1. An essential host dietary fatty acid promotes TcpH inhibition of TcpP proteolysis promoting virulence gene expression in Vibrio cholerae

Abstract: ABSTRACT Vibrio cholerae is a Gram-negative gastrointestinal pathogen responsible for the diarrheal disease cholera. Expression of key virulence factors, cholera toxin and toxin-coregulated pilus, is regulated directly by ToxT and indirectly by two transmembrane transcription regulators (TTRs), ToxR and TcpP, that promote the expression of toxT. TcpP abundance and activity are controlled by TcpH, a single-pass transmembrane protein, which protects TcpP from a two-step proteolytic process known as regulated intramembrane proteolysis (RIP). The mechanism of TcpH-mediated protection of TcpP represents a major gap in our understanding of V. cholerae pathogenesis. The absence of tcpH leads to unimpeded degradation of TcpP in vitro and a colonization defect in a neonate mouse model of V. cholerae colonization. Here, we show that TcpH protects TcpP from RIP via direct interaction. We also demonstrate that α-linolenic acid, a dietary fatty acid, promotes TcpH-dependent inhibition of RIP via co-association of TcpP and TcpH molecules within detergent-resistant membranes (DRMs) in a mechanism requiring the TcpH transmembrane domain. Taken together, our data support a model where V. cholerae cells use exogenous α-linolenic acid to remodel the phospholipid bilayer in vivo, leading to co-association of TcpP and TcpH within DRMs where RIP of TcpP is inhibited by TcpH, thereby promoting V. cholerae pathogenicity.IMPORTANCEVibrio cholerae continues to pose a significant global burden on health and an alternative therapeutic approach is needed, due to evolving multidrug resistance strains. Transcription of toxT, stimulated by TcpP and ToxR, is essential for V. cholerae pathogenesis. Our results show that TcpP, one of the major regulators of toxT gene expression, is protected from proteolysis by TcpH, via direct interaction. Furthermore, we identified a gut metabolite, α-linolenic acid, that stimulates the co-association of TcpP and TcpH within detergent-resistant membranes (also known as lipid-ordered membrane domains), thereby supporting TcpH-dependent antagonism of TcpP proteolysis. Data presented here extend our knowledge of RIP, virulence gene regulation in V. cholerae, and, to the best of our knowledge, provides the first evidence that lipid-ordered membranes exist within V. cholerae. The model presented here also suggests that TTRs, common among bacteria and archaea, and co-component signal transduction systems present in Enterobacteria, could also be influenced similarly.

2. Activation of a Vibrio cholerae CBASS anti-phage system by quorum sensing and folate depletion

Abstract: ABSTRACT A major challenge faced by bacteria is infection by bacteriophage (phage). Abortive infection is one strategy for combating phage in which an infected cell kills itself to limit phage replication, thus protecting neighboring kin. One class of abortive infection systems is the cyclic oligonucleotide based anti-phage signaling system (CBASS) which relies on two core enzymatic activities; an oligo-nucleotide cyclase that is activated following phage infection and a cyclic-oligo-nucleotide sensitive effector whose activity kills the infected cell. However, the mechanisms behind the deployment and activation of these lethal CBASS systems prior to and following infection have largely remained a mystery. While exploring unique genomic features of the current pandemic Vibrio cholerae biotype El Tor for clues underlying its pandemic success we found its CBASS was spuriously activated by the folate biosynthesis inhibitor sulfamethoxazole, but only after the population had reached a high-cell density. This population density-dependent activity revealed that transcription of both the oligo-nucleotide cyclase, dncV, and the CBASS phospholipase effector, capV, is enhanced at high-cell density by quorum sensing. Taken together, these results demonstrate that the V. cholerae CBASS is deployed when the environment is densely populated and activated in response to a perturbation in folate biosynthesis. IMPORTANCE To counteract infection with phage, bacteria have evolved a myriad of molecular defense systems. Some of these systems initiate a process called abortive infection, in which the infected cell kills itself to prevent phage propagation. However, such systems must be inhibited in the absence of phage infection to prevent spurious death of the host. Here, we show that the cyclic oligonucleotide based anti-phage signaling system (CBASS) accomplishes this by sensing intracellular folate molecules and only expressing this system in a group. These results enhance our understanding of the evolution of the seventh Vibrio cholerae pandemic and more broadly how bacteria defend themselves against phage infection.

Abstract: ABSTRACT To adapt and proliferate, bacteria must sense and respond to the ever-changing environment. Transmembrane transcription regulators (TTRs) are a family of one-component transcription regulators that respond to extracellular information and influence gene expression from the cytoplasmic membrane. How TTRs function to modulate expression of their target genes while localized to the cytoplasmic membrane remains poorly understood. In part, this is due to a lack of knowledge regarding the prevalence of TTRs among prokaryotes. Here, we show that TTRs are highly diverse and prevalent throughout bacteria and archaea. Our work demonstrates that TTRs are more common than previously appreciated and are enriched within specific bacterial and archaeal phyla and that many TTRs have unique transmembrane region properties that can facilitate association with detergent-resistant membranes. IMPORTANCE One-component signal transduction systems are the major class of signal transduction systems among bacteria and are commonly cytoplasmic. TTRs are a group of unique one-component signal transduction systems that influence transcription from the cytoplasmic membrane. TTRs have been implicated in a wide array of biological pathways critical for both pathogens and human commensal organisms but were considered to be rare. Here, we demonstrate that TTRs are in fact highly diverse and broadly distributed in bacteria and archaea. Our findings suggest that transcription factors can access the chromosome and influence transcription from the membrane in both archaea and bacteria. This study challenges thus the commonly held notion that signal transduction systems require a cytoplasmic transcription factor and highlights the importance of the cytoplasmic membrane in directly influencing signal transduction.

Abstract: Vibrio cholerae respires both aerobically and anaerobically and, while oxygen may be available to it during infection, other terminal electron acceptors are proposed for population expansion during infection. Unlike gastrointestinal pathogens that stimulate significant inflammation leading to elevated levels of oxygen or alternative terminal electron acceptors, V. cholerae infections are not understood to induce a notable inflammatory response. To ascertain the respiration requirements of V. cholerae during infection, we used Multiplex Genome Editing by Natural Transformation (MuGENT) to create V. cholerae strains lacking aerobic or anaerobic respiration. V. cholerae strains lacking aerobic respiration were attenuated in infant mice 105-fold relative to wild type, while strains lacking anaerobic respiration had no colonization defect, contrary to earlier work suggesting a role for anaerobic respiration during infection. Using several approaches, including one we developed for this work termed Comparative Multiplex PCR Amplicon Sequencing (CoMPAS), we determined that the bd-I and cbb3 oxidases are essential for small intestinal colonization of V. cholerae in the infant mouse. The bd-I oxidase was also determined as the primary oxidase during growth outside the host, making V. cholerae the only example of a Gram-negative bacterial pathogen in which a bd-type oxidase is the primary oxidase for energy acquisition inside and outside of a host.

Abstract: Abstract Pyrodictium delaneyi strain Hulk is a newly sequenced strain isolated from chimney samples collected from the Hulk sulfide mound on the main Endeavour Segment of the Juan de Fuca Ridge (47.9501 latitude, −129.0970 longitude, depth 2200 m) in the Northeast Pacific Ocean. The draft genome of strain Hulk shared 99.77% similarity with the complete genome of the type strain Su06T, which shares with strain Hulk the ability to reduce iron and nitrate for respiration. The annotation of the genome of strain Hulk identified genes for the reduction of several sulfur-containing electron acceptors, an unsuspected respiratory capability in this species that was experimentally confirmed for strain Hulk. This makes P. delaneyi strain Hulk the first hyperthermophilic archaeon known to gain energy for growth by reduction of iron, nitrate, and sulfur-containing electron acceptors. Here we present the most notable features of the genome of P. delaneyi strain Hulk and identify genes encoding proteins critical to its respiratory versatility at high temperatures. The description presented here corresponds to a draft genome sequence containing 2,042,801 bp in 9 contigs, 2019 protein-coding genes, 53 RNA genes, and 1365 hypothetical genes.

8. Achieving Single-Molecule Tracking of Subcellular Regulation in Bacteria during Real-Time Environmental Perturbations

Abstract: Bacteria rely on protein systems for regulation in response to external environmental signals. Single-molecule fluorescence imaging and tracking has elucidated the complex mechanism of these protein systems in a variety of bacteria. We recently investigated Vibrio cholerae, the Gram-negative bacterium responsible for the human cholera disease, and its regulation of the production of toxins and virulence factors through the membrane-localized transcription factors TcpP and ToxR. These experiments determined that TcpP and ToxR work cooperatively under steady-state conditions, but measurements of how these dynamical interactions change over the course of environmental perturbations were precluded by the traditional preparation of bacteria cells confined on agarose pads. Here, we address this gap in technology and access single-molecule dynamics during real-time changes by implementing two alternative sample preparations: device microfluidic device and chitosan-coated coverslips. We report the first demonstration of single-molecule tracking within live bacterial cells in a microfluidic device. Additionally, using the chitosan-coated coverslips, we show that real-time environmental changes impact TcpP-PAmCherry dynamics, activating a virulence condition in the bacteria about 45 minutes after dropping to pH 6 and about 20 minutes after inducing ToxR expression. These new technology advances open our ability for new experiments studying a variety of bacteria with single-molecule imaging and tracking during real-time environmental perturbations.

When growing without oxygen it may use other molecules that serve the same purpose as oxygen, acting as a terminal electron acceptor in an energy-generating process known as respiration. Given the largely anaerobic nature of the gastrointestinal tract, and the lack of significant inflammation during cholera infection, a process that can stimulate elevated levels of oxygen and other terminal electron acceptors, we sought to understand the respiratory mechanisms of V. cholerae during infection. We used a powerful genome-editing method to construct mutant strains of V. cholerae lacking some or all of the complement of proteins required for aerobic or anaerobic respiration. By analyzing these mutants in the laboratory and in intestinal colonization of infant mice, we determined that the ability to respire without oxygen is completely dispensable for V. cholerae to thrive during infection. We determined that two of the four oxygen-dependent respiration mechanisms are essential for V. cholerae to grow during infection, with the other two dispensable for wild type levels of colonization.

Abstract: Cholera is a diarrheal disease caused by the Gram-negative bacterium Vibrio cholerae. To reach the surface of intestinal epithelial cells, proliferate, and cause disease, V. cholerae tightly regulates the production of virulence factors such as cholera toxin (ctxAB) and the toxin-coregulated pilus (tcpA-F). ToxT is directly responsible for regulating these major virulence factors while TcpP and ToxR indirectly regulate virulence factor production by stimulating toxT expression. TcpP and ToxR are membrane-localized transcription activators (MLTAs) required to activate toxT expression. To gain a deeper understanding of how MLTAs identify promoter DNA while in the membrane, we tracked the dynamics of single TcpP-PAmCherry molecules in live cells using photoactivated localization microscopy and identified heterogeneous diffusion patterns. Our results provide evidence that (i) TcpP exists in three biophysical states (fast diffusion, intermediate diffusion, and slow diffusion), (ii) TcpP transitions between these different diffusion states, (iii) TcpP molecules in the slow diffusion state are interacting with the toxT promoter, and (iv) ToxR is not essential for TcpP to localize the toxT promoter. These data refine the current model of cooperativity between TcpP and ToxR in stimulating toxT expression and demonstrate that TcpP locates the toxT promoter independently of ToxR. IMPORTANCE Vibrio cholerae continues to be a public health threat throughout much of the world. Its ability to cause disease is governed by an unusual complex of regulatory proteins in the membrane of the cell, including ToxR and TcpP. These proteins collaborate to activate expression of the toxT gene, whose product activates genes for cholera toxin and other virulence factors. To study these membrane regulators, ToxR and TcpP, we applied superresolution imaging, which enables us to look at individual proteins in living cells. With this approach, we have uncovered dynamic intermolecular relationships between ToxR, TcpP, and toxT promoter DNA that dictate how toxT expression occurs. Because membrane regulators like ToxR and TcpP are broadly distributed in nature but poorly understood, this work describes mechanisms and approaches that will be of significant interest to a wide range of microbial scientists.

Abstract: Single-molecule fluorescence microscopy probes nanoscale, subcellular biology in real time. Existing methods for analyzing single-particle tracking data provide dynamical information, but can suffer from supervisory biases and high uncertainties. Here, we introduce a new approach to analyzing single-molecule trajectories: the Single-Molecule Analysis by Unsupervised Gibbs sampling (SMAUG) algorithm, which uses nonparametric Bayesian statistics to uncover the whole range of information contained within a single-particle trajectory (SPT) dataset. Even in complex systems where multiple biological states lead to a number of observed mobility states, SMAUG provides the number of mobility states, the average diffusion coefficient of single molecules in that state, the fraction of single molecules in that state, the localization noise, and the probability of transitioning between two different states. In this paper, we provide the theoretical background for the SMAUG analysis and then we validate the method using realistic simulations of SPT datasets as well as experiments on a controlled in vitro system. Finally, we demonstrate SMAUG on real experimental systems in both prokaryotes and eukaryotes to measure the motions of the regulatory protein TcpP in Vibrio cholerae and the dynamics of the B-cell receptor antigen response pathway in lymphocytes. Overall, SMAUG provides a mathematically rigorous approach to measuring the real-time dynamics of molecular interactions in living cells.Statement of SignificanceSuper-resolution microscopy allows researchers access to the motions of individual molecules inside living cells. However, due to experimental constraints and unknown interactions between molecules, rigorous conclusions cannot always be made from the resulting datasets when model fitting is used. SMAUG (Single-Molecule Analysis by Unsupervised Gibbs sampling) is an algorithm that uses Bayesian statistical methods to uncover the underlying behavior masked by noisy datasets. This paper outlines the theory behind the SMAUG approach, discusses its implementation, and then uses simulated data and simple experimental systems to show the efficacy of the SMAUG algorithm. Finally, this paper applies the SMAUG method to two model living cellular systems—one bacterial and one mammalian—and reports the dynamics of important membrane proteins to demonstrate the usefulness of SMAUG to a variety of systems.

Abstract: The momentum and helicity density distributions of the strange quark sea in the nucleon are obtained in leading order from charged-kaon production in deep-inelastic scattering on the deuteron. The distributions are extracted from spin-averaged K+/- multiplicities, and from K+/- and inclusive double-spin asymmetries for scattering of polarized positrons by a polarized deuterium target. The shape of the momentum distribution is softer than that of the average of the ubar and dbar quarks. In the region of measurement 0.02 < x < 0.6 and Q^2 > 1.0 GeV^2, the helicity distribution is zero within experimental uncertainties., 5 pages, 5 figures, 1 table; text slightly expanded, 10 references added

Abstract: A series of semi-inclusive deep-inelastic scattering measurements on deuterium, helium, neon, krypton, and xenon targets has been performed in order to study hadronization. The data were collected with the HERMES detector at the DESY laboratory using a 27.6 GeV positron or electron beam. Hadron multiplicities on nucleus A relative to those on the deuteron, R_A^h, are presented for various hadrons (\pi^+, \pi^-, \pi^0, K^+, K^-, p, and \bar{p}) as a function of the virtual-photon energy \nu, the fraction z of this energy transferred to the hadron, the photon virtuality Q^2, and the hadron transverse momentum squared p_t^2. The data reveal a systematic decrease of R_A^h with the mass number A for each hadron type h. Furthermore, R_A^h increases (decreases) with increasing values of \nu (z), increases slightly with increasing Q^2, and is almost independent of p_t^2, except at large values of p_t^2. For pions two-dimensional distributions also are presented. These indicate that the dependences of R_A^{\pi} on \nu and z can largely be described as a dependence on a single variable L_c, which is a combination of \nu and z. The dependence on L_c suggests in which kinematic conditions partonic and hadronic mechanisms may be dominant. The behaviour of R_A^{\pi} at large p_t^2 constitutes tentative evidence for a partonic energy-loss mechanism. The A-dependence of R_A^h is investigated as a function of \nu, z, and of L_c. It approximately follows an A^{\alpha} form with \alpha \approx 0.5 - 0.6., Comment: 33 pages, 12 figures

Abstract: Hard exclusive electroproduction of π + π − pairs off hydrogen and deuterium targets has been studied by the HERMES experiment at DESY. Legendre moments 〈 P 1 〉 and 〈 P 3 〉 of the angular distributions of π + mesons in the center-of-mass frame of the pair have been measured for the first time. Their dependence on the π + π − invariant mass can be understood as being due to the interference between relative P -wave (isovector) and S -, D -wave (isoscalar) states of the two pions. The increase in magnitude of 〈 P 1 〉 as Bjorken x increases is interpreted in the framework of generalized parton distributions as an enhancement of flavour non-singlet q q ¯ exchange for larger values of x , which leads to a sizable admixture of isoscalar and isovector pion pairs. In addition, the interference between P -wave and D -wave states separately for transverse and longitudinal pion pairs has been studied. The data indicate that in the f 2 ( 1270 ) region at 〈 Q 2 〉 = 3 GeV 2 higher-twist effects can be as large as the leading-twist longitudinal component.

Abstract: Azimuthal asymmetries in exclusive electroproduction of real photons are measured for the first time with respect to transverse target polarisation, providing new constraints on Generalized Parton Distributions. From the same data set on a hydrogen target, new results for the beam-charge asymmetry are also extracted with better precision than those previously reported. Abstract: The exclusive electroproduction of pi+ mesons was studied with the Hermes spectrometer at the DESY laboratory by scattering 27.6 GeV positron and electron beams off an internal hydrogen gas target. The virtual-photon cross sections were measured as a function of the Mandelstam variable t and the squared four momentum -Q^2 of the exchanged virtual photon. A model calculation based on Generalized Parton Distributions is in fair agreement with the data at low values of |t| if power corrections are included. Abstract: Precise measurements of the spin structure functions of the proton $g_1^p(x,Q^2)$ and deuteron $g_1^d(x,Q^2)$ are presented over the kinematic range $0.0041 \leq x \leq 0.9$ and $0.18 $ GeV$^2$ $\leq Q^2 \leq 20$ GeV$^2$. The data were collected at the HERMES experiment at DESY, in deep-inelastic scattering of 27.6 GeV longitudinally polarized positrons off longitudinally polarized hydrogen and deuterium gas targets internal to the HERA storage ring. The neutron spin structure function $g_1^n$ is extracted by combining proton and deuteron data. The integrals of $g_1^{p,d}$ at $Q^2=5$ GeV$^2$ are evaluated over the measured $x$ range. Neglecting any possible contribution to the $g_1^d$ integral from the region $x \leq 0.021$, a value of $0.330 \pm 0.011\mathrm{(theo.)}\pm0.025\mathrm{(exp.)}\pm 0.028$(evol.) is obtained for the flavor-singlet axial charge $a_0$ in a leading-twist NNLO analysis.

Abstract: A measurement of the beam-spin asymmetry in the azimuthal distribution of pions produced in semi-inclusive deep-inelastic scattering off protons is presented. The measurement was performed using the HERMES spectrometer with a hydrogen gas target and the longitudinally polarized 27.6 GeV positron beam of HERA. The sinusoidal amplitude of the dependence of the asymmetry on the angle ϕ of the hadron production plane around the virtual photon direction relative to the lepton scattering plane was measured for π + π + , π − π − and π 0 π 0 mesons. The dependence of this amplitude on the Bjorken scaling variable and on the pion fractional energy and transverse momentum is presented. The results are compared to theoretical model calculations.

Abstract: The first observation of an azimuthal cross section asymmetry with respect to the charge of the incoming lepton beam is reported from a study of hard exclusive electroproduction of real photons. The data have been accumulated by the HERMES experiment at DESY, in which the HERA 27.6 GeV electron or positron beam scattered off an unpolarized hydrogen gas target. The observed asymmetry is attributed to the interference between the Bethe-Heitler process and the deeply virtual Compton scattering (DVCS) process. The interference term is sensitive to DVCS amplitudes, which provide the most direct access to generalized parton distributions. © 2007 The American Physical Society.

Abstract: The transfer of polarization from a high-energy positron to a {lambda}{sup 0} hyperon produced in semiinclusive deep-inelastic scattering has been measured. The data have been obtained by the HERMES experiment at DESY using the 27.6 GeV longitudinally polarized positron beam of the HERA collider and unpolarized gas targets internal to the positron (electron) storage ring. The longitudinal spin-transfer coefficient is found to be D{sub LL{sup '}}{sup {lambda}}=0.11{+-}0.10(stat){+-}0.03(syst) at an average fractional energy carried by the {lambda}{sup 0} hyperon =0.45. The dependence of D{sub LL{sup '}}{sup {lambda}} on both the fractional energy z and the fractional longitudinal momentum x{sub F} is presented.

Abstract: Single-spin asymmetries in the semi-inclusive production of charged pions in deep-inelastic scattering from transversely and longitudinally polarized proton targets are combined to evaluate the subleading-twist contribution to the longitudinal case. This contribution is significantly positive for π + mesons and dominates the asymmetries on a longitudinally polarized target previously measured by Hermes . The subleading-twist contribution for π − mesons is found to be small.

Abstract: A search for an exotic baryon resonance with $S=\ensuremath{-}2$, $Q=\ensuremath{-}2$ has been performed in quasireal photoproduction on a deuterium target through the decay channel ${\ensuremath{\Xi}}^{\ensuremath{-}}{\ensuremath{\pi}}^{\ensuremath{-}}\ensuremath{\rightarrow}\ensuremath{\Lambda}{\ensuremath{\pi}}^{\ensuremath{-}}{\ensuremath{\pi}}^{\ensuremath{-}}\ensuremath{\rightarrow}p{\ensuremath{\pi}}^{\ensuremath{-}}{\ensuremath{\pi}}^{\ensuremath{-}}{\ensuremath{\pi}}^{\ensuremath{-}}$. No evidence for a previously reported ${\ensuremath{\Xi}}^{--}(1860)$ resonance is found in the ${\ensuremath{\Xi}}^{\ensuremath{-}}{\ensuremath{\pi}}^{\ensuremath{-}}$ invariant mass spectrum. An upper limit for the photoproduction cross section of 2.1 nb is found at the 90% confidence level. The photoproduction cross section for the ${\ensuremath{\Xi}}^{0}(1530)$ is found to be between 9 and 24 nb.

Abstract: The HERMES hydrogen and deuterium nuclear-polarized gas targets have been in use since 1996 with the polarized electron beam of HERA at DESY to study the spin structure of the nucleon. Polarized atoms from a Stern-Gerlach Atomic Beam Source are injected into a storage cell internal to the HERA electron ring. Atoms diffusing from the center of the storage cell into a side tube are analyzed to determine the atomic fraction and the atomic polarizations. The atoms have a nuclear polarization, the axis of which is defined by an external magnetic holding field. The holding field was longitudinal during 1996-2000, and was changed to transverse in 2001. The design of the target is described, the method for analyzing the target polarization is outlined, and the performance of the target in the various running periods is presented. © 2004 Elsevier B.V. Abstract: Our study aimed to assess the severity of severe acute respiratory syndrome coronavirus 2 infection in hospitalized infants under 40 days old, across 21 Belgian hospitals between 2020 and 2022. Of the 365 infants studied, 14.2% needed respiratory support. The median hospital stay was 3 days (interquartile range, 2-4), and there were no deaths. 32. Spontaneous Entrainment of Running Cadence to Music Tempo.

Abstract: Background: Since accumulating evidence suggests that step rate is strongly associated with running-related injuries, it is important for runners to exercise at an appropriate running cadence. As music tempo has been shown to be capable of impacting exercise performance of repetitive endurance activities, it might also serve as a means to (re)shape running cadence. The aim of this study was to validate the impact of music tempo on running cadence., Methods: Sixteen recreational runners ran four laps of 200 m (i.e. 800 m in total); this task was repeated 11 times with a short break in between each four-lap sequence. During the first lap of a sequence, participants ran at a self-paced tempo without musical accompaniment. Running cadence of the first lap was registered, and during the second lap, music with a tempo matching the assessed cadence was played. In the final two laps, the music tempo was either increased/decreased by 3.00, 2.50, 2.00, 1.50, or 1.00 % or was kept stable. This range was chosen since the aim of this study was to test spontaneous entrainment (an average person can distinguish tempo variations of about 4 %). Each participant performed all conditions., Results: Imperceptible shifts in musical tempi in proportion to the runner's self-paced running tempo significantly influenced running cadence ( p < .001). Contrasts revealed a linear relation between the tempo conditions and adaptation in running cadence ( p < .001). In addition, a significant effect of condition on the level of entrainment was revealed ( p < .05), which suggests that maximal effects of music tempo on running cadence can only be obtained up to a certain level of tempo modification. Finally, significantly higher levels of tempo entrainment were found for female participants compared to their male counterparts ( p < .05)., Conclusions: The applicable contribution of these novel findings is that music tempo could serve as an unprompted means to impact running cadence. As increases in step rate may prove beneficial in the prevention and treatment of common running-related injuries, this finding could be especially relevant for treatment purposes, such as exercise prescription and gait retraining., Key Points: Music tempo can spontaneously impact running cadence.A basin for unsolicited entrainment of running cadence to music tempo was discovered.The effect of music tempo on running cadence proves to be stronger for women than for men.

Abstract: Music is known to be capable of reducing perceived exertion during strenuous physical activity. The current interpretation of this modulating effect of music is that music may be perceived as a diversion from unpleasant proprioceptive sensations that go along with exhaustion. Here we investigated the effects of music on perceived exertion during a physically strenuous task, varying musical agency, a task that relies on the experience of body proprioception, rather than simply diverting from it. For this we measured psychologically indicated exertion during physical workout with and without musical agency while simultaneously acquiring metabolic values with spirometry. Results showed that musical agency significantly decreased perceived exertion during workout, indicating that musical agency may actually facilitate physically strenuous activities. This indicates that the positive effect of music on perceived exertion cannot always be explained by an effect of diversion from proprioceptive feedback. Furthermore, this finding suggests that the down-modulating effect of musical agency on perceived exertion may be a previously unacknowledged driving force for the development of music in humans: making music makes strenuous physical activities less exhausting.

Abstract: Three cytochrome P450 monooxygenases belonging to the CYP52 family were isolated from the genome of the sophorolipid-producing yeast Candida bombicola using degenerate PCR and genomic walking. One gene displayed high identity with the CYP52E members and was classified into this group (CYP52E3), whereas the other genes belonged to new groups: CYP52M and CYP52N. CYP52E3 and CYP52N1 turned out to be of no relevance for sophorolipid production, but show clear upregulation when the yeast cells are grown on alkanes as the sole carbon source. On the other hand, CYP52M1 is clearly upregulated during sophorolipid synthesis and very likely takes part in sophorolipid formation.

Abstract: Single-spin asymmetries for semi-inclusive electroproduction of charged pions in deep-inelastic scattering of positrons are measured for the first time with transverse target polarization. The asymmetry depends on the azimuthal angles of both the pion (phi) and the target spin axis (phi(S)) about the virtual-photon direction and relative to the lepton scattering plane. The extracted Fourier component sin((phi+phi(S))(pi)(UT) is a signal of the previously unmeasured quark transversity distribution, in conjunction with the Collins fragmentation function, also unknown. The component sin((phi-phi(S)(pi)(UT) arises from a correlation between the transverse polarization of the target nucleon and the intrinsic transverse momentum of quarks, as represented by the previously unmeasured Sivers distribution function. Evidence for both signals is observed, but the Sivers asymmetry may be affected by exclusive vector meson production.
