Summary: In leaves of C4 plants, the reactions of photosynthesis become restricted between two compartments. Typically, this allows accumulation of C4 acids in mesophyll (M) cells and subsequent decarboxylation in the bundle sheath (BS). In C4 grasses, proliferation of plasmodesmata between these cell types is thought to increase cell‐to‐cell connectivity to allow efficient metabolite movement. However, it is not known whether C4 dicotyledons also show this enhanced plasmodesmal connectivity and so whether this is a general requirement for C4 photosynthesis is not clear. How M and BS cells in C4 leaves become highly connected is also not known.We investigated these questions using 3D‐ and 2D‐electron microscopy on the C4 dicotyledon Gynandropsis gynandra as well as phylogenetically close C3 relatives.The M–BS interface of C4G. gynandra showed higher plasmodesmal frequency compared with closely related C3 species. Formation of these plasmodesmata was induced by light. Pharmacological agents that perturbed photosynthesis reduced the number of plasmodesmata, but this inhibitory effect could be reversed by the provision of exogenous sucrose.We conclude that enhanced formation of plasmodesmata between M and BS cells is wired to the induction of photosynthesis in C4G. gynandra. Like humans, many felid species suffer from teratozoospermia and frequently produce low numbers of normal spermatozoa. Male fertility can be affected by oxidative and dicarbonyl stress. Because of the high level of glycolytic activity in testes, reactive dicarbonyl metabolites may arise as side-products of glycolysis; their generation is further promoted by oxidative stress. Alpha-oxoaldehydes, including methylglyoxal (MG), are reactive dicarbonyl metabolites and substrates for the formation of advanced glycation end products. Elevated levels of both may lead to dicarbonyl stress and cause cellular dysfunction. However, MG and other α-oxoaldehydes can be converted to less dangerous molecules via the glyoxalase pathway. In this pathway, α-oxoaldehydes react with glutathione (GSH), forming a thioacetal, which becomes metabolized by glyoxalase I (GLO I) to S-D-lactoyl-glutathione (SLG). Glyoxalase II (GLO II) converts SLG to d -lactate upon the release of GSH. Nothing is known about the glyoxalase system in the feline testis and its capacity to mitigate an excess of dicarbonyl metabolites. To study whether GLO I and GLO II are present and have a specific function in the testis of the domestic cat, the gene expression of both enzymes were analyzed in testis samples of different developmental stages (prepubertal, pubertal, postpubertal). Furthermore, the presence of GLO I and GLO II proteins was investigated via immunohistochemistry. The GLO I gene expression does not change between developmental stages. Immunohistochemistry revealed strong signals for GLO I in the cytoplasm and nuclei of Sertoli and Leydig cells during all developmental stages. GLO I was described as catalyzing the rate-limiting step in the glyoxalase pathway. This implies a function on the part of this enzyme of sustaining the homeostasis of somatic testicular cells. For GLO II, we observed stage-dependent mRNA expression, which was significantly increased after puberty. In accordance with this observation, clear immunohistochemical GLO II signals were observed in nuclei of individual germ cells. The most intense signals were visible in spermatocytes. The different localizations of the strong GLO I and GLO II signals indicate that GLO II, in addition to the classical glyoxalase pathway, may have additional functions in meiotic germ cells, for example, providing lactate as an energy substrate and/or GSH as an antioxidant. On their way to the oocyte, sperm cells are subjected to oxidative stress, which may trigger the oxidation of phospholipids (PL). Applying MALDI-TOF MS, HPTLC and ESI-IT MS, we comparatively analyzed the PL compositions of semen and blood of species differing in their reproductive systems and types of nutrition (bull, boar, stallion, lion and man) with regard to the sensitivity to oxidation as well as the accumulation of harmful lyso-PL (LPL), transient products of lipid oxidation. In addition, the protective capacity of seminal fluid (SF) was also examined. The PL composition of erythrocytes and blood plasma is similar across the species, while pronounced differences exist for sperm and SF. Since the blood function is largely conserved across mammalian species, but the reproductive systems may vary in many aspects, the obtained results suggest that the PL composition is not determined by the type of nutrition, but by the relatedness of species and by functional requirements of cell membranes such as fluidity. Sperm motion and fertilization of oocytes require a rather flexible membrane, which is accomplished by significant moieties of unsaturated fatty acyl residues in sperm lipids of most species, but implies a higher risk of oxidation. Due to a high content of plasmalogens (alkenyl ether lipids), bull sperm are most susceptible to oxidation. Our data indicate that bull sperm possess the most effective protective power in SF. Obviously, a co-evolution of PL composition and protective mechanisms has occurred in semen and is related to the reproductive characteristics. Although the protective capacity in human SF seems well developed, we recorded the most pronounced individual contaminations with LPL in human semen. Perennial ryegrass (Lolium perenne L.) is a widely sown pasture species in temperate regions. Under heat and moisture-deficit stress, its production is suppressed and tiller mortality increases, particularly when plants are simultaneously exposed to other stresses such as intense defoliation. We investigated the effects of extended grazing exclusion periods in spring and summer on herbage and soils of rotationally grazed, perennial ryegrass-dominant pastures at a summer-wet site and a summer-dry site in sheep and beef hill country in New Zealand. Treatments comprised 'grazed' (conventional rotational grazing), 'early opening' (grazing exclusion from mid-spring to mid-summer to allow perennial ryegrass to flower), and 'deferred grazing' (grazing exclusion from mid-spring to late summer to allow perennial ryegrass to flower and set seed). At both sites, deferred grazing increased perennial ryegrass tiller density and total vegetation cover, and reduced the abundance of broadleaf species and Pseudopithomyces chartarum spores for up to 15 months after exclusion compared with the grazed control. Herbage production was reduced during the deferred period but there was a growth surge thereafter so that herbage production over the study period was similar in the deferred and grazed treatments. Pasture nutritive values were lower in the deferred than the grazed treatment during the exclusion period but were similar in all treatments by the end of the following winter. In the autumn after deferring, seedlings contributed ~50% of the perennial ryegrass tillers present at the summer-dry site but only ~10% at the summer-wet site, where regrowth tillering of existing plants contributed a high proportion of the perennial ryegrass tillers present. Overall, early opening had few effects on the pasture, whereas deferred grazing had numerous positive effects on the pasture but only short-term effects on soil quality. We investigated the impacts of deferred grazing (livestock excluded from mid-spring to late summer to enable perennial ryegrass to flower and set seed), early opening (livestock excluded from mid-spring to early summer to enable perennial ryegrass to flower) and standard rotational grazing on hill country pastures and soils under beef and sheep grazing. Compared with standard rotational grazing, deferred grazing had positive effects on pasture productivity but only short-term effects on soil quality, whereas early opening had few effects on the pastures or soils. Cryobanking of gametes in combination with artificial insemination is an essential option to support conservation programmes for endangered and threatened species. About two‐thirds of the felid species are classified as 'near threatened', 'vulnerable' or 'endangered' (www.cites.org), and mostly, epididymal sperm are collected from euthanized or castrated male felids and cryopreserved. However, epididymal compared with ejaculated and cryopreserved compared with fresh sperm have a limited potential to fertilize if vaginal non‐surgical insemination is applied in feline species. Missing or highly diluted seminal fluid in epididymal and cryopreserved sperm, as well as a potential interference of extender ingredients with the natural interactive properties of sperm in the female genital tract is discussed as potential drawback which hampers a proper sperm transit and fertilization besides the limited longevity of cryopreserved feline sperm. Individual components in seminal fluid as well as cryoextenders may adversely alter sperm properties and have a different impact on fertility and preservation success. The identification and investigation of beneficial as well as detrimental components is a precondition to deduce options for improving the process of cryopreservation in felids, particularly, if only epididymal sperm are available. Metaphors are linguistically dense images that transfer terms from their original usage to a different context and describe actions and objects beyond their literal meaning. This article uses Rudolf Schmitt's metaphor analysis (2017) to gain insight into the experiences of the so-called GDR children of Namibia. This term refers to a group of approximately 430 people who, as part of a solidarity project between the South West Africa People's Organisation (SWAPO) and the German Democratic Republic (GDR), were brought to the GDR between 1979 and 1989 to be trained as the elite for a future liberated Namibia. They grew up and attended school in the GDR until they were returned to Namibia in August 1990. Based on narrative interviews, we use metaphor analysis to show how the now-adult 'GDR children' experienced their lives between different national contexts. The interviewees talk about their collective education, feelings of heteronomy and the family bond that existed among the children themselves and between the children and their care staff. The spermadhesin AQN-3 is a major component of porcine seminal plasma. While various studies suggest that this protein binds to boar sperm cells, its attachment to the cells is poorly understood. Therefore, the capacity of AQN-3 to interact with lipids was investigated. For that purpose, AQN-3 was recombinantly expressed in E. coli and purified via the included His-tag. Characterizing the quaternary structure by size exclusion chromatography revealed that recombinant AQN-3 (recAQN-3) is largely present as multimer and/or aggregate. To determine the lipid specificity of recAQN-3, a lipid stripe method and a multilamellar vesicle (MLV)-based binding assay were used. Both assays show that recAQN-3 selectively interacts with negatively charged lipids, like phosphatidic acid, phosphatidylinositol phosphates, and cardiolipin. No interaction was observed with phosphatidylcholine, sphingomyelin, phosphatidylethanolamine, or cholesterol. The affinity to negatively charged lipids can be explained by electrostatic interactions because binding is partly reversed under high-salt condition. However, more factors have to be assumed like hydrogen bonds and/or hydrophobic forces because the majority of bound molecules was not released by high salt. To confirm the observed binding behavior for the native protein, porcine seminal plasma was incubated with MLVs comprising phosphatidic acid or phosphatidyl-4,5-bisphosphate. Attached proteins were isolated, digested, and analyzed by mass spectrometry. Native AQN-3 was detected in all samples analyzed and was – besides AWN – the most abundant protein. It remains to be investigated whether AQN-3, together with other sperm associated seminal plasma proteins, acts as decapacitation factor by targeting negative lipids with signaling or other functional roles in fertilization. The effects of soil water repellency (SWR) on runoff and nutrient losses are difficult to isolate. Hydrophobic organic substances coating soil particles can severely delay water infiltration and enhance runoff. We used a portable run-on simulator to investigate the effect of SWR on runoff and nutrient loss. Intact soil slabs, 0.48 m long and 0.19 m wide, were collected from a severely water-repellent Andosol under pasture. One day before simulating 60-min long run-on events with an intensity of 60 mm h −1 , superphosphate was applied at a rate of 45 kg P ha −1 . The effects of SWR were quantified by comparing runoff volumes and nutrient losses from run-on events conducted with water and a fully wetting aqueous ethanol solution as run-on liquids. Further, through conducting multiple consecutive water run-on events with the same soil slab, the hypothesis, that SWR is lost through the washing off of hydrophobic materials, was tested. Finally, runoff dynamics were visualised by adding a dye to the run-on water. In the first run-on experiment, 88% of the water applied was captured as runoff, while no runoff was observed when aqueous ethanol was used as run-on liquid, providing strong evidence that SWR governed runoff generation from this Andosol. In consecutive water run-on experiments, approximately 23% of the applied P was recovered in the runoff from the first event, while the cumulative P loss over ten consecutive run-on events was around 30% of the applied P. This confirms that nutrient losses were associated with SWR and the occurrence of runoff. After ten consecutive run-on events, the persistence of both actual and potential SWR in areas of the slab that had been wetted were significantly ( p  < 0.5) reduced. But the persistence of potential SWR of the soil was still classified as severe, suggesting only minor losses of hydrophobic materials from the soil surface. The persistence of potential SWR and the degree of SWR of the dry areas remained more or less unchanged. In accordance with this, visualisation of the wetted areas showed that runoff occurred as rivulets guided by surface topography, rather than as sheet flow, with the wetted area increasing from approximately 20% of the slab in the first event to around one third of the total slab area in the final event. This pattern is reflected in the cumulative pattern of the P losses over the ten events. Consequently, we conclude that SWR should be considered as a factor in hydrological modelling and should be included in models to address appropriately the risk of surface water contamination by solutes exogenously applied to water-repellent soils. Soils deliver the regulating ecosystem services of water infiltration and distribution, which can be controlled by macropores. Parameterizing macropore hydraulic properties is challenging due to the lack of direct measurement methods. With tension-disc infiltrometry hydraulic properties near saturation can be measured. Differentiating between hydrologically active and non-active pores, at a given water potential, indirectly assesses macropore continuity. Water flow through macropores is controlled by macropore size distribution, tortuosity, and connectivity, which can be directly derived by X-ray computed tomography (CT). Our objective was to parameterize macropore hydraulic properties based on the imaged macropore network of three horizons of an Andosol and a Gleysol. Hydraulic conductivity K unsat was derived from infiltration measurements. Soil cores from the infiltration areas were scanned with X-ray CT. K unsat was significantly higher in the Andosol than in the Gleysol at all water potentials, and decreased significantly with depth in both soils. The in situ measurements guided the definition of new macroporosity parameters from the X-ray CT reconstructions. For the Andosol, K unsat was best predicted using the imaged-limited macroporosity. A low total macroporosity, coupled with a high macropore density, indicated the abundance of smaller macropores, leading to homogeneous matrix flux. Imaged macropores were not well connected. In contrast, the Gleysol had a bi-modal macropore system with few very large, but well-connected macropores. K unsat was best predicted using the imaged macroporosity consisting only of macropores with diameters between 0.75 and 3 mm. Our research demonstrates that linking traditional soil physical measurements with soil-visualization techniques has a huge potential to improve parameterizing macropore hydraulic properties. The relevance of the relationships found in this study for larger scales and other soil types still needs to be tested, for example by a multi-scale investigation including a much wider range of different soils. In New Zealand a resurgence of irrigation development is underway. Considerable effort is focused on developing efficient irrigation technology to maximise production per unit of applied water while ensuring minimal environmental impacts. Most New Zealand pastoral soils have the potential to develop soil water repellency (WR). Its occurrence limits water infiltration, leads to ponding of water on the soil and may enhance runoff and macropore flow. Some limited New Zealand research has shown that subcritical WR may also be common, where water infiltration is impeded by WR and water absorption into the storage pores of the soil matrix may be restricted even though the soils appear to wet readily. The objective of this research was to assess the potential of arable soils to develop WR and to analyse potential tillage effects. Soil cores were collected from the 0–5 cm depth across the three dominant soil orders used for arable production in the Canterbury region, four common tillage practices and histories, as well as from two long-term tillage trials. At each site, duplicate soil cores were collected to compare the infiltration of water to ethanol using tension disc infiltrometers. Ethanol infiltration is not affected by repellent substances, so it is used as the reference infiltration liquid to assess the degree to which water infiltration is retarded. Our results showed that subcritical WR developed in arable soils irrespective of soil order. We found significantly higher WR with decreasing tillage intensity for the long-term tillage trial. In contrast, no such significant trend was found in the regional survey, which similarly only considered sites with known long-term tillage history. It is possible that any potential effect of tillage intensity on WR was masked by confounding site or soil factors in the survey. Tillage effects on subcritical WR were shown not to persist upon change of tillage system. Our results indicate that soil management affects the degree of subcritical WR and the resultant inhibition of early-time infiltration dynamics. Hydrological significance of the results was shown by comparing actual and potential short-term infiltration, showing that suboptimal infiltration into subcritical water-repellent soils limited soil wetting and water storage efficiency. Further research is recommended that develops soil and irrigation management practices that minimise the occurrence of subcritical WR and assist in capturing more water, resulting in increased irrigation efficiencies and reduced water footprints. Background: The hypothesis was that torasemide, due to more predictable pharmacokinetics/pharmacodynamics, induces greater improvements in functional and social limitation than furosemide and reduces the frequency of hospitalisations in primary care patients with chronic heart failure (CHF).Patients and Methods: Prospective, randomized, unblinded study in primary care, 237 patients with CHF (NYHA II-IV), all on ACE inhibitors. Randomisation: torasemide (n=122) or furosemide (n=115), treated for 9 months.Endpoints: Clinical efficacy, quality of life, safety, tolerability, hospitalisations.Results: Clinical improvement was observed in both groups, but the trend to improve by at least one NYHA class was significant only in torasemide- (P=0.014), but not in furosemide-treated patients. There were no differences with regard to adverse events and hospitalisation due to CHF. Overall, tolerability (P=0.0001) and improvement in daily restrictions (P=0.0002) were significantly higher, number of mictions at 3, 6 and 12 h after diuretic intake (P<0.001 at all time points) and urgency to urinate (P<0.0001) significantly lower in torasemide- vs. furosemide-treated patients.Conclusion: CHF patients treated with torasemide gain a higher benefit in quality of life than furosemide treated patients, due to torasemide's dual effect on both clinical status and social function. Soil water repellency (SWR) of soils is a property with significant consequences for agricultural water management, water infiltration, contaminant transport, and for soil erosion. It is caused by the presence of hydrophobic agents on mineral grain surfaces. Soils were samples in different depths at three forest sites in Japan and three pasture sites in New Zealand, covering soil organic carbon (SOC) contents between 1 and 26%. The SWR was measured over a range of water contents by three common methods; the water drop penetration time (WDPT) test, the molarity of an ethanol droplet (MED) method, and the sessile drop method (SDM). The aim to (i) compare the methods, (ii) characterize the soil-water repellency characteristic curves (SWRCC) being SWR as a function of the volumetric soil-water content ( θ ) or matric potential ( ψ ), and (iii) find relationships between SWRCC parameters and SOC content. The WDPT, MED, and SDM generally agreed well in predicting the θ range where SWR occurred, and there was close agreement between SWR results determined by average MED and SDM at similar θ . Generally, SWR was only found within the top 20 cm of the soil profiles. Six SWR parameters were introduced: (i) the area under the curve ( S WR ( θ ) ); (ii) θ at the maximum SWR ( θ WR-max ), (iii) θ where SWR ceased ( θ non-WR ), (iv) the maximum SWR ( CA i -max ), (v) pF at the maximum SWR ( pF WR-max ) and (vi) pF where SWR ceased ( pF non-WR ). The relationship between the first three parameters and SOC content were best described with Langmuir type equations (r 2 of 0.5–0.7), while the other three parameters changed linearly with SOC contents. Assessing the sustainability of orchards focuses on quantifying environmental impacts and resource consumption. Sustainable orchards also have to be profitable and socially responsible. We aimed to identify sustainable kiwifruit production in the Bay of Plenty, New Zealand, by considering orchards' environmental and economic performance. We conducted a survey of 40 orchards with different cultivars ( Actinidia deliciosa ‘Hayward’ (green) v. Actinidia chinensis ‘Hort16A’ (gold)) and management (integrated v. BioGro certified organic). Assessment of environmental performance was restricted to greenhouse gas emissions (carbon footprint of the orchard phase). We defined eco-efficiency on an area basis as NZD net profit per kg greenhouse gas emissions (1 NZD = 0.83 USD, 31/10/2013). Carbon footprints for the cultivars and management systems were comparable. The choice of functional unit, namely land area and 1 kg of produce, did not affect the result. Our analysis revealed fertilizer use and the N-associated emissions as hot spots for greenhouse gas emissions. Opportunities to reduce greenhouse gas emissions arise in the background system of fertilizer production, packaging, storage and transport, and the optimization of nutrient-use efficiency in the orchard. The integrated system had insignificantly higher greenhouse gas emissions than the organic system. Taking into account the profitability of the orchards, the eco-efficiency of organic orchards was significantly higher than that of integrated orchards. We demonstrated that the metric of eco-efficiency can enhance product differentiation for customers and can also assist orchardists to find the most sustainable management system. Management practices designed to increase carbon sequestration via perennial tree crops are potential tools to mitigate the consequences of climate change. Changes in orchard management could enable growers to meet eco-verification market demands for products with a low carbon footprint and potentially exploit the emerging business opportunity in carbon storage, while enhancing the delivery of ecosystem services that depend on soil carbon stocks. However, there is no standard methodology to verify any potential claims of carbon storage by perennial vine crops. We developed a robust methodology to quantify carbon storage in kiwifruit orchards. Soil carbon stocks (SCS) were determined in six depth increments to 1 m deep in two adjacent kiwifruit blocks, which had been established 10 (“young”) and 25 (“old”) years earlier. We used a space-for-time analysis. Our key results were the young and old kiwifruit block stored about 139 and 145 t C/ha to 1 m depth. Between 80–90 percent of the SCS were stored in the top 0.5 m, and 89–95 percent in the top 0.7 m; there was no significant difference between the SCS in row and alley to a depth of 0.5 m; a CV of 5–15 percent indicates that 4–10 cores are needed for 80 percent confidence in the estimated SCS; we recommend separating each core into the depths 0–0.1, 0.1–0.3, 0.3–0.5, and 0.5–1 m to allow the assessment of SCS dynamics; we detected a weak spatial pattern of the SCS only for the old kiwifruit block with a range of about 3 m. A sampling bay along a vine row should have a maximum length of 3 m. We then assessed SCS in more than sixty kiwifruit orchards throughout New Zealand. They stored on average 174.9 ± 3 t C ha−1to 1 m depth. On average, 51 percent of the SCS down to 1 m depth were stored in the top 0.3 m, which is the standard depth according to the Kyoto protocol. About 72 percent of the SCS to 1 m depth were captured when increasing the sampling depth to 0.5 m. These results underscore the necessity to analyze SCS in an orchard to at least 0.5 m deep. Using the same methodology to 1 m deep, we determined SCS in two wine grape vineyards on shallow, stony alluvial soils. We found a difference between vineyard and adjacent pasture SCS of nearly 16 t/ha. As the vines are 25 years old, this equates to carbon sequestration rates of 640 kg ha−1 yr−1. Our results of the space-for-time analysis also showed that all sequestration had occurred below 0.5 m. Therefore, we decided to sample C to a greater depth. In a 30-year old kiwifruit orchard and an adjacent pasture, SCS was measured to 9 m deep. The objective of this study was to evaluate the effects of diet composition on phytate (InsP6) degradation in dairy cows. In Experiment 1, four diets that differed in the amount and source of phosphorus (P) were fed to 24 lactating cows in a 4 × 4 Latin Square design. The control diet (Diet C) contained 4.18 g P/kg dry matter (DM). Diet MP contained additional mineral P (5.11 g P/kg DM), Diet RS contained rapeseed and rapeseed meal as organic P sources (5.26 g P/kg DM) and Diet RSM contained rapeseed meal and rapeseed oil (5.04 g P/kg DM). Total P (tP) and InsP6excretion in faeces were measured. In Experiment 2, we used a rumen simulation technique (Rusitec) to estimate ruminal disappearance of tP and InsP6from Diets C, MP and RSM. In Experiment 1, tP concentration in faeces increased with tP intake and was highest for Diets RS and RSM. The source of supplemented P had no influence on tP digestibility, but tP digestibility was reduced for Diets MP, RS and RSM in comparison to that for Diet C. InsP6disappearance decreased in Diet MP (85.0%) and increased in Diets RS (92.7%) and RSM (94.0%) compared to that in Diet C (90.0%). In Experiment 2, P source influenced ruminal tP disappearance (Diet MP, 78.6%; Diet RSM, 75.3%). InsP6disappearance for Diet C (98.1%) was higher than that for Diets MP (95.6%) and RSM (94.9%). The results confirmed the high potential of ruminants to degrade InsP6, but differences in diet composition influenced InsP6disappearance. Objective : Monocyte infiltration is a critical step in the pathophysiology of plaque instability in coronary artery disease (CAD). Macrophage migration inhibitory factor (MIF) is involved in atherosclerotic plaque progression and instability leading to intracoronary thrombosis. Gremlin-1 (Grem1) has been recently identified as endogenous inhibitor of MIF. To date there are no data on the clinical impact of this interaction in cardiovascular patients. Methods and results : Plasma levels of MIF and Grem1 were determined by enzyme-linked immunoassay in patients with acute coronary syndromes (ACS, n = 120; stable CAD, n = 166 and healthy control subjects, n = 25). MIF levels were significantly increased in ACS compared to stable CAD and healthy control (ACS: median 2.85; IQR 3.52 ng/ml; versus SAP: median 1.22; IQR 2.99 ng/ml; versus healthy control: median 0.10; IQR 0.09 ng/ml, p < 0.001). Grem1 levels were significantly higher in ACS and stable CAD patients compared to healthy control (ACS: median 211.00; IQR 130.47 ng/ml; SAP: median 220.20; IQR 120.93 ng/ml, versus healthy control: median 90.57; IQR 97.68 ng/ml, p < 0.001). Grem1/MIF ratio was independently associated with ACS, whereas the single parameters were not associated with the presence of ACS. Furthermore, Grem1/MIF ratio was associated with angiographic signs of intracoronary thrombi and severity of thrombus burden. Conclusion : These novel findings suggest a potential role of Grem1/MIF ratio to indicate acuity of CAD and the grade of plaque stability. Prospective angiographic cohort studies involving plaque imaging techniques are warranted to further characterize the prognostic role of this novel risk marker in CAD patients. Global agricultural ecosystems, the largest biospheric sources of atmospheric carbon dioxide (CO 2 ), may turn into considerable net carbon (C) sinks through adopting management strategies advised by research. As C sequestration is usually coupled with the silicon (Si) cycle, strategic manipulation of the biogeochemical Si cycle in agricultural ecosystems offers a not yet fully explored possibility to enhance C sequestration. This review summarizes current knowledge of C sequestration coupled with the Si cycle and its management in agricultural ecosystems. Carbon sequestration is coupled with the Si cycle through many processes including dynamics of phytoliths and aggregates, and silicate weathering at different temporal and spatial scales. Cultivation of deep rooting crops, erosion mitigation with buffer strips, fertilization of Si-rich materials are some of the potential management strategies to increase both crop production and C sequestration coupled with the Si cycle. Further questions such as identifying the controlling factors of bioavailable Si pools and C sequestration, and quantifying the cost-efficiency of different management strategies to manipulate the Si cycle with the aim to enhance C sequestration should be investigated. Disease resistant crop varieties are important for both disease control and for reducing chemical pesticide use. However, there is often a trade-off between disease resistance and yield. While the European Union has banned the use of some pesticides in agriculture, large amounts are still applied to various crops, including wheat. Market-based instruments could motivate farmers to adopt more environmentally friendly production methods. In this study we aimed to explore farmers' adoption of resistant varieties in different wheat price scenarios and their willingness to forgo yield when cultivating more resistant varieties. We conducted face-to-face interviews with 192 farmers using a choice experiment. Results showed that farmers preferred traits of yield stability (fungal disease resistance, lodging resistance and drought tolerance) over yield. Preferences for traits varied with the production system; livestock production was negatively associated with the importance of yield; cultivated land area was positively associated with protein content. The market price scenario did not change preferences. We calculate that farmers are willing to forgo more yield by cultivating highly disease resistant varieties than they are likely to recuperate from reducing costs of pesticides. Overall, we conclude that farmers' choices are based more on a combination of the production system, drought experience, climate change belief and the potential impact of further regulations to reduce the use of chemical pesticides than on the expected wheat price. We developed a laboratory-scale run-off measurement apparatus (ROMA) to quantify for the first time directly the impact of soil water repellency (SWR) on run-off from undisturbed soil slabs in the laboratory. We tested and evaluated the performance of ROMA with multiple consecutive run-off experiments using water followed by a fully-wetting liquid, namely an ethanol solution. We found that a 30% (v/v) ethanol solution was needed to ensure that the soil hydrophobicity had no influence on the infiltration rate of the liquid. The results demonstrated the ROMA is a robust and reproducible tool that performs at a high standard with instrument errors below 2%. We conducted ROMA run-off experiments with air-dried soil slabs (480mm long×190mm wide×50mm deep) collected from four pastoral sites, representing three major soil orders in the North Island, New Zealand. They were the Kashmir-Recent Soil (Fluvisol), Hawke's Bay-Recent Soil (Fluvisol), Taranaki-Gley Soil (Gleysol), and Taranaki-Organic Soil (Histosol). These soils had a high degree and persistence of SWR. The contact angles were 97, 97, 98 and 104°, the potential water drop penetration times (WDPT) were 4, 42, 54 and 231min, and the run-off fractions were 16, 19, 28, and 96% respectively. However, even the extremely hydrophobic Taranaki-Organic Soil, which had a runoff coefficient of 96%, only lost 13% of the applied bromide via run-off. This demonstrates that run-off occurred in rivulets covering only a small fraction of the surface. Multivariate regression analyses showed that the soil organic carbon content and the degree of SWR explained 89% of the variability of the run-off coefficients. We identified difficulties around the meaningfulness of the persistence of SWR, as determined by the WDPT test, since it just measures SWR at a single point. Alternatively, our ROMA experiments integrate the spatial variability of SWR of an undisturbed soil slab. In addition, the method is faster for extremely hydrophobic soils once the ROMA is set up. Renal transplant recipients (RTR) are considered at high risk for influenza-associated complications due to immunosuppression. The efficacy of standard influenza vaccination in RTRs is unclear. Hence, we evaluated activation of the adaptive immunity by the pandemic influenza A(H1N1) 2009 (A(H1N1)pdm09) vaccine in RTRs as compared to healthy controls. To determine cross-reactivity and/or bystander activation, seasonal trivalent influenza vaccine and tetanus/diphteria toxoid (TT/DT) vaccine-specific T cells along with allospecific T cells were quantified before and after A(H1N1)pdm09 vaccination. Vaccination-induced alloimmunity was additionally determined by quantifying serum creatinine and proinflammatory protein IP-10. Contrary to healthy controls, RTRs required a booster vaccination to achieve seroconversion (13.3 % day 21; 90 % day 90). In contrast to humoral immunity, sufficient A(H1N1)pdm09-specific T-cell responses were mounted in RTRs already after the first immunization with a magnitude comparable with healthy controls. Interestingly, vaccination simultaneously boosted T cells reacting to seasonal flu but not to TT/DT, suggesting cross-activation. No alloimmune effects were recorded. In conclusion, protective antibody responses required booster vaccination. Abstract: Agglomeration dramatically affects many aspects of nanoparticle–cell interactions. Here we show that hydroxyapatite nanoparticles formed large agglomerates in biological medium resulting in extensive particle uptake and dose-dependent cytotoxicity in human macrophages. Particle citration and/or the addition of the dispersant Darvan 7 dramatically reduced mean agglomerate sizes, the amount of particle uptake and concomitantly cytotoxicity. More surprisingly, agglomeration governed the mode of particle uptake. Agglomerates were sequestered within an extensive, interconnected membrane labyrinth open to the extracellular space. In spite of not being truly intracellular, imaging studies suggest particle degradation occurred within this surface-connected compartment (SCC). Agglomerate dispersion prevented the SCC from forming, but did not completely inhibit nanoparticle uptake by other mechanisms. The results of this study could be relevant to understanding particle–cell interactions during developmental mineral deposition, in ectopic calcification in disease, and during application of hydroxyapatite nanoparticle vectors in biomedicine. The reform of the European Common Fisheries Policy ( CFP) in 2013 decides whether the European fisheries can be put on a sustainable course in the next 10 years. The CFP reform must halt overexploitation of fish resources and stop harm to the marine environment by fishing activities. The European Commission published a reform proposal just recently. This proposal is already an important first step in the right direction, but is under threat of being watered down by the Fisheries Council. Necessary improvements of the current CFP include among others a binding definition of a sustainable stock management, a discard ban for all fish species, transferable fishing concessions and a cessation of all aid for building new vessels. These modifications pave the way for placing micromanagement in the hands of regions and fisheries so as to foster self-responsibility among fishers and give greater consideration to regional circumstances. Abstract: This paper analyzes the effects of social influence and participation in collective action initiatives on soil conservation effort among smallholder farmers in Lake Naivasha basin, Kenya. We apply binary and ordered probit models in a two stage regression procedure to cross-sectional data collected through a household survey among randomly selected smallholder farmers. Smallholder farming systems in the research area are associated with practices that render farmlands susceptible to soil erosion causing negative impacts on land and the environment. Therefore, strategies that encourage soil conservation are likely to also offer solutions for dealing with agri-environmental challenges and poverty alleviation. Results indicate that social capital facilitates participation in collective action initiatives which then influence individual soil conservation efforts. Neighborhood social influences, subjective norms, gender, education level, farm size, access to credit and livestock ownership also emerge as key determinants of soil conservation effort. Policy implications drawn by this study encourage strategies to increase participation and effectiveness in collective action initiatives as a boost to soil conservation. Implementation of soil conservation practices could also be encouraged through awareness increasing instruments, facilitating access to agricultural micro-credit and paying attention to gender related challenges on knowledge access and rights over land and other natural resources. Abstract: Biodiversity is an essential resource, which we classify as conditionally-renewable. In order to achieve conservation and sustainable use of biodiversity virtually all nation states signed the United Nations Convention on Biological Diversity. In this paper we investigate how the heterogeneity of countries in regard to ecosystems and wealth influences the stability of international biodiversity conservation agreements both without and with transfers. We further examine the effect of different degrees of ecosystem substitutability. We model a coalition formation game with players that have a continuous conservation choice. The conservation benefit is dependent on wealth and ecosystem quality. Aggregation of global benefits respects differences in ecosystem substitutability. In case of transfers, a fund redistributes coalition benefits according to a sharing rule. The main finding is that in the absence of transfers, compared to the homogeneous situation, heterogeneity in ecosystems and wealth reduces the size of a stable coalition. The destabilising effect is stronger the higher the ecosystem substitutability. Optimal transfers facilitate a large stable coalition. [Copyright &y& Elsevier]

Aims Cyclophilin A (CyPA) represents a ubiquitous intracellular protein, which is secreted by inflammatory and by dying/necrotic cells. The aim of this study was to evaluate the prognostic relevance of CyPA expression in endomyocardial biopsies of consecutive patients with congestive heart failure. Methods and results A total of 227 unselected patients (age 53.9 ± 15 years) with congestive heart failure undergoing endomyocardial biopsy for diagnostic reasons were enrolled. Biopsies were analysed using established histopathological and immunohistological criteria together with CyPA staining. Virus genome was studied by polymerase chain reaction. CyPA was significantly enhanced in patients with inflammatory cardiomyopathy (n = 127) as compared with patients with non-inflammatory cardiomyopathy (n = 100, P < 0.0001). During a mean follow-up of 16.3 months, 60 patients (26.4%) reached the primary endpoint, a composite of all-cause death, heart transplantation, malignant arrhythmia, and heart failure-related rehospitalization. Of all clinical (ejection fraction, New York Heart Association functional class), laboratory (brain natriuretic peptide), and immunohistological parameters (CyPA, extracellular matrix metalloproteinase inducer, CD68, CD3, major hisocompatibility complex II, and virus genome) tested, only CyPA was identified as an independent predictor for the composite endpoint [hazard ratio (HR) 2.4; 95% confidence interval (CI) 1.2–5.2; P = 0.019] as well as for all-cause death and heart transplantation alone (HR 4.7; 95% CI 1.1–19.8; P = 0.036). Subgroup analysis revealed CyPA as a predictor in patients with non-inflammatory cardiomyopathy for the composite endpoint (HR 3.0; 95% CI 1.3–6.6; P = 0.007) as well as all-cause death or heart transplantation alone (HR 6.4; 95% CI 1.4–28.1; P = 0.014). Background: Owing to its variable course from asymptomatic cases to sudden death risk stratification is of paramount importance in newly diagnosed non-ischemic cardiomyopathy. We tested whether late gadolinium enhancement (LGE) assessed by cardiac magnetic resonance (CMR) imaging is a prognostic marker in consecutive patients with newly diagnosed non-ischemic cardiomyopathy. Methods: We enrolled 185 patients who presented for evaluation of newly diagnosed non-ischemic cardiomyopathy. Coronary artery disease was excluded by coronary angiography. Following risk markers were additionally assessed: NYHA functional class (≥II), brain natriuretic peptide (>100 ng/l), troponin I (TnI, ≥0.03 µg/l), left ventricular ejection fraction (LVEF, ≤40%), left ventricular enddiastolic diameter (>55 mm) and QRS duration (>98 ms). Endpoint of the study was the composite of all-cause mortality, heart transplantation, aborted sudden death, sustained ventricular tachycardia or hospitalization due to decompensated heart failure within three years of follow-up. Results: During median follow-up of 21 months, 54 patients (29.2%) reached the composite endpoint. Ninety-four of the 185 patients (50.8%) were judged LGE-positive. Prognosis of LGE-positive patients was significantly worse than that of LGE-negative patients (cumulative 3-year event rates of 67.4% in LGE-positive and 27.2% in LGE-negative patients, respectively; p = 0.021). However, in multivariable analysis, presence of LGE was not an independent predictor of outcome. Only LVEF ≤40% and TnI ≥0.03 µg/l were independent risk predictors of the composite endpoint yielding relative risks of 3.9 (95% CI 1.9–8.1; p<0.0001) and 2.2 (95% CI 1.2–4.0; p = 0.014), respectively. Conclusions: In consecutive patients presenting with newly diagnosed non-ischemic cardiomyopathy, LGE-positive patients had worse prognosis. Abstract: Testosterone influences sexual differentiation in early development, and activates sexual maturation and sex-related behavior in males during puberty. Testosterone can also influence the expression of male alternative reproductive tactics, by either organizational effects (fixed tactics) or by activational effects (plastic tactics). However, the roles of testosterone in sexual maturation and at the same time the expression of alternative reproductive tactics have been little investigated experimentally, and studies of free-ranging mammals are lacking. We conducted a field experiment in free-ranging juvenile African striped mice (Rhabdomys pumilio), a species with alternative reproductive tactics. Juvenile male striped mice reaching puberty can remain in their family as philopatric group-living males with low testosterone levels, or they can disperse and become solitary living roamers with much higher testosterone levels. We tested whether experimentally increased testosterone levels in non-scrotal juvenile males induces puberty and leads to the expression of the roaming tactic. Testosterone-treated males received the hormone for 15days by silastic implants which were empty in control-treated males. When compared to control-treated males, testosterone-treated males had higher testosterone levels, lower corticosterone levels, and became scrotal with descended testes. Testosterone-treated males also had larger testes, larger epididymides, and showed indication of spermatogenesis. Testosterone-treated males did not become solitary-living roamers, but had larger home ranges than control males. We conclude that testosterone can induce sexual maturation and causes juvenile males to increase their home ranges, maybe to search for dispersal opportunities. [Copyright &y& Elsevier]

This paper shows the strong relation between the factors that lead to the resource curse ( RC) and factors that lead to a decline of genuine savings ( GS). There is substantial empirical evidence that economies that rely predominantly on their natural resources are also characterized by slower economic growth. This so-called RC is commonly traced back to the fact that natural resources' generate rents that are independent of a country's economic performance, which can lead to suboptimal reinvestments of this consumed natural capital. We argue that the factors responsible for the RC also have a negative effect on GS, a concept that measures 'weak' sustainable development by considering reinvestment of natural capital rents in physical and human capital. We discuss whether the RC hampers possibilities for resource abundant countries to obtain sufficiently high rates of GS, and find indeed many reasons why resource-dependent countries have problems achieving positive GS rates. We survey both areas of research, emphasizing the influence of the exogenous and endogenous determinants of economic growth, which are usually used to theoretically and empirically explain the RC on the three different forms of capital considered by GS. We specify why most countries suffering from the RC have negative GS rates and explain in detail where the linkages are. Background: Chronic inflammation promotes atherosclerosis in cardiovascular disease and is a major prognostic factor for patients undergoing percutaneous coronary intervention (PCI). Macrophage migration inhibitory factor (MIF) is involved in the progress of atherosclerosis and plaque destabilization and plays a pivotal role in the development of acute coronary syndromes (ACS). Little is known to date about the clinical impact of MIF in patients with symptomatic coronary artery disease (CAD). Methods and Results: In a pilot study, 286 patients with symptomatic CAD (n = 119 ACS, n = 167 stable CAD) undergoing PCI were consecutively evaluated. 25 healthy volunteers served as control. Expression of MIF was consecutively measured in patients at the time of PCI. Baseline levels of interleukin 6 (IL-6), "regulated upon activation, normal T-cell expressed, and secreted" (RANTES) and monocyte chemoattractant protein-1 (MCP-1) were measured by Bio-Plex Cytokine assay. C-reactive protein (CRP) was determined by Immunoassay. Patients with ACS showed higher plasma levels of MIF compared to patients with stable CAD and control subjects (median 2.85 ng/mL, interquartile range (IQR) 3.52 versus median 1.22 ng/mL, IQR 2.99, versus median 0.1, IQR 0.09, p<0.001). Increased MIF levels were associated with CRP and IL-6 levels and correlated with troponin I (TnI) release (spearman rank coefficient: 0.31, p<0.001). Patients with ACS due to plaque rupture showed significantly higher plasma levels of MIF than patients with flow limiting stenotic lesions (p = 0.002). Conclusion: To our knowledge this is the first study, demonstrating enhanced expression of MIF in ACS. It is associated with established inflammatory markers, correlates with the extent of cardiac necrosis marker release after PCI and is significantly increased in ACS patients with "culprit" lesions. Abstract: Calcium phosphate and hydroxyapatite nanoparticles are extensively researched for medical applications, including bone implant materials, DNA and SiRNA delivery vectors and slow release vaccines. Elucidating the mechanisms by which cells internalize nanoparticles is fundamental for their long-term exploitation. In this study, we demonstrate that hydrophilic hydroxyapatite nanoparticles are sequestered within a specialized compartment called SCC (surface-connected compartment). This membrane-bound compartment is an elaborate labyrinth-like structure directly connected to the extracellular space. This continuity is demonstrated by in vivo 2-photon microscopy of ionic calcium using both cell-permeable and cell-impermeable dyes and by 3-D reconstructions from serial block-face SEM of fixed cells. Previously, this compartment was thought to be initiated specifically by exposure of macrophages to hydrophobic nanoparticles. However, we show that the SCC can be triggered by a much wider range of nanoparticles. Furthermore, we demonstrate its formation in A549 human lung epithelial cells, which are considerably less phagocytic than macrophages. EDX shows that extensive amounts of hydroxyapatite nanoparticles can be sequestered in this manner. We propose that SCC formation may be a means to remove large amounts of foreign material from the extracellular space, followed by slow degradation, may be to avoid excessive damage to surrounding cells or tissues. Abstract: Many regions of the world are predicted to experience water scarcity due to more frequent and more severe droughts and increased water demands. Water use efficiency by plants can be negatively affected by soil water repellency (SWR). It is timely to review existing techniques to remedy SWR. Ideally remediation addresses the origins of a problem. However, the fundamental mechanisms of how and why SWR develops are still poorly understood. In this review it was hypothesized that SWR occurs where the balance of input–decomposition of organic matter is impaired, due to either increased input or decreased decomposition rates of hydrophobic substances. Direct and indirect strategies to remedy SWR were distinguished. While direct remediation aims at abolishing the causes of SWR, indirect strategies seek to manage sites with SWR by treating its symptoms. The 12 reviewed strategies include applying surfactants, clay, slow-release fertilizers, lime, and fungicides, bioremediation of SWR through stimulating earthworms, choosing adapted vegetation, irrigation, cultivation, soil aeration and compaction. Some of the techniques have been applied successfully only in laboratory experiments. Our review highlights that it is not straightforward to cure SWR based on easily measurable and site-specific soil and vegetation properties, and that long-term, large-scale field experiments are required to improve the understanding of the evolution of SWR as cornerstone to develop cost-effective and efficient remediation strategies. Abstract: Nanomedicine is one of the most promising areas of exploitation for multi-walled carbon nanotubes (MWNTs). These ‘needle-like’ nanovehicles are capable of carrying drug molecules via exo- and endohedral functionalisation and are steerable by an external magnetic field due to the presence of ferromagnetic nanoparticles in the nanotube core (up to 7.3wt.%). These properties make them promising candidates for drug targeting or MRI contrast agents. Particularly, oxidised and nitrogen-doped MWNTs exhibiting enhanced chemical reactivity compared to their unmodified precursors/analogues could be exploited in this field. Here, we assessed the toxicity and intracellular localisation of two different, chemically modified and unmodified nanotubes towards human macrophage cells using a range of toxicity and imaging techniques. Oxidised and N-doped MWNTs were not significantly toxic to HMMs in contrast to unmodified MWNTs. All types of MWNTs entered the cell via active phagocytosis/endocytosis, but also passively by ‘self-injection’ through the plasma membrane, and were ultimately found in the cytoplasm and possibly also the nucleus. Abstract: Background: There is cumulative evidence that the degree of inflammation correlates with prognosis after percutaneous coronary interventions (PCI). Additionally, there is a cross-link between platelet activation and inflammatory pathways. The aim of the present analysis was to evaluate the association of inflammatory markers and effects of dual antiplatelet therapy on platelet function and outcome in patients undergoing PCI. Methods and results: In a pilot study, 157 patients with symptomatic coronary artery disease (CAD) undergoing PCI were consecutively evaluated. Platelet response to clopidogrel and acetylsalicylic acid was assessed using whole blood multiple electrode aggregometry (MEA). Baseline levels of IL-6, RANTES and MCP-1 were measured by Bio-Plex Cytokine assay. C-reactive protein (CRP) was determined by Immunoassay. Levels of IL-6, RANTES, and CRP correlated well with ADP and arachidonic acid (AA)-induced MEA. In a second step, a retrospective analysis of a cohort of 903 PCI-patients was performed to evaluate the association of on-treatment residual platelet aggregation (RPA) and baseline CRP levels on the incidence of myocardial infarction (MI), death and stent thrombosis (ST). Patients suffering a subsequent event had a significantly higher level of baseline CRP and higher RPA compared to patients without events. After multivariate adjustment high baseline CRP and high RPA were independent predictors for combined major events and ST after PCI. Conclusion: To our knowledge this is the first study linking inflammation, antiplatelet drug responsiveness and outcome in a large CAD-patient cohort. Abstract: We hypothesized that elevated atmospheric carbon dioxide concentration [CO2] as a feature of climate change promotes the occurrence of soil water repellency (SWR) which reduces soil carbon mineralization and thus increases carbon sequestration. Soil surface transects under elevated (475μLL−1) and ambient atmospheric [CO2] in a long-term Free Air Carbon Dioxide Enrichment experiment were sampled at a high spatial resolution. All samples were hydrophobic at the time of the sampling. At the micro-scale, the differences in degree and persistence of SWR, soil organic matter (SOM) content, microbial respiration rates (MRR) and water content between the treatments were not significant. SWR was not correlated with SOM or MRR. A strong correlation between water contents and SWR parameters demonstrated the importance of SWR for water redistribution. At the meso-scale of disc infiltrometry, infiltration rates were reduced by SWR, and were higher under ambient than under elevated [CO2]. This corroborates the tendency of reduced SWR under elevated [CO2] observed at the micro-scale. SWR showed a spatial structure, exhibiting short ranges. SOM and MRR showed no spatial pattern at the scale analyzed, emphasizing that SWR did not contribute to an increase of the long-term terrestrial C sink in response to increased atmospheric [CO2]. Abstract: Multicellular spheroids have important applications in tumour studies, drug screening and tissue engineering. To enable simple manipulation of spheroids, magnetically labelled HeLa cells were cultured in hanging drops to generate magnetic spheroids. HeLa cells were labelled by biotinylating their cell membrane proteins and then binding streptavidin paramagnetic particles onto the biotinylated cell surface. Spheroids of different sizes were obtained by varying the seeding cell concentrations within the hanging drops and the spheroids had good cell viability. Characterisation of the F-actin distribution within the spheroids indicated a three dimensional reorganisation of the cellular cytoskeleton compared to monolayer cultures. The magnetic moment of the spheroids was measured and showed a superparamagnetic response in an applied field. Transmission electron microscopy analysis indicated that the paramagnetic particles were still present in the spheroids even after 21 days of culture. These spheroids could be easily and quickly separated magnetically without the need for centrifugation. The magnetic spheroids were also successfully manipulated and patterned using magnetic fields within a few seconds. The patterned spheroids then fused together to form a larger tissue construct. Abstract: Multi-walled carbon nanotubes (MWNTs) have been proposed for use in many applications and concerns about their potential effect on human health have led to the interest in understanding the interactions between MWNTs and human cells. One important technique is the visualisation of the intracellular distribution of MWNTs. We exposed human macrophage cells to unpurified MWNTs and found that a decrease in cell viability was correlated with uptake of MWNTs due to mainly necrosis. Cells treated with purified MWNTs and the main contaminant Fe2O3 itself yielded toxicity only from the nanotubes and not from the Fe2O3. We used 3-D dark-field scanning transmission electron microscopy (DF-STEM) tomography of freeze-dried whole cells as well as confocal and scanning electron microscopy (SEM) to image the cellular uptake and distribution of unpurified MWNTs. We observed that unpurified MWNTs entered the cell both actively and passively frequently inserting through the plasma membrane into the cytoplasm and the nucleus. These suggest that MWNTs may cause incomplete phagocytosis or mechanically pierce through the plasma membrane and result in oxidative stress and cell death. [Copyright &y& Elsevier]

Attracting and retaining competent teachers is a key concern when it comes to managing the supply and demand for teachers. This article examines the motivation that prompts people to enter or leave the teaching profession with the aim of identifying a decision framework for defining teacher policies. The results are based on the teacher workforce planning system put in place by the Education Department of the Swiss Canton of Geneva. Having identified the most influential factors affecting teacher motivation, we rated these with regard to their responsiveness to policy measures in a bid to identify levers for deriving effective teacher policies. This decision framework highlights three main issues to serve as a focus for policy measures-the characteristics of the job activities, working conditions and professional image. Bortezomib is the first approved member of a new class of anti-myeloma agents, the proteasome inhibitors. Further proteasome inhibitors are needed to optimise this promising treatment option. S-2209 [1-[1-{1-[(2,4-Dioxo-imidazolidin-1-ylimino)-methyl]-2-phenyl-ethylcarbamoyl}-2-(1H-indol-3-yl)-ethylcarbamoyl]-2-(1H-indol)] inhibits the chymotryptic activity of the human 20S proteasome (half maximal effective concentration, IC50∼220 nmol/l) which was determined by a proteasome inhibition assay. A nuclear factor κB inhibition assay revealed a half maximal effective concentration (EC50) of 0·9 μmol/l. The WST-1 growth assay showed inhibition of cell growth of all tested multiple myeloma (MM) cell lines with an IC50 between 100 nmol/l and 600 nmol/l. Strong induction of apoptosis was seen in MM cells at nanomolar concentrations (IC50∼300 nm) as well as in primary myeloma cells. No induction of apoptosis was detected in peripheral blood mononuclear cells from healthy humans. Upregulation of p53, activation of JNK protein, and downregulation of Mcl-1 was revealed. Despite the administration of 15 mg S-2209/kg/d in wistar rats, no toxicity with respect to body weight, hepatic enzymes, creatinine or haemoglobin was seen. Proteasome inhibition in white blood cells isolated from the treated rats was higher in the S-2209 treated animals in comparison with the control animals treated with 0·1 mg/kg/d bortezomib. Abstract: Ferumoxtran-10 is an ultrasmall superparamagnetic iron oxide nanoparticle potentially useful as a contrast material in magnetic resonance imaging for the diagnosis of inflammatory and degenerative disorders associated with high macrophage activity. In clinical trials, it is currently applied to monitor the effect of atorvastatin therapy on macrophage activity in human carotid plaques. A recent study reported the inhibition of iron oxide nanoparticle uptake in macrophages by lovastatin, an effect which could compromise the suitability of Ferumoxtran-10 as an MRI contrast material in patients on statin therapy. Therefore, we examined the effect of atorvastatin on human monocyte–macrophage uptake of Ferumoxtran-10 in vitro using biochemical assays, magnetic resonance imaging and transmission electron microscopy. Our study showed that non-toxic concentrations of atorvastatin did not affect the amount of Ferumoxtran-10 taken up by HMMs. Furthermore, the intracellular distribution of iron oxide nanoparticles and the resulting MRI signal intensities remained unchanged by statin treatment. These results were obtained using atorvastatin concentrations probably vastly exceeding those reached in patient plasma in vivo. Atorvastatin therapy itself is therefore unlikely to affect Ferumoxtran-10 based macrophage detection by MRI, a prerequisite for the use of this contrast material to monitor lesion macrophage burden during lipid-lowering therapy. This article discusses conservation payments to reindeer herders in Sweden for losses by carnivores. The payments are made for surviving offspring of wolverines, lynx, and wolves. According to the authors, the system works because payments are made based on conservation outcomes not numbers of livestock lost. They argue that this avoids moral issues and payments are made before there is damage so there is no delay. They state that payments are based on expected losses during the lifetime of the carnivore and the money is divided among the villagers in the area where the carnivore offspring are found.

Abstract: Objective: The purpose of this study was to investigate the effects of low power red laser associated with acidulated phosphate fluoride on the development of induced-dental caries in rats. Design: Dental caries were induced in molars of 40 rats divided into five groups: control group (CG), the teeth were not submitted to any treatment; laser group (LG), teeth were irradiated with a low power red laser (LPRL), power of 30mW and dose of 5J/cm2; fluoride group (FG), teeth were treated with topical acidulated phosphate fluoride (APF) 1.23% applied for 4min; laser+fluoride group (LFG), teeth were irradiated with LPRL followed by APF; fluoride+laser group (FLG), teeth were treated with APF followed by LPRL. The animals were killed after 48 days, and the first and second molars were extracted to analyze the caries lesion area, microhardness, and calcium and phosphorus ratio. Results: There were no statistical differences among FG, LFG, and FLG regarding to caries area and microhardness, although the caries area were smaller in LFG. Ca/P ratio did not show significant differences among all groups. Conclusions: Although LPRL before APF application appeared to diminish the caries progression, LPRL did not present any additional benefit compared with acidulated phosphate fluoride on the prevention of induced-dental caries in rats. Abstract: Ferumoxtran-10, a dextran-coated ultrasmall superparamagnetic iron oxide particle, has the potential to reveal macrophages in vivo using magnetic resonance imaging potentially acting as a marker of inflammatory status. Pending clinical trials, we examined the interactions of Ferumoxtran-10 with human monocyte-macrophages (HMMs) in vitro to assess its safety and lack of pro-inflammatory activity. After 72h, Ferumoxtran-10 was not toxic at 1mg/ml and may be only mildly toxic at 10mg/ml. Viability in cells with a high intracellular Ferumoxtran-10 load was not affected over 14 days. Ferumoxtran-10 did not interfere with baseline or stimulated cytokine (interleukin-12, interleukin-6, tumour necrosis factor-α or interleukin-1β) or superoxide anion production or with Fc-receptor-mediated phagocytosis. Similarly, Ferumoxtran-10 did not induce cytokine production and was not chemotactic. High-resolution electron microscopy and selected-area electron diffraction confirmed the core of Ferumoxtran-10 is composed of crystalline magnetite. Bright field transmission electron microscopy of thin sections demonstrated that Ferumoxtran-10 was retained in lysosomes of HMM for several days. Ferumoxtran-10 is not toxic to HMMs in vitro, does not activate them to produce pro-inflammatory cytokines or superoxide anions, is not chemotactic and does not interfere with Fc-receptor-mediated phagocytosis. Furthermore, extremely high intracellular Ferumoxtran-10 concentrations had only slight or no effects on these key activities. [Copyright &y& Elsevier]

Allophanic top‐ and subsoils from the Mexican and Newzealand Central Volcanic Plateau, as well as a nonallophanic sandy loam soil, were sampled to study the impact of organic matter and allophane content on 2,4‐D fate. High sorption rates were found, especially in the two topsoils from Mexico and New Zealand, with distribution coefficient (Kd) obtained from displacement experiments in packed columns equal to 7.61 and 8.43 L kg-1 respectively. 2,4‐Dichlorophenoxyacetic acid transfer through the soil columns was found to be in chemical nonequilibrium and was well predicted using a two‐site sorption model. For the two allophanic top soils, Kd obtained from batch was very different to the Kd obtained from column experiments. Either the equilibrium could not be reached in batch or the two‐site model was not able to describe the wide range of sorption sites present in the highly reactive organic matter and allophane components. Seminal plasma of mammalians contains, among others, proteins that are characterized by the fibronectin (Fn) type II module. Our knowledge about the structure and the physiological function of seminal Fn type II proteins mainly originates from studies on PDC-109, the bovine representative of this protein family. The present work focuses on the equine protein SP-1/2 (also named HSP-1/2) with particular emphasis on its interaction with lipid membranes by employing the intrinsic protein fluorescence and a number of spin-labeled and fluorescent lipid analogues. The results indicate that the interaction of SP-112 with (lipid) membranes is similar to that of PDC- 109 which can be explained by homologous amino acid sequences of both proteins. Like PDC-109, SP-1/2 has a specificity for phospholipids with the phosphocholine headgroup. Upon binding to lipid vesicles, the protein intercalates into the hydrophobic membrane core, resulting in a rigidification of the lipid phase and, at higher concentration, in a perturbation of membrane structure. However, compared with PDC-109, the impact of SP-1/2 on membranes is less intense in that the degree of protein-mediated immobilization of lipids was lower. Furthermore, different to PDC-109, SP-1/2 was not able to extract lipids from human red blood cells. In this study we demonstrated the combination of MALDI-TOF MS and TLC as a fast and powerful tool to investigate the phospholipid (PL) composition of organic extracts of bull spermatozoa. Since phosphatidylcholine (PC) is the dominant PL species, an adequate resolution of MALDI-TOF spectra for sphingomyelin (SM) or phosphatidylethanolamine (PE) was achieved only after previous PL separation by TLC. We found a poor diversity especially for PE and PC, mainly containing ether-linked fatty acids which were 1-palmityl-2-docosahexaenoyl-PL and the corresponding alkenyl-acyl compound (plasmalogen) 1-palmitenyl-2-docosahexaenoyl-PL. For PC, both lipids were quantified after phospholipase A2 digestion to represent 44.2 and 37.2%, respectively, of the total PC. In contrast, the diacyl-PC content of bull spermatozoa was comparatively low (18.6% of total PC).In the presence of trifluoroacetic acid (TFA), which is routinely added to the MALDI-TOF matrix to improve the signal to noise ratio, a high lysophospholipid (LPL) content was detected in the PL extracts of bull spermatozoa, whereas TLC did not reveal significant amounts of LPL. The TFA mediated hydrolysis of the acid-labile alkenyl-acyl PL to the corresponding LPL was shown to cause this discrepancy. This assumption was verified by analysing the PL composition by MALDI-TOF MS before and after (i) digestion of sperm cell lipids with phospholipase A2 and (ii) exposition of spermatozoa to HCl fumes. We conclude that the analysis of samples containing alkenyl-acyl-PL by MALDI-TOF has to be performed with great caution. [Copyright &y& Elsevier]

Developmental competence of oocytes is compromised if they originate from atretic follicles. Apoptosis is the underlying process of atresia. Apoptotic changes in follicular cells are thought to influence the outcome of IVF. The aim of this study was to investigate apoptosis in different compartments of single bovine follicles (follicular wall, granulosa and cumulus cells (CC)) in relation to COC morphology, and to determine whether the addition, in vitro, of exogenous follicular cells from atretic follicles to maturing cumulus oocyte complexes (COCs) influenced the development of oocytes.Antral follicles were dissected from bovine ovaries and opened to obtain COCs and free floating granulosa cells (GC). The COCs were classified according to morphology. Apoptosis was determined in cumulus and granulosa cells and in homogenates of the remaining follicular wall.For every morphological class of COCs, a large variability of apoptotic expression was found in all follicle compartments. Follicular wall apoptosis was not correlated to COC morphology or to the percentage of apoptotic granulosa or cumulus cells. In grade 1 (best morphology) COCs, the degree of apoptosis in granulosa cells was comparable to cumulus cell apoptosis (P<0.01). The overall expression of apoptosis in granulosa cells of follicles containing grade 3 COCs (median±median absolute deviation: 37.8±13.8%) was significantly higher (P<0.05) than in follicles with grade 1 (22.7±10.4%) or grade 2 COCs (20.0±17.0%). About 48.3% of grade 3 COCs possessed strongly apoptotic cumulus cells compared to 27.8 and 28.2% of grade 1 or grade 2 COCs, respectively. Nonapoptotic cumulus complexes were observed in grades 1 and 2 COCs only.Adding exogenous follicular cells from atretic follicles to bovine COCs (grades 1 and 2) during in vitro maturation (IVM) had no impact on fertilization, blastocyst formation or hatching after IVF. This is of particular practical relevance to embryo production after ovum pick up (OPU), as during this process, good quality COCs are cultured together with simultaneously collected slightly atretic COCs. Epidemiologically, a high-carotenoid intake via a fruit- and vegetable-rich diet is associated with a decreased risk of various forms of cancer. The mechanisms by which carotenoids exert this protective effect are controversial. In this study, we examined the potency of a range of carotenoids commonly found in human plasma to induce apoptosis in Jurkat E6.1 malignant T-lymphoblast cells. At a concentration of 20 μM, the order of potency to induce apoptosis after 24 h was: β-carotene > lycopene > lutein>β-cryptoxanthin=zeaxanthin. Canthaxanthin failed to induce apoptosis under these conditions. β-Carotene induced apoptosis in a time- and concentration-dependent manner with a lowest effective concentration of about 3 μM. Pre-conditioning of β-carotene for 72 h destroyed its pro-apoptotic activity almost completely, whereas degradation for 6 h or less did not, indicating that either β-carotene itself and/or an early degradation product of β-carotene are the death-inducing compounds. Apoptosis induced by β-carotene was characterized by chromatin condensation and nuclear fragmentation, DNA degradation, PARP cleavage and caspase-3 activation. The antioxidant BO-653 inhibited the degradation of β-carotene in vitro and significantly increased its cytotoxicity, indicating that a pro-oxidant effect of β-carotene is unlikely to cause its pro-apoptotic activity. The induction of apoptosis in transformed cells by carotenoids may explain their protective effect against cancer formation in humans. Mammalian seminal plasma contains a multitude of bioactive components, including lipids, glucose, mineral elements, metabolites, proteins, cytokines, and growth factors, with various functions during insemination and fertilization. The seminal plasma protein PDC-109 is one of the major soluble components of the bovine ejaculate and is crucially important for sperm motility, capacitation, and acrosome reaction. A hitherto underappreciated function of seminal plasma is its anti-microbial and antiviral activity, which may limit the sexual transmission of infectious diseases during intercourse. We have recently discovered that PDC-109 inhibits the membrane fusion activity of influenza virus particles and significantly impairs viral infections at micromolar concentrations. Here we investigated whether the antiviral activity of PDC-109 is restricted to Influenza or if other mammalian viruses are similarly affected. We focused on Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2), the etiological agent of the Coronavirus Disease 19 (COVID-19), thoroughly assessing PDC-109 inhibition with SARS-CoV-2 Spike (S)-pseudotyped reporter virus particles, but also live-virus infections. Consistent with our previous publications, we found significant virus inhibition, albeit accompanied by substantial cytotoxicity. However, using time-of-addition experiments we discovered a treatment regimen that enables virus suppression without affecting cell viability. We furthermore demonstrated that PDC-109 is also able to impair infections mediated by the VSV glycoprotein (VSVg), thus indicating a broad pan-antiviral activity against multiple virus species and families. Monitoring the temporal and spatial variation of soil properties is helpful to understand the evolution of soil properties and adjust the management method in time. Soil fertility evaluation is an urgent need to understand soil fertility level and prevent soil degradation. Here, we conducted an intensive field investigation in Chinese hickory (Carya cathayensis Sarg.) plantation to clarify the spatial and temporal variation of soil properties and its influencing factors, and to evaluate the change of soil fertility. The results showed that the soil pH and soil organic carbon (SOC) significantly increased from 2008 to 2018, while available nitrogen (AN) significantly decreased from 2008 to 2018. The semi-variance revealed that except available phosphorus (AP), the spatial dependencies of soil properties increased from 2008 to 2018. An increasing south-north gradient was found for soil AN, AP, available potassium (AK) and SOC and a decreasing south-north gradient was found for soil pH. The average soil fertility in the whole area was increased from 2008 to 2018. Our findings demonstrated that the changes of the management measures were the reason for the change of soil properties from 2008 to 2018. Preclinical management of patients with acute chest pain and their identification as candidates for urgent coronary revascularization without the use of high sensitivity troponin essays remains a critical challenge in emergency medicine. We enrolled 2760 patients (average age 70 years, 58.6% male) with chest pain and suspected ACS, who were admitted to the Emergency Department of the University Hospital Tübingen, Germany, between August 2016 and October 2020. Using 26 features, eight Machine learning models (non-deep learning models) were trained with data from the preclinical rescue protocol and compared to the "TropOut" score (a modified version of the "preHEART" score which consists of history, ECG, age and cardiac risk but without troponin analysis) to predict major adverse cardiac event (MACE) and acute coronary artery occlusion (ACAO). In our study population MACE occurred in 823 (29.8%) patients and ACAO occurred in 480 patients (17.4%). Interestingly, we found that all machine learning models outperformed the "TropOut" score. The VC and the LR models showed the highest area under the receiver operating characteristic (AUROC) for predicting MACE (AUROC = 0.78) and the VC showed the highest AUROC for predicting ACAO (AUROC = 0.81). A SHapley Additive exPlanations (SHAP) analyses based on the XGB model showed that presence of ST-elevations in the electrocardiogram (ECG) were the most important features to predict both endpoints.
