However, realistic numerical predictions are achieved only with a careful consideration of all the model input.In particular, a key ingredient is the choice of consistent constitutive models for the elasto-plastic behavior, identified using sound procedures from reliable and repeatable data sets. At present, the information provided in most benchmarks consists of the hardening law coefficients, along with limited experimental data such as, r-values, yield stresses and ultimate strengths extracted from uniaxial tension in certain loading directions, and possibly the coefficients of certain yield functions available in the material libraries of most commercial software. The identification of constitutive models is usually not included in the work requested to participants. Generally, there is no information provided concerning the experimental scatter because the “raw” data are not made available. While the behavior in tensile tests is essential, other loading paths (e.g. biaxial tension, uniaxial compression) may also be important for certain forming processes. Moreover, other types of data describing the initial material anisotropy and that induced by the deformation process, i.e. initial and post-test texture data, are rarely provided. For example, in a simple case of cup drawing, if the final cup test results are “blind data” describing the final geometry, the participants do not have the opportunity to analyze the quality/performance of their simulation results. In addition, if the tests are done in a single laboratory, the experimental scattering is limited to material variability but not to the characterization method nor to the raw data analysis procedure. This benchmark is a unique opportunity at providing an exhaustive analysis of the characterization and modeling of the elasto-plastic behavior of a sheet metal and its influence on the final virtual product. Aim: To affect the influence that osteoarthrosis has on the balance and gait of elderly patients. Method: This is an observational study in elderly people of both genders with diagnosed knee osteoarthritis. Initially, a lecture was held on the objectives of the study and after signing the Informed Consent Term, the collection of sociodemographic data was carried out; pain assessment using the visual analogue scale; balance assessment using the Berg Balance Scale, mobility and gait assessment using the Time Up and Go and Tinetti Test. Results: Eleven elderly people with osteoarthritis, all female, with a mean age of 75.6 years, were selected for the study. There was a correlation between increasing age and worsening mobility and balance in the elderly. Conclusion: It was concluded in this research, that aging is a physiological and progressive process, in which several body systems are compromised, mainly the musculoskeletal system. Aspects such as balance and morbidity have a strong relationship with advancing age and may have physical and psychological impairment. Objetivo: analizar la influencia que tiene la osteoartrosis en el equilibrio y la marcha de los pacientes ancianos. Metodología: Se trata de un estudio observacional en personas mayores de ambos sexos con osteoartritis de rodilla diagnosticada. Inicialmente se realizó una charla sobre los objetivos del estudio y luego de la firma del Término de Consentimiento Informado se llevó a cabo la recolección de datos sociodemográficos; evaluación del dolor mediante la escala analógica visual; evaluación del equilibrio mediante la escala de equilibrio de Berg, evaluación de la movilidad y la marcha mediante el test Time Up and Go y Tinetti. Resultados: Se seleccionaron para el estudio 11 ancianos con artrosis, todas mujeres, con una edad media de 75,6 años. Hubo una correlación entre el aumento de la edad y el empeoramiento de la movilidad y el equilibrio en los ancianos. Consideraciones finales: En esta investigación se concluyó que el envejecimiento es un proceso fisiológico y progresivo, en el que se ven comprometidos varios sistemas corporales, principalmente el sistema musculoesquelético. Aspectos como el equilibrio y la morbilidad tienen una fuerte relación con la edad avanzada y pueden tener un deterioro físico y psicológico. Objetivo: analisar a influência que a osteoartrose tem sobre o equilíbrio e a marcha de pacientes idosos. Metodologia: Trata-se de um estudo do tipo observacional em idosos de ambos os sexos com osteoartrite de joelho diagnosticada. Inicialmente foi realizado uma palestra sobre os objetivos do estudo e após assinatura do Termo de Consentimento Livre e Esclarecido foi realizado a coleta de dados sociodemográfico; avaliação da dor pela Escala visual analógia; avaliação do equilíbrio pela Escala de Equilíbrio de Berg, avaliação da mobilidade e marcha através do Time Up and Go e Teste de Tinetti. Resultados: Foram selecionados para o estudo 11 idosos com osteoartrose, todos do sexo feminino, com média de idade de 75,6 anos. Houve correlação entre o aumento da idade com a piora na mobilidade e equilíbrio dos idosos. Considerações finais: Concluiu-se nesta pesquisa, que o envelhecimento é um processo fisiológico e progressivo, no qual vários sistemas do organismo são comprometidos, principalmente o sistema musculoesquelético. Os aspectos como o equilíbrio e morbidade tem uma forte relação com o avanço da idade, podendo ter comprometimento físico e psicológico.

Aim: correlate the level of physical disability with pain, balance and gait in elderly with chronic low back pain. Method: observational study conducted at the Catholic University of Pernambuco. Initially, a lecture was held on the objectives of the study and after signing the Informed Consent Form, sociodemographic data were collected; assessment of the level of physical disability due to pain through Roland-Morris and Oswestry; Pain intensity by visual analog scale (VAS); Balance by Berg Balance Scale and Risk of Tinetti Fall. Results: the sample consisted of 20 elderly, with female predominance (80%), average age of 70.95 ± 6.08 years. There was a significant correlation between physical disability pain level, balance level and risk of falling. Conclusion: It was possible to show in the elderly with low back pain, an association between the questionnaires that assess physical disability. It can be concluded that there was a significant correlation between the level of physical disability with pain intensity and gait. In addition, the relevance of using assessment instruments for the development of therapeutic strategies aimed at autonomy and independence of the elderly person's health is also emphasized. Método: estudio observacional en hombres y mujeres ancianos con diagnóstico de lumbalgia crónica. Inicialmente se dio una charla sobre los objetivos del estudio y luego de firmar el Formulario de Consentimiento Libre e Informado se recolectaron datos sociodemográficos; evaluación del nivel de discapacidad física resultante del dolor utilizando Roland-Morris y Oswestry; Intensidad del dolor por escala analógica visual (EVA); Equilibrio de la balanza de Berg y Riesgo de caída de Tinetti. Resultados: la muestra estuvo conformada por 20 ancianos, predominantemente mujeres (80%), con una edad promedio de 70,95 ± 6,08 años. Hubo una correlación significativa entre el nivel de discapacidad física con el dolor, el nivel de equilibrio y la marcha. Conclusión: se pudo evidenciar en ancianos con lumbalgia una asociación entre los cuestionarios que evalúan la discapacidad física. También hubo una correlación significativa entre el nivel de discapacidad física con la intensidad del dolor y la marcha. Además, destaca la relevancia del uso de herramientas de evaluación para el desarrollo de estrategias terapéuticas orientadas a la autonomía e independencia de la salud de las personas mayores. Método: estudo do tipo observacional em idosos de ambos os sexos com lombalgia crônica diagnosticada. Inicialmente foi realizado uma palestra sobre os objetivos do estudo e após assinatura do Termo de Consentimento Livre e Esclarecido foi realizado a coleta de dados sociodemográfico; avaliação do nível de incapacidade física decorrente da dor através do Roland-Morris e Oswestry; Intensidade da dor pela escala visual analógica (EVA); Equilíbrio pela Escala de equilíbrio de Berg e Risco de queda pela Tinetti. Resultados: a amostra foi constituída por 20 idosos, com predominância feminina (80%), média de idade de 70,95 ± 6,08 anos. Houve correlação significativa entre nível de incapacidade física com dor, nível de equilíbrio e marcha. Conclusão: Foi possível evidenciar nos idosos com lombalgia, associação entre os questionários que avaliam incapacidade física. Também houve correlação significativa entre o nível de incapacidade física com intensidade da dor e com a marcha. Além disso, enfatiza-se, a relevância do uso de instrumentos de avaliação para o desenvolvimento de estratégias terapêuticas voltadas para autonomia e independência da saúde da pessoa idosa.

The influence of physical exercise on brain antioxidant defense mechanisms has been studied. Nevertheless, the effect of training volume on the brain`s redox balance remains unclear. In this meta-analysis, we compared the effect of training volume on antioxidant enzymatic resource and lipid peroxidation on various brain regions. The activities of the enzymes glutathione peroxidase (GPx), superoxide dismutase (SOD), catalase (CAT) and the levels of thiobarbituric acid reactive substances (TBARS) were also evaluated. The effects of training periods (weeks) and exercise duration were compared. Meta-analysis revealed that protocols over 8 weeks were associated with an increase in SOD (p = 0.0008) and CAT activities (p = 0.0001). Exercise durations for 30 and 60 min were associated with higher CAT activity (p = 0.04). Joint analysis revealed that moderate physical exercise over 4 and 8 weeks promoted a healthy enzymatic balance. However, high volumes of exercise over 8 weeks were associated with the increased antioxidant enzymatic activity, indicating higher reactive oxygen species (ROS) levels. The data also indicated that there is still limited research and inaccurate information, on the safety conditions of training periods that simulate tests of ultra resistance in humans.

Isolating high quality RNA is a limiting factor in molecular analysis, since it is the base for transcriptional studies. The RNA extraction method can directly affect the RNA quality and quantity, as well as, its overall cost. The industrial importance of the yeast genus Candida in several sectors comes from their capacity to produce Lipases. These enzymes are one of the main metabolites produced by some Candida species, and it has been shown that Candida yeast can biodegrade petroleum hydrocarbons and diesel oil from biosurfactants that they can produce, a feature that turns these organisms into potential combatants for bioremediation techniques. Thus, this study aimed to determine an efficient method for isolating high quality RNA from Candida viswanathii biomass. To achieve this aim, three different RNA extraction methods, TRIzol, Hot Acid Phenol, and CTAB (Cetyltrimethylammonium Bromide), were tested. The three tested methods allowed the isolation of high-quality RNA from C. viswanathii biomass and yielded suitable RNA quantity for carrying out RT-qPCR studies. In addition, all methods displayed high sensitivity for the expression analysis of the CvGPH1 gene through RT-qPCR, with TRIzol and CTAB showing the best results and the CTAB method displaying the best cost-benefit ratio (US$0.35/sample).

The hookah is a new trend of young people between the ages of 20 and 30, always present at parties or on conversation wheels, using glycerinated tobacco for their smoke, also known as essence. Knowing the harms of tobacco, used as the main component of hookah, such as cancer and breathing problems, it can also contain other possible causes for diseases such as pathogenic microorganisms. Thus, despite the widespread popularization of this practice, there is still little research on it. Therefore, the objective of this work was to perform microbiological analysis of the components of the hookah in order to find bacteria of pathological origin or not. For this, the in vitro inoculation methodology was used in petri dish containing solidified agar. In case of bacterial growth, the microorganisms were isolated and subjected to identification by biochemical tests. E. coli, E. aerogenes, K. pneumoniae were found in the hookah pieces. Therefore good hygiene and other countermeasures are required for smokers to prevent infections. La pipa de agua es una nueva tendencia para jóvenes de entre 20 y 30 años, siempre presente en fiestas o en círculos de conversación, utilizándose para fumar tabaco glicerinado, también conocido como esencia. Conociendo los efectos nocivos del tabaco, utilizado como principal componente de las pipa de agua, como el cáncer y los problemas respiratorios, también puede contener otras posibles causas de enfermedades, como los microorganismos patógenos. Por lo tanto, a pesar de la gran popularidad de esta práctica, todavía hay poca investigación al respecto. Por tanto, el objetivo de este trabajo fue realizar análisis microbiológicos de los componentes de la cachimba con el fin de encontrar bacterias de origen patológico o no. Para ello, se utilizó la metodología de inoculación in vitro, en placa de Petri conteniendo agar solidificado, en los casos de crecimiento bacteriano, los microorganismos fueron aislados y sometidos a identificación mediante pruebas bioquímicas. Se encontraron E. coli, E. aerogenes, K. pneumoniae en los trozos de narguile. Por lo tanto, los fumadores necesitan una buena higiene y otras contramedidas para evitar infecciones. O narguilé é uma nova tendência dos jovens entre 20 e 30 anos, sempre presente em festas ou em rodas de conversas, sendo usado tabaco glicerinado para o seu fumo, também conhecida como essência. Sabendo os malefícios do tabaco, utilizado como principal componente do narguilé, como câncer e problemas respiratórios, ele também pode conter outras possíveis causas para doenças, como micro-organismos patogênicos. Assim, apesar da grande popularização dessa prática, existem poucas pesquisas ainda sobre o mesmo. Diante disso, o objetivo desse trabalho foi realizar análises microbiológicas dos componentes do narguilé a fim de encontrar bactérias de origem patológica ou não. Para isso, foi utilizada a metodologia de inoculação in vitro, em placa de Petri contendo ágar solidificado, em casos de crescimento bacteriano, os microrganismos foram isolados e submetidos a identificação por provas bioquímicas. Foram encontradas E. coli, E. aerogenes, K. pneumoniae, nas peças do narguilé. Portanto é necessária uma boa higienização e outras contramedidas para os fumantes evitarem infecções.

Parkinson's disease (PD) induced by environmental toxins involves a multifactorial cascade of harmful factors, thus motivating the search for therapeutic agents able to act on the greatest number of molecular targets. This study evaluated the efficacy of 50 mg/kg purified anacardic acids (AAs), isolated from cashew nut shell liquid, on multiple steps of oxidative stress and inflammation induced by rotenone in the substantia nigra (SN) and striatum. Adult mice were divided into four groups: Control, rotenone, AAs + rotenone, and AAs alone. Lipoperoxidation, nitric oxide (NO) levels, and reduced glutathione (GSH)/oxidized gluthatione (GSSG) ratio were evaluated. NF-kB-p65, pro-IL-1β, cleaved IL-1β, metalloproteinase-9, Tissue Inhibitory Factor-1 (TIMP-1), tyrosine hydroxylase (TH), and glial fibrillary acidic protein (GFAP) levels were assessed by Western blot. In silico studies were also made using the SwissADME web tool. Rotenone increased lipoperoxidation and NO production and reduced TH levels and GSH/GSSG ratio in both SN and striatum. It also enhanced NF-kB-p65, pro, and cleaved IL-1β, MMP-9, GFAP levels compared to control and AAs groups. The AAs alone reduced pro-IL-1β in the striatum while they augmented TIMP1 and reduced MMP-9 amounts in both regions. AAs reversed rotenone-induced effects on lipoperoxidation, NO production, and GSH/GSSG ratio, as well as increased TH and attenuated pro-IL-1β and MMP-9 levels in both regions, NF-kB-p65 in the SN and GFAP in the striatum. Altogether, the in vivo and in silico analysis reinforced multiple and defined molecular targets of AAs, identifying that they are promising neuroprotective drug candidates for PD, acting against oxidative and inflammatory conditions induced by rotenone.

O SUS, Sistema Único de Saúde, é um dos sistemas de saúde público mais complexos do mundo, pautado por três princípios: universalidade, integralidade e equidade, ele se desdobra em fundamentais diretrizes organizativas, que buscam garantir o seu funcionamento conforme prevê a Constituição Federal e a lei 8080/90. Para uma prestação de serviço de qualidade a população e que atenda às necessidades, umas das diretrizes fundamentais do SUS é a descentralização de ações e serviços de saúde, a qual redistribui o poder e responsabilidade entre os três níveis de governo, ressaltando que os municípios devem ter condições gerenciais, técnicas, administrativas e financeiras para exercer esta função. O processo de descentralização alcançou até a municipalização da gestão dos serviços, o que certamente representou a quebra de um paradigma. Assim, para cada esfera de poder regional (União, Estado e Município) haveria um responsável local, mas articulado com as outras esferas. Na gestão descentralizada do SUS, surge a necessidade de fortalecer o processo de regionalização, hierarquização e integração das ações e serviços de saúde, bem como a necessidade de fortalecimento dos instrumentos de gestão que garantem a organização das redes e fluxos assistenciais. Para isso instituiu a Política Nacional de Regulação em 2008 que se dividiu em três dimensões: a Regulação de Sistemas de Saúde, a Regulação da Atenção à Saúde e a Regulação do Acesso à Assistência, denominado também por alguns como Regulação de Acesso ou Regulação Assistencial. É essa temática especificamente no sistema referência e contrarreferência que surgi a minha indagação e interesse sobre o assunto. O termo Regulação na área da saúde, tem vários desdobramentos, concepções e práticas. Trataremos nesse trabalho, a definição instituída pela Política Nacional de Regulação do Sistema Único de Saúde – SUS, focando na Regulação do Acesso à Assistência (regulação do acesso ou regulação assistencial), a qual possui como o objeto o acesso aos serviços de saúde e respectivos gestores. Afinal, para o Ministério da Saúde (2006), a regulação assistencial é o conjunto de relações, saberes, tecnologias e ações que intermedeiam a demanda das pessoas usuárias por serviços de saúde e o acesso a eles segundo diferentes perfis de demanda e de oferta, e a regulação do acesso é o estabelecimento de meios e ações, independente de pactuação prévia estabelecida na programação de ações e serviços de saúde e da disponibilidade de recursos financeiros. A regulação assistencial deve organizar a referência e contrarreferência entre os níveis de complexidade assistencial, orientando os processos de trabalho dos profissionais e seus gestores. Para isso, são necessárias sempre revisões e discussões da questão, de forma a permitir a sua utilização como instrumento aos profissionais da atenção básica, já que essa é a porta de entrada do SUS. Há necessidade de se instituir mecanismos mais eficazes da regulação assistencial e estabelecer uma operacionalidade melhor entre as instâncias, pois mesmo com portarias e decretos o acesso de serviços de saúde se mostra a cada dia mais difícil, mesmo considerando uma escassez nas ofertas, o que queremos tratar é a dificuldade do acesso visto que um dos papeis desse instrumento é ajudar no exercício da integralidade e não para cercear e dificultar a prática. Objetivo: Fazer uma revisão da literatura sobre o tema da regulação assistencial e a integralidade Método: O trabalho em questão trata-se de uma revisão bibliográfica, embora o interesse pelo tema escolhido ter partido da prática profissional. É uma revisão narrativa, sistematizada que utilizou três descritores para sua busca na literatura. De acordo com o objeto de estudo escolhido, no caso a “Regulação Assistencial” foram encontrados 5 (cinco) artigos governamentais extraídos do portal do Ministério da Saúde que nortearam e colaboraram para determinar as palavras chaves a serem pesquisadas nas bases dados da biblioteca virtual em saúde, isso porque no primeiro momento, a pesquisa pelo portal da BVS não trazia artigos que realmente tratassem o assunto desejado. Os descritores utilizados são: “Gestão em Saúde”, “Regulação em Saúde” e “Acesso aos serviços de Saúde”. No primeiro momento, foi realizado uma busca individual com cada descritor que trouxe o resultado abaixo “gestão em saúde” (2880) , “regulação em saúde”(42) e o acesso aos serviços de saúde(68448). Com as combinações dos descritores na BVS foi possível o seguinte retorno::” “Gestão em saúde” and “regulação em saúde” (8), “ Gestão em saúde” and “Acesso aos serviços de saúde” (70) e ““Regulacao em saúde” and “Acesso aos serviços de saúde”(6). Resultados: Para sistematizar e obter um resultado mais amplo, a sintaxe final do trabalho é: tw:(gestão em saúde)) AND (tw:(acesso aos serviços de saúde)) AND (tw:(regulação em saúde)) AND (instance:"regional"), a qual resultou em 153 artigos. Essa coleta de dados também não utilizou filtros e só após a busca selecionada que iniciou o processo de exclusão dos trabalhos que não estavam em acordo com o objetivo. Para excluir os trabalhos que não tratam a temática de estudo, foi feito a leitura de todos os títulos e resumos, após essa leitura foram excluídos os documentos que não abordavam sobre o sistema único de saúde brasileiro, a gestão da regulação de acesso aos serviços de saúde e os artigos duplicados. Esse processo de exclusão, resultou em 35 trabalhos, os quais foram submetidos ao registro na íntegra para um planilhamento e leitura completa posteriormente. Contudo, o assunto de interesse se mostrou um tema pouco discutido até o momento nas bases de dados e como já dito, por isso a necessidade de recorrer a uma pesquisa extra da plataforma BVS referenciando o assunto desejado, sendo incluindo cincos artigos governamentais extraídos do site do Ministério da Saúde,incluindo portarias e políticas.

Anacardic acids (AAs) are alkyl phenols mainly presenting in cashew nuts. The antioxidant effects of these compounds have been an area of interest in recent research, with findings suggesting potential therapeutic use for certain diseases. Nevertheless, none of these studies were performed in order to test the hypothesis of whether anacardic acids are capable of preventing behavioral changes and oxidative stress induced by the pesticide rotenone in experimental model of Parkinson's disease. In our research, adult male rats were treated orally with AAs (1, 3, 10, 25, 50, or 100 mg/kg/day) 1 h before rotenone (3 mg/kg; s.c.) for five consecutive days. The behavioral testing strategies, including tests for general locomotor activity (open field), motor coordination (rotarod), and spatial memory performance (elevated T-maze), were carried out. Lipoperoxidation levels and total superoxide dismutase (t-SOD) activity, as well as cytoplasmic and mitochondrial SOD gene expression, were assessed in the substantia nigra (SN), striatum, and cerebral cortex. The results showed that AAs dose-dependently prevented the rotenone-induced learning and motor impairment from 10 mg/kg/day. AAs also precluded rotenone-induced lipoperoxidation in all doses, acting directly on the mitochondria, and improved the t-SOD activity in the doses 25-100 mg/kg/day. AAs per se (100 mg/kg/day) increased SOD gene expression and t-SOD activity. Our findings indicate that the oral administration of AAs prevents rotenone-induced behavioral changes and oxidative stress, in part due to a modulatory action on the mitochondria and SOD gene expression. These data suggest that AAs have promising neuroprotective action against degenerative changes in Parkinson's disease.

Nutritional changes that lead to increase or deficiency of macro- or micronutrients may modify the normal course of neural development as a product of changes in epigenetic mechanisms. N-3 essential fatty acids play a key role in neural stem cells, astrocyte and neuron proliferation and differentiation, intercellular communication, exert antiinflammatory, and antioxidant effects. Emerging research supports the evidence that essential n-3 polyunsaturated fatty acids may modify the epigenome as one of the mechanisms involved in their beneficial actions during brain development and senescence. The purpose of this review is to identify epigenetic mechanisms related to the effects of these fatty acids on neural cells’ development and those implicated in their potential therapeutic use as a nutraceutic for some neurodegenerative diseases. The current data reinforce the importance of an adequate supplementation of these macronutrients on epigenetic modulation, either in physiological or pathological conditions.

The cerebellum is vulnerable to malnutrition effects. Notwithstanding, it is able to incorporate higher amount of docosahexaenoic acid (DHA) than the cerebral cortex (Cx) when low n-6/n-3 fatty acid ratio is present in a multideficient diet. Considering importance of DHA for brain redox balance, we hypothesize that this cerebellum feature improves its antioxidant status compared to the Cx. A chronic malnutrition status was induced on dams before mating and kept until weaning or adulthood (offspring). A group nutritionally rehabilitated from weaning was also analyzed. Morphometric parameters, total-superoxide dismutase (t-SOD) and catalase activities, lipoperoxidation (LP), nitric oxide (NO), reduced (GSH) and oxidized (GSSG) glutathione, reactive oxygen species (ROS), and reduced nicotinamide adenine dinucleotide/phosphate levels were assessed. Both ROS and LP levels were increased (∼53 %) in the Cx of malnourished young animals while the opposite was seen in the cerebellum (72 and 20 % of the control, respectively). Consistently, lower (∼35 %) and higher t-SOD (∼153 %) and catalase (CAT) (∼38 %) activities were respectively detected in the Cx and cerebellum compared to the control. In malnourished adult animals, redox balance was maintained in the cerebellum and recovered in the Cx (lower ROS and LP levels and higher GSH/GSSG ratio). NO production was impaired by malnutrition at either age, mainly in the cerebellum. The findings suggest that despite a multinutrient deficiency and a modified structural development, a low dietary n-6/n-3 ratio favors early antioxidant resources in the male cerebellum and indicates an important role of astrocytes in the redox balance recovery of Cx in adulthood.

Ionizing radiation can cause biological changes in different organisms such as mollusks from Biomphalaria glabrata species, in which alterations could be observed in the reproductive system of the specimens, prejudicing fertility and fecundity. As the changes may occur due to the lipid peroxidation caused by the action of free radicals on the gonads, the objective of this work was to evaluate the oxidative damage caused by the exposure of B. glabrata mollusks to different doses of 60Co gamma radiation. In addition, efforts were carried out to standardize a sensitive and low-cost technique for detecting negative effects caused by high doses of ionizing radiation. For this, each mollusk group (n = 10) was submitted to 0 (control), 10, 15, 20 and 25 Gy (gammacell 60Co, dose rate 3.53 kGy/h).
