The presence of sub-minimal inhibitory concentration (sub-MIC) antibiotics in our environment is widespread, and their ability to induce antibiotic resistance is inevitable. Acinetobacter baumannii, a pathogen known for its strong ability to acquire antibiotic resistance, has recently shown clinical resistance to the last-line antibiotic tigecycline. To unravel the complex mechanism of A. baumannii drug resistance, we subjected tigecycline-susceptible, -intermediate, and -mildly-resistant strains to successive increases in sub-MIC tigecycline and ultimately obtained tigecycline-resistant strains. The proteome of both key intermediate and final strains during the selection process was analyzed using nanoLC-MS/MS. Among the more than 2600 proteins detected in all strains, we found that RND efflux pump AdeABC was associated with the adaptability of A. baumannii to tigecycline under sub-MIC pressure. qRT-PCR analysis also revealed higher expression of AdeAB in strains that can quickly acquire tigecycline resistance compared with strains that displayed lower adaptability. To validate our findings, we added an efflux pump inhibitor, carbonyl cyanide m-chlorophenyl hydrazine (CCCP), to the medium and observed its ability to inhibit tigecycline resistance in A. baumannii strains with quick adaptability. The calculation method of the external fault short-circuit current of large units with clear mechanisms and practicality is of great value in engineering practice. Due to the special AC excitation structure and control mode, the external fault short-circuit characteristics of variable-speed pumped storage units (VSPSUs) are very different from those of conventional units. Therefore, this paper proposes a calculation method for external fault short-circuit current based on a time-domain dynamic circuit. According to the excitation and control characteristics of VSPSUs under external fault, the VSPSU under external fault is divided into two cases: continuous excitation and jumper action. Based on space vector analysis, the time-domain dynamic circuit models are established, respectively. On this basis, the analytical formula of the external fault short-circuit current of VSPSUs is derived by combining the transition boundary conditions of the two cases and the dynamic characteristics of the stator flux linkage. Finally, the comparison with PSCAD/EMTDC simulation results shows that the proposed VSPSUs' external fault short-circuit current calculation method has high accuracy and practical value, which can meet the needs of primary equipment selection and relay protection of variable-speed pumped storage power plants. Existing wideband beampattern synthesis methods usually require multiple beamformers, each corresponding to one frequency bin, which results in excessive resource consumption in engineering practice. To alleviate the resource consumption of wideband array beamforming, we propose a novel time-domain wideband digital beamformer that can control the beam shape over the whole bandwidth for interference mitigation by using only one set of amplitude–phase weights and integer time delay filters. To tackle the resultant optimization problem, two effective algorithms are introduced, and both can solve the problem within polynomial time. One is an alternating optimization algorithm that decomposes the original problem into multiple tractable subproblems iteratively. The other is the convex approximation algorithm that transforms the original problem into a series of second-order cone programming problems iteratively. The two algorithms exhibit their respective advantages in terms of convergence and computational complexity. Extensive numerical examples are presented to evaluate the proposed time-domain wideband beamforming strategy, highlighting that it can form a deep notch in any interested spatial–spectral region to suppress the strong interferences. Drainage networks, and the associated channel links and drainage basins, are fundamentalconcepts in earth science. Automatic extraction of drainage networks is an importantcomponent in digital terrain analysis and hydrology analysis such as distributed hydrologicalmodel, soil erosion, etc. Extensive researches have been conducted on extraction of drainagenetworks, but the problem of parallel drainage networks still exist in some flat areas by usingexisting methods. Because calculating flow direction by the cell slope is the vital point ofextracting drainage networks, proposing an effective method to get the right flow direction isnecessary to solve the problem of parallel flow lines. Hence, in this research, wepropose a new drainage networks extraction method combining Priority-Floodand D8 algorithms without filling depressions to resolve the problem of parallelflow lines. The most important advantage of Priority-Flood algorithm is ensuringthe correctness of the flow direction on macro terrain and avoiding the process ofdepression-filling to eliminate the parallel drainage networks is another significant reason.Meanwhile, advanced D8 algorithm takes suitable calculation methods of flowdirection based on different terrains to improve the accuracy of drainage networks,which ensure the water pour out from depression. We compare the results fromthe new method and the J&D algorithm of four different areas including the Alps,the Loess Plateau, the Rocky Mountains and the Qinghai-Tibet Plateau and wecan see that the new method has the obvious advantages of avoiding the parallelflow lines. To evaluate the accuracy of this new method and the J&D algorithm,we use drainage-networks-matching-difference to make quantitative descriptionof errors in the extraction of drainage networks, which is the ratio of the sum ofarea of sliver polygons to the total length of the real drainage networks. Accordingto the results of drainage-networks-matching-difference, it is shown that averagedrainage-networks-matching-difference of new method is better than it of the J&D algorithm,which means the drainage networks extracted by new method are closer to the real drainagenetworks. Chronic hepatitis E mostly occurs in organ transplant recipients and can lead to rapid liver fibrosis and cirrhosis. Previous studies found that the development of chronic hepatitis E virus (HEV) infection is linked to the type of immunosuppressant used. Animal models are crucial for the study of pathogenesis of chronic hepatitis E. We previously established a stable chronic HEV infection rabbit model using cyclosporine A (CsA), a calcineurin inhibitor (CNI)-based immunosuppressant. However, the immunosuppression strategy and timing may be optimized, and how different types of immunosuppressants affect the establishment of chronic HEV infection in this model is still unknown. Here, we showed that chronic HEV infection can be established in 100% of rabbits when CsA treatment was started at HEV challenge or even 4 weeks after. Tacrolimus or prednisolone treatment alone also contributed to chronic HEV infection, resulting in 100% and 77.8% chronicity rates, respectively, while mycophenolate mofetil (MMF) only led to a 28.6% chronicity rate. Chronic HEV infection was accompanied with a persistent activation of innate immune response evidenced by transcriptome analysis. The suppressed adaptive immune response evidenced by low expression of genes related to cytotoxicity (like perforin and FasL ) and low anti-HEV seroconversion rates may play important roles in causing chronic HEV infection. By analyzing HEV antigen concentrations with different infection outcomes, we also found that HEV antigen levels could indicate chronic HEV infection development. This study optimized the immunosuppression strategies for establishing chronic HEV infection in rabbits and highlighted the potential association between the development of chronic HEV infection and immunosuppressants.IMPORTANCEOrgan transplant recipients are at high risk of chronic hepatitis E and generally receive a CNI-based immunosuppression regimen containing CNI (tacrolimus or CsA), MMF, and/or corticosteroids. Previously, we established stable chronic HEV infection in a rabbit model by using CsA before HEV challenge. In this study, we further optimized the immunosuppression strategies for establishing chronic HEV infection in rabbits. Chronic HEV infection can also be established when CsA treatment was started at the same time or even 4 weeks after HEV challenge, clearly indicating the risk of progression to chronic infection under these circumstances and the necessity of HEV screening for both the recipient and the donor preoperatively. CsA, tacrolimus, or prednisolone instead of MMF significantly contributed to chronic HEV infection. HEV antigen in acute infection phase indicates the development of chronic infection.
