Dilated cardiomyopathy (DCM) is a heart condition that causes enlarged and weakened left ventricles and affects the heart’s ability to pump blood effectively. Most genetic etiology still needs to be understood. Previously, we have used the known germline hereditary fusion genes (HFGs) to identify HFGs associated with multiple myeloma and leukemia. In this study, we have developed a statistical model to study fusion transcripts discovered from the left ventricles of 122 DCM patients and 252 GTEx (Genotype Tissue Expression) healthy controls to discover novel HFGs, ranging from 4% to 87.7%, and EFGs, ranging from 4% to 99.2%, associated with DCM. This discovery of numerous novel HFGs and EFGs associated with DCM provides first-hand evidence that DCM results from interactive developmental consequences between germline genetic and environmental abnormalities and paves the way for future research and diagnostic and therapeutic applications, instilling hope for the future of DCM treatment.

Moringa oleifera Lam. leaves are a new raw food material rich in polysaccharides. These polysaccharides exhibit various biological properties, including antioxidant, hypoglycemic and immunoregulatory effects. However, the use of Moringa oleifera Lam. leaves polysaccharides (MOLP) may be limited by their large molecular weight (MW) and presence of numerous impurities, such as pigments. Research has indicated that degraded polysaccharides usually exhibit high biological activity because of changes in physical structure and chemical properties. In this study, we focused on the extraction of a degraded-modified fraction from MOLP using the Ultraviolet/ Hydrogen peroxide (UV/H2O2) method. Specifically, the physicochemical properties and glycosidic bond composition of a particular fraction (UV/H2O2 degraded Moringa oleifera Lam. leaves polysaccharides in 3 h called DMOLP-3) were investigated. In addition, in vitro simulated digestion experiments showed that DMOLP-3 was only partially degraded during gastrointestinal digestion, indicating that DMOLP-3 can be utilised by gut microorganisms. Furthermore, the prebiotic properties of MOLP and DMOLP-3 was studied using an in vitro faecal fermentation model. The results indicated that compared with MOLP, DMOLP-3 led to a decrease in both the colour and MW of the polysaccharides. In addition, this model exhibited enhanced solubility and antioxidant capabilities while also influencing the surface morphology. Moreover, DMOLP-3 can facilitate the proliferation of advantageous microorganisms and enhance the synthesis of short-chain fatty acids (SCFAs). These results provide valuable insights into the utilization of bioactive components in Moringa oleifera Lam. leaves for the intestinal health.

BackgroundLeptin Receptor (LEPR) has been suggested to have several roles in cancer metastasis. However, the role of LEPR and its underlying mechanisms in lymphatic metastasis of hepatocarcinoma have not yet been studied.MethodsWe performed bioinformatics analysis, qRT-PCR, western blotting, immunohistochemistry, immunofluorescence, enzyme-linked immunosorbent, coimmunoprecipitation assays and a series of functional assays to investigate the roles of LEPR in hepatocellular carcinoma.ResultsWe discovered that LEPR was highly expressed in liver cancer tissues, and the expression of LEPR in Hca-F cells was higher than that in Hca-P cells. Furthermore, LEPR promotes the proliferation, migration and invasion and inhibits the apoptosis of hepatocarcinoma lymphatic metastatic cells. Further studies indicated that LEPR interacts with ANXA7. Mechanistically, LEPR regulated ERK1/2 and JAK2/STAT3 expression via ANXA7 regulation.ConclusionsThese findings unveiled a previously unappreciated role of LEPR in the regulation of lymphatic metastatic hepatocellular carcinoma, assigning ANXA7-LEPR as a promising therapeutic target for liver cancer treatments.

A collector-generator (C-G) cell used for the in-situ detection of H2 generated through electrochemical catalysis was described. The cell was mainly assembled with two fluorine-doped tin oxide (FTO) electrodes deposited with Pt nanoparticles, the magnitude of the current generated by the electrocatalytic oxidation of H2 at the collector was used for quantitative analysis of H2 generated at the generator. When the generator potential was set at −0.5 ∼ −0.6 V vs. Ag/AgCl and the collector potential at 0.4 ∼ 0.5 V vs. Ag/AgCl, the total Faradaic efficiency of the C-G cell could stabilize about 70%, the detection limit was about 45 μmol/L and the sensitivity was about 1 mA/55 μmol L−1. This dual working electrode technology could provide a convenient and rapid method for H2 determination and evaluate the performance of H2 generation catalysts that assembled on the semiconductor thin-films.

In underwater optical communication systems, timely acquisition of distance information with the receiving end and high-speed communication are required. This paper proposes to integrate the monocular ranging function into the underwater blue-green laser communication system to realize the underwater optical communication ranging by integrating short-distance visual ranging and long-distance laser ranging. Using Monte Carlo method simulation, an underwater laser signal channel transmission model is established to simulate underwater laser spot images under different target sizes, distances and water qualities. We analyze the influence of these three factors on the accuracy of underwater monocular ranging. Considering the influence of spot diffusion and contrast reduction in simulated images on ranging accuracy, a monocular ranging algorithm is proposed that combines image enhancement and Zernike moments subpixel edge detection for small-sized underwater light source targets to improve the accuracy of the monocular ranging algorithm. Laboratory pool experiments were conducted to verify that the algorithm accuracy can meet the requirements.

A dual-wavelength transceiver separation method is proposed for the problem of self-interference limiting the system communication performance in Full-Duplex Underwater Wireless Optical Communication (FD-UWOC) systems. Based on the Monte Carlo numerical simulation, the effects of self-interference on the performance of 450 nm single-wavelength, 525 nm single-wavelength and dual-wavelength FD-UWOC systems are analyzed under three typical water qualities of Jerlov I, Jerlov IB and Jerlov II. The simulation results show that the poorer the water quality, the stronger the self-interference. It is also shown that the enhancement of self-interference will significantly reduce the Optical Signal to Noise Ratio (OSNR) of FD-UWOC, which limits the longest transmission distance of the system. The FD-UWOC system using the dual-wavelength transceiver separation method can bring about 60 dB improvement in OSNR compared with the single-wavelength FD-UWOC system, which can achieve a longer transmission distance. An 80 m improvement in Jerlov II water quality, and an effective transmission distance of more than 100 m in Jerlov I and Jerlov IB water quality are achieved. These results provide some reference significance to the design of FD-UWOC system.

Abstract Background/Aims Hepatitis E virus (HEV)-triggered acute-on-chronic liver failure (ACLF) has unacceptably high short-term mortality. However, it is unclear whether the existing predictive scoring models are applicable to evaluate the prognosis of HEV-triggered ACLF. Methods We screened datasets of patients with HEV-triggered ACLF from a regional tertiary hospital for infectious diseases in Shanghai, China, between January 2011 and January 2021. Clinical and laboratory parameters were recorded and compared to determine a variety of short-term mortality risk factors, which were used to develop and validate a new prognostic scoring model. Results Out of 4952 HEV-infected patients, 817 patients with underlying chronic liver disease were enrolled in this study. Among these, 371 patients with HEV-triggered ACLF were identified and allocated to the training set (n = 254) and test set (n = 117).
