In the last two decades, forensic pathology and crime scene investigations have seen a rapid increase in examination tools due to the implementation of several imaging techniques, e.g., CT and MR scanning, surface scanning and photogrammetry. These tools encompass relatively simple visualization tools to powerful instruments for performing virtual 3D crime scene reconstructions. A multi-modality and multiscale approach to a crime scene, where 3D models of victims and the crime scene are combined, offers several advantages. A permanent documentation of all evidence in a single 3D environment can be used during the investigation phases (e.g., for testing hypotheses) or during the court procedures (e.g., to visualize the scene and the victim in a more intuitive manner). Advanced computational approaches to understand what might have happened during a crime can also be applied by, e.g., performing a virtual animation of the victim in the actual context, which can provide important information about possible dynamics during the event. Here, we present an overview of the different techniques and modalities used in forensic pathology in conjunction with crime scene investigations. Based on our experiences, the advantages and challenges of an image-based multi-modality approach will be discussed, including how their use may introduce new visualization modalities in court, e.g., virtual reality (VR) and 3D printing. Clinical forensic medical examinations constitute an increasing proportion of our institution's tasks, and, concomitantly, the authorities are now requesting forensic life-threatening danger assessments based on our examinations. The aim of this retrospective study was to assess if a probability of survival (PS) trauma score could be useful for these forensic life-threatening danger assessments and to identify a cut-off PS score as a supporting tool for the forensic practice of assessing life-threatening danger. We compared a forensic database and a trauma database and identified 161 individuals (aged 15 years or older) who had both a forensic life-threatening danger assessment and a PS score. The life-threatening danger assessments comprised the following statements: was not in life-threatening danger (NLD); could have been in life-threatening danger (CLD); or was in life-threatening danger (LD). The inclusion period was 2012–2016. A statistically significant difference was found in the PS scores between NLD, CLD and LD (chi-square test: p < 0.0001). The usefulness of the PS score for categorizing life-threatening danger assessments was determined by a receiver-operator characteristic (ROC) curve. The area under the curve was 0.76 (95% CI, 0.69 to 0.84) and the ROC curve revealed that a cut-off PS score of 95.8 would appropriately identify LD. Therefore, a PS score below 95.8 would indicate life-threatening danger. We propose a further exploration of how the evidence-based PS score, including a cut-off value, might be implemented in clinical forensic medical statements to add to the scientific strength of these statements. Age assessment of migrants is crucial, particularly for unaccompanied foreign minors, a population facing legal, social, and humanitarian challenges. Despite existing guidelines, there is no unified protocol in Europe for age assessment. The Forensic Anthropology Society of Europe (FASE) conducted a comprehensive questionnaire to understand age estimation practices in Europe. The questionnaire had sections focusing on the professional background of respondents, annual assessment numbers, requesting parties and reasons, types of examinations conducted (e.g., physical, radiological), followed protocols, age estimation methods, and questions on how age estimates are reported. The questionnaire's findings reveal extensive engagement of the forensic community in age assessment in the living, emphasizing multidisciplinary approaches. However, there seems to be an incomplete appreciation of AGFAD guidelines. Commonalities exist in examination methodologies and imaging tests. However, discrepancies emerged among respondents regarding sexual maturity assessment and reporting assessment results. Given the increasing importance of age assessment, especially for migrant child protection, the study stresses the need for a unified protocol across European countries. This can only be achieved if EU Member States wholeheartedly embrace the fundamental principles outlined in EU Directives and conduct medical age assessments aligned with recognized standards such as the AGFAD guidelines. Fluctuating asymmetry are random deviations of an otherwise symmetrical body plan and arises from instability in development. Earlier studies suggest that levels of cranial fluctuating asymmetry may be influenced by lifestyle and quality of life in a population. It may, therefore, be useful as a stress indicator. We investigated whether cranial fluctuating asymmetry has changed in archaeological Danish populations over time, and between grave sites from the same time period. Our sample consisted of 219 adult individuals from the Neolithic Age (approx. 3000BC) to the Early Modern Age (approx. 1850). We collected 27 3-dimensional landmarks from the face, calvarium, and base of the cranium. Levels of shape variation were analyzed using Procrustes analysis of variance and principal component analysis. Cemeteries, time periods, and sex were compared using linear mixed models, one-way analysis of variance, and Kruskal-Wallis test. We found no statistically significant differences in cranial FA between grave sites from the same time period, nor did we find any statistically significant difference between time periods. We found that sex did not have an influence on levels of cranial FA. We found no measurable difference in levels of cranial FA between Danish populations over time. Further knowledge on genetics and other stress indicators in our sample may give more insight into the relationship between cranial fluctuating asymmetry and developmental instability. The most common method to predict nasal projection for forensic facial approximation is Gerasimov's two-tangent method. Ullrich H, Stephan CN ( J Forensic Sci, 2011; 56: 470) argued that the method has not being properly implemented and a revised interpretation was proposed. The aim of this study was to compare the accuracy of both versions using a sample of 66 postmortem cranial CT data. The true nasal tip was defined using pronasale and nasal spine line, as it was not originally specified by Gerasimov. The original guidelines were found to be highly inaccurate with the position of the nasal tip being overestimated by c. 2 cm. Despite the revised interpretation consistently resulting in smaller distance from true nasal tip, the method was not statistically accurate ( p > 0.05) in positioning the tip of the nose (absolute distance >5 mm). These results support that Gerasimov's method was not properly performed, and Ullrich H, Stephan CN ( J Forensic Sci, 2011; 56: 470) interpretation should be used instead. [ABSTRACT FROM AUTHOR]

Reports by minors of sexual relations against their will are not rare, and in some cases pornographic photography is a part of the abuse. Such material can be used to help identify the perpetrator. In this study we examined the efficacy of visual comparison between two databases. One consisted of blinded still images of the back of the right hand of 51 males from video recordings, the other of blinded high quality camera images of the back of the right hand from the same 51 individuals. The images were compared in pairs (2601 combinations) and a judgment was made about whether they were a highly likely, possible or unlikely match, using several anatomical features as markers for comparison. Using this method all 51 high quality images were correctly matched with the video image from the same person, though in some cases there was up to 5 other samples that could not be excluded as possible matches. In total there were no false positive 'highly possible' matches, but there were 50 false positive 'possible' matches. Visual comparison of the back of the hand is a valuable addition to the burden of evidence in a judicial setting, but should not be used as a standalone method to establish proof of identity. The applicability of the method is dependent on the existence of a database of images; the degree of certainty of the identification is directly correlated to the size of the database. This study examines what in Denmark may constitute evidence based on forensic anthropological gait analyses, in the sense of pointing to a match (or not) between a perpetrator and a suspect, based on video and photographic imagery. Gait and anthropometric measures can be used when direct facial comparison is not possible because of perpetrators masking their faces. The nature of judicial and natural scientific forms of evidence is discussed, and rulings dealing with the admissibility of video footage and forensic evidence in general are given. Technical issues of video materials are discussed, and the study also discusses how such evidence may be presented, both in written statements and in court. [ABSTRACT FROM AUTHOR]

Fatal lawn mower related injuries are a relatively rare occurrence. In a forensic setting, the primary aim is to reconstruct the injury mechanism and establish the cause of death. A relatively rare, but characteristic type of injury is a so-called projectile or missile injury. This occurs when the operator or a bystander is impacted by an object mobilized from the grass by the rotating mower blades. This type of injury often leaves only modest external trauma, which increases the risk of overlooking an entry wound. In this paper we present a case of a fatal lawn mower related projectile injury which was initially overlooked, later interpreted as a possible gunshot homicide, and finally identified as a lawn mower related projectile injury when autopsy revealed a piece of metal thread in the main bronchus to the right middle lobe, hemopericardium, and right-sided hemothorax. To our knowledge, this injury mechanism has not previously been reported as a cause of death. This case illustrates the importance of postmortem radiological imaging and interdisciplinary cooperation when establishing manner and cause of death in unusual cases. [ABSTRACT FROM AUTHOR]

Abstract: Weight but occasionally also the volume of cremated human remains may often be the primary basis for interpreting the many stages involved in past cremation ceremonies. However, methods used for describing volume for cremated remains are extremely varying and biased by many factors. Here we evaluate different methods for calculating and describing volume and propose a method for estimating the original prehistoric post-cremation weight. Our data suggests that low cremation weights reported for archaeologically recovered cremated remains are markedly underestimated, and that whole cremated bodies probably are represented more often than discussed in osteoarchaeology. Hence, this contests many suppositions on ritual selection of cremated human remains in prehistoric and early historical archaeology. This article presents the results of a validation study of a previously published method of sex determination from the temporal bone. The purpose of this study was to evaluate the lateral angle method for the internal acoustic canal for accurately determining the sex of human skeletal remains using measurements taken from computed tomography ( CT) scans. Previous reports have observed that the lateral angle size in females is significantly larger than in males. The method was applied to an independent series of 77 postmortem CT scans (42 males, 35 females) to validate its accuracy and reliability. The mean lateral angle of the internal acoustic canal was found to be larger in females (46.5°) than in males (43.4°). However, the difference was not statistically significant and the sex differences reported in previous studies were not substantiated. Abstract: Based on an actual case, where we were able to ascertain the year of birth of three dead babies found in a deep-freezer to within 1–2 years (1986, 1988 and 2004, respectively), we review the current state of forensic age determination/year of birth determination. The age of an individual (year of birth) is often a fundamental piece of data in connection with forensic identification of unidentified bodies. The methods most often used are based on determining various morphological, age-related, changes on the skeleton (or teeth, although odontological methods are not reviewed in this paper). As such, these methods are all relative, i.e. they do not furnish calendar ages or years, but an estimate of the age at death, with a rather large range, i.e. the methods rely on biological aging following the chronological aging. More recently, methods have been proposed using more direct ascertainment of age at death, e.g. protein racemisation, or, as in our case, radiocarbon methods. Especially the latter method may in fact yield absolute ages (years of birth), because 14C activity, as measured in specific proteins in specific cells or tissues in the body, were in equilibrium with the so-called bomb-pulse, when these proteins were formed (at birth). The bomb pulse reflects a dramatic change in atmospheric 14C content due to nuclear bomb testing, and these dramatic changes can be rather tightly related to single calendar years. Specifically, the presence of soft tissues may expand the scope of paleopathological studies. Many recent mummy studies have focused on the development and application of non-destructive methods for examining mummies, especially radiography and CT scanning with advanced 3D visualizations. Indeed, the development of commercially available CT scanners in the 1970s meant that for the first time the 3D internal structure of mummies and bog bodies could be studied non-destructively. This article describes the history of mummy radiography and CT scanning, and some of the problems and opportunities involved in applying these techniques, derived for clinical use, on naturally and artificially preserved ancient human bodies. Unless severely degraded, bone is quite readily visualized, but accurate imaging of preserved soft tissues, and pathological lesions therein, may require considerable post-image capture processing of CT data. Unbiased selection of regions of interest (ROIs) and unbiased definition of histological structures are needed to improve the repeatability of microscopic methods for age at death determination and to reduce operator subjectivity. We present results obtained by selecting ROIs according to stereological principles on a sample of 28 femoral cross sections of Caucasoid males aged 20-89 years. A regular grid was overlaid on the cross section, and the ROIs were selected as close as possible to the periosteum in the anterior, lateral, and medial regions. The areas consisting of all intact secondary osteons plus fragments were outlined and osteon population density, percent osteon population, area, and perimeter were calculated using stereological methods and software. Overall, the analyses of intra- and inter-section variability showed no significant difference between the ROIs, i.e., the location within the cross section of the ROIs does not affect the outcome of the analyses. Using established criteria for work with fossil DNA we have analysed mitochondrial DNA from 92 individuals from 18 locations in Denmark ranging in time from the Mesolithic to the Medieval Age. Unequivocal assignment of mtDNA haplotypes was possible for 56 of the ancient individuals; however, the success rate varied substantially between sites; the highest rates were obtained with untouched, freshly excavated material, whereas heavy handling, archeological preservation and storage for many years influenced the ability to obtain authentic endogenic DNA. While the nucleotide diversity at two locations was similar to that among extant Danes, the diversity at four sites was considerably higher. This supports previous observations for ancient Britons. The overall occurrence of haplogroups did not deviate from extant Scandinavians, however, haplogroup I was significantly more frequent among the ancient Danes (average 13%) than among extant Danes and Scandinavians (∼2.5%) as well as among other ancient population samples reported. Haplogroup I could therefore have been an ancient Southern Scandinavian type ''diluted'' by later immigration events. Interestingly, the two Neolithic samples (4,200 YBP, Bell Beaker culture) that were typed were haplogroup U4 and U5a, respectively, and the single Bronze Age sample (3,300-3,500 YBP) was haplogroup U4. These two haplogroups have been associated with the Mesolithic populations of Central and Northern Europe. Therefore, at least for Southern Scandinavia, our findings do not support a possible replacement of a haplogroup U dominated hunter-gatherer population by a more haplogroup diverse Neolithic Culture. Abstract: Computed tomography (CT) has been used routinely at the Department of Forensic Medicine, University of Copenhagen since 2002. A retrospective study was performed in order to correlate CT-scan based diagnoses of cranial and cerebral lesions with macroscopic autopsy diagnoses in 56 cases. The CT-scans were performed by a forensic pathologist. They were obtained by using two different CT-scan protocols. The results showed correct skull fracture diagnoses in 34/56 cases. Fractures were diagnosed partially (9) or missed totally (13) on CT-images in 22 cases. The agreement for fracture diagnoses of the anterior, medial and posterior cranial fossae was 20%, 52% and 60%, respectively. Fractures involving bilateral bones were diagnosed correctly more frequently. The diagnostic agreement regarding brain injuries varied from 0% to 79%. Both the autopsy-reports and CT-scan descriptions need to be standardized in order to secure more exact comparisons in the future. [Copyright &y& Elsevier]

The article presents a study which explored mummification and gave a presentation of various key mummy finds. The different ways of natural mummification include dessication, freezing and bog bodies, among other others. While artificial indicates that mummification is the result of intentional human activity. Other topics discussed include a brief history of mummy studies, methods in mummy research that include mummy autopsy and endoscopy, and relationship between mummies and paleopathology.

A police case with a strangulated woman with fingermarks on the neck and two suspects identifying each other as the perpetrator set off a laboratory experiment. Twenty-one males participated in the study. Blue paint was applied to their fingers, after which they grasped a neck dummy and pressed hard as if strangulating someone. The imprint was removed from the dummy, and their hands were photographed. Five imprints were randomly chosen and superimposed on the hand photographs in blind trials. In no cases did we match an imprint to the correct hand. However, in four cases we matched the imprint with several hands, one of which was the correct one. This means we were able to exclude nonmatches in 4/5 cases. Overall, matching of hands and fingermarks is difficult and inconclusive. Objective criteria for matching are difficult to establish, and matching is probably best suited for cases with specific anatomical features. [ABSTRACT FROM AUTHOR]

We report on the results of applying the so-called lateral angle method for sex determination on skeletal remains. The lateral angle denotes the angle of the internal auditory canal in relation to the medial surface of the petrous part of the temporal bone. The method involves making a small cast of the proximal part of the internal acoustic canal and determining the angle at which the canal opens up to the surface of the petrous bone. The method has the great advantage of utilizing one of the sturdiest bone elements of the human skeleton, and may thus be especially suited for analyses of very fragmented skeletal remains or cremated bones, where the petrous bone may still be readily recognizable. The method was tested using a forensic sample of 113 petrous bones with known sex. Intra- and interobserver testing was also performed. We found a statistically significant difference in angle size between males and females (mean angle size of males, 39.3°; mean angle size of females, 48.2°; P < 0.001). There was no bilateral difference in angle size. In blind trials, 83.2% of petrous bones were assigned to the correct sex. We also tested the lateral angle method against an archaeological skeletal sample. True sex was not known for this sample; instead, sexing had been carried out by assessing pelvic and cranial morphology in independent trials. We found a higher concordance between the lateral angle and "pelvic" sex than for lateral angle and "cranial" sex. Finally, we note that subadult sexing may also be possible with this method. Background: Earlier studies have addressed the human total cranial vault thickness and generally found no correlation with sex, age or body weight. However, the thickness of the diploe has not been investigated. Our study has determined the diploeic thickness of the human cranial vault using modern autopsy material. Methods: The diploeic bone thickness was measured in 64 individuals (43 males, 21 females) autopsied at our institute. The thickness was measured by X-raying biopsies trephined at four specific locations on the skull. Complete medical records and pathologic autopsy results were available. Results: There was a statistically significant difference in diploeic thickness between males and females in the frontal region only. Diploeic thickness was highly correlated with total cranial vault bone thickness, except for the left euryon in females. Subsequent analyses failed to reveal any correlations between the diploeic thickness and age and height and weight of the individual. Conclusion: Males overall have a thicker diploe, albeit this difference is statistically significant only in the frontal region. We could not discern any trends as pertains to diploeic thickness versus age, height or weight. Since the thickness of the diploe may be an important parameter in biomechanical modelling of the cranial vault, this means that the diploe can be built into such models based on the total cranial thickness, except for the frontal region where the sexual dimorphism must be taken into account. Our findings are consistent with previous studies relating the total cranial thickness to the same parameters, in that we found a high correlation between diploeic and total cranial thickness (except at the left euryon for females). Finally, we recommend that future studies try to incorporate CT or MR scan imaging, rather than point sampling, in order to achieve a total assessment of the dimensionalities of the diploe. Skeletal remains of four Dorset Palaeo-Eskimo individuals were found at Alarnerk (NhHd-1), Melville Peninsula, in 1954 by the joint Danish National Museum-University of Pennsylvania Expedition, which was excavating Dorset and preDorset sites. These remains included one complete mandible, two fragmentary mandibles, and a cranial fragment. One of the mandibular fragments, found in a grave, was from a child approximately nine months old. The other remains were found in middens at the site. These remains add to the otherwise very sparse Dorset human material. Analysis of the dental morphology, as well as morphometric analysis of the complete mandible, shows Eskimoid characters and resemblance with later period Greenlandic human skeletal remains. We present the results of a preliminary study on the use of 3-D software (Photomodeler®) for identification purposes. Perpetrators may be photographed or filmed by surveillance systems. The police may wish to have these images compared to photographs of suspects. The surveillance imagery will often consist of many images of the same person taken from different angles. We wanted to see if it was possible to combine such a suite of images in useful 3-D renderings of facial proportions.Fifteen male adults were photographed from four different angles. Based on these photographs, a 3-D wireframe model was produced by Photomodeler®. The wireframe models were then rotated to full lateral and frontal views, and compared to like sets of photographs of the subjects. In blind trials, 9/15 of the wireframe models were assigned to the correct sets of photographs. In five/15 cases, the wireframe models were assigned to several sets, including the correct set. Only in one case was a wireframe model not assigned to a correct set of photographs at all. [Copyright &y& Elsevier]

The fractal dimensions of human sagittal and coronal sutures were calculated on 31 complete skulls from the Terry Collection. The aim was to investigate whether the fractal dimension, relying on the whole sutural length, might yield a better description of age-related changes in sutural morphology, as opposed to other methods of quantification, which generally rely on more arbitrary scoring systems. However, the fractal dimension did not yield better age correlations than other previously described methods. At best, the results reflected the general observation that young adults below age 40 years display an age-related development, but that it is impossible to arrive at any precise age determinations for older adults. It seems that for some individuals, suture obliteration simply does not take place, even at an advanced age, whereas for others, suture obliteration progresses rapidly. Until a better understanding of sutural biology is reached, this will render cranial sutures only marginally useful in age determination. It is believed by many that reference data for age estimation purposes must be imaging-modality specific. A study from our department has however proven otherwise. We therefore found it interesting to investigate this further by looking at the level of agreement between different imaging modalities. The aim of this study was to investigate the level of agreement between the three radiological modalities, computed tomography (CT), magnetic resonance imaging (MRI), and digital radiography (DR), in assessing the ossification of the epiphyses of the knee. A total of 34 deceased individuals of 10–25 years of age, brought in for a medicolegal autopsy at our department, were scanned by CT, MRI, and DR. The ossification stages of the three bones of the right knee, distal femoral, proximal tibial, and proximal fibular epiphysis were assessed using the established combined staging method by Schmeling et al. and Kellinghaus et al. Analysis of the results by Cohen's weighted kappa showed a good agreement between CT and DR (K = 0.61–0.70), and MRI and DR (K = 0.68–0.79) but only moderate agreement between CT and MRI (K = 0.55–0.57). This leads us to conclude that different radiological images cannot be used interchangeably for age estimation purposes, so reference material needs to be imaging-modality specific. Background. Lens crystallines are special proteins in the eye lens. Because the epithelial basement membrane (lens capsule) completely encloses the lens, desquamation of aging cells is impossible, and due to the complete absence of blood vessels or transport of metabolites in this area, there is no subsequent remodelling of these fibers, nor removal of degraded lens fibers. Human tissue ultimately derives its 14C content from the atmospheric carbon dioxide. The 14C content of the lens proteins thus reflects the atmospheric content of 14C when the lens crystallines were formed. Precise radiocarbon dating is made possible by comparing the 14C content of the lens crystallines to the so-called bomb pulse, i.e. a plot of the atmospheric 14C content since the Second World War, when there was a significant increase due to nuclear-bomb testing. Since the change in concentration is significant even on a yearly basis this allows very accurate dating. Methodology/Principal Findings. Our results allow us to conclude that the crystalline formation in the lens nucleus almost entirely takes place around the time of birth, with a very small, and decreasing, continuous formation throughout life. The close relationship may be further expressed as a mathematical model, which takes into account the timing of the crystalline formation. Conclusions/Significance. Such a lifelong permanence of human tissue has hitherto only been described for dental enamel. In confront to dental enamel it must be held in mind that the eye lens is a soft structure, subjected to almost continuous deformation, due to lens accommodation, yet its most important constituent, the lens crystalline, is never subject to turnover or remodelling once formed. The determination of the 14C content of various tissues may be used to assess turnover rates and degree of substitution (for example for brain cell DNA). Potential targets may be nervous tissues in terms of senile or pre-senile degradation, as well as other highly specialised structures of the eyes. Severe mental illness (SMI) is associated with a reduced life expectancy of up to 20 years. One possible contributor to this fact is dysregulation of the hypothalamus-pituitary-adrenal (HPA)-axis. Looking at the morphology of effector organs, such as the adrenal glands themselves, could reveal insights into organ function and response to possible HPA-dysregulation. This forensic autopsy-based study investigated if there were any morphological changes in adrenal glands between decedents who had previously been submitted to a psychiatric hospital with a diagnosis of schizophrenia (n = 34), bipolar (n = 5), or depressive disorder (n = 20), any other psychiatric diagnosis (n = 36) compared with decedents who had no previous psychiatric admission (n = 40). Length of admissions to psychiatric wards and admission in the 180 days preceding death was included in regression as proxy variables for severity of illness. On the macroscopic level, we found no difference in gland weight or volume. On the microscopic level, we found a 25% increase in cross-sectional area of the zona fasciculata (ZF) in decedents who had a diagnosis of schizophrenia compared with controls (p = 0.033). Other diagnosis groups did not differ from controls. Total admission length was positively correlated with area of the ZF. Lay Summary People with a severe mental disorder may be in a constant state of increased stress, which is harmful. This study looked at the adrenal gland, which produces stress hormones, to see if they were different in deceased persons who had suffered from a severe mental illness. We found that the part of the adrenal gland that produces stress hormones is larger in deceased patients who suffered from schizophrenia, but not other types of psychiatric illnesses, compared to deceased persons with no history of psychiatric illness. Age at death estimation of human skeletal remains is one of the key issues in constructing a biological profile both in forensic and archaeological contexts. The traditional adult osteological methods evaluate macroscopically the morphological changes that occur with increasing age of specific skeletal indicators, such as the cranial sutures, the pubic bone, the auricular surface of the ilium and the sternal end of the ribs. Technologies such as computed tomography and laser scanning are becoming more widely used in anthropology, and several new methods have been developed. This review focuses on how the osteological age‐related changes have been evaluated in digital data. First, the 3D virtual copies of the bones have been used to mimic the appearance of the dry bones and the application of the traditional methods. Secondly, the information directly extrapolated from CT scans has been used to assess qualitatively or quantitatively the changes of the trabecular bones, the thickness of the cortical bones, and to perform morphometric analyses. Lastly, the most innovative approach has been the mathematical quantification of the changes of the pelvic joints, calculating the complexity of the surface. Accurate and precise documentation of lesions is an important aspect of the forensic pathologists' work. Photogrammetry provides a useful tool to take precise measurements from photographs. These photographs are normally acquired with single camera photographs, but the procedure is quite time-consuming. Video recording has the potential to record a larger amount of image data faster. We documented 33 cadaveric skin lesions, using photographs and video recordings. The dimensions of the lesions ranged between 0.27 and 21.8 cm. The measurements of the lesions were extracted with both manual and automatic point measurements from photographs and from video frames, respectively. Very small differences (mean and median < 1 mm) were found between measurements taken in photographs versus video frames. Video frames were often blurred, preventing clear demarcation of the edges of the lesions and presenting a larger amount of noise in the 3D models. The differences between the manual point and automatic point measurements were very small (mean and median < 1 mm), but the manual procedure is to be preferred, since automatic points were not always located on the edges of the lesions. The only aspect in which video frames were superior to photographs was the recording time: video recording was almost five times faster than the photo sessions. In conclusion, this study shows that precise and comparable measurements can be extracted both from photographs and video frames. The exhumation of Danish astronomer Tycho Brahe (1546–1601) was performed in 2010 to verify speculative views on the cause of his death. Previous analyses of skeletal and hair remains recovered from his grave refuted the presumption that he died from poisoning. These studies also outlined the possibility that he actually died from an acute illness, echoing the rather vague and inaccurate testimony of some historical records. We performed a detailed paleopathological analysis of Tycho Brahe’s skeletal remains, along with a reconstruction of his diet based on carbon and nitrogen stable isotopes analysis and an estimate of his physical status (relative body fat) based on medullar and cortical dimensions of the femoral shaft. The astronomer’s remains exhibit bone changes indicative of diffuse idiopathic skeletal hyperostosis (DISH). The study further allows us to classify him as obese (100% reliability according to our decision tree designed from Danish males), and points out his rich diet (high input of animal protein and/or marine resources) and high social status. Comorbidities of DISH and obesity are reviewed, and their influence on health status is discussed. We further consider some conditions associated with metabolic syndrome as possible causes of Tycho Brahe’s final symptoms (urinary retention, renal failure and coma), including diabetes, alcoholic ketoacidosis and benign prostatic hypertrophy. Although a definite and specific diagnosis cannot be established, our study points to today’s civilization diseases often associated with DISH and metabolic syndrome as the possible cause of death of Tycho Brahe. Simple Summary: Leprosy, a chronic infectious disease, leads to blood mineral imbalances: low levels of zinc, calcium, magnesium, and iron and high levels of copper. Interestingly, in late medieval Europe, minerals were used to treat leprosy. We investigated physiological responses to leprosy and possible evidence of treatment in dental tissues of leprosy sufferers from medieval Denmark and early 20th century Romania when multidrug therapy was not then yet invented. Using Synchrotron Fluorescence (SXRF) and laser ablation (LA-ICP-TOFMS), we show marked covariations in the zinc, calcium, and magnesium distributions, which are compatible with clinical studies but cannot be directly attributed to leprosy. Minerals used historically as a treatment for leprosy show no detectable intake (arsenic, mercury) or a diffuse distribution (lead) related to the daily consumption of contaminated water and food. Intense lead enrichments indicate acute incorporations, potentially through the administration of lead-enriched medication or the mobilization of lead from bone stores to the bloodstream during intense physiological stress related to leprosy. However, comparisons with a healthy control group are needed to ascertain these interpretations. The positive correlations and the patterns observed between lead and essential elements may indicate underlying pathophysiological conditions, demonstrating the potential of the two techniques for investigating diseases in past populations. Leprosy can lead to blood depletion in Zn, Ca, Mg, and Fe and blood enrichment in Cu. In late medieval Europe, minerals were used to treat leprosy. Using SXRF and LA-ICP-TOFMS, 12 elements were mapped in 15 tooth thin sections, and the statistical covariation of paired elements was computed to assess their biological relevance. The results show marked covariations in the Zn, Ca, and Mg distributions, which are compatible with clinical studies but cannot be directly attributed to leprosy. Abstract: The microscopic method of age at death determination was introduced by Kerley in 1965 [E.R. Kerley, The microscopic determination of age in human bone, Am. J. Phys. Anthropol, 23 (1965) 149–163.]. However, even though the method has been revised several times, there remain some fundamental issues concerning the reliability of the methods. This is because several basic histological features seen in a cross section of a bone have to be quantified according to the method, but the definition, and hence quantification, of these features leaves room for subjectivity. In a previous study we found that some of these features (osteon fragments and Haversian canals) could not be identified reliably. Only secondary osteons could be identified with a low inter and intra observer error. Furthermore, since the histological features are quantified in only parts of an entire bone cross section, the selection of these parts or areas is a potential source of bias. Finally, unless an unbiased method is used for addressing features on the borders of the selected areas, this will also introduce error. These issues have not been addressed specifically in previous studies. In this study, we used the methods of stereology to choose the regions of interest, as well as for dealing with border phenomena, and we only counted secondary osteons. Our results show a statistically significant increase in the median number of osteons per area unit with increasing age at death. However, this was after exclusion of one outlier. This result is probably due to the limited sample size (N =24). As such, this study is preliminary, but does warrant applying the described techniques to a larger sample. [Copyright &y& Elsevier]

Background: The resulting statement of forensic age estimations in the living is of great consequence for the examinees in question. Concerns have been raised about the nature of these examinations, e.g. the potentially harmful radiographic exposures, as well as the accuracy of the estimated age. Aim: This manuscript investigates the reported age estimates performed at the Section of Forensic Pathology, Institute of Forensic Medicine, Copenhagen, in 2012. Subjects and methods: The results of the individual assessments, i.e. the physical examination, and the radiographic examinations of bone age and dental age, were investigated as well as the correlation between the age estimates of the two latter examinations. Results: In 139 out of the 174 statements, a conclusive chronological age estimate of at least 18 years was given. Although bone age was assessed within the dental age-range in a total of 152 out of 162 cases, some variations in the combined assessments were seen. Conclusion: The current procedure is not mathematically or statistically satisfactory. Instead, the most likely age including the variation and error of the age estimate should be reported in statements. The introduction of a Bayesian approach and the use of post-mortem-imaging-based databases may be used for this purpose. [ABSTRACT FROM PUBLISHER]

We examined the utility of CT scans in the evaluation of degree of ectocranial suture closure. Five cranial points (left and right midcoronal positions, left and right midlambdoidal positions, and the lambda) were evaluated in 231 CT scans using a three-point scoring system (open, partial closed, and closed). The slice increment and thickness varied between three groups of the scans. The results showed a good correlation between degree of suture closure and increasing age in each group. Young individuals (<40 years) and old individuals (>60 years) could be clearly distinguished from the middle-aged individuals. ANOVA test revealed no difference between two groups of scans and between left and right sides ( p > 0.05). Interobserver agreement was good, especially considering the score by sides. Background:An introduction to a forensic medical homicide database established at the Department of Forensic Medicine in Copenhagen. Content: The database contains substantial clinical and demographic data obtained in conjunction with medico-legal autopsies of victims and forensic clinical examinations of perpetrators in homicide cases in eastern Denmark. Validity and coverage: The database contains information on all homicide cases investigated at the Department of Forensic Medicine in Copenhagen since 1971. Coverage for the catchment area of the department is assumed to be very good because of a medico-legal homicide autopsy rate close to 100%. Regional differences might exist however, due to the fact that the catchment area of the department is dominated by the city of Copenhagen. Conclusions: The strength of the database includes a long running time, near complete regional coverage and an exhaustive list of registered variables. Carbon monoxide (CO) inhalation is a well-known method of committing suicide. There has been a drastic increase in suicide by inhalation of CO, produced from burning charcoal, in some parts of Asia, and a few studies have reported an increased number of these deaths in Europe. CO-related deaths caused by charcoal burning have, to our knowledge, not been recorded in the Danish population before. In this retrospective study we present all autopsied cases of CO poisoning caused by charcoal burning in the period 2008-2012. 19 autopsied cases were identified, comprising 11 suicides, 4 accidents, and 2 cases of maternal/paternal filicide-suicide. The mean age of decedents was 38.2 years and the majority of the decedents were men. In 16 cases carboxyhemoglobin levels were above 50 % and in 14 cases we found distinctive cherry red livor mortis. Various concentrations of ethanol and drugs were found in 9 cases. Data suggest that this method of death has increased significantly in Denmark. Therefore, it is highly relevant to draw attention to the subject, to increase awareness as well as prevent future escalation. [ABSTRACT FROM AUTHOR]

In this pilot study, the authors tested whether photogrammetry can replace or supplement physical measurements made during autopsies and, based on such measurements, whether virtual computer models may be applicable in forensic reconstructions. Photogrammetric and physical measurements of markers denoting wounds on five volunteers were compared. Virtual models of the volunteers were made, and the precision of the markers' locations on the models was tested. Twelve of 13 mean differences between photogrammetric and physical measurements were below 1 cm, which indicates that the photogrammetric method has a high accuracy. The precision of the markers' location on the models was somewhat less, although the method is still promising and potentially superior to the current procedures used for reconstructions. The possibility to measure any distance on a body, even after the autopsy is concluded and the corpse is no longer available, is one of the biggest benefits of photogrammetry. [ABSTRACT FROM AUTHOR]

A systematic dietary investigation during Danish Roman Iron Age (1-375AD) is conducted by analyzing stable isotope ratios of carbon (δ13C) and nitrogen (δ15N) in the collagen of human and animal bone. The human sample comprises 77 individuals from 10 burial sites. In addition 31 samples of mammals and fish were analyzed from same geographical area. The investigation characterizes the human diet among different social groupings and analyses dietary differences present between sex, age, and site phase groups. Diachronically, the study investigates the Roman influences that had an effect on social structure and subsistence economy in both the Early and Late Period. Geographically the locations are both inland and coastal. The isotopic data indicate extremely uniform diet both between and within population groups from Early and Late Roman periods and the data are consistent throughout the Roman Iron Age. Protein consumption was dominated by terrestrial animals with no differences among social status, age, sex, or time period, while terrestrial plant protein only seems to have contributed little in the diet. Furthermore, the consumption of marine or aquatic resources does not seem to have been important, even among the individuals living next to the coast. [ABSTRACT FROM AUTHOR]

Intraskeletal variation in the composition of carbon (δ13C) and nitrogen (δ15N) stable isotopes measured in collagen is tested from various human bones and dentine. Samples were taken from the femur, rib, and petrous part of the temporal bone from well-preserved skeletons of both adults (n = 34) and sub-adults (n = 24). Additional samples of dentine from the root of 1st molars were taken from 16 individuals. The skeletal material is from a medieval cemetery (AD 1200-1573) in Holbæk, Denmark. Our results indicate that the petrous bone has an isotopic signal that differs significantly from that of femur and rib within the single skeleton (P < 0.001 and P < 0.001, respectively), with only minor variation seen between femur and rib. On the other hand, there was no significant difference between the petrous bone and the 1st molar. The intraskeletal variation may reflect differences in turnover rate among skeletal elements. The inner periosteal layer of the petrous bone is formed in uterus and does not undergo any further remodelling after the age of 2 years, whereas the rib and femur have a continuous turnover rate of ~5 and 10-20 years, respectively. From the results of this study it is believed the petrous bone may be a new useful bone element and a supplement or a proxy for teeth in the analysis of early dietary patterns as it may reflect diet in fetal stages and early years of life. Recordings from video surveillance systems are used as evidence from crime scenes. It would be useful to perform comparisons between disguised perpetrators and suspects based on their gait. We applied functional anatomical and biomechanical knowledge to analyze the gait of perpetrators, as recorded on surveillance video. Using a structured checklist, which addresses the single body segments during gait, we were able to give a statement concerning the gait patterns. Characteristic parameters were, e.g., varus instability in the knee at heel strike, and larger lateral flexion of the spinal column to one side than the other. Based on these characteristic features, we are able to state with reasonable certainty whether the suspect could be the perpetrator, but it is not possible to identify the perpetrator positively. Nevertheless, we have been involved in several cases where the court has found that this type of gait analysis was a valuable tool. [ABSTRACT FROM AUTHOR]

Background: Given the relative abundance of modern human DNA and the inherent impossibility for incontestable proof of authenticity, results obtained on ancient human DNA have often been questioned. The widely accepted rules regarding ancient DNA work mainly affect laboratory procedures, however, pre-laboratory contamination occurring during excavation and archaeological-/anthropological handling of human remains as well as rapid degradation of authentic DNA after excavation are major obstacles. Methodology/Principal Findings: We avoided some of these obstacles by analyzing DNA from ten Viking Age subjects that at the time of sampling were untouched by humans for 1,000 years. We removed teeth from the subjects prior to handling by archaeologists and anthropologists using protective equipment. An additional tooth was removed after standard archaeological and anthropological handling. All pre-PCR work was carried out in a ''clean- laboratory'' dedicated solely to ancient DNA work. Mitochondrial DNA was extracted and overlapping fragments spanning the HVR-1 region as well as diagnostic sites in the coding region were PCR amplified, cloned and sequenced. Consistent results were obtained with the ''unhandled'' teeth and there was no indication of contamination, while the latter was the case with half of the ''handled'' teeth. The results allowed the unequivocal assignment of a specific haplotype to each of the subjects, all haplotypes being compatible in their character states with a phylogenetic tree drawn from present day European populations. Several of the haplotypes are either infrequent or have not been observed in modern Scandinavians. The observation of haplogroup I in the present study (,2% in modern Scandinavians) supports our previous findings of a pronounced frequency of this haplogroup in Viking and Iron Age Danes. Conclusion: The present work provides further evidence that retrieval of ancient human DNA is a possible task provided adequate precautions are taken and well-considered sampling is applied. Abstract: The specific purpose of this study was to compare three different collagen extraction methods commonly used in isotope laboratories conducting dietary studies. We evaluated their resultant differences in δ13C and δ15N, collagen quality and collagen yield. Our study was based on well-preserved skeletal material from the medieval period in Denmark. Our study shows that there is a systematic significant difference in the yield and the δ13C values between the three methods. Using the method of DeNiro and Epstein [DeNiro, M.J., Epstein, S., 1981. Influence of diet on the distribution of nitrogen isotopes in animals. Geochimica et Cosmochimica Acta 45, 341–351] with NaOH as cleaning agent, will, according to our study, give δ13C values that are on average ±0.32‰ more positive than using the ultra-filtration method [Brown, T.A., Nelson, D.E., Vogel, J.S., Southon, J.R., 1988. The third method, which is a modified version of the second method, excluded the ultra-filtration step. This method seems to give δ13C values that lie in between the other methods. Our study did not show any significant difference in δ15N values. Although the differences between the methods are very small, we conclude that the use of stable isotope analysis in food determination studies requires adherence to routine methods for preparing and measuring samples. Objectives: By focusing on two Danish leprosaria (Næstved and Odense; 13th–16th c. CE) and using diet and origin as proxies, we follow a multi‐isotopic approach to reconstruct life histories of patients and investigate how leprosy affected both institutionalized individuals and the medieval Danish community as a whole. Materials and Methods: We combine archaeology, historical sources, biological anthropology, isotopic analyses (δ13C, δ15N, δ34S, 87Sr/86Sr) and radiocarbon dating, and further analyze bones with different turnover rates (ribs and long bones). Results: The δ13C, δ15N and δ34S results indicate a C3 terrestrial diet with small contributions of marine protein for leprosy patients and individuals from other medieval Danish sites. A similar diet is seen through time, between males and females, and patients with and without changes on facial bones. The isotopic comparison between ribs and long bones reveals no significant dietary change. The δ34S and 87Sr/86Sr results suggest that patients were local to the regions of the leprosaria. Moreover, the radiocarbon dates show a mere 50% agreement with the arm position dating method used in Denmark. Conclusions: A local origin for the leprosy patients is in line with historical evidence, unlike the small dietary contribution of marine protein. Although only 10% of the analyzed individuals have rib/long bone offsets that undoubtedly show a dietary shift, the data appear to reveal a pattern for 25 individuals (out of 50), with elevated δ13C and/or δ15N values in the ribs compared to the long bones, which points toward a communal type of diet and reveals organizational aspects of the institution. Clinical forensic assessments of injuries' life-threatening danger may have an impact on the legal aftermath following a violent assault. The pursuit of evidence-based guidelines should ensure a user-independent and reproducible forensic practice. However, does it? The aim of this study was to evaluate the forensic life-threatening danger assessments after a protocol implementation in 2016. The evaluation concerned usability and reproducibility of the protocol, and its influence on assessment severity. We analyzed the level of inter- and intra-rater agreement using 169 blinded, prior-protocol cases that were reassessed by two forensic specialists. We compared assessment made the year before and after protocol implementation (n = 262), and the forensic specialists' reassessments with the prior-protocol cases' original assessments (n = 169). Whether to make an assessment, the levels of agreement varied between weak agreement (inter-rater, Κ = 0.43; assessor 1, Κ = 0.57) and strong agreement (assessor 2, Κ = 0.90). Regarding severity, the levels of agreement varied between strong agreement (inter-rater, Κ = 0.87; assessor 1: Κ = 0.90) and almost perfect agreement (assessor 2: Κ = 0.94). The assessments were statistically significant redistributed after the implementation (chi-square test: p < 0.0001). The proportion of cases assessed as having not been in life-threatening danger increased from 9 to 43%, and moderate severity assessments decreased from 55 to 23%. Of the moderate severity assessments, 55% were reassessed as having not been in life-threatening danger. The protocol ensured independent and reproducible assessments when the forensic specialists agreed on making one. The protocol resulted in less severe assessments. Variation in the trace element chemistry of cortical bone microstructure is delineated for interred and non-interred human femora. This was done to investigate the range of element concentrations that might occur within single bones, specifically the original laminar bone and later osteons, and its potential for investigating chemical life histories. To do so, femora were chosen from individuals who experienced quite different ways of life over the past two millennia. The distributions of Sr, Ba, Cu, and Pb, mostly in partial (early) and complete (late) osteons, in cross-sections of proximal femora were characterized through Laser Ablation Inductively Coupled Plasma Mass Spectrometry. Absolute calibrations of these data were obtained using solution Inductively Coupled Plasma Mass Spectrometry on adjacent dissolved bulk samples. Chemical life histories were approximated by classifying bone microstructure into four categories: laminar bone and 1st, 2nd, and 3rd generation osteons. This four-part sequence, on average, charts the temporal dimension of an individual's life. Consistent with recent studies of medieval bones, Sr and Ba are thought to be mainly responsive to diet, presumably related to the consumption of mostly locally produced food, while Cu and Pb do the same for heavy metal exposure often attributable to social status or occupation. No systematic differences in these elements were found between interred and non-interred individuals. The effect of diagenesis on interpretations of life histories based on archaeological bone, therefore, are minimized by plotting element concentrations across cortical bone cross-sections. Objective: This study investigates the expression of mRNA encoding vasopressin in the hypothalamus of autopsy brains of individuals diagnosed with schizophrenia. Methods: Ten brains of individuals with schizophrenia and 10 brains from individuals without any disease were examined during autopsy. The hypothalamic block was dissected and immersion fixed in paraformaldehyde, sucrose substituted, frozen, and cut into 20‐µm‐thick coronal cryostat sections. The sections were hybridized with an S‐35‐labeled DNA antisense oligo probe and after washing covered by an X‐ray film. The hybridization signals on the films were transferred to a computer and densitometrically quantified. Results: The densitometry signals showed a statistically significant lower mRNA expression (53% decrease; p = 0.014) in the paraventricular nucleus of the individuals with schizophrenia compared to the controls. In the supraoptic nucleus, the decrease in the group with schizophrenia was 39% compared to the controls, but this decrease was not statistically significant (p = 0.194). Conclusions: Our results show a low expression of mRNA encoding vasopressin in the paraventricular nucleus of the individuals with schizophrenia. We suggest that vasopressin is not directly involved in the pathogenesis of schizophrenia, but might influence schizophrenic symptoms via vasopressin receptors located in the social behavioral neural network in the forebrain.
