Cyanobacterial mats supplanting coral and spreading coral diseases in tropical reefs, intensified by environmental shifts caused by human‐induced pressures, nutrient enrichment, and global climate change, pose grave risks to the survival of coral ecosystems. In this study, we characterized Okeanomitos corallinicola gen. and sp. nov., a newly discovered toxic marine heterocyte‐forming cyanobacterium isolated from a coral reef ecosystem of the South China Sea. Phylogenetic analysis, based on the 16S rRNA gene and the secondary structure of the 16S–23S rRNA intergenic region, placed this species in a clade distinct from closely related genera, that is, Sphaerospermopsis stricto sensu, Raphidiopsis, and Amphiheterocytum. The O. corallinicola is a marine benthic species lacking gas vesicles, distinguishing it from other members of the Aphanizomenonaceae family. The genome of O. corallinicola is large and exhibits diverse functional capabilities, potentially contributing to the resilience and adaptability of coral reef ecosystems. In vitro assays revealed that O. corallinicola demonstrates notable cytotoxic activity against various cancer cell lines, suggesting its potential as a source of novel anticancer compounds. Furthermore, the identification of residual saxitoxin biosynthesis function in the genome of O. corallinicola, a marine cyanobacteria, supports the theory that saxitoxin genes in cyanobacteria and dinoflagellates may have been horizontally transferred between them or may have originated from a shared ancestor. Overall, the identification and characterization of O. corallinicola provides valuable contributions to cyanobacterial taxonomy, offering novel perspectives on complex interactions within coral reef ecosystems. Coral–bacterial interaction is a major driver in coral acclimatization to the stressful environment. 16S rRNA High‐throughput sequencing was used to classify the role of different coral reef compartments; sediment, water, and tissue; in the South China Sea (SCS), as well as different locations in shaping the microbial community. The majority of OTUs significantly shifted at impacted sites and indicated distinction in the relative abundance of bacteria compartment/site‐wise. Richness and diversity were higher, and more taxa were enriched in the sediment communities. Proteobacteria dominated sediment samples, while Cyanobacteria dominated water samples. Coral tissue showed a shift among different sites with Proteobacteria remaining the dominant Phylum. Moreover, we report a dominance of Chlorobium genus in the healthy coral tissue sample collected from the severely damaged Site B, suggesting a contribution to tolerance and adaptation to the disturbing environment. Thus, revealing the complex functionally diverse microbial patterns associated with biotic and abiotic disturbed coral reefs will deliver understanding of the symbiotic connections and competitive benefit inside the hosts niche and can reveal a measurable footprint of the environmental impacts on coral ecosystems. The family Peridiniopsidaceae encompasses mainly freshwater species of the genera Peridiniopsis, Palatinus and Parvodinium. Only one benthic, marine species 'Scrippsiella' hexapraecingula has been attributed to this family. Here we established five strains by isolating single Parvodinium-like cells from the marine Gulf of Thailand, Hainan Island waters (China), off Manado (Indonesia) and from a freshwater reservoir in Fuzhou (China). All strains were examined with light, scanning and transmission electron microscopy, and their SSU, ITS-5.8S and partial LSU rRNA regions were sequenced. Four marine strains share a plate formula of Po, cp, X, 4′, 2a, 6′′, 6C, 4S, 5′′′, 2′′′′ and are herein attributed to a new genus Johsia as J. chumphonensis gen. et sp. nov. Its theca is characterized by an epitheca 1.5 times as long as the hypotheca in dorsal view and a 2a plate about half the size of 1a. A type B eyespot was observed in J. chumphonensis comprising two rows of lipid globules within a chloroplast with a single row of crystals overlying the eyespot. Production of spherical cysts was observed in culture. The freshwater strain shows a plate pattern of Po, cp, X, 4′, 2a, 7′′, 6C, 5S, 5′′′, 2′′′′, characterized by two unequal antapical plates and a lack of antapical spines, fitting the description of Peridinium parvulum, which was transferred to Parvodinium as P. parvulum comb. nov. In this species, a type A eyespot was observed comprising four rows of lipid globules within a chloroplast. A molecular phylogeny was inferred based on concatenated data from SSU, ITS-5.8S and partial LSU rRNA gene sequences using maximum likelihood and Bayesian inference. Our results show that Johsia is nested within the Peridiniopsidaceae and is a sister clade to Peridiniopsis borgei and the strain UTEX1948 identified as 'Scrippsiella' hexapraecingula with three anterior intercalary plates. The gonyaulacean family Protoceratiaceae is characterised by five precingular plates. It currently encompasses the type genus Ceratocorys and the fossil genus Atopodinium. Fourteen strains of Ceratocorys, Pentaplacodinium, and Protoceratium were established from Malaysian and Hawaiian waters, and their morphologies were examined using light and scanning electron microscopy. Two new species, Ceratocorys malayensis sp. nov. and Pentaplacodinium usupianum sp. nov., were described from Malaysian waters. They share a Kofoidean plate formula of Po, Pt, 3ʹ, 1a, 6ʹʹ, 6C, 6S, 5ʹʹʹ, 1p, 1ʹʹʹʹ. Ceratocorys malayensis has a short first apical plate (1ʹ) with no direct contact with the anterior sulcal plate (Sa) whereas Pentaplacodinium usupianum had a parallelogram-shaped 1ʹ plate which often contacted the Sa plate. The genera Ceratocorys and Pentaplacodinium were emended accordingly to incorporate species bearing five or six precingular plates. The Protoceratium strain from Hawaii was morphologically similar to P. reticulatum, but differed in the lack of a ventral pore in plate 1′ and slight or lack of contact between plates 1′ and Sa, and is here designated as P. cf. reticulatum. The maximum-likelihood and Bayesian inference analyses based on SSU, LSU and ITS ribosomal DNA sequences revealed that these three genera are monophyletic and form a well-resolved group. Our results support Protoceratium and Pentaplacodinium as members of the family Protoceratiaceae, characterised by the presence of one anterior intercalary plate. Seven strains of Protoceratium cf. reticulatum, Ceratocorys malayensis and Pentaplacodinium usupianum were examined for yessotoxin production by LC-MS/MS but none produced a detectable amount of toxin. SUMMARY: The small athecate dinoflagellate Karlodinium zhouanum is a species recently described in the coastal waters of China. K. zhouanum is morphologically similar to Karlodinium veneficum, a typical ichthyotoxic blooming karlotoxin‐producing species, and it is impossible to distinguish between these two species based on light microscopy. In this study, strains of K. zhouanum isolated from the East China Sea were studied. By analyzing toxins, toxicity, lipid characteristics and typical molecular and physiological traits of this species, K. zhouanum was shown to be nontoxic to brine shrimp and widely spread over the coastal waters of China. No karlotoxin‐like toxin was detected by liquid chromatography‐mass spectrometry (LC–MS). Instead of gymnodinosterol, the critical sterol in toxic K. veneficum, 27(nor)‐24S‐4α‐Methyl‐5α‐ergosta‐8(14)‐en‐3β‐ol (NEE) was dominant in K. zhouanum, while gymnodinosterol was absent. These sterol characteristics may provide not only support for the species separation between toxic and nontoxic species of Karlodinium but also environmental survey tools to differentiate the contribution of nontoxic Karlodinium strains, which has been unclear until now. SUMMARY: A strain of Pseliodinium pirum was isolated from Jiaozhou Bay, China, identified based on a recently emended classification, and further characterized for its morphology using light, scanning electron, and transmission electron microscopy, and phylogeny based on SSU and partial LSU rDNA sequences. The pigment composition, life history, and potential effects on aquatic animals were also examined. We observed the typical characteristics of Ceratoperidiniaceae, in which its apical structure complex (ASC) formed a circular loop, and the ASC of our isolate comprised four to five rows of vesicles, and connected with sulcal intrusion. Epifluorescence and transmission electron microscopy revealed a bean‐shaped, centrally located nucleus, with at least 76 chromosomes. Numerous rod‐shaped chloroplasts were in connection to the irregularly shaped pyrenoids. Pigment analysis showed that peridinin was the most abundant among all carotenoids and other pigments. Phylogenetic analyses using maximum likelihood and Bayesian inference indicated that our isolate is conspecific with the entity Cochlodinium cf. helix (accession No. KF245459), but different from Ceratoperidinium, Kirithra, and other unidentified species of the family Ceratoperidiniaceae. Pseliodinium pirum could produce sexual, thin‐walled cyst, with subspherical and spherical shape and smooth surface (without spines or rough projections). The cyst could germinate within 3 days. Bioassays did not show adverse effects of P. pirum on the finfish Oryzias melastigma and the rotifer Brachionus plicatilis, indicating it may not be a harmful species. [ABSTRACT FROM AUTHOR]
