Abstract

Bartonellae are hemotropic bacteria, agents responsible for emerging zoonoses. These Alphaproteobacteria are heme auxotroph which must import heme for supporting their growth, as they cannot synthesize it. Therefore, Bartonella genome encodes for a complete heme uptake system allowing the transportation of this compound in the cytoplasm. Heme has also been shown to be used as an iron source for Bartonella. Similarly to other bacteria which use heme as an iron source, Bartonellae must degrade it to allow the release of iron. For Bartonella, the genes cluster devoted to the heme uptake system contains a gene encoding for a polypeptide that shares homologies with heme trafficking or degrading enzymes. Using complementation of an E. coli mutant strain impaired in heme degradation, we demonstrated that HemS from Bartonella henselae allows the release of iron from heme. Recombinant HemS of B. henselae binds heme and can degrade it in the presence of a suitable electron donor. Knocking down the expression of HemS in B. henselae reduces B. henselae ability to face H2O2 exposure. Bartonellae encode for four or five outer membrane Heme binding proteins. The structural genes of these highly homologous proteins are expressed differently according to oxygen, temperature, and heme concentration. These proteins were hypothesized to be involved in various cellular processes according to their ability to bind heme and their regulation profile. In this report, we investigated the roles of the four Heme binding proteins of Bartonella henselae. We show that these proteins are involved in the defense against oxidative stress, colonization of endothelial cells and survival inside the flea, Les Bartonelles sont des bactéries hémotropes responsables de zoonoses émergentes. Ces Alphaprotéobactéries sont auxotrophes pour l'hème et doivent donc l'importer du milieu extérieur pour croître. Les Bartonelles possèdent un système complet de transport de l'hème permettant de transporter ce composé dans le cytoplasme. Chez Bartonella, il a été montré que l'hème pouvait être utilisé comme source de fer. Comme pour d'autres bactéries utilisant l'hème comme une source de fer, Bartonella doit dégrader l'hème pour libérer le fer. Chez Bartonella, un ensemble de gènes codant pour le système de transport de l'hème, contient un gène codant pour un polypeptide (HemS) présentant des homologies avec des protéines liant ou dégradant l'hème. En utilisant des expériences de complémentation de mutants d'E. coli incapables de dégrader l'hème, nous avons mis en évidence que HemS de Bartonella henselae permet la libération du fer de l'hème. HemS purifié lie l'hème et le dégrade en présence de donneurs d'électrons. La diminution du niveau de HemS chez B. henselae décroit sa capacité de survivre à une exposition à H2O2. Les Bartonelles expriment quatre ou cinq protéines de la membrane externe ayant la capacité de fixer l'hème. Les gènes de structure de ces protéines sont exprimés différemment en fonction de paramètres comme la température ainsi que la concentration en oxygène ou en hème. Ces protéines ont été proposées comme étant impliquées dans divers processus cellulaires étant donné leur profil d'expression. Dans ce manuscrit, nous montrons que ces protéines sont impliquées dans la défense contre le stress oxydatif, la colonisation des cellules endothèliales et la survie dans la puce

20. Various Profiles of tet Genes Addition to tet (X) in Riemerella anatipestifer Isolates From Ducks in China.

Abstract

To investigate tetracycline resistance and resistant genotype in Riemerella anatipestifer , the tetracycline susceptibility of 212 R. anatipestifer isolates from China between 2011 and 2017 was tested. The results showed that 192 of 212 (90.6%) R. anatipestifer isolates exhibited resistance to tetracycline (the MICs ranged from 4 to 256 μg/ml). The results of PCR detection showed that, 170 of 212 (80.2%) R. anatipestifer isolates possessed the tet (X) gene. Other genes, including tet (A), tet (M), tet (Q), tet (O), tet (B), and tet (O/W/32/O), were found at frequencies of 20.8, 4.7, 1.4, 0.9, 0.9, and 0.5%, respectively. However, tet (C), tet (E), tet (G), tet (K), and tet (W) were not detected in any isolate. In these tet gene positive strains, 31 (14.6%), 2 (0.9%), 5 (2.4%), 1 (0.5%), 3 (1.4%) were detected containing tet (A)/ tet (X), tet (M)/ tet (O), tet (M)/ tet (X), tet (O)/ tet (X), and tet (Q)/ tet (X) simultaneously, respectively. One isolates, R131, unexpectedly contained three tet genes, i.e., tet (M), tet (O), and tet (X). Sequence analysis of the tet gene ORFs cloned from R. anatipestifer isolates confirmed that tet (A), tet (B), tet (M), tet (O), tet (Q) and an unusual mosaic tet gene tet (O/W/32/O) were present in R. anatipestifer . The MIC results of R. anatipestifer ATCC 11845 transconjugants carrying tet (A), tet (B), tet (M), tet (O), tet (O/W/32/O), tet (Q), and tet (X) genes exhibited tetracycline resistance with MIC values ranging from 4 to 64 μg/ml. Additionally, the tet (X) gene could transfer into susceptible strain via natural transformation (transformation frequencies of ~10 -6 ). In conclusion, the tet (A), tet (B), tet (M), tet (O), tet (O/W/32/O), tet (Q), and tet (X) genes were found and conferred tetracycline resistance in R. anatipestifer isolates. Moreover, the tet (X) is the main mechanism of tetracycline resistance in R. anatipestifer isolates. To our knowledge, this is the first report of tet (A), tet (B), tet (M), tet (O), tet (Q), and mosaic gene tet (O/W/32/O) in R. anatipestifer .
