Abstract The diets of the eight species of ursids range from carnivory (e.g., polar bears, Ursus maritimus) to insectivory (e.g., sloth bears, Melursus ursinus), omnivory (e.g., brown bears, U. arctos), and herbivory (e.g., giant pandas, Ailuropoda melanoleuca). Dietary energy availability ranges from the high-fat, highly digestible, calorically dense diet of polar bears (~ 6.4 kcal digestible energy/g fresh weight) to the high-fiber, poorly digestible, calorically restricted diet (~ 0.7) of giant pandas. Thus, ursids provide the opportunity to examine the extent to which dietary energy drives evolution of energy metabolism in a closely related group of animals. We measured the daily energy expenditure (DEE) of captive brown bears in a relatively large, zoo-type enclosure and compared those values to previously published results on captive brown bears, captive and free-ranging polar bears, and captive and free-ranging giant pandas. We found that all three species have similar mass-specific DEE when travel distances and energy intake are normalized even though their diets differ dramatically and phylogenetic lineages are separated by millions of years. For giant pandas, the ability to engage in low-cost stationary foraging relative to more wide-ranging bears likely provided the necessary energy savings to become bamboo specialists without greatly altering their metabolic rate.

Abstract Declining Arctic sea ice is increasing polar bear land use. Polar bears on land are thought to minimize activity to conserve energy. Here, we measure the daily energy expenditure (DEE), diet, behavior, movement, and body composition changes of 20 different polar bears on land over 19–23 days from August to September (2019–2022) in Manitoba, Canada. Polar bears on land exhibited a 5.2-fold range in DEE and 19-fold range in activity, from hibernation-like DEEs to levels approaching active bears on the sea ice, including three individuals that made energetically demanding swims totaling 54–175 km. Bears consumed berries, vegetation, birds, bones, antlers, seal, and beluga. Beyond compensating for elevated DEE, there was little benefit from terrestrial foraging toward prolonging the predicted time to starvation, as 19 of 20 bears lost mass (0.4–1.7 kg•day−1). Although polar bears on land exhibit remarkable behavioral plasticity, our findings reinforce the risk of starvation, particularly in subadults, with forecasted increases in the onshore period.

Updating predictions of the response of high-profile, at-risk species to climate change and anthropogenic stressors is vital for informing effective conservation action. Here, we review two prior generations of Bayesian network probability models predicting changes in global polar bear (Ursus maritimus) population status, and provide a contemporary update based on recent research findings and sea-ice projections by newer climate models. We compare predictions of polar bear population response from all 3 models among four circumpolar Arctic ecoregions, using sea ice projections based on three IPCC greenhouse gas emissions scenarios (SSP2.6, 4.5, 8.5). Consistent with the previous two model generations, polar bears will continue to experience increasing probability of declining or greatly declining populations throughout the 21st century, varying by emission scenario. Populations within the Polar Basin Divergent Ice Ecoregion have the highest predicted probability of declines, but predictions were slightly less dire relative to the previous model generation. Most of the influence, denoted by model sensitivity analysis, is from expected degradation and loss of sea ice and reduced access to marine prey. The lack of terrestrial prey adequate to substitute for loss of access to marine prey, as well as human-caused bear morality associated with hunting and defense of life and property encountered when polar bears are increasingly forced ashore also contributed to predicted declines. Although some tidewater glacial fjords and other localized onshore resources may provide local refugia, their benefit is transient. Our findings continue to inform priorities for inventory, monitoring, and research needs, and suggest that similar updates to models of other at-risk species can capitalize on the comparison framework we present here.

Abstract Within optimal foraging theory animals should maximize their net energy gain while minimizing energetic costs. Energetic expenditure in wild animals is therefore key to measure proxies of fitness. Accelerometers are an effective tool to study animal movement-based energetics, but retrieval of the device is usually required and often difficult. Accelerometers measure movement across three axes (x, y, and z) and can be calibrated to measures of oxygen consumption from captive animals, providing estimates of overall energy expenditure. Measuring energetic expenditures using a global positioning system (GPS) approach could provide an alternative method to study energetic ecology. This technique uses locomotor speeds across a range of slopes from successive GPS locations, which can be linked to the energy expenditure from captive individuals. We compared accelerometer and GPS methods of energetic expenditures in free-roaming brown bears (Ursus arctos) on the Kodiak Archipelago, Alaska, USA. We then applied the GPS method to examine how multiple factors influenced brown bear movement-based daily energetic expenditures (MDEE). A new geological-structural map of the southern Serre Massif (SM), in the south-central part of the Calabrian-Peloritani-Orogen (CPO), is provided. CPO is a ribbon-like microplates puzzle, originally belonging to the southern European Variscan Belt and, later involved into the Alpine geodynamics of the central Mediterranean Area. The SM represents one of the key European Variscan basement relicts, because of its exhumation mechanisms as well as for the absence of any Alpine metamorphic overprint. This map has the aim to better delineate the sequence of the Variscan blasto-deformational relationships consisting in a prograde multistage history, followed by an extensional/transpressional multistage retrograde evolution, which triggered the intrusion of the former plutonic products. The mylonitic fabric resulted finally replaced by the effects of the late- to post-kinematic plutonic intrusions coeval with a former late-Variscan exhumation stage, followed, during Mesozoic, by carbonate platform sedimentation, before to be completed exhumed during the Oligocene-Miocene Alpine stages.

Abstract Background Environmental conditions can influence animal movements, determining when and how much animals move. Yet few studies have quantified how abiotic environmental factors (e.g., ambient temperature, snow depth, precipitation) may affect the activity patterns and metabolic demands of wide-ranging large predators. We demonstrate the utility of accelerometers in combination with more traditional GPS telemetry to measure energy expenditure, ranging patterns, and movement ecology of 5 gray wolves (Canis lupus), a wide-ranging social carnivore, from spring through autumn 2015 in interior Alaska, USA. Results Wolves exhibited substantial variability in home range size (range 500–8300 km2) that was not correlated with daily energy expenditure. Mean daily energy expenditure and travel distance were 22 MJ and 18 km day−1, respectively. Wolves spent 20% and 17% more energy during the summer pup rearing and autumn recruitment seasons than the spring breeding season, respectively, regardless of pack reproductive status. Wolves were predominantly crepuscular but during the night spent 2.4 × more time engaged in high energy activities (such as running) during the pup rearing season than the breeding season. Conclusion Integrating accelerometry with GPS telemetry can reveal detailed insights into the activity and energetics of wide-ranging predators. Heavy precipitation, deep snow, and high ambient temperatures each reduced wolf mobility, suggesting that abiotic conditions can impact wolf movement decisions. Identifying such patterns is an important step toward evaluating the influence of environmental factors on the space use and energy allocation in carnivores with ecosystem-wide cascading effects, particularly under changing climatic conditions.

Abstract Retinoid X receptors are members of the nuclear receptor family that regulate gene expression in response to retinoic acid and related ligands. Group 1 metabotropic glutamate receptors are G-protein coupled transmembrane receptors that activate intracellular signaling cascades in response to the neurotransmitter, glutamate. These two classes of molecules have been studied independently and found to play important roles in regulating neuronal physiology with potential clinical implications for disorders such as depression, schizophrenia, Parkinson’s and Alzheimer’s disease. Here we show that mice lacking the retinoid X receptor subunit, RXRγ, exhibit impairments in group 1 mGluR-mediated electrophysiological responses at hippocampal Schaffer collateral-CA1 pyramidal cell synapses, including impaired group 1 mGluR-dependent long-term synaptic depression (LTD), reduced group 1 mGluR-induced calcium release, and loss of group 1 mGluR-activated voltage-sensitive currents. These animals also exhibit impairments in a subset of group 1 mGluR-dependent behaviors, including motor performance, spatial object recognition, and prepulse inhibition. Together, these observations demonstrate convergence between the RXRγ and group 1 mGluR signaling pathways that may function to coordinate their regulation of neuronal activity. They also identify RXRγ as a potential target for the treatment of disorders in which group 1 mGluR signaling has been implicated.

Pharmaceuticals are now considered to be established contaminants, and their presence in water poses a real risk not only to the marine ecosystem, as they may adversely affect non-target organisms that are exposed to them, but also indirectly to humans. This is particularly true for the model organism considered in this work, Mytilus galloprovincialis (Lamarck, 1819), a suspensivore and bioaccumulating organism that enters the human food chain. Among the most commonly used over-the-counter medicines, anti-inflammatory drugs certainly feature prominently, with acetylsalicylic acid (ASA) at the top. In this work, M. galloprovincialis specimens were exposed to two concentrations of ASA (10 and 100 μg/L) for 10 and 20 days to evaluate possible alterations in the decrease in regulatory volume (RVD) in digestive gland cells and cell viability of both these cells and hemocytes. In addition, the histopathological condition index of the gills and digestive gland was evaluated. The data obtained showed that chronic exposure to ASA did not alter the cell viability of hemocytes and digestive gland cells but alters the physiological mechanisms of volume regulation in the digestive gland and, in addition, a time-dose reaction to ASA in the gills and digestive gland showing numerous alterations such as lipofuscin deposits and hemocyte infiltration was found. These results confirm the potential toxicity to the marine biota, highlighting the necessity to deepen the knowledge regarding the link between over-the-counter pharmaceuticals and non-target organisms.

In this work, the best alternative design of Heat Exchange Network (HEN) applied to the process of esterification and transesterification with methanol to obtain biodiesel from waste vegetable oils (WVO) was selected through a differential economic analysis between the alternatives with and without HEN. Both options with heat integration presented a higher net present value (NPV) than those where no energy integration was applied, mainly due to the decrease in operating costs. The NPV for options 1 and 2 was US$ 29,821,901 and US$ 26,004,006, respectively. Therefore, Option 1 turned out to be the most economically convenient technological alternative.

The present study studies the ideal concentration of natural sweetener Stevia in jams developed from grapes reduced in their natural carbohydrates, through a Central Location Test, from a panel of 123 consumers from the city of Olavarria (Argentina), in an age range of 15-65 years. Each of the consumers received 6 samples of jam with different concentrations of Stevia sweetener. For each sample, consumers had to answer according to their degree of acceptability if: "It is less sweet than I like it", "It is sweet as I like it" or "It is sweeter than I like it". The optimal concentration was 3.74% (w / w), which means that 5.61 g of the Stevia sweetener is equivalent to 100 g of common sugar, for which purpose the Cut Point technique was applied applying the Statistical Survival Methodology.

The PEACETIME cruise (May–June 2017) was a basin-scale survey covering the Provencal, Algerian, Tyrrhenian, and Ionian basins during the post-spring bloom period and was dedicated to tracking the impact of Saharan dust deposition events on the Mediterranean Sea pelagic ecosystem. Two such events occurred during this period, and the cruise strategy allowed for the study of the initial phase of the ecosystem response to one dust event in the Algerian Basin (during 5 d at the so-called “FAST long-duration station”) as well as the study of a latter response to another dust event in the Tyrrhenian Basin (by sampling from 5 to 12 d after the deposition). This paper documents the structural and functional patterns of the zooplankton component during this survey, including their responses to these two dust events. The mesozooplankton were sampled at 12 stations using nets with two different mesh sizes (100 and 200 µm) that were mounted on a Bongo frame for vertical hauls within the depth layer from 0 to 300 m. The Algerian and Tyrrhenian basins were found to be quite similar in terms of hydrological and biological variables, which clearly differentiated them from the northern Provencal Basin and the eastern Ionian Basin. In general, total mesozooplankton showed reduced variations in abundance and biomass values over the whole area, with a noticeable contribution from the small size fraction (<500 µm) of up to 50 % with respect to abundance and 25 % with respect to biomass. This small size fraction makes a significant contribution (15 %–21 %) to the mesozooplankton fluxes (carbon demand, grazing pressure, respiration, and excretion), which is estimated using allometric relationships to the mesozooplankton size spectrum at all stations. The taxonomic structure was dominated by copepods, mainly cyclopoid and calanoid copepods, and was completed by appendicularians, ostracods, and chaetognaths. Zooplankton taxa assemblages, analyzed using multivariate analysis and rank frequency diagrams, slightly differed between basins, which is in agreement with recently proposed Mediterranean regional patterns. However, the strongest changes in the zooplankton community were linked to the abovementioned dust deposition events. A synoptic analysis of the two dust events observed in the Tyrrhenian and Algerian basins, based on the rank frequency diagrams and a derived index proposed by Mouillot and Lepretre (2000), delivered a conceptual model of a virtual time series of the zooplankton community responses after a dust deposition event. The initial phase before the deposition event (state 0) was dominated by small-sized cells consumed by their typical zooplankton filter feeders (small copepods and appendicularians). The disturbed phase during the first 5 d following the deposition event (state 1) then induced a strong increase in filter feeders and grazers of larger cells as well as the progressive attraction of carnivorous species, leading to a sharp increase in the zooplankton distribution index. Afterward, this index progressively decreased from day 5 to day 12 following the event, highlighting a diversification of the community (state 2). A 3-week delay was estimated for the index to return to its initial value, potentially indicating the recovery time of a Mediterranean zooplankton community after a dust event. To our knowledge, PEACETIME is the first in situ study that has allowed for the observation of mesozooplankton responses before and soon after natural Saharan dust depositions. The change in the rank frequency diagrams of the zooplankton taxonomic structure is an interesting tool to highlight short-term responses of zooplankton to episodic dust deposition events. Obviously dust-stimulated pelagic productivity impacts up to mesozooplankton in terms of strong but short changes in taxa assemblages and trophic structure, with potential implications for oligotrophic systems such as the Mediterranean Sea.

In the Arctic, warming air and ocean temperatures have resulted in substantial changes to sea ice, which is primary habitat for polar bears (Ursus maritimus). Reductions in extent, duration, and thickness have altered sea ice dynamics, which influences the ability of polar bears to reliably access marine mammal prey. Because nutritional condition is closely linked to population vital rates, a progressive decline in access to prey or an increase in the energetic cost of accessing prey has the potential to adversely affect polar bear population dynamics. We examined long-term (1983–2015) patterns of spring body condition (indexed using residual body mass) and maternal investment (i.e., litter mass of cubs-of-the-year and yearlings; COY and YRL) of polar bears from Alaska’s southern Beaufort Sea to evaluate potential relationships with regional- and circumpolar-scale sea ice conditions and atmospheric patterns. The length of the summer open-water (OW) season (i.e., the period of time the sea ice is mostly absent from the continental shelf) increased at a rate of 18 days decade-1 over the study period. However, the OW season duration was not a strong determinant of spring residual body mass or litter mass. Residual body mass of independent (i.e., subadults and adults) female bears varied relative to age class, reproductive status, and the strength of the prior winter’s Arctic Oscillation (i.e., a circumpolar-scale mode of climate variability driven by long-term atmospheric patterns). Spring residual mass of independent males varied with age class and variation in wind speed (i.e., regional-scale short-term atmospheric patterns) during the winter of the year preceding capture. Over the study period, mean annual body mass of adult females unaccompanied by COY declined by 4 kg/ decade-1, while no temporal trends were evident in the mean annual body mass of adult females with COY, adult males, and subadults. Litter mass of COY varied relative to capture date, maternal age class and mass, litter size, and year of capture. Litter mass of YRL varied with capture date, maternal age class and mass, litter size, variation in winter wind speed (the year of and year preceding capture), and the strength of the prior winter’s Arctic Oscillation. Mean annual litter mass of COY decreased at a rate of 2.6 kg decade-1 and declined 0.68 kg for every 10 kg reduction in maternal mass. No trend was evident in the mean annual litter mass of yearlings. These findings suggest a nuanced response of the southern Beaufort Sea polar bears to environmental change, where some demographic groups (e.g., adult males and subadults) are presently more resilient than others to changes in the Arctic marine ecosystem.

Abstract Animals responding to habitat loss and fragmentation may increase their home ranges to offset declines in localized resources or they may decrease their home ranges and switch to alternative resources. In many regions of the Arctic, polar bears (Ursus maritimus) exhibit some of the largest home ranges of any quadrupedal mammal. Polar bears are presently experiencing a rapid decline in Arctic sea ice extent and a change in sea ice composition. For the Southern Beaufort Sea subpopulation of polar bears, this has resulted in a divergent movement pattern where most of the subpopulation remains on the sea ice in the summer melt season while the remainder move to land. We evaluated the effects of summer land use and maternal denning on the annual and seasonal utilization distribution size (i.e., home range) of adult female polar bears in the Southern Beaufort Sea subpopulation over 30 yr (1986–2016) during a period of rapid sea ice decline. For bears that remained on the summer sea ice, model‐derived mean annual utilization distributions were 64% larger in 1999–2016 (x̄ = 176,000 km2) relative to 1986–1998 (x̄ = 107,000 km2). This increase was primarily driven by increases in summer utilization distributions that encompassed increased amounts of open water and decreased amounts of preferred sea ice. The mean centroid of summer utilization distributions for bears that remained on the sea ice was 193 km further north‐northeast in 1999–2016. In contrast, bears that summered on land during 1999–2016 exhibited 88% smaller mean annual utilization distribution sizes (x̄ = 22,000 km2) relative to bears that remained on the summer sea ice during the same period. Our findings highlight the impacts of sea ice declines on polar bear space use and the increasing importance of land as an alternative summer refuge.

Introduction Neural stem/progenitor cells derived from olfactory neuroepithelium (hereafter OE-NS/PCs) are emerging as a viable proxy and a valuable tool for translational studies on severe mental illnesses (SMI). In this respect, the use of OE-NS/PCs as a surrogate cellular model of schizophrenia (SZ) has enabled insights into cell signaling and cell cycle dynamics in this disease. Objectives We explored whether mitochondrial dysfunction, which has been already associated with SZ, may have a role in the altered proliferation pattern previously observed in OE-NS/PCs of SZ patients. Methods OE-NS/PCs were collected from 20 patients and 20 healthy controls (Hcs) by nasal brushing, cultured in proper medium and expanded. Fresh OE-NS/PCs at passage 3 of both groups underwent BrdU proliferation assays or were frozen for later use. Mitochondrial ATP production was measured in both fresh and thawed OE-NS/PCs by using the ATPlite Luminescence Assay kit. The western tropical South Pacific (WTSP) is one of the most understudied oceanic regions in terms of the planktonic food web, despite supporting some of the largest tuna fisheries in the world. In this stratified oligotrophic ocean, nitrogen fixation may play an important role in supporting the plankton food web and higher trophic level production. In the austral summer (February–April) of 2015, the OUTPACE (Oligotrophy to UlTra-oligotrophy PACific Experiment) project conducted a comprehensive survey of 4000 km along 20∘ S, from New Caledonia to Tahiti, to determine the role of N2 fixation on biogeochemical cycles and food web structure in this region. Here, we characterize the zooplankton community and plankton food web processes at 15 short-duration stations (8 h each) to describe the large-scale variability across trophic gradients from oligotrophic waters around Melanesian archipelagoes (MAs) to ultra-oligotrophic waters of the South Pacific gyre (GY). Three long-duration stations (5 days each) enabled a more detailed analysis of processes and were positioned (1) in offshore northern waters of New Caledonia (MA), (2) near Niue Island (MA), and (3) in the subtropical Pacific gyre (GY) near the Cook Islands. At all stations, meso-zooplankton was sampled with a bongo net with 120 µm mesh size to estimate abundance, biomass, community taxonomy and size structure, and size fractionated δ15N. Subsequently, we estimated zooplankton carbon demand, grazing impact, excretion rates, and the contribution of diazotroph-derived nitrogen (DDN) to zooplankton biomass. The meso-zooplankton community showed a general decreasing trend in abundance and biomass from west to east, with a clear drop in the GY waters. Higher abundance and biomass corresponded to higher primary production associated with complex mesoscale circulation in the Coral Sea and between 170–180∘ W. The taxonomic structure showed a high degree of similarity in terms of species richness and abundance distribution across the whole region, with, however, a moderate difference in the GY region, where the copepod contribution to meso-zooplankton increased. The calculated ingestion and metabolic rates allowed us to estimate that the top–down (grazing) and bottom–up (excretion of nitrogen and phosphorous) impact of zooplankton on phytoplankton was potentially high. Daily grazing pressure on phytoplankton stocks was estimated to remove 19 % to 184 % of the total daily primary production and 1.5 % to 22 % of fixed N2. The top–down impact of meso-zooplankton was higher in the eastern part of the transect, including GY, than in the Coral Sea region and was mainly exerted on nano- and micro-phytoplankton. The regeneration of nutrients by zooplankton excretion was high, suggesting a strong contribution to regenerated production, particularly in terms of N. Daily NH4+ excretion accounted for 14.5 % to 165 % of phytoplankton needs for N, whereas PO43- excretion accounted for only 2.8 % to 34 % of P needs. From zooplankton δ15N values, we estimated that the DDN contributed to up to 67 % and 75 % to the zooplankton biomass in the western and central parts of the MA regions, respectively, but strongly decreased to an average of 22 % in the GY region and down to 7 % in the easternmost station. Thus, the highest contribution of diazotrophic microorganisms to zooplankton biomass occurred in the region of highest N2 fixation rates and when Trichodesmium dominated the diazotrophs community (MA waters). Our estimations of the fluxes associated with zooplankton were highly variable between stations and zones but very high in most cases compared to literature data, partially due to the high contribution of small forms. The highest values encountered were found at the boundary between the oligotrophic (MA) and ultra-oligotrophic regions (GY). Within the MA zone, the high variability of the top–down and bottom–up impact was related to the high mesoscale activity in the physical environment. Estimated zooplankton respiration rates relative to primary production were among the highest cited values at similar latitudes, inducing a high contribution of migrant zooplankton respiration to carbon flux. Despite the relatively low biomass values of planktonic components in quasi-steady state, the availability of micro- and macronutrients related to physical mesoscale patterns in the waters surrounding the MA, the fueling by DDN, and the relatively high rates of plankton production and metabolism estimated during OUTPACE may explain the productive food chain ending with valuable fisheries in this region.

Zooplankton play a key role in the regeneration of nitrogen and phosphorus in the ocean through grazing and metabolism. This study investigates the role of the organic and inorganic nitrogen and phosphorus compounds released by copepods on biogeochemical processes and on the microbial community composition during the OUTPACE cruise (18 February–3 April 2015) at three long-duration stations (LD). Two LD stations were located in the Melanesian Archipelago region (MA; LD A and LD B) and one in the South Pacific Gyre (SG; LD C), which represent oligotrophic and ultra-oligotrophic regions respectively. At each station, on-board microcosm experiments were performed with locally sampled organisms, comprising a mix of epipelagic copepods fed with their natural food and then incubated along with wild microbial assemblages. In the presence of copepods, ammonium and dissolved organic nitrogen showed a significant increase compared to a control in two situations: in ammonium concentration (rate: 0.29 µmol L−1 h−1 after 4 h of incubation) in LD C and in dissolved organic nitrogen concentration (rate: 2.13 µmol L−1 h−1 after 0.5 h of incubation) in LD A. In addition, during the three experiments, an enhanced remineralization (ammonification and nitrification) was observed when adding copepods compared to the controls. A shift in the composition of the active bacterial community was observed for the experiments in LD A and LD B, which were mainly characterized by an increase in Alteromonadales and SAR11, respectively, and linked with changes in nutrient concentrations. In the experiment performed in LD C, both groups increased but at different periods of incubation. Alteromonadales increased between 1 and 2 h after the beginning of the experiment, and SAR 11 at the end of incubation. Our results in near in situ conditions show that copepods can be a source of organic and inorganic compounds for bacterial communities, which respond to excretion pulses at different timescales, depending on the initial environmental conditions and on their community composition. These processes can significantly contribute to nutrient recycling and regenerated production in the photic zone of ultra-oligotrophic and oligotrophic oceanic regions.

Abstract Background Satellite telemetry studies provide information that is critical to the conservation and management of species affected by ecological change. Here we report on the performance and retention of two types (SPOT-227 and SPOT-305A) of ear-mounted Argos-linked satellite transmitters (i.e., platform transmitter terminal, or PTT) deployed on free-ranging polar bears in Eastern Greenland, Baffin Bay, Kane Basin, the southern Beaufort Sea, and the Chukchi Sea during 2007–2013. Results Transmissions from 142 out of 145 PTTs deployed on polar bears were received for an average of 69.3 days. The average functional longevity, defined as the number of days they transmitted while still attached to polar bears, for SPOT-227 was 56.8 days and for SPOT-305A was 48.6 days. Thirty-four of the 142 (24%) PTTs showed signs of being detached before they stopped transmitting, indicating that tag loss was an important aspect of tag failure. Furthermore, 10 of 26 (38%) bears that were re-observed following application of a PTT had a split ear pinna, suggesting that some transmitters were detached by force. All six PTTs that were still on bears upon recapture had lost the antenna, which indicates that antenna breakage was a significant contributor to PTT failure. Finally, only nine of the 142 (6%) PTTs—three of which were still attached to bears—had a final voltage reading close to the value indicating battery exhaustion. This suggests that battery exhaustion was not a major factor in tag performance. Conclusions The average functional longevity of approximately 2 months for ear-mounted PTTs (this study) is poor compared to PTT collars fitted to adult female polar bears, which can last for several years. Early failure of the ear-mounted PTTs appeared to be caused primarily by detachment from the ear or antenna breakage. We suggest that much smaller and lighter ear-mounted transmitters are necessary to reduce the risk of tissue irritation, tissue damage, and tag detachment, and with a more robust antenna design. Our results are applicable to other tag types (e.g., iridium and VHF systems) and to research on other large mammals that cannot wear radio collars.

We report a novel case of an infant with neurofibromatosis type 1 (NF1) who presented with new onset presumed focal impaired awareness seizures with motor onset followed by rapid progression to infantile spasms (IS). Electroencephalography (EEG) captured evolution from focal epileptiform discharges to multifocal and generalized discharges, then to hypsarrhythmia over three days. Development of IS within days of focal seizure onset is rapid, and to our knowledge, has not been demonstrated electrographically. The pattern of rapid ictal transition to hypsarrhythmia is essential for neurologists to be able to recognize as it can help lead to early treatment, which is necessary for improved outcomes in IS.

Animal structural body size and condition are often measured to evaluate individual health, identify responses to environmental change and food availability, and relate food availability to effects on reproduction and survival. A variety of condition metrics have been developed but relationships between these metrics and vital rates are rarely validated. Identifying an optimal approach to estimate the body condition of polar bears is needed to improve monitoring of their response to decline in sea ice habitat. Therefore, we examined relationships between several commonly used condition indices (CI), body mass, and size with female reproductive success and cub survival among polar bears (Ursus maritimus) measured in two subpopulations over three decades. To improve measurement and application of morphometrics and CIs, we also examined whether CIs are independent of age and structural size-an important assumption for monitoring temporal trends-and factors affecting measurement precision and accuracy. Maternal CIs and mass measured the fall prior to denning were related to cub production. Similarly, maternal CIs, mass, and length were related to the mass of cubs or yearlings that accompanied her. However, maternal body mass, but not CIs, measured in the spring was related to cub production and only maternal mass and length were related to the probability of cub survival. These results suggest that CIs may not be better indicators of fitness than body mass in part because CIs remove variation associated with body size that is important in affecting fitness. Further, CIs exhibited variable relationships with age for growing bears and were lower for longer bears despite body length being related to cub survival and female reproductive success. These results are consistent with findings from other species indicating that body mass is a useful metric to link environmental conditions and population dynamics.

Tracking data have led to evidence-based conservation of marine megafauna, but a disconnect remains between the many 1000s of individual animals that have been tracked and the use of these data in conservation and management actions. Furthermore, the focus of most conservation efforts is within Exclusive Economic Zones despite the ability of these species to move 1000s of kilometers across multiple national jurisdictions. To assist the goal of the United Nations General Assembly’s recent effort to negotiate a global treaty to conserve biodiversity on the high seas, we propose the development of a new frontier in dynamic marine spatial management. We argue that a global approach combining tracked movements of marine megafauna and human activities at-sea, and using existing and emerging technologies (e.g., through new tracking devices and big data approaches) can be applied to deliver near real-time diagnostics on existing risks and threats to mitigate global risks for marine megafauna. With technology developments over the next decade expected to catalyze the potential to survey marine animals and human activities in ever more detail and at global scales, the development of dynamic predictive tools based on near real-time tracking and environmental data will become crucial to address increasing risks. Such global tools for dynamic spatial and temporal management will, however, require extensive synoptic data updates and will be dependent on a shift to a culture of data sharing and open access. We propose a global mechanism to store and make such data available in near real-time, enabling a holistic view of space use by marine megafauna and humans that would significantly accelerate efforts to mitigate impacts and improve conservation and management of marine megafauna.

Este trabajo describe la optimización del proceso de esterificación de aceites vegetales usados (AVUs) para la producción de biodiesel a partir de un análisis estructural mediante simulación . L a optimización energética se determin ó mediante una programación matemática, y posterior resolución mediante el software GAMS ® . Se analizaron dos alternativas tecnológicas . La Alternativa 1 incluye el proceso de esterificación aplicando una torre de lavado a la salida del reactor , mientras que la Alternativa 2 aplica un decantador a la salida del reactor . Se determinó que la A lternativa 2 fue la mejor opción debido a que el costo de servicio s totales anual es fue menor. Para la optimización energética del proceso seleccionado , se incorporó una bomba de calor entre las corrientes del condensador y del rehervidor de la torre de destilación y se diseñó una red de intercambio calórico entre las corrientes del proceso , esto permitiría un ahorro energético de servicios externos de 64,21% .

In oligotrophic tropical and subtropical oceans, where strong stratification can limit the replenishment of surface nitrate, dinitrogen (N2) fixation by diazotrophs can represent a significant source of nitrogen (N) for primary production. The VAHINE (VAriability of vertical and tropHIc transfer of fixed N2 in the south-wEst Pacific) experiment was designed to examine the fate of diazotroph-derived nitrogen (DDN) in such ecosystems. In austral summer 2013, three large ( ∼ 50 m3) in situ mesocosms were deployed for 23 days in the New Caledonia lagoon, an ecosystem that typifies the low-nutrient, low-chlorophyll environment, to stimulate diazotroph production. The zooplankton component of the study aimed to measure the incorporation of DDN into zooplankton biomass, and assess the role of direct diazotroph grazing by zooplankton as a DDN uptake pathway. Inside the mesocosms, the diatom–diazotroph association (DDA) het-1 predominated during days 5–15 while the unicellular diazotrophic cyanobacteria UCYN-C predominated during days 15–23. A Trichodesmium bloom was observed in the lagoon (outside the mesocosms) towards the end of the experiment. The zooplankton community was dominated by copepods (63 % of total abundance) for the duration of the experiment. Using two-source N isotope mixing models we estimated a mean ∼ 28 % contribution of DDN to zooplankton nitrogen biomass at the start of the experiment, indicating that the natural summer peak of N2 fixation in the lagoon was already contributing significantly to the zooplankton. Stimulation of N2 fixation in the mesocosms corresponded with a generally low-level enhancement of DDN contribution to zooplankton nitrogen biomass, but with a peak of ∼ 73 % in mesocosm 1 following the UCYN-C bloom. qPCR analysis targeting four of the common diazotroph groups present in the mesocosms (Trichodesmium, het-1, het-2, UCYN-C) demonstrated that all four were ingested by copepod grazers, and that their abundance in copepod stomachs generally corresponded with their in situ abundance. 15N2 labelled grazing experiments therefore provided evidence for direct ingestion and assimilation of UCYN-C-derived N by the zooplankton, but not for het-1 and Trichodesmium, supporting an important role of secondary pathways of DDN to the zooplankton for the latter groups, i.e. DDN contributions to the dissolved N pool and uptake by nondiazotrophs. This study appears to provide the first evidence of direct UCYN-C grazing by zooplankton, and indicates that UCYN-C-derived N contributes significantly to the zooplankton food web in the New Caledonia lagoon through a combination of direct grazing and secondary pathways.

A healthy dehydrated food of high nutritional-quality and added-value was developed: low-calories raisin obtained by an ultrasonic assisted combined-dehydration with two-stage osmotic treatment (D3S) complemented by drying. Pink Red Globe grape produced at Mendoza (Argentina), experienced a substitution of sugar by natural sweetener Stevia in two osmotic stages under different conditions (treatment with/without ultrasound; sweetener concentration 18, 20, 22% w/w; time 35, 75, 115 minutes), evaluating soluble solids (SS), moisture (M), total polyphenols (PF), antioxidant efficiency (AE) and sugar profile. The multiple optimization of the process by response surface methodology and desirability analysis, allowed to minimize M, maximize SS (Stevia incorporation), and preserve the maximum amount of PF. After the first stage, the optimal treatment reduced the majority sugars of the grape in 32% (sucrose, glucose), and the 57% at the end of the dehydration process.

Se desarrolló un alimento deshidratado saludable de eleva da calidad nutricional y valor agregado: uva pasa de bajo contenido calórico, por deshidratación combinada asistida por ultrasonido mediante un tratamiento osmótico dual (D3S) de dos etapas complementado por secado. U va rosada Red Globe producida en Mendoz a (Argentina) se sometió a sustitución de sus azúcares por edulcorante natural Stevia en dos etapas osmóticas bajo diferentes condiciones ( tratamiento con/sin ultrasonido; concentración del edulcorante 18, 20, 22%p/p ; tiempo 35, 75 , 115 minutos) , evaluando sólidos solubles (SS), humedad (M) , polifenoles totales ( P F), eficiencia antioxidante (A E ) y perfil de azúcares. L a optimización múltiple del proceso por la metodología de superficie de respuesta y análisis de deseabilidad , permitió minimiza r M, maximizan do SS ( incorporación de Stevia ) y conservando la máxima cantidad PF. El tratamiento óptimo redujo un 32% los azúcares mayoritarios de la uva (sacarosa, glucosa) tras la primera etapa, y el 57% de ellos al final del proceso.

The beer brewing process is one of the most polluting industrial processes, generating a huge amount of wastewater effluent and solid wastes (i.e. spent grain and yeast), which must be disposed of or treated in the least costly way to meet strict discharge regulations set by government entities. Particularly, spent grain, the leftover malt and adjuncts after the mash has extracted most of the sugars, proteins, and nutrients, can constitute as much as 85 % of a brewery’s total by-product. However, sustainability is a hallmark of the craft beer industry and therefore there is a great interest to find innovative ways to prevent spent grain from going to waste. At this regard, the “Birraverde” project, coordinated by CREA and funded by the National Rural Network, aimed to develop some technical solutions for the recovery and reuse of brewer’s wastes (spent grain, spent yeast and wastewaters), according to the principles of circular economy. In particular, for the recovery and valorisation of spent grain two alternative solutions were proposed: 1) conversion of dried spent grain into pellets that can be used for heat generation to be reused in the beer production cycle; 2) production of biochar (charcoal) from dried spent grain through a thermochemical process of pyro-gasification. In the latter case, a preliminary trial was carried out using a lab-scale pyrolytic reactor. The first proposed model, if integrated within the brewing process pipeline, can reduce the costs of buying pellets on the market, producing gain margins even under low rate of plant utilisation (160 h y-1). On the other hand, the second model, the conversion of spent grain into biochar, a new organic amendmen, can offer the possibility to realize a diversification of the farm activities opening to new markets.

This paper presents the heat exchanger networks (HEN) application to the esterification of used vegetable oils (UVOs) with acid catalysis for the biodiesel production. Different methods are applied to obtain minimum heat quantities and the HEN for a processes capacity of 0,19 kg/s of UVOs. A minimum temperature difference (∆Tmin) of 10 °C is considered and the minimum requirements of warming and cooling, resulting in 4629, 87 W and 10066, 30 W, respectively. When the heat exchanger net is applied, the required steam service in the process decreases in 78,92% and the cooling water service in 62,48%, demonstrating that the integration reduces the energetic requirements in relation to the non-integrated process

ABSTRACT Historically, biologists used brood‐pair ratios (BPRs) as an index to waterfowl productivity to help guide management decisions and evaluate conservation practices. However, BPRs are biased by imperfect detection probabilities, especially for broods. We conducted roadside surveys for breeding waterfowl pairs on 7–8 study sites in the springs of 2006–2008 in northeastern North Dakota, USA. Later each year, we conducted replicate counts of broods on the same wetlands and used mark–recapture methods to estimate sightability‐adjusted BPRs (SA‐BPRs). Traditional roadside brood surveys detected only 30–45% of the available broods, depending on species. We explored the potential for using SA‐BPRs to measure hen success (i.e., the probability a female hatches ≥1 egg across all nesting attempts) for mallards (Anas platyrhynchos) and other upland‐nesting dabbling ducks (Anas spp.). We found that SA‐BPRs explained 40% of the variation in hen success over 5 species of dabbling ducks, and we were able to detect an effect of predator reduction on hen success in combined dabblers, but not in mallards alone. However, we found no relationship between SA‐BPRs and mallard fledging rates (hen success × initial brood size × duckling survival). Our results suggest that SA‐BPRs can provide a cost‐effective alternative to traditional measures of productivity such as nesting success, but not to measures of duckling survival. Nevertheless, SA‐BPRs may be useful in areas where traditional measures of waterfowl productivity are logistically or financially challenging. © 2014 The Wildlife Society.

E n este trabajo se presenta la aplicación de redes de intercambio de calor (RIC) a la esterificación de aceites vegetales usados (AVUs ) con catálisis ácida para la producción de biodiesel. Se aplican distintos métodos para obtener las cantidades mínimas de calor y la RIC para una capacidad de procesamiento de 0,19 kg/s de AVUs. Se considera una diferencia de temperatura mínima (∆Tmin) de 10°C y los requerimientos mínimos de calentamiento y enfriamiento, resultan ser de 4629,87 W y de 10066,30 W, respectivamente. Al aplicar la red de intercambio calórico disminuye en un 78,92% el servicio de vapor requerido en el proceso y en un 62,48% el servicio de agua de enfriamiento, lo que demuestra que la integración reduce los requerimientos energéticos respecto al proceso no integrado.

Se estudió el efecto de la humedad sobre propiedades físicas de granos de maíz Colorado Duro en el rango 17− 35% base seca, y la cinética de secado a temperaturas entre 30− 90°C y a velocidad de 0.43 m/s. El 12.91x10-3 m), diámetro contenido de humedad tuvo efecto significativo sobre la longitud (12.059x10-3− -3 -3 -3 -3 geométrico (7.8x10 − 8.281x10 m), diámetro equivalente (8.18x10 − 8.63x10 m), peso de mil granos (363.6− 421.9 g), ángulo de reposo (10.11− 24.66°), y densidades aparente (0.801− 0.684 g/mL) y real (1.339− 1.170 g/mL); pero el ancho, espesor, porosidad y esfericidad no se vieron significativamente afectados. El secado ocurrió en el período de velocidad decreciente, siendo el tiempo de secado y la temperatura las variables significativas sobre la humedad del grano. Los modelos de Page, ecuación difusiva de “tiempos cortos” y una modificación introducida a éste ultimo resultaron adecuados (R2>0.99) para describir los datos. Los coeficientes de difusión de agua estuvieron entre 1.907x10-11 y 6.495x10-11 m2/s, y la energía de activación fue 17.74 kJ/mol.

The diversity and distribution of epipelagic metazooplankton across the Mediterranean Sea was studied along a 3000 km long transect from the eastern to the western basins during the BOUM cruise in summer 2008. Metazooplankton were sampled using both a 120 μm mesh size bongo net and Niskin bottles in the upper 200 m layer at 17 stations. Here we report on the stock, the composition and the structure of the metazooplankton community. The abundance was 4 to 8 times higher than in several previously published studies, whereas the biomass remained within the same order of magnitude. An eastward decrease in abundance was evident, although biomass was variable. Spatial (horizontal and vertical) distribution of metazooplankton abundance and biomass was strongly correlated to chlorophyll-a concentration. In addition, a clear association was observed between the vertical distribution of nauplii and small copepods and the depth of the deep chlorophyll maximum. The distinction between the communities of the eastern and western basins was clearly explained by the environmental factors. The specific distribution pattern of remarkable species was also described.

SELEX, the process of selecting aptamers, is often hampered by the difficulty of preparing target molecules in their native forms and by a lack of a simple yet quantitative assay for monitoring enrichment and affinity of reactive aptamers. In this study, we sought to discover DNA aptamers against human serum markers for potential therapeutic and diagnostic applications. To circumvent soluble expression and immobilization for performing SELEX, we ectopically expressed soluble growth factors on the surface of yeast cells to enable cell-SELEX and devised a flow cytometry-based method to quantitatively monitor progressive enrichment of specific aptamers. High-throughput sequencing of selected pools revealed that the emergence of highly enriched sequences concurred with the increase in the percentage of reactive aptamers shown by flow cytometry. Particularly, selected DNA aptamers against VEGF were specific and of high affinity (K(D) = ∼ 1 nM) and demonstrated a potent inhibition of capillary tube formation of endothelial cells, comparable to the effect of a clinically approved anti-VEGF antibody drug, bevacizumab. Considering the fact that many mammalian secretory proteins have been functionally expressed in yeast, the strategy of implementing cell-SELEX and quantitative binding assay can be extended to discover aptamers against a broad array of soluble antigens.

The four-subunit Negative Elongation Factor (NELF) is a major regulator of RNA Polymerase II (Pol II) pausing. The subunit NELF-E contains a conserved RNA Recognition Motif (RRM) and is proposed to facilitate Poll II pausing through its association with nascent transcribed RNA. However, conflicting ideas have emerged for the function of its RNA binding activity. Here, we use in vitro selection strategies and quantitative biochemistry to identify and characterize the consensus NELF-E binding element (NBE) that is required for sequence specific RNA recognition (NBE: CUGAGGA(U) for Drosophila). An NBE-like element is present within the loop region of the transactivation-response element (TAR) of HIV-1 RNA, a known regulatory target of human NELF-E. The NBE is required for high affinity binding, as opposed to the lower stem of TAR, as previously claimed. We also identify a non-conserved region within the RRM that contributes to the RNA recognition of Drosophila NELF-E. To understand the broader functional relevance of NBEs, we analyzed promoter-proximal regions genome-wide in Drosophila and show that the NBE is enriched +20 to +30 nucleotides downstream of the transcription start site. Consistent with the role of NELF in pausing, we observe a significant increase in NBEs among paused genes compared to non-paused genes. In addition to these observations, SELEX with nuclear run-on RNA enrich for NBE-like sequences. Together, these results describe the RNA binding behavior of NELF-E and supports a biological role for NELF-E in promoter-proximal pausing of both HIV-1 and cellular genes.
