Congeneric species often share ecological niche space resulting in competitive interactions that either limit co-occurrence or lead to niche partitioning. Differences in fundamental nutritional niches mediated through character displacement or isolation during evolution are potential mechanisms that could explain overlapping distribution patterns of congenerics. We directly compared nutritional requirements and tolerances that influence the fundamental niche of mule (Odocoileus hemionus) and white-tailed deer (O. virginianus), which occur in allopatry and sympatry in similar realized ecological niches across their ranges in North America. Digestible energy and protein requirements and tolerances for plant fiber and plant secondary metabolites (PSMs) of both deer species were quantified using in vivo digestion and intake tolerance trials with six diets ranging in content of fiber, protein, and PSMs using tractable deer raised under identical conditions in captivity. We found that compared with white-tailed deer, mule deer required 54% less digestible protein and 21% less digestible energy intake per day to maintain body mass and nitrogen balance. In addition, they had higher fiber, energy, and dry matter digestibility and produced glucuronic acid (a byproduct of PSM detoxification) at a slower rate when consuming the monoterpene α-pinene. The mule deers’ enhanced physiological abilities to cope with low-quality, chemically defended forages relative to white-tailed deer might minimize potential competitive interactions in shared landscapes and provide a modest advantage to mule deer in habitats dominated by low-quality forages.

Sagebrush ecosystems (Artemisia spp.) face many threats including large wildfires and conversion to invasive annuals, and thus are the focus of intense restoration efforts across the western United States. Specific attention has been given to restoration of sagebrush systems for threatened herbivores, such as Greater Sage-Grouse (Centrocercus urophasianus) and pygmy rabbits (Brachylagus idahoensis), reliant on sagebrush as forage. Despite this, plant chemistry (e.g., crude protein, monoterpenes and phenolics) is rarely considered during reseeding efforts or when deciding which areas to conserve. Near-infrared spectroscopy (NIRS) has proven effective in predicting plant chemistry under laboratory conditions in a variety of ecosystems, including the sagebrush steppe. Our objectives were to demonstrate the scalability of these models from the laboratory to the field, and in the air with a hyperspectral sensor on an unoccupied aerial system (UAS). Sagebrush leaf samples were collected at a study site in eastern Idaho, USA. Plants were scanned with an ASD FieldSpec 4 spectroradiometer in the field and laboratory, and a subset of the same plants were imaged with a SteadiDrone Hexacopter UAS equipped with a Rikola hyperspectral sensor (HSI). All three sensors generated spectral patterns that were distinct among species and morphotypes of sagebrush at specific wavelengths. The amount and composition of phytochemicals in forage plants influences habitat quality for wild herbivores. However, evaluating forage quality at fine resolutions across broad spatial extents (i.e., foodscapes) is challenging. Unmanned aerial systems (UAS) provide an avenue for bridging this gap in spatial scale. We evaluated the potential for UAS technology to accurately predict nutritional quality of sagebrush (Artemisia spp.) across landscapes. We mapped seasonal forage quality across two sites in Idaho, USA, with different mixtures of species but similar structural morphotypes of sagebrush. We classified the sagebrush at both study sites using structural features of shrubs with object-based image analysis and machine learning and linked this classification to field measurements of phytochemicals to interpolate a foodscape for each phytochemical with regression kriging. We compared fine-scale landscape patterns of phytochemicals between sites and seasons. Classification accuracy for morphotypes was high at both study sites (81–87%). Forage quality was highly variable both within and among sagebrush morphotypes. Coumarins were the most accurately mapped (r2 = 0.57–0.81), whereas monoterpenes were the most variable and least explained. Patches with higher crude protein were larger and more connected in summer than in winter. UAS allowed for a rapid collection of imagery for mapping foodscapes based on the phytochemical composition of sagebrush at fine scales but relatively broad extents. However, results suggest that a more advanced sensor (e.g., hyperspectral camera) is needed to map mixed species of sagebrush or to directly measure forage quality.

Red deer (Cervus elaphus) are mixed feeders that consume both herbaceous and woody plants. As a consequence, intensive browsing on trees where red deer populations are particularly dense often leads to serious conflicts between the interests of forestry and hunting stakeholders. Therefore, understanding the density of deer that forest system can bear requires the ability to measure amount of potential forage provided by non-commercial tree species within a stand that serve as forage. Our objective was to build models that estimate forage potential (i.e., maximum biomass edible and accessible for consumption by red deer per tree) based on tree size [i.e., stem base diameter (d0)]. We developed models for three tree species commonly consumed by red deer in Central Europe, aspen (Populus tremula), goat willow (Salix caprea) and rowan (Sorbus aucuparia). To construct models of forage potential, we harvested 380 young (age of 2–15 years) trees on 14 sites in the Western Carpathians of Slovakia. Tree biomass was first divided into three components (stem bark, branches and foliage), each of which was subdivided into edible and non-edible portions based on branch diameter and height from the ground and then dried and weighed. We then quantified edible biomass by tree species, tree component and season (growing vs. dormant). The total amount of tree mass (forage potential) that could be consumed by red deer generally increased with tree size, but the relative contributions of different tree components varied by tree size, tree species and season. Our models predicted that the maximum forage potential per tree was ~ 500 g in aspen, 350 g in goat willow and 300 g in rowan in summer, and nearly 500 g in aspen, 300 g in goat willow and 250 g in rowan in winter. Together with theoretical knowledge and practical experience, our forage potential models can be used to help forest and wildlife managers both to better measure edible tree biomass for red deer populations and how edible tree biomass might be used to minimize the risk of deer damaging commercially valuable trees.

Vegetation in the forest understory is a key food resource for wild ungulates like deer (Odocoileus spp.) because the amount of nutritious forage influences animal productivity and density. Therefore, measuring the abundance of understory vegetation available to wildlife populations is often a key objective for wildlife managers. Field-based methods for measuring understory vegetation across remote landscapes are time- and resource-intensive, so we compared estimates of understory vegetation density derived from airborne light detection and ranging (LiDAR) returns with vegetation biomass sampled directly on 65 field plots across 4 years and >250,000 hectares of xeric conifer forests in northeastern Washington. Large wild herbivores are important and natural components of forest ecosystems, but through their browsing activities have the potential to influence the structure and composition of forest communities, thus timber production and ecosystem dynamics. To examine the effects of browsing by wild herbivores on a young post-disturbance forest in the Kysuce region of northwestern Slovakia, we established two sets of 2 m radius plots, 15 within a fenced area (5.12 ha) that excluded large wild herbivores, and 15 within an adjacent unfenced area. In each plot, we recorded the species, tree height, stem base diameter, and mutual geographic positions of trees. When we compared tree community characteristics between the unfenced and fenced plots, we found fewer and smaller broadleaved tree species, except silver birch (Betula pendula Roth.) in the unfenced plots. Although common rowan (Sorbus aucuparia L.) was the dominant species within fenced plots, where some individuals were over 6.0 m tall, this species was rare outside the fenced area and usually did not exceed 1.5 m. In contrast, Norway spruce (Picea abies Karts L.) was more abundant and taller within the unfenced area, likely released from competition by suppression of broadleaved trees by herbivores. In addition, fenced plots also showed twice the tree species richness (Shannon index) of unfenced ones. Despite changes in tree communities, total aboveground biomass stock was only slightly but significantly lower in the unfenced than the fenced plots (29.6 kg per 10 m2 vs. 33.5 kg per 10 m2). Our study suggested that browsing pressure by large wild herbivores that focused on most broadleaved trees weakened interspecies competition and allowed the expansion of Norway spruce. As a consequence, converting spruce monocultures to mixed species stands is likely unrealistic when faced with heavy browsing pressure by wild large herbivores.

How intensely animals use habitat features depends on their functional properties (i.e., how the feature influences fitness) and the spatial and temporal scale considered. Herbivores that forage on chemically defended plants consume complex mixtures of plant secondary metabolites (PSMs). However, the mechanisms by which herbivores tolerate mixtures of PSMs are relatively poorly understood. As such, it remains difficult to predict how PSMs, singly or as complex mixtures, influence diet selection by herbivores. Although relative rates of detoxification of PSMs have been used to explain tolerance of PSMs by dietary specialist herbivores, few studies have used the rate of detoxification of individual PSMs to understand dietary preferences of individual herbivores for individual versus mixtures of PSMs. We coupled in vivo experiments using captive feeding trials with in vitro experiments using enzymatic detoxification assays to evaluate the dietary preferences and detoxification capacities of pygmy rabbits (Brachylagus idahoensis), dietary specialists on sagebrush (Artemisia spp.), and mountain cottontails (Sylvilagus nuttallii), dietary generalists. We compared preference for five single PSMs in sagebrush compared to a mixture containing those same five PSMs. We hypothesized that relative preference for individual PSMs would coincide with faster detoxification capacity for those PSMs by specialists and generalists. Pygmy rabbits generally showed little preference among individual PSMs compared to mixed PSMs, whereas mountain cottontails exhibited stronger preferences. Pygmy rabbits had faster detoxification capacities for all PSMs and consumed higher concentrations of individual PSMs versus a mixture than cottontails. However, detoxification capacity for an individual PSM did not generally coincide with preferences or avoidance of individual PSMs by either species. Cottontails avoided, but pygmy rabbits preferred, camphor, the PSM with the slowest detoxification rate by both species. Both species avoided β-pinene despite it having one of the fastest detoxification rate. Taken together our in vivo and in vitro results add to existing evidence that detoxification capacity is higher in dietary specialist than generalist herbivores. However, results also suggest that alternative mechanisms such as absorption and the pharmacological action of individual or mixtures of PSMs may play a role in determining preference of PSMs within herbivore species.

Merck & Co, Inc (Kenilworth, NJ) is investing in approaches to enrich clinical trial data and augment decision making through use of digital health technologies, outpatient sampling, and real‐time data access. As part of this strategy, a phase I study was conducted to explore a few technologies of interest. In this fixed‐sequence two‐period trial, 16 healthy subjects were administered 50‐mg once‐daily sitagliptin packaged in a bottle that electronically captured the date and time study medication was dispensed (period 1) and in a traditional pharmacy bottle (period 2). Dried blood spot samples were collected for sitagliptin concentration analysis on select study days, both in clinic and at home, with collection time recorded using an electronic diary in period 1 and by clinic staff in period 2. Study results demonstrated the feasibility and subject acceptance of collecting digital adherence data and outpatient dried blood spot samples in clinical trials and highlighted areas for future improvements.

Large herbivores prefer to consume plants with high concentration of digestible carbohydrates and protein, whereas fire spread is favored by foliage characteristics that induce rapid ignition and high radiant energy. We tested in eight Patagonian woody plants whether foliar traits that determine dry matter and nitrogen digestibility to browsing herbivores were related to four variables that determine their flammability. We found a negative relation between digestibility and flammability. Fiber content (i.e. Neutral and Acid Detergent Fiber), was positively related with heat emission, maximum temperature and flame duration, being negatively related to time to ignition. PCA indicated that digestibility was negatively related with all variables associated with flammability. Digestibility decreased throughout the growing season, whereas flammability showed no clear seasonal trend. Fiber content was correlated with time to ignition and maximum temperature during most of the growing season, except in late spring. We conclude that highly digestible plants are likely to be less flammable than fiber rich ones, and that fiber content may serve as an indicator of seasonal changes in flammability of foliage. Because leaf traits are likely to influence habitat selection and movement by herbivores as well as spread and severity of fires, further studies linking foliar shape variables and plant architecture with their chemical composition may improve current mechanistic models of herbivore and fire behavior.

1.Assessing habitat quality is a primary goal of ecologists. However, evaluating habitat features that relate strongly to habitat quality at fine-scale resolutions across broad-scale extents is challenging. Unmanned aerial systems (UAS) provide an avenue for bridging the gap between relatively high spatial resolution, low spatial extent field-based habitat quality measurements and lower spatial resolution, higher spatial extent satellite-based remote sensing. Our goal in this study was to evaluate the potential for UAS structure from motion (SfM) to estimate several dimensions of habitat quality that provide potential security from predators and forage for pygmy rabbits (Brachylagus idahoensis) in a sagebrush-steppe environment. 2.At the plant and patch scales, we compared UAS-derived estimates of vegetation height, volume (estimate of food availability), and canopy cover to estimates from ground-based terrestrial laser scanning (TLS), and field-based measurements. Then, we mapped habitat features across two sagebrush landscapes in Idaho, USA, using point clouds derived from UAS SfM. 3.At the individual plant scale, the UAS-derived estimates matched those from TLS for height (r2 = 0.85), volume (r2 = 0.94), and canopy cover (r2 = 0.68). However, there was less agreement with field-based measurements of height (r2 = 0.67), volume (r2 = 0.31), and canopy cover (r2 = 0.29). At the patch scale, UAS-derived estimates provided a better fit to field-based measurements (r2 = 0.51-0.78) than at the plant scale. Landscape-scale maps created from UAS were able to distinguish structural heterogeneity between key patch types. 4.Our work demonstrates that UAS was able to accurately estimate habitat heterogeneity for a key terrestrial vertebrate at multiple spatial scales. Given that many of the vegetation metrics we focus on are important for a wide variety of species, our work illustrates a general remote sensing approach for mapping and monitoring fine-resolution habitat quality across broad landscapes for use in studies of animal ecology, conservation, and land management. Lay SummaryBehavior of pygmy rabbits varied seasonally to accommodate both thermal challenges and predation risk. We quantified selection of rest sites and activity during summer and winter to evaluate behavioral tradeoffs between thermal and security resources. Our work suggests that behavior might buffer individuals of this species against a changing climate, and it highlights the need for incorporating behavior into models of how individuals and populations of animals respond to climate change.

This chapter presents a qualitative study about a graduate-level design thinking course which resulted in students recognizing themselves as empowered designers in their respective fields. An empowered designer is someone who recognizes themselves as a designer and is able to engage in design purposefully. The study took place within the context of a nontraditional interdisciplinary instructional design program. Participants of this study were a group of students from diverse professional backgrounds including K-12 and higher education, nursing, graphic design, and corporate training. Student reflective journals and an assignment paper provided descriptive data for this study. The data was analyzed using naturalistic thematic methods. The results suggest that adult learners, who do not identify themselves as designers, become empowered designers after being introduced to design concepts. The study implications discuss how empowered designers become active participants of future design projects who take initiative in finding new design problems. In the implications, we recommend that educators place more emphasis on recognizing, nurturing, and empowering student habits and beliefs related to being a designer.

Forested lands in the western USA have undergone changes in management and condition that are resulting in a shift towards climax vegetation. These changes can influence the quality and quantity of forage for herbivores that rely on early-seral plants. To evaluate how management of forested landscapes might affect nutrition for Shiras moose (A. a. shirasi) at large spatial scales, we focused on shrubs and evaluated summer diet composition, forage availability, and forage quality across 21 population management units encompassing >36,000 km2 in northern Idaho, USA. We identified 17 shrub species in the diets of moose, 11 of which comprised the bulk of the diets. These forage shrubs varied markedly in both energy (mean digestible energy for leaves ranged from 9.62 to 12.89 kJ/g) and protein (mean digestible protein for leaves ranged from 1.73 to 7.90%). By adapting established field sampling methods and integrating recent advances in remote sensing analyses in a modeling framework, we predicted approximations of current and past (i.e., 1984) quantities of forage shrubs across northern Idaho. We also created a qualitative index of population trend for moose across population management units using harvest data. Predicted quantities of forage shrubs varied widely across the study area with generally higher values at more northern latitudes. The quantity of forage shrubs was estimated to have declined over the past 30 years in about half of the population management units, with the greatest declines predicted for high-energy forage species. The population trend index was correlated with the percent change in availability of moderate-energy forage shrubs, indicating that availability of forage shrubs and change in availability over time might be affecting population dynamics for moose in northern Idaho. Our study highlights the importance of assessing how changes in forest management across broad spatiotemporal extents could affect wildlife and their habitats.

Mule (Odocoileus hemionus) and white‐tailed deer (Odocoileus virginianus) are congeneric and share similar life histories, yet their distribution is segregated across much of North America. Extensive research on both species within and outside their zone of co‐occurrence has not fully explained these distribution patterns, especially the potential role of diet and foraging behavior. Therefore, we used a common garden experiment to compare diet composition, diet quality, foraging behavior, and intake of tractable mule and white‐tailed deer foraging together within the dry Douglas‐fir (Pseudotsuga menziesii)/ponderosa pine (Pinus ponderosa) forests of northeastern Washington. We sampled at 21 0.5‐ha sites from June to August 2016. We used standard bite count techniques coupled with forage biomass sampling, behavioral observations, and nutritional analyses to compare the foraging ecology of the two species. Mule and white‐tailed deer had similar activity patterns. However, mule deer took larger bites and harvested food faster than white‐tailed deer, and white‐tailed deer consumed more diverse but higher‐quality diets than mule deer. These differences resulted in mule deer acquiring ~25% more dry matter and digestible energy per day. About 90% of the diets consumed by both deer species consisted of deciduous shrubs and forbs, and they selected many of the same plant species. However, overall diet composition was 38% dissimilar, with mule deer consuming diets that were more likely to contain shrubs with higher levels of tannins and lower levels of dry matter digestibility than diets eaten by white‐tailed deer. Dietary overlap was greatest at both very low and very high forage biomass, indicating potential for modest resource competition or partitioning. Our research provides evidence that differences in diet composition of mule and white‐tailed deer do not merely reflect differences in habitat selection, but also suggest the species differ in their fundamental nutritional niches.

Sagebrush ( Artemisia spp.) habitat in the Intermountain West is one of the most endangered ecosystems in North America due, in part, to fire, climate change, and anthropogenic disturbances. However, restoration efforts rarely consider the dietary quality of sagebrush that is conserved or restored despite growing evidence that it is an influential parameter explaining habitat use by many important wild and domestic herbivores. The objective of this study was to evaluate the capacity of near-infrared reflectance spectroscopy (NIRS) to measure and monitor the dietary quality of sagebrush. Leaf samples were collected from two sagebrush species over two seasons and three sites in Idaho, USA. We developed calibration equations for crude protein (CP), dry matter digestibility (DMD), 1,8-cineole (cineole), and total polyphenolics. The coefficient of determination ( r 2 ) and ratio of performance to deviation (RPD) were 0.93 and 3.5 for CP, 0.83 and 1.8 for DMD, 0.64 and 1.5 for cineole, and 0.64 and 1.6 for total polyphenolics. These results indicate that NIRS may offer a rapid, noninvasive, diagnostic tool for assessing dietary quality of sagebrush, but future research should explore the potential for development of improved prediction equations and in situ analysis of sagebrush dietary quality with field spectroscopy.

Understanding habitat use by animals requires understanding the simultaneous tradeoffs between food and predation risk within a landscape. Quantifying the synergy between patches that provide quality food and those that are safe from predators at a scale relevant to a foraging animal could better reveal the parameters that influence habitat selection. To understand more thoroughly how animals select habitat components, we investigated tradeoffs between diet quality and predation risk in a species endemic to sagebrush Artemisia spp. communities in North America, the pygmy rabbit Brachylagus idahoensis. This species is a rare example of a specialist herbivore that relies almost entirely on sagebrush for food and cover. We hypothesized that pygmy rabbits would forage in areas with low food risk (free of plant secondary metabolites, PSMs) and low predation risk (high concealment). Summary Small herbivores face risks of predation while foraging and are often forced to trade off food quality for safety. Life history, behaviour, and habitat of predator and prey can influence these trade‐offs. We compared how two sympatric rabbits (pygmy rabbit, Brachylagus idahoensis; mountain cottontail, Sylvilagus nuttallii) that differ in size, use of burrows, and habitat specialization in the sagebrush‐steppe of western North America respond to amount and orientation of concealment cover and proximity to burrow refuges when selecting food patches. We predicted that both rabbit species would prefer food patches that offered greater concealment and food patches that were closer to burrow refuges. However, because pygmy rabbits are small, obligate burrowers that are restricted to sagebrush habitats, we predicted that they would show stronger preferences for greater cover, orientation of concealment, and patches closer to burrow refuges. We offered two food patches to individuals of each species during three experiments that either varied in the amount of concealment cover, orientation of concealment cover, or distance from a burrow refuge. Both species preferred food patches that offered greater concealment, but pygmy rabbits generally preferred terrestrial and mountain cottontails preferred aerial concealment. Only pygmy rabbits preferred food patches closer to their burrow refuge. Different responses to concealment and proximity to burrow refuges by the two species likely reflect differences in perceived predation risks. Because terrestrial predators are able to dig for prey in burrows, animals like pygmy rabbits that rely on burrow refuges might select food patches based more on terrestrial concealment. In contrast, larger habitat generalists that do not rely on burrow refuges, like mountain cottontails, might trade off terrestrial concealment for visibility to detect approaching terrestrial predators. This study suggests that body size and evolutionary adaptations for using habitat, even in closely related species, might influence anti‐predator behaviors in prey species.

The increasing intensity of forestry practices, especially using herbicides to promote early survival and growth of planted conifers, has the potential to influence the abundance of nutritious, shade-intolerant forbs and shrubs on which black-tailed deer (Odocoileus hemionus columbianus) and other forest ungulates depend. Therefore, we compared summer forage resources and nutrition of female black-tailed deer in stands that received herbicide treatments paired with stands that did not in Douglas-fir/western hemlock forests of western Washington. We investigated how these responses changed as forests aged from early seral to canopy closure (2–20 years post-harvest) in both treatments, and also in unmanaged mid-seral stands (30–90 years old). We measured the biomass and nutritional quality of understory vegetation and the diet composition, diet quality and nutrient intake of tractable black-tailed deer within each paired stand. We found that herbicides did not affect the rate of closure of the overstory canopy, but influenced plant species composition of the understory vegetation and reduced total and non-conifer understory biomass, primarily in the first 9 years before canopy closure. Herbicides also reduced the biomass of two of the six plant functional groups, evergreen shrubs and deciduous shrubs and trees, but did not reduce forb, graminoid, or fern biomass. Herbicides reduced bite size, harvest rate, daily dry matter intake and daily digestible energy (DE) intake of deer, but increased dietary digestible protein (DP) content, and these effects occurred primarily in the first 3 years post-treatment. Biomass of plants suitable for black-tailed deer, and the deers’ bite rate, daily feeding time, dietary DE, and daily digestible DP intake did not respond to herbicides. Our yearling and sub-adult, non-lactating deer met requirements for yearling growth and adult maintenance for dietary DE in 78%, and DE intake in 27% of plots, but only met the higher requirements for lactating females in 5% of plots. Range contractions in the Great Basin over the last century suggest that American pikas (Ochotona princeps) might be highly sensitive to climate change. However, documentation of pikas residing at relatively warm, low-elevation sites has recently shed new light on the possible resilience of pika populations to warmer ambient conditions when they have access to cooler microhabitats for thermoregulation. To provide insight into the possible behavioral mechanisms of adaptation to warmer habitats, we investigated activity patterns, foraging behavior, and space use of a population of pikas living in an atypical, warmer habitat at Craters of the Moon National Monument and Preserve, an extensive lava flow surrounded by high desert grassland and sagebrush communities in southern Idaho. We compared pika behavior at this site to that of a population in a more typical alpine habitat at Grays Peak in the nearby Pioneer Mountains in Idaho. Specifically, we evaluated and compared activity patterns and foraging behavior of pikas in relation to temperature, time of day, and month. Pikas at Craters of the Moon were less active aboveground during all times of the day from late May until mid-August, compared to those at Grays Peak. Surface temperatures were warmer across the entire season at Craters of the Moon than at Grays Peak, and pikas exhibited a crepuscular activity pattern that coincided with variations in surface temperature at Craters of the Moon. Pikas were the least active during the midday, when the surface temperatures were highest and the insulating effect of the lava tubes (i.e., the difference between surface and crevice temperatures) was most pronounced. Pikas at Craters of the Moon spent less time haying and displayed fewer territorial behaviors than pikas at Grays Peak, but both groups filled a similar number of hay piles. The vegetation community was less diverse and sparser at Craters of the Moon than at Grays Peak, and consequently, the vegetation that was consumed and cached reflected these differences. Our results expand the body of literature about American pikas at their environmental limits, and this study is the first step in identifying the unique suite of behaviors that pikas use to persist in a seemingly inhospitable environment at Craters of the Moon.

Dried blood spot (DBS) sample collection has gained increased interest across the pharmaceutical industry as a potential alternative to plasma for pharmacokinetic (PK) evaluations. However, regulatory guidelines and examples of late-stage clinical trial applications in the literature are lacking. This paper communicates Merck’s strategy for the implementation of DBS exemplified by experience on a late-stage program (MK-8931). In this program, DBS was proposed as the sole matrix for phase 3 studies to decrease logistical burden in an aging target patient population (Alzheimer’s disease). In vitro and bioanalytical tests demonstrated initial method feasibility and suitability for further evaluations in the clinic. An in vivo dataset was developed initially in healthy subjects (phase 1 study) and then in patients (phase 2/3 study) to establish a quantitative relationship between the blood and plasma concentrations (bridging dataset) using descriptive and population PK analyses. This allowed for PK conclusions to be seamlessly drawn across the clinical program without impact from the choice of matrix. This integrated information package (in vitro, bioanalytical and clinical) was presented to major regulatory agencies (FDA and EMA) for regulatory input. Based on this package, regulatory concurrence was gained on accepting DBS as the sole matrix in late-stage clinical trials.

When selecting habitats, herbivores must weigh multiple risks, such as predation, starvation, toxicity, and thermal stress, forcing them to make fitness trade-offs. Here, we applied the method of paired comparisons (PC) to investigate how herbivores make trade-offs between habitat features that influence selection of food patches. The method of PC measures utility and the inverse of utility, relative risk, and makes trade-offs and indifferences explicit by forcing animals to make choices between two patches with different types of risks. Using a series of paired-choice experiments to titrate the equivalence curve and find the marginal rate of substitution for one risk over the other, we evaluated how toxin-tolerant (pygmy rabbit Brachylagus idahoensis) and fiber-tolerant (mountain cottontail rabbit Sylviagus nuttallii) herbivores differed in their hypothesized perceived risk of fiber and toxins in food. Pygmy rabbits were willing to consume nearly five times more of the toxin 1,8-cineole in their diets to avoid consuming higher levels of fiber than were mountain cottontails. Fiber posed a greater relative risk for pygmy rabbits than cottontails and cineole a greater risk for cottontails than pygmy rabbits. Our flexible modeling approach can be used to (1) quantify how animals evaluate and trade off multiple habitat attributes when the benefits and risks are difficult to quantify, and (2) integrate diverse risks that influence fitness and habitat selection into a single index of habitat value. This index potentially could be applied to landscapes to predict habitat selection across several scales.

Greater sage-grouse (Centrocercus urophasianus) and Columbian sharp-tailed grouse (Tympanuchus phasianellus columbianus) have declined substantially in Washington, USA, primarily because native shrub-steppe has been converted to agriculture. In response, state and federal agencies have acquired and restored habitat, and augmented and reintroduced grouse to suitable areas. We examined how sympatric, translocated sage-grouse and sharp-tailed grouse used space and selected habitats within their spring–summer home ranges and at nest sites within remnant shrub-steppe surrounded by a matrix of cropland in eastern Washington. Because their life-history requirements differ, we expected extensive habitat partitioning between species. Using radiolocations of ≥43 birds of each species, we found that sage-grouse had larger spring–summer home ranges than sharp-tailed grouse, and the composite of home ranges for sharp-tailed grouse fell almost completely within the composite of home ranges for sage-grouse. By creating resource utilization function models using radiolocations of ≥53 birds of each species, we found that areas of highest predicted intensity of use for both species overlapped by >50%, even at the top 5% quantile. Both species used restored fields and areas farther from trees and roads or distribution lines more intensely. Sage-grouse used less rugged areas more intensely, and both species used 3 levels of shrub cover equally. To compare selection of nest sites relative to available sites for nesting in both species, we created resource selection function models for ≥30 birds of each species and found that sage-grouse selected areas farther from distribution lines, whereas sharp-tailed grouse selected restored fields. When we examined vegetation characteristics used by female sage-grouse and sharp-tailed grouse at nest sites using a case-control, use versus non-use design for ≥26 birds of each species, we found sage-grouse used areas with greater shrub cover, lower annual forb cover, and taller perennial grasses, whereas sharp-tailed grouse used areas with greater perennial grass cover and taller perennial grasses and forbs. When we compared habitat features measured at nest sites between species, we found sage-grouse used areas with greater moderate and dense shrub cover, lower sparse shrub cover, less restored fields, higher patch diversity, and areas farther from distribution lines than sharp-tailed grouse. These differences resulted in only 38% overlap of areas within the top quartile of relative selection values for nest sites by the 2 species, and

Small mammals in habitats with strong seasonal variation in the thermal environment often exhibit physiological and behavioral adaptations for coping with thermal extremes and reducing thermoregulatory costs. Burrows are especially important for providing thermal refuge when above-ground temperatures require high regulatory costs (e.g., water or energy) or exceed the physiological tolerances of an organism. Our objective was to explore the role of burrows as thermal refuges for a small endotherm, the pygmy rabbit (Brachylagus idahoensis), during the summer and winter by quantifying energetic costs associated with resting above and below ground. We used indirect calorimetry to determine the relationship between energy expenditure and ambient temperature over a range of temperatures that pygmy rabbits experience in their natural habitat. We also measured the temperature of above- and below-ground rest sites used by pygmy rabbits in eastern Idaho, USA, during summer and winter and estimated the seasonal thermoregulatory costs of resting in the two microsites. Although pygmy rabbits demonstrated seasonal physiological acclimatization, the burrow was an important thermal refuge, especially in winter. Thermoregulatory costs were lower inside the burrow than in above-ground rest sites for more than 50% of the winter season. In contrast, thermal heterogeneity provided by above-ground rest sites during summer reduced the role of burrows as a thermal refuge during all but the hottest periods of the afternoon. Our findings contribute to an understanding of the ecology of small mammals in seasonal environments and demonstrate the importance of burrows as thermal refuge for pygmy rabbits.

When animals select habitats, they integrate a suite of behaviors that are influenced by multiple competing resource requirements. Resources that influence decisions about habitat use are likely to differ across spatial scales and hierarchical behaviors. At coarser scales, animals are expected to select resources that are critical to fitness, and at finer scales, to intensively use resources that enhance fitness. Our goal was to contrast habitat selection at two hierarchical behavioral levels (patch selection and intensity of patch use) to test hypotheses about how resources shape habitat use. We applied a two-stage hurdle model to quantify both initial selection and intensity of use of resource patches by a burrowing herbivore, the pygmy rabbit (Brachylagus idahoensis). We expected security from predation to influence initial selection of patches, and forage availability to influence intensity of use of selected patches. We monitored locations of adults fitted with radio-collars during winter and summer. We measured vegetation, burrow characteristics, and concealment cover within patches that were used and unused by rabbits, and we quantified burrow densities surrounding patches in a GIS. Selection of used patches from available patches was largely influenced by security resources (presence and proximity of burrows, shrub height, and woody ground cover) during both seasons. Intensity of patch use was also influenced by forage availability and, consequently, differed between seasons. During winter, sagebrush is the primary forage, and greater sagebrush canopy within a patch was associated with increased intensity of use. During summer, rabbits more intensively used patches with greater availability of herbaceous forage. Elucidating how animals make choices about habitats across a diversity of spatial and temporal scales will continue to increase our understanding of the factors that govern distribution of populations and movements of individuals. By extending these concepts to hierarchical behaviors, we can enhance insights into the processes that shape the patterns of habitat use we observe.

Both temperature and diet quality play an important role in the time and energy budgets of small mammalian herbivores. However, little is known about how temperature and diet quality interact to influence diet selection, nutrient extraction, and energetics, and how these effects might differ among species. Therefore, we examined the effects of diet quality and temperature on aspects of the foraging ecology of two species of lagomorphs, pygmy rabbits (Brachylagus idahoensis), which are small dietary specialists, and mountain cottontail rabbits (Sylvilagus nuttallii), which are larger dietary and habitat generalists. In a series of feeding experiments, we investigated 1) the effects of temperature on selection of plant fiber and the plant secondary metabolite 1,8 cineole in their diets, 2) effects of temperature and plant fiber on daily intake, digestion, and passage of food, 3) effects of plant fiber and 1,8 cineole on resting metabolic rate, and 4) how these interactions differ between the rabbit species. Both species chose to eat more total food and a greater proportion of high fiber food that passed more quickly through the digestive system in colder temperatures. However, temperature did not affect how much 1,8 cineole the rabbits consumed nor how thoroughly they digested food. Food quality affected how well they digested the dry matter in the food, but not their resting metabolic rate. Understanding how the interactions between ambient temperature and food quality affect selection of diets and intake by small mammalian herbivores, and the physiological mechanisms governing these choices, is useful for predicting how these species might respond to changes in both temperature and food quality and inform conservation and restoration strategies.

Because small ruminants (

Fire suppression has increased the canopy cover, stocking density, and fuel loads of dry forests in the inland northwestern United States. These changes not only lead to high-severity, stand-replacing wildfires that may threaten human safety and further alter natural forest communities, but also reduce light transmission to the forest floor, which can limit production of herbaceous and woody understory vegetation that serves as forage for wild ungulates. Therefore, forest managers have implemented fuels reduction treatments that reduce the density of standing trees in mature stands to restore historic conditions, promote healthy forests, and reduce wildfire risk. To test the hypothesis that these treatments also improve the quality and quantity of forage resources for wild ungulates, we measured biomass, plant species composition, and nutritional quality of understory vegetation in 106 untreated and treated 0.5-ha plots that ranged from 1 to 22 years since treatment and 2–100% canopy cover in Douglas-fir (Pseudotsuga menziesii)/ponderosa pine (Pinus ponderosa) forests in the Colville National Forest of northeastern Washington. On average, fuels reduction treatments halved basal area and overstory canopy cover, doubling understory vegetation biomass. The most nutritious plants (i.e., composing suitable biomass) and plants most likely to be consumed by deer (i.e., acceptable biomass) increased with decreasing forest canopy cover, and increased with time since treatment for approximately 14 years post-harvest, but suitable biomass began declining thereafter. Understory plant biomass also responded to a range of site, weather, and environmental variables, especially those influencing moisture gradients. Our results indicate that active management prescriptions intended to reduce fuel loads posed by overstocked conifers can also increase the amount of high quality forage for deer and other wild ungulates, especially when carefully planning the timing, intensity, and rotation schedules of fuels reduction treatments.

Heterogeneous vegetation structure can create a variable landscape of predation risk—a fearscape—that influences the use and selection of habitat by animals. Mapping the functional properties of vegetation that influence predation risk (e.g., concealment and visibility) across landscapes can be challenging. Traditional ground-based measures of predation risk are location specific and limited in spatial resolution. We demonstrate the benefits of terrestrial laser scanning (TLS) to map the properties of vegetation structure that shape fearscapes. We used TLS data to estimate the concealment of prey from multiple vantage points, representing predator sightlines, as well as the visibility of potential predators from the locations of prey. TLS provides a comprehensive data set that allows an exploration of how habitat changes may affect prey and predators. Together with other remotely sensed imagery, TLS could facilitate the scaling up of fearscape analyses to promote the management and restoration of landscapes.

For herbivores, nutrient intake is limited by the relatively low nutritional quality of plants and high concentrations of potentially toxic defensive compounds (plant secondary metabolites [PSMs]) produced by many plants. In response to phytochemical challenges, some herbivores selectively forage on plants with higher nutrient and lower PSM concentrations relative to other plants. Pygmy rabbits (Brachylagus idahoensis) are dietary specialists that feed on sagebrush (Artemisia spp.) and forage on specific plants more than others within a foraging patch. We predicted that the plants with evidence of heavy foraging (browsed plants) would be of higher dietary quality than plants that were not browsed (unbrowsed). We used model selection to determine which phytochemical variables best explained the difference between browsed and unbrowsed plants. Higher crude protein increased the odds that plants would be browsed by pygmy rabbits and the opposite was the case for certain PSMs. Additionally, because pygmy rabbits can occupy foraging patches (burrows) for consecutive years, their browsing may influence the nutritional and PSM constituents of plants at the burrows. In a post hoc analysis, we did not find a significant relationship between phytochemical concentrations, browse status, and burrow occupancy length. We concluded that pygmy rabbits use nutritional and chemical cues while making foraging decisions.
