9. Application of vacuum‐sealing drainage in buttonhole‐related arteriovenous fistula infection: A case report.

Abstract

Few studies have reported the application of vacuum‐sealing drainage of infected dialysis vascular access wounds. Herein, we present a case of buttonhole‐related arteriovenous fistula infection treated with vacuum‐sealing drainage. A 53‐year‐old female hemodialysis patient was hospitalized with an inflamed arteriovenous fistula. The patient underwent non‐tunneled catheterization for dialysis and was treated with moxifloxacin and vancomycin for staphylococcal infection. On Day 3, the skin overlying the inflamed fistula was ulcerated, resulting in severe hemorrhage. Emergency surgery was performed along with vacuum‐sealing drainage for fistula reconstruction. Vacuum‐sealing drainage accelerated the recovery of the wound without complications. No further access complications occurred during over a 3‐year follow‐up. [ABSTRACT FROM AUTHOR]

10. Fast response/recovery and sub-ppm ammonia gas sensors based on a novel V2CTx@MoS2 composite.

Abstract

Ammonia, a colorless gas with a strong irritating odor, is widely found in natural environments. Continuous exposure to ammonia, even at low concentrations, can cause irritation of the eyes, nose, and throat. In severe cases, it may even lead to respiratory distress, lung damage, and death. Therefore, detecting ammonia in a low concentration range is crucial. This study presents the synthesis and characterization of a novel V2CTx@MoS2 composite, focusing on morphology, components, and structure to confirm the successful integration of both materials. Subsequently, its sensing performance for ammonia gas in a low concentration range (0.2–10 ppm) was tested at room temperature (RT, ∼25 °C). The developed V2CTx@MoS2 composite sensor exhibited nearly 8-fold improved response to ammonia with a shorter recovery time of more than 30 s and a detection limit of 129 ppb compared with pure V2CTx. In addition, V2CTx@MoS2 demonstrated satisfactory long-term stability and high selectivity for ammonia detection. These results show that the V2CTx@MoS2-based gas sensor is promising for the rapid and stable detection of trace ammonia at RT, thereby contributing remarkably to environmental monitoring, health protection, and energy management. Abstract

Lupus nephritis (LN) is a significant cause of various acute and chronic renal diseases, which can eventually lead to end-stage renal disease. The pathogenic mechanisms of LN are characterized by abnormal activation of the immune responses, increased cytokine production, and dysregulation of inflammatory signaling pathways. LN treatment is an important issue in the prevention and treatment of systemic lupus erythematosus. Mesenchymal stem cells (MSCs) have the advantages of immunomodulation, anti-inflammation, and anti-proliferation. These unique properties make MSCs a strong candidate for cell therapy of autoimmune diseases. MSCs can suppress the proliferation of innate and adaptive immune cells, such as natural killer cells (NKs), dendritic cells (DCs), T cells, and B cells. Furthermore, MSCs suppress the functions of various immune cells, such as the cytotoxicity of T cells and NKs, maturation and antibody secretion of B cells, maturation and antigen presentation of DCs, and inhibition of cytokine secretion, such as interleukins (ILs), tumor necrosis factor (TNF), and interferons (IFNs) by a variety of immune cells. MSCs can exert immunomodulatory effects in LN through these immune functions to suppress autoimmunity, improve renal pathology, and restore kidney function in lupus mice and LN patients. Herein, we review the role of immune cells and cytokines in the pathogenesis of LN and the mechanisms involved, as well as the progress of research on the immunomodulatory role of MSCs in LN.

Abstract

OBJECTIVE: Obesity, particularly child obesity, is one of the most common public health problems in the world and raises the risk of end-stage renal disease. Zinc (Zn) is essential for multiple organs in terms of normal structure and function; however, effects of Zn deficiency or supplementation among young individuals with obesity have not been well studied. METHODS: Weaned mice were fed high-fat diets (HFD) with varied contents of Zn (Zn deficient, adequate, and supplemented) for 3 or 6 months. This study examined associations between renal pathogenesis and dietary Zn levels, specifically assessing inflammatory pathways by utilizing P38 MAPK inhibitor SB203580. RESULTS: HFD feeding induced typical syndromes of obesity-related renal disorders, which worsened by Zn marginal deficiency. The progression of obesity-related renal disorders was delayed by Zn supplementation. HFD induced renal inflammation, reflected by increased P38 MAPK phosphorylation along with increases of inflammatory cytokines MCP-1, IL-1β, IL-6, and TNF-α. P38 MAPK inhibition prevented renal pathological changes in mice fed with HFD and HFD/Zn deficiency. CONCLUSIONS: P38 MAPK mediated the renal inflammatory responses, which played a central role in the pathogenesis of HFD-induced renal disorders. Zn could delay the progression of obesity-related kidney disease by down-regulating P38 MAPK-mediated inflammation.

Abstract

Diabetic nephropathy (DN), as one of the chronic complications of diabetes, is the major cause of end-stage renal disease. However, the pathogenesis of this disease is not fully understood. In recent years, research on microRNAs (miRNAs) has become a hotspot because of their critical role in regulating posttranscriptional levels of protein-coding genes that may serve as key pathogenic factors in diseases. Several miRNAs were found to participate in the pathogenesis of DN, while others showed renal protective effects. Therefore, targeting miRNAs that are involved in DN may have a good prospect in the treatment of the disease. The aim of this review is to summarize DN-related miRNAs and provide potential targets for diagnostic strategies and therapeutic intervention.

Abstract

Z basic -ΔI-CM is a novel intein-based self-cleavable tag we developed to accelerate the soluble expression of recombinant proteins in Escherichia coli (E. coli). Previously we found that intein activity could be interfered by its flanking exteins, and thus reducing the production efficiency and final yield. In this work, we used CXC-chemokine 9 (CXCL9) as a model C-extein, which fusion with Z basic -ΔI-CM showed high intein activity. When the fusion protein got soluble expression, CXCL9 was released immediately and purified directly from cell lysis supernatant. The results demonstrated that Z basic -ΔI-CM tag had successfully mediated the efficient production of high-quality CXCL9 with reduced time and resources consumption in comparison with inclusion bodies expression. Molecular dynamics simulations suggested that the improved cleavage activity of Z basic -ΔI-CM upon fusion with CXCL9 may be due to the higher dynamics of the first half loop and stabilization of the second half loop of intein. Our results proved that the self-cleavable Z basic -ΔI-CM mediated soluble expression could be a feasible process for cytokines like CXCL9, thus of attractive potentials for production of therapeutic proteins using E. coli expression system., (Copyright © 2019 Elsevier Inc.
