Background: The R2R3-MYB transcription factors in plants participate in various physiological and biochemical processes and responds to various external stimuli. Prunus sibirica (known as Siberian apricot) is a drupe tree species that produces extremely high nutritional value kernels. However, it is susceptiblility to frost damage during the flowering period, results in a marked reduction in kernel yield. Results: In this study, the MYB gene family of P. sibirica (PsMYB) was systematically analyzed, and 116 R2R3-MYB genes that were distributed unevenly over eight chromosomes were ultimately screened. Phylogenetic analysis divided these 116 genes into 30 subgroups. We discovered that 37 PsMYBs had cold stress-responsive promoters, and six PsMYBs were annotated to be associated with cold response. Intraspecific homology analysis identified segmental duplication as the primary gene amplification mechanism, and homology analysis of the PsMYB genes with those of five other species revealed phylogenetic relationships with Rosaceae species. Protein interaction studies revealed collaborative regulation of the PsMYB proteins with Arabidopsis protein, and transcriptome analysis identified PsMYB genes that were highly expressed at low temperatures. Additionally, the expression levels of 22 PsMYBs in different tissue parts of P. sibirica and under different low-temperature stress conditions were evaluated using quantitative real-time PCR, with the results verifying that PsMYBs are specifically expressed in different plant parts and may be involved in the growth and development of P. sibirica species. Genes upregulated after exposure to low-temperature stress and likely involved in cold response were identified. Conclusion: This study lays a foundation for understanding the molecular biology of PsMYBs in P. sibirica and provides a theoretical basis for the future study of transgenic lines with cold resistance during the flowering period of this tree. Continual climate change strongly influences temperature conditions worldwide, making ectothermic animals as suitable species for studying the potential impact of climate change on global biodiversity. However, the study of how lizards distributed at different latitudes respond to climate change at the transcriptome level is still insufficient. According to the Climatic Variability Hypothesis (CVH), the range of climate fluctuations experienced by terrestrial animals throughout the year increases with latitude, so individuals at higher latitudes should exhibit greater thermal plasticity to cope with fluctuating environments. Mitochondria, as the energy center of vertebrate cells, may indicate species' plasticity through the sensitivity of gene expression. In this study, we focused on the changes in transcript levels of liver mitochondrial protein-coding genes (PCGs) in skinks from the genus Plestiodon (P. capito and P. elegans) and the genus Scincella (S. modesta and S. reevesii) under low-temperature conditions of 8 °C, compared to the control group at 25 °C. Species within the same genus of skinks exhibit different latitudinal distribution patterns. We found that the two Plestiodon species, P. elegans and P. capito, employ a metabolic depression strategy (decreased transcript levels) to cope with low temperatures. In contrast, the two Scincella species show markedly different patterns: S. modesta exhibits significant increases in the transcript levels of six genes (metabolic compensation), while in S. reevesii, only two mitochondrial genes are downregulated (metabolic depression) compared to the control group. We also found that P. capito and S. modesta, which live at mid-to-high latitudes, exhibit stronger adaptive responses and plasticity at the mitochondrial gene level compared to P. elegans and S. reevesii, which live at lower latitudes. Low temperatures pose a critical challenge to aquaculture operations, as they can significantly affect fish health and overall productivity. The study investigated the impact of low-temperature stress on juvenile golden pompano, focusing on serum indicators, intestinal microbiome, and intestinal transcriptome. With 180 fish divided into low-temperature (18 ± 0.5) ℃ and control (28 ± 0.5) ℃ groups, the gradual temperature decrease mimicked natural seasonal changes. Each group had 3 replicates, with 30 fish per replicate. Results indicated significant effects on serum indicators, including enzyme activity and ion concentrations. While intestinal flora richness was not drastically affected, the dominant flora composition changed. Transcriptome analysis revealed 1752 differentially expressed genes, with enrichment in pathways related to circadian rhythm, proteasome, metabolism, and signaling pathways. This research enhances understanding of cold stress responses in juvenile golden pompano, offering insights for tailored aquaculture management to promote robust and stress-resistant fish stocks. Global climate warming and frequent extreme low-temperature events have made it essential to investigate the impact of low temperatures on parasitic wasps to protect and strengthen farmland biodiversity, which in turn enhances the biological control potential of natural enemies such as parasitic wasps. We systematically examined how low-temperature stress affects the parasitic functional response of Trichopria drosophilae to Drosophila suzukii (Diptera: Drosophilidae) pupae. Our findings indicate that the parasitic behavior of T. drosophilae towards D. suzukii pupae aligns with the Holling II functional response model following exposure to different temperatures. Within the temperature range of 8 °C to −8 °C, lower temperatures correlated decreased instantaneous attack rate of T. drosophilae and an increase in processing time. The search constant Q initially increased and then decreased with declining temperatures. Short-term low-temperature stress negatively impacted the parasitic and searching abilities of T. drosophilae but did not alter its parasitic functional response model. Notably, short-term low-temperature stress had minimal effects on the water content, protein content, and total sugar content of male and female T. drosophilae adults. However, as temperatures decreased, the activities of key enzymes, including GAPDH, SOD, T-AOC, and malondialdehyde (MDA), exhibited an initial increase followed by a decrease. Conversely, the activities of LDH and HOAD decreased, while the activities of CAT and POD increased. Further study on the effect of short-term low temperature on T. drosophilae can provide a research basis for the large-scale production and low-temperature refrigeration technology of T. drosophilae , and provide a scientific basis for its efficient use in the field. Although the gene family has been characterized in various species, such as Oryza sativa, Arabidopsis thaliana, and Populus trichocarpa, studies on the Prunus sibirica AP2/ERF (PsAP2/ERF) gene family are lacking. In this study, PsAP2/ERFs in P. sibirica were characterized by genomic and transcriptomic analyses. Results: In the study, 112 PsAP2/ERFs were identified and categorized into 16 subfamilies. Within each subfamily, PsAP2/ERFs exhibited similar exon-intron structures and motif compositions. Additionally, 50 pairs of segmentally duplicated genes were identified within the PsAP2/ERF gene family. Our experimental results showed that 20 PsAP2/ERFs are highly expressed in leaves, roots, and pistils under low-temperature stress conditions. Among them, the expression of PsAP2/ERF21, PsAP2/ERF56 and PsAP2/ERF88 was significantly up-regulated during the treatment period, and it was hypothesised that members of the PsAP2/ERF family play an important role inlow temperature stress tolerance. Conclusions: This study improves our understanding of the molecular basis of development and low-temperature stress response in P. sibirica and provides a solid scientific foundation for further functional assays and evolutionary analyses of PsAP2/ERFs. Low-temperature (LT) is one of the major abiotic stresses that restrict the growth and development of maize seedlings. Brassinolides (BRs) have been shown to enhance LT tolerance in several plant species; the physiological and molecular mechanisms by which BRs enhance maize tolerance are still unclear. Here, we characterized changes in the physiology and transcriptome of N192 and Ji853 seedlings at the three-leaf stage with or without 2 μM 2,4-epibrassinolide (EBR) application at 25 and 15 °C environments via high-performance liquid chromatography and RNA-Sequencing. Physiological analyses revealed that EBR increased the antioxidant enzyme activities, enhanced the cell membrane stability, decreased the malondialdehyde formation, and inhibited the reactive oxygen species (ROS) accumulation in maize seedlings under 15 °C stress; meanwhile, EBR also maintained hormone balance by increasing indole-3-acetic acid and gibberellin 3 contents and decreasing the abscisic acid level under stress. Transcriptome analysis revealed 332 differentially expressed genes (DEGs) enriched in ROS homeostasis, plant hormone signal transduction, and the mitogen-activated protein kinase (MAPK) cascade. These DEGs exhibited synergistic and antagonistic interactions, forming a complex LT tolerance network in maize. Additionally, weighted gene co-expression network analysis (WGCNA) revealed that 109 hub genes involved in LT stress regulation pathways were discovered from the four modules with the highest correlation with target traits. In conclusion, our findings provide new insights into the molecular mechanisms of exogenous BRs in enhancing LT tolerance of maize at the seedling stage, thus opening up possibilities for a breeding program of maize tolerance to LT stress. Low temperature is a significant abiotic stress factor that not only impacts plant growth, development, yield, and quality but also constrains the geographical distribution of numerous wild plants. Kohlrabi (Brassica oleracea L. var. caulorapa L.) belongs to the Brassicaceae family and has a short growing period. In this study, a total of 196,642 unigenes were obtained from kohlrabi seedlings at low temperatures; of these, 52,836 unigenes were identified as differentially expressed genes. Transcription factor family members ARR-B, C3H, B3-ARF, etc. that had a high correlation with biochemical indicators related to low temperature were identified. A total of nineteen BocARR-B genes (named BocARR-B1–BocARR-B19) were obtained, and these genes were distributed unevenly across seven chromosomes. Nineteen BocARR-B genes searched four conserved motifs and were divided into three groups. The relative expression level analysis of 19 BocARR-B genes of kohlrabi showed obvious specificity in different tissues. This study lays a foundation and provides new insight to explain the low-temperature resistance mechanism and response pathways of kohlrabi. Denphal-type Dendrobium is the famous cut and potted flower in the world, and most cultivated in tropical and subtropical regions. However, it often suffers from cold in winter in subtropical regions. To verify the physiological response of Denphal-type Dendrobium under low temperature in this study, the mature and young plantlets of Dendrobium Udomsri Beauty were treated under 15, 10, and 5 °C, respectively. And then the electrical conductivity (EC), soluble protein, soluble sugar, free proline, malondialdehyde (MDA), chlorophyll content, and the rate of defoliation after regrowth were measured. The results showed that both mature plant and young seedlings of Dendrobium Udomsri Beauty, the EC, soluble protein, soluble sugar, free proline, MDA content, and defoliation rate were increased with the decrease of treatment temperature and the extension of treatment times. The content of chlorophyll decreased gradually with the decrease of treatment temperature and the extension of treatment times. The correlation analysis showed that soluble sugar, free proline, MDA, chlorophyll content, and defoliation rate were significantly correlated with the semi-lethal temperature. It is indicated that the content of free proline, MDA, chlorophyll, and defoliation rate could be used as the effective indexes for the comprehensive assessment of cold tolerance of Dendrobium Udomsri Beauty. Out-of-season breeding is an effective method for addressing seasonal shortages of fry in aquaculture species such as largemouth bass (LMB) for year-round production. Off-season breeding of LMB can be achieved by subjecting breeding LMB to prolonged low-temperature conditions; however, this can alter reproductive rhythms, affecting the quality of their sperm and leading to a decrease in reproductive efficiency. Therefore, it is crucial to investigate issues such as the damage to the testes and the related mechanisms caused by low-temperature stress during out-of-season breeding. In this experiment, we assessed the changes in the testes during this time in LMB by comparing reproductive rhythms, testicular histomorphology, ultrastructure, antioxidant capacity and apoptosis. We synthesized measurements of LMB from three identically treated cement ponds and fish exposed to water temperatures of 13–16 °C to assess the changes in the testes. The results showed that (1) out-of-season reproduction delayed sperm production and promoted sperm redevelopment in LMB, various hormone levels have changed over time (e.g., LH, FSH, and T). (2) The head plasma membrane of LMB spermatozoa was separated, and the middle mitochondria were swollen. (3) The expression levels of antioxidant enzymes (cat, sod, and gpx) were upregulated, and oxidative stress occurred in LMB. (4) The expression levels of apoptosis genes (e.g., bax, bcl2, and caspase3) were upregulated, and apoptosis occurred in LMB due to off-season breeding. Moreover, important genes of the mitochondrial apoptosis pathway (bid, CYT-C) were upregulated, indicating that spermatozoan apoptosis in LMB was probably achieved through the mitochondrial apoptosis pathway. These results suggest the delays, damage, and regeneration of LMB testes. Abstract The cold tolerance of Litopenaeus vannamei is important for breeding in specific areas. To explore the cold tolerance mechanism of L. vannamei, this study analyzed biochemical indicators, cell apoptosis, and metabolomic responses in cold-tolerant (Lv-T) and common (Lv-C) L. vannamei under low-temperature stress (18 °C and 10 °C). Abstract Background The heavy metal-associated isoprenylated plant protein (HIPP) is an important regulatory element in response to abiotic stresses, especially playing a key role in low-temperature response. Results This study investigated the potential function of PavHIPP16 up-regulated in sweet cherry under cold stress by heterologous overexpression in tobacco. The results showed that the overexpression (OE) lines’ growth state was better than wild type (WT), and the germination rate, root length, and fresh weight of OE lines were significantly higher than those of WT. In addition, the relative conductivity and malondialdehyde (MDA) content of the OE of tobacco under low-temperature treatment were substantially lower than those of WT. In contrast, peroxidase (POD), superoxide dismutase (SOD), catalase (CAT) activities, hydrogen peroxide (H2O2), proline, soluble protein, and soluble sugar contents were significantly higher than those of WT. Yeast two-hybrid assay (Y2H) and luciferase complementation assay verified the interactions between PavbHLH106 and PavHIPP16, suggesting that these two proteins co-regulated the cold tolerance mechanism in plants. The research results indicated that the transgenic lines could perform better under low-temperature stress by increasing the antioxidant enzyme activity and osmoregulatory substance content of the transgenic plants. Conclusions This study provides genetic resources for analyzing the biological functions of PavHIPPs, which is important for elucidating the mechanisms of cold resistance in sweet cherry.

Pinus massoniana L. is one of the most important conifer species in southern China and is the mainstay of the forest ecosystem and timber production, yet low temperatures limit its growth and geographical distribution. This study used 30-day-old seedlings from families of varying cold-tolerance to examine the morphological traits of needles and stems, chlorophyll fluorescence characteristics, protective enzymes, and changes in starch and lignin under different low-temperature stresses in an artificial climate chamber. The results showed that the seedlings of Pinus massoniana exhibited changes in phenotypic morphology and tissue structure under low-temperature stress. Physiological and biochemical indexes such as protective enzymes, osmoregulatory substances, starch, and lignin responded to low-temperature stress. The cold-tolerant family increased soluble sugars, starch grain, and lignin content as well as peroxidase activity, and decreased malondialdehyde content by increasing the levels of actual photochemical efficiency (ΦPSII), electron transport rate (ETR), and photochemical quenching (qP) to improve the cold tolerance ability. This study provides a reference for the selection and breeding of cold-tolerant genetic resources of Pinus massoniana and the mechanism of cold-tolerance, as well as the analysis of the mechanism of adaptation of Pinus massoniana in different climatic regions of China. Plant secondary metabolism is profoundly influenced by environmental stress. Low-temperature stress can induce the production of NO and ROS. The root of Astragalus membranaceus Bge. var. mongolicus Hsiao is commonly used as a medicinal herb in Asia. This study aimed to regulate the secondary metabolism and improve the quality of cultivated A. membranaceus var. mongolicus by subjecting it to different temperatures (5, 20, and 5/20°C). The activities and gene expressions of enzymes catalyzing the synthesis of secondary metabolites, including PAL, CHS, HMGR, SQS, were determined. Nitric oxide synthase (NOS) activity and the contents of NO, superoxide radical (), hydrogen peroxide (H2O2), and malondialdehyde (MDA) and five secondary metabolites were measured. Low-temperature stress of 5°C significantly increased NOS activities and NO contents, which led to a considerable surge in the levels of , H2O2, and MDA. Furthermore, the activities of SOD, CAT, and POD, increased rapidly and peaked on the 2nd and 3rd days. The gene expressions and activities of PAL, CHS, HMGR, and SQS were also markedly enhanced, which led to a substantial increase in all secondary metabolites. In the 5°C treatment group, calyculin-7-glucoside, ononin, calycosin, formononetin, and astragaloside IV were increased by 45.6, 36.6, 97.9, 123.4, and 74.9%, respectively, compared with the 20°C treatment group. Thus, low-temperature stress can significantly improve the secondary metabolism of A. membranaceus var. mongolicus, enhance the accumulation of medicinal chemical components, and augment the quality of the herb. Cold stress causes considerable damage to tender tea seedlings. Previous studies have explored changes in the physiological and biochemical factors of tea in response to cold stress; however, the mechanisms of cold resistance in ancient tea tree plants are unclear. The aim of this study was to analyze the effects of 0 °C cold stress for 15 days and 24 °C ambient temperature recovery for 5 days on the physiological and biochemical characteristics of two representative old tea varieties: Dali tea and Siqiu tea. The results revealed significant changes in antioxidant, photosynthetic efficiency, and physiological and biochemical indicators in response to cold stress, with the two species exhibiting different patterns. Cold stress decreased chlorophyll and carotene content, Fv/Fm, Y(II), non-photochemical quenching coefficient, photochemical quenching, and superoxide dismutase (SOD) activity, and increased intercellular CO2 concentration and ascorbate peroxidase activity. Siqiu tea showed a higher increase in soluble sugar content and antioxidant enzyme activity and a lower accumulation of malondialdehyde and minimal fluorescence (F0) than Dali, indicating a greater tolerance to cold stress. Based on partial least-squares discriminant analysis, six key differential physiological indicators of cold resistance—water-soluble sugar, F0, peroxidase, catalase, SOD, and gas conductance—were identified. Hyperspectral imaging was used to predict the total polyphenol content in low-temperature stressed tomato seedlings for the development of a multispectral image sensor. The spectral data with a full width at half maximum (FWHM) of 5 nm were merged to obtain FWHMs of 10 nm, 25 nm, and 50 nm using a commercialized bandpass filter. Using the permutation importance method and regression coefficients, we developed the least absolute shrinkage and selection operator (Lasso) regression models by setting the band number to ≥11, ≤10, and ≤5 for each FWHM. The regression model using 56 bands with an FWHM of 5 nm resulted in an R2 of 0.71, an RMSE of 3.99 mg/g, and an RE of 9.04%, whereas the model developed using the spectral data of only 5 bands with a FWHM of 25 nm (at 519.5 nm, 620.1 nm, 660.3 nm, 719.8 nm, and 980.3 nm) provided an R2 of 0.62, an RMSE of 4.54 mg/g, and an RE of 10.3%. These results show that a multispectral image sensor can be developed to predict the total polyphenol content of tomato seedlings subjected to low-temperature stress, paving the way for energy saving and low-temperature stress damage prevention in vegetable seedling production. Whiteleg shrimp Penaeus vannamei, a highly valuable commercial species in aquaculture, is known to be susceptible to environmental stressors. In this study, we examined how dietary astaxanthin impacts the survival, behavior, and molecular responses of shrimp under acute cold stress. Shrimp were split into two groups: one fed a standard diet, the other fed an astaxanthin-enriched diet. Following feeding periods of 2 and 4 weeks, the shrimp were exposed to an acute temperature stress of 10 °C. Survival rates, behavior, and gene expression (heat shock protein [HSP]70, HSP90, hepatic lectin-like, prophenoloxidase [proPO], lysozyme, hemocyanin C chain-like, catalase [CAT], superoxide dismutase [SOD]) were studied. Astaxanthin as dietary supplement significantly improved survival under cold stress, suggesting enhanced stress tolerance. Molecular analysis showed slightly lower HSP70 and HSP90 expression in the astaxanthin group across various tissues, indicating a potential mechanism by which astaxanthin aids in stress mitigation. The astaxanthin-supplemented diet positively influenced immune and antioxidant gene expression, implying a stronger immune response and defense. These findings highlight astaxanthin's potential to boost stress resilience and overall health in Whiteleg shrimp in aquaculture, resulting in enhanced immune responses and increased disease resistance. To investigate the low-temperature adaptability of different provenances of Ziziphus jujuba var. spinosa, we used 21 clones from seven provenances as experimental materials and observed the changes in physiological and biochemical indicators and the characteristics of anatomical structures under low-temperature stress. A comprehensive evaluation of their cold resistance was conducted using the membership function method. As the temperature decreased, the relative electrical conductivity (REC) of clone 89 became stable and had the lowest LT50 value (−44.04 °C). The cold-resistant Z. jujuba var. spinosa had a higher bound water/free water (BW/FW) ratio and antioxidant enzyme activity and accumulated large quantities of osmotic regulatory substances. Higher xylem, phloem, and xylem–cortex ratios and greater conduit density enhanced the cold resistance of Z. jujuba var. spinosa. The membership function values of clones 89, 90, 91, 604, and 612 were greater than 0.6, indicating that they could be evaluated as resources with the potential for low-temperature resistance. The cold resistance rankings for the different provenances were as follows: Kazuo, Liaoning > Jiaxian, Shaanxi > Fuxing, Heibei > Changqing, Shandong > Neiqiu, Heibei > Yanchuan, Shaanxi > Xiaxian, Shanxi. These results provide a scientific basis for the rapid and accurate identification of cold resistance in Z. jujuba var. spinosa resources and the breeding and cultivation of new cold-resistant varieties of this subspecies. Background: The heavy metal-associated isoprenylated plant protein (HIPP) is an important regulatory element in response to abiotic stresses, especially playing a key role in low-temperature response. Results: This study investigated the potential function of PavHIPP16 up-regulated in sweet cherry under cold stress by heterologous overexpression in tobacco. The results showed that the overexpression (OE) lines' growth state was better than wild type (WT), and the germination rate, root length, and fresh weight of OE lines were significantly higher than those of WT. In addition, the relative conductivity and malondialdehyde (MDA) content of the OE of tobacco under low-temperature treatment were substantially lower than those of WT. In contrast, peroxidase (POD), superoxide dismutase (SOD), catalase (CAT) activities, hydrogen peroxide (H2O2), proline, soluble protein, and soluble sugar contents were significantly higher than those of WT. Yeast two-hybrid assay (Y2H) and luciferase complementation assay verified the interactions between PavbHLH106 and PavHIPP16, suggesting that these two proteins co-regulated the cold tolerance mechanism in plants. The research results indicated that the transgenic lines could perform better under low-temperature stress by increasing the antioxidant enzyme activity and osmoregulatory substance content of the transgenic plants. Simple Summary: In order to withstand extreme temperatures, organisms can change their behavior, physiology, and biochemistry prior to or in reaction to environmental variations. This allows them to enhance their chances of survival under challenging conditions. However, lizards are ectothermic, relying on environmental temperature for body heat and making them highly susceptible to climate change. Mitochondria play a crucial role in maintaining body temperature, regulating metabolic rates, and preventing cellular oxidative damage. If mitochondrial respiratory chain complexes become damaged or disrupted, this can be fatal. Mitochondrial DNA also serves as a sentinel for cell stress. Lizards, as a large group of reptiles, are ectothermic. Their body temperatures are predominantly regulated by their environment and temperature variations directly impact their behavior and physiological activities. Frequent cold periods and heat waves can affect their biochemistry and physiology, and often their ability to maintain their body temperature. Mitochondria, as the center of energy metabolism, are crucial for maintaining body temperature, regulating metabolic rate, and preventing cellular oxidative damage. Here, we used RT-qPCR technology to investigate the expression patterns and their differences for the 13 mitochondrial PCGs in Sphenomorphus incognitus (Squamata:Scincidae), also known as the brown forest skink, under extreme temperature stress at 4 °C, 8 °C, 34 °C, and 38 °C for 24 h, compared to the control group at 25 °C. In southern China, for lizards, 4 °C is close to lethal, and 8 °C induces hibernation, while 34/38 °C is considered hot and environmentally realistic. Results showed that at a low temperature of 4 °C for 24 h, transcript levels of ATP8, ND1, ND4, COI, and ND4L significantly decreased, to values of 0.52 ± 0.08, 0.65 ± 0.04, 0.68 ± 0.10, 0.28 ± 0.02, and 0.35 ± 0.02, respectively, compared with controls. By contrast, transcript levels of COIII exhibited a significant increase, with a mean value of 1.86 ± 0.21. However, exposure to 8 °C for 24 h did not lead to an increase in transcript levels. Indeed, transcript levels of ATP6, ATP8, ND1, ND3, and ND4 were significantly downregulated, to 0.48 ± 0.11, 0.68 ± 0.07, 0.41 ± 0.08, 0.54 ± 0.10, and 0.52 ± 0.07, respectively, as compared with controls. Exposure to a hot environment of 34 °C for 24 h led to an increase in transcript levels of COI, COII, COIII, ND3, ND5, CYTB, and ATP6, with values that were 3.3 ± 0.24, 2.0 ± 0.2, 2.70 ± 1.06, 1.57 ± 0,08, 1.47 ± 0.13, 1.39 ± 0.56, and 1.86 ± 0.12, respectively, over controls. By contrast, ND4L exhibited a significant decrease (to 0.31 ± 0.01) compared with controls. When exposed to 38 °C, the transcript levels of the 13 PCGs significantly increased, ranging from a 2.04 ± 0.23 increase in ND1 to a 6.30 ± 0.96 rise in ND6. Under two different levels of cold and heat stress, the expression patterns of mitochondrial genes in S. incognitus vary, possibly associated with different strategies employed by this species in response to low and high temperatures, allowing for rapid compensatory adjustments in mitochondrial electron transport chain proteins in response to temperature changes. Low temperatures pose a significant threat to plant growth and development. Studies have shown that aquaporins (AQPs), as the main functional proteins on the cell membrane regulating water ingress and egress, play a vital role in maintaining dynamic water balance when plants face cold stress. Catalpa bungei, an important timber and ornamental tree species, has its cultivation range significantly limited by its poor cold tolerance. However, no study has been found aiming to identify its aquaporin gene family. This study aims to fill this gap using two C. bungei cultivars with differing cold tolerance as experimental material: "Qiuza 1", which is less cold-tolerant, and "Qiuza 2", which is more cold-tolerant. The plants were subjected to low-temperature stress at 4 °C for 24 h. Using high-throughput molecular sequencing technology, a transcriptome sequencing of the leaves was performed at 0, 6, 12, and 18 h of cold stress. Fifteen candidate aquaporin genes in C. bungei (CbAQP) were identified. Phylogenetic analysis showed that the CbAQP gene family is divided into five subfamilies: 5 PIPs, 4 TIPs, 3 NIPs, 2 SIPs, and 1 XIP. By analyzing AQPs related to cold stress in other plants and the expression patterns of CbAQP genes, 12 CbAQP genes related to cold stress were identified. The genes that responded positively include CbPIP2;5, CbPIP1;2, CbTIP4;1, and CbNIP2;1. The results provide a foundation for further analysis of the biological functions of candidate CbAQP genes related to cold tolerance and offer theoretical support for improving seedling quality, cold-resistant genetic breeding, and expanding its distribution range. Abiotic stressors are the main factors limiting the expansion of territories occupied by grape plantations. Industrial viticulture is concentrated in the south of Russia and is limited by climatic factors that do not allow large-scale production in other regions of the country.The present review considers the molecular mechanisms of resistance to low-temperature stress and discusses the role of the main genes determining the ability of plants to survive and acclimatize during a critical temperature drop.One of the most studied ways of responding to cold stress is the interaction of genes in the ICE-CBF-COR cascade, however, a more accurate understanding of the genes responsible for resistance to abiotic environments specifically in grapes requires additional studies. A series of studies of functions of transcription factors and related genes of response to low-temperature stress in various species (Arabidopsis, tea, orange, blueberry, and grape) have identified four main regulons: 1) CBF/DREB, 2) NAC/ZF-HD, 3) AREB/ABF, and 4) MYC/MYB. Studies have demonstrated the function of the HOS1 gene, which negatively regulates the work of ICE1 (a key resistance factor). The review considers candidate genes in various species of annual plants: ICE1, HOS1, SIZ1, MPK3, MPK6, in families of genes: CBF, COR, RD 29A, LTI78, ERD, LEA; DREB1, ADREB1B; WRKY10, and in perennial crops: ICE1, CBF1, HSP70, SUS1, GST, DHN1, BMY5, BHLH102, GR-RBP3, ICE1, GOLS1, GOLS3; CBF; COR27, RD29B, NCED1, ERF105, ZAT10, SAP15, WRKY3, and LEA.Until recently, interspecific hybridization was the leading method for obtaining cold-resistant grape varieties. The main donor of resistance is V. аmurensis Rupr. Recently, the research focused on the genetic basis of grape resistance to low temperatures is actively developing. For instance, a comparative analysis of the transcriptomes of two species contrasting in this trait, i.e. V. amurensis, resistant to low temperatures, and V. vinifera L. with low cold resistance, made it possible to identify three additional candidate genes with an increased expression in response to exposure to low temperatures, namely CBF3, ERF105 and ZAT10. At the same time, the practical application of modern accelerated breeding methods requires the identification of all additional key genes responsible for resistance to low-temperature stress. The components from the cascade of sequentially expressing ICE–CBF–COR genes (ICE1, ICE2, CBF1, CBF2, CBF3, and HOS1) have been selected as candidate genes.

Abstract The Golden2-like (GLK) transcription factor family is a significant group of transcription factors in plantae. The currently available studies have shown that GLK transcription factors have been studied mainly in chloroplast growth and development, with fewer studies in abiotic stress regulation. In this study, all tea plant GLK transcription factors were identified for the first time in tea plants, and genome-wide identification, phylogenetic analysis, and thematic characterization were performed to identify 66 GLK transcription factors in tea plants. These genes are categorized into seven groups, and an amino acid sequence comparison analysis is performed. This study revealed that the structure of GLK genes in tea plants is highly conserved and that these genes are distributed across 14 chromosomes. Collinearity analysis revealed 17 pairs of genes with fragment duplications and one pair of genes with tandem duplications, and the analysis of Ka/Ks ratios indicated that most of the genes underwent negative purifying selection. Analysis of promoter cis-elements revealed that the promoters of tea plant GLK genes contain a large number of cis-acting elements related to phytohormones and stress tolerance. In addition, a large number of genes contain LTR elements, suggesting that tea plant GLK genes are involved in low-temperature stress. qRT‒PCR analysis revealed that the expression of CsGLK17, CsGLK38, CsGLK54, CsGLK11 and CsGLK60 significantly increased and that the expression of CsGLK7 and CsGLK13 decreased in response to low-temperature induction. Taken together, the results of the transcription profile analysis suggested that CsGLK54 may play an important regulatory role under low-temperature stress. The subcellular localization of CsGLK54 was in the nucleus. Furthermore, CsGLK54 positively regulated the transcription levels of the NbPOD and NbSOD genes under low-temperature stress, which led to an increase in POD and SOD enzyme activities and a decrease in MDA content. Abstract Background Quinoa (Chenopodium quinoa Willd.) is valued for its nutritional richness. However, pre-harvest sprouting poses a significant threat to yield and grain quality. This study aims to enhance our understanding of pre-harvest sprouting mitigation strategies, specifically through delayed sowing and avoiding rainy seasons during quinoa maturation. The overarching goal is to identify cold-resistant varieties and unravel the molecular mechanisms behind the low-temperature response of quinoa. We employed bioinformatics and genomics tools for a comprehensive genome-wide analysis of polyamines (PAs) and ethylene synthesis gene families in quinoa under low-temperature stress. Results This involved the identification of 37 PA biosynthesis and 30 PA catabolism genes, alongside 227 ethylene synthesis. Structural and phylogenetic analyses showcased conserved patterns, and subcellular localization predictions indicated diverse cellular distributions. The results indicate that the PA metabolism of quinoa is closely linked to ethylene synthesis, with multiple genes showing an upregulation in response to cold stress. However, differential expression within gene families suggests a nuanced regulatory network. Conclusions Overall, this study contributes valuable insights for the functional characterization of the PA metabolism and ethylene synthesis of quinoa, which emphasize their roles in plant low-temperature tolerance and providing a foundation for future research in this domain.

Low temperatures are one of the major abiotic stresses that affect alfalfa's development and yield. Enhancing frost resistance through resistance-related genes is one of the most effective ways to address this issue in alfalfa. Therefore, exploring cold-resistant gene resources and the cultivation of cold-resistant alfalfa cultivars is inevitable in order to achieve high yield and quality. In this study, we conducted transcriptome profiling of roots obtained from two alfalfa genotypes, i.e., Qingda No.1 for freeze tolerance and Gannong No.9 for freeze sensitivity. We observed that Qingda No.1 had more lateral roots and a more developed root system after overwintering, while Gannong No.9 had fewer lateral roots and an underdeveloped root system. After overwintering, Qingda No.1 exhibited higher superoxide dismutase (SOD) activity compared to Gannong No.9, while Gannong No.9 showed higher perosuperoxide dismutasexidase (POD) activity than Qingda No.1. We identified 25,935 differentially expressed genes, with 12 979 and 12 956 differential genes found in the freeze-tolerant variety Qingda No.1 group and the freeze-sensitive Gannong No.9 group, respectively. The enrichment of the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways also differed between the two groups. We also discovered several gene family members, and the most frequent transcription factors were bHLH, B3, NAC, WRKY, and MYB_related. These findings provide comprehensive information to further understand the molecular mechanisms of adaptation to freezing stress in alfalfa and offer potential functional candidate genes for adaptation to abiotic stress.

Introduction: MYC transcription factors are the basic regulators of the jasmonic acid signaling pathway and play important roles in plant growth and development and the response to adverse stress. In recent years, severe winter freezing and late spring frost in the main planting area of walnut in Xinjiang have affected the growth and development of walnut, which has become a prominent problem restricting walnut production. Xinjiang wild walnut is the only remaining wild species of walnuts in China, which contains a lot of genes with excellent traits, and is important for the cultivation and breeding. Methods: In this paper, the physicochemical properties and bioinformatics of MYC transcription factor members in walnut were analyzed, and the nine MYC were screened from the transcriptome data under low temperature stress. At last, we study the subcellular localizations and the expression patterns of the nine MYC members in Xinjiang wild walnut. Results: The results revealed that 30 MYC members were identified from published walnut whole-genome data, and their evolutionary relationships with Arabidopsis and poplar were divided into six groups according to clustering analysis, among which JrMYC22 and JrMYC23 had high homology with PtrMYC2b, which is induced by jasmonic acid in response to low- temperature stress. Walnut MYC members are unevenly distributed on 12 chromosomes. The prediction of promoter cis-acting elements of walnut MYC transcription factor family members revealed that cis-acting elements related to jasmonic acid and low temperature stress were the ones with the greatest number of members, with 12. In addition, all nine MYC family members in Xinjiang wild walnut plants responding to low-temperature stress exhibited strong fluorescence responses in the nucleus. The expression levels of these members in response to low-temperature stress revealed that JrMYC28, JrMYC31, JrMYC33, JrMYC34, and JrMYC35 were highly expressed, and it was hypothesized that JrMYC28, JrMYC31, JrMYC33, JrMYC34, and JrMYC35 might play a key role in the response to low temperature stress. Low temperature and cold damage seriously hinder the growth, development, and morphogenesis of cotton seedlings. However, the response mechanism of cotton seedlings under cold stress still lacks research. In this study, transcriptome sequencing, gas exchange parameters, and rapid chlorophyll fluorescence parameters were analyzed in leaves of cold-tolerant upland cotton variety "ZM36" under different temperature stress [25°C (T25, CK), 15°C (T15), 10°C (T10), and 4°C (T4)]. The results showed that the net photosynthetic rate (Pn), stomatal conductance (Gs), transpiration rate (Tr), PSII potential maximum photochemical efficiency (Fv/Fm), and performance index (PIabs) of cotton leaves significantly decreased, and the intercellular CO2 concentration (Ci) and Fo/Fm significantly increased under cold stress. The transcriptome sequencing analysis showed that a total of 13,183 DEGs were involved in the response of cotton seedlings at each temperature point (T25, T15, T10, and T4), mainly involving five metabolic pathways--the phosphatidylinositol signaling system, photosynthesis, photosynthesis antenna protein, carbon fixation in photosynthetic organisms, and carotenoid synthesis. The 1,119 transcription factors were discovered among all the DEGs. These transcription factors involve 59 families, of which 15.8% of genes in the NAC family are upregulated. Through network regulatory analysis, the five candidate genes GhUVR8 (GH_A05G3668), GhPLATZ (GH_A09G2161), GhFAD4-1 (GH_A01G0758), GhNFYAl (GH_A02G1336), and GhFAD4-2 (GH_D01G0766) were identified in response to cold stress. Furthermore, suppressing the expression level of GhPLATZ by virus-induced gene silencing led to the reduction of low temperature resistance, implying GhPLATZ as a positive regulator of low temperature tolerance. The findings of the study revealed a piece of the complex response mechanism of the cold-tolerant variety "ZM36" to different cold stresses and excavated key candidate genes for low temperature response, which provided support for accelerating the selection and breeding of cotton varieties with low temperature tolerance. Dendrobium officinale is a valuable traditional Chinese herbal plant that is both medicinal and edible. However, the yield of wild Dendrobium officinale is limited. Adverse stress affects the growth, development, and yield of plants, among which low temperature is the primary limiting factor for introducing Dendrobium officinale to high-latitude areas and expanding the planting area. Therefore, this study aims to explore the variations in growth ability, cold resistance, and contents of bioactive compounds among different Dendrobium officinale strains. Four strains of Dendrobium officinale were selected as experimental materials and were subjected to low-temperature stress (4 °C). The agronomic traits, physiological indices, as well as the expressions of cold resistance-related genes (HSP70, DcPP2C5, DoCDPK1, and DoCDPK6) in the roots and leaves of Dendrobium officinale, were determined. The contents of bioactive compounds, including polysaccharides, flavonoids, and phenols were also measured. Compared with the other strains, Xianju had the highest seed germination and transplantation-related survival rates. Under low-temperature stress, Xianju exhibited the strongest cold resistance ability, as revealed by the changes in water contents, chlorophyll levels, electrical conductivities, enzyme activities, and expressions of the cold resistance-related genes. Additionally, the polysaccharide content of Xianju increased the most, while the stem flavonoid and leaf phenol contents were elevated in all four strains under cold treatment. Therefore, selecting excellent performing strains is expected to expand the planting area, improve the yield, and increase the economic benefits of Dendrobium officinale in high latitude areas with lower temperatures. Simple Summary: Global climate change is a growing threat to the lives and ranges of reptile species. The present study examines three lizard species (Calotes versicolor, Ateuchosaurus chinensis, and Hemidactylus bowringii) that share a sympatric distribution in the Guangzhou region of Guangdong, China. Mitochondria act as energy-producing centers, and the present study analyses the effects of low temperatures in the context of warm winters on lizards, by examining their responses to low-temperature stress at the level of mitochondrial gene expression. This present study analyses the effects of low temperatures in the context of warm winters on the mitochondrial gene expression of lizards and reveals the implications of the distribution within it. Ecological factors related to climate extremes have a significant influence on the adaptability of organisms, especially for ectotherms such as reptiles that are sensitive to temperature change. Climate extremes can seriously affect the survival and internal physiology of lizards, sometimes even resulting in the loss of local populations or even complete extinction. Indeed, studies have shown that the expression levels of the nuclear genes and mitochondrial genomes of reptiles change under low-temperature stress. At present, the temperature adaptability of reptiles has rarely been studied at the mitochondrial genome level. In the present study, the mitochondrial genomes of three species of lizards, Calotes versicolor, Ateuchosaurus chinensis, and Hemidactylus bowringii, which live in regions of sympatry, were sequenced. We used RT-qPCR to explore the level of mitochondrial gene expression under low-temperature stress, as compared to a control temperature. Among the 13 protein-coding genes (PCGs), the steady-state transcript levels of ND4L, ND1, ATP6, and COII were reduced to levels of 0.61 ± 0.06, 0.50 ± 0.08, 0.44 ± 0.16, and 0.41 ± 0.09 in C. versicolor, respectively, compared with controls. The transcript levels of the ND3 and ND6 genes fell to levels of just 0.72 ± 0.05 and 0.67 ± 0.05 in H. bowringii, compared with controls. However, the transcript levels of ND3, ND5, ND6, ATP6, ATP8, Cytb, and COIII genes increased to 1.97 ± 0.15, 2.94 ± 0.43, 1.66 ± 0.07, 1.59 ± 0.17, 1.46 ± 0.04, 1.70 ± 0.16, and 1.83 ± 0.07 in A. chinensis. Therefore, the differences in mitochondrial gene expression may be internally related to the adaptative strategy of the three species under low-temperature stress, indicating that low-temperature environments have a greater impact on A. chinensis, with a small distribution area. In extreme environments, the regulatory trend of mitochondrial gene expression in reptiles is associated with their ability to adapt to extreme climates, which means differential mitochondrial genome expression can be used to explore the response of different lizards in the same region to low temperatures. Low temperature is one of the main abiotic stresses that affects plant growth, causing serious damage or even death to plants. The differential expression of the TaEXPA19 gene in the above and underground parts of winter wheat and the implications for cold resistance remain unclear. In this study, the TaEXPA19 gene was cloned and analysed for expression in winter wheat, and transgenic Arabidopsis thaliana was constructed to investigate the effect of the TaEXPA19 gene in response to low‐temperature stress on plant growth. The TaEXPA19‐A and TaEXPA19‐D genes have different response patterns in the above and underground parts of transgenic A. thaliana. When plants were subjected to low‐temperature stress, the leaves were quickly upregulated and the roots were downregulated, and then upregulated to respond to low‐temperature stress to promote the growth of leaf length and leaf width petiole length. The results indicated that TaEXPA19 genes could improve low‐temperature tolerance in plants. The results of this study laid a foundation for the study of the cold resistance of winter wheat. The NAC (NAM, ATAF1/2, CUC2) family of transcription factors (TFs) is a vital transcription factor family of plants. It controls multiple parts of plant development, tissue formation, and abiotic stress response. We cloned the FvNAC29 gene from Fragaria vesca (a diploid strawberry) for this research. There is a conserved NAM structural domain in the FvNAC29 protein. The highest homology between FvNAC29 and PaNAC1 was found by phylogenetic tree analysis. Subcellular localization revealed that FvNAC29 is localized onto the nucleus. Compared to other tissues, the expression level of FvNAC29 was higher in young leaves and roots. In addition, Arabidopsis plants overexpressing FvNAC29 had higher cold and high-salinity tolerance than the wild type (WT) and unloaded line with empty vector (UL). The proline and chlorophyll contents of transgenic Arabidopsis plants, along with the activities of the antioxidant enzymes like catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD) under 200 mM NaCl treatment or −8 °C treatment, were higher than those activities of the control. Meanwhile, malondialdehyde (MDA) and the reactive oxygen species (ROS) content were higher in the WT and UL lines. FvNAC29 improves transgenic plant resistance to cold and salt stress by regulating the expression levels of AtRD29a, AtCCA1, AtP5CS1, and AtSnRK2.4. It also improves the potential to tolerate cold stress by positively regulating the expression levels of AtCBF1, AtCBF4, AtCOR15a, and AtCOR47. These findings suggest that FvNAC29 may be related to the processes and the molecular mechanisms of F. vesca response to high-salinity stress and LT stress, providing a comprehensive understanding of the NAC TFs. The growth and development of apricot flower organs are severely impacted by spring frosts. To better understand this process, apricot flowers were exposed to temperatures ranging from 0 °C to −8 °C, including a control at 18 °C, in artificial incubators to mimic diverse low-temperature environments. We aimed to examine their physiological reactions to cold stress, with an emphasis on changes in phenotype, membrane stability, osmotic substance levels, and antioxidant enzyme performance. Results reveal that cold stress induces significant browning and cellular damage, with a sharp increase in browning rate and membrane permeability below −5 °C. Soluble sugars and proteins initially rise as osmoprotectants, but their content decreases at lower temperatures. Proline content consistently increases, suggesting a protective role. Antioxidant enzyme activities, including catalase (CAT), peroxidase (POD), superoxide dismutase (SOD), and ascorbate peroxidase (APX), exhibit a complex pattern, with initial increases followed by declines at more severe cold conditions. Correlation and principal component analyses highlight the interplay between these responses, indicating a multifaceted adaptation strategy. The occurrence of early spring and late frost damage brought severe challenges to the production of walnuts in China, seriously affecting their growth, development, yield and revenue. In order to clarify the harmful characteristics and influencing mechanisms of late frost on walnuts in China, the influencing factors of the degree of damage caused by late frost on walnuts, as well as the changes in physiological and biochemical indicators and molecular mechanisms in response to low temperature stress were reviewed. It was found that research on physiological changes focused on the three aspects including cell membrane system (relative conductivity), antioxidant system (malondialdehyde, superoxide dismutase, peroxidase and catalase) and osmoregulation substances (soluble sugars, soluble protein and proline), in addition, the expression levels of genes such as JrPAL, JrJAZ, JrGRAS, JrICE1, JrCBF/JrDREB1, JrCOR, JrWRKY, JrDHN, JrGST, and JrPUB were induced by low temperature stress. And prospects were put for future research directions, which provided scientific basis for further exploring the response mechanism of walnuts to late frost damage and adopting effective production measures. Suboptimal temperatures at sowing and emergence affect the early development of maize, with potentially irreversible effects later in the growing season. We studied recovery from cold stress of an inbred line (B73) and 13 Doubled Haploid lines derived from a European flint maize landrace. After a cold treatment (20–12 °C, day–night) from sowing to seedling establishment, seedlings were transplanted and grown in the greenhouse until the V8 stage (eight leaves fully developed), when we measured agronomically relevant plant traits and spectral indices of mature leaves. Survival rates of transplanted seedlings after cold treatment ranged from 10% to 100%. After a strong delay in early development due to cold, the surviving plants were able to compensate for this delay at later stages of recovery. They reached the V8 stage after only five more growing degree days than plants grown under the control treatment (25–18 °C, day–night). Plants from the most cold-tolerant genotypes (PE0401 and PE0100) accumulated more root and shoot biomass at the end of the recovery phase compared with the same genotypes exposed to the control treatment. The genotypes with the most plastic leaf morphological traits (PE0161 and PE0072) had little reduction in leaf biomass at the end of the recovery phase in comparison with less responsive genotypes such as PE0171. We conclude that genotypes that survived cold treatment with minimal cold damage of seedling leaves can be candidates for further cold recovery studies and breeding. Crop growth models, such as the WOrld FOod STudies (WOFOST) model, mimic the mechanistic processes involved in crop development, growth, and yield production. The accuracy of simulation is decreased in unfavorable low‐temperature settings because these models do not accurately represent crop response processes in low‐temperature stress. Enhancing the WOFOST crop growth model's accuracy in simulating crops' responses to cold temperatures is the aim of this work. Given its vulnerability to low temperatures, the inquiry uses winter wheat in Henan Province as a focal point. It integrates the WHEATGROW wheat phenology model with the Frost model of Lethal Temperature 50 (FROSTOL) inside the framework of the crop growth model. This link aims to improve simulation accuracy and supplement the model's mechanisms, particularly when it comes to the impact of low temperatures on crop development. The study uses Long Short‐Term Memory networks to build a yield model that integrates remote sensing data with information from simulated crop models. Under low temperatures, the leaf area index, total above ground biomass, and total weight of storage organs of the model WWF—which combines FROSTOL and WHEATGROW with WOFOST—show a considerable decline. It was discovered that there is a greater improvement in simulation accuracy of the linked model WWF relative to the WOFOST model in frost years than in normal years, based on a comparison analysis between typical frost years and normal years. To be more precise, the improvement is 8.03% in frost years and 1.98% in regular years. In the Non-chernozem zone of Russia, the recurrent spring cold up to 1-2 C can cause damage and death of thermophilic amaranth seedlings. Pre-sowing treatment of seeds with growth regulators is an inexpensive and effective method to reduce the negative effect of hypothermia on seed germination. The aim of the research was to study the effect of low-temperature stress on etiolated and de-etiolated seedlings of amaranth cv. Valentina ( A. tricolor L.) grown from seeds treated with growth stimulants. Seeds were pretreated with aqueous solutions of Albit (1 g/L), hydrogen peroxide (H2O2) - 5 mM and succinic acid (ScA) - 500 mg/L. Seeds were germinated in peat pots at 23 2 C (T23) for 7 days. On the 7th day, peat pots with seedlings grown in the light and in the dark were moved to thermostat at 2.0 0.5 C (T2) for 8 hours. Determination of the amount of amaranthine, chlorophylls and carotenoids were carried out according to generally accepted methods. Pretreatment of seeds with the growth regulators Albit, H2O2, and ScA increased hypocotyl length, root length, and biomass of light and etiolated seedlings. Low positive temperatures increased the content of amaranthine and carotenoids but reduced the content of chlorophylls. It was shown that all used growth regulators - H2O2, Albit and ScA trigger or at least maintain the system of antioxidant protection in light and etiolated seedlings of amaranth cv. Valentina under low positive temperatures.

Extreme low-temperature incidents have become more frequent and severe as climate change intensifies. In Huang-Huai-Hai wheat growing area of China, the late spring coldness occurring at the jointing-booting stage (the anther interval stage) has resulted in significant yield losses of winter wheat. This study attempts to develop an economical, feasible, and efficient cultivation technique for improving the low-temperature (LT) resistance of wheat by exploring the effects of twice-split phosphorus application (TSPA) on wheat antioxidant characteristics and carbon and nitrogen metabolism physiology under LT treatment at the anther interval stage using Yannong 19 as the experimental material. The treatments consisted of traditional phosphorus application and TSPA, followed by a –4°C LT treatment and natural temperature (NT) control at the anther interval stage. Our analyses showed that, compared with the traditional application, the TSPA increased the net photosynthetic rate (Pn), stomatal conductance (Gs), and transpiration rate (Tr) of leaves and reduced the intercellular carbon dioxide concentration (Ci). The activity of carbon and nitrogen metabolism enzymes in the young wheat spikes was also increased by the TSPA, which promoted the accumulation of soluble sugar (SS), sucrose (SUC), soluble protein (SP), and proline (Pro) in young wheat spike and reduced the toxicity of malondialdehyde (MDA). Due to the improved organic nutrition for reproductive development, the young wheat spikes exhibited enhanced LT resistance, which reduced the sterile spikelet number (SSN) per spike by 11.8% and increased the spikelet setting rate (SSR) and final yield by 6.0 and 8.4%, respectively, compared to the traditional application. The positive effects of split phosphorus application became more pronounced when the LT treatment was prolonged.

Abstract Background The RNA recognition motif (RRM) is primarily engaged in the processing of mRNA and rRNA following gene transcription as well as the regulation of RNA transport; it is critical in preserving RNA stability. Results In this study, we identified 102 members of the RRM1 gene family in Brassica rapa, which were dispersed across 10 chromosomes with the ninth chromosome being the most extensively distributed. The RRM1 gene family members of Brassica rapa and Arabidopsis thaliana were grouped into 14 subclades (I–XIV) using phylogenetic analysis. Moreover, the results of transcriptome analysis and RT-qPCR indicated that the expression of Brapa05T000840 was upregulated in the cultivars ‘Longyou 7’ and ‘Longyou 99’ following exposure to cold stress at a temperature of 4 °C for 24 h. The levels of expression in the leaves and growth cones of the ‘Longyou 7’ variety were found to be significantly higher than those observed in the ‘Longyou 99’ variety under conditions of low temperature and NaCl stress. It illustrates the involvement of the RRM1 gene in the physiological response to both low temperature and salt stress. In addition, it was observed that the survival rate of transgenic BrRBP (Brapa05T000840) Arabidopsis thaliana plants was notably higher compared to that of wild-type plants when subjected to varying durations of low temperature treatment. Furthermore, the expression of the BrRBP gene in transgenic plants exhibited an upward trend as the duration of low temperature treatment increased, reaching its peak at 24 h. The in-vivo enzymatic activity of reactive oxygen species-scavenging enzymes were found to be significantly elevated in comparison to wild-type plants, suggesting that the BrRBP gene may enhance the cold tolerance of Arabidopsis thaliana. Conclusions This study offers a significant foundation for comprehending the regulation mechanism of the RRM1 gene family in winter Brassica rapa subjected to cold stress, as well as for finding key genes associated with cold resistance.

Variation in fish temperature is determined by changes in the external aquatic environment, and temperature serves as a crucial regulatory factor for the behavior, physiology, and molecular processes of fish. A rapid decline in water temperature can elicit a strong stress response in fish, potentially inhibiting growth and even leading to mortality. Low temperature stress significantly impacts various life processes of organisms, including growth and development, energy metabolism, and reproductive development. In recent years, there has been increasing research on the effects of low-temperature stress on organisms, with a greater focus at the molecular level. Low temperatures induce a decrease in cellular activity and even growth arrest, resulting in apoptosis. Apoptosis is a form of programmed cell death ubiquitous in the biological world, and it plays a vital role in maintaining the normal physiological function of an organism. Once apoptosis occurs, it is irreversible and cannot be stopped. Temperature regulation is crucial for fish, and current knowledge of the role of leptin in fish primarily focuses on the regulation of feeding, lipid energy metabolism, and reproduction, with limited reports on its role in temperature regulation.Leptin, a protein hormone produced by adipocytes, has long been recognized as a product of the obese gene, which regulates organismal metabolism, neuroendocrine function, and other physiological processes. In animals, it exerts diverse physiological functions related to cell growth, proliferation, apoptosis, and metabolism. The homology of the leptin gene in fish is relatively low compared to that in mammals, and multiple leptin proteins encoded by multiple leptin genes can be produced due to genomic duplication events. Regarding the role of leptin in temperature regulation, more research has been conducted on mammals, mainly related to the regulation of energy metabolism and the influence on temperature adaptation evolution. There has been limited research on the regulatory mechanism of leptin in fish during cold stress.Dissostichus mawsoni belongs to the group of Antarctic vertebrates and has lived in the cold and isolated environment of the Southern Ocean at a temperature of –1.9 ℃ for ~30 million years. It has adapted to the extremely low temperatures of the surrounding Antarctic waters and is an excellent material and living fossil for studying temperature adaptation mechanisms in extreme environments. Compared with other fish species at the same depth, it has a large amount of lipid deposition in its subcutaneous and muscle tissues, mainly triglycerides (TGs). Predictions based on 3D structural models suggest that the partial absence of the lepB structure in D. mawsoni leads to only three α-helices, and lepA has four α-helices and a short and twisted E-helix as well as several irregular turns, forming a hollow barrel-like structure, which differs from the protein structure of all other known leptins. Moreover, previous studies have found a close correlation between lepA and temperature evolution, leading to speculation that Antarctic fish lepB may play an essential role in low-temperature adaptation.In this study, a eukaryotic expression vector of the Antarctic toothfish lepb gene was constructed and transfected into ZFL cells to establish a model of overexpression of the D. mawsoni lepb (DMLB) gene in the ZFL cells. After conducting a cell culture temperature experiment, 10 ℃ was selected as the significant difference temperature. Following 2 weeks of cold stimulation, the DMLB experimental group cells remained viable, whereas the control group cells died. The results indicated that the overexpression of the DMLB gene provided strong resistance to low-temperature conditions. By detecting changes in cell proliferation, apoptosis, reactive oxygen species (ROS) level, and ATP content under low-temperature stress, it was discovered that DMLB maintained cell growth and proliferation, reduced ROS production, and inhibited cell apoptosis. DMLB effectively maintained ATP levels in cells under low-temperature stress, which helps maintain the mitochondrial status and reduce the effects of apoptosis and necrosis caused by low-temperature stress, thereby protecting cells from cold stress. Additionally, the results of Oil red O staining and TG detection suggested that the DMLB gene may slow down the depletion of cell lipids under low-temperature stress, perhaps by lowering lipid metabolism to preserve lipids to cope with low-temperature damage. Consequently, the DMLB gene may be functional in the cold resistance of D. mawsoni by protecting cells from damage at low temperatures, but its specific molecular regulatory mechanism needs to be further explored. In this study, 10 ℃ was used as the key temperature, providing a basis for understanding the regulation of energy homeostasis by the lepb gene in D. mawsoni under low-temperature stress. This study explored the role of the lepB gene in the adaptation of D. mawsoni to extremely low-temperature environments and provided fundamental data for further study of the evolution of similar species in the Antarctic region. Furthermore, this study enriched the basic data related to low-temperature tolerance and provided reference materials for further investigation of the low-temperature tolerance mechanism of leptin in Notothenioids, as well as laid a foundation for further study of the mechanism of leptin in temperature regulation.

Continual climate change strongly influences temperature conditions worldwide, making ectothermic animals as suitable species for studying the potential impact of climate change on global biodiversity. However, the study of how lizards distributed at different latitudes respond to climate change at the transcriptome level is still insufficient. According to the Climatic Variability Hypothesis (CVH), the range of climate fluctuations experienced by terrestrial animals throughout the year increases with latitude, so individuals at higher latitudes should exhibit greater thermal plasticity to cope with fluctuating environments. Mitochondria, as the energy center of vertebrate cells, may indicate species’ plasticity through the sensitivity of gene expression. In this study, we focused on the changes in transcript levels of liver mitochondrial protein-coding genes (PCGs) in skinks from the genus Plestiodon (P. capito and P. elegans) and the genus Scincella (S. modesta and S. reevesii) under low-temperature conditions of 8 °C, compared to the control group at 25 °C. Species within the same genus of skinks exhibit different latitudinal distribution patterns. We found that the two Plestiodon species, P. elegans and P. capito, employ a metabolic depression strategy (decreased transcript levels) to cope with low temperatures. In contrast, the two Scincella species show markedly different patterns: S. modesta exhibits significant increases in the transcript levels of six genes (metabolic compensation), while in S. reevesii, only two mitochondrial genes are downregulated (metabolic depression) compared to the control group. We also found that P. capito and S. modesta, which live at mid-to-high latitudes, exhibit stronger adaptive responses and plasticity at the mitochondrial gene level compared to P. elegans and S. reevesii, which live at lower latitudes. We suggest that this enhanced adaptability corresponds to more significant changes in a greater number of genes (plasticity genes).

Out-of-season breeding is an effective method for addressing seasonal shortages of fry in aquaculture species such as largemouth bass (LMB) for year-round production. Off-season breeding of LMB can be achieved by subjecting breeding LMB to prolonged low-temperature conditions; however, this can alter reproductive rhythms, affecting the quality of their sperm and leading to a decrease in reproductive efficiency. Therefore, it is crucial to investigate issues such as the damage to the testes and the related mechanisms caused by low-temperature stress during out-of-season breeding. In this experiment, we assessed the changes in the testes during this time in LMB by comparing reproductive rhythms, testicular histomorphology, ultrastructure, antioxidant capacity and apoptosis. We synthesized measurements of LMB from three identically treated cement ponds and fish exposed to water temperatures of 13–16 °C to assess the changes in the testes. The results showed that (1) out-of-season reproduction delayed sperm production and promoted sperm redevelopment in LMB, various hormone levels have changed over time (e.g., LH, FSH, and T). (2) The head plasma membrane of LMB spermatozoa was separated, and the middle mitochondria were swollen. (3) The expression levels of antioxidant enzymes (cat, sod, and gpx) were upregulated, and oxidative stress occurred in LMB. (4) The expression levels of apoptosis genes (e.g., bax, bcl2, and caspase3) were upregulated, and apoptosis occurred in LMB due to off-season breeding. Moreover, important genes of the mitochondrial apoptosis pathway (bid, CYT-C) were upregulated, indicating that spermatozoan apoptosis in LMB was probably achieved through the mitochondrial apoptosis pathway. These results suggest the delays, damage, and regeneration of LMB testes. Our findings provide new insights into the molecular mechanisms that trigger changes in sperm quality during out-of-season breeding in fish.

Relatively high-latitude coral reefs could be potential "refuges" for corals under climate change. One of the most important aspects limiting their availability as refuges is low-temperature stress. However, the mechanisms underlying the response of coral holobionts to low-temperature stress is unclear. In this study, we aimed to explore the underlying mechanisms by recording the maximum quantumyields of photosystemII (Fv/Fm) and transcriptome responses of Porites lutea under acute (1--2 weeks) and chronic (6--12 weeks) low-temperature stress at 20°C and 14°C. The P. lutea samples were collected from a relatively high- latitude coral reef in the South China Sea (109°00'--109°15'E and 21°00'--21°10' N). The study suggested that: (1) Under acute low-temperature stress, the Fv/Fm of Symbiodiniaceae dropped by 64%, which was significantly higher than the 49% observed under chronic stress. Low-temperature stress inhibited photosystem II (PSII) functioning, with greater inhibition under acute stress. (2) Downregulation of sugar metabolism-related genes under low-temperature stress implied that the decrease in energy was due to obstruction of PSII. (3) Under low-temperature stress, calcification-related genes were downregulated in coral hosts, possibly because of energy deprivation caused by inhibited photosynthesis, Symbiodiniaceae expulsion, and oxidative phosphorylation uncoupling in mitochondria. (4) Acute low-temperature stress induced the upregulation of genes related to the TNF signaling pathway and endoplasmic reticulum stress, promoting apoptosis and coral bleaching. However, these phenomena were not observed during chronic stress, suggesting acclimation to chronic low- temperature stress and a greater survival pressure of acute low-temperature stress on coral holobionts. In conclusion, low-temperature stress inhibits Symbiodiniaceae PSII functioning, reducing energy production and affecting calcification in coral holobionts. Low temperature is a critical environmental stress factor that restricts crop growth and geographical distribution, significantly impacting crop quality and yield. When plants are exposed to low temperatures, a series of changes occur in their external morphology and internal physiological and biochemical metabolism. This article comprehensively reviews the alterations and regulatory mechanisms of physiological and biochemical indices, such as membrane system stability, redox system, fatty acid content, photosynthesis, and osmoregulatory substances, in response to low-temperature stress in plants. Furthermore, we summarize recent research on signal transduction and regulatory pathways, phytohormones, epigenetic modifications, and other molecular mechanisms mediating the response to low temperatures in higher plants. In addition, we outline cultivation practices to improve plant cold resistance and highlight the cold-related genes used in molecular breeding. Last, we discuss future research directions, potential application prospects of plant cold resistance breeding, and recent significant breakthroughs in the research and application of cold resistance mechanisms. The effects of selenium supplementation in Tachysurus fulvidraco (T. fulvidraco) on low-temperature stress are not known. In this study, 280 healthy T. fulvidraco were divided into two groups, the G0 group (a control group) and the T0 group (a selenium treatment group on a 0.22 mg/kg diet), for a 6-week feeding time. Then, low-temperature stress (water temperature dropped from 26 to 13 °C, with a rate of 1 °C/h) was administered after that. The feeding results showed that selenium increased the percent weight gain (PWG), specific growth rate (SGR), and survival rate (SR) of T. fulvidraco and decreased the feed conversion rate (FCR), but these differences were not significant (p > 0.05). Under low temperatures, selenium still has no significant effects on antioxidant indexes such as glutathione peroxidase (GSH-Px) activity, superoxide dismutase (SOD) activity, and malondialdehyde (MDA) content in serum (p > 0.05). However, metabolomic analysis revealed that selenium caused changes in lipids and lipid-like molles, organic acids and their derivatives, and fatty acyls. Choline, linoleic acid, and glycerophospholipid metabolism pathways; d-arginine and d-ornithine metabolism; valine, leucine, and isoleucine degradation; and biosynthesis pathways, as well as pyrimidine metabolism pathways, were activated to produce these metabolites to combat against this stress. In addition, selenium increased the diversity of intestinal microbes in T. fulvidraco and decreased the relative abundance of Plesiomonas. However, the combined analysis showed the intestinal microbe changes did not affect metabolite production. In summary, selenium activated lipid, carbohydrate, and amino acid metabolism for energy substance provision, reduced the oxidation and production of other harmful substances, and increased the intestinal microbe diversity of T. fulvidraco to improve resistance to low-temperature stress. Pouring semi-flexible pavement material (PSFM) is widely used as a wearing layer material or below pavement due to its excellent resistance to deformation at high temperatures and under heavy loads. However, in cold regions, the material exhibits severe cracking issues. The primary objective of this study is to enhance the resistance of pouring semi-flexible pavements (SFPs) to low-temperature cracking in cold regions by strategically designing pavement structures that incorporate PSFM. To achieve this goal, we conducted indoor tests to determine the relaxation modulus and temperature shrinkage coefficient of PSFM and simulated a pavement structure using COMSOL finite element simulation. The impacts of different application layers and layer thicknesses on low-temperature stresses were investigated based on these findings. The research findings indicate that when PSFM is used as the wearing layer material, the low-temperature stress is 4.7% lower than that of typical materials used in the pavement-wearing layer. When used as the binder layer material, the low-temperature stress on the wearing layer material increases by 3.5%. As the thickness of the wearing layer increases, the low-temperature stress within the pavement structure decreases, but the low-temperature stress on the pavement surface increases. Therefore, it is recommended to use PSFM as the binder layer material and appropriately increase the thickness of the wearing layer to enhance the pavement's resistance to low-temperature cracking.
