Der cGMP-Signalweg ist als einer der Hauptregulatoren von diversen Funktionen in Eukaryoten bekannt; allerdings ist seine Funktionsweise in Protozoen wenig verstanden. Im Rahmen dieser Arbeit wurde eine Guanylatcyclase, gekoppelt mit N-terminalen P4-ATPase, in intrazellulären Parasiten Toxoplasma gondii gemeldet. Eine in silico-Analyse wies auf eine Aktivierung der Guanylatcyclase durch Heterodimerisierung ihrer Cyclasedomänen hin und ermöglichte wertvolle Einsichten in mögliche Funktionen ihrer ATPase-Domäne. Dieses Protein (477-kDa) bezeichnet als TgATPaseP-GC in dieser Studie, lokalisiert in der Plasmamembran am apikalen Pol des Parasiten. TgATPaseP-GC ist unempfänglich gegenüber genetischer Deletion und seine CRISPR/Cas9 unterstützte Spaltung beendet den lytischen Zyklus von T. gondii vorzeitig. Darüber hinaus reduzierte ein Cre/loxP-vermittelter Knockdown von TgATPaseP-GC die Synthese von cGMP im Tachyzoiten und inhibierte das Parasitenwachstum aufgrund von Beeinträchtigungen Motilitäts-abhängiger Prozesse des Austretens und Eindringens. Trotz seiner zeitlich beschränkten Funktion ist TgATPaseP-GC konstitutiv während des ganzen lytischen Zyklus exprimiert, welches eine post-translationale Regulierung des cGMP-Signalweges bedingt. Nicht zuletzt impliziert das Vorhandensein von TgATPaseP-GC-Orthologen in anderen Alveolata eine divergente Umfunktionierung der cGMP-Signalwege in Protozoen. Darüber hinaus wurde ein optogenetischer Ansatz verwendet, um den cGMP-Weg durch eine photo-aktivierte Rhodopsin-Guanylat-Cyclase (RhoGC) in T. gondii zu exprimiert. Dieses System erlaubte eine kontrollierte Erhöhung von cGMP durch Licht in einer schnellen und reversiblen Weise. Die Anregung von RhoGC stimulierte signifikant die Parasitenmotilität, deren Auswirkung auch mit erhöhten Eindringen und Austreten überwacht wurde; im Gegensatz zum genetischen Knockdown von TgATPaseP-GC. Das System ermöglicht die Vermittler des cGMP-Signalwegs durch Phosphoproteomics zu identifizieren., cGMP signaling is known as one of the master regulators of diverse functions in eukaryotes; however, its architecture and functioning in protozoans remain poorly understood. In the scope of this thesis, an exclusive guanylate cyclase coupled with N-terminal P4-ATPase was reported in an obligate intracellular parasite Toxoplasma gondii. In silico analysis indicated an activation of the guanylate cyclase by heterodimerization of its two cyclase domains and offered valuable insights into possible functions of its ATPase domain. This bulky protein (477-kDa), termed in this study as TgATPaseP-GC to reflect its envisaged multifunctionality, localizes in the plasma membrane at the apical pole of the parasite. TgATPaseP-GC is refractory to genetic deletion, and its CRISPR/Cas9-assisted disruption aborts the lytic cycle of T. gondii. Besides, Cre/loxP-mediated knockdown of TgATPaseP-GC reduced the synthesis of cGMP in tachyzoites and inhibited the parasite growth due to impairments of motility-dependent egress and invasion events. Notably, despite its temporally restricted function, TgATPaseP-GC is expressed constitutively throughout the lytic cycle, entailing a post-translational regulation of cGMP signaling. Not least, the occurrence of TgATPaseP-GC orthologs in several other alveolates implies a divergent functional repurposing of cGMP signaling in protozoans. Furthermore, an optogenetic approach was utilized to induce cGMP pathway by a photo-activated rhodopsin-guanylate cyclase (RhoGC) in T. gondii. The system enabled a light-control of cGMP elevation on crucial steps of lytic cycle in a fast, spatial and reversible manner. Excitation of RhoGC significantly stimulated the parasite motility of which impact was also monitored with an increased host-cell invasion and egress; as opposed to the genetic knockdown of TgATPaseP-GC. Having an established optogenetic system in the parasite allows to identify downstream targets of cGMP signaling via phosphoproteomic analysis.

Das zu den Protozoen gehörende Phylum der Apicomplexa umfasst nahezu 6000 Parasitenarten. Die meisten Apicomplexa haben sich an eine obligat intrazelluläre Lebensweise angepasst und infizieren verschiedenste Tiere und den Menschen. Zu den bedeutendsten Vertretern der Apicomplexa zählen Toxoplasma, Plasmodium und Eimeria. In dieser Arbeit lag der Schwerpunkt auf den drei repräsentativen Organismen Toxoplasma gondii, Eimeria falciformis und Plasmodium berghei, welche sich alle in einem etablierten Wirt (der Maus) reproduzieren, sich allerdings hinsichtlich ihrer Wirtszellen, Persistenz sowie in ihrem Reproduktionsverhalten deutlich unterscheiden. Somit ermöglichen die genannten Parasiten eine umfassende Untersuchung der Biologie der Apicomplexa. Die meisten Entwicklungsstufen dieser Pathogene sind sehr eng mit der Wirtszelle assoziiert, was auch metabolische Wechselwirkungen beinhaltet. Das Verständnis dieser Interaktionen ist unerlässlich, um die Evolution von Parasiten zu ergründen. Grundsätzlich war das Ziel dieser Arbeit, die metabolischen Netzwerke der genannten Parasiten zu eruieren und den Einfluss des Metabolismus auf Wachstum, Pathogenese und Adaption in verschiedenen Nährstoffumgebungen zu untersuchen. Unsere Vorgehensweise verband biochemische, revers-genetische, zellbiologische und optogenetische Bottom-Up-Methoden mit Top-Down-Methoden wie Lipidomics, Metabolomics und Transcriptomics um folgende Prämissen anzugehen: • Vergleichender Entwurf der metabolischen Netzwerke in den obengenannten Parasiten • Nährstoff-Plastizität für die Überlebensfähigkeit des Parasiten in verschiedenen Milieus • Umregulierung oder Ausbeutung des Wirtsmetabolismus durch intrazelluläre Parasiten • Stadien-spezifische Regulation des Metabolismus während der asexuellen Reproduktion • Identifizierung und Validierung potentieller anti-parasitischer Wirkstoffe, The protozoan phylum apicomplexa comprises nearly 6000 parasitic species. Most apicomplexans have adapted to obligate intracellular parasitism in a wide range of organisms, including animals and humans. Some notable members of the phylum include Toxoplasma, Plasmodium and Eimeria species. This study focused on three representative parasites, namely Toxoplasma gondii, Eimeria falciformis and Plasmodium berghei, all of which infect a common and well-established model host organism (i.e. mouse), but have diverged from each other considerably with respect to the target host cells, persistence and reproduction behavior. These parasites together therefore enable a fairly inclusive study of the apicomplexan biology. Most developmental stages of these pathogens intimately associate with host cells, involving a metabolic crosstalk between the two entwined entities. A germane understanding of such interactions is vital to appreciate the evolution of parasites. In a nutshell, this work aimed to determine the design of metabolic networks in indicated parasites and the impact of metabolism on growth, pathogenesis and adaptation in discrete nutritional milieus. Our approach blended bottom-up methods of biochemistry, reverse genetics, cell biology and optogenetics with the top-down lipidomics, metabolomics and transcriptomics to address the following major premises: • Comparative design of the selected metabolic networks in aforementioned parasites • Nutritional plasticity underlying the parasite survival in variable environments • Subversion or exploitation of host metabolism by intracellular parasites • Stage-specific rewiring of parasite metabolism during asexual reproduction • Identification and endorsement of potential anti-parasitic drug targets

Toxoplasmose ist weltweit eine der am häufigsten auftretenden parasitären Zoonosen mit einer geschätzten Infektionsrate von über 30%. Toxoplasma gondii (Phylum: Apicomplexa) besitzt ein Plastid ähnliches Organell, den Apicoplasten. In diesem befindet sich das einzig bekannte Redoxsystem, welches aus der Ferredoxin-NADP+-Reduktase und Ferredoxin (Fd) besteht. Fd als Elektonendonator liefert Elektronen an verschiedene essentielle Stoffwechselwege, wie der Isoprenoidvorstufen- und Liponsäuresynthese. Um die bei einem Elektronentransfer benötigte direkte Protein-Protein-Interaktion eingehend zu analysieren, wurde ein bakterielles Reverse Two Hybrid System verwendet, womit die Interaktion von TgFd und TgLipA gezeigt werden konnte. Da angenommen wird, dass Fd eine zentrale Rolle in verschiedenen Stoffwechselwegen übernimmt, ist für einen Fd Knockout ein komplexer biochemischer Phänotyp zu erwarten, der möglicherweise zum Absterben der Parasiten führt. Zur Untersuchung dessen wurden zwei komplementäre Wege verfolgt. Eine der Strategien basierte auf dem grundsätzlichen Nachweis, dass Fd unerlässlich für das Überleben von T. gondii ist. Mit Hilfe des DiCre Systems sollte ein definierter genetischer Fd Knockout hergestellt werden, welcher jedoch nicht zweifelsfrei generiert werden konnte. Bei der zweiten Strategie kam ein konditionales Knockdown System zur Anwendung, bei welchem die Expression des Fd Gens nach Induktion herabreguliert wird. Mit Hilfe dessen konnten weitreichende Auswirkungen der Fd Defizienz auf T. gondii gezeigt werden: die Fettsäuresynthese der im Apicoplasten synthetisierten Fettsäuren ist reduziert sowie die Motilität durch eine beeinträchtigte Isoprenoidsynthese verringert, wodurch insgesamt drastische Auswirkungen auf das Parasitenwachstum gezeigt werden konnten. Beide Stoffwechsel sind vom Elektronendonator Fd abhängig und durch die Fd Herabregulation betroffen. Die Ergebnisse unterstreichen die essentielle Rolle des Fd-Redoxsystems von T. gondii., Toxoplasmosis is one of the most common parasitic zoonoses world-wide, around 30% of human beings are infected. Toxoplasma gondii (phylum: Apicomplexa) contains a unique intracellular organelle derived from plastids, called apicoplast. The only known redox system in the apicoplast consists of the ferredoxin NADP+-reductase and its redox partner, ferredoxin (Fd). The latter donates electrons to different essential metabolic pathways in the apicoplast like the last two enzymes of the isoprenoid precursor biosynthesis and the lipoic acid synthesis. To dissect protein protein interactions for an electron transfer a bacterial reverse two hybrid system was used. The physical interaction of both proteins TgFd and TgLipA could be shown. Fd is supposed to play an important role in diverse metabolic pathways, hence a knock-out of the Fd gene is expected to generate a complex biochemical phenotype and be lethal to the parasite. Therefore two complementary approaches were used to analyze the role of TgFd in this context. The first strategy shall verify the essentiality of TgFd for the survival of T. gondii. It is based on the DiCre system whereby a defined genetic knock out of TgFd is produced. Respectives parasites have been generated, but at the end no genetic Fd knock out could be produced. In the second approach a conditional knock-down was generated, where the expression of the TgFd gene is repressed after induction. The Fd deficiency has wide ranging effects on T. gondii: The fatty acid synthesis of the apicoplast-synthesized fatty acids is reduced as well as the motility is decreased due to an affected isoprenoid synthesis. In total this leads to a dramatic inhibition of parasite growth. Both metabolic pathways depend upon the electron carrier Fd and thus are affected by Fd deficiency. The results underline the essential role of the ferredoxin redoxsystem of T. gondii.

Das Überleben und die Vermehrung der parasitär lebenden Apicomplexa setzen eine effiziente Synthese von Phospholipiden während ihres gesamten Lebenszyklus voraus. In dieser Arbeit nutzten wir zunächst Eimeria falciformis um den Prozess der Lipid-Biogenese in Sporozoiten zu untersuchen. Durch Lipidomics-Analysen wurde das Auftreten von zwei exklusiven Lipiden, Phosphatidylthreonin (PtdThr) und Inositolphosphorylceramid. Der Parasit exprimiert fast das gesamte Lipid-Biogenese- Netzwerk aus eukaryotischen und prokaryotischen Enzymen. Toxoplasma gondii diente als genmanipulierbarer Ersatz für die Untersuchung der Eimeria-Enzyme, mit dem wir ein stark räumlich segmentiertes Netzwerk der Lipidsynthese im Apicoplast, ER, Golgi und Mitochondrium zeigen konnten. Ebenso legte die Komplementierung einer T. gondii-Mutante mit einer PtdThr-Synthase von E. falciformis eine konvergente Funktion von PtdThr für den lytischen Zyklus von Kokzidien-Parasiten nahe. Außerdem setzten wir T. gondii als etablierten Modelorganismus ein, um die De- novo-Synthese und die metabolische Rolle eines bedeutenden Lipidvorläufers, CDP- Diacylglycerin (CDP-DAG), zu untersuchen. Wir konnten zwei phylogenetisch divergente CDP-DAG-Synthase (CDS) Enzyme in T. gondii nachweisen. Das eukaryotisch-typische TgCDS1 und das prokaryotisch-typische TgCDS2 lokalisieren im ER bzw. im Apicoplast. Der konditionierte Knockdown von TgCDS1 bremst das Parasitenwachstum stark ab, was den fast vollständigen Verlust der Virulenz im Mausmodell hervorruft. Das restliche marginale Wachstum der TgCDS1 Mutante wird durch zusätzliche Deletion der TgCDS2 verhindert. Lipidomics-Analysen zeigten eine signifikante und spezifische Abnahme der Phosphatidylinositol (PtdIns)- und Phosphatidylglycerol (PtdGro)-Level bei Verlust der TgCDS1- bzw. TgCDS2-Gene. Zusammengenommen zeigt unsere Arbeit ein Phospholipid-Biogenese-Modell mit erstaunlicher Kooperation verschiedener Organellen und einem extensiven Lipidtransport im Parasiten., The survival and proliferation of apicomplexan parasites oblige efficient synthesis of phospholipids throughout their life cycles. Here, we first deployed Eimeria falciformis to investigate the process of lipid biogenesis in sporozoites. Lipidomics analyses demonstrate the occurrence of two exclusive lipids phosphatidylthreonine (PtdThr) and inositol phosphorylceramide along with other prototypical lipids. The parasite expresses nearly the entire lipid biogenesis network, which is an evolutionary mosaic of eukaryotic- and prokaryotic-type enzymes. Using Toxoplasma gondii as a gene- tractable surrogate to examine the Eimeria enzymes, we show a highly compartmentalized network of lipid synthesis distributed primarily in the apicoplast, ER, Golgi and mitochondrion. Likewise, trans-species complementation of a T. gondii mutant with a PtdThr synthase from E. falciformis suggests a convergent function of PtdThr in promoting the lytic cycle in coccidian parasites. We also employed the well-established model parasite T. gondii to explore de novo synthesis and metabolic roles of one major lipid precursor CDP-diacylglycerol (CDP- DAG). We report the occurrence of two phylogenetically divergent CDP-DAG synthase (CDS) enzymes in T. gondii. Eukaryotic-type TgCDS1 and prokaryotic-type TgCDS2 reside in the ER and apicoplast, respectively. Conditional knockdown of TgCDS1 severely attenuates parasite growth, which translates into a nearly complete loss of virulence in a mouse model. Residual growth of the TgCDS1 mutant is abolished by subsequent deletion of TgCDS2. Lipidomics analyses reveal significant and specific decline in phosphatidylinositol (PtdIns) and phosphatidylglycerol (PtdGro) upon loss of TgCDS1 and TgCDS2, respectively. Taken together, our work establishes a phospholipid biogenesis model involving significant inter-organelle cooperation and lipid trafficking in apicomplexan parasites.

Erreger des Leishmania donovani Komplexes (LDC) verursachen viszerale Leishmaniose (VL), eine der häufigsten durch Sandmücken übertragenen Infektionskrankheiten beim Menschen. Die vorliegende von der EU geförderte Dissertation untersucht die weltweite Populationsstruktur des LDC mit besonderem Schwerpunkt auf dem Indischen Subkontinent (ISC), wo das gehäufte Auftreten von Therapieversagen ein Problem für die geplante Eliminierung von VL darstellt. Zwei hoch diskriminierende molekulare Typisierungsverfahren wurden angewendet. 845 LDC-Isolate wurden mittels Multi-Lokus-Mikrosatelliten-Typisierung (MLMT) charakterisiert. Die Parasiten wurden in Gebieten mit endemischer VL aus unterschiedlichen Wirten isoliert und repräsentieren verschiedene klinische Formen der Leishmaniose. Eine 125 Parasiten umfassende Teilprobe wurde vollständig sequenziert und in einem next-generation Multi-Lokus-Sequenz-Ansatz (ng-MLSA) typisiert, welcher auf der Analyse von genomweiten Single-Nukleotid-Polymorphismen (SNP) beruht. Sowohl die MLMT- als auch die SNP-Daten wurden mit den gleichen populationsgenetischen Methoden ausgewertet. Der ng-MLSA Ansatz bestätigte weitgehend die Populationsstrukturen des mit dem MLMT analysierten größeren Datensatzes, die genetische Struktur korrelierte mit der geographischen Herkunft der Isolate. Die Unempfänglichkeit der Parasiten gegenüber Antimon- oder Miltefosin sowie die in vitro gemessene Resistenz der Isolate vom ISC konnten nicht auf einen spezifischen Genotyp zurückgeführt oder mit einem spezifischen genetischen Merkmal verknüpft werden. Die Gesamtgenomsequenzierung konnte bisher keine Mutationen im Parasitengenom nachweisen, die in Zusammenhang mit der Antimon- und Miltefosin-Unempfindlichkeit bzw. dem Therapieversagen gebracht werden könnten. Analysen basierend auf ausgewählten Sequenzen deuten auf eine variable chromosomale Ploidie und eine erhöhte Kopienzahl einiger Gene hin, die zur Entstehung von Arzneimittelresistenzen beitragen könnten., Parasites of the Leishmania donovani species complex (LDC) cause most cases of visceral leishmaniasis (VL), one of the most fatal vector-borne parasitic human diseases. As part of an EU funded project, this dissertation has investigated the worldwide genetic population structure of parasites of the LDC, with special focus on the Indian subcontinent (ISC) where unresponsiveness to anti-leishmanial drugs has recently become an urgent problem for the containment of VL. Two types of highly discriminatory approaches have been used. Multi-locus microsatellite typing (MLMT) has been applied to 845 LDC isolates from numerous Old and New World foci of VL, from different clinical forms of the disease and from various hosts. A subset of 125 fully sequenced isolates, reflecting the worldwide distribution of the LDC, was analysed using a next-generation multi-locus sequence approach (ng-MLSA) including single nucleotide polymorphisms (SNP). Both microsatellite and SNP data sets were analysed using, in general, the same population genetic tools. The ng-MLSA approach has, in general, corroborated the population structures obtained with MLMT for the larger data set. With the exception of non MON-1 parasites, the genetic structure revealed was largely associated with the geographic origin of the isolates, but not with the clinical presentation, host specificity and the immune status of the host or year of parasite isolation. Unresponsiveness to antimony or miltefosine treatment as well as the respective resistances measured in vitro could not be linked to a specific genotype or genetic trait. Wg sequencing also failed, so far, to identify mutations, which could be related to the unresponsiveness of LDC isolates from the ISC to antimony and miltefosine therapy. Analyses of selected targets have revealed extensive variation in chromosomal ploidy in all wg sequenced isolates under study and copy number variations for some genes possibly involved in drug resistance.

Objective: The aim of this study was to examine whether the natural protease inhibitor Av-cystatin (rAv17) of the parasitic nematode Acanthocheilonema viteae exerts anti-inflammatory effects in an in vitro model of lipopolysaccharide (LPS)-activated microglia. Methods: Primary microglia were harvested from the brains of 2-day-old Wistar rats and cultured with or without rAv17 (250 nMmlcap>). After 6 and 24 h the release of nitric oxide (Griess reagent) and TNF-α (ELISA) was measured in the supernatant. Real-time PCR was performed after 2, 6 and 24 h of culture to measure the mRNA expression of IL-1β, IL-6, TNF-α, COX-2, iNOS and IL-10. To address the involved signaling pathways, nuclear NF-ĸB translocation was visualized by immunocytochemistry. Morphological changes of microglia were analyzed by Coomassie blue staining. Differences between groups were calculated using one-way ANOVA with Bonferroni's post hoc test. Results: Morphological analysis indicated that LPS-induced microglial transformation towards an amoeboid morphology is inhibited by rAv17. Av-cystatin caused a time-dependent downregulation of proinflammatory cytokines, iNOS and COX-2 mRNA expression, respectively. This was paralleled by an upregulated expression of IL-10 in resting as well as in LPS-stimulated microglia. Av-cystatin reduced the release of NO and TNF-α in the culture supernatant. Immunocytochemical staining demonstrated an attenuated translocation of NF-ĸB by Av-cystatin in response to LPS. In addition, Western blot analysis revealed a rAv17-dependent reduction of the LPS-induced ERK1/2-pathway activation. Toxoplasma gondii ist ein weit verbreiteter, obligat-intrazellulärer, einzelliger Parasit, der die lebensbedrohliche Krankheit Toxoplasmose in Menschen und Tieren hervorrufen kann. Der schnell replizierende Parasit benötigt erhebliche Mengen an Phospholipiden zur Biogenese intra- und extrazellulärer Membranen. Phosphatidylethanolamin (PtdEtn) ist ein wichtiges und ubiquitäres Phospholipid in Pro- und Eukaryoten und das zweithäufigste Lipid in T. gondii. Dieses kann de novo über den CDP-Ethanolamin Stoffwechselweg oder durch Decarboxylierung von Phosphatidylserin synthetisiert werden. Im Rahmen dieser Arbeit konnte die Expression von zwei distinkten Phosphatidylserin Decarboxylasen (PSDs) in T. gondii nachgewiesen werden: TgPSD1pv ist partiell löslich und wird über Dichte Granula in die Parasitophore Vakuole sekretiert, während sich TgPSD1mt im Mitochondrium von Tachyzoiten befindet. TgPSD1mt ist in der Lage einen Ethanolamin-auxotrophen S. cerevisiae Stamm zu komplementieren. Ein Knock-down von TgPSD1mt in T. gondii verursacht eine verlangsamte Parasitenreplikation, welche zu einem verminderten in vitro Wachstum führt. Der PtdEtn-Gehalt in der Mutante bleibt unverändert, was auf eine stringente Homöostase des zellulären PtdEtn Reservoirs durch alternative Lipidbiogenesewege hindeutet. Tatsächlich verfügt T. gondii zusätzlich über einen aktiven CDP-Ethanolamin Stoffwechselweg im Endoplasmatischen Retikulum, welcher den Verlust von TgPSD1mt partiell kompensieren kann. Das zweite, sekretierte TgPSD1pv-Enzym hingegen scheint für das Parasitenwachstum in vitro entbehrlich zu sein. Infektionsversuche mit radioaktiv markierten Wirtszellen zeigten zudem eine Aufnahme von PtdEtn oder PtdEtn-Derivaten in intrazellulär replizierenden Tachyzoiten. Diese Ergebnisse demonstrieren eine außergewöhnliche Kompartmentalisierung und Plastizität der PtdEtn-Synthese in T. gondii., Toxoplasma gondii is a remarkably successful and widespread obligate intracellular protozoan parasite, which can cause the potentially life threatening disease Toxoplasmosis in humans and animals. The fast proliferating parasite requires a significant amount of phospholipids for biogenesis of organelles and enclosing vacuolar membranes. Phosphatidylethanolamine (PtdEtn) is one of the most ubiquitous phospholipids and the second most abundant lipid in T. gondii. It can be produced de novo by the CDP-ethanolamine pathway or by decarboxylation of phosphatidylserine. This work revealed the expression of two distinct PtdSer decarboxylase (PSD) enzymes in T. gondii: One of which is Coccidia-specific and partially soluble and secreted into the parasitophorous vacuole via dense granules (TgPSD1pv), and a second enzyme that localizes in the mitochondrion (TgPSD1mt) of tachyzoites. The mitochondrial PSD can complement a S. cerevisiae mutant auxotrophic for ethanolamine. A conditional knockdown of the TgPSD1mt gene impairs the parasite growth in vitro. Surprisingly, the mutant displayed an unaltered total PtdEtn content, which suggests a stringent homeostasis of the cellular PtdEtn pool by alternative routes of lipid biogenesis. Consistently, the parasite encodes an active CDP-ethanolamine pathway in the endoplasmic reticulum. Metabolic labeling of the TgPSD1mt mutant displayed an increased utilization of ethanolamine into PtdEtn, indicating an upregulation of the de novo CDP-ethanolamine pathway. Likewise, exogenous ethanolamine partially restored the growth phenotype of the mutant. In contrast, the TgPSD1pv enzyme is dispensable for the parasite growth. Host cell pre-labeling with radioactive ethanolamine indicated a potential uptake of host-derived PtdEtn or PtdEtn-derivates by intracellular parasites. Taken together, these results demonstrate an exceptional compartmentalization and plasticity of the PtdEtn synthesis in T. gondii.

The major membrane phospholipid classes, described thus far, include phosphatidylcholine (PtdCho), phosphatidylethanolamine (PtdEtn), phosphatidylserine (PtdSer), and phosphatidylinositol (PtdIns). Here, we demonstrate the natural occurrence and genetic origin of an exclusive and rather abundant lipid, phosphatidylthreonine (PtdThr), in a common eukaryotic model parasite, Toxoplasma gondii. The parasite expresses a novel enzyme PtdThr synthase (TgPTS) to produce this lipid in its endoplasmic reticulum. Genetic disruption of TgPTS abrogates de novo synthesis of PtdThr and impairs the lytic cycle and virulence of T. gondii. The observed phenotype is caused by a reduced gliding motility, which blights the parasite egress and ensuing host cell invasion. Notably, the PTS mutant can prevent acute as well as yet-incurable chronic toxoplasmosis in a mouse model, which endorses its potential clinical utility as a metabolically attenuated vaccine. Together, the work also illustrates the functional speciation of two evolutionarily related membrane phospholipids, i.e., PtdThr and PtdSer.

Wir fanden konservierte molekulare Signaturen der Regulation durch Δ7-DA und Ascarosid bei Dauer- und infektiösen Larven. Danach wurde die hohe Konservierung durch unsere Analyse in Dauer- und Postdauer-Stadium zwischen den zwei nah verwandten freilebenden Arten C. elegans und C. briggsae identifiziert. Das heißt, dass die relative Veränderung auf mRNA- oder Protein- Ebene zwischen zwei Arten stark korreliert ist. Aber die relative Veränderung innerhalb derselben Art zeigt keine hochgradige Korrelation zwischen mRNA- und Protein-Ebene. Unsere Ergebnisse zeigen in C. elegans Dauerlarven die signifikante Reduzierung der RNA-Mengen in 20 Stoffwechselwegen. Im Gegensatz dazu speicherten Dauerlarven reichlich RNA-Mengen in GO Termen wie Ribosome und Aminoacyl-tRNA biosynthesis. Auf Protein-Ebene sind die Stoffwechselwege von Proteinsynthese und Proteinverarbeitung im endoplasmatischen Retikulum in Dauerlarven herunterreguliert und GO Terme wie Lysosome sind hochreguliert. Durch die Zeitreihenanalyse der Proteom-Remodellierung der molekularen Signaturen beim Austritt aus dem Dauer-Stadium fand wir, dass GO Terme wie metal ion binding signifikant herunterreguliert sind und der Proteinabbau hochreguliert ist. Unsere Ergebnisse vom pSILAC Experiment deuten an, dass die Proteine für Energieerzeugung und Chaperone/Proteinfaltung beim Daueraustritt schnell verbraucht sind und wieder hergestellt werden. Zum Schluss haben wir als Erste den popomR-Assay in C. elegans etabliert und ein Screening der vermeintlichen Proteinbindestellen auf poly-A-RNA durchgeführt, um in der Zukunft die konservierten Mechanismen der post-transkriptionellen Regulation durch RBPs im Dauer-Stadium zu analysieren., We found the conservation of molecular signatures by regulating with Δ7-DA and Ascarosid in dauer larvae and infective larvae. Then by our comparative analysis, the high degree of conservation between two closely related free-living species C. elegans and C. briggsae was identified in dauer and post-dauer stages. This means that the relative changes are strongly correlated on the mRNA or the protein level between two species. But the relative changes in the same species don’t show any strong correlation between the mRNA and the protein levels. Our results showed a significantly reduced amount of RNA in 20 metabolic pathways in C. elegans dauer larvae. In contrast, dauer larvae stored a large amount of RNA in GO terms such as ribosome and aminoacyl-tRNA biosynthesis. On the protein level, the metabolic pathways of protein synthesis and protein processing in endoplasmic reticulum were downregulated in dauer larvae and the term of lysosome was up-regulated. Due to time course analysis for proteome remodeling of molecular signatures during exit process from dauer stage, we found that GO terms such as metal ion binding were significantly downregulated during dauer exit and at the same time the protein degradation was up-regulated. Our results of pSILAC experiment suggest that the proteins for energy generation and chaperone/protein folding are quickly spent and rebuilded during dauer exit. Finally, we were the first to establish the popomR assay in C. elegans and performed a screening of the putative protein binding sites on poly-A RNA to analyze the conserved mechanisms of post-transcriptional regulation by RBPs in dauer larvae in the future.

Leishmania tropica ist der Auslöser von kutaner Leishmaniose beim Menschen und kommt in Afrika über den Mittleren Osten bis nach Nordindien vor. Mittels Mikrosatellitentypisierung (MLMT) wurde die weltweite Populationsstruktur dieser Spezies und der nahe verwandten Spezies L. aethiopica aufgedeckt, indem sowohl Methoden angewandt wurden, die auf genetischen Distanzen beruhen als auch solche, die auf der Analyse von Allelfrequenzen basieren. Die 195 Stämme von L. tropica sowie die acht Stämme von L. aethiopica gruppierten hauptsächlich gemäß ihrer geographischen Herkunft. Die Stämme von L. aethiopica stellten eine eigene Gruppe dar, die allerdings innerhalb der afrikanischen Stämme von L. tropica gruppierte. Vorläufige Ergebnisse einer genomweiten SNP-Analyse haben die Ergebnisse der Mikrosatellitenanalyse weitgehend bestätigt. Um die Gründe für die hohe genetische Variabilität innerhalb der Spezies L. tropica herauszufinden, wurde eine Funktionelle Klonierung durchgeführt, in der N-methyl-N’-nitro-N-nitrosoguanidin (MNNG) als Indikator für ein funktionierendes oder eingeschränktes Mismatch Repair (MMR)-System eingesetzt wurde. Dafür wurde ein Akzeptorstamm (hohe MNNG-Toleranz) mit einer Cosmidbibliothek, die genomische DNA eines Donorstammes (niedrige MNNG-Toleranz) enthielt, transfiziert. Die erhaltenen Transfektanten wurden dann auf ihre MNNG-Toleranz getestet. Die zeit- und kosteneffiziente Identifizierung von großen Mengen an Sandmücken ist wichtig für Feldstudien, in denen mehrere Tausend Mücken gefangen werden. Hier wird eine multiplexe Technik zur ligationsabhängigen Sondenamplifizierung vorgestellt, die die Identifizierung von Sandmücken-Spezies im Mittleren Osten ermöglicht. Die Spezies Phlebotomus syriacus, P. arabicus und P. papatasi können durch diese Methode mit spezies-spezifischen Sonden eindeutig identifiziert werden. Außerdem kann diese Methode dazu genutzt werden, weitere Spezies zu diskriminieren und auf gepoolte Sandmücken angewandt werden., Leishmania tropica is the causative agent of human cutaneous leishmaniasis in foci ranging from Africa through the Middle East to northern India. By multilocus microsatellite typing (MLMT), the world-wide population structure of this species and its closely related species L. aethiopica has been revealed applying methods based on both genetic distances and allele frequencies. The 195 strains of L. tropica and eight strains of L. aethiopica largely clustered according to their geographical origins. The strains of L. aethiopica formed a distinct group, although clustering among other African strains of L. tropica. Preliminary data obtained through a whole genome sequencing approach including strains of L. tropica, L. aethiopica and L. major have largely corroborated the results of the MLMT approach. To reveal the reasons for the high genetic variability among strains of L. tropica, a Functional Cloning approach was conducted using N-methyl-N’-nitro-N-nitrosoguanidin (MNNG) as an indicator for a functioning or impaired mismatch repair (MMR) system. The transfectants retrieved from the transfection of an acceptor strain exhibiting high MNNG tolerance with a cosmid library bearing the genomic DNA of a donor strain with a reduced MNNG tolerance were screened for the restored phenotype of the donor strain. The time- and cost-efficient identification of a large amount of sand flies is important since several thousands are caught during field studies. Here, a multiplex ligation-dependent probe amplification approach (MLPA) for the identification of sand flies endemic to the Middle East is introduced. The unambiguous identification of Phlebotomus syriacus, P. arabicus and P. papatasi was possible with this approach using species-specific probes. Furthermore, this technique has the potential to discriminate more species and to be applied to pooled sand fly specimens.

In C. elegans bietet die frühe Embryogenese ein attraktives Modellsystem, um Wechselwirkungen von Proteinen in vivo zu entschlüsseln. Zur präzisen Identifizierung von spezifischen Interaktionen im C. elegans Embryo wurde ein neuer quantitativer Ansatz entwickelt, welcher die Expression von Fusionsproteinen an grün fluoreszierendes Protein in vivo mit markierungsfreier Interaktionsproteomik kombiniert. Diese Strategie wurde angewandt, um die Interaktionspartner von acht Proteinen zu untersuchen, die in essentiellen biologischen Prozessen während der frühen Embryogenese involviert sind. Diese Studie liefert als Ergebnis ein erstes embryonales in vivo Interaktionsnetzwerk bestehend aus 559 Interaktionen zwischen 472 Proteinen. Dieses Netzwerk erfasst nicht nur bekannte Bindungen, sondern auch neue Interaktionen von hoher funktioneller Relevanz. Die Netzwerkinformationen wurden mit Experimenten auf Basis der Ribonukleinsäuren-Interferenz kombiniert um neue Regulatoren der sogenannten „P granules” ausfindig zu machen. Infolgedessen wurde das fadenwurmspezifische Protein GEI-12 als neuer Interaktionspartner der DYRK-Kinase MBK-2 und als wichtiger Regler für die Dynamik der „P granules“ und für die Aufrechterhaltung der Keimbahn identifiziert. Dies führt zu einem hypothetischen Modell in welchem der Phosphorylierungszustand von GEI-12 den Auf- und Abbau der „P granules“ während der frühen Embryogenese vermittelt. Darüber hinaus veranlasst GEI-12 auch die Entstehung von „P granules“ in Säugetierzellen und bindet an PP2A-Phosphatasen, was darauf hindeutet, dass die grundlegenden biophysikalischen Eigenschaften die zur Entstehung der Ribonukleoprotein-Körperchen notwendig sind, im Laufe der Evolution zwischen Spezies konserviert geblieben sind. Zusammenfassend stellt die in vivo Interaktionskartierung ein vielseitiges Werkzeug dar, welches nicht nur die funktionelle Organisation des Proteoms aufdeckt, sondern auch Einsichten in die tierische Entwicklungsbiologie liefert., In C. elegans, early embryogenesis provides an attractive model system for mapping in vivo protein interactions. In order to accurately identify specific interactions in C. elegans embryos, a new quantitative approach was developed combining in vivo expressed GFP fusion proteins with label-free interaction proteomics. This strategy was applied to studying the interaction partners of eight bait proteins involved in essential biological processes during early embryogenesis. As a result, this study generated a pilot embryo in vivo interaction network composed of 559 interactions among 472 proteins. Importantly, this network captures not only well-characterized bindings but also new interactions of high functional relevance. Further utility of the network is demonstrated by combining it with RNAi perturbation to search for new regulators of P granule formation in early embryos. Consequently, a worm-specific protein GEI-12 was discovered as a novel interaction partner of the DYRK kinase MBK-2 and as an important regulator of P granule dynamics and germline maintenance. This leads to a hypothetical model in which the phosphorylation state of GEI-12 mediates P granule assembly and disassembly during early embryogenesis. In addition, GEI-12 also induces granule formation in mammalian cells and interacts with PP2A phosphatases, indicating that the fundamental biophysical properties required for ribonucleoprotein granule formation are conserved across species during evolution. In summary, in vivo interactome mapping is a versatile approach that not only unravels the functional organization of the proteome but also can reveal insights into animal development.

Infektionen mit dem intestinalen Parasiten Giardia duodenalis, verursachen weltweit eine der häufigsten humanen Parasitosen. Bislang konnten keine eindeutigen Virulenz- oder Pathogenitätsmarker des Erregers beschrieben werden. Es wird allerdings vermutet, dass potentielle G. duodenalis Virulenzfaktoren Enzyme sind, die während des Kontaktes des Erregers mit den Dünndarmepithelzellen sezerniert werden. Eines dieser Enzyme ist die Arginin Deiminase (ADI), die Arginin zu Citrullin umwandelt. Ziel dieser Arbeit war es Merkmale zu identifizieren, die für die ADI als Virulenzfaktor sprechen. Dazu wurde das Enzym zunächst hinsichtlich seiner Bedeutung für die Wirt-Pathogen-Interaktion untersucht. Die mit rekombinanter, katalytisch aktiver ADI (Assemblage A) behandelten LPS-stimulierten humanen moDC zeigten eine Veränderung in ihrem Phänotyp als auch in ihrer Cytokinsekretion. Diese ließ sich auf die durch das Enzym hervorgerufene Arginindepletion und/oder auf die Bildung der Metabolite, Citrullin und NH4+, zurückführen. Weiterhin konnte gezeigt werden, dass Parasitenisolate verschiedener G. duodenalis Assemblage A-Subtypen, vermutlich durch die katalytische Aktivität der ADI, die Stickstoffmonoxid-Bildung einer intestinalen Epithelzelllinie inhibiert. Neben dem Einfluss auf die Wirtsimmunantwort wurde auch die Variabilität in der kodierenden Sequenz des Enzyms in verschiedenen Parasitenisolaten analysiert. Anschließend erfolgte die funktionelle Charakterisierung des nativen (verschiedene Assemblage A-Subtypen) als auch des rekombinant aufgereinigten Enzyms (Assemblage A, B und E). Dabei zeigten sich Unterschiede in der Substrataffinität der ADI für Arginin, sowohl zwischen unterschiedlichen Assemblage A-Subtypen als auch unterschiedlichen Assemblage-Klassen. Zusammenfassend wurde gezeigt, dass die G. duodenalis ADI immunmodulatorische Effekte hat und das vermutlich eine Korrelation zwischen der Variation in der Primärstruktur und der Funktion des Enzyms besteht., Giardia duodenalis (G. duodenalis) is an intestinal protozoan parasite that causes giardiasis, one of the most prevalent parasitic diseases worldwide. So far, little is known concerning host-parasite interaction, in particular what determines the parasite’s pathogenicity. Several potential virulence factors, among them the arginine deiminase (ADI) that hydrolyzes arginine into citrulline and NH4+, are discussed. The ADI was identified to be released upon contact with intestinal epithelium by Giardia trophozoites and was recognized as an immunoreactive protein during acute human giardiasis. Aim of the study was to identify hints for G. duodenalis ADI to be a virulence factor. First, to analyze the enzyme’s impact on host-parasite interplay, its influence on human monocyte-derived dendritic cells (moDC) was investigated. Treatment of LPS-stimulated cells with recombinant ADI of assemblage A changed DC phenotype (CD83, CD86) and cytokine secretion (TNF-α, IL-10, IL-12p40). These immunomodulatory changes in DC response were due to arginine depletion and the formation of reaction products, in particular, ammonium ions. Furthermore, trophozoites of different assemblage subtypes were shown, probably due to consumption of arginine by ADI, to reduce nitric oxide formation by intestinal epithelial cells in vitro. Second, variation in the ADI coding sequence of different G. duodenalis isolates being collected in a Giardia biobank was analyzed by sequencing. Subsequently, functional genetics were performed with native ADI of different assemblage A subtypes expressed by these strains as well as with purified, recombinant ADI of assemblage A, B and E. It was recognized that enzymes of the same subtype as well as of different assemblages types had different substrate affinities for arginine. To sum up, this report identified G. duodenalis ADI to be immunomodulatory and gives first indications of a correlation between enzyme function and variation of the protein primary structure.
