Abstract Background Zinc finger E-box binding homEeobox 1 (ZEB1) and ZEB2 are two anoikis-related transcription factors. The mRNA expressions of these two genes are significantly increased in kidney renal clear cell carcinoma (KIRC), which are associated with poor survival. Meanwhile, the mechanisms and clinical significance of ZEB1 and ZEB2 upregulation in KIRC remain unknown. Methods Through the Cancer Genome Atlas (TCGA) database and Gene Expression Omnibus (GEO) database, expression profiles, prognostic value and receiver operating characteristic curves (ROCs) of ZEB1 and ZEB2 were evaluated. The correlations of ZEB1 and ZEB2 with anoikis were further assessed in TCGA-KIRC database. Next, miRTarBase, miRDB, and TargetScan were used to predict microRNAs targeting ZEB1 and ZEB2, and TCGA-KIRC database was utilized to discern differences in microRNAs and establish the association between microRNAs and ZEBs. TCGA, TIMER, TISIDB, and TISCH were used to analyze tumor immune infiltration. Results It was found that ZEB1 and ZEB2 expression were related with histologic grade in KIRC patient. Kaplan-Meier survival analyses showed that KIRC patients with low ZEB1 or ZEB2 levels had a significantly lower survival rate. Meanwhile, ZEB1 and ZEB2 are closely related to anoikis and are regulated by microRNAs. We constructed a risk model using univariate Cox and LASSO regression analyses to identify two microRNAs (hsa-miR-130b-3p and hsa-miR-138-5p). Furthermore, ZEB1 and ZEB2 regulate immune cell invasion in KIRC tumor microenvironments. Conclusions Anoikis, cytotoxic immune cell infiltration, and patient survival outcomes were correlated with ZEB1 and ZEB2 mRNA upregulation in KIRC. ZEB1 and ZEB2 are regulated by microRNAs.

Tanshinone IIA has a potential therapeutic effect on cerebral ischemia/reperfusion injury (CIRI). In this study, tanshinone IIA was encapsulated in poly(lactic-co-glycolic acid)-block-poly (ethylene glycol)-carboxylic acid (PLGA-PEG-COOH) nanoparticles, and its therapeutic efficacy on CIRI was investigated. Morphology and dynamic light scattering analyses were performed to identify and optimize nano-formulations. A drug release test was conducted using the dialysis method. The cytotoxic effect of tanshinone IIA on human neuroblastoma cells (SH-SY5Y) and brain endothelial capillary cells (hCMEC/D3) was measured using the MTT assay. The protective effect of PLGA-PEG-COOH-encapsulated tanshinone IIA against CIRI was evaluated in oxygen and glucose deprivation/reoxygenation-induced SH-SY5Y/IR cells and middle cerebral artery occlusion (MCAO) rats. Results showed that PLGA-PEG-COOH-encapsulated tanshinone IIA promoted viability and inhibited apoptosis of SH-SY5Y/IR cells (P < 0.01). Moreover, PLGA-PEG-COOH-encapsulated tanshinone IIA facilitated the invasion of SH-SY5Y/IR cells and repressed inflammation in MCAO rats (P < 0.01). Noteworthy, PLGA-PEG-COOH-encapsulated tanshinone IIA combined with angiopep-2 peptide presented a better inhibitory effect on CIRI than tanshinone IIA alone (P < 0.01). Angiopep-2 peptide contributes to traversing blood–brain barrier by recognizing lipoprotein-related protein expressed in the brain capillary endothelial cells. In conclusion, PLGA-PEG-COOH-encapsulated tanshinone IIA plus angiopep-2 peptide holds promising therapeutic potential toward CIRI.

In this paper，the differences between neutral salt spray test and cyclic tests (including GM GMW 14872，Volkswagen PV1210，JASO M610) in the evaluation of corrosion effect of bare sheet and galvanized sheet were analyzed，and the effects of different accelerated corrosion tests on the corrosion morphology and corrosion mark expansion value of different coating materials were determined.By comparing and analyzing the evaluation results of the corrosion resistance of several coating materials obtained by different corrosion test methods，it could be found that the macroscopic morphology of paint damage in salt spray test and the above-mentioined cyclic tests presented as follows: GA board was the best，followed by GI board，and CR board was the worst.For the neutral salt spray test，the expansion value of single side erosion mark of GI board was the largest，followed by that of GA board，and CR board had the smallest value.For the JASO M610 and PV1210 cyclic corrosion tests，that value of GI board was the largest，followed by CR board，and GA board had the smallest value.For the GMW14872 cycle test，that value of GA board was the smallest，followed by GI board，and CR board had the largest value.

Centromere protein M (CENPM) is essential for chromosome separation during mitosis. However, its roles in lung adenocarcinoma (LUAD) progression and metastasis remain unknown. In this study, we aimed to explore the effects of CENPM on LUAD progression as well as the underlying mechanisms. We analyzed the expression of CENPM and its correlation with clinicopathological characteristics using GEO LUAD chip datasets and TCGA dataset. We further investigated the impact of CENPM on LUAD in vitro and in vivo. In silico analysis and qRT-PCR revealed that CENPM is upregulated in LUAD compared with that in normal lung tissues. Via gain/loss-of-function assays, we further found that CENPM promotes the LUAD cell cycle, cell proliferation, migration and invasion, and inhibits cell apoptosis in vitro. The in vivo study showed that loss of CENPM inhibits the growth of A549 xenografts. Furthermore, we found that CENPM can promote the phosphorylation of mTOR rather than directly affect the mTOR content. Inhibition of mTOR activity abrogates the promoting effects of CENPM on cell cycle progression, cell proliferation, migration and invasion. Taken together, these results show that CENPM plays an important role in the growth and metastasis of LUAD and may be a promising therapeutic target in LUAD.

NaNbO3-based lead-free energy storage ceramics are highlighted as essential components for the next–generation pulsed power capacitors, especially in the realm of eco-friendly renewable energy sources. Nevertheless, their large electric hysteresis loss and low breakdown strength (Eb) lead to difficulties in simultaneously achieving high recoverable energy density (Wrec) and efficiency (η), severely hindering their further development. Herein, a multistep synergistic optimization strategy is proposed to balance Wrec and η in NaNbO3-based ceramics. The addition of CaZrO3 (CZ) and Bi2/3HfO3 (BH) into NaNbO3 ceramic generates a stabilized antiferroelectric phase, highly dynamic polar nanoregions, refined grains, and increased band-gap, resulting in delayed polarization saturation and ultrahigh Eb. With the deliberate multistep synergistic route, the 0.85NNCZ–0.15BH ceramics attain an extraordinary Wrec of 9.2 J cm−3 and high η of 82.5% under 920 kV cm−1, accompanied by superior stability of temperature, frequency, and fatigue resistance. The novel and easily applicable multistep synergistic optimization strategy presented in this work could be extended to other high-performance ceramic systems, paving a new and effective route for the development of high-performance capacitors. The SmFLS gene was cloned from eggplant and has an ORF of 1014 bp encoding 337 amino acids. The deduced protein sequence of SmFLS was 88.07% and 84.94% identical to homologs encoded by StFLS in Solanum tuberosum and SlFLS in S. lycopersicum, respectively. SmFLS contains typical DIOX_N and 2OG-Fe (II)_Oxy functional domains, as well as five strictly conserved amino acid residues (H223, D225, H279, [R.sup.2]89, and S291) related to FLS enzyme activity. Phylogenetic tree analysis showed that SmFLS had the closest genetic relationship with the FLS genes in potato and tomato. At a high temperature of 35 [degrees]C, the expression level of SmFLS was higher than that of the control in the same period, and it reached extremely significant levels on 15DAF and 20DAF, at which the eggplant peel color became lighter accordingly. Upon overexpression of SmFLS in eggplant, the flavonol content of transformed plants was significantly higher than that of untransformed plants, and the peel color was lighter than that of the control. The results indicate that SmFLS negatively regulates eggplant peel coloration under high temperature. Key words: eggplant, FLS, flavonol, peel coloring, high temperature. SmFLS contient des domaines fonctionnels typiques [DIOX_N et 2OG-Fe (II)_Oxy] ainsi que cinq acides amines strictement conserves (H223, D225, H279, [R.sup.2]89 et S291) qui sont impliques dans l'activite enzymatique FLS. Une analyse phylogenetique a montre que SmFLS est plus proche des FLS chez la pomme de terre et la tomate. A une temperature elevee (35 [degrees]C), le niveau d'expression de SmFLS etait superieur a celui chez les plantes temoins au meme stade et il a atteint des niveaux tres eleves a 15DAF et 20DAF, un stade auquel la peau de l'aubergine a pali. Lors de la sur-expression de SmFLS chez l'aubergine, le contenu en flavonols chez les plantes transformees etait significativement plus eleve que chez les temoins non-transformes et la couleur de la peau etait plus claire que chez les temoins. Ces resultats indiquent que SmFLS regule negativement la coloration de la peau de l'aubergine en conditions chaudes.
