Importin ��s from Oryza sativa (Os), Lycopersicon esculentum (Le), and Arabidopsis thaliana (At) were compared. Multiple sequence alignments of amino acid sequences were generated using ClustalW in MEGA7.0. The sequence alignments obtained were used as input for the neighbor-joining method using MEGA7.0 to construct the phylogenetic tree. For phylogenetic tree construction, a bootstrap method with 1,000 replications was used for test of phylogeny. Scale bar indicates 0.2 amino acid substitution per site. Fig. S3. During cavern leaching for salt cavern gas storage in China, cavern shape irregularity has become a growing problem. It reduces cavern volume and the utilization ratio of the salt formations. The collapse of the irregular parts may cause downhole accidents and threaten cavern stability. In this paper, shape repair of an irregular salt cavern by re-leaching under gas blanket is proposed for the first time in China. The technological process of the cavern repair under gas blanket is introduced. A mathematical model is set up to predict the real-time three-dimensional shape of the salt cavern during the repair process. The upper and lower limit of the gas-brine interface, the depth of the leaching tubing and the salt-brine mass transfer rates are included in the model. The coupled equations for shape prediction are derived, which consider the effects of brine concentration and velocity. A VC++ program is developed based on these equations. To verify the proposed model, the repair process of a gourd-shaped salt cavern is carried out in the laboratory and is simulated by the program. The comparison of the experimental and simulation results shows that the proposed model has a high accuracy. The prediction model and the VC++ program are used in the field repair work of JT86 cavern of Jintan underground gas storage. The irregular parts are well repaired, and the cavern is enlarged by about 9000 m3. The sonar imaging and injection-production data of the repaired cavern coincide well with the simulation results obtained by the proposed model, which further validates the reliability and precision of the model.

ZNF580, a newly found C2H2 zinc finger transcription factor, was first described by Zhang (GenBank ID: AF184939). Emerging evidence has suggested that reactive oxygen species (ROS) play an important role in redox-sensitive signal transduction, and the vascular endothelium plays a critical role in the vascular inflammatory response. In this communication, we present evidence for the potential role of ZNF580 in hydrogen peroxide (H2O2)-regulated inflammation-related signaling pathways. In a human endothelial cell hybridoma line (EA.hy926), ZNF580 levels were markedly upregulated with H2O2 stimulation in different concentrations (0–400 μM) and at different time-points (0–6 h). H2O2 promoted the rapid translocation of p65 from the cytoplasm into the nucleus according to immunocytochemistry staining. In subsequent research, inhibition of NF-κB by pyrrolidine dithiocarbamate (PDTC, a selective chemical inhibitor of NF-κB) was shown to block the upregulated expression of ZNF580 that was induced by H2O2. Furthermore, transient transfection of ZNF580 resulted in an increase of the pro-inflammatory cytokine interleukin-8 (IL-8) 3.01 ± 0.05 folds according to real-time RT-PCR and ELISA assays, which also showed significantly enhanced motility of human acute monocytic leukemia cells (THP-1). These results suggest that H2O2 upregulates the expression of ZNF580 via the NF-κB signaling pathway, and overexpression of ZNF580 plays a critical role in augmenting the release of pro-inflammatory cytokine IL-8.
