Stroke is a severe medical condition resulting from an interruption in the blood supply to the brain, ultimately compromising tissue homeostasis. Currently, stroke stands as the second leading cause of death worldwide and the third leading cause when considering both mortality and disability together. Periodontitis is characterized by persistent inflammation in hard and soft tissues which support the teeth, primarily caused by bacterial biofilms, and is one of the most common causes of tooth loss in adults and can contribute to a systemic inflammatory burden. In the light of this, the present study investigated the effects of inducing focal ischemia in the motor cortex in rats undergoing experimental periodontitis. Adult Wistar rats were divided into four groups (control, ischemia, periodontitis, and periodontitis + ischemia) and were evaluated for motor performance, basic histology, and the volume and microarchitecture of alveolar bone. The results showed that the comorbidity between ischemia and periodontitis aggravates the spontaneous locomotion of rats, although the motor performance of adult rats had not been altered. Nonetheless, they revealed significant tissue impairment in the motor cortex. Additionally, there was a meaningful alteration in both the volume and microarchitecture of alveolar bone in this group. Our results indicate that the model of comorbidity between ligature-induced experimental periodontitis and focal ischemia was capable of inducing greater neurological impairment and alveolar bone loss in rats, attributable to diminished bone quality, when compared to each condition individually.

Abstract The objective was to evaluate the sensitivity of Piptadenia gonoacantha seeds to desiccation and storage conditions. The seeds were subjected to artificial drying in a forced air convection oven (39.7 °C ± 0.78 and 28.1% ± 1.90 RH) for different periods. After each drying period, evaluation methods were performed to determine the seeds water content and germination (germination test). The seeds were divided into two lots in the storage experiment: with previous drying (at 6.0% water content) and without previous drying (control, at 11% water content), and were stored for 36 months in a plastic bag in three conditions: refrigerator at 5 °C, freezer at -20 °C and natural condition (29 °C). The seeds were removed every three months and subjected to water content measurement and germination tests. The reduction of the initial water content of the seeds from 14% to 6% upon artificial drying resulted in the loss of 1/3 of their viability, suggesting that the seeds are sensitive to desiccation. The best practice to store seeds of P. gonoacantha found in this study was without previous drying, into the freezer. The seeds lost only 14% of their germination after 36 months under these conditions.

Abstract Multiple Myeloma (MM) is a hematological malignancy characterized by the clonal proliferation of plasma cells within the bone marrow. Diagnosing MM presents considerable challenges, involving the identification of plasma cells in cytology examinations on hematological slides. At present, this is still a time-consuming manual task and has high labor costs. These challenges have adverse implications, which rely heavily on medical professionals’ expertise and experience. To tackle these challenges, we present an investigation using Artificial Intelligence, specifically a Machine Learning analysis of hematological slides with a Deep Neural Network (DNN), to support specialists during the process of diagnosing MM. In this sense, the contribution of this study is twofold: in addition to the trained model to diagnose MM, we also make available to the community a fully-curated hematological slide dataset with thousands of images of plasma cells. Taken together, the setup we established here is a framework that researchers and hospitals with limited resources can promptly use. Our contributions provide practical results that have been directly applied in the public health system in Brazil. Given the open-source nature of the project, we anticipate it will be used and extended to diagnose other malignancies.

Non-hegemonic perspectives on sustainability are grounded in environmental sustainability based on justice. We argue about the potential of co-operatives for learning framed through degrowth epistemological visions promoting pedagogical changes in Higher Education. The context is a public university in Catalonia (Spain) implementing an innovative teaching methodology merging service learning and co-operatives for learning in an Environmental Sciences degree. The research question was: What are the experiences of undergraduate students participating in co-operatives for learning about environmental education? Data collection used focus group interviews and individual reflective narratives. Based on the construct of dialectical tensions, we identify three breaking points for the transition to teaching justice-based environmental sustainability using co-operatives for learning: (1) decommodification of work (tradition/innovation; theory/practice); (2) reconstruction of relationships with 'the other' (individual/collective); and (3) with nature (capitalism/degrowth). We conclude that students' environmental praxis is promoted when addressing new perspectives of development and sustainability in university teaching.

The aim of this paper is to analyse facial similarity and apply it to identify the individuals depicted in the mural paintings of the apse of St. Leocadia Church, located in Chaves Municipality (North of Portugal), which were painted during the first quarter of the 16th century. This study also compares the portraits of this mural paintings with the oil paintings by the Proto-Renaissance Portuguese painter Nuno Gonçalves. Through this research, the feasibility of face recognition technology is explored to answer many ambiguities about Manueline stylistic identity and iconography. Additionally, it aims to associate historical events, artistic discoveries, and the expansion of portraiture as propaganda of power during the Portuguese Proto-Renaissance and Early Renaissance. On the other hand, it focuses on the prevalence of the religious and devotional over the sacred in Manueline painting. A proposal was made to identify the characters that are fundamental to the meaning of the mural paintings. An experiment was conducted on seven characters from the paintings at St. Leocadia Church, which were then compared to Nuno Gonçalves’ portraits. Facial similarity analysis was conducted on the faces portrayed in the Panels of St. Vincent, a remarkable portrait gallery from 15th-century Portugal, which has been the subject of national and international research for 130 years. Other paintings that were analysed were the oil paintings of St. Peter and St. Paul and of Infanta St. Joana, which were created by the same Quattrocento master. The purpose of the mural paintings of St. Leocadia Church could be catechetical in nature or related to the ritual practices of royal ancestor worship in royal portrait apses of the churches. It could also be associated with the Portuguese maritime expansion and the macro-imperial ideology of D. Manuel I.

Sixteenth-century mural paintings of Nossa Senhora das Neves in Vilar de Perdizes (Galicia—North Portugal Euroregion) were analysed. An iconographic study has allowed us to understand the meaning of the seven scenes that constitute the mural painting. X-ray fluorescence (XRF) and Raman spectroscopies determined the compounds used in this mural painting, both in the original and in later repaintings. The black paint was bone black. Hydroxyapatite characteristic bands and those of the associated phosphates have been identified. White lime was used as white paint. Lepidocrocite and goethite were used to make yellows, and hematite was used to make red shades. Cinnabar has been used for a later red repaint. Carbon-based compounds and rutile were used to create different tonalities by darkening or lightening colours. It is of great significance to obtain accurate and reliable mural painting information through scientific means, since preservation, restoration, and repainting without detailed information can be harmful to mural paintings.

Nuclear receptor Nur77 plays a pivotal role in immune regulation across various tissues, influencing pro-inflammatory signaling pathways and cellular metabolism. While cellular mechanics have been implicated in inflammation, the contribution of Nur77 to these mechanical processes remains elusive. Macrophages exhibit remarkable plasticity in their morphology and mechanics, enabling them to adapt and execute essential inflammatory functions, such as navigating through inflamed tissue and pathogen engulfment. However, the precise regulatory mechanisms governing these dynamic changes in macrophage mechanics during inflammation remain poorly understood. To establish the potential correlation of Nur77 with cellular mechanics, we compared bone marrow-derived macrophages (BMDMs) from wild-type (WT) and Nur77-deficient (Nur77-KO) mice and employed cytoskeletal imaging, single-cell acoustic force spectroscopy (AFS), migration and phagocytosis assays, and RNA-sequencing. Our findings reveal that Nur77-KO BMDMs exhibit changes to their actin networks compared to WT BMDMs, which is associated with a stiffer and more rigid phenotype. Subsequent in vitro experiments validated our observations, showcasing that Nur77 deficiency leads to enhanced migration, reduced adhesion, and increased phagocytic activity. The transcriptomics data confirmed altered mechanics-related pathways in Nur77-deficient macrophage that are accompanied by a robust pro-inflammatory phenotype. Utilizing previously obtained ChIP-data, we revealed that Nur77 directly targets differentially expressed genes associated with cellular mechanics. In conclusion, while Nur77 is recognized for its role in reducing inflammation of macrophages by inhibiting the expression of pro-inflammatory genes, our study identifies a novel regulatory mechanism where Nur77 governs macrophage inflammation through the modulation of expression of genes involved in cellular mechanics. Our findings suggest that immune regulation by Nur77 may be partially mediated through alterations in cellular mechanics, highlighting a potential avenue for therapeutic targeting.

Abstract The family Hymenochaetaceae includes a diversity of 893 species described around the world. Its representatives are known by their usually rusty colored basidiomes with a poroid hymenial surface, hydnoid or smooth, woody consistency, and wide morphological variation regarding the arrangement on the substrate. They behave as saprophytic, parasitic, ectomycorrhizal and play a fundamental role in the decomposition of wood in forest ecosystems. In the Brazilian Amazonia region, approximately 40 species of Hymenochaetaceae are currently recorded. The main goal of this study was to increase the knowledge on Hymenochaetaceae from the Brazilian Amazonia. Collections were carried out between October 2021 and April 2022 in the state of Pará, municipalities of Tomé-Açu and Bujaru, to expanding the knowledge of this fungal family to the Brazilian Amazonia. A total of 15 specimens were identified, distributed in seven genera and 12 species. Four species are new records for the state of Pará (Fomitiporia apiahyna, Phellinus neocallimorphus, Phellinus sancti-georgii, and Sclerotus extensus) and two of them are new records for the Brazilian Amazonia (P. neocallimorphus and P. sancti-georgii). Our findings contribute to taxonomic knowledge of this family in the Brazilian Amazonia and reduce the information gaps about the diversity of species.

The last glacial inception was characterised by rapid changes in temperature, atmospheric pCO2, and changes in the water mass geometry of the major ocean basins. Although several climatic feedback mechanisms have been proposed to explain the glacial/interglacial cycles witnessed in the Quaternary, the exact mechanistic responses of these processes are still under constrained. In this study we use proxies including planktonic foraminifera compositional assemblages and oxygen stable isotopes to reconstruct past changes in sea surface productivity, stratification, and carbonate dissolution. We use core SIS-249 (2,091 mbsl, western South Atlantic 30°S 47°W), spanning 30–110 thousand years ago (ka), and currently bathed by modern Northern Component Water. We test existing hypotheses suggesting that the orbital obliquity cycle modulates the biological pump in the study area. Spectral analysis run on our synthesised productivity proxies recognises a ~43 kyr-cycle, related to the obliquity cycle. We propose that the enhanced productivity is produced by two mechanisms: i) the glacial upwelling of subsurface nutrient-rich waters and, ii) the continental (wind-driven dust and riverine outflows) fertilisation of the photic zone, with the latter process being obliquity-paced. We also suggest that not only the increased organic matter export but also a change in its bioavailability (from refractory to labile) led to calcium carbonate dissolution, as the degradation of the more soluble organic matter decreased the pH of the glacial bottom water, partially dissolving the calcium carbonate. Abstract Coffea sp. is cultivated in many tropical countries. Brazil has always adopted intensive agricultural practices, but organic coffee farming is an alternative system based on the non-use of agrochemicals and the rational management of soils. Metabarcoding 16S analysis using next-generation sequencing has been developed to identify and compare the diversity of the Coffea arabica L. rhizospheric bacterial community in two farming areas in São Paulo, Brazil. Dourado uses conventional farming, while Ribeirão Corrente uses organic. We found broad taxonomic composition, with sequences from 24 phyla, 55 classes, 61 orders, 146 families, and 337genus. The three most abundant phyla were Proteobacteria (38.27%), Actinobacteria (15.56%), and Acidobacteria (16.10%). In organic farming, the top 3 were the family Sphingomonadaceae, order Rhizobiales, genus Nocardioides, and Gp6. The genus Gp2 and the phylum Candidatus Saccharibacteria were the most abundant OTUs exclusively present in conventional farming. In the organic farming practice, Proteobacteria, Actinobacteria, and Acidobacteria were also present among the exclusive OTUs; we also found OTUs belonging to Bacteroidetes, Firmicutes, and Verrucomicrobia. Our study indicates a positive effect of organic farming on microbial communities. Fertilization may directly affect soil microbiota, suggesting that a large and active microbial community low in functional diversity might not adapt to new climatic conditions. A diverse community could provide better resilience to environmental changes, improving the productivity of this important crop.

Trace elements such as cobalt (Co), molybdenum (Mo), and zinc (Zn) play necessary roles in different biological functions. Co is a microelement that influences the vascular system. Mo works as an enzymatic cofactor of three enzymes (aldehyde oxidase, sulfite oxidase, and xanthine oxidase dehydrogenase). However, these elements are difficult to detect, since the analytical methods developed have a high cost, which restrict their applicability. In this sense, fluorescent sensors are an alternative for detecting trace elements, such as Mo4+ ions. Herein, a new multichannel trace elements sensor has been proposed to detect Mo entities. In this sense, two different N-CQDs were synthesized and fully characterized. The N-CQDs presented quantum yield values of 25.93% and 6.02% and excellent solubility in water. Also, a mixture of these two carbon-based nanoparticles was used to identify and to quantify Mo in water between seven different trace elements. The method was found to reach 1.28 and 3.88 ppm for limit of detection (LOD) and quantification (LOQ), respectively. To further verify the potential of the detection platform, the multichannel sensor was applied to identify the different concentrations of metal ions (Fe2+, Co2+, Mn2+, Cu2+, Zn2+, Mg2+, and Mo4+) in water. The data matrix was treated using different algorithms, such as K-Means and Discriminant Analysis (DA). The detection strategy has successfully identified the molybdenum ions at 5 ppm. This result shows the potential application of a multichannel sensor toward the detection of Mo entities, since it is comparable with the molybdenum test already available on the market.

Objective: The present study aims to evaluate the viability of adult human neural cells in culture obtained from traumatized brain tissues collected in emergency surgery procedures. Methods: Exploratory, descriptive, quantitative and cross-sectional study evaluating samples obtained from patients who underwent traumatic brain injury with extrusion of brain tissue submitted to cell culture in a standardized medium, being preserved during 168h. After observation under phase contrast microscopy and immunohistochemical processing for neuronal (MAP-2) and glial (GFAP) markers, morphometric parameters of neural cells (cell body area, dendritic field length and fractal dimension) were evaluated using ImageJ software, with data obtained after 24, 72 and 168h being compared using non-parametric Kruskal Wallis test, followed by Dunn’s post hoc test. Results: The explant of the nervous tissue revealed a consolidated pattern of cell migration into the culture medium. Cell proliferation, upon reaching confluence, presented an aspect of cellular distribution juxtaposed along the culture medium at all time points analyzed. Both neurons and glial cells remained viable after 168h in culture, with their morphologies not varying significantly throughout the time points evaluated. Immunohistochemistry for MAP-2 showed a relatively well-preserved cytoskeletal organization. GFAP immunoreactivity revealed activated astrocytes especially at the later time point. Conclusions: Our results point out the viability of cell culture from traumatized human nervous tissue, opening up perspectives for the use of substances of natural origin that may contribute neuroprotectively to neuronal maintenance in culture, allowing future translational approach.

The basal ganglia are a subcortical collection of interacting clusters of cell bodies, and are involved in reward, emotional, and motor circuits. Within all the brain processing necessary to carry out voluntary movement, the basal nuclei are fundamental, as they modulate the activity of the motor regions of the cortex. Despite being much studied, the motor circuit of the basal ganglia is still difficult to understand for many people at all, especially undergraduate and graduate students. This review article seeks to bring the functioning of this circuit with a simple and objective approach, exploring the functional anatomy, neurochemistry, neuronal pathways, related diseases, and interactions with other brain regions to coordinate voluntary movement.

Paradigma e epistemologia são conceitos recorrentemente citados por estudiosos e pesquisadores que debatem a via da complexidade, dividindo-lhes a opinião sobre que concepção melhor se relaciona e mais adequadamente reflete a noção complexa em sua caraterização mais essencial. Neste artigo, então, temos como objetivo refletir sobre essa questão terminológica, buscando distinguir o termo que, em nossa opinião, melhor defina os princípios fundantes da complexidade, revelando seus propósitos e características. A partir de uma discussão sobre o conceito de paradigma e suas implicações, exploramos os matizes do que se denomina evolução paradigmática, para chegar à concepção de epistemologia e delinear suas contribuições à visão de complexidade sob o enfoque moriniano. Também refletimos sobre uma reforma do pensamento e da educação que, indicadoras de metamorfoses, podem levar à esperança da transparadigmatologia. Ao longo de nossa discussão, encaminhamos nossas considerações para as contribuições dessa argumentação para o processo ensino-aprendizagem e para a formação docente. De forma sucinta, aqui promovemos um exercício de compreensão do próprio processo de conhecimento à luz do pensamento complexo, com base em Edgar Morin, em direção ao metaponto de vista, como contribuição às ciências da Educação e da Linguística Aplicada, entre outras.

Abstract Sub‐Saharan Africa has been the last continent to experience a significant number of cases in the novel Coronavirus (COVID‐19). Studies suggest that air pollution is related to COVID‐19 mortality; poor air quality has been linked to cardiovascular, cerebrovascular, and respiratory diseases, which are considered co‐morbidities linked to COVID‐19 deaths. We examine potential connections between country‐wide COVID‐19 cases and environmental conditions in Senegal, Cabo Verde, Nigeria, Cote D'Ivorie, and Angola. We analyze PM2.5 concentrations, temperatures from cost‐effective in situ measurements, aerosol optical depth (AOD), and fire count and NO2 column values from space‐borne platforms from 1 January 2020 through 31 March 2021. Our results show that the first COVID‐19 wave in West Africa began during the wet season of 2020, followed by a second during the dry season of 2020. In Angola, the first wave starts during the biomass burning season but does not peak until November of 2020. Overall PM2.5 concentrations are the highest in Ibadan, Nigeria, and coincided with the second wave of COVID‐19 in late 2021 and early 2022. The COVID‐19 waves in Cabo Verde are not in phase with those in Senegal, Nigeria, and Cote, lagging by several months in general. Overall, the highest correlations occurred between weekly new COVID‐19 cases meteorological and air quality variables occurred in the dry season.

Objective: To evaluate the correlation between functional dependence and emotional state, cognitive function, and ambulation of the elderly people accompanied by a family health team. Methods: A cross-sectional, quantitative, and prospective study was conducted in the city of Quixeré/CE between August 2020 and April 2021. Participated in 229 elderly people registered in the Family Health Strategy teams evaluated using the Lawton scale, MMSE, GDS-15, and Tinneti test. The association of variables was verified using the chi-square or Fisher's exact tests. Odds Ratio (95%CI) were calculated, followed by logistic regression using the Hosmer and Lemeshow test. Results: The sample was primarily composed of women with low education, average age of 70 years, and a high demand for primary health services. Women were more vulnerable to depression, with no difference in falls related to sex. Age was a risk factor for all aspects evaluated. There was a correlation between functional dependence and all aspects analyzed. There was a relationship between the level of functional dependence and education (OR: 2.0; 95%CI: 0.05 - 78.32), living in the rural area (OR: 2.83; 95%CI: 1.6 - 5.1), depression (OR: 5.18; 95%CI: 1.2 – 22.8) and risk of falls (OR: 1.67 CI: 0.8 – 3.5), with the last two factors remaining significant in the logistic regression. Conclusion: Functional dependence to more complex daily activities among the elderly was significant, with an essential relationship with emotional and cognitive aspects and risk of falls.

Multiple Myeloma (MM) is a cytogenetically heterogeneous clonal plasma cell proliferative disease whose diagnosis is supported by analyses on histological slides of bone marrow aspirate. In summary, experts use a labor-intensive methodology to compute the ratio between plasma cells and non-plasma cells. Therefore, the key aspect of the methodology is identifying these cells, which relies on the experts' attention and experience. In this work, we present a valuable dataset comprising more than 5,000 plasma and non-plasma cells, labeled by experts, along with some patient diagnostics. We also share a Deep Neural Network model, as a benchmark, trained to identify and count plasma and non-plasma cells automatically. The contributions of this work are two-fold: (i) the labeled cells can be used to train new practitioners and support continuing medical education; and (ii) the design of new methods to identify such cells, improving the results presented by our benchmark. Believed to cause damage to the nervous system and possibly being associated with neurodegenerative diseases, deltamethrin (DM) is a type II pyrethroid used in pest control, public health, home environment, and vector control. The objective of this study was to evaluate the motor, cognitive and emotional changes associated with dopaminergic and BDNF imbalance after DM exposure in rats. Sixty Wistar rats (9–10 months-old) were used, under Ethics Committee on Animal Research license (ID 19/2017). The animals were randomly divided into four groups: control (CTL, 0.9% saline), DM2 (2 mg DM in 1.6 mL 0.9% saline), DM4 (4 mg of DM in 1.6 mL of 0.9% saline), and DM8 (8 mg of DM in 1.6 mL of 0.9% saline). DM groups were submitted to 9 or 15 inhalations, one every 48 h. Half of the animals from each group were randomly selected and perfused 24 h after the 9th or 15th inhalation. Throughout the experiment, the animal’s behavior were evaluated using catalepsy test, open field, hole-board test, Modified Elevated Plus Maze, and social interaction. At the end of the experiments, the rats were perfused transcardially and their brains were processed for Tyrosine Hydroxylase (TH) and Brain derived neurotrophic factor (BDNF) immunohistochemistries. The animals submitted to 9 inhalations of DM showed a reduction in immunoreactivity for TH in the Substantia nigra pars compacta (SNpc), ventral tegmental area (VTA), and dorsal striatum (DS) areas, and an increase in BDNF in the DS and CA1, CA3 and dentate gyrus (DG) hippocampal areas. Conversely, the animals submitted to 15 inhalations of DM showed immunoreactivity reduced for TH in the SNpc and VTA, and an increase in BDNF in the hippocampal areas (CA3 and DG). Our results indicate that the DM inhalation at different periods induce motor and cognitive impairments in rats. Such alterations were accompanied by dopaminergic system damage and a possible dysfunction on synaptic plasticity.

Objective: To characterize social communication in children up to 12 years old diagnosed with Autistic Spectrum Disorder (ASD) treated at the Child Psychosocial Care Center (CAPSi) in Mossoró/RN. Methods: Exploratory, descriptive, quantitative, cross-sectional study conducted at CAPSi in Mossoró/RN. The data collection was based on the application of questionnaires to parents/guardians to characterize their socioeconomic conditions and their perception of their children. The Language Development Assessment (LDA) instrument for children was used, developed to assess the acquisition and development of language content (semantics) and structure (morphology and syntax). The questionnaires were coded, and the data were tabulated for further statistical analysis. Results: 33 children and 73 parents/guardians were studied. Most parents/guardians were female (95.9%), aged 30-39 years (mean age 37.6 years predominantly), with a partner (60%), with high school education (45%), and monthly income around one minimum wage (76.7%), having positive attitudes toward their children and noticing difficulties in their communication. In applying the questionnaire to the children, 63.7% presented results within the normal range, while 3% presented mild and 33.3% severe language alterations. Conclusion: Children with ASD attending CAPSi have language disorders in the pragmatic sublevel, and more than half of the participants did not have morphosyntactic and semantic changes.

Autism Spectrum Disorder (ASD) encompasses a highly variable set of neurodevelopmental disorders with early onset and non-degenerative pattern. According to the World Health Organization's International Classification of Diseases (11th Revision), ICD-11, ASD is characterized as “persistent deficits in the ability to initiate and to sustain reciprocal social interaction and social communication, and by a range of restricted, repetitive, and inflexible patterns of behaviour, interests or activities that are clearly atypical or excessive for the individual's age and sociocultural context.”

The environmental contamination by methylmercury (MeHg) is a major concern for public health. The effects of MeHg in the central nervous system (CNS) of adult animals have been extensively investigated; however, little is known about the effects of MeHg exposure during intrauterine and lactation periods on motor and cognitive functions of adolescent rats. Therefore, this study aimed to investigate the effect of MeHg exposure during intrauterine life and lactation on both motor and cognitive functions of offspring rats. Ten female Wistar rats were exposed to 40 μg/kg/day of MeHg through cookie treats from the first day of pregnancy until the last day of breastfeeding. Both motor and cognitive functions of offspring male rats were assessed by open field, rotarod, and step-down inhibitory avoidance tests. Forty-one days after birth, the hippocampus and cerebellum were collected to determine total Hg content, antioxidant capacity against peroxyl radicals (ACAP), reduced glutathione (GSH) levels, lipid peroxidation (LPO), and nitrite levels. MeHg exposure during CNS development increased Hg levels in both hippocampal and cerebellar parenchymas, triggered oxidative stress throughout ACAP and GSH decrease, increased LPO and nitrite levels. These alterations resulted in reduced spontaneous and stimulated locomotion and short- and long-term memory deficits. Therefore, damages triggered by MeHg exposure during intrauterine life and lactation had detrimental effects on oxidative biochemistry and motor and cognitive functions of offspring rats.

Herein, we have displayed an easy way to produce monodisperse spinel nanoparticles (NPs) and the antifungal activity of CoFe2O4, Co0.5Zn0.5Fe2O4 and ZnFe2O4 nanostructures. Firstly, the structural, morphological and magnetic properties of each NP were investigated through x-ray diffraction (XRD), Transmission Electron Microscopy (TEM) and Vibrating Sample Magnetometer (VSM). The XRD data showed diffraction peaks related to the crystalline spinel phase. The TEM micrographs displayed monodisperse NPs with spherical morphology. The average sizes of CoFe2O4, Co0.5Zn0.5Fe2O4 and ZnFe2O4 NPs were 6.87 ± 0.05 nm, 5.18 ± 0.01 nm and 11.52 ± 0.09 nm, respectively. The VSM data indicated that the nanostructures are superparamagnetic at room temperature. Afterward, the antifungal properties of the Co/Zn-based ferrite NPs against Botrytis cinerea were tested. So, the inhibition of mycelial growth by different concentrations (45 – 360 ppm) of NPs was measured. The most effective nanostructure was CoFe2O4, with an EC50 value of 265 ppm. Further, to elucidate how the NPs are affecting B. cinerea, reactive oxygen species (ROS) production was measured. The results indicated that the CoFe2O4 monodisperse NPs could induce a burst of ROS in B. cinerea, promoting cellular damage.

Abstract The cleaning or removal of manganese stains on Cultural Heritage has not been much tested or successful so far. The aim of this article was to assess a new green cleaning gel for Mn-rich black-blue stains on different substrates. The black-blue stains were characterized at optical and chemical level through colour-related data, optical microscope, FTIR, XRF and XPS. Mn-stained granite found on historical churches at Vila Real (North of Portugal) and glass jars of Leclanché cells, belonging to the ISEP’s Museum (Portugal) collection, were the ideal case studies to test the efficiency of chitosan: thiourea dioxide (TD) cleaning gel. TD proved to be the best candidate to reduce insoluble manganese oxides, over Hydroxylamine Hydrochloride and Hydroxymethanesulfinic Acid. Cleaning assays performed on stained granite samples collected at a historical quarry and in situ application on stained granite churches allowed removal of the stains to a satisfactory level. Similar results were obtained on stained glass jars.

Abstract We present a measurement of the cosmic-ray spectrum above 100 PeV using the part of the surface detector of the Pierre Auger Observatory that has a spacing of 750 m. An inflection of the spectrum is observed, confirming the presence of the so-called second-knee feature. The spectrum is then combined with that of the 1500 m array to produce a single measurement of the flux, linking this spectral feature with the three additional breaks at the highest energies. The combined spectrum, with an energy scale set calorimetrically via fluorescence telescopes and using a single detector type, results in the most statistically and systematically precise measurement of spectral breaks yet obtained. These measurements are critical for furthering our understanding of the highest energy cosmic rays.

Human activity has required, since its origins, stones as raw material for carving, construction and rock art. The study, exploration, use and maintenance of building stones is a global phenomenon that has evolved from the first shelters, manufacture of lithic tools, to the construction of houses, infrastructures and monuments. Druids, philosophers, clergymen, quarrymen, master builders, naturalists, travelers, architects, archaeologists, physicists, chemists, curators, restorers, museologists, engineers and geologists, among other professionals, have worked with stones and they have produced the current knowledge in heritage stones. They are stones that have special significance in human culture. In this way, the connotation of heritage in stones has been acquired over the time. That is, the stones at the time of their historical use were simply stones used for a certain purpose. Therefore, the concept of heritage stone is broad, with cultural, historic, artistic, architectural, and scientific implications. A historical synthesis is presented of the main events that marked the use of stones from prehistory, through ancient history, medieval times, and to the modern period. In addition, the main authors who have written about stones are surveyed from Ancient Roman times to the middle of the twentieth century. Subtle properties of stones have been discovered and exploited by artists and artisans long before rigorous science took notice of them and explained them.

Traumatic brain injury (TBI) is one of the leading causes of long-lasting morbidity and mortality worldwide, being a devastating condition related to the impairment of the nervous system after an external traumatic event resulting in transitory or permanent functional disability, with a significant burden to the healthcare system. Harmful events underlying TBI can be classified into two sequential stages, primary and secondary, which are both associated with breakdown of the tissue homeostasis due to impairment of the blood–brain barrier, osmotic imbalance, inflammatory processes, oxidative stress, excitotoxicity, and apoptotic cell death, ultimately resulting in a loss of tissue functionality. The present study provides an updated review concerning the roles of brain edema, inflammation, excitotoxicity, and oxidative stress on brain changes resulting from a TBI. The proper characterization of the phenomena resulting from TBI can contribute to the improvement of care, rehabilitation and quality of life of the affected people.

Abstract Background KRAS is the most frequently mutated oncogene in cancer, however efforts to develop targeted therapies have been largely unsuccessful. Recently, two small-molecule inhibitors, AMG 510 and MRTX849, have shown promising activity in KRAS G12C-mutant solid tumors. The current study aims to assess the molecular profile of KRAS G12C in colorectal (CRC) and non-small-cell lung cancer (NSCLC) tested in a clinical certified laboratory. Methods CRC and NSCLC samples submitted for KRAS testing between 2017 and 2019 were reviewed. CRC samples were tested for KRAS and NRAS by pyrosequencing, while NSCLC samples were submitted to next generation sequencing of KRAS, NRAS, EGFR, and BRAF. Results The dataset comprised 4897 CRC and 4686 NSCLC samples. Among CRC samples, KRAS was mutated in 2354 (48.1%). Most frequent codon 12 mutations were G12D in 731 samples (14.9%) and G12V in 522 (10.7%), followed by G12C in 167 (3.4%). Abstract The objective of this work was to evaluate the sensitivity of Melanoxylon brauna Schott. tree legume seeds to desiccation and storage. In the drying experiment, the Melanoxylon brauna seeds were submitted to two drying conditions: a forced air circulation chamber (40.18 °C ± 0.13 and 28.48% ± 3.95 RH) and a silica gel desiccator (27.19 °C ± 1.28 and 26.19% ± 0.94 RH) for different times (0, 12, 24, 36, 72, and 144 hours). A completely randomized design in a 2 (drying methods) × 5 (drying times) factorial scheme plus control and 4 replications of 25 seeds was used. The following variables were evaluated before and after drying: seed moisture content, percentage of germinated seeds, germination speed index, percentage of mortality, normal and abnormal seedlings. In the storage experiment the seeds were divided into two batches: pre-dried (at 5.0% humidity) and without drying (control at 8.9% humidity). The seeds were then stored in plastic bags in three environments: refrigerator at 5 °C, freezer at –20 °C and room temperature (29 °C). The seeds were removed every four months and submitted to the humidity and germination test for 24 months. Data from this storage experiment were analyzed considering a randomized block design in a 2 (drying levels: presence and absence) × 3 (storage environments: refrigerator, freezer or room temperature) factorial scheme + 2 controls (with and without drying at baseline) and 4 repetitions of 25 seeds. Drying reduced initial seed water content from 8.9% to 5.0%, without loss of viability. Drying in the chamber at 40 °C was faster and more efficient than in silica gel. The results enable classifying the seeds of this species as orthodox, i.e. tolerant to desiccation. The fridge and freezer were efficient for storing the Melanoxylon brauna seeds up to 24 months, independent of previous drying, while storing the seeds at room temperature with previous drying makes them last longer than without drying, as the seeds can last up to 16 months with drying, or 12 months without drying.

Abstract The hybrid design of the Pierre Auger Observatory allows for the measurement of the properties of extensive air showers initiated by ultra-high energy cosmic rays with unprecedented precision. By using an array of prototype underground muon detectors, we have performed the first direct measurement, by the Auger Collaboration, of the muon content of air showers between $$2\times 10^{17}$$ 2×1017 and $$2\times 10^{18}$$ 2×1018 eV. We have studied the energy evolution of the attenuation-corrected muon density, and compared it to predictions from air shower simulations. The observed densities are found to be larger than those predicted by models. We quantify this discrepancy by combining the measurements from the muon detector with those from the Auger fluorescence detector at $$10^{{17.5}}\, {\mathrm{eV}} $$ 1017.5eV and $$10^{{18}}\, {\mathrm{eV}} $$ 1018eV . We find that, for the models to explain the data, an increase in the muon density of $$38\%$$ 38% $$\pm 4\% (12\%)$$ ±4%(12%) $$\pm {}^{21\%}_{18\%}$$ ±18%21% for EPOS-LHC, and of $$50\% (53\%)$$ 50%(53%) $$\pm 4\% (13\%)$$ ±4%(13%) $$\pm {}^{23\%}_{20\%}$$ ±20%23% for QGSJetII-04, is respectively needed.
