Human papillomavirus (HPV) integration is a critical step in cervical cancer development; however, the oncogenic mechanism at the genome-wide transcriptional level is still poorly understood. In this study, we employed integrative analysis on multi-omics data of six HPV-positive and three HPV-negative cell lines. Through HPV integration detection, super-enhancer (SE) identification, SE-associated gene expression and extrachromosomal DNA (ecDNA) investigation, we aimed to explore the genome-wide transcriptional influence of HPV integration. We identified seven high-ranking cellular SEs generated by HPV integration in total (the HPV breakpoint-induced cellular SEs, BP-cSEs), leading to intra-chromosomal and inter-chromosomal regulation of chromosomal genes. The pathway analysis revealed that the dysregulated chromosomal genes were correlated to cancer-related pathways. Importantly, we demonstrated that BP-cSEs existed in the HPV–human hybrid ecDNAs, explaining the above transcriptional alterations. Our results suggest that HPV integration generates cellular SEs that function as ecDNA to regulate unconstrained transcription, expanding the tumorigenic mechanism of HPV integration and providing insights for developing new diagnostic and therapeutic strategies.

Adjusting the interlayer structure of carbon nitride is expected to adjust its physicochemical properties, thereby enhancing photocatalytic activity. In this paper, alkaline metal ion-intercalated carbon nitride was prepared by molten salt co-pyrolysis. Alkali metal ions acted as mediators to provide a new migration path for the photogenerated electrons of g-C3N4. The intercalation of K+ and Na+ reduced the bandgap value of carbon nitride, expanded the visible light absorption range, and promoted the separation and migration of charge carriers. As a result, the hydrogen production rate of DCN1 reached 500 μmol g−1 h−1, which was 10 times than that of pristine g-C3N4. Experimental and theoretical methods were used to explain the improvement of photocatalytic efficiency after K+ and Na+ were intercalated into the interlayer of g-C3N4. Our work provided a new idea for the design of high-efficiency hydrogen evolution photocatalysts via intercalation method.

Hydrophilic hydrogels exhibit good mechanical properties and biocompatibility, whereas hydrophobic elastomers show excellent stability, mechanical firmness, and waterproofing in various environments. Hydrogel–elastomer hybrid material devices show varied application prospects in the field of bioelectronics. In this paper, the research progress in hydrogel–elastomer adhesion in recent years, including the hydrogel–elastomer adhesion mechanism, adhesion method, and applications in the bioelectronics field, is reviewed. Finally, the research status of adhesion between hydrogels and elastomers is presented.

OBJECTIVE: To investigate the mechanism by which long non-coding RNA TUG1 affects bladder cancer cell migration and invasion. METHODS: The expressions of TUG1 and miR-29c-3p were examined by quantitative RT-PCR (qRT-PCR) in 10 bladder cancer tissues and 5 bladder cancer cell lines. Trans-well assay was used to detect the changes in migration and invasion abilities of bladder cancer T24 cells after TUG1 knockdown using RNA interference technique, and the alteration in the expression of CAPN7 was also detected. The expression of CAPN7 was examined in T24 cells overexpressing mir-29c-3p by Western blotting, and luciferase reporter assay was performed to confirm the targeting of miR-29c-3p to TUG1 and CAPN7. The effects of CAPN7 overexpression and sh-TUG1 on the migration and invasion of T24 cells were investigated. RESULTS: The expression of TUG1 was up-regulated and mir-29c-3p was down-regulated significantly in bladder cancer tissue with a negative correlation between their expressions. TUG1 knockdown significantly inhibited the migration and invasion of T24 cells (P < 0.01). Overexpression of mir-29c-3p in T24 cells obviously down-regulated the expression of CAPN7 protein, whose expression was positively correlated with TUG1 expression (r=0.4081, P=0.0139). The results of luciferase reporter assay confirmed both TUG1 and CAPN7 as the targets of mir-29c-3p. CAPN7 overexpression could partially reverse the tumor suppressing effect of sh-TUG1 in T24 cells. CONCLUSION: Mir-29c-3p targeting TUG1 affects the migration and invasion of bladder cancer cells by regulating the expression of CAPN7.

Purpose To examine sleep disturbances in older adults in Macau and Guangzhou, China and their associated factors. Design and Methods Four‐hundred and thirty seven subjects in Guangzhou and 244 subjects in Macau were interviewed. Findings In total, 681 older adults participated in the study, and 27.8% reported sleep disturbance, with 43.9% in Macau and 18.8% in Guangzhou. Physical quality of life was negatively associated with sleep disturbances. Severe depressive symptoms were positively related but living in Guangzhou was negatively related to sleep disturbances. Sleep disturbances are more common in Macau compared to Guangzhou. Practice Implications Appropriate screening and treatment strategies are needed to address sleep disturbance in this population.

BACKGROUND: Little is known about the characteristics of older adults with cognitive impairment in Macao. This study aimed to determine the prevalence of cognitive impairment and the quality of life (QOL) of older adults living in the community and nursing homes. METHODS: A consecutive sample of 413 subjects (199 from the community; 214 from nursing homes) was recruited and interviewed using standardized instruments. Cognition was measured with the Repeatable Battery for the Assessment of Neuropsychological Status and QOL with the brief version of the World Health Organization Quality of Life instrument. RESULTS: Altogether 87 subjects (21.0%) had cognitive impairment. On multivariate analyses, advanced age (P < 0.001, OR = 1.06, 95%CI: 1.03-1.1) and depressive symptoms (P = 0.03, OR = 1.07, 95%CI: 0.005-1.1) were positively associated with cognitive impairment. Married marital status (P = 0.01, OR = 0.3, 95%CI: 0.1-0.7) and higher education level (P < 0.001, OR = 0.1, 95%CI: 0.06-0.3) were negatively associated with cognitive impairment. After the confounders were controlled for, cognitive impairment was significantly associated with the lower psychological (F (11,412) = 6.3, P = 0.01) and social relationship domains of QOL (F (11,412) = 4.0, P = 0.04). CONCLUSION: Cognitive impairment was found to be common in community-dwelling and nursing home resident older adults in Macao. Given cognitive impairment's negative impact on QOL, appropriate strategies should be implemented to improve access to treatment in this population.

In order to meet the requirement of IETM’s content display in different application scenarios, this paper takes the requirement of S1000D standard as the object, analyses the characteristics of its display content, divides the data module into general structure and special structure, and extracts common elements and private elements from the data module by taking six common data modules such as descript, proced, fault, schedul, IPD and crew as examples. The three-tier style architecture of business layer, abstract layer and implementation layer is designed. Based on this architecture, the online browser, online editor and PDF publishing style framework are implemented. Its advantages are easy to understand, modify and customize the style.

Data management is the core of IETM development. This paper designs IETM data management software based on GJB6600 standard. IETM data management software implements manual project management, manual data planning, data module / data entity / basis data and other manual data management, supports manual editing, manual layout release and delivery management, achieves data exchange in the upstream and downstream manufacturer manual production process, realizes the task assignment, proofreading, approval and finalization in the manual compilation process, realizes data version management and security control, and provide a collaborative working environment for the manual production.

In order to convert S1000D standard data into SCORM standard data, this paper modeled interactive electronic manual data (IETP) from two dimensions of SNS and PM, and generated SNS-IETP and PM-IETP models. The data of electronic training system was modeled to generate SCORMP model. Three models were compared and analyzed according to the business scenarios, and the PM-IETP model was selected as the conversion input to build the mapping model between PM-IETP and SCORMP. The common elements of SCORM standard are extracted to form a prior knowledge template, based on which a data conversion model is constructed and a dynamic update algorithm is designed. Through practice, this data conversion method is feasible and effective, which can solve the one-way conversion between the two standard data.

Technical literature is the bearing of the core knowledge of equipment, and it is the basis and support for the development of comprehensive equipment support. It is of great significance for equipment support. How to make the technical literature better play the inheritance, promotion and use efficiency of knowledge is the key problem to be solved in the comprehensive equipment support. The interactive electronic technical manual uses digital forms to store and manage technical literature in the form of text, forms, images, engineering graphics, sound, video, animation, and more. It is also viewed and used interactively to provide fault reasoning guidance and knowledge push services. In 2010, China released the interactive electronic technical manual National Military Standard GJB6600, which provides a good guide and specification for the use of military equipment technical literature, but lacks the support of a fully independent and controllable publishing platform. Under the background of domestically controllable domestic control, this paper analyzes the interactive publishing requirements of equipment developers and users for technical literature, gives the index of independent controllable technical literature publishing platform, and proposes an independent and controllable interactive publishing scheme based on GJB6600 technical literature. And proposed an interactive publishing platform construction facility that can guide technical literature.

Background: The incidence rate of endometrial cancer has increased significantly in the past two decades. The current chemotherapy of endometrial cancer can cause intolerable side effects. Recently, both cordycepin and gold nanostars have demonstrated the ability to inhibit the growth and differentiation of cancers. The aim of this study was to investigate the anti-endometrial cancer effects of cordycepin, gold nanostars and the combination of two. Methods: The gold nanostars were made by seed-mediated reduction, and their morphology was confirmed by laser particle size analysis, spectrophotometry and electron microscopy. MTT cytotoxicity assays, clonogenic assays and flow cytometry were employed to evaluate the inhibition rate, survival fraction and apoptosis in HEC-1A cells under different concentrations of cordycepin, Au NS and their combination. Results: This study revealed that both cordycepin and Au NS with 808 nm light exposure could inhibit cell proliferation, cause cell apoptosis, and inhibit clone ability of endometrial cancer cells. The combined Au NS-cordycepin solution produced greater anti-cancer effects than each treatment alone. Conclusion: This study supports further investigations into Au NS-cordycepin in the development of new treatments for endometrial cancer.

Antibiotic resistance has become a serious global concern, and the discovery of antimicrobial herbal constituents may provide valuable solutions to overcome the problem. In this study, the effects of therapies combining antibiotics and four medicinal herbs on methicillin-resistant Staphylococcus aureus (MRSA) were investigated. Specifically, the synergistic effects of Magnolia officinalis, Verbena officinalis, Momordica charantia, and Daphne genkwa in combination with oxacillin or gentamicin against methicillin-resistant (ATCC43300) and methicillin-susceptible (ATCC25923) S. aureus were examined. In vitro susceptibility and synergistic testing were performed to measure the minimum inhibitory concentration and fractional inhibitory concentration (FIC) index of the antibiotics and medicinal herbs against MRSA and methicillin-susceptible S. aureus. To identify the active constituents in producing these synergistic effects, in silico molecular docking was used to investigate the binding affinities of 139 constituents of the four herbs to the two common MRSA inhibitory targets, penicillin binding proteins 2a (PBP2a) and 4 (PBP4). The physicochemical and absorption, distribution, metabolism, and excretion properties and drug safety profiles of these compounds were also analyzed. D. genkwa extract potentiated the antibacterial effects of oxacillin against MRSA, as indicated by an FIC index value of 0.375. M. officinalis and V. officinalis produced partial synergistic effects when combined with oxacillin, whereas M. charantia was found to have no beneficial effects in inhibiting MRSA. Overall, tiliroside, pinoresinol, magnatriol B, and momorcharaside B were predicted to be PBP2a or PBP4 inhibitors with good drug-like properties. This study identifies compounds that deserve further investigation with the aim of developing therapeutic agents to modulate the effect of antibiotics on MRSA. Impact statement Antibiotic resistant is a well-known threat to global health and methicillin-resistant Staphylococcus aureus is one of the most significant ones. These resistant bacteria kill thousands of people every year and therefore a new effective antimicrobial treatment is necessary. This study identified the herbs and their associated bioactive ingredients that can potential the effects of current antibiotics. These herbs have long history of human usage in China and have well-defined monograph in the Chinese Pharmacopeia. These indicate their relatively high clinical safety and may have a quicker drug development process than that of a new novel antibiotic. Based on the results of this study, the authors will perform further in vitro and animal studies, aiming to accumulate significant data for the application of clinical trial.

Aim There have been no previous studies of quality of life (QOL) in older adults in Macao. This study aimed to examine QOL in relation to the sociodemographic and clinical characteristics of adults aged ≥50 years in Macao. Methods A sample of 451 subjects (203 living in the community, 248 living in nursing homes) was interviewed using standardized instruments. Basic sociodemographic and clinical data including QOL were collected. Result There were no significant differences between the community and nursing home groups in any of the QOL domains. Multiple linear regression analyses revealed that poor physical QOL was significantly predicted by severe depressive symptoms, insomnia, major medical conditions, unmarried status, and lower education ( F 11,438 = 26.2, P < 0.001), which accounted for 38.2% of the variance. Poor psychological QOL was significantly predicted by severe depressive symptoms and lower educational level ( F 11,438 = 24.3, P < 0.001), which accounted for 36.4% of the variance. Poor social QOL was significantly predicted by severe depressive symptoms, male gender, and unmarried status ( F 11,438 = 5.6, P < 0.001), which accounted for 12.5% of the variance. Poor environment QOL was significantly predicted by lower educational level, severe depressive symptoms, and younger age ( F 11,438 = 6.6, P < 0.001), which accounted for 12.1% of the variance. Conclusion Older Macanese adults had poorer scores on physical and social QOL domains than the general Hong Kong Chinese population. Their QOL was more strongly related to severe depressive symptoms, major medical conditions, and insomnia.

An activated carbon (AC)-supported nanocomposite was prepared by precipitating β-FeOOH onto KOH activated soybean meal-derived biochar (SYBK). The as-prepared β-FeOOH/SYBK composites were characterized by N2-Brunauer–Emmett–Teller (BET), scanning electron microscopy (SEM), X-ray diffractions (XRD) and X-ray photoelectron spectroscopy (XPS). XRD results confirmed that β-FeOOH was impregnated by AC. The chromate (Cr(VI)) removal capacity was investigated in a batch experiment with different conditions. The ratios of β-FeOOH and AC were compared for Cr(VI) removal and a loading quantity of 20 wt% β-FeOOH was considered as the most efficient amount. This was possibly ascribed to it having the highest surface area (670.65 m2 g−1) of the β-FeOOH/SYBK nanocomposites. It was found that 20β-FeOOH/SYBK could remove as much as 96% Cr(VI) at pH 1–2 with 2 mmol L−1 EDTA and 2.0 g L−1 nanocomposites. The maximal Cr(VI) removal by 20β-FeOOH/SYBK was 37.04 g kg−1, as estimated by a Langmuir isotherm model. The removal mechanisms were examined by studying the speciation of Cr on sorbents as well as in aqueous solution. The XPS analysis of spent sorbents and chemical speciation of Cr in aqueous solutions revealed that partial Cr(VI) was reduced to Cr(III) on sorbents and in aqueous solution. This suggests that Cr(VI) can be removed by simultaneous sorption and reduction by the as-prepared nanocomposites.

BACKGROUND The objective of this study was to determine whether miR-24 can regulate malignant proliferation and chemotherapy sensitivity of EC cells by targeted silencing of the S100 Calcium Binding Protein A8 (S100A8) gene. MATERIAL AND METHODS The expression of miR-24 in EC tissues was detected by quantitative real-time PCR. The proliferation ability and chemotherapy sensitivity were analyzed by MTT assay. Bioinformatics software was used to predict some potential target genes of miR-24. Luciferase activity assay was used to verify the relationship between target genes and miR-24. S100A8 protein expression was detected by Western blot analysis. RESULTS The low expression of miR-24 in EC tissues compared with normal control tissues suggests miR-24 might play a role in tumorigenesis of EC. EC HEC-1A cells were transfected with miR-24 agonist (agomiR-24) to up-regulate the expression of miR-24. Up-regulation of miR-24 inhibited the cell proliferation and advanced the chemotherapy sensitivity to paclitaxel in HEC-1A cells significantly. We used several types of bioinformatic software to predict that miR-24 could specifically combine with the 3' untranslated region (3'UTR) of the S100A8 gene, and this prediction was verified by Western blot and luciferase activities assay. The regulation effects of miR-24 enhancement on cell proliferation and chemotherapy sensitivity were largely reversed by S100A8 up-regulation. CONCLUSIONS miR-24 acts as a tumor-suppressing gene to inhibit malignant proliferation and advance chemotherapy sensitivity to paclitaxel in EC by targeted silencing of the S100A8 gene.

Endometrial cancer (EC) is the most common malignancy of the female reproductive tract. In this study, we clarified the clinical significance of CDKN2B antisense RNA 1 (CDKN2B-AS) gene, and its effects on paclitaxel sensitivity in EC. Firstly, CDKN2B-AS gene was highly expressed in EC tissues and cell lines. The high-expression of CDKN2B-AS gene was associated with high pathological grade and low paclitaxel sensitivity of EC tissues. Knockdown of CDKN2B-AS gene sensitized Ishikawa/PA and HEC1A/PA cells to paclitaxel, and promoted paclitaxel-induced cytotoxicity. Secondly, the low-expression of miR-125a-5p was closely associated with low paclitaxel sensitivity of EC cells, and up-regulation of miR-125a-5p could increase paclitaxel sensitivity of Ishikawa/PA and HEC1A/PA cells. MiR-125a-5p also mediated the suppressive effects of knockdown of CDKN2B-AS on paclitaxel resistance in EC cells. Thirdly, B-cell lymphoma-2 (Bcl2) and Multidrug Resistance-Associated Protein 4 (MRP4) genes were target genes of miR-125a-5p, which modulated paclitaxel resistance of Ishikawa/PA and HEC1A/PA cells through targeted silencing Bcl2 and MRP4. In conclusion, high-expression of CDKN2B-AS is associated with a poor response to paclitaxel of EC patients, and knockdown of CDKN2B-AS inhibits paclitaxel resistance through miR-125a-5p-Bcl2/MRP4 pathway in EC patients. Our findings help elucidate the molecular mechanisms of chemoresistance in EC patients.

As the equipment of modern agriculture becomes more and more complex, fault reasoning and fault maintenance support have turned into two important core functions. And how to improve the fault reasoning task success rate is an unavoidable problem for Interactive Electronic Technical Manual (IETM) that has been used in modern agriculture support information. Aiming to solve the poor interactive, inaccurate location, one-sided guidance problems in fault reasoning and fault maintenance of complex agricultural equipment, we use process data module and fault data module to describe the fault and full process for fault diagnose, fault reasoning and fault maintenance, and then according to interactive fault reasoning principle based on process data module and fault data module to guide fault maintenance staff complete fault reasoning, isolation and maintenance in shorter time. As a result, this is a new approach to improve the accuracy and efficiency of fault remedy and maintenance, and it also improves the efficiency of fault reasoning of complex agricultural equipment by IETM in an interactive way.

Spare parts are common in many industries, and spare parts management is a very important part of equipment maintenance. The level of spare parts management directly affects whether the entire equipment system can operate normally under limited funding conditions. This paper research the related literatures on spare parts management, and conclude the research status on spare parts classification method, spare parts demand forecasting and spare parts management model. And put forward some of my own opinion on spare parts management.

This paper describes the concept of interactive electronic technology manual (IETM), which combined with the information technique development trend and equipment maintenance demand. It discusses the development trend of IETM and the relationship between equipment maintenance, IETM and equipment integrated support design. The development framework of IETM is presented in this paper. The IETM production process and main work contents of each stage are described in detail, which has great application value. It can provide technical support for the information construction of equipment support.

Fault isolation and fault maintenance support is an important function, and how to improve the fault isolation mission success rate is an unavoidable problem for IETM using in integrated logistic support informatization. Aim to solve the low interactive, location inaccurate, one-sided guidance problems in fault isolation and fault maintenance, this paper use process data module to describe full process for fault diagnose, fault isolation and fault maintenance, and then according to interactive fault reasoning principle based on process data module to guide fault maintenance man accomplish fault reasoning, isolation and maintenance in shorter time. As a result, this improves the accuracy and efficiency of fault remedy and maintenance, also upgrade the efficiency of integrated logistic support.

This paper implements a transformation algorithm, which is used to transform XML Schema into Java codes. Based on the algorithm design, we first defined a Java class, which is used to implement paring unit of the algorithm. Secondly, we defined a Java class for each of the twenty-eight kinds of elements and twelve kinds of facet elements, and build import or other relation between these classes according to any possible nested relation between elements. These classes are used to create Java objects for elements. Thirdly, in order to increase code reusability or define data type of Java class variables conveniently, we defined a set of Java abstract classes or interfaces. Related Java classes extend or implement these abstract classes or interfaces to realize specific functions. Finally, we performed numerous experiments to validate the algorithm. The algorithm can transform any non-empty and well-formed XML Schema documents into Java objects, and can be applied to any XML related software development.

An efficient method for transforming XML Schema into programming language is the development basis of XML related software. In this paper, we propose an algorithm design, which is used to transform XML Schema document into Java codes. Firstly, we studied XML Schema elements and nested relation between elements in all cases. Then we studied DOM API and used DOM parser to parse XML Schema document. Thirdly, we studied Deep First Search algorithm and used it to traverse parsed result. During the traversing process, create corresponding Java objects for XML Schema elements. Finally, we got a collection of Java objects, which is organized according to nested relation between elements. Except notation and anyAttribute, the algorithm can transform the other elements contained in XML Schema document into Java objects, which makes sure that the collection of transformational Java objects is an equivalent representation of XML Schema document.

Apply Integrated Logistics Support (ILS) to weapon equipment can efficiently improve equipment's automation and digital level. ILS needs support of various integrated support systems, which have demands for data editing. Nowadays, most of data editing software used in these systems are customized and provide a form-based editing approach, which becomes an obstacle to carry out ILS. This paper brings forward to design a data-driven based universal data editing framework. In this framework, data models are taken as input and only corresponding data models need to be modified when data change. Firstly, the whole development process of data editing software that adopt form-based development mode is analyzed in detail to find out problems exists in this development mode. Then the data-driven based universal data editing framework is designed. New idea of data-driven architecture is proposed. Data-driven architecture takes data models as input and processes all data models in a universal way. Finally, key technology for framework realization is given.

There are limited available data on elder abuse and its impact on quality of life (QOL) in China. This study investigated the prevalence of elder abuse in nursing homes and its associated demographic, clinical factors and QOL in Macau and Guangzhou, China. A total of 681 subjects (244 in Macau and 437 in Guangzhou) were consecutively recruited. The prevalence of elder abuse was 11.48% and 8.24% in Macau and Guangzhou, respectively. Multivariate analyses revealed that having a religion and depressive symptoms were independently and positively associated with elder abuse. No significant association between elder abuse and any QOL domain was found. Elder abuse is common in nursing homes in both Macau and Guangzhou. Appropriate strategies and educational programs should be developed for health professionals to reduce the risk of elder abuse.

Endometrial carcinoma is the most common malignant tumor of the female genital tract worldwide. TUSC7 (tumor suppressor candidate 7) is an antisense long non-coding RNA and is downregulated and acts as a potential tumor suppressor in several malignant tumors. In this study, the low expression of TUSC7 was confirmed in endometrial carcinoma tissues and was associated with high pathological stages of endometrial carcinoma, which revealed that TUSC7 might be involved in tumorigenesis and progression of endometrial carcinoma. Moreover, the expression of TUSC7 in endometrial carcinoma tissues and cell lines resistant to CDDP and Taxol was lower than that in sensitive endometrial carcinoma tissues and cell lines, which indicated that the TUSC7 expression level was positively correlated with the response of endometrial carcinoma patients to chemotherapy with CDDP and Taxol. TUSC7 upregulation inhibited proliferation, blocked cells at G1 phase, and advanced apoptosis and chemotherapy sensitivity to CDDP and Taxol in HEC1A/CR cell line. Furthermore, miR-23b was upregulated in endometrial carcinoma and negatively correlated with the expression of TUSC7. RNA pull-down assay indicated that TUSC7 could specifically silence the expression of miR-23b in HEC1A/CR cell line; miR-23b was a target gene of TUSC7. MiR-23b upregulation mostly reversed the TUSC7-induced regulatory effects on HEC1A/CR cell line. In summary, long non-coding RNA TUSC7 was underexpressed in endometrial carcinoma, especially in endometrial carcinoma chemotherapy-resistant tissues and cell lines and acted as a potential tumor suppressor gene to inhibit cell growth as well as advance the chemotherapy sensitivity through targeted silencing of miR-23b, which might provide a new therapeutic target to endometrial carcinoma.

Ultra-thin layered structures and modified bandgaps are two efficient strategies to increase the photocatalytic performance of various materials for the semiconductor industry. In the present study, we combined both strategies in one material to form carbon-doped graphitic carbon nitride (g-C3N4) nano-layered structures by the method of melamine thermal condensation, in the presence of different mass ratios of biochar. The characterization showed that the composite with the best ratio retained the g-C3N4 polymeric framework and the bond with g-C3N4. The biochar was established via π-π stacking interactions and ether bond bridges. The π-conjugated electron systems provided from biochar can elevate charge separation efficiency. The ultra-thin structure also curtailed the distance of photogenerated electrons migrating to the surface and enlarge specific surface area of materials. The presence of carbon narrowed the bandgap and increased light absorption at a wider range of wavelengths of g-C3N4. The biochar/melamine ratio of 1:15 presented the best performance, 2.8 and 5 times faster than g-C3N4 degradating Rhodamine and Methyl Orange, respectively. Moreover, the catalyst presented a good stability for 4 cycles. In addition to that, biochar from waste biomass can be considered a sustainable, cost-effective, and efficient option to modify g-C3N4-based photocatalysts.

Helicobacter pylori infection is a WHO class 1 carcinogenic factor of gastric adenocarcinoma. In the past decades, many studies have demonstrated the increasing trend of antibiotic resistance and pointed out the necessity of new effective treatment. This study was aimed at identifying phytochemicals that can inhibit H. pylori and possibly serve as adjuvant treatments. Here, in silico molecular docking and drug-like properties analyses were performed to identify potential inhibitors of urease, shikimate kinase and aspartate-semialdehyde dehydrogenase. These three enzymes are targets of the treatment of H. pylori. Susceptibility and synergistic testing were performed on the selected phytochemicals and the positive control antibiotic, amoxicillin. The in-silico study revealed that oroxindin, rosmarinic acid and verbascoside are inhibitors of urease, shikimate kinase and aspartate-semialdehyde dehydrogenase, respectively, in which, oroxindin has the highest potency against H. pylori, indicated by a minimum inhibitory concentration (MIC) value of 50 &mu, g/mL. A combination of oroxindin and amoxicillin demonstrated additive effects against H. pylori, as indicated by a fractional inhibitory concentration (FIC) value of 0.75. This study identified phytochemicals that deserve further investigation for the development of adjuvant therapeutic agents to current antibiotics against H. pylori.

Endometrial cancer drug treatments often produce undesirable effects. Thus, discovering new drugs with fewer side effects is required. Cordycepin is a constituent of Cordyceps sinensis, which has been proven to inhibit tumor growth by stimulating the adenosine A3 receptor (A3R). However, cordycepin is rapidly degraded by adenosine deaminase (ADA) and has a clinically unacceptable short half-life. One of its derivatives, MRS5698, was predicted to exhibit antitumor effects with a poor affinity to ADA by our previous validated in silico experiments. The purpose of this study was to explore the possibilities of using MRS5698 as a novel antitumor agent through experiments on Ishikawa and HEC-1A cells. The detection of inhibition and apoptotic rate of MRS5698 and cisplatin, and their combination, on Ishikawa and HEC-1A cells were performed by MTT assays and flow cytometry, respectively. The inhibition rates of MRS5698 on Ishikawa and HEC-1A cells were both significantly higher than the control groups ( P < 0.05). MRS5698 produced a higher inhibitory effect on HEC-1A cells than on Ishikawa cells with IC50 values of 20.55 and 27.25 μg/mL, respectively. MRS5698 had a stronger inhibitory effect than cisplatin on HEC-1A cells. The Annexin V-FITC/propidium iodide assays demonstrated that the total rate of apoptosis of MRS5698 on HEC-1A cells was higher than that on Ishikawa cells. The results of MTT assay and cellular apoptosis showed that the combined use of MRS5698 and cisplatin produces dose-independent antagonistic effects. MRS5698 produced antitumor effects on both cell lines, which were better than that of cordycepin. However, the combined use of MRS5698 and cisplatin produced an antagonistic effect. A further in vivo study could be considered for investigating the antitumor effects of either MRS5698 monotherapy or MRS5698 in combination with other nonplatinum-based chemotherapeutic drugs in treating endometrial cancer.

BACKGROUND The aim of this study was to investigate the sensitivity to rapamycin of endometrial cancer cells with different phosphatase and tensin homologue (PTEN) expression to understand the mechanism of resistance to mammalian target of rapamycin (mTOR) inhibitors in the treatment of endometrial cancer. MATERIAL AND METHODS Twenty specific pathogen-free female BALB/c mice received transplants of either HEC-1A (PTEN-positive) or Ishikawa (PTEN-negative) cells. Mice in the treatment group were injected intraperitoneally once a week for 4 consecutive weeks. The control group was injected weekly with phosphate buffer saline (PBS) for 4 consecutive weeks. Tumor volume, tumor mass, growth curves, and inhibition rate were measured, after which the mice were killed. RESULTS Both tumor growth rate and size were slower in the treatment group than in the control group for all mice that received transplants of either HEC-1A or Ishikawa cells. The tumor inhibition rates in the treatment group were 48.1% and 67.1% in mice transplanted with HEC-1A and Ishikawa cells, respectively. CONCLUSIONS The inhibitory effects of rapamycin were enhanced in PTEN-negative Ishikawa tumor cells compared with PTEN-positive HEC-1A cells, which could explain the reduced effect of rapalogues in some endometrial cancer patients and help to understand the mechanism of resistance to this drug.

Although the molecular therapeutics targeting key biomarkers such as epithelial growth factor receptor (EGFR), PI3K/AKT/mTOR, and vascular endothelial growth factor (VEGF) shows some success in clinical trials, some internally existing challenges in endothelial cancer biology hinder the drug effects. One of the major challenges stems from cancer stem cell-derived drug resistance. CD133 positive cells are well believed as cancer stem cells (CSC) in endometrial cancers and NOTCH pathway plays a critical role in retaining CD133+ cells by promoting CSC self-renewal and chemoresistance. Here, we initiated a therapeutic strategy to improve effects of EGFR inhibition by targeting NOTCH pathway of CD133+ cells in endometrial cancers. We first detected and purified the CD133+ cell fraction in endometrial cancer cell line Ishikawa (IK), and validated activation of NOTCH pathway in the CD133+ cells that have higher proliferation rate and lower apoptosis rate, comparing to CD133- cells. Results of nude mouse xenograft experiments further demonstrated CD133+ cells retain higher tumorigenesis capacity than CD133- cells, indicating their tumor-initiating property. Last, we applied both NOTCH inhibitor DAPT and EGFR inhibitor AG1478 treatment on endometrial cancer lines IK and HEC-1A and the results suggested improvement effects of the combination therapy compared to the treatments of DAPT or AG1478 alone. These findings indicated targeting NOTCH pathway in CD133+ cells, combining with EGFR inhibition, which provides a novel therapeutic strategy for endometrial cancer diseases.

The requirements of data exchange, with XML as the main format, use structural-centric and object-centric data binding frameworks. Through analysis of the current data binding framework, we present a data-centric XML binding framework in order to establish interfaces between relational databases and XML files. This framework solves the problems of generating large amounts of temporary Java code or producing complex XML schema files. The functions of marshalling and unmarshalling meditate the conversion between relational databases and XML files. In this model, objects and files are completely decoupled. Therefore, it implements a non-invasive from of data binding.

Purpose To examine the prevalence of sleep disturbances (difficulty initiating sleep [DIS], difficulty maintaining sleep [DMS], and early morning awakening [EMA]), their socio-demographic and clinical correlates, and quality of life (QOL) in older adults in Macao. Design and methods Four hundred fifty-one subjects were interviewed using standardized instruments. Findings The prevalence of at least one type of sleep disturbance was 38.1%; the figures of DIS, DMS, and EMA were 18.6, 31.3, and 23.9%, respectively. Female sex and depressive symptoms were independently associated with more frequent sleep disturbances. Sleep disturbances were independently associated with lower physical QOL. Practice implications Sleep disturbances are common in older adults in Macao. Appropriate strategies should be implemented to prevent and treat sleep disturbances and concerted attempts should be made to improve access to treatment.

Rosiglitazone is a synthetic ligand of peroxisome proliferator-activated receptor γ (PPARγ), and it can induce apoptosis and autophagy in a variety of cancer cells. In the present study, we aimed to investigate the influence of rosiglitazone on the proliferation and apoptosis of the 5637 and T24 human bladder cancer cell lines. The results demonstrated that the level of growth inhibition rate was gradually increased by treating the 5637 and T24 cells with higher doses of rosiglitazone and longer incubation time. Rosiglitazone exerted a potent inhibiting effect on migration of the 5637 and T24 cell lines. Moreover, rosiglitazone exerted a antineoplastic activity by inducing apoptosis and cell cycle arrest. Furthermore, treatment with rosiglitazone led to decrease the anti-apoptotic protein Bcl-2 level and increase the pro-apoptotic protein caspase 3 level in 5637 and T24 cells. Importantly, the protein expression of PPAR γ was significantly increased in the present of rosiglitazone in 5637 and T24 cells as compared to control group. In conclusion, the present study demonstrates that rosiglitazone has a potential antineoplastic activity in human bladder cancer cell lines, and the underlying mechanism was mediated, at least partially, through regulation of apoptosis-related protein and PPAR γ expression.

Purpose To evaluate memory impairment associated with electroconvulsive therapy (ECT)-antipsychotic (AP) combination in comparison to AP monotherapy in schizophrenia. Design and methods A systematic literature search of randomized controlled trial (RCTs) was performed. Findings Eleven RCTs that compared ECT-AP combination (n = 508) with AP monotherapy (n = 510) were analyzed. ECT-AP combination was associated with greater impairment than AP monotherapy in (1) endpoint memory quotient (MQ) of the Wechsler Memory Scale (WMS)-Revised at the end of the ECT course; and (2) picture recall, counting, recognition, and associative learning of the WMS. However, no group difference was found in MQ at 1 and 2 weeks post-ECT. Practice implications The ECT-AP combination was associated with greater transient memory impairment compared to AP monotherapy.

Summary Background MicroRNAs (miRNAs) are small non-coding nucleotides that regulate mRNA stability and protein expression by imperfect base pairing with the 3′-untranslated region (3′UTR) of target mRNAs. Many miRNAs have been documented to be aberrantly expressed in human cancers, but the role of miRNAs in endometrioid endometrial cancer (EEC) remains poorly understood. The objective of this study was to investigate the effect of miR-125b on EEC development and to explore its molecular mechanism in EEC carcinogenesis. Material/Methods Real-time quantitative PCR was applied to evaluate the expression level of miRNA-125b in EEC and normal endometrium (NE) samples. The invasion ability of miR-125b in EEC HEC1B cells was analyzed by Transwell assay after pre-miR-125b or anti-miR-125b transfection. For the invasion mechanism analysis of miR-125b on HEC1B cells, miRBase, TargetScan, miRanda and PicTar were used to predict the possible target gene of miR-125b. Luciferase activities assay, cotransfection and Western blot were used to reveal that the predicted target genes of miR-125b were direct and specific. RNA interference technology was used to confirm that the invasion inhibition of miR-125b was directly induced by ERBB2. Results Our study showed that miR-125b was down-regulated in human EEC specimens compared to that in NC specimens. Over-expression of miR-125b in HEC1B cells inhibited EEC invasion and this inhibitory effect on HEC1B cells could be restored by miR-125b knock down. Mechanism analysis revealed that ERBB2 was a direct and specific target of miR-125b. The inhibitory effect on EEC cell invasion was mediated by miR-125b inhibition of the translation of a proto-oncogene, ERBB2. Conclusions Aberrantly expressed miR-125b contributes to HEC1B cells invasion partly through directly down-regulating ERBB2 protein expression in EEC. This miRNA signature offers a novel potential therapeutic strategy for EEC.

Loss or mutation of the PTEN (phosphatase and tensin homologue deleted on chromosome 10) gene is associated with resistance to epidermal growth factor receptor (EGFR) inhibitors. However, the mechanism underlying remains elusive. In this study, we aimed to explore whether sensitivity to the EGFR tyrosine kinase inhibitor (TKI) is affected by PTEN status in endometrial cancer cells. PTEN siRNA and the PTEN gene were transfected into HEC-1A and Ishikawa endometrial cancer cells using lentiviral vectors. Cells were treated under various concentrations of RG14620 and rapamycin, which are EGFR and mammalian target of rapamycin (mTOR) inhibitors, respectively. The IC(50) of RG16420 was determined by using the MTT method. Cell apoptosis and the cell cycle were studied, and activation of EGFR, AKT, and p70S6 were detected by Western blot analysis. Loss of PTEN promoted cell proliferation and led to significant increases in the levels of EGFR, phospho-EGFR, AKT, phospho-AKT, and phospho-mTOR proteins. Ishikawa and HEC-1A(PTENkd) cells that displayed loss and inactivation of PTEN function were resistant to RG14620. HEC-1A and Ishikawa(PTEN) cells with intact PTEN were sensitive to RG14620. The combination of two inhibitors was more effective than both monotherapies, particularly in carcinoma cells with PTEN dysfunction. Decreased phospho-EGFR protein expression was observed in all cell lines that were sensitive to RG14620. Decreased phospho-AKT and phospho-p70S6 protein expression was observed in PTEN-intact cells that were sensitive to RG14620. PTEN loss results in resistance to EGFR TKI, which was reversed by PTEN reintroduction or mTOR inhibitor treatment. The combined treatment of EGFR TKI and the mTOR inhibitor provided a synergistic effect by promoting cell death in PTEN-deficient and PTEN-intact endometrial cancer cells, particularly in PTEN-deficient carcinoma cells with up-regulated EGFR activation.

Introduction: Human potassium chloride cotransporter-1 (KCC1) gene is expressed in endometrial cancer and related to metastasis of endometrial cancer. However, whether KCC1 contributes to invasion and metastasis of endometrial cancer has not been thoroughly investigated. The purpose of this study is to research the alternation effect of insulin-like growth factor I (IGF-I) on the expression of KCC1 in endometrial cancer HEC-1B cells and to explore the mechanism of how KCC1 regulates the invasion ability of HEC-1B cells through the extracellular signal-regulated kinase (ERK) signaling pathway. Methods: First, the inhibitive effect of RNA interference to KCC1 was detected by semiquantitative reverse transcriptase-polymerase chain reaction. Western blot was used to measure expression changes of KCC1 after exposure to IGF-I in the HEC-1B cells. The change in quantity of phosphorylated ERK1/2 (p-ERK1/2) and cell invasion ability also were measured. After RNA interference and treatment with U0126, the quantity of p-ERK1/2 and the cell invasion ability were measured again. Results: After the application of IGF-I on the HEC-1B cells, the expression of KCC1 and p-ERK1/2 increased dramatically, and the cell invasion ability advanced. RNA interference could inhibit the expression of KCC1, and the quantity of p-ERK1/2 and the cell invasion ability decreased even under the effect of IGF-I. Furthermore, after treatment with U0126, the cell invasion ability no longer advanced even under the effect of IGF-I either. Conclusions: Insulin-like growth factors I can induce the upregulation of KCC1 gene, and KCC1 gene participates in the invasion ability of HEC-1B cells through the ERK signaling pathway.

Bladder carcinoma is a common malignancy with complicated treatment methods due to its heterogeneity. In this study, we focused on two bladder carcinoma cell lines, 5637 and T24, to compare their differences from the transcriptome level. RNA sequencing was used to generate the transcriptome data of the two cell line and the control cell line SV-HUC-1. Differentially expressed genes (DEGs) and differentially expressed microRNAs (miRNAs) of cell line 5637 and T24 were screened. Their annotation and analyses were conducted using gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) to predict their possible functions and pathways involved. Number of DEGs specific in cell line 5637, specific in cell line T24 and in both the cell lines was 880, 1512 and 1412, respectively. Number of differentially expressed miRNAs of the three categories was 7, 20 and 18, respectively. These DEGs and miRNAs participated in different biological processes and pathways, among which some were further verified by qRT-PCR. Interferon-stimulated genes (ISGs), including STAT1, TMEM173 and OAS3, were down-regulated in cell line 5637 compared to SV-HUC-1. NDOR1 and NDUFV1, genes related to mitochondrial metabolism, were up-regulated in cell line T24. miR-4257, miR-6733 and gene WNT9A and WNT10A were down-regulated in both the cell lines. Thus cell line 5637 might have lower chemotherapy resistance while T24 might exhibit abnormal mitochondrial metabolism. These results uncovered major differences between cell line 5637 and T24, which indicated the two cell lines, should be selectively used in bladder carcinoma research.
