Interpretation and inversion of P-T (Pressure-Temperature) paths of high-pressure (HP) and ultrahigh-pressure (UHP) metamorphic rocks are critical to understanding the subduction and exhumation histories of an orogen. The Sulu orogen constitutes the eastern part of the Dabie-Sulu orogen, in which, HP metamorphic rocks are relatively poorly as studied compared to the well-documented UHP metamorphic rocks. This study presents the P-T paths of HP eclogites constructed by petrography, mineral chemistry, and pseudosection modelling by using the GeoPS software within the MnNCFMASKHTO (Mn-CaO-FeO-MgO-Al2O3-SiO2-K2O-H2O-TiO2-O2) system. Sample 19LR70–1 mainly consists of omphacite, garnet, quartz, minor rutile, ilmenite, and amphibole. Sample 19LR71–1 is mainly composed of omphacite, garnet, kyanite, and accessory minerals of rutile and amphibole. These two eclogite samples recorded clockwise P-T paths with peak P-T conditions of P ≥ 21.3 kbar, T ≥ 755°C and P ≥ 23.0 kbar, T ≥ 871°C, respectively. Well-correlated U-Pb ages and trace element data show that the protoliths of eclogites were 655–699 Ma and 676–754 Ma, the metamorphic ages were 212–232 Ma, and the time of partial melting were 221–225 Ma. Combined with regional geological data, we conjectured that before 240 Ma, the South China plate was subducted to the North China craton; subsequently, at 240–220 Ma, HP eclogite facies metamorphism occurred at a depth of 60–70 km. Isoprenoids are used in many commercial applications and much work has gone into engineering microbial hosts for their production. Isoprenoids are produced either from acetyl-CoA via the mevalonate pathway or from pyruvate and glyceraldehyde 3-phosphate via the 1-deoxy-D-xylulose 5-phosphate (DXP) pathway. Saccharomyces cerevisiae exclusively utilizes the mevalonate pathway to synthesize native isoprenoids and in fact the alternative DXP pathway has never been found or successfully reconstructed in the eukaryotic cytosol. There are, however, several advantages to isoprenoid synthesis via the DXP pathway, such as a higher theoretical yield, and it has long been a goal to transplant the pathway into yeast. In this work, we investigate and address barriers to DXP pathway functionality in S. cerevisiae using a combination of synthetic biology, biochemistry and metabolomics. We report, for the first time, functional expression of the DXP pathway in S. cerevisiae. Under low aeration conditions, an engineered strain relying solely on the DXP pathway for isoprenoid biosynthesis achieved an endpoint biomass 80% of that of the same strain using the mevalonate pathway.

X-ray free-electron lasers (XFELs) provide very intense X-ray pulses suitable for macromolecular crystallography. Each X-ray pulse typically lasts for tens of femtoseconds and the interval between pulses is many orders of magnitude longer. Here we describe two novel acoustic injection systems that use focused sound waves to eject picoliter to nanoliter crystal-containing droplets out of microplates and into the X-ray pulse from which diffraction data are collected. The on-demand droplet delivery is synchronized to the XFEL pulse scheme, resulting in X-ray pulses intersecting up to 88% of the droplets. We tested several types of samples in a range of crystallization conditions, wherein the overall crystal hit ratio (e.g., fraction of images with observable diffraction patterns) is a function of the microcrystal slurry concentration. We report crystal structures from lysozyme, thermolysin, and stachydrine demethylase (Stc2). Additional samples were screened to demonstrate that these methods can be applied to rare samples.

The Anshan–Benxi area, situated in the northeast of the North China Craton (NCC), is home to not only the oldest rocks in China (~3.8 Ga) but also a diverse range of granitoids dated between 3.8 and 2.5 Ga. The Lianshanguan batholith, covering an area of approximately 250 km2 with an east–west trend, predominantly consists of syenogranites (K2O > 4 wt. % and K2O/Na2O ratios > 1.3). Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) U-Pb analyses of the two syenogranites yielded concordant ages of 2541 ± 22 and 2512 ± 13 Ma, respectively. These syenogranites had zircon εHf(t) values ranging from −20 to +4.9 with two-stage Hf model ages (TDM2(Hf)) spanning 3.9–2.7 Ga. Based on petrological, geochemical, and isotopic characteristics, we conclude that the Lianshanguan syenogranites are mainly resulted from the reworking of complicated Eoarchean–Mesoarchean crustal materials, possibly with a small proportion of ~2.7 Ga juvenile crustal materials. When compared with coeval syenogranites from the Northern Liaoning and Western Liaoning–Eastern Hebei areas, ~2.5 Ga syenogranites from the Anshan–Benxi area displayed more complicated TDM2(Hf) ages, hinting at a pronounced late Neoarchean reworking of the Eoarchean to Mesoarchean continental crust (including metasedimentary sources) primarily in the Anshan–Benxi region of the North China Craton.
