Abstract Background Acer truncatum Bunge is an economic, ecological, oil, and medicinal tree, and its kernel oil is rich in nervonic acid. It is crucial to explore the transcriptional expression patterns of genes affecting fatty acid synthesis to improve the quality of Acer truncatum oil. Results This study used the seeds from high fatty acid strain YQC and those from low fatty acid strain Y38 as the test materials. Specifically, we performed a comparative transcriptome analysis of Y38 seeds and YQC to identify differentially expressed genes (DEGs) at two time points (seeds 30 days after the blooming period and 90 days after the blooming period). Compared with YQC_1 (YQC seeds at 30 days after the blooming period), a total of 3,618 DEGs were identified, including 2,333 up-regulated and 1,285 downregulated DEGs in Y38_1 (Y38 seeds at 30 days after blooming period). In the Y38_2 (Y38 seeds at 90 days after the blooming period) versus YQC_2 (YQC seeds at 90 days after the blooming period) comparison group, 9,340 genes were differentially expressed, including 5,422 up-regulated and 3,918 down-regulated genes. The number of DEGs in Y38 compared to YQC was significantly higher in the late stages of seed development. Gene functional enrichment analyses showed that the DEGs were mainly involved in the fatty acid biosynthesis pathway. And two fatty acid synthesis-related genes and seven nervonic acid synthesis-related genes were validated by qRT-PCR. Conclusions This study provides a basis for further research on biosynthesizing fatty acids and nervonic acidnervonic acids in A. truncatum seeds.

Abstract Background In addition to functioning as a precise monitoring mechanism in cell cycle, the anaphase-promoting complex/cyclosome (APC/C) is reported to be involved in regulating multiple metabolic processes by facilitating the ubiquitin-mediated degradation of key enzymes. Fatty acid oxidation is a metabolic pathway utilized by tumor cells that is crucial for malignant progression; however, its association with APC/C remains to be explored. Methods Cell cycle synchronization, immunoblotting, and propidium iodide staining were performed to investigate the carnitine palmitoyltransferase 1 C (CPT1C) expression manner. Proximity ligation assay and co-immunoprecipitation were performed to detect interactions between CPT1C and APC/C. Flow cytometry, 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2 H-tetrazolium, inner salt (MTS) assays, cell-scratch assays, and transwell assays and xenograft transplantation assays were performed to investigate the role of CPT1C in tumor progression in vitro and in vivo. Immunohistochemistry was performed on tumor tissue microarray to evaluate the expression levels of CPT1C and explore its potential clinical value. Results We identified CPT1C as a novel APC/C substrate. CPT1C protein levels exhibited cell cycle-dependent fluctuations, peaking at the G1/S boundary. Elevated CPT1C accelerated the G1/S transition, facilitating tumor cell proliferation in vitro and in vivo. Furthermore, CPT1C enhanced fatty acid utilization, upregulated ATP levels, and decreased reactive oxygen species levels, thereby favoring cell survival in a harsh metabolic environment. Clinically, high CPT1C expression correlated with poor survival in patients with esophageal squamous cell carcinoma. Conclusions Overall, our results revealed a novel interplay between fatty acid utilization and cell cycle machinery in tumor cells. Additionally, CPT1C promoted tumor cell proliferation and survival by augmenting cellular ATP levels and preserving redox homeostasis, particularly under metabolic stress. Therefore, CPT1C could be an independent prognostic indicator in esophageal squamous cell carcinoma.

Abstract Background Reintroduction represents an effective strategy for the conservation of endangered wildlife, yet it might inadvertently impact the native ecosystems. This investigation assesses the impact of reintroducing endangered Przewalski's horses into the desert grassland ecosystem of the Kalamaili Nature Reserve (KNR), particularly its effect on the spatial distribution of ticks. In a 25 km2 core area of Przewalski's horse distribution, we set up 441 tick sampling sites across diverse habitats, including water sources, donkey trails, and grasslands, recording horse feces and characteristics to analyze the occurrence rate of ticks. Additionally, we gathered the data of 669 fresh feces of horses. To evaluate the spatial dynamics between these feces and ticks, we used methods such as Fixed Kernel Estimation (FKE), Moran’s I spatial autocorrelation index, and Generalized Linear Models (GLM). Results The dominant species of ticks collected in the core area were adult Hyalomma asiaticum (91.36%). Their occurrence rate was higher near donkey trails (65.99%) and water sources (55.81%), particularly in areas with the fresh feces of Przewalski's horses. The ticks’ three risk areas, as defined by FKE, showed significant overlap and positive correlation with the distribution of Przewalski's horses, with respective overlap rates being 90.25% in high risk, 33.79% in medium risk, and 23.09% in low risk areas. Moran's I analysis revealed a clustering trend of the fresh feces of Przewalski's horses in these areas. The GLM confirmed a positive correlation between the distribution of H. asiaticum and the presence of horse fresh feces, alongside a negative correlation with the proximity to water sources and donkey trails. Conclusions This study reveals the strong spatial correlation between Przewalski's horses and H. asiaticum in desert grasslands, underlining the need to consider interspecific interactions in wildlife reintroductions. The findings are crucial for shaping effective strategies of wildlife conservation and maintaining ecological balance.

Objective: A non-invasive predictive model was developed using radiomic features to forecast CXCL9 expression level in breast cancer patients. Methods: CXCL9 expression data and MRI images of breast cancer patients were obtained from The Cancer Genome Atlas (TCGA) and The Cancer Imaging Archive (TCIA) databases, respectively. Local tissue samples from 20 breast cancer patients were collected to measure CXCL9 expression levels. Radiomic features were extracted from MRI images using 3DSlicer, and the minimum Redundancy Maximum Relevance and Recursive Feature Elimination (mRMR_RFE) method was employed to select the most pertinent radiomic features associated with CXCL9 expression levels. Support vector machine (SVM) and Logistic Regression (LR) models were utilized to construct the predictive model, and the area under the receiver operating characteristic curve (AUC) was calculated for performance evaluation. Results: CXCL9 was found to be upregulated in breast cancer patients and linked to breast cancer prognosis. Nine radiomic features were ultimately selected using the mRMR_RFE method, and SVM and LR models were trained and validated. The SVM model achieved AUC values of 0.748 and 0.711 in the training and validation sets, respectively. The LR model obtained AUC values of 0.771 and 0.724 in the training and validation sets, respectively. Conclusion: The utilization of MRI radiomic features for predicting CXCL9 expression level provides a novel non-invasive approach for breast cancer Prognostic research.

Gut microbiome studies based on fecal samples can non-invasively reflect the survival and health status of endangered species. This study presents the first characterization of the gut microbiome in the semi-wild population of the critically endangered Saiga antelope (Saiga tatarica), unveiling the existence of two distinct enterotypes within the semi-wild Saiga: the Oscillospiraceae UCG-005 enterotype and the Arthrobacter enterotype. These enterotypes show significant differences in microbiome diversity, composition, and predicted functions. The Arthrobacter enterotype has lower alpha diversity, fewer short-chain fatty acids (SCFAs)-producing bacteria, more potential pathogens, lower energy metabolism and higher lipid metabolism abundance. The findings imply that Saiga with the Arthrobacter enterotype have lower energy utilization efficiency and higher disease risks, particularly for obesity. Microbial correlation network analysis indicates SCFAs-producing bacteria play a key role in the formation of these enterotypes. This suggests that next-generation probiotics, primarily composed of SCFAs-producing bacteria, could be used to regulate and improve the health of the Saiga. Additionally, the similarity of the enterotype associated with a healthier survival state to that of wild counterparts suggests that incorporating gut microbiome monitoring into the key indicators for the reintroduction of endangered species could be significant for wildlife conservation and management.

Microwave heating has attracted interests in many industrial applications, yet heating inhomogeneity remains a critical issue. This study proposes a novel approach to enhance microwave heating uniformity employing a switchable frequency selective surface (FSS). The PIN diodes in each FSS unit cell can be toggled ON or OFF by controlling voltage biases, enabling the FSS to either reflect or transmit incident electromagnetic waves. Placing this switchable FSS in front of a heating cavity wall creates a dynamic reflecting boundary, which, through periodic alterations of the FSS and metallic wall, effectively improves the heating uniformity. To validate this method, we simulated the heating of a potato sample at 2450 MHz using COMSOL Multiphysics and conducted experimental tests. Both simulation and experimental results demonstrate that the proposed method significantly enhances microwave heating uniformity across various materials and sample sizes. This work provides a promising solution for designing microwave heating reactors.

Background: When marathon runners break the 2-h barrier at the finishing line, it attracts global attention. This study is aimed to conduct a bibliometric analysis of publications in the field of marathon running, analyze relevant research contributors, and visualize the historical trends of marathon performance research over the past 15 years.Methods: On 8 December 2023, we extracted high-quality publication data from the Web of Science Core Collection spanning from 1 January 2009 to 30 November 2023. We conducted bibliometric analysis and research history visualization using the R language packages biblioshiny, VOSviewer, and CiteSpace.Results: A total of 1,057 studies were published by 3,947 authors from 1,566 institutions across 63 countries/regions. USA has the highest publication and citation volume, while, the University of Zurich being the most prolific research institution. Keywords analysis revealed several hotspots in marathon research over the past 3 years: (1) physiology of the elite marathon runners, (2) elite marathon training intensity and pacing strategies, (3) nutritional strategies for elite marathon runners, (4) age and sex differences in marathon performance, (5) recovery of inflammatory response and muscle damage.Conclusion: This study presents the first comprehensive bibliometric analysis of marathon performance research over the past 15 years. It unveils the key contributors to marathon performance research, visually represents the historical developments in the field, and highlights the recent topical frontiers. The findings of this study will guide future research by identifying potential hotspots and frontiers.

The CCCH zinc finger proteins play critical roles in a wide variety of growth, development, and stress responses. Currently, limited reports are available about the roles of animal CCCH proteins in plants. In this study, we report the identification of human TTP (hTTP) with functional similarity to PdC3H17 in a hybrid poplar. hTTP and PdC3H17 shared highly similar tandem CCCH zinc-finger RNA-binding domains. The fragments excluding the CCCH domain of both hTTP and PdC3H17 possessed transcriptional activation activities in yeast cells. Compared to the controls, ectopic expression of hTTP in poplar caused dwarfism, and resulted in significant increases in stem xylem vessel number and photosynthetic and ROS-scavenging abilities, thereby enhancing plant tolerance to drought stress. Our results suggest that hTTP may perform a function in poplar through the PdC3H17-mediated system, and provide an example for the application of animal genes in plants.

Uncertainty quantification (UQ) in polynomial chaos expansion (PCE) suffers from the curse of dimensionality, which is fundamentally reflected in that the number of the PCE coefficients to be found grows rapidly as the number of random inputs increases, and thus the number of samples required increases dramatically. Regardless of the approach taken to alleviate this disaster, we are usually faced with the technical challenge of automatically and programmatically determining the basis functions during the PCE implementation, which are composed of a large number of multivariate polynomials. To address this problem, this paper proposes an algorithm based on the recursive idea for automatically determining the basis functions, and then combines this algorithm with the latest weight-based regression called least squares polynomial chaos regression (LSPCR), develops the six algorithms for the LSPCR in the medium- and high-dimensional cases, and evaluates their performance in the electromagnetic coupling problem of the transmission lines (TLs) in this case. Subsequently, the performance of six algorithms for LSPCR are compared in the medium- and high-dimensional, low-order cases, each algorithm consisting of a sampling strategy selected from asymptotic sampling (AS), standard sampling (SS), and coherence-optimal sampling (COS), paired with a norm problem chosen from either least squares optimization (LSO) or $\ell _{\mathbf {1}}$ -minimization ( $\ell _{\mathbf {1}}$ -M) problems. Numerical experiments demonstrate the excellence of the proposed algorithm. Furthermore, the results also show that only the algorithms that employ the SS (SSs) and the algorithms based on the COS (COSs) require no more than 1.4% of the total Monte Carlo (MC) computation time when producing similarly accurate results to the MC, regardless of the chosen norm problem.

A coverage prediction model helps network operators find coverage gaps, plan base station locations, evaluate quality of service, and build radio maps for spectrum sharing, interference management, localization, etc. Existing coverage prediction models rely on the height and transmission power of the base station, or the assistance of a path loss model. All of these increase the complexity of large-scale coverage predictions. In this paper, we propose a multi-modal model, DNN-SS, which combines a DNN (deep neural network) and SS (semantic segmentation) to perform coverage prediction for mobile networks. Firstly, DNN-SS filters the samples with a geospatial-temporal moving average filter algorithm, and then uses a DNN to extract numerical features. Secondly, a pre-trained model is used to perform semantic segmentation of satellite images of the measurement area. Thirdly, a DNN is used to extract features from the results after semantic segmentation to form environmental features. Finally, the prediction model is trained on the dataset consisting of numerical features and environmental features. The experimental results on campus show that for random location prediction, the model achieves a RMSE (Root Mean Square Error) of 1.97 dB and a MAE (Mean Absolute Error) of 1.41 dB, which is an improvement of 10.86% and 10.2%, respectively, compared with existing models. For the prediction of a test area, the RMSE and MAE of the model are 4.32 dB and 3.45 dB, respectively, and the RMSE is only 0.22 dB lower than that of existing models. However, the DNN-SS model does not need the height, transmission power, and antenna gain of the base station, or a path loss model, which makes it more suitable for large-scale coverage prediction.

This paper presents two sensitivity analyses, one of which is for the time-domain transmission line (TL) model using the latest weight-based algorithm that is a combination of the coherence-optimal sampling (COS) and the l1-minimum problem (l1-M) and the other is for the high-frequency field line coupling model based on the algorithm that combines the Latin hypercube sampling (LHS) with the l1-M for the first time, where the COS and the LHS are employed to draw samples and the polynomial chaos expansion (PCE) coefficients are obtained by solving the l1-M. Both algorithms are within the PCE framework. The results show that for the time-domain TL model, the capacitance has the greatest impact on the voltage response, followed by the resistance. The height and the radius have the least impact, while there is little difference between them. For the high-frequency field line coupling model, the azimuth angle has the greatest impact on the current in the TL, followed by the elevation angle. The terminal load, the conductivity, the field amplitude, the relative permittivity, and the TL radius have the least effect, and there is little difference among them. Additionally, both algorithms only require no more than 0.60% of the total Monte Carlo (MC) computation time when yielding solutions with almost identical accuracy. Compared to performing a sensitivity analysis using the MC method, the use of two algorithms significantly improves efficiency.

Abstract The consequences of sub-lethal levels of ambient air pollution are underestimated for insects, for example, the accumulation of particulate matter on sensory receptors located on their antennae may have detrimental effects to their function. Here we show that the density of particulate matter on the antennae of houseflies (Musca domestica) collected from an urban environment increases with the severity of air pollution. A combination of behavioural assays, electroantennograms and transcriptomic analysis provide consistent evidence that a brief exposure to particulate matter pollution compromises olfactory perception of reproductive and food odours in both male and female houseflies. Since particulate matter can be transported thousands of kilometres from its origin, these effects may represent an additional factor responsible for global declines in insect numbers, even in pristine and remote areas.

Abstract Background The accumulation of fatty acids in plants covers a wide range of functions in plant physiology and thereby affects adaptations and characteristics of species. As the famous woody oilseed crop, Acer truncatum accumulates unsaturated fatty acids and could serve as the model to understand the regulation and trait formation in oil-accumulation crops. Here, we performed Ribosome footprint profiling combing with a multi-omics strategy towards vital time points during seed development, and finally constructed systematic profiling from transcription to proteomes. Additionally, we characterized the small open reading frames (ORFs) and revealed that the translational efficiencies of focused genes were highly influenced by their sequence features. Results The comprehensive multi-omics analysis of lipid metabolism was conducted in A. truncatum. We applied the Ribo-seq and RNA-seq techniques, and the analyses of transcriptional and translational profiles of seeds collected at 85 and 115 DAF were compared. Key members of biosynthesis-related structural genes (LACS, FAD2, FAD3, and KCS) were characterized fully. More meaningfully, the regulators (MYB, ABI, bZIP, and Dof) were identified and revealed to affect lipid biosynthesis via post-translational regulations. The translational features results showed that translation efficiency tended to be lower for the genes with a translated uORF than for the genes with a non-translated uORF. They provide new insights into the global mechanisms underlying the developmental regulation of lipid metabolism. Conclusions We performed Ribosome footprint profiling combing with a multi-omics strategy in A. truncatum seed development, which provides an example of the use of Ribosome footprint profiling in deciphering the complex regulation network and will be useful for elucidating the metabolism of A. truncatum seed oil and the regulatory mechanisms.

In satellite remote sensing (SRS), there is a demand for large-power microwave components. A Butler matrix is essential to a transmitting antenna array in SRS. This article illustrates the electrical and mechanical design, simulation, and test results of a large-power planar beamforming network for SRS at C-band. It is a 4 × 4 Butler matrix based on square coaxial lines. Short-ended stubs are used in the Butler matrix to broaden its bandwidth by 10%, support inner conductors, and enhance heat transfer in vacuum environments. The simulation results are consistent with the measured results. The reflection coefficient is less than −18 dB, and the isolation is more than 23 dB from 3.8 GHz to 4.2 GHz. The insertion losses are less than 0.6 dB, and the phase errors are better than ±6°. The measured peak microwave power of the proposed Butler matrix is 9 kW. Its size is 440 × 400 × 40 mm3. The proposed Butler matrix beamforming network can be applied to SRS systems.

Hepatitis-hydropericardium syndrome (HHS), caused by fowl adenovirus serotype 4 (FAdV-4), has been widely spread across China, resulting in great financial losses in the poultry industry. Therefore, efficient vaccines against this disease urgently need to be developed. In our study, the fiber-2 and penton base proteins derived from the FAdV-4 JS strain were expressed in a prokaryotic system (E. coli) in a soluble form. Then, the efficacy of the two recombinant proteins formulated with cheap and widely used adjuvants (Marcol™ 52 white oil) were respectively tested, and the minimum immune doses and safety of the above proteins were also determined. It was indicated that the fiber-2 (20 µg/bird, 200 µg/bird) and penton base (200 µg/bird) could provide complete protection against the highly pathogenic FAdV-4 and suppress its replication and shedding. Unfortunately, only the fiber-2 protein could induce complete protection (10/10) at a low dose (10 µg/bird). In addition, we confirmed that the fiber-2 subunit vaccine formulated with oil adjuvants was safe for vaccinated chickens. Conclusively, all of our results suggest that we successfully prepared an efficient and cheap fiber-2 subunit vaccine with few side effects.

Abstract Objectives Our aim was to assess the ability of the Whole-genome sequencing (WGS) in predicting drug resistance profile of multidrug-resistant mycobacterium tuberculosis (MDR-MTB) from newly diagnosed cases in China. Methods We validated the Phenotypic drug Sensitivity Test (pDST) for 12 anti-tuberculosis drugs using the Bactec MGIT 960 system. We described the characteristics of the isolates enrolled and compared the pDST results with resistance profiles predicted by WGS. Results The pDST showed that of the 43 isolates enrolled, 25.6% were sensitive to rifabutin (RFB); 97.7%、97.7%、93.0% and 93.0% were sensitive to cycloserine (Cs), amikacin/kanamycin (Ak/Km), para-aminosalicylic acid (Pas) and ethionamide Eto), respectively; 18.6% were resistant to fluoroquinolones (FQs) or second-line injections. Genotype DST determined by WGS of Ak/Km、Eto and RFP reached high consistency to 97.7% compared with pDST, followed by moxifloxacin (Mfx) 95.3%, levofloxaci (Lfx) and Pas 93%, streptomycin (Sm) 90.3%. The genotype DST of RFB and EMB showed low consistency with the pDST of 67.2 and 79.1%. WGS also detected 27.9% isolates of pyrazinamide(PZA)-related drug-resistant mutation. No mutations associated with linezolid (Lzd), bedaquiline (Bdq) and clofazimine (Cfz) were detectd. Conclusions WGS has the potential to infer resistance profiles without time-consuming phenotypic methods, which could be provide a basis to formulate reasonable treatment in high TB burden areas.

Traffic congestion has been a hot topic of research in the field of intelligent transportation, which can be alleviated by efficient route navigation. Most of the existing route planning methods are non-negotiated algorithms, which do not take into account the route conflicts and collaborative relationships between multiple vehicles. Also, most negotiated algorithms have not been comprehensively considered dynamic route collaboration between vehicles, large-scale efficient computation, environmental pollution, etc. Therefore, an ecological multivehicle real-time route selection model (EMR2SM) for urban road networks is firstly proposed in this paper, which combines real-time traffic conditions of the road network with travel time, distance, and exhaust emissions as optimization indicators. In order to solve the large-scale computation problem of traditional negotiated algorithms, an adaptive multiswarm bee colony (AMSBC) algorithm is designed, which efficiently solves the multivehicle dynamic route selection problem. AMSBC searches the optimal route for each vehicle in parallel through multiple population division and self-adaption mechanism, to make multivehicle route selection reach Nash equilibrium. Compared with three non-negotiated optimization algorithms based on swarm technology, EMR2SM is verified by experiments that it improves the efficiency and accuracy of the optimal route selection for multiple vehicles and reduces vehicle emissions, which can effectively reduce traffic congestion and environmental pollution.

Taxi sharing is a promising method to save resource consumption and alleviate traffic congestion while satisfying people’s commuting needs. Existing research methods include taxi dispatching methods based on intelligent algorithms, single vehicle route recommendation algorithms, and route recommendation algorithms based on vehicle traffic history. However, these studies either focus on how to efficiently dispatch satisfactory vehicles for passengers, ignoring the effect of efficient routes on vehicle travel efficiency, or let vehicles follow the shortest detour distance recommended by the system, ignoring the traffic congestion caused by the influx of vehicles into the same road. To address the abovementioned problems, a noncooperative game for a taxi sharing model (NCG-TSM) in urban road networks is proposed in this paper combined with the traffic conditions of the optional routes, and a distribution estimation algorithm for the shared taxi game is designed to make multivehicle route selections reach Nash equilibrium. The effectiveness of NCG-TSM is verified through simulation experiments. When the number of vehicles reaches the congestion capacity of the road segment, compared to the three common frameworks, the travel time cost and fuel consumption cost can be reduced by 5.8% to 9.1% and 3.5% to 8.9%, respectively. Besides, the occupancy rate has been improved, especially compared to the BMP framework, by 5.5% to 40%.

This paper mainly studies the multiclass stochastic user equilibrium problem considering the market share of battery electric vehicles (BEVs) and random charging behavior (RCB) in a mixed transport network containing electric vehicles and gasoline vehicles (GVs). In order to analyze the random charging and path choice behaviors of BEV users and extract the differences in travel behaviors between BEV and GV users, an improved logit-based model, multilabel algorithm, and queuing theory are applied. The influencing factors of charging possibility mainly include the initial state of charge (SOC), the SOC at the beginning of charging, and the psychologically acceptable safe SOC threshold arriving at the destination. Diversity choices of user paths and charging locations will result in changes in queuing traffic and differences in queuing time. Conversely, different stations have different queuing dwell times, which will also affect the routing and charging locations for BEVs with RCB. The path-based method of successive averages (MSA) is adopted to solve the model. Through the simulation of the test network Sioux Falls, the equilibrium traffic flow and possible charging flow under different market shares and initial SOC are predicted, and the properties of the model and the feasibility of the algorithm are verified.

Myiasis is an infestation of maggots on living tissue in humans and animals all over the world. It is known to occur in wild animals, while no information is reported in forest musk deer (Moschus berezovskii). During our research on the conservation of forest musk deer, we found a new record of traumatic myiasis of an injured forest musk deer. The flies are likely Lucilia caesar (Linnaeus, 1758) according to the results of DNA barcoding technology. We report traumatic myiasis of forest musk deer for the first time, which expands the information on parasite and myiasis of forest musk deer and confirms the potential risk of traumatic myiasis of forest musk deer.

Abstract Tachinidae are one of the most speciose families of Diptera and the largest group of non-hymenopteran parasitoids. Little is known about their diversity, distribution patterns, and seasonal variation in most ecosystems. This study reports on tachinid flies collected by a Malaise trap over 73 weeks in Baihua Mountain Reserve, northern China, and investigates the patterns of local species richness and its temporal distribution. The most species-rich season was summer, but the majority of specimens were recovered in spring. A total of 755 tachinid specimens were collected, consisting of 144 species in 85 genera, comprising 26.5% of the species and 49.7% of the genera recorded from northern China. A total species richness of 243 was estimated, indicating that only a portion of the community of tachinid flies was collected at this location and suggesting that the diversity of tachinids might be underestimated across Beijing and northern China. This work is a first step in assessing patterns of tachinid diversity in China using quantitative sampling and establishes a baseline for comprehending the temporal and spatial diversity of these ecologically significant parasitoids.

Proprotein convertase subtilisin kexin type 9 (PCSK9) inhibits the clearance of low-density lipoprotein (LDL) cholesterol (LDL-C) from plasma by directly binding with the LDL receptor (LDLR) and sending the receptor for lysosomal degradation. As the interaction promotes elevated plasma LDL-C levels, and therefore a predisposition to cardiovascular disease, PCSK9 has attracted intense interest as a therapeutic target. Despite this interest, an orally bioavailable small-molecule inhibitor of PCSK9 with extensive lipid-lowering activity is yet to enter the clinic. We report herein the discovery of NYX-PCSK9i, an orally bioavailable small-molecule inhibitor of PCSK9 with significant cholesterol-lowering activity in hyperlipidemic APOE∗3-Leiden.CETP mice. NYX-PCSK9i emerged from a medicinal chemistry campaign demonstrating potent disruption of the PCSK9-LDLR interaction in vitro and functional protection of the LDLR of human lymphocytes from PCSK9-directed degradation ex vivo. APOE∗3-Leiden.CETP mice orally treated with NYX-PCSK9i demonstrated a dose-dependent decrease in plasma total cholesterol of up to 57%, while its combination with atorvastatin additively suppressed plasma total cholesterol levels. Importantly, the majority of cholesterol lowering by NYX-PCSK9i was in non-HDL fractions. A concomitant increase in total plasma PCSK9 levels and significant increase in hepatic LDLR protein expression strongly indicated on-target function by NYX-PCSK9i. Determinations of hepatic lipid and fecal cholesterol content demonstrated depletion of liver cholesteryl esters and promotion of fecal cholesterol elimination with NYX-PCSK9i treatment. All measured in vivo biomarkers of health indicate that NYX-PCSK9i has a good safety profile. NYX-PCSK9i is a potential new therapy for hypercholesterolemia with the capacity to further enhance the lipid-lowering activities of statins.

In the year of 2016 to 2017, record-breaking marine heatwave (MHW) events occurred along the Chinese coast, exerting severe impacts on coastal fishes and fisheries. However, the response of the nekton community to MHW events still remains poorly understanding. Thus, to investigate the influences of the MHW events on species composition dynamics, this study conducted a survey on nekton communities in the northern East China Sea in the summers during 2014 to 2017. The recently-developed “heatwaveR” package was adopted to describe the variation characteristics of heatwaves within the study area, based on high-resolution remote-sensing sea surface temperature products. We also compared variations in the community structures of different biological groups before (2014–2015) and during the occurrence of the MHW events (2016–2017). During the MHW events, temperature increased by approximately 2.3°C in both 2016 and 2017. Compared with those before the MHW events, the biomass and dominance of several important crustaceans decreased, and the biomass of warm water affinity species increased significantly, while their richness and diversity decreased; the biomass of warm temperature affinity species declined significantly, while their diversity increased slightly. These results show the difference in the response rates of species with different thermal preferences and different species compositions to the extreme climate change. Beta diversity analysis revealed that the inter-group variation before and during the MHW event exceeded the intra-group variation, proving that MHWs changed the community structure to a certain extent. PERMANOVA test showed that the species compositions of planktivorous fish (FG1) and small-omnivorous fish (FG2) in the feeding-functional groups were significantly different before and during the MHW event, which may be attributed to the impact of MHWs on oligotrophic fishes through thermal preference and the trophic cascade effect. There was no significant change in the species compositions of other functional groups, indicating that medium-trophic and eutrophic fishes possessed certain adaptability to MHWs. Regarding fisheries management under extreme warming events, it would be advisable to diagnose temperature anomalies in the early stage, monitor changes in species abundance in the early life cycle, and then rapidly develop suitable fishing strategies.

Metformin is widely used to surmount insulin resistance (IR) and type 2 diabetes. Accumulating evidence suggests that metformin may improve IR through regulating gut microbiota and bile acids. However, the underlying mechanisms remain unclear. Our metabolomic analysis showed that metformin significantly increased the accumulation of tauroursodeoxycholic acid (TUDCA) in intestine and liver from high-fat diet (HFD)-induced IR mice. TUDCA also alleviated IR, and reduced oxidative stress and intestinal inflammation in ob/ob mice. TUDCA blocked KEAP1 to bind with Nrf2, resulting in Nrf2 translocation into nuclear and initiating the transcription of antioxidant genes, which eventually reduced intracellular ROS accumulation and improved insulin signaling. Analysis of gut microbiota further revealed that metformin reduced the relative abundance of Bifidobacterium, which produces bile salt hydrolase (BSH). The reduction in BSH was probably crucial for the accumulation of TUDCA. Metformin also increased the proportion of Akkermanisia muciniphlia in gut microbiota of ob/ob mice via TUDCA. These beneficial effects of metformin in remodeling gut microbiota, reducing oxidative stress and improving insulin sensitivity were partly due to the accumulation of TUDCA, suggesting that TUDCA may be a potential therapy for metabolic syndrome.

The mitochondrial genome of Anthomyia illocata Walker, 1857 belonging to the Anthomyiidae, was obtained using a next-generation sequencing approach. This 16,236 bp complete mitogenome consists of 13 protein-coding, two ribosomal RNA, and 22 transfer RNA genes, as well as a non-coding control region. The Anthomyiidae are reconstructed as a paraphyletic group, with the genera Pegomya recovered as a sister group of the Scathophagidae.

Abstract Background Blowflies are ubiquitous insects, often shiny and metallic, and the larvae of many species provide important ecosystem services (e.g., recycling carrion) and are used in forensics and debridement therapy. Yet, the taxon has repeatedly been recovered to be para- or polyphyletic, and the lack of a well-corroborated phylogeny has prevented a robust classification. Results We here resolve the relationships between the different blowfly subclades by including all recognized subfamilies in a phylogenomic analysis using 2221 single-copy nuclear protein-coding genes of Diptera. Maximum likelihood (ML), maximum parsimony (MP), and coalescent-based phylogeny reconstructions all support the same relationships for the full data set. Based on this backbone phylogeny, blowflies are redefined as the most inclusive monophylum within the superfamily Oestroidea not containing Mesembrinellidae, Mystacinobiidae, Oestridae, Polleniidae, Sarcophagidae, Tachinidae, and Ulurumyiidae. The constituent subfamilies are re-classified as Ameniinae (including the Helicoboscinae, syn. nov.), Bengaliinae, Calliphorinae (including Aphyssurinae, syn. nov., Melanomyinae, syn. nov., and Toxotarsinae, syn. nov.), Chrysomyinae, Luciliinae, Phumosiinae, Rhiniinae stat. rev., and Rhinophorinae stat. rev. Metallic coloration in the adult is shown to be widespread but does not emerge as the most likely ground plan feature. Conclusions Our study provides the first phylogeny of oestroid calyptrates including all blowfly subfamilies. This allows settling a long-lasting controversy in Diptera by redefining blowflies as a well-supported monophylum, and blowfly classification is adjusted accordingly. The archetypical blowfly trait of carrion-feeding maggots most likely evolved twice, and the metallic color may not belong to the blowfly ground plan.

The global epidemic has been controlled to some extent, while sporadic outbreaks still occur in some places. It is essential to summarize the successful experience and promote the development of new drugs. This study aimed to explore the common mechanism of action of the four Chinese patent medicine (CPMs) recommended in the Medical Observation Period COVID-19 Diagnostic and Treatment Protocol and to accelerate the new drug development process. Firstly, the active ingredients and targets of the four CPMs were obtained by the Chinese medicine composition database (TCMSP, TCMID) and related literature, and the common action targets of the four TCMs were sorted out. Secondly, the targets of COVID-19 were obtained through the gene-disease database (GeneCards, NCBI). Then the Venn diagram was used to intersect the common drug targets with the disease targets. And GO and KEGG pathway functional enrichment analysis was performed on the intersected targets with the help of the R package. Finally, the results were further validated by molecular docking and molecular dynamics analysis. As a result, a total of 101 common active ingredients and 21 key active ingredients of four CPMs were obtained, including quercetin, luteolin, acacetin, kaempferol, baicalein, naringenin, artemisinin, aloe-emodin, which might be medicinal substances for the treatment of COVID-19. TNF, IL6, IL1B, CXCL8, CCL2, IL2, IL4, ICAM1, IFNG, and IL10 has been predicted as key targets. 397 GO biological functions and 166 KEGG signaling pathways were obtained. The former was mainly enriched in regulating apoptosis, inflammatory response, and T cell activation. The latter, with 92 entries related to COVID-19, was mainly enriched to signaling pathways such as Coronavirus disease—COVID-19, Cytokine-cytokine receptor interaction, IL-17 signaling pathway, and Toll-like receptor signaling pathway. Molecular docking results showed that 19/21 of key active ingredients exhibited strong binding activity to recognized COVID-19-related targets (3CL of SARS-CoV-2, ACE2, and S protein), even better than one of these four antiviral drugs. Among them, shinflavanone had better affinity to 3CL, ACE2, and S protein of SARS-CoV-2 than these four antiviral drugs. In summary, the four CPMs may play a role in the treatment of COVID-19 by binding flavonoids such as quercetin, luteolin, and acacetin to target proteins such as ACE2, 3CLpro, and S protein and acting on TNF, IL6, IL1B, CXCL8, and other targets to participate in broad-spectrum antiviral, immunomodulatory and inflammatory responses.

Backward-in-time Lagrangian model can identify potential spawning areas by reconstructing egg drift trajectories, contributing to accurately designing potential priority conservation plans for spawning areas. In this study, we apply this approach to investigate the small yellow croaker (Larimichthys polyactis) with commercial value in China. A two-step spatial random forest (RF) model is used to predict the occurrence probability and abundance of their eggs and describe the optimal ecological range of environmental factors. A priority protection index (BPPI) of the spawning areas is established by combining the sites with the optimal occurrence and abundance and integrating backward tracking pathways. The result indicates that the model with 1-2 day time lags of environmental variables shows the optimum explanatory power. Temperature and salinity are the most important factors affecting oogenesis and show a regime shift in the response curve. They reflect the physiological regulation of parental sexual maturation by the environment. In addition, egg abundance correlates more strongly with chlorophyll-a (Chl a) concentration and depth, suggesting that parents prefer environments with shallow water and high prey density for spawning activities. The egg retrieval shows that the potential spawning sources are distributed near the southeastern part of the oogenesis site, with a maximum egg dispersal distance of no more than 30 km. This finding confirms that the coastal regions of Jiangsu Province are an important spawning ground for the small yellow croaker, making a significant contribution to the productivity and resilience of the fish.

Forest musk deer (Moschus berezovskii) are endangered ruminants whose adult males secrete musk. China has been breeding forest musk deer artificially since the 1950s in an effort to restore wild populations, with Shaanxi and Sichuan provinces as the two main sites for captive breeding. Genetic diversity is a significant indicator that determines the long-term viability and status of a population, particularly for species at risk of extinction. In this study, we analyzed the current genetic makeup of seven captive forest musk deer populations in the Shaanxi province, using the mitochondrial DNA (mtDNA) control region (CR) as the molecular marker. We sequenced 604 bp of mtDNA CR, with an average content of A+T higher than G+C. We observed 111 variable sites and 39 different haplotypes from 338 sequences. The nucleotide diversity (Pi) and haplotype diversity (Hd) were 0.02887 and 0.908, respectively. Genetic differentiation between these populations was not significant, and the populations might not have experienced rapid growth. By combining our sequences with previous ones, we identified 65 unique haplotypes with 26 rare haplotypes and estimated a total of 90 haplotypes in Shaanxi province captive populations. The Shaanxi province and Sichuan province obtained 88 haplotypes, the haplotypes from the two populations were mixed together, and the two populations showed moderate genetic differentiation. Our findings suggested that captive forest musk deer populations in the Shaanxi province had high genetic diversity, with a rich founder population of about 90 maternal lines. Additionally, managers could develop genetic management plans for forest musk deer based on the haplotype database. Overall, our study will provide insights and guidelines for the conservation of genetic diversity in captive forest musk deer populations in the Shaanxi province.

Abstract Background The effect of time delay from diagnosis to surgery on the prognosis of elderly patients with liver cancer is not well known. We investigated the effect of surgical timing on the prognosis of elderly hepatocellular carcinoma patients undergoing surgical resection and constructed a Nomogram model to predict the overall survival of patients. Methods A retrospective analysis was performed on elderly patients with primary liver cancer after hepatectomy from 2012 to 2018. The effect of surgical timing on the prognosis of elderly patients with liver cancer was analyzed using the cut-off times of 18 days, 30 days, and 60 days. Cox was used to analyze the independent influencing factors of overall survival in patients, and a prognostic model was constructed. Results A total of 232 elderly hepatocellular carcinoma patients who underwent hepatectomy were enrolled in this study. The cut-off times of 18, 30, and 60 days were used. The duration of surgery had no significant effect on overall survival. Body Mass Index, Child-Pugh classification, Tumor size Max, and Length of stay were independent influencing factors for overall survival in the elderly Liver cancer patients after surgery. These factors combined with Liver cirrhosis and Venous tumor emboli were incorporated into a Nomogram. The nomogram was validated using the clinical data of the study patients, and exhibited better prediction for 1-year, 3-year, and 5-year overall survival. Conclusions We demonstrated that the operative time has no significant effect on delayed operation in the elderly patients with hepatocellular carcinoma, and a moderate delay may benefit some patients. The constructed Nomogram model is a good predictor of overall survival in elderly patients with hepatectomy.

Gastric ulcer (GU) is one of the most commonly diagnosed diseases worldwide, threatening human health and seriously affecting quality of life. Reports have shown that the Chinese herbal medicine Sarcandra glabra (Thunb.) Nakai (SGN) can treat GU. However, its pharmacological effects deserve further validation; in addition, its mechanism of action is unclear. An acute gastric ulcer (AGU) rat model induced by alcohol was used to evaluate the gastroprotective effect of SGN by analysis of the histopathological changes in stomach tissue and related cytokine levels; the potential mechanisms of action of SGN were investigated via serum metabolomics and network pharmacology. Differential metabolites of rat serum were identified by metabolomics and the metabolic pathways of the identified metabolites were enriched via MetaboAnalyst. Furthermore, the critical ingredients and candidate targets of SGN anti-AGU were elucidated. A compound-reaction-enzyme-gene network was established using Cytoscape version 3.8.2 based on integrated analysis of metabolomics and network pharmacology. Finally, molecular docking was applied to verify the acquired key targets. The results showed that SGN exerted a certain gastroprotective effect via multiple pathways and targets. The effects of SGN were mainly caused by the key active ingredients isofraxidin, rosmarinic, and caffeic acid, which regulate hub targets, such as PTGS2, MAPK1, and KDR, which maintain the homeostasis of related metabolites. Signal pathways involved energy metabolism as well as immune and amino acid metabolism. Overall, the multi-omics techniques were proven to be promising tools in illuminating the mechanism of action of SGN in protecting against diseases. This integrated strategy provides a basis for further research and clinical application of SGN.

To explore how the living environment influences the establishment of gut microbiota in different species, as well as the extent to which changes in the living environment caused by captive breeding affect wildlife’s gut microbiota and health, we used 16S rRNA gene amplicon sequencing and shotgun metagenomic sequencing to compare the gut microbiome of two species of threatened equids, the Przewalski’s Horse and the Asian wild ass, in the wild and captivity. The results revealed that different species of Equidae living in the same environment showed remarkable convergence of gut microflora. At the same time, captive populations exhibited significantly “unhealthy” microbiota, such as low Alpha diversity, high levels of potentially pathogenic bacteria and biomarkers of physical or psychological disease, and enrichment of microbial functions associated with exogenous exposure and susceptibility, implying that the artificial environment created by captivity may adversely impact the health of wildlife to some extent. Our findings demonstrate the importance of the environmental factors for the establishment of gut microbiota and host health and provide new insights into the conservation of wildlife in captivity from the perspective of the microbiome.

The bat fly genus Ascodipteron Adensamer, 1896 currently contains 15 species, all of which occur in tropical and subtropical areas of the Eastern Hemisphere. A new species of endoparasitic bat fly, Ascodipteron sanmingensis sp. nov., was collected from the Great Himalayan Leaf-nosed Bat, Hipposideros armiger (Hodgson, 1853), during ecological studies on bats in Fujian, China.A new species, Ascodipteron sanmingensis sp. nov., is described based on dealate neosomic females and is supported by molecular data from a 368 bp fragment of the cytochrome B (Cytb) gene. Habitus and diagnostic details as well as the attachment sites on the host are documented with photographs. A detailed comparison of the new species with related species is provided and the new species is accommodated in the most recent key to the world species of Ascodipteron.

Abstract Background Rapid identification of pathogenic Mycobacterium species is critical for a successful treatment. However, traditional method is time-consuming and cannot discriminate isolated non-tuberculosis mycobacteria (NTM) at species level. In the retrospective study, we evaluated the clinical applicability of PCR-reverse blot hybridization assay (PCR-REBA Myco-ID) with clinical specimens for rapid detection and differentiation of mycobacterial species. Methods A total of 334 sputum and 362 bronchial alveolar lavage fluids (BALF) from 696 patients with mycobacterium pulmonary disease (MPD) and 210 patients with non-mycobacterium pulmonary disease used as controls were analyzed. Sputum or BALF were obtained for MGIT 960-TBc ID test and PCR-REBA Myco-ID assay. High resolution melt analysis (HRM) was used to resolve inconsistent results of MGIT 960-TBc ID test and PCR-REBA Myco-ID assay. Results A total of 334 sputum and 362 BALF specimens from 696 MPD patients (292 MTB and 404 NTM) were eventually analyzed. In total, 292 MTBC and 436 NTM isolates (mixed infection of two species in 32 specimens) across 10 Mycobacterium species were identified. The most frequently isolated NTM species were M. intracellulare (n = 236, 54.1%), followed by M. abscessus (n = 106, 24.3%), M. kansasii (n = 46, 10.6%), M. avium (n = 36, 8.3%). Twenty-two cases had M. intracellulare and M. abscessus mixed infection and ten cases had M. avium and M. abscessus mixed infection. A high level of agreement (n = 696; 94.5%) was found between MGIT 960-TBc ID and PCR-REBA Myco-ID (k = 0.845, P = 0.000). PCR-REBA Myco-ID assay had higher AUC for both MTBC and NTM than MGIT 960-TBc ID test. Conclusion PCR-REBA Myco-ID has the advantages of rapid, comparatively easy to perform, relatively low cost and superior accuracy in mycobacterial species identification compared with MGIT 960-TBc ID. We recommend it into workflow of mycobacterial laboratories especially in source-limited countries.

Objective: The aim of this study was to assess the clinical utility of metagenomic next-generation sequencing (mNGS) on smear-negative extrapulmonary specimens collected in China. Methods: Specimens were tested by mNGS and other routine tests for tuberculosis (TB). The diagnostic accuracy of mNGS was calculated and compared with the final clinical diagnosis. Results: The sensitivity of mNGS was found to be significantly higher than the sensitivities of the other routine TB tests. Receiver operating characteristic curve analysis showed that mNGS achieved the highest area under the curve (AUC) value of 0.79. The mNGS positive rate was highest for tuberculous meningitis. All non-tuberculous extrapulmonary pathogens were directly and simultaneously detected. Conclusions: mNGS appeared to be superior to all previous etiological tests for TB on smear-negative extrapulmonary specimens and could identify all possible pathogens at once within 48 h.

Abstract Background H7 subtype avian influenza has caused great concern in the global poultry industry and public health. The conventional serological subtype-specific diagnostics is implemented by hemagglutination inhibition (HI) assay despite lengthy operation time. In this study, an efficient, rapid and high-throughput competitive enzyme-linked immunosorbent assay (cELISA) was developed for detection of antibodies against H7 avian influenza virus (AIV) based on a novel monoclonal antibody specific to the hemagglutinin (HA) protein of H7 AIV. Results The reaction parameters including antigen coating concentration, monoclonal antibody concentration and serum dilution ratio were optimized for H7 antibody detection. The specificity of the cELISA was tested using antisera against H1 ~ H9, H11 ~ H14 AIVs and other avian viruses. The selected cut-off values of inhibition rates for chicken, duck and peacock sera were 30.11, 26.85 and 45.66% by receiver-operating characteristic (ROC) curve analysis, respectively. With HI test as the reference method, the minimum detection limits for chicken, duck and peacock positive serum reached 20, 21 and 2− 1 HI titer, respectively. Compared to HI test, the diagnostic accuracy reached 100, 98.6, and 99.3% for chicken, duck and peacock by testing a total of 400 clinical serum samples, respectively. Conclusions In summary, the cELISA assay developed in this study provided a reliable, specific, sensitive and species-independent serological technique for rapid detection of H7 antibody, which was applicable for large-scale serological surveillance and vaccination efficacy evaluation programs.

Abstract Background Neuropathic pain belongs to chronic pain and is caused by the primary dysfunction of the somatosensory nervous system. Long noncoding RNAs (lncRNAs) have been reported to regulate neuronal functions and play significant roles in neuropathic pain. DLEU1 has been indicated to have close relationship with neuropathic pain. Therefore, our study focused on the significant role of DLEU1 in neuropathic pain rat models. Methods We first constructed a chronic constrictive injury (CCI) rat model. Paw withdrawal threshold (PWT) and paw withdrawal latency (PWL) were employed to evaluate hypersensitivity in neuropathic pain. RT-qPCR was performed to analyze the expression of target genes. Enzyme-linked immunosorbent assay (ELISA) was conducted to detect the concentrations of interleukin‐6 (IL-6), tumor necrosis factor‐α (TNF-α) and IL-1β. The underlying mechanisms of DLEU1 were investigated using western blot and luciferase reporter assays. Results Our findings showed that DLEU1 was upregulated in CCI rats. DLEU1 knockdown reduced the concentrations of IL‐6, IL‐1β and TNF‐α in CCI rats, suggesting that neuroinflammation was inhibited by DLEU1 knockdown. Besides, knockdown of DLEU1 inhibited neuropathic pain behaviors. Moreover, it was confirmed that DLEU1 bound with miR-133a-3p and negatively regulated its expression. SRPK1 was the downstream target of miR-133a-3p. DLEU1 competitively bound with miR-133a-3p to upregulate SRPK1. Finally, rescue assays revealed that SRPK1 overexpression rescued the suppressive effects of silenced DLEU1 on hypersensitivity in neuropathic pain and inflammation of spinal cord in CCI rats. Conclusion DLEU1 regulated inflammation of the spinal cord and mediated hypersensitivity in neuropathic pain in CCI rats by binding with miR-133a-3p to upregulate SRPK1 expression.

Soil salinization is a major environmental problem that seriously threatens the sustainable development of regional ecosystems and local economies. Fraxinus velutina Torr. is an excellent salt-tolerant tree species, which is widely planted in the saline-alkaline soils in China. A growing body of evidence shows that microRNAs (miRNAs) play important roles in the defense response of plants to salt stress; however, how miRNAs in F. velutina exert anti-salt stress remains unclear. We previously identified two contrasting F. velutina cuttings clones, salt-tolerant (R7) and salt-sensitive (S4) and found that R7 exhibits higher salt tolerance than S4. To identify salt-responsive miRNAs and their target genes, the leaves and roots of R7 and S4 exposed to salt stress were subjected to miRNA and degradome sequencing analysis. The results showed that compared with S4, R7 showed 89 and 138 differentially expressed miRNAs in leaves and roots, respectively. Specifically, in R7 leaves, miR164d, miR171b/c, miR396a, and miR160g targeting NAC1, SCL22, GRF1, and ARF18, respectively, were involved in salt tolerance. In R7 roots, miR396a, miR156a/b, miR8175, miR319a/d, and miR393a targeting TGA2.3, SBP14, GR-RBP, TCP2/4, and TIR1, respectively, participated in salt stress responses. Taken together, the findings presented here revealed the key regulatory network of miRNAs in R7 responding to salt stress, thereby providing new insights into improving salt tolerance of F. velutina through miRNA manipulation.

Fraxinus velutina Torr with high salt tolerance has been widely grown in saline lands in the Yellow River Delta, China. However, the salt-tolerant mechanisms of F. velutina remain largely elusive. Here, we identified two contrasting cutting clones of F. velutina, R7 (salt-tolerant), and S4 (salt-sensitive) by measuring chlorophyll fluorescence characteristics (Fv/Fm ratio) in the excised leaves and physiological indexes in roots or leaves under salt treatment. To further explore the salt resistance mechanisms, we compared the transcriptomes of R7 and S4 from leaf and root tissues exposed to salt stress. The results showed that when the excised leaves of S4 and R7 were, respectively, exposed to 250 mM NaCl for 48 h, Fv/Fm ratio decreased significantly in S4 compared with R7, confirming that R7 is more tolerant to salt stress. Comparative transcriptome analysis showed that salt stress induced the significant upregulation of stress-responsive genes in R7, making important contributions to the high salt tolerance. Specifically, in the R7 leaves, salt stress markedly upregulated key genes involved in plant hormone signaling and mitogen-activated protein kinase signaling pathways; in the R7 roots, salt stress induced the upregulation of main genes involved in proline biosynthesis and starch and sucrose metabolism. In addition, 12 genes encoding antioxidant enzyme peroxidase were all significantly upregulated in both leaves and roots. Collectively, our findings revealed the crucial defense pathways underlying high salt tolerance of R7 through significant upregulation of some key genes involving metabolism and hub signaling pathways, thus providing novel insights into salt-tolerant F. velutina breeding.

Objective: Metagenomic Next-Generation Sequencing (mNGS) has been applied as a novel method of detection pathogens for infectious diseases, but its value in the rapid diagnosis of tuberculous meningitis(TBM)has not been clarified based on large samples. Methods: A retrospective analysis was conducted on 51 inpatients with suspected TBM who underwent mNGS and four other tests in cerebrospinal fluid (CSF). Results: Among 51 included patients, 45 cases were diagnosed as TBM (38 definite, 5 probable, 2 possible) and 6 cases as non-TBM. Using final diagnosis as reference standard, the sensitivity, specificity, PPV (positive predictive value), and NPV (negative predictive value) of mNGS in CSF for TBM were 84.44%(38/45, 69.94%–93.01%), 100%(6/6, 51.68%–100%), 100%(40/40, 88.57%–100%) and 46.15%(6/13, 20.40%–73.88%). The diagnostic sensitivity of mNGS(84.4%)was significantly higher than that of AFB (0%, P = 0.000), MGIT960 culture(22.2%, P = 0.000), MTB PCR(24.4%, P = 0.000) and Xpert MTB/RIF(40%, P = 0.000). The ROC curve showed that CSF protein quantification and CSF cell count might be valuable in the prediction of NGS positive detection of MTB (Mycobacterium tuberculosis). Conclusion: CSF mNGS had high sensitivity, specificity and PPV in the diagnosis of TBM. Patients with a significant increase in CSF cell number and protein quantification might have a higher likelihood of positive MTB detection of NGS.

A novel metal–organic framework (MOF) based on La(III) ions as the metal nodes with the formula of {[LaL(H2O)2]·H2O}n (1) was prepared under the hydrothermal conditions by using 1-(4-carboxybenzyl)-1H-pyrazole-3,5-dicarboxylic acid (H3L) as the three connected ligand. Complex 1 shows excellent water stabilities as well as solvent-free conditions, and reveals microporous nature as established via the CO2 sorption at 195 K. The resulting activated 1a shows good heterogeneous catalytic activity for the cyanosilylation of the aromatic aldehydes, which could be reused for at least four times without losing the framework integrality. Furthermore, anti-bacterial activity of the compound was deep studied within the research. Firstly, the RT-PCR was utilized to determine pi3k and akt relative expression in macrophages after tuberculosis (TB) infection and compound treatment. And the ELISA detection was enrolled for IFN-γ and TNF-α detection. Next, the production of active nitrogen mediator (RNIs) was measured by the RNIs detection kit. Finally, compound protective function in vivo was assessed within the TB infected mice model, the bacterial colony numbers in the lung tissue was determined by diluting ager plate method, the pathological injury of lung was observed by HE staining. In this research, we aim to reveal a new candidate for the TB infection treatment.

Abstract Background Pathogens including duck-origin avian influenza virus (AIV), duck-origin Newcastle disease virus (NDV) and duck Tembusu virus (DTMUV) posed great harm to ducks and caused great economic losses to the duck industry. In this study, we aim to develop a triplex real-time polymerase chain reaction (PCR) assay to detect these three viruses as early as possible in the suspicious duck flocks. Results The detection limit of the triplex real-time PCR for AIV, NDV, and DTMUV was 1 × 101 copies/μL, which was at least 10 times higher than the conventional PCR. In addition, the triplex assay was highly specific, and won’t cross-react with other duck pathogens. Besides, the intra-day relative standard deviation and inter-day relative standard deviation were lower than 4.44% for these viruses at three different concentrations. Finally, a total of 120 clinical samples were evaluated by the triplex real-time PCR, the conventional PCR and virus isolation, and the positive rates for these three methods were 20.83, 21.67, 19.17%, respectively. Taking virus isolation as the gold standard, the diagnostic specificity and positive predictive value of the three viruses were all above 85%, while the diagnostic sensitivity and negative predictive value of the three viruses were all 100%. Conclusion The developed triplex real-time PCR is fast, specific and sensitive, and is feasible and effective for the simultaneous detection of AIV, NDV, and DTMUV in ducks.

Abstract Background Tuberculous pleurisy (TBP) is the most common form of extrapulmonary tuberculosis (TB). However, rapid diagnostic methods with high accuracy for tuberculous pleurisy are urgently needed. In the present study, we evaluated the diagnostic accuracy of Xpert MTB/RIF, LAMP and SAT-TB assay with pleural fluids from culture-positive TBP patients. Methods We prospectively enrolled 300 patients with exudative pleural effusions used as the samples for Xpert MTB/RIF, LAMP and SAT-TB assay. Of these, 265 including 223 patients diagnosed with TBP and 42 non-TBP patients used as controls were analyzed. As an important tool in the field of mathematics, higher-order derivation problems are widely used in differentials, quantum mechanics, and engineering applications. However, in the electronic computer (EC), due to the existence of the carry in the calculation, the computational efficiency is low when solving the higher-order derivation problem. In response to this problem, the ternary optical computer (TOC) has the advantages of no carry-in and the characteristics of numerous data bits, reconfigurable processors and parallel computing. Solve the higher-order derivation problems with complex operations by constructing multipliers and adders on the TOC platform, and copying multiple composite operator units (COUs). This article introduces the design of the higher-order derivative algorithm based on TOC in detail, the reconfiguration process of the multiplier and adder, and the number of bits of the multiplier and adder required in the implementation is given. Besides, the hardware resources and clock cycles in the operation are analyzed. The feasibility of the implementation scheme is verified by experiments. Compared with the traditional higher-order derivative, the higher-order derivative based on the TOC is superior in time performance, computational efficiency, and processing of complex operations. Due to the limitation of the research stage, the algorithm is only applicable to the function of polynomials, which lays a foundation for the further research of higher-order derivative algorithms, and has certain application significance.

The measurement of radiated emission (RE) in an anechoic chamber becomes very challenging at high frequencies, up to 60 GHz, because the scanning plane of the receiver is in measurement standard deviation from the actual wavefront. As a result, the RE intensity of the devices may be underestimated, resulting in electromagnetic interference. The deviation between the electric field at the far-field vertical scanning point and the actual wavefront is researched. Then, in an anechoic chamber, a hybrid deep learning amendment model of convolutional neural network (CNN) and transformer is proposed to correct the RE measurement at a 3 m distance. The results indicate that the correction is reliable, with an average error of 6.35% for a 3 m distance in a semianechoic chamber and less than 4.83% for other test scenarios. The proposed method provides a promising solution for RE measurement at a millimeter wave band in an anechoic chamber.

Identifying the spatio-temporal distribution hotspots of fishes and allocating priority conservation areas could facilitate the spatial planning and efficient management. As a flagship commercial fishery species, Largehead hairtail (Trichiurus japonicus) has been over-exploited since the early 2000s. Therefore, the spatio-temporal management of largehead hairtail nursery grounds could effective help its recovery. This study aims to predict juvenile largehead hairtail distribution patterns and identify priority conservation areas for nursery grounds. A two-stage hierarchical Bayesian spatio-temporal model was applied on independent scientific survey data (Catch per unit effort, CPUE) and geographic/physical variables (Depth, Distance to the coast, Sea bottom temperature, Dissolved oxygen concentration and Net primary production) to analyze the probability of occurrence and abundance distribution of juvenile largehead hairtail. We assessed the importance of each variable for explaining the occurrence and abundance. Using persistence index, we measured the robustness of hotspots and identified persistent hotspots for priority conservation areas. Selected models showed good predictive capacity on occurrence probability (AUC = 0.81) and abundance distribution (r = 0.89) of juvenile largehead hairtail. Dissolved oxygen, net primary production, and sea bottom temperature significantly affected the probability of occurrence, while distance to the coast also affected the abundance distribution. Three stable nursery grounds were identified in Zhejiang inshore waters, the largest one was located on the east margin of the East China Sea hairtail national aquatic germplasm resources conservation zones (TCZ), suggesting that the core area of nursery grounds occurs outside the protected areas. Therefore, recognition of these sites and their associated geographic/oceanic attributes provides clear targets for optimizing largehead hairtail conservation efforts in the East China Sea. We suggested that the eastern and southern areas of TCZ should be included in conservation planning for an effective management within a network of marine protected areas.

Lispe Latreille (Diptera: Muscidae) are a widespread group of predatory flies that inhabit semi-aquatic environments. Previous studies on this genus have mainly focused on morphological classification, so molecular data are entirely lacking, and there has been no attempt at a phylogenetic placement of the genus or the resolution of intragenic relationships. To address the phylogenetic placement of Lispe and to fill its gap in the Tree of Life Web Project, 58 Lispe spp. (covering 11 out of 13 acknowledged Lispe species groups) were selected to reconstruct a phylogeny using Maximum likelihood (ML) estimates, Maximum Parsimony (MP) analyses, and Bayesian inference (BI) based on two mitochondrial protein-coding genes (cytochrome c oxidase subunit I (COXI) and cytochrome b gene (CYTB)) and one nuclear gene (a fragment of the carbamoyl phosphate synthetase region of the CAD gene). The phylogenetic analyses indicated that the monophyletic Lispe is the sister group of the monophyletic Limnophora, together forming the tribe Limnophorini under the subfamily Coenosiinae. Three generic categories are proven obsolete: Chaetolispa Malloch, Lispacoenosia Snyder, and Xenolispa Malloch. Within the genus, the validity of 11 species groups is clarified by both molecular and morphological data. This study provides a sound basis for continuing intergeneric and intrageneric research into this fascinating and widespread genus.

The complete mitochondrial genome of Lepidotrigla alata was sequenced in this study. The genome sequence is 16,512 bp in length, comprising 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and one control region. Overall base composition is 26.46% A, 25.05% T, 17.22% G, and 31.27% C. Phylogenetic analysis indicated that L. alata was close to L. guentheri and L. microptera, and each genus in the family Triglidae was monophyletic. This study can provide valuable information for future evolutionary studies on L. alata and Triglidae.
