In order to study the effect of pre - oxidation treatment on the quality of the nitriding layer of stainless steel parts, quenched and tempered samples were treated with different pre - oxidation and sand blowing treatments before nitriding. The color of the preoxidized film was observed visually. The microstructure, depth, hardness, and brittleness of nitriding layer were measured by metallographic microscope and hardness tester. Results showed that the passivation film on the surface of 1Cr11Ni2W2MoV stainless steel could be destroyed if the steel was preoxidized at 550 ℃ for 5～20 min before nitriding. So the hardness, depth and structure of the nitriding layer could meet the technical requirements for nitriding if treated so. Furthermore, the uniformity and nitriding speed of the layer were better than that of the layer with only sand blowing treatment before nitriding.

Abstract Background The scales serve as an ideal model for studying the regulatory mechanism of bone homeostasis in fish. To explore the effect of salinity acclimation on bone metabolism of juvenile rainbow trout (Oncorhynchus mykiss), three sampling time points during salinity acclimation (7D, 14D and 21D) were selected to detect variations in histological characteristics. In the histological analysis, osteoblast marker enzymes alkaline phosphatase (ALP), osteoclast marker tartrate-resistant acid phosphatase (TRAcP) and calcium salt deposit areas (Von Kossa′s) were detected. Changes in calcium (Ca), phosphorus (P) and the molar mass ratio of calcium to phosphorus (Ca/P) in the scales were also detected by Inductively Coupled Plasma Mass Spectrometry (ICP-MS). In addition, the global MicroRNA (miRNA) expression profiles during salinity acclimation were examined using Illumina sequencing platform because of their important regulatory roles in teleost biological processes. Results Twelve independent miRNA libraries were constructed, a total of 664 known and 92 putative novel miRNAs were identified. A total of 290 differentially expressed (DE) miRNAs were found in clusters with significant trends in the cluster analysis, and five types of clustering patterns were obtained; 22,374 DE predicted target genes of the aforementioned 290 DE miRNAs were obtained, 5957 of which clustered in six types of clustering patterns with a significant trend. To better understand the functions of the DE miRNAs, GO and KEGG analysis was performed on the 5957 target genes, as a result, they were significantly enriched in bone metabolism related signaling pathways such as MAPK signaling pathway, Calcium signaling pathway, Wnt signaling pathway, Mineral absorption and NF-kappa B signaling pathway. Six DE miRNAs were randomly selected and their expression were verified by quantitative real-time PCR (qRT-PCR), the expression trends were consistent with the results of transcriptome sequencing. Conclusions The DE miRNAs and DE target genes identified in this study might play an important role in regulation of bone metabolism during salinity acclimation, relative genes or pathways could serve as key candidates for further studies to elucidate molecular mechanism of teleost bone metabolism, and help performing salinity acclimation and developing marine culture of salmonid species.

Hepatitis C virus (HCV) heterogeneity accounts for the failure of effective vaccine development and the lack of successful anti-viral therapy in some patients. Little is known about the immune response to HCV peptides and the region or race specific genotypes in China. The objective of this study was to characterize HCV antibody immune response to HCV peptides and HCV genotypes in different regions and races of China. A total of 363 serum samples were collected from HCV carriers in 6 regions in China. The immune response to HCV peptides was evaluated by ELISA. HCV genotypes were examined using nested RT-PCR. We found that the anti-HCV antibody neutralization rates were significantly different among the serum samples from different areas or from different races in the same area. For samples from Tibet and Sinkiang, the rates of neutralization by HCV peptides were only 3.2% and 30.8%, respectively. The genotypes of samples from Tibet and Sinkiang were apparently heterogeneic and included type I, II, III and multiple types (I/II/III, I/II, I/III, II/III). One specific sample with multiple-genotype (I/II/III) HCV infection was found to consist of type I, II, III, II/III and an unclassified genotype. These studies indicate that the anti-HCV antibody immune response to HCV peptides varied across regions and among races. The distribution of HCV genotypes among Tibetans in Tibet and Uighurs in Sinkiang was different from that in the inner areas of China. In addition, a “master” genotype, type II, was found to exist in HCV infection with multiple HCV genotypes.

Two methods of coagulation and adsorption were used to pretreat a simulated micro-polluted ground water to study the relationship between ultrafiltration (UF) membrane and pollution levels of different water samples and analyze the effect of particles characteristics on flux decline. The experimental results indicate that powdered activated carbon could reduce the concentration of organics effectively, but the effect of wiping out turbidity is not obvious. Coagulation could reduce the turbidity of water effectively although it has low efficiency on organics removal. The coagulation pretreatment could improve the flux of raw water, but the adsorption has no influence on flux. The correlation fitting results of fouling index of turbidity and dissolved organic matter (DOC) show that fouling index of UF has an obvious correlation with turbidity, but no obvious correlation could be found with DOC concentration. The turbidity-causing pollutants are the major foulant under experimental condition. The colloidal and particles sized between 0.01 µm and 1.2 µm are the main pollutant which lead to flux decline, meanwhile, it is cumulatively on fouling index for turbidity-causing pollutants during UF process.
