Abstract

Computer-assisted systems are becoming broadly used in medicine. In endoscopy, most research focuses on the automatic detection of polyps or other pathologies, but localization and navigation of the endoscope are completely performed manually by physicians. To broaden this research and bring spatial Artificial Intelligence to endoscopies, data from complete procedures is needed. This paper introduces the Endomapper dataset, the first collection of complete endoscopy sequences acquired during regular medical practice, making secondary use of medical data. Its main purpose is to facilitate the development and evaluation of Visual Simultaneous Localization and Mapping (VSLAM) methods in real endoscopy data. The dataset contains more than 24 hours of video. It is the first endoscopic dataset that includes endoscope calibration as well as the original calibration videos. Meta-data and annotations associated with the dataset vary from the anatomical landmarks, procedure labeling, segmentations, reconstructions, simulated sequences with ground truth and same patient procedures. Abstract

Landmark recognition and matching is a critical step in many Image Navigation and Registration (INR) models for geostationary satellite services, as well as to maintain the geometric quality assessment (GQA) in the instrument data processing chain of Earth observation satellites. Matching the landmark accurately is of paramount relevance, and the process can be strongly impacted by the cloud contamination of a given landmark. This paper introduces a complete pattern recognition methodology able to detect the presence of clouds over landmarks using Meteosat Second Generation (MSG) data. The methodology is based on the ensemble combination of dedicated support vector machines (SVMs) dependent on the particular landmark and illumination conditions. This divide-and-conquer strategy is motivated by the data complexity and follows a physically-based strategy that considers variability both in seasonality and illumination conditions along the day to split observations. In addition, it allows training the classification scheme with millions of samples at an affordable computational costs. The image archive was composed of 200 landmark test sites with near 7 million multispectral images that correspond to MSG acquisitions during 2010. Results are analyzed in terms of cloud detection accuracy and computational cost. We provide illustrative source code and a portion of the huge training data to the community.

Abstract

Dealing with land cover classification of the new image sources has also turned to be a complex problem requiring large amount of memory and processing time. In order to cope with these problems, statistical learning has greatly helped in the last years to develop statistical retrieval and classification models that can ingest large amounts of Earth observation data. Kernel methods constitute a family of powerful machine learning algorithms, which have found wide use in remote sensing and geosciences. However, kernel methods are still not widely adopted because of the high computational cost when dealing with large scale problems, such as the inversion of radiative transfer models or the classification of high spatial-spectral-temporal resolution data. This paper introduces an efficient kernel method for fast statistical retrieval of bio-geo-physical parameters and image classification problems. The method allows to approximate a kernel matrix with a set of projections on random bases sampled from the Fourier domain. The method is simple, computationally very efficient in both memory and processing costs, and easily parallelizable. We show that kernel regression and classification is now possible for datasets with millions of examples and high dimensionality. Examples on atmospheric parameter retrieval from hyperspectral infrared sounders like IASI/Metop; large scale emulation and inversion of the familiar PROSAIL radiative transfer model on Sentinel-2 data; and the identification of clouds over landmarks in time series of MSG/Seviri images show the efficiency and effectiveness of the proposed technique.

Abstract

The initial excitement generated more than two decades ago by the discovery of drugs targeting fatty acid synthase (FASN)‐catalyzed de novo lipogenesis for cancer therapy was short‐lived. However, the advent of the first clinical‐grade FASN inhibitor (TVB‐2640; denifanstat), which is currently being studied in various phase II trials, and the exciting advances in understanding the FASN signalome are fueling a renewed interest in FASN‐targeted strategies for the treatment and prevention of cancer. Here, we provide a detailed overview of how FASN can drive phenotypic plasticity and cell fate decisions, mitochondrial regulation of cell death, immune escape and organ‐specific metastatic potential. We then present a variety of FASN‐targeted therapeutic approaches that address the major challenges facing FASN therapy. These include limitations of current FASN inhibitors and the lack of precision tools to maximize the therapeutic potential of FASN inhibitors in the clinic. Rethinking the role of FASN as a signal transducer in cancer pathogenesis may provide molecularly driven strategies to optimize FASN as a long‐awaited target for cancer therapeutics. Abstract

Aquest treball s’ha portat a terme a partir d’un estudi bibliogràfic, que tracta sobre els materials híbrids compostos de polímers conductors bioabsorbibles, biocompatibles i biodegradables. També s’estudia com a evolucionat les aplicacions biomèdiques durant els darrers anys. Els polímers són una classe de biomaterials que es poden dissenyar per satisfer requisits específics d’ús final. Es poden seleccionar segons les característiques clau, com ara resistència mecànica, degradabilitat, permeabilitat, solubilitat i transparència, però els polímers disponibles actualment han de millorar-se alterant les seves propietats superficials i massives. En el primer capítol del projecte, es fa una introducció sobre els polímers conductors: que són, quines aplicacions tenen, les seves propietats i la seva evolució històrica. També s’ha estudiat els polímers conductors més habituals; poliacetilè, politiofè, polianilina , el polipirrol i poli (3,4-etilendioxitiofè) (PEDOT). Seguidament, s’explica sobre els polímers no conductors amb biopropietats (biodegradable, biocompatible i bioabsorbible), per així fer un anàlisis d’aquests termes i després veure com afecta aquestes biopropietats en polímers conductors. Per últim, s’estudia les aplicacions biomèdiques, sobretot dels sensors implantables durant els darrers anys. This project has been carried out through a bibliographic study, which deals with hybrid materials composed of bioabsorbable, biocompatible and biodegradable conductive polymers. It also studies how the biomedical applications have evolved over the years. Polymers are a class of biomaterials that can be designed to meet specific end-use requirements. They can be selected according to key characteristics, such as mechanical strength, degradability, permeability, solubility and transparency, but currently available polymers have to alter their surface and bulk properties. In the first chapter of the project, an introduction to conductive polymers is given: what they are, what applications they have, their properties and their historical evolution. The most common conductive polymers are also studied: polyacetylene, polyoxymethylene, polyaniline, polypyrrole and poly(3,4-ethylenedioxythiophene) (PEDOT). Next, non-conductive polymers with bio properties (biodegradable, biocompatible and bioabsorbable) are explained, followed by an analysis of these terms and learning how they affect these bio properties in conductive polymers. Abstract

Simple Summary: An increase of sialic acid in the cancer cells' surface is a hallmark of tumours, including pancreatic cancer, and it has been related to tumour malignancy and immune suppression. In this work, we have assessed for the first time the potential of the sialyltransferase inhibitor, Ac53FaxNeu5Ac, to reduce tumor sialylation in pancreatic cancer and to revert its malignant phenotype. We have shown that Ac53FaxNeu5Ac treatment on human pancreatic cancer cells decreased their sialic acid, reduced their E-selectin adhesion—the prior step to tumour extravasation—and their migration and invasion capabilities. In addition, subcutaneous pancreatic tumours generated on immunocompetent mice that were treated with Ac53FaxNeu5Ac showed a reduced growth and increased tumour infiltrating lymphocytes. These results show that the targeting of tumour sialoglycans in pancreatic cancer reverts its malignant phenotype and favours anti-tumour immune surveillance, which opens the way to use sialyltransferase inhibitors as a novel therapeutic strategy against this dismal disease. Hypersialylation is a feature of pancreatic ductal adenocarcinoma (PDA) and it has been related to tumor malignancy and immune suppression. In this work, we have evaluated the potential of the sialyltransferase inhibitor, Ac53FaxNeu5Ac, to decrease tumor sialoglycans in PDA and to revert its malignant phenotype. Sialoglycans on PDA cells were evaluated by flow cytometry, and the functional impact of Ac53FaxNeu5Ac was assessed using E-selectin adhesion, migration, and invasion assays. PDA tumors were generated in syngeneic mice from KC cells and treated with Ac53FaxNeu5Ac to evaluate tumor growth, mice survival, and its impact on blocking sialic acid (SA) and on the tumor immune component. Ac53FaxNeu5Ac treatment on human PDA cells decreased α2,3-SA and sialyl-Lewisx, which resulted in a reduction in their E-selectin adhesion, and in their migratory and invasive capabilities. Subcutaneous murine tumors treated with Ac53FaxNeu5Ac reduced their volume, their SA expression, and modified their immune component, with an increase in CD8+ T-lymphocytes and NK cells. In conclusion, Ac53FaxNeu5Ac treatment weakened PDA cells' malignant phenotype, thereby reducing tumor growth while favoring anti-tumor immune surveillance. Altogether, these results show the positive impact of reducing SA expression by inhibiting cell sialyltransferases and open the way to use sialyltransferase inhibitors to target this dismal disease.
