The safety of microplastics (MPs) and associated health effects has been one of the major concerns worldwide. However, the role of photoaging toward the risk of MPs in water ecosystems remains inconclusive yet. In this study, the size of polyamide (PA, ∼32.50 μm) MPs was obviously decreased after photoaging in water containing fulvic acid (FA) and humic acid (HA) (∼19.75 and ∼24.30 μm, respectively). Nanoplastics were formed (4.65% and 2.03%, respectively) and hydrophilia and colloidal stability was improved due to the formation of oxygen-containing functional groups. FA-aged PA exhibited higher inhibition on body length and weight of developing zebrafish than HA-aged and pristine PA. Photoaged MPs in intestine were more difficult to be depurated by zebrafish, leading to the disappearance of intestinal folding, shedding of more enterocytes, and emaciation of intestinal microvilli. Dietary lipid digestion in larvae was inhibited by aged PA due to oxidative stress-triggered lipid peroxidation and inhibition of lipase activities and bile acids secretion. Exposure of photoaged MPs down-regulated genes (cd36, dgat1a, dgat2, mttp, etc.) associated with triglyceride resynthesis and transportation, resulting in lipid maladsorption and growth inhibition. Our findings highlight the potential negative effects of environmentally aged MPs on diet digestion and nutrient assimilation in fish.

Several studies have documented the hypolipidemic effect of anthocyanin-rich plants in vitro and in vivo. The objective of this study was to elucidate the inhibitory activity of anthocyanin-rich fraction from Thai berries against fat digestive enzymes. The ability of Thai berries to bind bile acid, disrupt cholesterol micellization and the cholesterol uptake into Caco-2 cells was also determined. The content of total phenolics, flavonoid and anthocyanin in Prunus domestica L. (TPE), Antidesma bunius (L.) Spreng, Syzygium cumini (L.) Skeels, and Syzygium nervosum A. Cunn. Ex DC was 222.7–283.5 mg gallic acid equivalents, 91.2–184.3 mg catechin equivalents, and 37.9–49.5 mg cyanidin-3-glucoside equivalents/g extract, respectively. The anthocyanin-rich fraction of all extracts inhibited pancreatic lipase and cholesterol esterase with the IC50 values of 90.6–181.7 μg/mL and 288.7–455.0 μg/mL, respectively. Additionally, all extracts could bind primary and secondary bile acids (16.4–36.6%) and reduce the solubility of cholesterol in artificial micelles (53.0–67.6%). Interestingly, TPE was the most potent extract on interfering the key steps of lipid digestion among the tested extracts. In addition, TPE (0.10–0.50 mg/mL) significantly reduced the cholesterol uptake into Caco-2 cells in a concentration-dependent manner. These results demonstrate a new insight into the role of anthocyanin-rich Thai berry extract on interfering the key steps of lipid digestion and absorption.

Abstract The objective of the present study was to investigate the influences of dietary lipid composition on the gastrointestinal digestion and postprandial serum lipid profiles, and the connections between them. The in-vitro digestion results showed that maximum free fatty acid (FFA) release level of different lipid samples was PO (Palm oil) > RO (Rapeseed oil) > LINO (Linseed oil) > SO (Sunflower oil) > LO (Lard oil), and the first-order kinetics apparent rate constant was PO > SO ≈ RO > LO ≈ LINO, this may probably be ascribed to their specific lipid fatty acid composition and TAG structure. The individual FFA released during 240 min in-vitro digestion time was measured, and it showed that the release rate of short-chain saturated fatty acids (e.g. C16:0 in PO) were higher than the long-chain poly-unsaturated fatty acids (e.g. C18:3n-3 in LINO). Besides, the position of fatty acids within TAG molecules could also impose influences on the lipid hydrolysis process upon pancreas lipase in gastrointestinal tract using in-vitro digestion model. The postprandial serum fatty acid composition of the adult SD male rats were examined within 240 min after oral gavage administration, and the Pearson correlations between lipid fatty acid composition and the serum fatty acid profiles were analyzed. Certain correlations were summarized between lipid compositions (i.e. fatty acid composition and TAG structure), lipid digestion fates and serum fatty acid content in postprandial. The present work may provide some basic understandings of the connections among lipid compositions, lipid gastrointestinal digestion differences and the postprandial serum lipid profiles, and provide useful information about their nutritional and functional evaluation. Our recently published paper "Small Intestine Microbiota Regulate Digestive and Absorptive Adaptive Responses to Dietary Lipids" in Cell Host & Microbe explored the neglected small intestine microbiota and demonstrated its critical role as a regulator of fat digestion and absorption. This work generated the following important take home messages: 1) small intestinal microbes are particularly sensitive to high fat diets and turn on host processes regulating fat digestion and transport, 2) this action is very likely orchestrated by a consortium of microbes, each having different specific effects and targets, and 3) the actions of this consortium appear to be mediated by bacteria-derived small molecules or bioactive components. These findings are expected to provide insight into developing treatments for conditions of under- or over-nutrition. The goal of this addendum is to summarize our findings, address issues related to gut microbiota and gnotobiotic research specifically regarding technology and experimental design, discuss this work in the context of relevant literature, and lastly provide considerations for future research.
