Considering growing participatory turns in regulatory scientific risk analysis, this paper compares how social scientists use participatory and analytical methods to understand risk perceptions and meet competing demands for representativeness and inclusiveness. Drawing on case studies of how three European risk agencies use participatory and analytic methods in the context of biotechnology, it confirms difficulties of analytic methods to shed light on perceptions when applied to unfamiliar topics. It also shows the potential of participatory in particular deliberative formats to engage affected populations in the risk analysis process, despite challenges in promoting inclusiveness. The cases call for the integration of methods, while remaining aware of the need to understand the mutual interplay in the constructions of risks and structural inequalities.

Secondary metabolites (SMs) are compounds with relevant biological activities. Their production under laboratory conditions, especially in broth, is still challenging. An example is the pedopeptins, which are nonribosomal peptides active against some bacteria listed by the WHO for which new antibiotics are urgently needed. Their biosynthesis is inhibited by high concentrations of peptone from casein (PC) in tryptic soy broth (TSB), and we applied a RNA-seq approach to identify Pedobacter lusitanus NL19 cellular pathways modulated by this condition. Results were validated by qPCR and revealed 261 differentially expressed genes (DEGs), 46.3% of them with a predicted biological function. Specifically, high concentration of PC significantly repressed the de novo biosynthesis of biotin (- 60X) and the production of nonribosomal peptide synthetases (NRPS) of pedopeptins (about - 14X), but no effect was observed on the expression of other NRPS. Através da primeira edição do Orçamento Participativo Portugal, o ano de 2017 viu nascer uma oportunidade para promover o envelhecimento ativo através de um projeto pioneiro em Portugal, intitulado “Grupos de Canto para Seniores”. Na conceção do programa deste projeto foram tidas em consideração recomendações teóricas e práticas consolidadas, baseadas em evidências empíricas, atendendo ainda ao parecer e à experiência das equipas interdisciplinares que muito contribuíram com as suas competências científicas, técnicas e sociais e que foram, por isso, fundamentais para uma implementação bem-sucedida. O presente livro apresenta a memória deste projeto, evidenciando as diferentes fases de implementação do projeto e dando voz a vários dos seus intervenientes.

Zika virus (ZIKV) has caused an unprecedented epidemic linked to severe congenital syndromes. Transmission of ZIKV in the Americas was first confirmed in May 2015 in northeast Brazil, though the virus was likely introduced 1–2 years prior to its detection. Manaus, the capital city of the Amazonas State, the largest territory of any state in Brazil and the main economic center in the northern region, reported between 2016 and 2017 more than 2,327 suspected cases of ZIKV infection. To gain insights into the timing, source, and likely route(s) of ZIKV introduction in the Amazonas State, we tracked the virus by sequencing ZIKV genomes from infected patients. Using nanopore sequencing technology, we generated 56 Brazilian ZIKV genomes from Manaus city in the Amazonas state, sampled from human cases. On the basis of available sequences of isolates from the Americas, the Manaus sequences, we analyzed fell within a single strongly supported monophyletic clade (bootstrap support = 99%, posterior support = 1.00) that belongs to the Asian genotype. Genetic analysis suggests the outbreak most likely originated from transmission cycles not previously identified in North Brazil and not from a separate introduction into the Americas. Molecular dating analysis indicates that the outbreak was caused by a single founder strain that is estimated to have arrived in Manaus around February 2015. By analyzing surveillance and genetic data, we discovered that ZIKV moved among transmission zones in Manaus. Geographical analysis further indicates that the Northern part of the Manaus regions has a high transmission potential for ZIKV. Our work illustrates that near-real time genomics in the field can augment traditional approaches to infectious disease surveillance and control. Estimated dates for the international spread of ZIKV from the north region indicate the persistence of the virus transmission in recipient regions. Our study provides an understanding of how ZIKV initiates transmission in new regions.

No âmbito da investigação de doutoramento “Jornalismo de Cinema em Portugal”, financiada pela FCT, procuramos conhecer as características da cobertura jornalística aos filmes portugueses Diamantino, Variações, A Herdade e Vitalina Varela. O Jornalismo de Cinema constitui um objecto de estudo ainda por explorar pelas ciências sociais e da comunicação. Encarado como um subgénero do Jornalismo Cultural, o Jornalismo de Cinema assume-se como um campo de sentido informativo, crítico e pedagógico. A presente investigação procura, a partir da análise de conteúdo aos artigos dos principais órgãos de comunicação social portugueses (Público, Correio da Manhã, Jornal de Notícias, Diário de Notícias, Expresso e Observador), programas de rádio (Cinemax e A Grande Ilusão) e televisão (Janela Indiscreta, Cinebox e Cartaz Cinema) especializados em cinema, identificar as características da cobertura mediática ao cinema português, especificamente quatro filmes estreados em 2019: Diamantino de Gabriel Abrantes, Variações de João Maia, A Herdade de Tiago Guedes e Vitalina Varela de Pedro Costa. O Jornalismo de Cinema, considerado um subgénero do Jornalismo Cultural, apresenta-se-nos como um objecto de estudo ainda por explorar nas ciências sociais e da comunicação, nomeadamente em Portugal onde a investigação sobre este subgénero é quase inexistente. Uma vez que o cinema é, de entre as manifestações culturais e artísticas, uma das que tem maior presença nos media portugueses (de acordo com dados do projecto A Cultura na Primeira Página [Baptista, 2014; 2017a]), importa aprofundar a investigação sobre esta prática jornalística. No âmbito de uma investigação mais alargada, parece-nos fundamental percorrer os principais títulos da imprensa portuguesa sobre cinema que têm acompanhado a recepção do cinema nacional e internacional e que têm sido cruciais para a construção da cultura cinematográfica dos portugueses. A primeira publicação dedicada em exclusivo ao cinema surgiu na década de 1910 e é a partir da segunda metade do séc. XX que se começa a constatar a aceleração do gosto cinematográfico (Barroso,2008:26) e a consolidação das publicações sobre cinema em Portugal. Durante o processamento do café, por via úmida ou seca, ocorrem processos fermentativos. Estes, podem ser naturais ou induzidos e influenciam a qualidade de bebida. O objetivo desse trabalho foi testar o efeito de métodos de fermentação anaeróbica sobre a qualidade de bebida do Robusta Amazônico (Coffea canephora) produzido no estado Rondônia. A pesquisa foi realizada em propriedade rural localizada no município de Novo Horizonte do Oeste - RO, utilizando o clone 25, amplamente cultivado na região. O estudo se baseou em dois experimentos conduzidos em delineamento de parcelas subdividas para quantificação dos efeitos da fermentação por maceração semicarbônica e da fermentação com inoculação de levedura, considerando os frutos íntegros ou despolpados. Os cafés foram avaliados de acordo com a metodologia Protocolo de degustação de Robustas Finos- PDRF. Não foi observada diferença significativa nas notas de qualidade de bebida entre os tratamentos, indicando que tanto a fermentação quanto o processamento natural têm potencial para produção de bebida fina. Apesar de não ter sido observada diferenças significativas nas notas finais de qualidade da bebida, observa-se expressiva diferença nos descritores de qualidade sensorial, entre os tratamentos fermentados e naturais. O agrupamento a partir dos descritores da análise sensorial indica a formação de dois grupos distintos, formados por tratamentos de processamento fermentado e natural. Os processos de fermentação induzida, via Maceração semicarbônica por 20 dias, se destacaram dos demais, sendo caracterizado como bebida de aspectos frutado, doce e caramelo. The objective of this work was to test the effect of anaerobic fermentation methods on the beverage quality of the Amazonian Robustas (Coffea canephora) grown in the state of Rondônia. This research was carried out on a rural property located in the municipality of Novo Horizonte do Oeste - RO, using the clone 25 cultivated in the region. The study was based on two experiments conducted in the design of subdivided plots to quantify the effects of fermentation by semicarbonic maceration and by fermentation with yeast inoculation, considering whole and pulped fruits. The beverage quality were evaluated according to the Fine Robust Tasting Protocol - PDRF protocol. There was no significant difference in the beverage quality scores between treatments, indicating that fermentation and natural processing have the potential to produce fine drink. The postharvest procedures were performed appropriately considering the estimates of the coefficient of experimental variation and the mean beverage quality score. Although there were no significant differences in the beverage quality scores, there is a significant difference in the descriptors of sensory quality, between fermented and natural treatments. The grouping from the descriptors of the sensory analysis indicates the formation of two distinct groups, formed by fermented and natural processing treatments. The induced fermentation processes, via semicarbonic maceration for 20 days, stood out from the others for its positive attributes, being characterized as a fruity, sweet and caramel drink. Changes in oral health status following treatment should be assessed clinically as well as by patients' reported outcomes. This study investigated changes on oral-health-related quality of life (OHRQoL) of adolescents enrolled in a comprehensive oral health care program for caries and gingival conditions. The sensitivity of the Child Perception Questionnaire11-14 (CPQ11-14) to detect clinical changes, that is, its responsiveness, was assessed. A total of 618 10-15-year adolescents answered the questionnaire before treatment and 560 at follow-up after 1 year. In the follow-up, 2 additional global transition judgment questions were asked. The adolescents were clinically and radiographically examined. At the baseline, 374 adolescents needed only nonoperative treatment, whereas 274 needed non- and operative treatments. After 1 year, the adolescents were regrouped according to the fulfillment of their treatment needs in Group 1 (n = 363) needing/receiving nonoperative treatment only; Group 2 (n = 98) needing/receiving both non- and operative treatments; Group 3 (n = 99) receiving nonoperative treatment only, while needing also operative treatment. The CPQ11-14 total mean change scores by the global transition judgment on self-perceived oral health status indicated significant differences (p < 0.001, ANOVA; internal responsiveness). The effect of the program for the treatment groups was significant (p = 0.014, ANCOVA; external responsiveness). The effect size for the change scores was of moderate magnitude. In conclusion, an overall improvement of adolescents' OHRQoL was observed following 1-year oral health care program for caries and gingival conditions. The CPQ11-14 was internally and externally sensitive to detect substantial clinical changes. The CPQ11-14 seems appropriate for measuring long-term changes on adolescents' OHRQoL.
