Abstract

Mice are the most commonly used model animals for itch research and for development of anti-itch drugs. Most laboratories manually quantify mouse scratching behavior to assess itch intensity. This process is labor-intensive and limits large-scale genetic or drug screenings. In this study, we developed a new system, Scratch-AID (Automatic Itch Detection), which could automatically identify and quantify mouse scratching behavior with high accuracy. Our system included a custom-designed videotaping box to ensure high-quality and replicable mouse behavior recording and a convolutional recurrent neural network trained with frame-labeled mouse scratching behavior videos, induced by nape injection of chloroquine. The best trained network achieved 97.6% recall and 96.9% precision on previously unseen test videos. Remarkably, Scratch-AID could reliably identify scratching behavior in other major mouse itch models, including the acute cheek model, the histaminergic model, and a chronic itch model. Moreover, our system detected significant differences in scratching behavior between control and mice treated with an anti-itch drug. Taken together, we have established a novel deep learning-based system that could replace manual quantification for mouse scratching behavior in different itch models and for drug screening.

Abstract

High leakage current density and relatively low piezoelectric activity have become one of the main obstacles to expanding the practical application of Bi4Ti3O12 (BIT) high-temperature piezoelectric ceramics. Although ion doping can improve electrical resistivity and piezoelectric response, it often lowers the Curie temperature. In this work, by introducing CaBi2Nb2O9 (CBN) with higher Curie temperature to BIT, a composite ceramic (1−x)BIT-xCBN was designed, and the effect of CBN content on the structure and electrical properties of the ceramics was investigated. With the increase in the x value, the intensity of the highest peak (117) gradually decreased until disappearing, while the intensity of peak (115) gradually increased, and the X-ray diffraction (XRD) refinement results showed that some non-stoichiometric compounds, Bi1.74Ti2O6.624 and Ca0.5Bi2.5Ti0.5Nb1.5O9, were produced. The sheet-like grains were effectively suppressed, while the granular grains became prominent with high CBN doping content, which should contribute to improving the electrical resistivity of ceramics. The optimal electrical properties were obtained in 0.8BIT-0.2CBN composite ceramics as follows: the piezoelectric coefficient d33 = 13.8 pC/N, the Curie temperature TC = 765°C, and the electrical resistivity ρdc = 8.52 × 105 Ω·cm (@ 500°C). In addition, the d33 maintained 89.1% of the initial value after annealing at 550°C, showing good thermal stability for high-temperature sensing applications. [ABSTRACT FROM AUTHOR]

Abstract

N6‐methyl‐2′‐O‐methyladenosine (m6Am), occurring adjacent to the 7‐methylguanosine (m7G) cap structure and catalyzed by the newly identified writer PCIF1 (phosphorylated CTD interacting factor 1), has been implicated in the pathogenesis of various diseases. However, its involvement in renal cell carcinoma (RCC) remains unexplored. Here, significant upregulation of PCIF1 and m6Am levels in RCC tissues are identified, unveiling their oncogenic roles both in vitro and in vivo. Mechanically, employing m6Am‐Exo‐Seq, LPP3 (phospholipid phosphatase 3) mRNA is identified as a key downstream target whose translation is enhanced by m6Am modification. Furthermore, LPP3 is revealed as a key regulator of phosphatidic acid metabolism, critical for preventing its accumulation in mitochondria and facilitating mitochondrial fission. Consequently, Inhibition of the PCIF1/LPP3 axis significantly altered mitochondrial morphology and reduced RCC tumor progression. In addition, depletion of PCIF1 sensitizes RCC to sunitinib treatment. This study highlights the intricate interplay between m6Am modification, phosphatidic acid metabolism, and mitochondrial dynamics, offering a promising therapeutic avenue for RCC. Abstract

Ca0.94Ce0.06Bi4Ti4-x(W1/3Cr2/3)xO15 (CCBT-WCx, x = 0.00–0.10) ceramics were prepared by a solid-state sintering method, and the effect of W/Cr co-doping on the structure and electrical properties of the ceramics was systematically investigated. An enhanced piezoelectric constant d33 (22.1 pC/N) was achieved at the optimized composition with x = 0.04, which increased by 35% as compared to that of pure CCBT ceramics due to more effective distortion of the titanium-oxygen octahedron induced by W/Cr co-doping. Moreover, after annealing at 500 °C, the d33 (21.0 pC/N) of CCBT-WC0.04 ceramics still remained 95% of its initial value, exhibiting excellent annealing thermal stability. 49. Boosting energy storage properties of BNT-based relaxor ferroelectric ceramics via (Zn1/3Nb2/3)4+ complex ion doping.

Abstract

High power density electrostatic capacitor is a fundamental component of advanced electrical and electronic systems. Herein, the (Zn1/3Nb2/3)4+ complex ion was introduced into the B-site of Bi0.385Na0.325Ba0.105Sr0.155TiO3 relaxor ferroelectric ceramics to improve energy storage properties and dielectric temperature stability. All pseudo-cubic structured ceramics have clear grain boundaries with an average grain size of 1 ~ 2 μm. In the optimized composition, a recoverable energy density of 2.4 J/cm3 with an energy efficiency of 78% can be achieved under a relatively low electric field of 160 kV/cm, together with excellent stability and reliability of energy storage in temperature, frequency, and cycling fields, as well as fast charging–discharging rate. This work provides guidance for the design of high-performance energy storage dielectric materials by enhancing the B-site disorder of relaxor ferroelectric ceramics via complex ion doping.
