A porous molecular crystal (TSCl) was found to crystallize from solution during the synthesis of tetrakis(4-sulfophenylmethane) from dichloromethane and water. Crystal structure prediction (CSP) allowed us to understand the driving force behind the formation of this porous TSCl phase and the intermolecular interactions that drive its formation. Gas sorption analysis showed that TSCl is permanently porous with selective adsorption of CO2 over N2, H2 and CH4 and a maximum CO2 uptake of 74 cm3/g at 195 K. Calculations revealed that TSCl assembles via a combination of weak hydrogen bonds and strong dispersive interactions. This illustrates that CSP can underpin new approaches to crystal engineering that do not involve more classical directional interactions, such as hydrogen bonding.

The anticipated shift in the focal point of interest of solid-state chemists, crystal engineers, and crystallographers from structure to properties to function parallels the need to apply our accumulated understanding of the intricacies of crystal structure to explaining the related properties, with the ultimate goal of harnessing that knowledge in applications that require soft, lightweight, or biocompatible organic solids. In these developments, the adaptive molecular crystals warrant particular attention as an alternative choice of materials for light, flexible, and environmentally benign devices, primarily memories, capacitors, sensors, and actuators. Some of the outstanding requirements for the application of these dynamic materials as high-efficiency energy-storage devices are strongly induced polarization, a high switching field, and narrow hysteresis in the case of reversible dynamic processes. However, having been studied almost exclusively by chemists, molecular crystals still lack the appropriate investigations that reliably evaluate their reproducibility, scalability, and actuating performance, and some important drawbacks have diverted the interest of engineers from these materials in applications. United under the umbrella term crystal adaptronics, the recent research efforts aim to realistically assess the appositeness of dynamic crystals for applications that require fast, reversible, and continuous operation over prolonged periods of time. With the aim of highlighting the most recent developments, this Perspective discusses their assets and pitfalls. It also provides some hints on the likely future developments that capitalize on the untapped, sequestered potential of this distinct materials class for applications.

Flexible organic single crystals are evolving as new materials for optical waveguides that can be used for transfer of information in organic optoelectronic microcircuits. Integration in microelectronics of such crystalline waveguides requires downsizing and precise spatial control over their shape and size at the microscale, however that currently is not possible due to difficulties with manipulation of these small, brittle objects that are prone to cracking and disintegration. Here we demonstrate that atomic force microscopy (AFM) can be used to reshape, resize and relocate single-crystal microwaveguides in order to attain spatial control over their light output. Using an AFM cantilever tip, mechanically compliant acicular microcrystals of three N-benzylideneanilines were bent to an arbitrary angle, sliced out from a bundle into individual crystals, cut into shorter crystals of arbitrary length, and moved across and above a solid surface. When excited by using laser light, such bent microcrystals act as active optical microwaveguides that transduce their fluorescence, with the total intensity of transduced light being dependent on the optical path length. This micromanipulation of the crystal waveguides using AFM is non-invasive, and after bending their emissive spectral output remains unaltered. The approach reported here effectively overcomes the difficulties that are commonly encountered with reshaping and positioning of small delicate objects (the "thick fingers" problem), and can be applied to mechanically reconfigure organic optical waveguides in order to attain spatial control over their output in two and three dimensions in optical microcircuits.

We report the first organic semiconductor crystal with a unique combination of properties that can be used as a multifunctional optoelectronic device. Mechanically flexible single crystals of 9,10-bis(phenylethynyl)anthracene (BPEA) can function as a phototransistor, photoswitch, and an optical waveguide. The material can exist as two structurally different solid phases, with single crystals of one of the phases being elastic at room temperature while those of the other are brittle and become plastic at higher temperature. The output and transfer characteristics of the devices were characterized by measuring the generation and temporal response of the switching of the photogenerated current. The current-voltage characteristics of both phases exhibit linearity and symmetry about the positive and negative voltages. The crystals transmit light in the telecommunications range with significantly low optical loss for an organic crystalline material.

The versatility in mechanical properties and the capability of optical waveguiding of molecular crystals have attracted research on the potential application of these materials in optomechanical transduction. Organic crystals are emerging as mechanically compliant, light-weight and chemically versatile alternatives to the commonly used silica and polymer waveguides. However, the previously reported organic crystals were shown to be able to transmit visible light, whereas actual implementation in telecommunication devices requires transparency in the near-infrared spectral range. Here we demonstrate that single crystals of the amino acid L-threonine could be used as optical waveguides and filters with high mechanical and thermal robustness for transduction of signals in the telecommunications range. On their (00\documentclass[12pt]{minimal} \usepackage{amsmath} \usepackage{wasysym} \usepackage{amsfonts} \usepackage{amssymb} \usepackage{amsbsy} \usepackage{mathrsfs} \usepackage{upgreek} \setlength{\oddsidemargin}{-69pt} \begin{document}$$\bar 1$$\end{document}1¯) face, crystals of this material have an extraordinarily high Young’s modulus (40.95 ± 1.03 GPa) and hardness (1.98 ± 0.11 GPa) for an organic crystal. First-principles density functional theory calculations, used in conjunction with analysis of the energy frameworks to correlate the structure with the anisotropy in the Young’s modulus, showed that the high stiffness arises as a consequence of the strong charge-assisted hydrogen bonds between the zwitterions. The crystals have low optical loss in the O, E, S and C bands of the spectrum (1250−1600 nm), while they effectively block infrared light below 1200 nm. This property favors these and possibly other related organic crystals as all-organic fiber-optic waveguides and filters for transduction of information., Fiber-optics based on organic crystals could have potential for unique telecommunications applications but typically transmit visible wavelengths. Here the authors present mechanically robust organic crystals with favourable optical properties across the main telecommunication bands in the near-infrared.

Summary Precise control over topologically intricate molecular architectures remains an open challenge for chemists due to their inherent structural complexity. We report a simple solvent-free strategy to selectively prepare two multi-component supramolecular crystalline systems based on the halogen bond, endowed with either an unknot topology or a Borromean-type entanglement. Real-time in situ monitoring of the solvent-free mechanochemical synthesis of these three-component halogen-bonded ionic co-crystals reveals that the choice of milling conditions leads to a switch in the supramolecular reaction trajectory, resulting in the selective formation of an open halogen-bonded network or a halogen-bonded Borromean-type assembly.

Simple Summary Studying the metabolome, the complete set of metabolites found in a biological sample, helps to understand how metabolism differs between healthy and diseased individuals. Malignant skin cancers (melanomas) differ greatly in their mutational landscapes and metabolism among individuals. Understanding these differences is important for accurate treatment of melanoma patients. The aim of our study was to characterize the metabolomes of genetically different human melanoma cell lines transplanted into mice. We found that melanomas, regardless of mutation status, differed greatly in their lipid profiles, which are important metabolites for energy production. Moreover, we identified seven metabolites with the potential to distinguish healthy from melanoma-bearing mice. We hope that the metabolomic differences detected in the mouse model may be reproducible in humans and potentially lead to metabolism-based therapeutic approaches for melanoma patients. Abstract Melanomas are genetically and metabolically heterogeneous, which influences therapeutic efficacy and contributes to the development of treatment resistance in patients with metastatic disease. Metabolite phenotyping helps to better understand complex metabolic diseases, such as melanoma, and facilitates the development of novel therapies. Our aim was to characterize the tumor and plasma metabolomes of mice bearing genetically different melanoma xenografts. We engrafted the human melanoma cell lines A375 (BRAF mutant), WM47 (BRAF mutant), WM3000 (NRAS mutant), and WM3311 (BRAF, NRAS, NF1 triple-wildtype) and performed a broad-spectrum targeted metabolomics analysis of tumor and plasma samples obtained from melanoma-bearing mice as well as plasma samples from healthy control mice. Differences in ceramide and phosphatidylcholine species were observed between melanoma subtypes irrespective of the genetic driver mutation. Furthermore, beta-alanine metabolism differed between melanoma subtypes and was significantly enriched in plasma from melanoma-bearing mice compared to healthy mice. Moreover, we identified beta-alanine, p-cresol sulfate, sarcosine, tiglylcarnitine, two dihexosylceramides, and one phosphatidylcholine as potential melanoma biomarkers in plasma. The present data reflect the metabolic heterogeneity of melanomas but also suggest a diagnostic biomarker signature for melanoma screening.

Understanding of polymorphism of organic semiconducting materials is the key to structural control of their electrical and mechanical properties. Motivated by the ambipolar n-type charge transport and electroluminescence of thienopyrrolyldione end-capped oligothiophenes, here we studied the propensity of one representative to crystallize as different polymorphs which display distinctly different mechanically properties. The crystal structures of the two polymorphs (denoted "α" and "β") of the material, 2,2′-(2,2′-thiophene-5,5′-diyl)bis(5-butyl-5H-thieno[2,3-c]pyrrole-4,6)-dione (C4-NT3N), were determined. In the α phase, the molecules interact strongly by Ï-stacking, forming columns which are bonded via C-H-O and chalcogen bonds, and this packing is consistent with the elastic behavior observed with the crystals. Instead, the β phase has the molecules aligned along their core forming layers. While the molecules interact strongly within the layers, they are practically unbound between the layers. The presence of slip planes in this form explains the plastic deformation induced by applying a force perpendicular to the (001). The thermal behavior and the enantiotropic relationship of the polymorphs are reported.

Various aspects of molecular motion in crystals have been extensively studied in different research fields of chemistry as a valuable source of structural and dynamic information en route to new smart materials and solid-state molecular machines. Recent research efforts have been directed towards engineering crystalline media with specific motile components, namely, amphidynamic crystals, whose dynamics can be exploited to achieve specific functions such as sensing, gas separation and switchable dielectrics. The most promising structural models within this line of pursuit are based upon anisotropic Brownian rotary trajectories. In this highlight, we review the recent advances in this field, with particular emphasis on potential applications. This summary should provide useful guidelines for further development of this remarkable class of materials.

He then earned his PhD in supramolecular chemistry from the Polytechnic University of Milan under the supervision of Professor Pierangelo Metrangolo (2017). After a first postdoctoral appointment at New York University Abu Dhabi, working with Professor Pance Naumov on molecular crystalline materials, he joined the lab of Professor Andy Cooper at the University of Liverpool, where he is currently Postdoctoral Research Associate working on porous liquids and crystals.

Based on rotational dynamics measurements carried out with isomorphic co-crystals formed by halogenbonding (XB) between tritylacetylene halides (TrX) and diazabicyclo[2.2.2]octane (dabco), we were able to distinguish the sources of the enthalpic and entropie components in the rotational free energy barrier. We describe the formation of the 1:1 co-crystals (TrX···N(R)3N) obtained from 1 equiv of dabco and 1 equiv of either Tri or TrBr, respectively, to give 4a and 4b instead of the potential 2:1 complexes. The co-crystals were prepared by solvent evaporation and mechanochemical synthesis. No co-crystal with TrCl was obtained, reflecting the weaker nature of the TrCl···NR3 interaction. Single-crystal X-ray diffraction confirmed structures that resemble a spinning top on a tripod and revealed that the two XB co-crystals are isomorphous, with slightly different C-X···NR3 (X = I, Br) distances and packing interactions. Quadrupolar-echo2H NMR experiments with2H-labeled samples showed that fast rotation of dabco in these co-crystals follows a six-fold potential energy surface with three lowest energy minima. Variable-temperature 1H NMR spin-lattice relaxation (VT 1H T1) data revealed rotational dynamics with indistinguishable pre-exponential factors and small but distinguishable activation energies. The activation energy of 4b (Ea = 0.71 kcal mol-1) is the lowest reported in the field of amphidynamic crystals. Using the Eyring equation, we established that their activation entropy for rotation is small but negative (ΔS‡ = -3.0 cal mol-1 K-1), while there is almost a 2-fold difference in activation enthalpies, with 4a having a higher barrier (ΔH‡ = 0.95 kcal mol-1) than 4b (ΔH‡ = 0.54 kcal mol-1)-Analysis of the rotator cavity in the two co-crystals revealed subtle differences in steric interactions that account for their different activation energies.

Background Cancer is one of the greatest public health challenges worldwide, and we still lack complementary approaches to significantly enhance the efficacy of standard anticancer therapies. The ketogenic diet, a high-fat, low-carbohydrate diet with adequate amounts of protein, appears to sensitize most cancers to standard treatment by exploiting the reprogramed metabolism of cancer cells, making the diet a promising candidate as an adjuvant cancer therapy. Scope of review To critically evaluate available preclinical and clinical evidence regarding the ketogenic diet in the context of cancer therapy. Furthermore, we highlight important mechanisms that could explain the potential antitumor effects of the ketogenic diet. Major conclusions The ketogenic diet probably creates an unfavorable metabolic environment for cancer cells and thus can be regarded as a promising adjuvant as a patient-specific multifactorial therapy. The majority of preclinical and several clinical studies argue for the use of the ketogenic diet in combination with standard therapies based on its potential to enhance the antitumor effects of classic chemo- and radiotherapy, its overall good safety and tolerability and increase in quality of life. A fluorescent hollow crystal of 9,10-dicyanoanthracene was filled with a perylene diimide derivative with complementary emissive features. The obtained crystal-in-crystal hybrid was used as a multiresponsive optical waveguide, which showed passive and active modes of light transduction and a bandwidth that extends from the visible to the infrared region.

Chemiluminescence, a process of transduction of energy stored within chemical bonds of ground-state reactants into light via high-energy excited intermediates, is known in solution, but has remained undetected in macroscopic crystalline solids. By detecting thermally induced chemiluminescence from centimeter-size crystals of an organic peroxide here we demonstrate direct transduction of heat into light by thermochemiluminescence of bulk crystals. Heating of crystals of lophine hydroperoxide to ~115 °C results in detectable emission of blue-green light with maximum at 530 nm with low chemiluminescent quantum yield [(2.1 ± 0.1) × 10‒7 E mol‒1]. Spectral comparison of the thermochemiluminescence in the solid state and in solution revealed that the solid-state thermochemiluminescence of lophine peroxide is due to emission from deprotonated lophine. With selected 1,2-dioxetane, endoperoxide and aroyl peroxide we also establish that the thermochemiluminescence is common for crystalline peroxides, with the color of the emitted light varying from blue to green to red., Chemiluminescence is known in solution, but has remained undetected in macroscopic crystalline solids so far. Here the authors demonstrate direct transduction of heat into light by thermochemiluminescence in a centimeter-size lophine hydroperoxide crystal.

Halogen bonding has recently gained well deserved attention in present-day research for its importance in many fields of supramolecular science and crystal engineering. Although generally overlooked in comprehensive studies in the past, halogen bonding has become an important tool also in the field of materials science. An increased number of scientific reports are published every year where halogen bonding is exploited in soft materials rather than in crystal engineering. Here, we focus on a description of the most exciting contemporary developments in the field of halogen-bonded functional soft materials, assembled using the guiding principles of crystal engineering. We give a particular emphasis to those published in the past few years.

Solid-state nuclear magnetic resonance (SSNMR) spectroscopy is a versatile characterization technique that can provide a plethora of information complementary to single crystal X-ray diffraction (SCXRD) analysis. Herein, we present an experimental and computational investigation of the relationship between the geometry of a halogen bond (XB) and the SSNMR chemical shifts of the non-quadrupolar nuclei either directly involved in the interaction (15 N) or covalently bonded to the halogen atom (13 C). We have prepared two series of X-bonded co-crystals based upon two different dipyridyl modules, and several halobenzenes and diiodoalkanes, as XB-donors. SCXRD structures of three novel co-crystals between 1,2-bis(4-pyridyl)ethane, and 1,4-diiodobenzene, 1,6-diiodododecafluorohexane, and 1,8-diiodohexadecafluorooctane were obtained. For the first time, the change in the 15 N SSNMR chemical shifts upon XB formation is shown to experimentally correlate with the normalized distance parameter of the XB. The same overall trend is confirmed by density functional theory (DFT) calculations of the chemical shifts. 13 C NQS experiments show a positive, linear correlation between the chemical shifts and the C-I elongation, which is an indirect probe of the strength of the XB. These correlations can be of general utility to estimate the strength of the XB occurring in diverse adducts by using affordable SSNMR analysis.

An anthracene derivative, 9,10-dicyanoanthracene, crystallizes as fluorescent needle-like single crystals that can be readily plastically bent in two directions. Spatially resolved photoluminescence analysis revealed that this material has robust optoelectronic properties that are preserved upon extreme crystal deformation. The highly flexible crystals were successfully tested as efficient switchable optical waveguiding elements for both active and passive light transduction, and the mode of operation depends on the wavelength of the incident light. This prototypical dual-mode organic optical crystalline fiber brings mechanically compliant molecular organic crystals closer to applications as novel light-transducing media for wireless transfer of information in all-organic micro-optoelectronic devices.

A series of 4-pyperidinyl-1,8-naphthalimide derivatives containing at the N-position an n-methylpyridine (n = 2, 3, or 4) have been synthesized and isolated as crystalline materials. These isomers were further reacted, in the solid state, with the co-former 1,4-diiodotetrafluorobenzene (I2F4) to give three new co-crystals of the general formula n2·I2F4. All crystalline materials have been thoroughly characterized in the solid state via single-crystal and powder X-ray diffraction, infrared spectroscopy and thermal methods; in addition, the photophysical properties of all compounds have been investigated in solution and in the solid state. The fluorescence features of the crystalline solids depend on the different arrangement of the molecules in the lattice and co-crystallization leads to important changes in the emission properties of the crystalline compounds. Emission quantum yields of the solids vary between 0.11 and 0.50.

A novel class of superfluorinated and NIR-luminescent gold nanoclusters were obtained starting from a branched thiol, bearing 27 equivalent 19F atoms per molecule. These unprecedented clusters combine in a unique nanosystem both NIR photoluminescence and 19F NMR properties, thus representing a promising multimodal platform for bioimaging applications.

A small library of 1:1 cocrystals formed on halogen bonding driven self-assembly of α,ω-diiodoperfluoroalkanes with pyridine derivatives is synthesized and characterized through single crystal X-ray analyses. A strict correlation exists between the metrics of the starting tectons and the metrics of the self-assembled architectures. The pitch along the infinite chains, formed on XB driven self-assembly, and the thickness of the fluorocarbon layers, formed due to perfluorocarbon/hydrocarbon segregation, both correlate with the number of difluoromethylene groups in diiodoperfluoroalkanes.

Halogen bonds occur when electrophilic halogens (Lewis acids) attractively interact with donors of electron density (Lewis bases). This term is commonly used for interactions undertaken by monovalent halogen derivatives. The aim of this chapter is to show that the geometric features of the bonding pattern around iodine in its hypervalent derivatives justify the understanding of some of the longer bonds as halogen bonds. We suggest that interactions directionality in ionic and neutral λ(3)-iodane derivatives is evidence that the electron density distribution around iodine atoms is anisotropic, a region of most positive electrostatic potential exists on the extensions of the covalent bonds formed by iodine, and these positive caps affect, or even determine, the crystal packing of these derivatives. For instance, the short cation-anion contacts in ionic λ(3)-iodane and λ(5)-iodane derivatives fully match the halogen bond definition and geometrical prerequisites. The same holds for the short contacts the cation of ionic λ(3)-iodanes forms with lone-pair donors or the short contacts given by neutral λ(3)-iodanes with incoming nucleophiles. The longer and weaker bonds formed by iodine in hypervalent compounds are usually called secondary bondings and we propose that the term halogen bond can also be used. Compared to the term secondary bond, halogen bond may possibly be more descriptive of some bonding features, e.g., its directionality and the relationships between structure of interacting groups and interaction strength.

Six halogen-bonded cocrystals involving aromatic donors have been studied by far-IR spectroscopy. Characteristic redshift and intensity enhancement of the C-I stretching band have been observed, which provided a distinct signature of the halogen bond involving iodopentafluorobenzene.

Amyloid supramolecular assemblies have found widespread exploitation as ordered nanomaterials in a range of applications from materials science to biotechnology. New strategies are, however, required for understanding and promoting mature fibril formation from simple monomer motifs through easy and scalable processes. Noncovalent interactions are key to forming and holding the amyloid structure together. On the other hand, the halogen bond has never been used purposefully to achieve control over amyloid self-assembly. Here we show that single atom replacement of hydrogen with iodine, a halogen-bond donor, in the human calcitonin-derived amyloidogenic fragment DFNKF results in a super-gelator peptide, which forms a strong and shape-persistent hydrogel at 30-fold lower concentration than the wild-type pentapeptide. This is remarkable for such a modest perturbation in structure. Iodination of aromatic amino acids may thus develop as a general strategy for the design of new hydrogels from unprotected peptides and without using organic solvents., Amyloid assemblies lie at the heart of many physiological functions, as well as being the cause of numerous diseases. Here, the authors subtly modify wild-type pentapeptides with halides, and discover that the new halogen bonding interactions have a remarkable influence on their physical properties.
