 95% CI 0.49–0.67), and objective response rate (OR 2.13, 95% CI 1.23–3.67) in patients with uHCC. However, TACE plus TKIs caused a higher incidence of AEs, especially hand-foot skin reactions (OR 87.17%, 95%CI 42.88–177.23), diarrhea (OR 18.13%, 95%CI 9.32–35.27), and hypertension (OR 12.24%, 95%CI 5.89–25.42). Conclusions: Our meta-analysis found that TACE plus TKIs may be beneficial for patients with uHCC in terms of TTP, OS, and tumor response rates. However, combination therapy is also associated with a significantly increased risk of adverse reactions. Therefore, we must evaluate the clinical benefits and risks of combination therapy. In intensive pig production, the fighting behavior of weaning piglets after merging pens is relatively common. Fighting behavior not only easily causes injury in pigs but also affects the production performance of pigs. To reduce fighting behavior in farms, this study aimed to explore the possible effect of odorous substances on piglet fighting behavior after merging into a large pen. Six different sprays were tested: original creamy, cheese flavor, orange flavor, truffle, vanilla and pigpen flavor. In each experiment, two groups were set (one odor‐sprayed and no sprayed control), and 12 pigs were used per group. After mixing, the frequency of occurrence of various piglet behaviors in different pens was recorded. During this period, salivary cortisol levels and skin lesion scores were evaluated. As a result, the piglets sprayed with the original creamy, cheese flavor and vanilla substances obtained significantly higher average daily gain and feed intake and showed a significantly lower incidence of fighting behavior, and the skin lesion score and salivary cortisol of piglets were also reduced significantly. Background: Recent genome-wide association studies in Caucasians suggested that an association exists between the ABO gene locus and soluble levels of P-selectin (sP-selectin). However, it is unclear if the relationship corresponds to the phenotypic expression of ABO groups or is present in different ethnic groups. The aim of this study was to verify this observation at both genotypic and phenotypic levels in a healthy Chinese population.Study Design and Methods: The ABO blood groups were determined by both phenotypes and genotypes in 440 healthy Chinese Han volunteers, while P-selectin levels were evaluated for sP-selectin and total platelet P-selectin (pP-selectin).Results: ABO phenotyping and quantitative analysis of individual sP-selectin plasma levels were combined to demonstrate that individuals phenotypically expressing the A antigen have approximately 20% lower sP-selectin plasma levels than those carrying the B or O phenotype (p < 0.0001), but that no difference exists between A and AB and between B and O phenotypes. Genotyping data revealed that the presence of the A gene could be attributed to the observed difference in phenotype comparison, with no difference between A/A, A/B, and A/O genotypes. There were also no associations between ABO blood groups, either phenotypes or genotypes, and pP-selectin levels.Conclusion: This study demonstrated an association between sP-selectin levels and ABO groups in a Chinese Han population, implicating its generalizability to other ethnic groups. To meet the increasing need for point-of-care testing (POCT), simple and portable readout strategies would be highly desirable. Thermometer with high accuracy and straightforward readout is an ideal tool for the development of new POCT methods. The exploration of new thermometer-based detection methods is of great significance. In this study, a simple biosensor for glucose based on the photothermal effect of gold nanorods using a simple thermometer as readout has been developed. In the presence of glucose oxidase, glucose can react with the dissolved oxygen to produce H 2 O 2. With the help of Fe2+, H 2 O 2 can etch gold nanorods (AuNRs) to different aspect ratios. The decrease of the aspect ratio of AuNRs leads to the blue-shift of the localized surface plasmon resonance peak, resulting in a decrease of photothermal effect in the near-infrared regions and the temperature of the system decreased. The change of the temperature has a linear relationship with the logarithm of glucose concentration in the range of 1.0–10.0 mM with a detection limit of 0.8 mM. The proposed method possesses a bias offset of −0.03 mM for glucose detection compared to the hospital method. Since many enzymatic reactions can produce H 2 O 2 , the principle can be modified to detect different targets by simply change of the enzyme used. We demonstrated a facile approach, by adjusting the solvent ratio of water/acetone binary mixture, to alter the intermolecular interactions between Enzalutamide (ENZ) and hydroxypropyl methylcellulose acetate succinate (HPMC-AS) for spray drying process, which can be readily implemented to produce spray-dried dispersions (SDD) with enhanced stability and bioavailability. The prepared SDD of ENZ/HPMC-AS were examined systematically in terms of particle size, morphology, dissolution, solubility, stability, and bioavailability. Our results show that the introduction of water (up to 30% volume fraction) can effectively reduce the hydrodynamic diameter of HPMC-AS from approximately 220 nm to 160 nm (a reduction of c.a. 20%), which increases the miscibility of the drug and polymer, delaying or inhibiting the crystallization of ENZ during the spray drying process, resulting in a homogeneous amorphous phase. The benefits of using acetone/water binary mixture were subsequently evidenced by an increased specific surface area, improved dissolution profile and relative bioavailability, enhanced stability, and elevated drug release rate. This fundamental finding underpins the great potential of using binary mixture for spray drying process to process active pharmaceutical ingredients (APIs) that are otherwise challenging to handle. Prussian blue analogues as cathode material for sodium-ion batteries suffer from poor cycling stability and inferior rate performance because of crystal defects and sluggish ion diffusion kinetics. The Na 1.02 Fe[Fe(CN) 6 ] 0.95 ·□ 0.05 Prussian blue nanoparticles with few lattice vacancies and a yolk-shell structure are designed and synthesized to address these two problems. The hierarchical structure endows yolk-shelled Prussian blue nanoparticles with large specific surface area, large pore volume and short ion pathways. As a result, the yolk-shell Prussian blue nanoparticles exhibit a high discharge capacity (140 mA h g−1 at 0.1C), high-rate performance (51 mA h g−1 at 10C) and excellent cycling stability (95.8% capacity retention after 500 cycles at 1C). Directly coating an active pharmaceutical ingredient (API) onto excipient granules has been a common approach to prepare solid dosage forms. The combination of supercritical anti-solvent (SAS) and fluidized bed (FB) coating technology (SAS-FB) has the advantages of preventing nanoparticles aggregation, oxidation and light exposure. However individual operating parameters and factors which contribute to the overall coating efficiency remain to be defined. Sirolimus is an immunosuppressive agent for preventing the rejection of organ transplants and this drug is sensitive to light exposure and high temperature. Our study used sirolimus as the model API to evaluate parameters including temperature, pressure, drug concentration, mass, material and diameter of carrier, CO 2 flow rate and solvent in the SAS-FB process. By optimizing these parameters, we achieved a 3.5-fold enhancement of the coating efficiency over the standard condition. A series of characterizations of the sirolimus coated particles were performed from which scanning electron microscopy and Raman mapping confirmed that the sirolimus particles were uniformly coated on carriers as cuboid particles; X-ray powder diffraction showed that processed sirolimus is crystalline but has lower crystallinity than the API, and fourier transform infrared spectroscopy and differential scanning calorimeter confirmed that there is no chemical interaction between sirolimus and carriers after SAS-FB processing. Finally compared to sirolimus alone, sirolimus coated particles displayed a faster dissolution and higher bioavailability. Collectively, our optimized operation parameters for SAS-FB coating technique provide a useful guidance for achieving higher efficiency of drug coating and faster release rate of sirolimus pellets, which has the potential to apply to other APIs.
