Abstract Slime expelled by velvet worms entraps prey insects within seconds in a hardened biopolymer network that matches the mechanical strength of industrial polymers. While the mechanic stimuli-responsive nature and building blocks of the polymerization are known, it is still unclear how the velvet worms’ slime hardens so fast. Here, we investigated the slime for the first time, not only after, but also before expulsion. Further, we investigated the slime’s micro- and nanostructures in-depth. Besides the previously reported protein nanoglobules, carbohydrates, and lipids, we discovered abundant encapsulated phosphate and carbonate salts. We also detected CO2 bubbles during the hardening of the slime. These findings, along with further observations, suggest that the encapsulated salts in expelled slime rapidly dissolve and neutralize in a baking-powder-like reaction, which seems to accelerate the drying of the slime. The proteins’ conformation and aggregation are thus influenced by shear stress and the salts’ neutralization reaction, increasing the slime’s pH and ionic strength. These insights into the drying process of the velvet worm’s slime demonstrate how naturally evolved polymerizations can unwind in seconds, and could inspire new polymers that are stimuli-responsive or fast-drying under ambient conditions.

BackgroundAutism spectrum disorder (ASD) is behaviorally and biologically heterogeneous and likely represents a series of conditions arising from different underlying genetic, metabolic, and environmental factors. There are currently no reliable diagnostic biomarkers for ASD. Based on evidence that dysregulation of branched-chain amino acids (BCAAs) may contribute to the behavioral characteristics of ASD, we tested whether dysregulation of amino acids (AAs) was a pervasive phenomenon in individuals with ASD. This is the first article to report results from the Children's Autism Metabolome Project (CAMP), a large-scale effort to define autism biomarkers based on metabolomic analyses of blood samples from young children.MethodsDysregulation of AA metabolism was identified by comparing plasma metabolites from 516 children with ASD with those from 164 age-matched typically developing children recruited into the CAMP. ASD subjects were stratified into subpopulations based on shared metabolic phenotypes associated with BCAA dysregulation.ResultsWe identified groups of AAs with positive correlations that were, as a group, negatively correlated with BCAA levels in ASD. Imbalances between these two groups of AAs identified three ASD-associated amino acid dysregulation metabotypes. The combination of glutamine, glycine, and ornithine amino acid dysregulation metabotypes identified a dysregulation in AA/BCAA metabolism that is present in 16.7% of the CAMP subjects with ASD and is detectable with a specificity of 96.3% and a positive predictive value of 93.5% within the ASD subject cohort.ConclusionsIdentification and utilization of metabotypes of ASD can lead to actionable metabolic tests that support early diagnosis and stratification for targeted therapeutic interventions.

Following the vision of the European Commission, organisations and workers establishing Industry 5.0 approaches aspire to more future-proof, resilient, sustainable, and human-centred European industries. In this contribution, we explore how technological innovations that contribute to a "win–win" interaction with involved stakeholders may be advanced in a human-centred and transparent proceeding supported by impartial expert translators who provide information or knowledge-based guidance for decision-makers, initiators and implementers in manufacturing innovation driven by sustainability. We elaborate a stepwise procedure for agreeing on milestones and conjointly treading the path towards solving innovation challenges during a translation process. We exemplify the technological aspects of such a process using an innovation case aiming at identifying parameters for enhancements in a vacuum-bagging process applied to the manufacturing of composite parts from prepregs based on condensation-curing matrix resins made from renewable resources. In detail, we present a straightforward design of an experimental approach varying the dwelling temperature and the temperature ramps during the curing of stacked prepregs. In this way, we demonstrate that for cured composites comprising a poly(furfuryl alcohol)-based matrix, the porosity and connected mechanical properties achieved with autoclave-free curing processes sensitively depend on these process parameters. Introduction: A professional and valid ultrasound examination is dependent upon an extensively experienced examiner. Recognition and classification of rare or minimally distinctive findings are a particular source of uncertainty on the part of the examiner. By creating an online-based video database of sonographic findings, we tried to share our own experience for clinical and educational purposes. Material and methods: More than a thousand video clips of documented ultrasound findings were anonymized and cut to a practicable length of between 10 and 25 sec. The findings were critically evaluated and labeled with a primary diagnosis. They were also classified by organ systems and various keywords. Results: An online portal that currently contains more than 1,000 video sequences of typical, non-typical, especially vivid or rare ultrasound findings has been created. The portal is free of charge and accessible for any Internet-capable PC. It is also optimized for use on mobile devices (smartphones, iPads, etc.). Search and location of relevant findings is performed using keywords or a diagnosis-based search function. The Internet address is www.sono.gallery. Conclusions: The video portal is a fast and universally accessible non-commercial platform. Its moving images can be used as an aid in resolving problematic differential diagnoses of typical, non-typical or especially rare ultrasound findings or in verifying one’s own findings.

Pancreatic cancer is one of the most lethal malignancies due to its late diagnosis and limited response to treatment. Tractable methods to identify and interrogate pathways involved in pancreatic tumorigenesis are urgently needed. We established organoid models from normal and neoplastic murine and human pancreas tissues. Pancreatic organoids can be rapidly generated from resected tumors and biopsies, survive cryopreservation, and exhibit ductal- and disease-stage-specific characteristics. Orthotopically transplanted neoplastic organoids recapitulate the full spectrum of tumor development by forming early-grade neoplasms that progress to locally invasive and metastatic carcinomas. Due to their ability to be genetically manipulated, organoids are a platform to probe genetic cooperation. Comprehensive transcriptional and proteomic analyses of murine pancreatic organoids revealed genes and pathways altered during disease progression. The confirmation of many of these protein changes in human tissues demonstrates that organoids are a facile model system to discover characteristics of this deadly malignancy.

Real-world labs and transdisciplinary urban research aim to initiate transand support sustainability transformations. This paper explores the categories of societal impacts and their connections, as well as chalkey principles to facilitate these impacts. The study is based on the socieempirical analysis of48 projects by a series of surveys and interviews within an independent, accompanying research program. All projects were conducted under the heading of urban sustainability transformations and pursued a transdisciplinary real-world lab approach. Three categories of societal impacts were identified: 1. social and individual changes, 2. governance changes, and 3. changes in the physical environment. Six of the projects were selected as in-depth case studies, revealing three patterns of impact pathways. The case studies also explore the relationships between processes, results, and impacts leading to key principles for fostering societal impacts and thus, sustainability transformations.
