Objective To explore the influencing factors for short-term prognosis after endovascular treatment in elderly patients with posterior circulation infarction (POCI). Methods A retrospective analysis was performed on the clinical data of 50 elderly patients with POCI who received endovascular treatment in the Nuclear Industry 416 Hospital from January 2019 to December 2020. The clinical data included patients’ preoperative general status (age, sex, smoking history, and alcohol abuse history), past disease history (diabetes, hypertension, hyperuricemia, and transient ischemic attack), post-onset indicators [onset-to-door time (ODT), door-to-needle time (DNT), NIH Stroke Scale (NIHSS) on admission], and postoperative conditions (immediate postoperative NIHSS, revascularization, and postoperative complications). The patients were divided into good prognosis group and poor prognosis group according to modified Rankin Scale (mRS) score 90 d after endovascular treatment and were analyzed for the differences in the above indicators. Meanwhile, a multivariate logistic regression analysis was performed on the influencing factors for patients’ short-term prognosis after endovascular treatment. Results There were 20 and 30 patients in the good prognosis group and poor prognosis group, respectively, demonstrating significant differences between the two groups in nine indicators [systolic blood pressure, glycosylated hemoglobin (HbA1c), serum uric acid, total cholesterol, postoperative modified Thrombolysis in Cerebral Infarction (mTICI) grade, preoperative NIHSS score, immediate postoperative NIHSS score, ODT, and postoperative stroke-related pneumonia] (t=2.30-4.13,χ2=6.35,7.07,P

Forkhead box O transcriptional factors, especially FoxO1 and FoxO3a, play critical roles in physiologic and pathologic immune responses. However, the function of FoxO4, another main member of the FoxO family, in lymphoid cells is still poorly understood. Here, we showed that loss of FoxO4 in T cells augmented IFN-[gamma] production of Th1 cells in vitro. Correspondingly, conditional deletion of FoxO4 in CD4* T cells enhanced T cell-specific responses to Listeria monocytogenes infection in vivo. Genome-wide occupancy and transcriptomic analyses identified Dkk3 (encoding the Dickkopf-3 protein) as a direct transcriptional target of FoxO4. Consistent with the FoxO4-DKK3 relationship, recombinant DKK3 protein restored normal levels of IFN-[gamma] production in FoxO4-deficient Th1 cells through the downregulation of lymphoid enhancer-binding factor 1 (Left1) expression. [Purpose / Significance] In order to quickly obtain a massive amount of data of land transfer on the Internet, with the help of the current popular method of data collection and analysis-web crawler technology, the new hot spots of rural land transfer in China are excavated and sorted out from the perspective of news and information. [Method / Process] Taking People's Daily as the target web page, we search keywords on the web page and use Python to write and design programs to crawl the news documents of farmland circulation on the web page. and We then use the obtained more than 2 500 pieces of news as data to analyze the status, content and problems of agricultural land transfer by using text analysis methods and drawing Python word cloud diagrams. [Results / Conclusions] Crawling the web news information, through statistical analysis of the collected news content, after hot spot mining, it is found that China is attaching more importance to the transfer of agricultural land and increased publicity in this regard, The construction of agricultural land transfer and precision poverty alleviation, and rural tourism and cooperatives are closely connected with one another. At last we analyze the existing problems in above-mentioned three aspects and make recommendations.

Follicular regulatory T (Tfr) cells are generated with the responses of germinal center (GC). However, the molecular mechanism on the cell generation is still unknown. Our study found that sclerostin domain-containing protein 1, is essential in the process of Tfr cell generation. With the aid of reporter mice, the results of fate tracking and transcriptome assessment indicated that the Tfh cells expressing SOSTDC1 are identified as a unique T cell population, are developed from SOSTDC1- Tfh cells, and have lost their ability to help B cells in the production of antibodies. It is worth noting that lack of SOSTDC1 results in substantially decreased Tfr cell numbers, and enhanced GC responses. T cells become dysfunctional when they encounter self antigens or are exposed to chronic infection or to the tumour microenvironment.sup.1. The function of T cells is tightly regulated by a combinational co-stimulatory signal, and dominance of negative co-stimulation results in T cell dysfunction.sup.2. However, the molecular mechanisms that underlie this dysfunction remain unclear. Here, using an in vitro T cell tolerance induction system in mice, we characterize genome-wide epigenetic and gene expression features in tolerant T cells, and show that they are distinct from effector and regulatory T cells. Notably, the transcription factor NR4A1 is stably expressed at high levels in tolerant T cells. Overexpression of NR4A1 inhibits effector T cell differentiation, whereas deletion of NR4A1 overcomes T cell tolerance and exaggerates effector function, as well as enhancing immunity against tumour and chronic virus. Mechanistically, NR4A1 is preferentially recruited to binding sites of the transcription factor AP-1, where it represses effector-gene expression by inhibiting AP-1 function. NR4A1 binding also promotes acetylation of histone 3 at lysine 27 (H3K27ac), leading to activation of tolerance-related genes. This study thus identifies NR4A1 as a key general regulator in the induction of T cell dysfunction, and a potential target for tumour immunotherapy.
