30. Gene regulation of iron-deficiency responses is associated with carbon monoxide and heme oxydase 1 in Chlamydomonas reinhardtii.

Abstract

Carbon monoxide (CO) as an endogenous gaseous molecule regulates a variety of biological processes in animals. However, CO regulating nutrient stress responses in green alga is largely unknown. On the other hand, heme oxydase (HO1 as a rate-limiting enzyme of the first step for heme degration and to catalyze heme into biliverdin (BV), which is concomitant with releasing of CO and ferrous ions, probably participates in the process of CO-regulating response to nutrient stress in green alga. In this paper, we described an observation that CO could regulate iron-homeostasis in iron-starving Chlamydomonas reinhardtii. Exogenous CO at 8 µM was able to prevent the iron deficient-inducing chlorosis and improve chlorophyll accumulation. Expression pattern of FOX1, FTR1 and ferredoxin was up-regulated by CO exposure in iron-deficient mediam. treatment with external CO increasing iron accumulation in iron-deficient C. reinhardtii. Moreover, to get insights into the regulatory role of HO1, we constructed a transgenic alga overexpressing HO1 and HO1 knock-out mutants. The results show that there was no significant influence on chlorosis with HO1 overexpression of C. reinhardtii under iron-deficiency and the chlorophyll accumulation, and gene expression associated with iron deficiency of mutant were greatly improved. Otherwise, those results from HO1 knock-out mutants were opposite to HO1 overexpression mutants. Finally, CO exposure induced NO accumulation in cells. However, such an action could be blocked by NO scavenger cPTIO. These results indicate that CO/HO1 may play an important role in improving green algae adaptation to iron deficiency or cross-talking with NO under the iron deficiency.

Abstract

A 6-week feeding trail was conducted to study the effect of density stress on growth performance,plasma cortisol,blood glucose and activities of related enzymes of juvenile Elineophelelus malabaricus.Three hundred juveniles,body weight (15±0.5) g were randomly distributed in 4 experimental groups:G10 (10 ind/tank,1.1 kg/m 3),G20 (20 ind/tank,2.1 kg/m 3),G30 (30 ind/tank,3.2 kg/m 3),and G40 (40 ind/tank,4.2 kg/m 3).The results showed:(1) weight gain (WG) and specific growth rate (SGR) of G10 were the lowest among the experimental groups,and WG and SGR of G20 were the highest.Feed conversion ratio (FCR) increased with the increase of density.(2) No significant difference was observed in plasma cortisol levels between G30 and G40 (P 0.05),plasma cortisol levels of G10 and G20 were significantly lower than those of G 30 and G 40 (P 0.05),and plasma cortisol level of G10 was the lowest.Similar trend was recorded in blood glucose levels.(3)Glutamic pyruvic transaminase (GPT) activities of G30 and G40 were significantly higher than those of G10 and G20 ;glutamic oxaloacetic transaminase (GOT) activities and lactic dehydrogenase (LDH) activities increased with the increase of the stocking density,total choline esterase (TChE) activities decreased with the increase of the stocking density.Hence,too high or too low stocking density would affect the growth performance and metabolism of juvenile Epinephelus malabaricus,and 20 ind/tank (2.1 kg/m3) was considered to be a suitable stocking density.

Abstract

The rate of hyperglycaemia in people around the world is increasing at an alarming rate at present, and innovative methods of alleviating hyperglycaemia are needed. The effects of Jerusalem artichoke inulin on hyperglycaemia, liver-related genes and the intestinal microbiota in mice fed a high-fat diet (HFD) and treated with streptozotocin (STZ) to induce hyperglycaemia were investigated. Inulin-treated hyperglycaemic mice had decreased average daily food consumption, body weight, average daily water consumption and relative liver weight and blood concentrations of TAG, total cholesterol, HDL-cholesterol and fasting blood glucose. Liver-related gene expressions in hyperglycaemic (HFD-fed and STZ-treated) compared with control mice showed eighty-four differentially expressed genes (forty-nine up-regulated and thirty-five down-regulated). In contrast, hyperglycaemic mice treated with inulin had twenty-two differentially expressed genes compared with control ones. Using Illumina high-throughput sequencing technology, the rarefaction and the rank abundance curves as well as the α diversity indices showed the treatment-induced differences in bacterial diversity in intestine. The linear discriminant analysis of effect size showed that the inulin treatment improved intestinal microbiota; in particular, it significantly increased the number of Bacteroides in the intestine of mice. Abstract

This study was aimed at evaluating the physiological and metabolic responses of juvenile hybrid grouper ♀ Epinephelus fuscoguttatus × ♂ Epinephelus lanceolatus to stocking density. Hybrid grouper juveniles (mean ± SE = 25.43 ± 2.36 g live mass) were stocked for 22 weeks in a recirculating aquaculture system (RAS) under four different densities: low stocking density (LD; 1.03 kg m−3), medium stocking density (MD; 2.06 kg m−3), high stocking density (HD; 3.09 kg m−3) and extra‐high stocking density (EHD; 4.11 kg m−3). Biometric variables were recorded and plasma, liver, intestine and stomach samples were taken for biochemical analysis at the end of the experimental period. Final stocking densities were 6.27, 16.04, 23.77 and 28.32 kg m−3, respectively, with significant differences in growth performance. Our results showed that the best growth rates and feed utilisation occurred in the MD group. Higher plasma cortisol and glucose levels and lower triglyceride levels reflected the stress responses in the EHD group. Moreover, the activity of aspartate and alanine transaminases was elevated in the HD and EHD groups due to enhanced gluconeogenesis. The activity of the digestive enzyme pepsin significantly increased in the MD group. We found that 2.06–3.09 kg m−3 is the most suitable starting density for culturing juvenile hybrid grouper in recirculating aquaculture systems. [ABSTRACT FROM AUTHOR]

Abstract

The effects of exogenous calcium ion (Ca2+) on malondialdehyde (MDA), relative conductivity (RC), net photosynthetic rate (Pn) and chlorophyll content were investigated in leaves of salt-stressed Jerusalem artichoke (Helianthus tuberosus L.). The plants were subjected to the following five treatments: (1) control (half-strength Hoagland solution alone), (2) lOmmol L-1 CaCl2, (3) 30% seawater, (4) 30% seawater +10mmol L-1 CaCl2, and (5) 30% seawater+5 mmol L-1 EGTA (a chelant of calcium ion). The results showed that seawater stress resulted in the accumulation of active oxygen species damage to photosynthetic apparatus of Jerusalem artichoke leaves. Compared with the seawater alone, enrichment of challenge solutions with relatively higher concentration of Ca2+ significantly protected the photosynthetic system from oxidative damage by inhibiting lipid peroxidation, retarding electrolyte leakage from cell and increasing chlorophyll content. Therefore, high F/Fm [maximal photochemical efficiency of photosystem II (PSII) while all PSII reaction centers are open], Fm/F0 (another expression for the maximal photochemical efficiency of PSII), ΦPSII (actual quantum yield of PSII under actinic irradiation), qP (photochemical quenching coefficient), and Pn were maintained, and lower qNP (non-photochemical quenching coefficient) of the leaves was kept under seawater stress. EGTA had the opposite effect. Thus Ca ion may help protect the photosynthetic system of Jerusalem artichoke leaves from oxidative damage induced by the seawater stress.
