Many regulatory genes that affect cellular development in Streptomyces, such as the canonical bld genes, have already been identified. However, in this study, we identified sven_5003 in Streptomyces venezuelae as a major new developmental regulatory gene, the deletion of which leads to a bald phenotype, typical of bld mutants, under multiple growth conditions. Our data indicated that disruption of sven_5003 also has a differential impact on the production of the two antibiotics jadomycin and chloramphenicol. Enhanced production of jadomycin but reduced production of chloramphenicol were detected in our sven_5003 mutant strain (S. venezuelae D5003). RNA-Seq analysis indicated that SVEN_5003 impacts expression of hundreds of genes, including genes involved in development, primary and secondary metabolism, and genes of unknown function, a finding confirmed by real-time PCR analysis. Transcriptional analysis indicated that sven_5003 is an auto-regulatory gene, repressing its own expression. Despite the evidence indicating that SVEN_5003 is a regulatory factor, a putative DNA-binding domain was not predicted from its primary amino acid sequence, implying an unknown regulatory mechanism by SVEN_5003. BACKGROUND: It is recommended to use non-invasive positive pressure ventilation in elderly patients with heart failure combined with obstructive sleep apnea syndrome (OSAS). OBJECTIVE: To study the therapeutic effect of non-invasive positive pressure ventilation on elderly patients with heart failure complicated with OSAS. METHODS: Using the random number table method, 119 elderly patients with heart failure complicated with OSAS who were admitted to our hospital from April 2020 to April 2021 were divided into the observation (60 cases) and control (59 cases) groups. On the basis of conventional drug treatment, patients in the observation group were treated with non-invasive positive pressure ventilation, and patients in the control group were treated with low-flow oxygen inhalation. The sleep quality, hemoglobin, red blood cells, erythropoietin, pro-brain natriureticpeptide (pro-BNP) levels and blood pressure were compared. RESULTS: After treatment, levels of hemoglobin, erythrocytes, erythropoietin, pro-BNP, blood pressure and sleep apnea-hypopnea index in the observation group were lower before treatment and in the control group in the same period. The oxygen saturation was higher before treatment and the control group in the same period (P < 0.05). The overall satisfaction with sleep quality in the observation group was higher (P < 0.05). CONCLUSION: Non-invasive positive pressure ventilation can improve blood oxygen saturation and sleep quality in elderly patients with heart failure complicated with OSAS, and reduce pro-BNP level. This study aimed to explore and develop data mining models for adult age estimation based on CT reconstruction images from the sternum. Maximum intensity projection (MIP) images of chest CT were retrospectively collected from a modern Chinese population, and data from 2700 patients (1349 males and 1351 females) aged 20 to 70 years were obtained. A staging technique within four indicators was applied. Several data mining models were established, and mean absolute error (MAE) was the primary comparison parameter. The intraobserver and interobserver agreement levels were good. Within internal validation, the optimal data mining model obtained the lowest MAE of 9.08 in males and 10.41 in females. For the external validation (N = 200), MAEs were 7.09 in males and 7.15 in females. In conclusion, the accuracy of our model for adult age estimation was among similar studies. Lyman-alpha (Ly α) emission from galaxies is currently our most promising probe for constraining when and how reionization began, and thus when the first galaxies formed. At z > 7, the majority of galaxies detected with Ly α are in candidate overdensities. Here, we quantify the probability of these galaxies residing in large ionized bubbles. We create (1.6 Gpc)3 intergalactic medium (IGM) simulations: sufficient volume to robustly measure bubble size distributions around UV-bright galaxies and rare overdensities. We find |${M_{\small UV}}\lesssim -16$| galaxies and overdensities are ≳10–1000 × more likely to trace ionized bubbles compared to randomly selected positions. The brightest galaxies and strongest overdensities have bubble size distributions with highest characteristic size and least scatter. We compare two models: gradual reionization driven by numerous UV-faint galaxies versus rapid reionization by rarer brighter galaxies, producing larger bubbles at fixed neutral fraction. We demonstrate that recently observed z ∼ 7 overdensities are highly likely to trace large ionized bubbles, corroborated by their high Ly α detection rates. However, Ly α detections at z ≈ 8.7 in EGS and z  = 10.6 in GN-z11 are unlikely to trace large bubbles in our fiducial model – 11 and 7 per cent probability of >1 proper Mpc bubbles, respectively. Ly α detections at such high redshifts could be explained by: a less neutral IGM than previously expected; larger ionized regions at fixed neutral fraction; or if intrinsic Ly α flux is unusually strong in these galaxies. Disturbances in the circadian rhythm have been reported in patients following traumatic brain injury (TBI). However, the rhythmic expression of circadian genes in peripheral blood leukocytes (PBL) following TBI has not yet been studied. The messenger ribonucleic acid (mRNA) expression of period 1 (Per1), Per2, Per3, cryptochrome 1 (Cry1), Cry2, brain and muscle aryl hydrocarbon receptor nuclear translocator-like 1 (Bmal1), and circadian locomotor output cycles kaput (Clock) was quantified in PBLs from sham-operated rats and rats with acute subdural hematoma (ASDH) over a 48-h period. The rectal temperature of the animals was measured every 4 h over 2 days. The mesor, rhythm, amplitude, and acrophase were estimated using cosinor analysis. Cosinor analysis revealed that Per2, Cry1, and Bmal1 mRNAs were rhythmically expressed in the PBLs of sham-operated rats. In contrast, fluctuations in rhythmic expression were not observed following ASDH. The rectal temperature of sham-operated rats also exhibited rhythmicity. ASDH rats had a disrupted rectal temperature rhythm, a diminished amplitude, and an acrophase shift. TBI with ASDH results in dysregulated expression of some circadian genes and changes in body temperature rhythm. Further research is required to understand the pathophysiology of altered circadian networks following TBI. Equus Linnaeus animals, as large non-ruminant herbivores, harbor a diverse array of microorganisms in their gastrointestinal tract, and the microbial composition varies across different segments of the gut. These microorganisms play a crucial role in the efficient utilization of feed, especially for nutrients that cannot be readily degraded by endogenous enzymes or are minimally susceptible to endogenous enzymatic breakdown. The intestinal microbiota of Equus Linnaeus animals interacts with the host, participating in energy metabolism and immune regulation. External factors (such as diet composition and environmental conditions) and internal factors (including age, gender, and genetic background) can disrupt the balance of intestinal microorganisms, leading to digestive system disorders such as diarrhea, colic, and colitis. The article provides an overview of core microbial communities in the Equus Linnaeus animals intestinal tract, community structures in different gut segments, factors influencing gut microbiota, and the relationship between gut microbiota and overall health. Rare and vulnerable endemic plants represent different evolutionary units that occur at different times, and protecting these species is a key issue in biological protection. Understanding the impact of the history of endangered plant populations on their genetic diversity helps to reveal evolutionary history and is crucial for guiding conservation efforts. Saussurea involucrata, a perennial alpine species mainly distributed in the Tianshan Mountains, is famous for its medicinal value but has become endangered due to over-exploitation. In the present study, we employed both nuclear and chloroplast DNA sequences to investigate the genetic distribution pattern and evolutionary history of S. involucrata. A total of 270 individuals covering nine S. involucrata populations were sampled for the amplification and sequencing of nrDNA Internal Transcribed Spacer (ITS) and chloroplast trnL-trnF, matK and ndhF-rpl32 sequences. Via calculation, we identified 7 nuclear and 12 plastid haplotypes. Among the nine populations, GL and BA were characterized by high haplotype diversity, whereas BG revealed the lowest haplotype diversity. Molecular dating estimations suggest that divergence among S. involucrata populations occurred around 0.75 Ma, coinciding with the uplift of Tianshan Mountains. Our results reveal that both isolation-by-distance (IBD) and isolation-by-resistance (IBR) have promoted genetic differentiation among populations of S. involucrata. The results from the ecological niche modeling analyses show a more suitable habitat for S. involucrata in the past than at present, indicating a historical distribution contraction of the species. Objective: Adult age estimation (AAE) is a challenging task. Deep learning (DL) could be a supportive tool. This study aimed to develop DL models for AAE based on CT images and compare their performance to the manual visual scoring method. Methods: Chest CT were reconstructed using volume rendering (VR) and maximum intensity projection (MIP) separately. Retrospective data of 2500 patients aged 20.00–69.99 years were obtained. The cohort was split into training (80%) and validation (20%) sets. Additional independent data from 200 patients were used as the test set and external validation set. Different modality DL models were developed accordingly. Comparisons were hierarchically performed by VR versus MIP, single-modality versus multi-modality, and DL versus manual method. Mean absolute error (MAE) was the primary parameter of comparison. Results: A total of 2700 patients (mean age = 45.24 years ± 14.03 [SD]) were evaluated. Of single-modality models, MAEs yielded by VR were lower than MIP. Multi-modality models generally yielded lower MAEs than the optimal single-modality model. The best-performing multi-modality model obtained the lowest MAEs of 3.78 in males and 3.40 in females. On the test set, DL achieved MAEs of 3.78 in males and 3.92 in females, which were far better than the MAEs of 8.90 and 6.42 respectively, for the manual method. For the external validation, MAEs were 6.05 in males and 6.68 in females for DL, and 6.93 and 8.28 for the manual method. Conclusions: DL demonstrated better performance than the manual method in AAE based on CT reconstruction of the costal cartilage. Clinical relevance statement: Aging leads to diseases, functional performance deterioration, and both physical and physiological damage over time. Accurate AAE may aid in diagnosing the personalization of aging processes. We fabricated a microfluidic channel with a graphene oxide (GNO)-doped polydimethylsiloxane photothermal heater for thermophoretically shifting particles. Under 808 nm near infrared (NIR) irradiation, GNO doped in one side of the channel walls would be heated through the photothermal effect, which would create a temperature gradient in the channel and drive particles shifting toward the cold side. By controlling NIR intensity and irradiation area, the temperature distribution in the channel can be conveniently adjusted, which can be used to separate micro- and nano-particles by changing the shifting distance of the particles. Meanwhile, as GNO was directly doped in channel walls, this method can fit channels with complex shapes well, which is useful for improving the separation efficiency by adding complex structures in channels. In arid and semi-arid regions, rainfall takes on a critical significance to both agricultural and engineering construction activities, and the transport process and driving mechanism of soil water under rainfall conditions are in need of further investigation. To clarify the variations in soil moisture, temperature, and liquid and vapor flux under various rainfall scenarios, the Mu Us Sandy Land was selected as the study region, and a water–vapor–heat transport model was established using the Hydrus-1D software with in situ observed soil and meteorological data. The simulated results were in good agreement with the measured data during both the calibration and validation periods, suggesting that the model was accurate and applicable to the study region. The variations in the selected dry and rainy periods proved the significant effect of rainfall events on soil matric potential, temperature, and driving forces. When rainfall occurred, the hydraulic conductivity for liquid water rose by three to five orders of magnitude, driving the liquid water flow downward. In contrast, the vapor flux played a vital role in soil water movement, accounting for about 15% of the total water flux in the shallow layer when the soil was dry, while it became non-significant during rainy periods due to the reduction in hydraulic conductivity for vapor and the temperature gradient. These results clarified the mechanisms of soil liquid water and vapor movement in arid areas, which could provide scientific support for future studies on vegetation restoration and ecosystem sustainability in ecologically fragile areas. Objectives: Gingivoperiosteoplasty is often used for reconstruction of alveolar defects in infants with cleft lip and palate. This study aimed to examine outcomes of tertiary gingivoperiosteoplasty, which has not previously been investigated. Materials and methods: This prospective study included 11 adults with complete cleft lip and palate (n = 12 sites) who consecutively underwent segmental Le Fort I osteotomy and concomitant gingivoperiosteoplasty for correction of skeletal class III deformity, nasoalveolar fistula and alveolar cleft. Outcomes included clinical and radiographic evaluations of gingivoperiosteoplasty at the time of osteotomy (presence of nasoalveolar fistula, residual cleft defect and unsupported root ratio of cleft-adjacent teeth), and determination of influencing factors for the clinical success of alveolar cleft repair. Study variables included age, gender, pre-surgical orthodontic treatment and alveolar cleft width of cleft-adjacent canine and angulation between cleft-adjacent teeth before surgery and 1-week postsurgery. Results: Posttreatment, no nasoalveolar fistula remained. The residual cleft defect decreased significantly (p < 0.01). The unsupported root ratio of cleft-adjacent teeth did not differ (p > 0.05); eight cleft sites reached Bergland I or II (67% success). One-week postsurgery, the minimal alveolar cleft width of cleft-adjacent canine was significantly less in the success group compared with the failed group (p = 0.01). Conclusions: Tertiary gingivoperiosteoplasty and segmental Le Fort I osteotomy decreased nasoalveolar fistulas and induced alveolar bone formation. The minimal alveolar cleft width immediately after surgery was the major influencing factor of clinical success. Generating ultrafast pulses with better spectrotemporal control is crucial for optimizing and characterizing nonlinear light–matter responses, yet it is limited by the gain bandwidth of laser media or the phase‐matching geometry of nonlinear processes. This work proposes a simple approach to independently manage a femtosecond source's spectral location and bandwidth. Self‐phase‐modulation‐enabled spectral broadening is first analyzed, which is potentially energy‐scalable using hollow‐core capillaries or multipass cells. It is demonstrated that the outmost lobes in the broadened spectrum show different dependencies on the initial pulse energy and duration. A simple yet effective toy model is introduced that successfully predicts broadband spectral tuning, and the impact of other nonlinear effects, dispersion, and input pulse asymmetry on the experimental scenario is also discussed. Thus a fiber‐based versatile source is demonstrated, which is compressible down to its transform‐limit duration, as short as 12.2 fs centered at 920 nm. In addition, bandwidth‐dependent third‐harmonic generation spectroscopy is performed from a dielectric metasurface with an optimized nonlinear response, and the dependency of laser bandwidth and pulse duration is investigated on the signal‐to‐background ratio of two‐photon images. Pupillary light reflex (PLR) assessment is a crucial examination for evaluating brainstem function, particularly in patients with acute brain injury and neurosurgical conditions. The PLR is controlled by neural pathways modulated by both the sympathetic and parasympathetic nervous systems. Altered PLR is a strong predictor of adverse outcomes after traumatic and ischemic brain injuries. However, the assessment of PLR needs to take many factors into account since it can be modulated by various medications, alcohol consumption, and neurodegenerative diseases. The development of devices capable of measuring pupil size and assessing PLR quantitatively has revolutionized the non-invasive neurological examination. Automated pupillometry, which is more accurate and precise, is widely used in diverse clinical situations. This review presents our current understanding of the anatomical and physiological basis of the PLR and the application of automated pupillometry in managing neurocritical patients. Chemoradiation-induced hearing loss (CRIHL) is one of the most devasting side effects for nasopharyngeal carcinoma (NPC) patients, which seriously affects survivors' long-term quality of life. However, few studies have comprehensively characterized the risk factors for CRIHL. In this study, we found that age at diagnosis, tumor stage, and concurrent cisplatin dose were positively associated with chemoradiation-induced hearing loss. We performed a genome-wide association study (GWAS) in 777 NPC patients and identified rs1050851 (within the exon 2 of NFKBIA), a variant with a high deleteriousness score, to be significantly associated with hearing loss risk (HR = 5.46, 95% CI 2.93–10.18, P = 9.51 × 10–08). The risk genotype of rs1050851 was associated with higher NFKBIA expression, which was correlated with lower cellular tolerance to cisplatin. According to permutation-based enrichment analysis, the variants mapping to 149 hereditary deafness genes were significantly enriched among GWAS top signals, which indicated the genetic similarity between hereditary deafness and CRIHL. Pathway analysis suggested that synaptic signaling was involved in the development of CRIHL. Additionally, the risk score integrating genetic and clinical factors can predict the risk of hearing loss with a relatively good performance in the test set. Collectively, this study shed new light on the etiology of chemoradiation-induced hearing loss, which facilitates high-risk individuals' identification for personalized prevention and treatment. Background: Patients suffered from severe traumatic brain injury (TBI) have twice the risk of developing into neurodegenerative diseases later in their life. Thus, early intervention is needed not only to treat TBI but also to reduce neurodegenerative diseases in the future. Physiological functions of neurons highly depend on mitochondria. Thus, when mitochondrial integrity is compromised by injury, neurons would initiate a cascade of events to maintain homeostasis of mitochondria. However, what protein senses mitochondrial dysfunction and how mitochondrial homeostasis is maintained during regeneration remains unclear. Results: We found that TBI-increased transcription of a mitochondrial protein, phosphoglycerate mutase 5 (PGAM5), during acute phase was via topological remodeling of a novel enhancer-promoter interaction. This up-regulated PGAM5 correlated with mitophagy, whereas presenilins-associated rhomboid-like protein (PARL)-dependent PGAM5 cleavage at a later stage of TBI enhanced mitochondrial transcription factor A (TFAM) expression and mitochondrial mass. To test whether PGAM5 cleavage and TFAM expression were sufficient for functional recovery, mitochondrial oxidative phosphorylation uncoupler carbonyl cyanide 4-(trifluoromethoxy) phenylhydrazone (FCCP) was used to uncouple electron transport chain and reduce mitochondrial function. As a result, FCCP triggered PGAM5 cleavage, TFAM expression and recovery of motor function deficits of CCI mice. Conclusions: Findings from this study implicate that PGAM5 may act as a mitochondrial sensor for brain injury to activate its own transcription at acute phase, serving to remove damaged mitochondria through mitophagy. Subsequently, PGAM5 is cleaved by PARL, and TFAM expression is increased for mitochondrial biogenesis at a later stage after TBI. Introduction: Since being designated as medicines by World Health Organization (WHO), blood components are subject to pharmacovigilance reporting. Using VigiBase, the WHO global database of individual case safety reports (ICSRs), we characterized reports of adverse reactions for all blood products. Study Design and Methods: ICSRs involving blood products as the suspected medicine in VigiBase between 1968 and 2021 were extracted. MedDRA preferred terms and the International Society of Blood Transfusion haemovigilance definitions were used to stratify adverse reactions. Descriptive statistics were used to characterize ICSR demographics. Results: A total of 111,033 ICSRs containing 577,577 suspected adverse reactions with 6152 MedDRA preferred terms were reported for 34 blood products. There were 12,153 (10.9%) reports for blood components, 98,135 (88.4%) reports for plasma‐derived medicines, and 745 (0.7%) reports for recombinant products. The majority of reports (21.0% and 19.7%, respectively) were from patients aged 45–64 and over 65 years. The Americas contributed the most ICSRs (49.7%). Top reported suspected adverse reactions were for the following MedDRA preferred terms: headache (3.5%), pyrexia (2.8%), chills (2.8%), dyspnoea (1.8%), and nausea (1.8%). Conclusion: VigiBase already has a large number of reports on blood products. When compared to other existing haemovigilance databases, our study found reports from a broader range of countries and reporters. Hyperspectral image (HSI) provides rich spectral–spatial information and the light detection and ranging (LiDAR) data reflect the elevation information, which can be jointly exploited for better land-cover classification. However, due to different imaging mechanisms, HSI and LiDAR data always present significant image difference, current pixel-wise feature fusion classification methods relying on concatenation or weighted fusion are not effective. To achieve accurate classification result, it is important to extract and fuse similar high-order semantic information and complementary discriminative information contained in multimodal data. In this paper, we propose a novel coupled adversarial learning based classification (CALC) method for fusion classification of HSI and LiDAR data. In specific, a coupled adversarial feature learning (CAFL) sub-network is first trained, to effectively learn the high-order semantic features from HSI and LiDAR data in an unsupervised manner. On one hand, the proposed CAFL sub-network establishes an adversarial game between dual generators and discriminators, so that the learnt features can preserve detail information in HSI and LiDAR data, respectively. On the other hand, by designing weight-sharing and linear fusion structure in the dual generators, we can simultaneously extract similar high-order semantic information and modal-specific complementary information. Meanwhile, a supervised multi-level feature fusion classification (MFFC) sub-network is trained, to further improve the classification performance via adaptive probability fusion strategy. In brief, the low-level, mid-level and high-level features learnt by the CAFL sub-network lead to multiple class estimation probabilities, which are then adaptively combined to generate a final accurate classification result. Both the CAFL and MFFC sub-networks are collaboratively trained by optimizing a designed joint loss function, which consists of unsupervised adversarial loss and supervised classification loss. Overall, by optimizing the joint loss function, the proposed CALC network is pushed to learn highly discriminative fusion features from multimodal data, leading to higher classification accuracies. Extensive experiments on three well-known HSI and LiDAR data sets demonstrate the superior classification performance by the proposed CALC method than several state-of-the-art methods. Frequency conversion is an essential tool in modern communication devices. Traditionally, frequency conversion is achieved through parametric coupling via nonlinear inductors or capacitors whose reactance is modulated by a carrier wave. In this study, nonlinear acoustic Lamb wave devices are explored for simultaneous signal filtering and frequency conversion. Three sets of interdigitated transducers are fabricated on a piezoelectric aluminum nitride (AlN) thin film to provide a carrier wave, a low-power signal wave, and to receive a frequency converted mixed wave. Two devices are fabricated and tested to demonstrate frequency upconversion and downconversion by utilizing mechanical nonlinearity of AlN, and the results are compared to a nonlinear circuit model. The nonlinear circuit model is used to link experimentally observed phenomenon to the acoustic material's intrinsic nonlinearity. The nonlinearity of AlN reaches a maximum of 2.8% with a carrier wave power at 28 dBm. An analytical model is used to predict device performance along with physical dimensions. These analytical results show that nonlinear acoustic mixers and filters can approach sub-millimeter sizes, which is orders-of-magnitude smaller than conventional structures using nonlinear inductors and capacitors. Simple Summary: Mood disorders including depression and bipolar were associated with an increased hazard ratio of cervical cancer. The cumulative incidence of cervical cancer was significantly higher in participants with mood disorders than in the non-mood-disorder cohort. Thus, mood disorders could potentially increase the risk of developing subsequent cervical cancer. Objective: Psychological distress is considered a factor for cancer development. However, the impact of mood disorders (depression and bipolar) on the development of cervical cancer remains uncertain. We conducted a nationwide population-based retrospective cohort study to investigate the association between mood disorders and the subsequent risk of developing cervical cancer. Methods: A total of 138,130 participants' profiles between 2000 and 2012 were extracted from the National Health Insurance Research Database and subdivided into a mood-disorder cohort (27,626 participants) and a non-mood-disorder cohort (110,504 participants). Cohorts were propensity-matched for a 1:4 ratio according to age and index year. The Cox proportional hazards regression model was utilized for assessing cervical cancer risk between cohorts. Results: Kaplan–Meier analysis revealed that the mood-disorder cohort had a higher cumulative incidence of cervical cancer. The mood-disorder cohort was also associated with an increased risk of cervical cancer after adjustments for potential confounders. Subgroup analysis revealed a negative impact of mood disorders on cervical cancer, especially in the 30–50 years and white-collar groups. Conclusions: Our findings demonstrated that mood disorders were associated with an increased risk of cervical cancer development, which provide helpful information for clinical strategies to reduce the incidence of cervical cancer in this vulnerable population. Revealing the cosmic hydrogen reionization history is one of the main goals of the modern cosmology. z > 5 quasars (QSOs) have been used as back-lights to investigate the evolution of the intervening intergalactic medium (IGM) during the cosmic reionization since their first discovery. However, due to the small population of luminous QSOs (∼130 QSOs known to date), a tight constraint on the reionization history has not yet been placed. In this work, we aim to tighten the constraint using the 93 QSOs (5.5 < z < 7.1) recently discovered in the Subaru High-z Exploration of Low-Luminosity Quasars (SHELLQS) project. This is the largest QSO sample used to constrain the epoch of reionization. We measure the mean IGM Ly α transmission and the QSO near-zone size using the UV spectra of these QSOs. The mean IGM Ly α transmission rises above zero at z ≲ 6, indicating the end of the reionization. The near-zone sizes of the SHELLQs QSOs are consistent with sizes spanned by QSOs of lifetime t q ∼ 1–100 Myr in simulations. Due to the scatter created by the low signal-to-noise spectra and large Ly α redshift uncertainty, we cannot conclude whether the redshift evolution of the near-zone size is affected by the reionization effect. Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by progressive weakness and muscular atrophy in the upper or lower limbs, ultimately leading to paralysis and death. Genetic studies have demonstrated that mutation in the gene encoding fused in sarcoma (FUS) is an uncommon cause of ALS. Here, we report a case of a 31-year-old Asian man with ALS with rare onset of dropped-head syndrome. Symptoms, including asymmetric proximal weakness of the upper limbs, hoarseness, dysphagia, and nocturnal dyspnea, emerged over a period of 5 months. After genetic testing, the patient was confirmed to harbor a novel pathogenic heterozygous mutation, c.1558C > T (p.R520C). Backgroud: Postoperative nausea and vomiting (PONV) is one of the most common complications after total thyroidectomy under general anesthesia. Total intravenous anesthesia (TIVA) has been documented to prevent PONV in patients undergoing total thyroidectomy. Penehyclidine, an anticholinergic agent with an elimination half-life of over 10 h, is widely used as premedication to reduce glandular secretion. This study aimed to explore the preventative effects of penehyclidine with propofol-remifentanil-TIVA to single-TIVA on PONV in patients undergoing total thyroidectomy. Methods: A total of 100 patients scheduled for total thyroidectomy were randomly assigned to either the penehyclidine group (n = 50) or TIVA group (n = 50). Propofol and remifentanil were was used for TIVA in all patients. No patients who received premedication. Patients were administrated with either 5 ml of normal saline or 0.5 mg of penehyclidine soon after anesthesia induction. The incidence of nausea and vomiting, the severity of nausea, the requirement of rescue antiemetics, and adverse effects were investigated during the first 24 h in two time periods (0–2 h and 2–24 h). Results: The overall PONV incidence during the 24 h after surgery was significantly lower in the penehyclidine group compared with the TIVA group (12% vs 36%, P < 0.005). Besides, the incidence of nausea and the incidence of vomiting were significantly lower in the penehyclidine group compared with the TIVA group at 2–24 h after surgery. However, there was no significant difference between the two groups at 0–2 h after surgery. Conclusions: Administration of penehyclidine under TIVA with propofol-remifentanil is more effective for prevention of PONV than TIVA alone, especially 2–24 h after total thyroidectomy. By adopting atomic layer deposition (ALD) sidewall passivation, the electroluminescence (EL) and communication performances of mini-light emitting diodes (mini-LEDs) of different sizes were improved. In particular, the most significant improvement in the electroluminescence properties of the external quantum efficiency (EQE) (a 7.2% increase), leakage current, and the communication properties of the modulation bandwidth (a 20% increase) transmission rate and bit error rate (BER) was found in the smallest mini-LEDs (80 $\mu \text{m}\,\,\times $ 120 $\mu \text{m}$). According to the results of time-resolved photoluminescence (TRPL) measurements, the carrier lifetime of the samples can be affected by both the size and ALD sidewall passivation. Removal of CO2 or C2H4 from C2H2 is still a challenging task due to their similar physical–chemical properties. Here, a bifunctional ligand decorated with amino and sulfoxide groups, 5′,5′′′′-sulfonylbis (2′-amino-[1,1′:3′,1′′-terphenyl]-4,4′′-dicarboxylic acid) (H4L), was employed to construct a new microporous iron-organic framework (Fe-MOF) with the formula [(Fe3O)(L)1.5(H2O)3]n. This MOF can serve as a parent structure to obtain the isostructural Cr-MOF by solvent-assisted metal metathesis. Furthermore, the gas adsorption and separation performance of these two MOFs were systematically investigated. Compared to Fe-MOF, Cr-MOF shows a moderately higher CO2, C2H2 and C2H4 uptake capacity. Additionally, Cr-MOF can selectively adsorb C2H2 over CO2 and C2H4. The separation potential towards C2H2/C2H4 and C2H2/CO2 was further established via IAST calculations of mixture adsorption equilibrium. Additionally, the increased specific surface area and pore structure provided numerous adsorption and active sites. Radical scavenger experiments and electron spin resonance experiments further revealed that O 2 − and h+ played crucial roles in the degradation process. —Machine learning technologies have gained significant popularity in rechargeable battery research in recent years, and have been extensively adopted to construct data-driven solutions to tackle multiple challenges for energy storage in embedded computing systems. An important application in this area is the machine learning-based battery lifetime prediction, which formulates regression models to estimate the remaining lifetimes of batteries given the measurement data collected from the testing process. Due to the non-idealities in practical operations, these measurements are usually impacted by various types of interference, thereby involving noise on both input variables and regression labels. Therefore, existing works that focus solely on minimizing the regression error on the labels cannot adequately adapt to the practical scenarios with noisy variables. To address this issue, this study adopts total least squares (TLS) to construct a regression model that achieves superior regression accuracy by simultaneously optimizing the estimation of both variables and labels. Furthermore, due to the expensive cost for collecting battery cycling data, the number of labeled data samples used for predictive modeling is often limited. It, in turn, can easily lead to overfitting, especially for TLS, which has a relatively larger set of problem unknowns to solve. To tackle this difficulty, the TLS method is investigated conjoined with stepwise feature selection in this work. Hepatic steatosis, a lipid disorder characterized by the accumulation of intrahepatic fat, is more prevalent in the elderly population. This study investigates the role of miR-155–5p in the autophagy dysregulation of aging hepatic steatosis. We established an aging mouse model in vivo and a hepatocellular senescence model induced by low serum and palmitic acid in vitro. The fluctuations of microRNAs were derived from RNA-seq data and confirmed by qPCR in 4- and 18-month-old mouse liver tissues. Hematoxylin-eosin (H&E) staining observed pathological changes. Markers of senescence, autophagy, and lipolysis genes were analyzed using Western blot and qPCR. Bioinformatics analysis predicted miR-155–5p's target gene PICALM, confirmed by dual luciferase reporter assay and transfection of miR-155–5p mimic/inhibitor into senescent hepatocytes. Senescent markers (p21, p16, and p-P53) and miR-155–5p were up-regulated in aging liver tissues and senescent hepatocytes. Bioinformatics analysis identified PICALM as a target gene of miR-155–5p, a finding further supported by dual luciferase reporter assays. Inhibition of miR-155–5p reduced expression of senescent marker genes (p16, p21, p-P53), improved autophagy (evidenced by increased LC3B-II and ATG5, and decreased P62), and enhanced lipolysis (indicated by increased ATGL and p-HSL) in senescent hepatocytes. Oil red O staining confirmed that miR-155–5p inhibition significantly reduced lipid accumulation in these cells. Stroke is an acute cerebrovascular disease caused by the rapid rupture or blockage of intracranial blood vessels for a variety of reasons, preventing blood from flowing into the brain and causing damage to brain tissue. The global burden of stroke disease is quickly increasing, and ischemic stroke (IS) accounts for 60 percent to 70 percent of all strokes, owing to the prevalence of people's bad lifestyles and the intensity of global ageing. Although most IS patients have received effective treatment, many patients still have certain dysfunction or death after treatment, and the recurrence rate is about 18%, which brings a heavy economic burden to society and families. Therefore, it is urgent to build a postoperative prediction model for IS, so as to take targeted clinical intervention measures, which has extremely important practical significance for improving the prognosis of IS. The following work has been done in this paper: (1) the theoretical background for the BP prediction model and logistic regression prediction model suggested in this work is offered, as well as the research progress and related technologies of IS recurrence prediction by domestic and foreign academics. (2) The basic principles of BPNN and logistic regression are introduced, and the logistic multifactor predictor is constructed. (3) The experimental results show that the consistency rate, sensitivity, and specificity of the prediction results of BPNN are higher than those of logistic regression, indicating that for diseases such as IS, which have many pathogenic factors and complex relationships between factors, the fitting effect of BPNN model is better than that of the logistic regression model. With the continuous improvement of medical technology and the aging of the population, the death rate of stroke is gradually decreasing, but the recurrence rate is still high, and the number of recurrences is increasing, resulting in disability and other symptoms, which brings great burden and distress to patients and their families. As the number of strokes increases, neurological impairment becomes more and more severe, affecting patients' ability to live, socialize, and work, and seriously reducing their quality of life. Objectives: To assess the performance of knee MRI for forensic age prediction and classification for 12-, 14-, 16-, and 18-year thresholds. Methods: The ossification stages of distal femoral epiphyses and proximal tibial epiphyses were assessed using an integrated staging system by Schmeling et al. and Kellinghaus et al. for knee 3.0T MRI with T1-weighted turbo spin-echo (T1-TSE) in sagittal orientation among 852 Chinese Han individuals (483 males and 369 females) aged 7–30 years. Regression models for age prediction were constructed and their performances were evaluated based on mean absolute deviation (MAD) values. In addition, the performances of age classification were assessed using receiver operating characteristic (ROC) analyses. Results: The intra- and inter-observer agreement levels were very good (κ > 0.80). The complete fusion of those two types of epiphyses took place before 18.0 years in our study participants. The minimum MAD values were 2.51 years (distal femur) and 2.69 years (proximal tibia) in males, and 2.75 years (distal femur) and 2.87 years (proximal tibia) in females. The specificity values of constructed prediction models were all above 90% for the 12-, 14-, and 16-year thresholds, compared to the 74.8–84.6% for the 18-year threshold. Better performances of age prediction and classification were observed in males by distal femoral epiphyses. Conclusions: Ossification stages via 3.0T MRI of the knee with T1-TSE sequence using an integrated staging system could be a reliable noninvasive method for age prediction or for age classification for 12-, 14-, and 16-year thresholds, especially in males by distal femoral epiphyses. However, assessments based on the full bony fusion of the distal femoral epiphysis and proximal tibial epiphysis seemed not reliable for age classification for the 18-year threshold in the Chinese Han population. Microwave radiation is a relatively new energy source that is being considered for several applications in mineral processing and extractive metallurgy. In the present research, various gibbsite-type bauxite ores were subjected to microwave radiation. The main objective was to assess the effect of microwave dewatering on the compressive strength indices of the ores and to compare the results obtained to those for conventional heating. Firstly, the fundamental interactions of the microwaves with the ores were evaluated by determining both the real and the imaginary permittivities as a function of temperature, and these were related to the water content. Secondly, the microwave heating behavior was modeled using a 24 factorial statistical analysis. Thirdly, the effect of dewatering by microwave heating on the compressive strength indices of roughly spherical bauxite ore pisoids was studied, and these results were compared to those obtained using conventional heating. Fourthly, the effect of particle size on the compressive strengths of irregular-shaped single particles of bauxite ore was investigated using both heating techniques. Finally, the energy requirements for dewatering of the ores, and hence reducing their compressive strengths, were compared for both processes. On the laboratory scale, the results showed that in comparison to conventional dewatering, microwave dewatering resulted in lower strength indices at both lower moisture removals and energy inputs. In the rapid promotion of China's electricity spot market, a large number of electricity retailers and large consumers participate in power trading, of which medium- and long-term power trading accounts for a large proportion. In the electricity spot market, the previous medium- and long-term transactions need to be closely combined with the current spot market transaction settlement rules. This paper analyzes the trading strategy of large retailers in the power market. In order to effectively reduce the total electricity cost, it is necessary to optimize the medium- and long-term transactions based on three aspects: electricity quantity and benchmark price decisions of medium- and long-term contracts, the daily electricity decomposition method in the day-ahead (DA) market, and the daily load curve decomposition strategy. According to load history characteristics that are extracted by the X12 method, daily electricity is decomposed from the medium- and long-term electricity quantity in the DA market. This paper introduces three methods of decomposing the daily load curve and proves that the particle swarm algorithm is the best method for effectively minimizing the cost in the DA market. Through analyzing the total electricity cost change pattern, we prove that the basic component of decision making is the relative relationship between the electricity price of medium- and long-term contracts and the equivalent kWh price of medium- and long-term electricity in the DA market, which is determined by the decomposition daily curve method. If the equivalent kilowatt-hour price obtained by the decomposition method in the DA market is greater than the electricity price of medium- and long-term contracts, the larger the electrical energy of medium- and long-term contracts, the lower the costs. Based on the above principles, electricity retailers can carry out planning for medium- and long-term transactions, as well as the decomposition and declaration of the daily electricity quantities and daily load curves. Noninvasive photothermal therapy (PTT) represents a promising direction for more modern and precise medical applications. However, PTT efficacy is still not satisfactory due to the existence of heat shock proteins (HSPs) and poorly targeted delivery. Herein, the design of a nanosystem with improved delivery efficacy for anticancer treatment employing the synergetic effects of reactive oxygen species (ROS)-driven chemodynamic therapy (CDT) to inactivated HSPs with photothermal-hyperthermia was therefore achieved through the development of pH-targeting glycol chitosan/iron oxide enclosed core polypyrrole nanoclusters (GCPI NCs). The designed NCs effectively accumulated toward cancer cells due to their acidic microenvironment, initiating ROS generation via Fenton reaction at the outset and performing site-specific near infrared (NIR)-photothermal effect. Store-operated channels (SOCs) are highly selective Ca2+ channels that mediate Ca2+ influx in non-excitable and excitable (i.e., skeletal and cardiac muscle) cells. These channels are triggered by Ca2+ depletion of the endoplasmic reticulum and sarcoplasmic reticulum, independently of inositol 1,4,5-trisphosphate (InsP3), which is involved in cell growth, differentiation, and gene transcription. Exploring effective spectral–spatial feature extraction methods is one of the most focused problems in current hyperspectral image (HSI) classification research. However, complex spectral–spatial structure characteristics in HSIs, e.g., shape-variable spatial structure and nonlinear spectral structure, are difficult to be effectively extracted and jointly represented. To overcome this issue, a novel superpixel-based Brownian descriptor (SBD) method for HSI classification is proposed in this article. In specific, superpixel segmentation is first used to extract shape-adaptive spatial structure information from dimension-reduced HSI, leading to generate nonoverlapping homogeneous 3-D image blocks. Then, similar pixels within the 3-D image block are jointly represented by a new local spectral–spatial feature based on the Brownian descriptor (BD). This is the first time that the BD is introduced to measure both linear and nonlinear correlations among different spectral bands in HSI. On one hand, the integration of superpixel and BD helps to provide much richer and more valuable information for better discrimination between different categories. On the other hand, the SBD can effectively represent the internal structure characters within each 3-D image block of different spatial shapes by a symmetric positive definite matrix of a united form. Finally, considering that the SBD lies on the Riemannian manifold space, a log-Euclidean kernel sparse representation (LKSR) classifier is introduced to obtain the classification results. The plausible biogenetic pathway of two unprecedented indole diterpenoids, penpaxilloids A and B (1 and 2), was postulated. Compound 1 acted as a noncompetitive inhibitor against protein tyrosine phosphatase 1B (PTP1B) with IC 50 value of 8.60 ± 0.53 μM. Compound 17 showed significant α -glucosidase inhibitory activity with IC 50 value of 19.96 ± 0.32 μM. Participants who consumed soy product of 1–6 portions/week, versus no consumption, had significantly lower risks of all-cause and CVD mortality (adjusted HR (95% CI) 0.91 (0.86, 0.97) and 0.87 (0.79, 0.96), respectively). In participants who consumed soy product of ≥ 7 portions/week, the association of higher intake with lower CVD mortality was modestly mediated by total cholesterol (4.2%, 95% CI 1.0–16.6%). Updated meta-analyses showed that the highest level of soy product intake, versus the lowest, was associated with lower risks of all-cause and CVD mortality (pooled HR (95% CI) 0.92 (0.88, 0.96) and 0.92 (0.87, 0.98), respectively).Moderate and high soy product intake were associated with lower risks of all-cause and CVD mortality. Damage to the blood–brain barrier (BBB) is an important factor leading to intracerebral hemorrhage (ICH)-induced secondary brain injury (SBI). Krüppel-like transcription factor 2 (KLF2) plays an important role in the maintenance of the BBB. This study aims to detect the changes of KLF2 after ICH and evaluate the potential effects of fraxinellone on ICH-induced SBI and its correlation with KLF2. An ICH model was established by injecting autologous blood into the right basal ganglia of Sprague-Dawley (SD) rats. First, after ICH induction, the protein levels of KLF2 were reduced. Then, we found that the decrease of KLF2 protein levels induced by ICH could be effectively reversed with the treatment of fraxinellone in vascular endothelial cells. Furthermore, fraxinellone treatment effectively alleviated brain edema, decreased the levels of TNF-α and IL-1β, and improved neuronal cell degeneration induced by ICH. Meanwhile, fraxinellone ameliorated neurobehavioral disorders, motor and sensory impairments, and neurobehavioral disorders and memory loss caused by ICH. Collectively, these findings reveal that KLF2 may be a potential target for fraxinellone to exert neuroprotective effects after ICH, and fraxinellone could be a potential therapeutic agent for SBI after ICH. • The protein levels of KLF2 decreased after ICH. • Fraxinellone attenuates the decrease of KLF2 protein levels induced by ICH. • Fraxinellone rescues ICH induced-brain injury. • Fraxinellone ameliorates ICH-induced behavioral and cognitive dysfunction. Forensic age estimation in living individuals is mainly based on radiological features, but direct radiography and computed tomography lead to a rise in ethical concerns due to radiation exposure. Thus, the contribution of magnetic resonance imaging (MRI) to age estimation of living individuals is a subject of ongoing research. In the current study, MRIs of shoulder were retrospectively collected from a modern Chinese Han population and data from 835 individuals (599 males and 236 females) in the age group 12 to 30 years were obtained. A staging technique based on (Schmidt et al. Int J Legal Med 121(4):321-324, 2007) and (Kellinghaus et al. Int J Legal Med 124(4):321–325, 2010) was used and all images were evaluated with T1-wieghted turbo spin echo (T1-TSE) sequence and T2-weighed fat suppression (T2-FS) sequence. One-sided images were assessed because data from both sides were considered coincidental, as no significant differences were found (P > 0.05). Two MRI sequences were evaluated separately and subsequently compared. Regression models and supportive vector classification (SVC) models were established accordingly. The intraobserver and interobserver agreement levels were good. Compared with T1-TSE sequence, the R2 values of T2-FS sequence were generally higher, while the mean absolute deviation (MAD) values were slightly lower. For T2-FS sequence, the MAD value was 1.49 years in males and 2.19 years in females. With two MRI sequences incorporated, the SVC model obtained with 85.7% correctly classified minors and 96.2% correctly classified adults in males, while 83.3% and 98.0% respectively in females. In conclusion, T2-FS sequence may slightly outperform the T1-TSE sequence in shoulder MRI analysis for age estimation, while shoulder MRIs could be a reliable prediction indicator for the 18-year threshold and two MRI sequences incorporated are encouraged. This study was to explore the effect of computed tomography (CT) images processed by image edge detection technology based on the improved Canny algorithm in the diagnosis of stroke patients with mobility dysfunction and to evaluate the clinical application value of early rehabilitation nursing (ERN). 114 patients who were diagnosed and treated in hospital and were suspected of having stroke movement dysfunction were selected as the research objects, and they were randomly divided into two groups, each with 57 patients. Patients in the control group were diagnosed with conventional CT examination, and the patients in observation group were diagnosed based on the CT images processed by the image edge detection technology based on the improved Canny algorithm. Patients in the observation group were divided into a group C and a group O. Patients (27 cases) in group O received rehabilitation training within 3 days after their vital signs were stabilized, and patients (30 cases) in group C received rehabilitation training within 3∼7 days after their condition was stabilized. The CT image diagnosis effects on patients of the control group and the observation group were analyzed, and the ERN effect on patients of the C group and the O group was compared. The results showed that the mean square error (MSE) of the improved Canny algorithm (233.78) was smaller than that of the traditional Canny algorithm and Sobel and Prewitt algorithm, and the peak signal-to-noise ratio (PSNR) (27.89) was greater than that of the traditional Canny algorithm and Sobel and Prewitt algorithm (P < 0.05). Compared with the control group, the sensitivity (85.00% vs. 62.12%), specificity (70.59% vs. 36.36%), and accuracy (80.70% vs. 54.39%) of the examination method of the observation group were much higher (P < 0.05). In addition, the total effective rate of patients in group O was 89.47%, which was greatly higher than that of group C (70.18%), and the scores of Meyer index and Barthel index were also higher in contrast to those of group C (P < 0.05). In conclusion, the improved Canny algorithm showed a clearer display on the edge detection of CT images and good application effect. It showed the effect of making conventional CT more accurate in the examination and diagnosis of stroke patients, and it was worthy of clinical application and promotion. Background. Mutations of the Ectodysplasin-A (EDA) gene are generally associated with syndrome hypohidrotic ectodermal dysplasia or nonsyndromic tooth agenesis. The influence of EDA mutations on dentinogenesis and odontoblast differentiation has not been reported. The aim of this study was to identify genetic clues for the causes of familial nonsyndromic oligodontia and explore the underlying mechanisms involved, while focusing on the role of human dental pulp stem cells (hDPSCs). Materials and Methods. Candidate gene sequences were obtained by PCR amplification and Sanger sequencing. Functional analysis was conducted, and the pathogenesis associated with EDA mutations in hDPSCs was investigated to explore the impact of the identified mutation on the phenotype. Capillary electrophoresis (CE) was used to detect X-chromosome inactivation (XCI) in the blood of female carriers. Results. In this study, we identified an EDA mutation in a Chinese family: the missense mutation c.1013C>T (Thr338Met). Transfection of hDPSCs with a mutant EDA lentivirus decreased the expression of EDA and dentin sialophosphoprotein (DSPP) compared with transfection of control EDA lentivirus. Mechanistically, mutant EDA inhibited the activation of the NF-κB pathway. The CE results showed that symptomatic female carriers had a skewed XCI with a preferential inactivation of the X chromosome that carried the normal allele. Conclusions. In summary, we demonstrated that EDA mutations result in nonsyndromic tooth agenesis in heterozygous females and that, mechanistically, EDA regulates odontogenesis through the NF-κB signalling pathway in hDPSCs. Background: Autoimmune polyendocrine syndrome type 1 (APS-1) is a rare monogenic inherited disease caused by mutations of the autoimmune regulator gene (AIRE). The three major components of this syndrome are chronic mucocutaneous candidiasis, hypoparathyroidism and adrenocortical insufficiency. Case presentation: We report a 20-year-old male who was clinically diagnosed with APS-1 at the age of 15. He was admitted to our department this time for suffering from polyuria and polydipsia for 6 months and was finally diagnosed with diabetes insipidus. Whole-exome sequencing (WES) revealed a novel compound heterozygous mutation of the AIRE gene —the c.239 T > G (p.Val80Gly) variant on one allele and the copy number variant (CNV) of 21q22.3(chr21:45,670,150–45,706,528)*1 on the other. Objective: Segmental Le Fort I osteotomy through the cleft is a common strategy to narrow the alveolar cleft in adults. This study compared skeletal stability between single and segmental Le Fort I osteotomies in patients with unilateral cleft lip and palate (UCLP). Materials and methods: This retrospective analysis examined 45 adults with complete UCLP-associated class III deformities who underwent bimaxillary surgery with either single (n = 30) or segmental (n = 15) Le Fort I advancement. Cone beam computed tomography (CBCT) scans of the facial skeleton were acquired before surgery, 1-week postsurgery, and at follow-up. Measures of landmarks from the CBCT images for the two treatment groups were compared for translation (left/right, posterior/anterior, superior/inferior) and rotation (yaw, roll, pitch). Results: Postsurgery, the downward movement of the maxilla was larger in the segmental group than the single group. At follow-up, the maxilla moved backward in both groups, and upward in the segmental group. The mandible moved forward and upward and rotated upward in both groups. The amount of upward movement and rotation was larger in the segmental group than the single group. Conclusions: Two years after bimaxillary surgery in patients with UCLP-associated class III deformity, greater relapse was found after segmental Le Fort I osteotomies in vertical translation of the maxilla and mandible, and pitch rotation of the mandible compared with single Le Fort I osteotomies. In this Letter, we explore a non-degenerate phase independent parametric quality factor (Q)-enhancement technique for aluminum nitride (AlN) Lamb wave resonators. Unlike other active Q-enhancement techniques which require precise phase control of the electronic feedback loop, this technique is implemented by parametrically pumping AlN material stiffness to realize a negative resistance seen at the signal path. The negative resistance is dependent on the nonlinear material modulation and multi-resonance coupling in the device. A nonlinear circuit model is developed to simulate the parametric coupling of each resonance and extract the nonlinearity of AlN from experimental data. With proper pump frequency and pump power, the device quality factor is boosted in both simulation and experiment. The demonstrated Q-enhancement method is simple to implement and can be applied to other types of resonators that have nonlinear behavior and support multi-resonance operation. Through transporting of substances to regulate the physiological and pathological functions of various cells, it can affect the progress of malignant tumors. Natural killer (NK) cell is an indispensable part of the human immune system, and is important in the development of cancers. In the tumor microenvironment, tumor-derived exosomes (TDEXs) can act on NK cells, adjusting their functions, and modifying the interaction between NK cells and other immune cells, resulting in the change of the immune response. In recent years, the research on the relationship between NK cells and tumor exosomes has become a hot topic. The grand challenge of cross-domain sentiment analysis is that classifiers trained in a specific domain are very sensitive to the discrepancy between domains. A sentiment classifier trained in the source domain usually have a poor performance in the target domain. One of the main strategies to solve this problem is the pivot-based strategy, which regards the feature representation as an important component. However, part-of-speech information was not considered to guide the learning of feature representation and feature mapping in previous pivot-based models. Therefore, we present a fused part-of-speech vectors and attention-based model (FAM). In our model, we fuse part-of-speech vectors and feature word embeddings as the representation of features, giving deep semantics to mapping features. And we adopt Multi-Head attention mechanism to train the cross-domain sentiment classifier to obtain the connection between different features. Objectives: Autosomal-dominant hypocalcification amelogenesis imperfecta (ADHCAI) is a hereditary disease characterized by enamel defects. ADHCAI is mainly caused by nonsense mutations in a gene called family with sequence similarity 83 member H (FAM83H). To study the pathogenesis of ADHCAI, a Chinese ADHCAI family was investigated. Materials and methods: The ultrastructure of enamel was analyzed by micro-CT and scanning electron microscopy. Whole-exome sequencing (WES) was performed to identify the pathogenic gene. The function of the mutant FAM83H was studied by real-time PCR, western blotting, subcellular localization, and protein degradation pathway analyses. Results: WES identified a known nonsense mutation (c.1915A > T) in exon 5 of the FAM83H gene, causing a truncated protein (p.Lys639*). However, the cases reported herein exhibited significant differences in the clinical phenotype compared with that the previously reported case. An abnormal enamel rod head structure was observed in affected teeth. In vitro functional studies showed altered protein localization and a decreased protein degradation rate for mutant FAM83H. Conclusions: We verified the FAM83H p.Lys639* protein as a gain-of-function variant causing ADHCAI. Abnormal enamel rod head structure was observed in teeth with mutant FAM83H proteins. We also investigated the molecular pathogenesis and presented data on the abnormal degradation of mutant FAM83H proteins. Clinical relevance: This study helped the family members to understand the disease progression and provided new insights into the pathogenesis of ADHCAI.
