In order to enhance China’s competitiveness in international ice and snow athletic competitions, this paper proposes a study on the cultivation of ice and snow athletic reserve talents supported by structural equation modeling. The initial selection of the driving factors for the cultivation of ice and snow athletic reserve talents is carried out by means of reviewing the information, and the driving factors for the cultivation model of ice and snow athletic reserve talents in a province are finally determined after expert selection, common factor extraction, and principal component analysis. The fitting index and covariance matrix were solved using the partial least squares method, and then the construction of the structural equation model was completed. Research data were obtained to empirically analyze the cultivation of ice and snow athletic talent reserves using the structural equation model. The results showed that the contribution of the academic training factor to the drivers of ice and snow competitive reserve talent cultivation was 0.891 for the path coefficient of Q6, 0.826 for the path coefficient of Q7, 0.848 for the path coefficient of Q9, and 0.914 for the path coefficient of Q10. This study provides a reference for solving real problems, such as the shortage of competitive ice and snow talents in China, which is of great significance for the strategic development of China’s sports industry.

Drug resistance contributes to treatment failure and relapse in acute lymphoblastic leukemia (ALL). G1P3 (also known as IFI6, interferon, alpha-inducible protein 6) has been regarded as an antiapoptotic protein in myeloma cells and contributes to chemoresistance in breast cancer. However, the role of G1P3 in the proliferation and chemosensitivity of ALL is largely unknown. Data from colony formation and bromo-deoxyuridine (BrdU) incorporation assays showed that siRNA-mediated downregulation of G1P3 repressed cell proliferation of glucocorticoids-resistant human leukemic cells (CEM-C1), while overexpression of G1P3 promoted the cell proliferation. Cell apoptosis of CEM-C1 was suppressed by G1P3 overexpression accompanied by a decrease in cleaved caspase-3 and caspase-9. Knockdown of G1P3 increased protein expression of cleaved caspase-3 and caspase-9 to promote the cell apoptosis of CEM-C1. Moreover, silencing of G1P3 reduced cell viability and promoted cell apoptosis of CEM-C1 exposed to dexamethasone. The proapoptotic protein B-cell lymphoma 2 interacting mediator of cell death (Bim) was enhanced by the interference of G1P3 in CEM-C1. Silencing of Bim attenuated G1P3 interference-induced decrease in cell viability and increase in cell apoptosis in CEM-C1 exposed to dexamethasone. Conclusively, knockdown of G1P3 inhibited cell proliferation of ALL and sensitized glucocorticoid-resistant ALL cells to dexamethasone through upregulation of Bim-mediated cell apoptosis.

In situ depot gel is a type of polymeric long-acting injectable (pLAI) drug delivery system; compared to microsphere technology, its preparation process is simpler and more conducive to industrialization. To ensure the chemical stability of peptide ACTY116, we avoided the use of harsh conditions such as high temperatures, high shear mixing, or homogenization; maintaining a water-free and oxygen-free environment was also critical to prevent hydrolysis and oxidation. Molecular dynamics (MDs) simulations were employed to assess the stability mechanism between ACTY116 and the pLAI system. The initial structure of ACTY116 with an alpha helix conformation was constructed using SYBYL-X, and the copolymer PLGA was generated by AMBER 16; results showed that PLGA-based in situ depot gel improved conformational stability of ACTY116 through hydrogen bonds formed between peptide ACTY116 and the components of the pLAI formulation, while PLGA (Poly(DL-lactide-co-glycolide)) also created steric hindrance and shielding effects to prevent conformational changes. As a result, the chemical and conformational stability and in vivo long-acting characteristics of ACTY116 ensure its enhanced efficacy. In summary, we successfully achieved our objective of developing a highly stable peptide-loaded long-acting injectable (LAI) in situ depot gel formulation that is stable for at least 3 months under harsh conditions (40 °C, above body temperature), elucidating the underlying stabilisation mechanism, and the high stability of the ACTY116 pLAI formulation creates favourable conditions for its in vivo pharmacological activity lasting for weeks or even months. Hypokalemia and hyponatremia are common but easily ignored adverse events in treatment with voriconazole (VCZ) that can lead to serious consequences. We intend to investigate the incidence of VCZ‐induced hypokalemia and hyponatremia and their risk factors based on real‐world data. A prospective study was conducted. A total of 272 patients with 414 VCZ plasma trough concentrations (C0) and VCZ N‐oxide concentrations (CN) were included. The incidence of hypokalemia was 18.0% (48/266). A total of 81.2% (39/48) of patients developed hypokalemia within 14 days, whereas 56.2% (27/48) of patients developed hypokalemia within 1 week. The proportion of female patients in the hypokalemia group was higher than that in the nonhypokalemia group, as was the proportion of patients receiving intravenous VCZ. In the multivariate analysis, the independent risk factors for hypokalemia were sex, combined use of antibiotics, and VCZ CN/C0. The incidence of hyponatremia was 7.9% (21/266). The proportion of patients over 47 years of age in the hyponatremia group was 71.4% (15/21). The number of days of VCZ use in the hyponatremia group was greater than that in the nonhyponatremia group. A total of 47.6% (10/21) of patients in the hyponatremia group had supratherapeutic VCZ C0 (>5.0 µg/mL). In conclusion, hypokalemia is more likely to occur in females, in patients receiving intravenous VCZ, and in patients with the combined use of antibiotics. Hyponatremia is more likely to occur in patients older than 47 years who have been using VCZ for a long time and have higher VCZ C0 values. [ABSTRACT FROM AUTHOR]

Objective To investigate the changes in endogenous metabolites in a rat model of depression and the modulatory effect of Baihe Zhimu decoction (BZD) on the abnormal metabolites through a metabolomic approach. Methods Forty rats were randomly divided into normal control group, depression model group, fluoxetine treatment group and BZD treatment group. In all but the control group, rat models of depression were established by keeping the rats in separate cages combined with exposure to chronic unpredictable mild stress. A metabolomic approach was used to identify the endogenous metabolites and metabolic pathways that were related to the antidepressive effects of BZD. Results BZD could significantly increase the body weight, sucrose preference and behavioral scores in the rat models of depression (P < 0.05). BZD treatment resulted in the recovery of normal levels for 23 metabolites that showed abnormal serum levels in the depressive rats, and these metabolites were associated with fatty acid amide metabolism, phenylalanine metabolism, glycine metabolism and lipid metabolism. By comparison, fluoxetine treatment only restored the normal levels for 17 abnormal metabolites, which were related with glycine metabolism and lipid metabolism. Conclusion BZD can modulate more metabolic pathways than fluoxetine for treatment of depression.

Molybdenum disulfide as a supercapacitor material has attracted great attention owing to its low price and high theoretical capacity. However, the low conductivity and relatively few active sites restrict its practical application. In this work, a 3D bacterial cellulose intercalated MoS2@rGO nanocomposite is fabricated via the freeze-drying method and an in situ carbonization process. The BC can prevent the stacking of rGO and expose more electrochemical active sites. Benefitting from the synergistic effect of BC and rGO, the prepared MoS2@rGO@CF-2 displays a high specific capacitance of 345 F g−1 at the current density of 0.5 A g−1. [ABSTRACT FROM AUTHOR]
