10. Identification and characterization of a novel Pavlovskyella(Acari: Argasidae) from Chile, parasite of the culpeo fox (Lycalopex culpaeus)

Abstract

Soft ticks (Argasidae) of the subgenus PavlovskyellaPospelova-Shtrom are worldwide distributed parasites of medical importance. However, the systematics of the subgenus are currently under debate because genetic data shows that the group is paraphyletic. Meanwhile, species of Pavlovskyellacontinue to be discovered. In this study a novel species of the subgenus is described from specimens collected on a fox in central Chile. The larva of this new species differentiates from other Pavlovskyellaspp. by having the following combination of characters: subpyriform dorsal plate; 15 pairs of setae, 7 anterolateral, 3 central, and 5 posterolateral, and hypostome with denticles in the distal third. Nymphs and adults of the species lack cheeks, eyes or bulging structures on the flank, but exhibit dorsoventral grooves, and humps on tarsi I, II, and III. Moreover, a patch of glabrous integument appears on the distal portion of coxal folds. A phylogenetic analysis using the mitogenome indicates a monophyletic group composed by Ornithodoros(Pavlovskyella) brasiliensisAragão, Ornithodoros(Pavlovskyella) furcosusNeumann, Ornithodoros(Pavlovskyella) improvisusMuñoz-Leal & Venzal, and Ornithodoros(Pavlovskyella) rostratusAragão. Interestingly, a phylogeny using 18S–28S rDNA sequences shows that South American Pavlovskyellaspp. are paraphyletic, as depicted in previous studies. Adding species of the subgenus from the Neotropical region to phylogenetic analyses could aid to solve this paraphyly. Furthermore, this is the fifth species of Pavlovskyelladescribed in South America, and the second in Chile.

Abstract

Adaptation is key for survival as vector-borne pathogens transmit between the arthropod and vertebrate, and temperature change is an environmental signal inducing alterations in gene expression of tick-borne spirochetes. While plasmids are often associated with adaptation, complex genomes of relapsing fever spirochetes have hindered progress in understanding the mechanisms of vector colonization and transmission. We utilized recent advances in genome sequencing to generate the most complete version of the Borrelia turicatae 150 kb linear megaplasmid (lp150). Additionally, a transcriptional analysis of open reading frames (ORFs) in lp150 was conducted and identified regions that were up-regulated during in vitro cultivation at tick-like growth temperatures (22°C), relative to bacteria grown at 35°C and infected murine blood. Evaluation of the 3' end of lp150 identified a cluster of ORFs that code for putative surface lipoproteins. With a microbe's surface proteome serving important roles in pathogenesis, we confirmed the ORFs expression in vitro and in the tick compared to spirochetes infecting murine blood. Transcriptional evaluation of lp150 indicates the plasmid likely has essential roles in vector colonization and/or initiating mammalian infection. These results also provide a much needed transcriptional framework to delineate the molecular mechanisms utilized by relapsing fever spirochetes during their enzootic cycle.

Abstract

RESUMEN Las espiroquetas causantes de la fiebre recurrente (FR) son patógenos del géneroBorreliatransmitidos por garrapatas blandas y duras. Esta enfermedad se considera como desatendida y emergente. Aunque la enfermedad se ha descrito y diagnosticado en México, al momento no se ha logrado el aislamiento de la bacteria causante de la misma. De tal forma, el objetivo de este trabajo fue obtener el primer aislado mexicano deBorreliaspp. causante de FR transmitida por garrapatas de vectores colectados en México. En marzo del 2022 se realizó una colecta de garrapatas en los estados de Sonora y Sinaloa donde previamente se tenían reportes serológicos de exposición de personas a esta enfermedad. Las garrapatas vivas agrupadas en lotes se pusieron en contacto con ratones DBA/2J para su alimentación y posterior vigilancia, se realizaron frotis de sangre periférica y tinción Giemsa del día 02 al día 18 posterior a la alimentación. A las 4 semanas se obtuvo suero y se realizó un Western blot para detectar anticuerpos IgG contra la proteína recombinante glicerofosfodiesterasa (rGlpQ). La prueba de Western blot mostró que un suero fue positivo contra la rGlpQ, por lo que las garrapatasO. turicatapositivas se alimentaron en un nuevo ratón DBA/2J y a los 7 días se tomó sangre periférica y se inoculó en medio Barbour-Stoenner-Kelley IIb (mBSK IIb) en donde se observaron espiroquetas a los 9 días de la inoculación. Se realizó la extracción de DNA y se realizó la confirmación por PCR y secuenciación del gen que codifica para el rRNA 16S, que indicó que se trataba deBorrelia turicatae. Posteriormente, el DNA genómico de esta cepa se secuenció con las plataformas Ilumina y Nanopore, las secuencias resultantes se ensamblaron, se anotaron y se analizaron filogenéticamente con otros genomas depositados en el NCBI. El resultado indica que esta cepa deB. turicataeestá emparentada con otras aisladas en Norteamérica y que representa una cepa nueva; tiene un genoma de 1.57 Mpb con las siguientes características: 29.4% GC, 1 cromosoma lineal, 13 plásmidos lineales y 3 plásmidos circulares. En conclusión, se obtuvo una cepa deBorrelia turicataede garrapatasOrnithodoros turicatadel estado de Sinaloa, México. Estos resultados sugieren que se debe ampliar el estudio a otras áreas para conocer las zonas de distribución de las garrapatas blandas y de la bacteria causante de FR en México.

25. Description of a new Pavlovskyellaspecies (Acari: Argasidae) from Chile

Abstract

Soft ticks (Argasidae) of the PavlovskyellaPospelova-Shtrom subgenus are important vectors of relapsing fever spirochetes, which are agents of disease globally. South American representatives of the Pavlovskyellasubgenus include 3 species: Ornithodoros(Pavlovskyella) brasiliensisAragão, Ornithodoros(Pavlovskyella) furcosusNeumann, and Ornithodoros(Pavlovskyella) rostratusAragão. Here, we describe a fourth species based on morphological and mitogenomic evidence of ticks collected in burrows of unknown hosts in central Chile. The larva of the new species separates from other South American soft ticks by the following combination of characters: 13 pairs of dorsolateral setae, dorsal plate hexagonal, hypostome blunt with denticles from apex almost to the base. Adults of this new species lack cheeks, possess a dorsoventral groove, and have humps, similar to O.(P.) brasiliensis; however, they lack bulging structures on the flanks of idiosoma. Moreover, females and males differ from O.(P.) rostratusby having 3 humps instead of spurs in tarsi I and from O.(P.) furcosusbecause of their smaller size and thinner anterior lip of the genital aperture in females. The phylogenetic analysis performed with mitogenomes of the Argasidae family depicts the new Pavlovskyellaspecies from Chile in a monophyletic clade with other South American species in the subgenus, confirming a regional group.

Abstract

Borrelia hermsii is a blood-borne pathogen transmitted by the argasid tick Omithodoros hermsi. Since spirochaete clearance in mice is associated with an IgM-mediated response, an immunoproteomic analysis was used to identify proteins reactive with IgM. We report that IgM from both mice and human patients infected with B. hermsii not only reacted with the previously identified variable membrane proteins but also identified candidate antigens including heat-shock proteins, an adhesin protein, ABC transporter proteins, flagellar proteins, housekeeping proteins, an immune evasion protein, and proteins with unknown function. Furthermore, IgM reactivity to recombinant glycerophosphodiester phosphodiesterase was detected during early spirochaete infection and prior to a detectable IgG response. Lastly, a conserved hypothetical protein was produced in Escherichia coil and tested with immune serum against B. hermsii and Borrelia recurrentis. These results identify a much larger set of immunoreactive proteins, and could help in the early serodiagnosis of this tick-borne infection.
