Generative AI such as those with large language models have created opportunities for innovative assessment design practices. Due to recent technological developments, there is a need to know the limits and capabilities of generative AI in terms of simulating cognitive skills. Assessing student critical thinking skills has been a feature of assessment for time immemorial, but the demands of digital assessment create unique challenges for equity, academic integrity and assessment authorship. Educators need a framework for determining their assessments vulnerability to generative AI to inform assessment design practices. This paper presents a framework that explores the capabilities of the LLM ChatGPT4 application, which is the current industry benchmark. This paper presents the Mapping of questions, AI vulnerability testing, Grading, Evaluation (MAGE) framework to methodically critique their assessments within their own disciplinary contexts. This critique will provide specific and targeted indications of their questions vulnerabilities in terms of the critical thinking skills. This can go on to form the basis of assessment design for their tasks.

Many real-world spatially related problems, including river-basin planning and management, give rise to geographical information system based decision making, since the performance of spatial policy alternatives were traditionally and are still often represented by thematic maps. Advanced technologies and approaches, such as geographical information systems (GIS), offer a unique opportunity to tackle spatial problems traditionally associated with more efficient and effective data collection, analysis, and alternative evaluation. This paper discusses the advantages and challenges of the use of digital spatial data and geographical information systems in river basis management. Spatial data on social, environmental and other spatial conditions for the study area of 451.77 km2, the Slovenian part of the Sotla river basin, are used to study the GIS capabilities of supporting spatial decisions in the framework of river basin management.

8. Chinese Students' Metaphorical Descriptions of Thai Teachers at a Thai University

- Abstract
Purpose: This paper discusses Chinese students' negative and positive written feedback about Thai teachers using metaphorical descriptions and the links between it and their classroom experiences. Methodology: An open-response questionnaire was employed to collect the data from 21 Chinese female students. The questionnaire provided both positive and negative "people" or "thing" metaphors of Thai teachers for students' selection. In addition, students were allowed to use their own metaphors to describe their teachers. Data was analyzed by using open and axial coding techniques. Findings: The results revealed that Chinese students were able to compare Thai teachers with either a "person" or "thing" metaphor and could write a metaphorical description that reflected different aspects of their instructors' teaching, both positively and negatively. The quality of their descriptions was rich enough to link with their classroom learning experiences. Both positive and negative "people" and "thing" metaphorical descriptions were associated with three different viewpoints: academic, power dynamics, and emotion. The positive "people" metaphorical descriptions were linked to four classroom issues: knowledge and experience, teaching style, motivation, and guardian/protector. In contrast, the positive "thing" and negative "people" and "thing" metaphorical descriptions were linked to three classroom issues: knowledge and experience, teaching style, and emotion. Significance: These findings help to strengthen Thai-Sino understanding of the relationship between Chinese students and Thai teachers. Findings also suggested that Chinese students' metaphorical feedback should be used with the non-metaphorical assessment form to evaluate and improve Thai teachers' instructional practices in the Thai-Chinese student exchange curriculum.

10. The influence of protected natural and cultural heritage on land management/market: The case of Slovenian natural protected areas

This article is focused on finding problems in land use domain in the areas of protected natural and cultural heritage. In the paper, the influence of special regulation in the natural and cultural protected areas on land management is presented. The paper gives an overview on history of cultural heritage and nature protection initiatives in Slovenia and provides a review on basic EU and international initiatives, conventions in this field. For the case of Slovenian rural land market, it highlights the problem of complex institutional regulations relating to land management in the protected areas, which affect mostly local people. Here, the impact of the protected regimes, the case of pre-emption right, on land management and consequently spatial development in local communities is stressed, which is an important topic in particular in less developed regions since restriction of land use often means more complex, costly and time lasting procedures in land management and less opportunities as the consequence.

AbstractThe increasing occurrence of disasters worldwide has motivated researchers to continuously evaluate potential technological advances to support disaster management (DM) as well as emergency response. The advent of Volunteered Geographic Information (VGI) offers the possibility of near real-time data collection or the possibility of massive disaster and post-disaster data collection. VGI is a type of geographic information provided by volunteers who have no formal training in geoinformatics and geographic information systems (GIS). The objective of this review aimed to examine research publications that address VGI in the context of DM, focusing on VGI data quality. From the collected metadata of publications published in the Web of Science (WoS) on crowdsourcing and VGI in the context of DM, we extracted and processed those articles related to data quality using the text mining method and a bibliometric approach. The research addresses the quality of VGI data and its fit for purpose for DM studies that rely on accurate and reliable geographic information for successful management through identified topics. The article concludes by highlighting the potential of VGI to provide valuable information for DM, while also pointing to the need for further research to identify and improve the quality of VGI data.

Abstract Background Disruption of Ca2+ homeostasis after calcium electroporation (CaEP) in tumors has been shown to elicit an enhanced antitumor effect with varying impacts on healthy tissue, such as endothelium. Therefore, our study aimed to determine differences in Ca2+ kinetics and gene expression involved in the regulation of Ca2+ signaling and homeostasis, as well as effects of CaEP on cytoskeleton and adherens junctions of the established endothelial cell lines EA.hy926 and HMEC-1. Methods CaEP was performed on EA.hy926 and HMEC-1 cells with increasing Ca2+ concentrations. Viability after CaEP was assessed using Presto Blue, while the effect on cytoskeleton and adherens junctions was evaluated via immunofluorescence staining (F-actin, α-tubulin, VE-cadherin). Differences in intracellular Ca2+ regulation ([Ca2+]i) were determined with spectrofluorometric measurements using Fura-2-AM, exposing cells to DPBS, ionomycin, thapsigargin, ATP, bradykinin, angiotensin II, acetylcholine, LaCl3, and GdCl3. Molecular distinctions were identified by analyzing differentially expressed genes and pathways related to the cytoskeleton and Ca2+ signaling through RNA sequencing. Results EA.hy926 cells, at increasing Ca2+ concentrations, displayed higher CaEP susceptibility and lower survival than HMEC-1. Immunofluorescence confirmed CaEP-induced, time- and Ca2+-dependent morphological changes in EA.hy926’s actin filaments, microtubules, and cell–cell junctions. Spectrofluorometric Ca2+ kinetics showed higher amplitudes in Ca2+ responses in EA.hy926 exposed to buffer, G protein coupled receptor agonists, bradykinin, and angiotensin II compared to HMEC-1. HMEC-1 exhibited significantly higher [Ca2+]i changes after ionomycin exposure, while responses to thapsigargin, ATP, and acetylcholine were similar in both cell lines. ATP without extracellular Ca2+ ions induced a significantly higher [Ca2+]i rise in EA.hy926, suggesting purinergic ionotropic P2X and metabotropic P2Y receptor activation. RNA-sequencing analysis showed significant differences in cytoskeleton- and Ca2+-related gene expression, highlighting upregulation of ORAI2, TRPC1, TRPM2, CNGA3, TRPM6, and downregulation of TRPV4 and TRPC4 in EA.hy926 versus HMEC-1. Moreover, KEGG analysis showed upregulated Ca2+ import and downregulated export genes in EA.hy926. Conclusions Our finding show that significant differences in CaEP response and [Ca2+]i regulation exist between EA.hy926 and HMEC-1, which may be attributed to distinct transcriptomic profiles. EA.hy926, compared to HMEC-1, displayed higher susceptibility and sensitivity to [Ca2+]i changes, which may be linked to overexpression of Ca2+-related genes and an inability to mitigate changes in [Ca2+]i. The study offers a bioinformatic basis for selecting EC models based on research objectives.

18. Changes in Black Truffle (Tuber melanosporum) Aroma during Storage under Different Conditions

The enticing aroma of truffles is a key factor for their culinary value. Although all truffle species tend to be pricy, the most intensely aromatic species are the most sought after. Research into the aroma of truffles encompasses various disciplines including chemistry, biology, and sensory science. This study focusses on the chemical composition of the aroma of black truffles (Tuber melanosporum) and the changes occurring under different storage conditions. For this, truffle samples were stored under different treatments, at different temperatures, and measured over a total storage time of 12 days. Measurements of the truffle aroma profiles were taken with SPME/GC–MS at regular intervals. To handle the ample data collected, a systematic approach utilizing multivariate data analysis techniques was taken. This approach led to a vast amount of data which we made publicly available for future exploration. Results reveal the complexity of aroma changes, with 695 compounds identified, highlighting the need for a comprehensive understanding. Principal component analyses offer initial insights into truffle composition, while individual compounds may serve as markers for age (formic acid, 1-methylpropyl ester), freshness (2-Methyl-1-propanal; 1-(methylthio)-propane), freezing (tetrahydrofuran), salt treatment (1-chloropentane), or heat exposure (4-hydroxy-3-methyl-2-butanone). This research suggests that heat treatment or salt contact significantly affects truffle aroma, while freezing and cutting have less pronounced effects in comparison. The enrichment of compounds showing significant changes during storage was investigated with a metabolomic pathway analysis. The involvement of some of the enriched compounds on the pyruvate/glycolysis and sulfur pathways was shown.

19. Sustainable Grassland-Management Systems and Their Effects on the Physicochemical Properties of Soil

Grassland covers approximately 17.4% of Europe’s land area, stores about 20% of the world’s soil carbon and has the potential to sequester carbon. With the help of sustainable management systems, grasslands could reduce greenhouse gases and act as a terrestrial sink for atmospheric CO2. In this study, we will investigate the effect of grassland management (cutting, grazing, and a combination of the two) and soil depth (0–10, 10–20, 20–30 cm) on the physical (volumetric water content—VWC, bulk density—BD, porosity—POR, mass consisting of coarse fragments—FC) and chemical properties of soil (organic carbon—SOC, inorganic carbon—SIC, total carbon—STC, total nitrogen—STN, organic matter—SOM, C/N ratio, pH) in Central European lowlands. The management system affected BD, SOC and STN and tended to affect VWC and STC in the first soil depth only. Grazing and the combined system stored greater amounts of STN, SOC and STC and had higher BDs at the surface (0–10 cm) compared to the cutting system. Most soil properties were influenced by soil depth, with C/N ratio and BD increasing and SOC, STC, STN, SOM, VWC and POR decreasing with depth. Our study highlights an opportunity for grassland users to improve soil quality, reduce fossil fuel usage and improve animal welfare through their management systems and argues that systems such as grazing and the combined system should be promoted to mitigate climate change.

A collaborative trial involving 16 participants from nine European countries was conducted within the NORMAN network in efforts to harmonise suspect and non-target screening of environmental contaminants in whole fish samples of bream (Abramis brama). Participants were provided with freeze-dried, homogenised fish samples from a contaminated and a reference site, extracts (spiked and non-spiked) and reference sample preparation protocols for liquid chromatography (LC) and gas chromatography (GC) coupled to high resolution mass spectrometry (HRMS). Participants extracted fish samples using their in-house sample preparation method and/or the protocol provided. Participants correctly identified 9–69 % of spiked compounds using LC-HRMS and 20–60 % of spiked compounds using GC-HRMS. From the contaminated site, suspect screening with participants’ own suspect lists led to putative identification of on average ∼145 and ∼20 unique features per participant using LC-HRMS and GC-HRMS, respectively, while non-target screening identified on average ∼42 and ∼56 unique features per participant using LC-HRMS and GC-HRMS, respectively. Within the same sub-group of sample preparation method, only a few features were identified by at least two participants in suspect screening (16 features using LC-HRMS, 0 features using GC-HRMS) and non-target screening (0 features using LC-HRMS, 2 features using GC-HRMS). The compounds identified had log octanol/water partition coefficient (KOW) values from −9.9 to 16 and mass-to-charge ratios (m/z) of 68 to 761 (LC-HRMS and GC-HRMS). A significant linear trend was found between log KOW and m/z for the GC-HRMS data. Overall, these findings indicate that differences in screening results are mainly due to the data analysis workflows used by different participants. Further work is needed to harmonise the results obtained when applying suspect and non-target screening approaches to environmental biota samples.

Summary: Background: While vaccines have established utility against COVID-19, phase 3 efficacy studies have generally not comprehensively evaluated protection provided by previous infection or hybrid immunity (previous infection plus vaccination). Individual patient data from US government-supported harmonized vaccine trials provide an unprecedented sample population to address this issue. We characterized the protective efficacy of previous SARS-CoV-2 infection and hybrid immunity against COVID-19 early in the pandemic over three-to six-month follow-up and compared with vaccine-associated protection. Methods: In this post-hoc cross-protocol analysis of the Moderna, AstraZeneca, Janssen, and Novavax COVID-19 vaccine clinical trials, we allocated participants into four groups based on previous-infection status at enrolment and treatment: no previous infection/placebo; previous infection/placebo; no previous infection/vaccine; and previous infection/vaccine. The main outcome was RT-PCR-confirmed COVID-19 >7–15 days (per original protocols) after final study injection. We calculated crude and adjusted efficacy measures. Findings: Previous infection/placebo participants had a 92% decreased risk of future COVID-19 compared to no previous infection/placebo participants (overall hazard ratio [HR] ratio: 0.08; 95% CI: 0.05–0.13). Among single-dose Janssen participants, hybrid immunity conferred greater protection than vaccine alone (HR: 0.03; 95% CI: 0.01–0.10). Too few infections were observed to draw statistical inferences comparing hybrid immunity to vaccine alone for other trials. Vaccination, previous infection, and hybrid immunity all provided near-complete protection against severe disease. Interpretation: Previous infection, any hybrid immunity, and two-dose vaccination all provided substantial protection against symptomatic and severe COVID-19 through the early Delta period. Thus, as a surrogate for natural infection, vaccination remains the safest approach to protection. Funding: National Institutes of Health.

Modern environmental and economic challenges in waste management require transition from linear to circular economic flow. In practice, this entails considerable challenges that include the change of material circle flux, the application of mathematical modelling and the use of life cycle thinking – also in the field of recovered wood (RW). To this end, the reverse logistics process model CATWOOD (CAscade Treatment of WOOD) with mechanistic modelling for detailed planning of the RW reverse flow with regular collection, innovative (cascade) sorting based on RW quality and environmentally sound recovery has been designed. As a decision support, the quantitative methods of life-cycle assessment (LCA) and societal life-cycle costing (SLCC) have been incorporated into the CATWOOD, which can choose among a few alternative scenarios. A case study has been performed in the Posavje region in Slovenia, which has discovered that reverse logistics scenarios for reuse are environmentally friendlier than those for recycling or energy recovery, but also more costly, mainly because of extensive manual labour needed and less heavy technology involved in sorting and recovery processes. Sensitivity analysis has exposed that modifying the values of the input parameters may change the final LCA and SLCC results in scenarios observed.

Summary: Background: Topical steroids are used widely to manage excessive inflammation and hypergranulation in burns; however, their use is controversial, and current evidence is largely anecdotal. Topical KENACOMB is a steroid preparation consisting of triamcinolone acetonide, neomycin, gramicidin, and nystatin, and it is standard of care at the Royal Brisbane and Women's Hospital burns unit. To our knowledge, there is no published literature that reports the use of KENACOMB to treat wound-associated inflammation and hypergranulation. Objective: To synthesise current evidence surrounding the efficacy and safety of topical steroid use in treating inflammation and hypergranulation in burns patients. We also describe the use of topical KENACOMB in our burns unit. Methods: A systematic review of PubMed, Cochrane, and EMBASE databases was performed. Articles published in English that reported the use of topical steroids for granulation tissue or inflammation in burn wounds or skin graft donor sites were included. Results: We identified 350 articles, of which six met inclusion criteria. Four studies presented primary patient data, and two studies reported the results of surveys of burns unit professionals. A total of 54 patients were included across all studies, and no control group was reported in any study. Studies reported rapid improvements in healing, with 86.6%–100% of wounds showing complete reepithelialisation following treatment. Reported adverse outcomes included skin thinning, atrophy of granulation tissue, systemic side effects, and local wound infection. Conclusions: This review highlights the paucity of conclusive evidence on the outcomes of topical steroids in treating inflammation and hypergranulation in burns and donor sites. While KENACOMB has shown efficacy in treating these wound types in our local experience, there is limited research available on the product. There is a clear need for quality research on the use of topical steroids in burns patients to better inform its ongoing clinical use.

IntroductionIn calcium electroporation (CaEP), electroporation enables the cellular uptake of supraphysiological concentrations of Ca2+, causing the induction of cell death. The effectiveness of CaEP has already been evaluated in clinical trials; however, confirmatory preclinical studies are still needed to further elucidate its effectiveness and underlying mechanisms. Here, we tested and compared its efficiency on two different tumor models to electrochemotherapy (ECT) and in combination with gene electrotransfer (GET) of a plasmid encoding interleukin-12 (IL-12). We hypothesized that IL-12 potentiates the antitumor effect of local ablative therapies as CaEP and ECT.MethodsThe effect of CaEP was tested in vitro as well as in vivo in murine melanoma B16-F10 and murine mammary carcinoma 4T1 in comparison to ECT with bleomycin. Specifically, the treatment efficacy of CaEP with increasing calcium concentrations alone or in combination with IL-12 GET in different treatment protocols was investigated. We closely examined the tumor microenvironment by immunofluorescence staining of immune cells, as well as blood vessels and proliferating cells.ResultsIn vitro, CaEP and ECT with bleomycin reduced cell viability in a dose-dependent manner. We observed no differences in sensitivity between the two cell lines. A dose-dependent response was also observed in vivo; however, the efficacy was better in 4T1 tumors than in B16-F10 tumors. In 4T1 tumors, CaEP with 250 mM Ca resulted in more than 30 days of growth delay, which was comparable to ECT with bleomycin. In contrast, adjuvant peritumoral application of IL-12 GET after CaEP prolonged the survival of B16-F10, but not 4T1-bearing mice. Moreover, CaEP with peritumoral IL-12 GET modified tumor immune cell populations and tumor vasculature.ConclusionsMice bearing 4T1 tumors responded better to CaEP in vivo than mice bearing B16-F10 tumors, even though a similar response was observed in vitro. Namely, one of the most important factors might be involvement of the immune system. This was confirmed by the combination of CaEP or ECT with IL-12 GET, which further enhanced antitumor effectiveness. However, the potentiation of CaEP effectiveness was also highly dependent on tumor type; it was more pronounced in poorly immunogenic B16-F10 tumors compared to moderately immunogenic 4T1 tumors.

Abstract Lifestyle modifications could prevent almost one-third to one-half of all cancer cases. The awareness of cancer risk factors could motivate people to make such changes in their behaviors and lifestyles. This work aims to investigate the cancer awareness level in the Pakistani population. Telephone interviews of 657 individuals in Pakistan were carried out using the Cancer Awareness Measure (CAM) and Cancer Awareness Measure–MYthical Causes Scale (CAM-MY). We observed that participants scored significantly better on the CAM scale than the CAM-MY scale, and CAM scores were negatively associated with CAM-MY scores. Years of formal education or a biology major at undergraduate or graduate level did not affect our population's cancer awareness levels. Age displayed a weak but statistically significant negative association with CAM scores. Most participants failed to identify modifiable cancer risk factors, e.g., low physical activity. Efforts should be made to improve awareness of modifiable risk factors. We observed that brief training sessions could markedly improve people's understanding of cancer risk factors and myths.

Non‐targeted screenings (NTS) are essential tools in different fields, such as forensics, health and environmental sciences. NTSs often employ mass spectrometry (MS) methods due to their high throughput and sensitivity in comparison to, for example, nuclear magnetic resonance–based methods. As the identification of mass spectral signals, called annotation, is labour intensive, it has been used for developing supporting tools based on machine learning (ML). However, both the diversity of mass spectral signals and the sheer quantity of different ML tools developed for compound annotation present a challenge for researchers in maintaining a comprehensive overview of the field. In this work, we illustrate which ML‐based methods are available for compound annotation in non‐targeted MS experiments and provide a nuanced comparison of the ML models used in MS data analysis, unravelling their unique features and performance metrics. Through this overview we support researchers to judiciously apply these tools in their daily research. This review also offers a detailed exploration of methods and datasets to show gaps in current methods, and promising target areas, offering a starting point for developers intending to improve existing methodologies. Present-day airline industry is quite a competitive field and crew scheduling represents one of the crucial problems due to significant impact on the airline’s cost. The crew scheduling problem is based on the assignment of crew members to operate different tasks of route. The main goal of this paper is to provide an analysis and a solution to one of the biggest problems detected on a small airport in the Serbia - the problem of ground crew scheduling. The paper presents the main characteristics, goals and limitations of a real-life problem identified at this small airport. In order to solve the problem, we developed a dynamic discrete simulation model. The model is developed in a spreadsheet environment of Microsoft Excel. Some of the main limitations found in the development of the model are strong constraints and multiple goals. The model presented in the paper is designed as a useful management tool for smaller airports and is aimed at the improvement of operative processes.

Establishing a multi-purpose cadastre, especially in terms of upgrading cadastral contents with the various spatial data, such as land use, is a challenge in Slovenia and internationally. Land use strongly affects spatial planning, development, and management, so high-quality spatial integration of the land cadastre with spatial plans data is crucial for effective land management. In the first part of the article, we reviewed the literature and documents that prescribe guidelines for the development of the land cadastre; we use these guidelines as a basis for developing a proposed method of linking and harmonising the data of the land cadastre with the spatial plan data. Land use is specified in spatial plans, and we linked it to the graphical and attribute land cadastre data layer. We tested the method in selected study areas in Prekmurje with a high-quality cadastre in the municipalities of Kramarovci and Nemčavci. As a result, we presented land use data directly in the land cadastre database, which requires simultaneous land use and cadastre maintenance. Based on the results for selected cadastral municipalities, we critically evaluated the proposed method.

Digital cadastral maps with accompanying land-related attributes have become a fundamental dataset for many application fields, e.g., spatial planning and development, protecting state lands, securing of land tenure, facilitating land reforms, agriculture, forestry, land management, taxation, etc. In order to fulfil its main objectives, cadastral data needs to be available and accessible, which is, among the others, emphasized also within the United Nations Framework for Effective Land Administration (FELA). This is not only important on the national level but also beyond, including at the European level where use cases and consequently demand for pan-European data sets have evolved in recent years. In order to satisfy these needs, several initiatives regarding cadastral and other geospatial data have been launched in the last 20 years. It started with the Permanent Committee on Cadastre in the European Union, the European Land Information Service, INSPIRE, UN-GGIM Europe and recent European policies on open data and high-value datasets. Our main question is, did those initiatives result in the possibility of building a cadastral map of Europe or not? Is it possible to create a cadastral map of Europe on the desktop or an open online GIS application? Within the paper, we take the opportunity to reflect on the development and implementation of European spatial data infrastructure (INSPIRE) with the main focus on the availability and accessibility of cadastral data. We also take into consideration other European initiatives related to cadastral data. The overall findings show that there is still work to be carried out. Technological developments and recent policy initiatives will certainly be drivers for future improvement.

Abstract Background Prevention of metastatic invasion is one of the main challenges in the treatment of alveolar rhabdomyosarcoma. Still the therapeutic options are limited. Methods Treatment effects were first evaluated by cell viability, migration, invasion, and colony forming assays on the alveolar rhabdomyosarcoma cell line RH-30 in comparison with healthy primary cells. Results Initial anti-tumor screenings of all substances analyzed in this study, identified the plant extract of Vincetoxicum arnottianum (VSM) as the most promising candidate, harboring the highest anti-metastatic potential. Those significant anti-motility properties were proven by a reduced ability for migration (60%), invasion (99%) and colony formation (61%) under 48 h exposure to 25 μg/ml VSM. The restricted motility features were due to an induction of the stabilization of the cytoskeleton – actin fibers were 2.5-fold longer and were spanning the entire cell. Decreased proliferation (PCNA, AMT, GCSH) and altered metastasis (e. g. SGPL1, CXCR4, stathmin) marker expression on transcript and protein level confirmed the significant lowered tumorigenicity under VSM treatment. Finally, significant alterations in the cell metabolism were detected for 25 metabolites, with levels of uracil, N-acetyl serine and propanoyl phosphate harboring the greatest alterations. Compared to the conventional therapy with cisplatin, VSM treated cells demonstrated a similar metabolic shutdown of the primary cell metabolism. Primary control cells were not affected by the VSM treatment. Conclusions This study revealed the VSM root extract as a potential, new migrastatic drug candidate for the putative treatment of pediatric alveolar rhabdomyosarcoma with actin filament stabilizing properties and accompanied by a marginal effect on the vitality of primary cells. The present paper discusses the heterogeneity of the apartment market. For this purpose, we have developed the model for the mass valuation of apartments in the Republic of Slovenia. The construction of the mass valuation model is based on the generalised additive model approach. In this paper, the development of the model is presented. In the experimental part, the analysis of the results of the two models is performed. The dependent variable (the price of an apartment) is distributed according to the Gaussian and the gamma distributions. Particular attention has been paid to the impact of the transaction time on the apartments’ transaction value. The results of the model are also compared with the results of the mass valuation model in the Republic of Slovenia, which is carried out cyclically and iteratively, the results of which depend on the results (and mass valuation models) of previous cycles.

VACTERL association is a rare malformation complex consisting of vertebral defects, anorectal malformation, cardiovascular defects, tracheoesophageal fistulae with esophageal atresia, renal malformation, and limb anomalies. According to current knowledge, VACTERL is based on a multifactorial pathogenesis including genomic alterations. This study aimed to improve the understanding of the genetic mechanisms in the development of VACTERL by investigating the genetic background with a focus on signaling pathways and cilia function. The study was designed as genetic association study. For this, whole-exome sequencing with subsequent functional enrichment analyses was performed for 21 patients with VACTERL or a VACTERL-like phenotype. In addition, whole-exome sequencing was performed for three pairs of parents and Sanger-sequencing was performed for ten pairs of parents. Analysis of the WES-data revealed genetic alteration in the Shh- and Wnt-signaling pathways. Additional performed functional enrichment analysis identified an overrepresentation of the cilia, including 47 affected ciliary genes with clustering in the DNAH gene family and the IFT-complex. The examination of the parents showed that most of the genetic changes were inherited. In summary, this study indicates three genetically determined damage mechanisms for VACTERL with the potential to influence each other, namely Shh- and Wnt-signaling pathway disruption, structural cilia defects and disruption of the ciliary signal transduction.

48. Quantification of Ergot Alkaloids via Lysergic Acid Hydrazide—Development and Comparison of a Sum Parameter Screening Method

Ergot alkaloids are a group of mycotoxins occurring in products derived from various grasses (e.g., rye) and have been regulated in the EU recently. The new maximum levels refer to the sum of the six most common ergot alkaloids in their two stereoisomeric forms in different food matrices. Typically, these twelve compounds are individually quantified via HPLC-MS/MS or -FLD and subsequently summed up to evaluate food safety in a time-consuming process. Since all these structures share the same ergoline backbone, we developed a novel sum parameter method (SPM) targeting all ergot alkaloids simultaneously via lysergic acid hydrazide. After extraction and clean-up, in analogy to the current European standard method EN 17425 (ESM) for ergot alkaloid quantitation, the samples were derivatized by an optimized hydrazinolysis protocol, which allowed quantitative conversion after 20 min at 100 °C. The new SPM was evaluated against another established HPLC-FLD-based method (LFGB) and the HPLC-MS/MS-based ESM using six naturally contaminated rye and wheat matrix reference materials. While the SPM provided comparable values to the ESM, LFGB showed deviating results. Determined recovery rates, limits of detection and quantification of all three employed methods confirm that the new SPM is a promising alternative to the classical approaches for ergot alkaloid screening in food.

49. Video Making to Support Science Investigations

- Abstract
Through the science curriculum, teachers can use students' passion for screen content to motivate them to explore innovative new ways to bring their science learning to life through video making. Communicating science concepts using multimodal literacies can be a particularly powerful way for students to learn about science, especially when they experience difficulties with traditional literacies (Dezuanni & Woods, 2014). With 'planning and conducting' and 'communicating' being two of the five sub-strands in the inquiry skills, students can record and share their investigation ideas through the use of different representations, text types and modes. With multimodal texts including typed text, still and moving images, and sound such as spoken voice, music and sound effects, there are many possibilities for how students can conduct and communicate investigation ideas. By extending students' skills for making multimodal texts using a range of technologies, their ability to engage audiences will further develop beyond the typical slideshow presentation. While slideshows will always be a valued example of a multimodal text, evolving digital technologies continue to provide 'easy to use' video and animation applications and software to make moving images with sound and text in addition to static slides. To support students in making videos, teachers can lead the way through making videos to support science education programs. Teacher-made videos produced in an hour or two can be more time efficient to use compared with spending several hours searching for appropriate videos made by others. Not only do students prefer screen content that is fun and engaging with youthful presenters, it also needs to be made with high production values and have educational relevance across learning areas to meet their ever-increasing high expectations (Cunningham et al. 2016, p.4). Teacher-made and student-made video texts can meet student learning interests and expectations within the science learning area, if made with care and attention for quality. The Curriculum Connections STEM (CC STEM) YouTube channel was developed by researchers at Queensland University of Technology based on findings from a national research project: the Australian Screen Content in Education Project. A key project finding was that YouTube is highly popular as a teaching tool and that video production is becoming a common activity in classrooms across the curriculum, as teachers and students turn to multimodal communication to develop and express knowledge in new ways. This article is an example of an activity inspired by CC STEM resources for science students in upper primary school.

Unmanned aerial vehicles, equipped with various sensors and devices, are increasingly used to acquire geospatial data in geodesy, geoinformatics, and environmental studies. In this context, a new research and professional field has been developed – UAV photogrammetry – dealing with photogrammetry data acquisition and data processing, acquired by unmanned aerial vehicles. In this study, we analyse the selected factors that impact the quality of data provided using UAV photogrammetry, with the focus on positional accuracy; they are discussed in three groups: (a) factors related to the camera properties and the quality of images; (b) factors related to the mission planning and execution; and (c) factors related to the indirect georeferencing of images using ground control points. These selected factors are analysed based on the detailed review of relevant scientific publications. Additionally, the influence of the number of ground control points and their spatial distribution on point clouds' positional accuracy has been investigated for the case study. As the conclusion, key findings and recommendations for UAV photogrammetric projects are given; we have highlighted the importance of suitable lighting and weather conditions when performing UAV missions for spatial data acquisition, quality equipment, appropriate parameters of UAV data acquisition, and a sufficient number of ground control points, which should be determined with the appropriate positional accuracy and their correct distribution in the field.
