In order to study and analyze the causes of perforation failure in 3003 aluminum tubes， used in automobile air conditioning， the reasons for surface perforation defects in these tubes were investigated.This investigation involved macro-morphology analysis，chemical composition analysis， scanning electron microscope micro-morphology analysis and energy spectrum analysis.Results showed that nonmetallic inclusions on the surfaces of the 3003 aluminum tubes damaged the integrity and continuity of these surfaces.This issue prevented the formation of a dense oxide film at the inclusion sites.Under the combined influence of the strongly corrosive elements chlorine (Cl) and sulfur (S)， pitting and perforation failures occurred in the 3003 aluminum tubes.It was suggested that the addition of refining agents should be carefully controlled during the melting process of the 3003 alloy.Additionally， improvements in the filtering of inclusions in the aluminum liquid， as well as enhancements in the surface cleanliness and storage environment of the aluminum tubes， were recommended.These research findings could provide a certain reference for the manufacturing of 3003 aluminum tubes.

Immunodeficiency, centromeric instability and facial anomalies type I (ICF1) syndrome is a rare genetic disease caused by mutations in DNA methyltransferase (DNMT) 3B, a de novo DNA methyltransferase. However, the molecular basis of how DNMT3B deficiency leads to ICF1 pathogenesis is unclear. Induced pluripotent stem cell (iPSC) technology facilitates the study of early human developmental diseases via facile in vitro paradigms. Here, we generate iPSCs from ICF Type 1 syndrome patient fibroblasts followed by directed differentiation of ICF1-iPSCs to mesenchymal stem cells (MSCs). By performing genome-scale bisulfite sequencing, we find that DNMT3B-deficient iPSCs exhibit global loss of non-CG methylation and select CG hypomethylation at gene promoters and enhancers. Further unbiased scanning of ICF1-iPSC methylomes also identifies large megabase regions of CG hypomethylation typically localized in centromeric and subtelomeric regions. RNA sequencing of ICF1 and control iPSCs reveals abnormal gene expression in ICF1-iPSCs relevant to ICF syndrome phenotypes, some directly associated with promoter or enhancer hypomethylation. Upon differentiation of ICF1 iPSCs to MSCs, we find virtually all CG hypomethylated regions remained hypomethylated when compared with either wild-type iPSC-derived MSCs or primary bone-marrow MSCs. Collectively, our results show specific methylome and transcriptome defects in both ICF1-iPSCs and differentiated somatic cell lineages, providing a valuable stem cell system for further in vitro study of the molecular pathogenesis of ICF1 syndrome. Whether human induced pluripotent stem cells (hiPSCs) are epigenetically identical to human embryonic stem cells (hESCs) has been debated in the stem cell field. In this study, we analyzed DNA methylation patterns in a large number of hiPSCs (n = 114) and hESCs (n = 155), and identified a panel of 82 CpG methylation sites that can distinguish hiPSCs from hESCs with high accuracy. We show that 12 out of the 82 CpG sites were subject to hypermethylation in part by DNMT3B. Notably, DNMT3B contributes directly to aberrant hypermethylation and silencing of the signature gene, TCERG1L. Overall, we conclude that DNMT3B is involved in a wave of de novo methylation during reprogramming, a portion of which contributes to the unique hiPSC methylation signature. These 82 CpG methylation sites may be useful as biomarkers to distinguish between hiPSCs and hESCs.

Mammalian pre-implantation development is a complex process involving dramatic changes in the transcriptional architecture. We report here a comprehensive analysis of transcriptome dynamics from oocyte to morula in both human and mouse embryos, using single-cell RNA sequencing. Based on single-nucleotide variants in human blastomere messenger RNAs and paternal-specific single-nucleotide polymorphisms, we identify novel stage-specific monoallelic expression patterns for a significant portion of polymorphic gene transcripts (25 to 53%). By weighted gene co-expression network analysis, we find that each developmental stage can be delineated concisely by a small number of functional modules of co-expressed genes. This result indicates a sequential order of transcriptional changes in pathways of cell cycle, gene regulation, translation and metabolism, acting in a step-wise fashion from cleavage to morula. Cross-species comparisons with mouse pre-implantation embryos reveal that the majority of human stage-specific modules (7 out of 9) are notably preserved, but developmental specificity and timing differ between human and mouse. Furthermore, we identify conserved key members (or hub genes) of the human and mouse networks. These genes represent novel candidates that are likely to be key in driving mammalian pre-implantation development. Together, the results provide a valuable resource to dissect gene regulatory mechanisms underlying progressive development of early mammalian embryos.

Processed by sulfuric acid anodizing, several unexpected strip defects exhibited on the anodizing film of AA5252 aluminium alloy sheets. Their formation mechanism was studied in detail. The normal zone and strip zone were compared, with respect to surface brightness, porosity of anodizing film, and microstructures of the corresponding aluminium substrate tested by spectroscopic colorimeter and SEM, and SEM-EBSD. Results showed that the brightness of strip zone was lower than that of normal zone. Additionally, compared with normal zone, the porosity of anodizing film was higher, and cubic grains fewer, which located in the corresponding surface layer of aluminium substrate in strip zone. In order to further clarify the effect of grain orientation on the brightness of anodizing film, single crystal aluminium sheet with (100), (110) or (111) orientation was anodized. Results showed that the anodized (100) oriented specimen, i.e., cubic grain was brighter, whose anodizing film had lower porosity, compared with (110) and (111) oriented ones. Therefore, the formation mechanism of strip defects was proposed as uneven distribution of cubic grains. The zones with fewer cubic orientated grains were darker, and were visible as strip defects after anodizing.

Infection with the necrotrophic fungus Diplodia sapinea (Fr.) Fuckel is among the economically and ecologically most devastating diseases of conifers in the northern hemisphere and is accelerated by global climate change. This study aims to characterize the changes mediated by D. sapinea infection on its pine host (Pinus sylvestris L.) that lead to the death of its needles. For this purpose, we performed an indoor infection experiment and inoculated shoot tips of pine seedlings with virulent D. sapinea. The consequences for foliar traits, including the phytohormone profile, were characterized at both the metabolite and transcriptome level. Our results showed that D. sapinea infection strongly affected foliar levels of most phytohormones and impaired a multitude of other metabolic and structural foliar traits, such as reactive oxygen species scavenging. Transcriptome analysis revealed that these changes are partially mediated via modified gene expression by fungal exposure. Diplodia sapinea appears to overcome the defense reactions of its pine host by reprogramming gene expression and post-transcriptional controls that determine essential foliar metabolic traits such as the phytohormone profile, cell wall composition and antioxidative system. In view of the current situation of gully in China, new type of environmentally-friendly gully with anti-mosquito and odor-reducing function has been developed. The control performance of new gully on the initial rainwater pollutants under the condition of different filter material ratio and filter bag thickness. The results show that the new environmentally-friendly gully has better removal effect of various pollutants in the initial rainwater under the condition of 1:3 filter material and 3cm filter bag. The removal rate of SS is over 80%, total nitrogen the removal rate is above 75%, and the removal rate of COD and total phosphorus is over 70%.
