However, the role of processing of precursor family (POP family) in clear cell renal cell carcinoma (ccRCC) has not been studied yet. Here, we analyzed the expression and prognostic value of POP family in ccRCC analyzed and subsequently revealed the relationship between POP7 and immune infiltration in ccRCC patients.Methods: POP family expression in cancer and normal tissues was analyzed in Cancer Genome Atlas pan-cancer (TCGA-pan-cancer). Kaplan–Meier (KM) survival analysis, univariable and multivariable analysis demonstrated the survival of ccRCC with POP family in Kidney Clear Cell Carcinoma (TCGA-KIRC). POP7 mRNA and protein expression were verified by Gene Expression Omnibus (GEO) data, the quantitative real-time polymerase chain reaction (qRT-PCR), and Office of Cancer Clinical Proteomics Research (CPTAC). The diagnostic ability of POP7 mRNA and protein expression was achieved with ROC curves. Gene Set Enrichment Analysis (GSEA) and TiMER2 evaluated pathogenesis role and immune infiltration of POP7in ccRCC.Results: There is a significant difference in expression of POP family in TCGA-pan-cancer, especially in ccRCC. KM survival analysis, univariable and multivariable analysis demonstrated low expression of POP7 and was associated with poor OS and poor DFS. GEO data, the qRT-PCR, and CPTAC verified the high expression of POP7 mRNA and protein in ccRCC. ROC curves verified a valuable diagnostic ability of POP7 in mRNA and protein expression. GSEA demonstrated POP7 was associated with CD8+cells, CD4+cells, natural killer (NK) cells, and helper T (TH1) cells. TiMER2 results indicated POP7 had a positive correlation with T cell regulatory (Tregs) and myeloid-derived suppressor cells (MDSC) in ccRCC and was an immunosuppressor for ccRCC.Conclusion: POP7 was a reliable immunosuppressor, predictor and biomarker for ccRCC.Keywords: POP7, ccRCC, immunotherapy, prognostic indicator

The deoxynucleotidyl transferase terminal interacting protein 1 (DNTTIP1) was reported to promote multiple tumor progression. However, there is no research about DNTTIP1 in ccRCC.Methods: Kaplan–Meier survival analysis, multivariate analysis demonstrated the prognostic indicator in overall survival (OS) and disease-free survival (DFS) of ccRCC with DNTTIP1 expression in the Cancer Genome Atlas Kidney Clear Cell Carcinoma (TCGA-KIRC). Receiver operator characteristic (ROC) curve analyzed diagnostic ability of DNTTIP1 in TCGA-KIRC and validation dataset. The quantitative real-time polymerase chain reaction (qRT-PCR) detected the DNTTIP1 expression in renal cancer tissues, and the Office of Cancer Clinical Proteomics Research (CPTAC) verified the protein expression of DNTTIP1. Moreover, nomogram predicted the role of DNTTIP1 in ccRCC patient. Single-sample Gene Set Enrichment Analysis (SsGSEA) and GSEA evaluated the pathogenesis role of DNTTIP1 in TCGA-KIRC.Results: DNTTIP1 expression was higher in ccRCC tumor tissues. High expression of DNTTIP1 was associated with poor OS (HR = 1.618, P < 0.0001), and poor DFS (HR = 1.789, P < 0.0001). SsGSEA and GSEA showed DNTTIP1 was associated with hypoxia, epithelial-mesenchymal transition (EMT), angiogenesis, G2M checkpoint. DNTTIP1 had a positive correlation with EMT biomarkers in ccRCC, and might be an effective target for ccRCC.Conclusion: This study provided that higher expression of DNTTIP1 predicted poor prognosis in ccRCC, and DNTTIP1 might be a novel detection biomarker and therapeutic target of tumor malignant in the future.Keywords: DNTTIP1, ccRCC, gene therapy, prognostic indicator

This study was designed to demonstrate the relationship between sarcopenia and systemic inflammatory response (neutrophil/lymphocyte ratio [NLR], platelet/lymphocyte ratio [PLR], and large platelet/lymphocyte ratio [LPLR]) prior to radical gastrectomy for gastric cancer. Patients and methods: We conducted a prospective study of gastric cancer patients who underwent radical gastrectomy. The clinical utility of the NLR, PLR, and LPLR was evaluated by receiver operating characteristic curves. Sarcopenia components including skeletal muscle index, handgrip strength, and 6 m usual gait speed were measured. Logistic analysis was used to identify the independent indices associated with sarcopenia. Results: A total of 670 patients were included, representing 504 men and 166 women. Of these, 104 patients (15.5%) were diagnosed with sarcopenia and 567 (84.5%) were non-sarcopenia. PLR has a diagnostic sensitivity of 91.3% for sarcopenia. 13. A cryogenic-helium pipe flow facility with unique double-line molecular tagging velocimetry capability

Abstract

Cryogenic helium-4 has extremely small kinetic viscosity, which makes it a promising material for high Reynolds ($Re$) number turbulence research in compact laboratory apparatuses. In its superfluid phase (He II), helium has an extraordinary heat transfer capability and has been utilized in various scientific and engineering applications. In order to unlock the full potential of helium in turbulence research and to improve our understanding of the heat transfer mechanism in He II, a flow facility that allows quantitative study of helium heat-and-mass transfer processes is needed. Here we report our work in assembling and testing a unique helium pipe flow facility that incorporates a novel double-line molecular tracking velocimetry (DL-MTV) system. This flow facility allows us to generate turbulent pipe flows with $Re$ above $10^7$, and it can also be adapted to produce heat-induced counterflow in He II. The DL-MTV system, which is based on the generation and tracking of two parallel thin He$^*_2$ molecular tracer lines with an adjustable separation distance, allows us to measure not only the velocity profile but also both the transverse and longitudinal spatial velocity structure functions. We have also installed a deferential pressure sensor to the flow pipe for pressure drop measurement. The testing results of the flow facility and the measurement devices are presented. Abstract

A preregistered meta-analysis, including 244 effect sizes from 85 field audits and 361,645 individual job applications, tested for gender bias in hiring practices in female-stereotypical and gender-balanced as well as male-stereotypical jobs from 1976 to 2020. A “red team” of independent experts was recruited to increase the rigor and robustness of our meta-analytic approach. A forecasting survey further examined whether laypeople (n = 499 nationally representative adults) and scientists (n = 312) could predict the results. Forecasters correctly anticipated reductions in discrimination against female candidates over time. However, both scientists and laypeople overestimated the continuation of bias against female candidates. Instead, selection bias in favor of male over female candidates was eliminated and, if anything, slightly reversed in sign starting in 2009 for mixed-gender and male-stereotypical jobs in our sample. Forecasters further failed to anticipate that discrimination against male candidates for stereotypically female jobs would remain stable across the decades.

Abstract

Back exosuits deliver mechanical assistance to reduce the risk of back injury, however, minimising restriction is critical for adoption. We developed the adaptive impedance controller to minimise restriction while maintaining assistance by modulating impedance based on the user's movement direction and nonlinear sine curves. The objective of this study was to compare active assistance, delivered by a back exosuit via our adaptive impedance controller, to three levels of assistance from passive elastics. Fifteen participants completed five experimental blocks (4 exosuits and 1 no-suit) consisting of a maximum flexion and a constrained lifting task. While a higher stiffness elastic reduced back extensor muscle activity by 13%, it restricted maximum range of motion (RoM) by 13°. The adaptive impedance approach did not restrict RoM while reducing back extensor muscle activity by 15%, when lifting. This study highlights an adaptive impedance approach might improve usability by circumventing the assistance-restriction trade-off inherent to passive approaches. Practitioner summary: This study demonstrates a soft active exosuit that delivers assistance with an adaptive impedance approach can provide reductions in overall back muscle activity without the impacts of restricted range of motion or perception of restriction and discomfort. 42. Development of Offline E-Modules to Improve Learners’ Academic Performance in Disciplines and Ideas in the Social Sciences

Abstract

As the education system in the Philippines has adapted to distance learning due to the global pandemic, self-learning modules also have become the primary learning resources of the learners. However, many modules distributed to learners were criticized due to erroneous contents, level of difficulty, number of tasks, and printing costs. Meanwhile, there were lack of modules for specialized subjects in some public senior high schools. In this regard, this study attempted to develop and validate offline yet electronic modules in one of the specialized subjects in senior high school, Disciplines and Ideas in the Social Sciences (DISS). In developing these, the study used Normative Design and Analysis, Design, Develop, Implement, and Evaluation (ADDIE) model. Twenty (20) Social Science and five (5) ICT teachers in the Division of Biñan City were asked to validate the developed e-modules, while thirty (30) learners from Biñan Integrated National High School were asked to participate in the Implementation Phase. An adopted questionnaire was given to the validators to assess the e-modules’ validity and acceptability while pretests and post-tests were given to the participating learners. Through the ADDIE model, the study was able to design and develop offline e-modules named Disciplines and Ideas in the Social Sciences Knowledge-Amplifying Remote Training E-Modules (DISSKARTE). Using the four-point Likert Scale and the simple mean, findings revealed that its objectives, concepts, topics, directions, exercises, and reflections were all highly valid. The findings showed that its functionality, accuracy, suitability, usability, and efficiency were all highly accepted. Pretest results showed that most of the participants did not meet expectations while the post-test results revealed otherwise as the majority got outstanding and very satisfactory remarks. T-test for dependent means revealed that there were significant differences in the pretests and post-tests. This implied that through DISSKARTE, the learners’ performance on DISS can improve. The study is limited to developing e-modules covering only a quarter of DISS subject. However, it is hoped that the study was able to contribute to the world of literature as there were only few to zero studies about developing instructional materials for DISS especially in the context of new normal.
