Float bay production of seedlings accounts for approximately 62% of tobacco (Nicotiana tabacum L.) transplant production in South Carolina. Float systems utilize styrofoam trays containing soilless media that float in plastic-lined bays filled with nutrient solution. Oomycete fungi with motile zoospores, such as Pythium spp., can devastate plants grown hydroponically (3). This poses a serious concern for growers because no fungicides are labeled for control of Pythium spp. in nutrient solutions for tobacco greenhouse float systems. In May 1996, Pythium myriotylum Drechs. was recovered from diseased seedlings produced in float bays in Walterboro, SC. The pathogen was isolated on PAR selective medium (2). Koch's postulates were fulfilled by inoculating plants in an experimental float bay and then reisolating the pathogen on PAR from symptomatic plants. Diseased plants appeared chlorotic and stunted, with water-soaked lesions on the stem. Root systems were severely necrotic and infected plants could not be used as transplants. Disease development was confined to plants sharing a common nutrient solution in individual bays. Within affected bays nearly 100% of plants were diseased, suggesting pathogen spread through the nutrient solution. At the time of disease onset, the grower was attempting a second crop of transplants. The high late-season temperatures were favorable for P. myriotylum, which has a growth optimum of 37°C. Primary inoculum could potentially arise from contaminated media or water and/or improper sanitation procedures in greenhouse maintenance. Commercially available strains of hybrid white, hybrid off-white, and brown Agaricus bisporus mushrooms were compared for resistance to green mold caused by Trichoderma harzianum biotype 4 (Th4). Seven mushroom spawn strains were assessed for total weight of mushrooms (grams per 0.1 m2) with or without the addition of an aqueous Th4 spore suspension added at spawning time. Cropping studies were conducted at the Mushroom Research Center (Pennsylvania State University) to emulate commercial growing operations. Excessive spawn handling had no significant effect on development of green mold. Severity of green mold was related to time between infestation and green mold appearance, with more significant yield losses occurring when green sporulation was detected early in production. Significant differences in yield were measured among mushroom strains in response to Th4 infestation. Hybrid white strains were extremely susceptible, with a mean yield loss of 96%. Hybrid off-white strains exhibited intermediate susceptibility, with mean yield losses of 56 to 73%. Brown strains were highly resistant, with mean yield losses of 9 to 16%. From these findings, we report the existence of green mold resistance, with a continuum of resistance among spawn strains. The findings suggest use of brown strains to manage green mold outbreaks, particularly where benomyl resistance in Trichoderma spp. is a threat.

The visual system is comprised of many specialized cell types that are essential for relaying sensory information about an animal's surroundings to the brain. The cells present in ocular tissue are notoriously delicate, making it particularly challenging to section thin slices of unfixed tissue. Maintaining the morphology of the native tissue is crucial for accurate observations by either conventional staining techniques or in this instance matrix-assisted laser desorption ionization (MALDI IMS) or imaging using mass spectrometry. As vision loss is a significantly debilitating condition, studying molecular mechanisms involved in the process of vision loss is a critically important area of research.

Pesticides are widely used in agriculture to control weeds, pests and diseases. Successful control is dependent on the compound reaching the target site within the organism after spray or soil application. Conventional methods for determining uptake and movement of herbicides and pesticides include autoradiography, liquid scintillation and chromatographic techniques such as high-performance liquid chromatography (HPLC). Autoradiography using radiolabelled compounds provides the best indication of a compound's movement within the plant system. Autoradiography is an established technique but it relies on the synthesis of radiolabelled compounds. The distribution of four sulfonylurea herbicides in sunflower plants has been studied 24 h after foliar application. The use of matrix-assisted laser desorption/ionisation mass spectrometry imaging (MALDI-MSI) images of protonated molecules and fragment ions (resulting from fragmentation at the urea bond within the sulfonylurea herbicides) has provided evidence for translocation above and below the application point. The translocation of nicosulfuron and azoxystrobin within the same plant system has also been demonstrated following their application to the plant stem. This study provides evidence that MALDI-MSI has great potential as an analytical technique to detect and assess the foliar, root and stem uptake of agrochemicals, and to reveal their distribution through the plant once absorbed and translocated.

AZD1152 is a highly selective Aurora B kinase inhibitor currently undergoing Phase I and II clinical evaluation in patients with acute myelogenous leukemia and advanced solid malignancies. We have established two AZD1152-resistant cell lines from SW620 colon and MiaPaCa pancreatic carcinoma lines, which are >100-fold resistant to the active metabolite of AZD1152, AZD1152 HQPA and interestingly, cross-resistant to the pan-Aurora kinase inhibitor, VX-680/MK0457. Using whole-genome microarray analysis and comparative genomic hybridization, we were able to identify MDR1 and BCRP as the causative genes that underlie AZD1152 HQPA-resistance in these models. Furthermore, the upregulation of either of these genes is sufficient to render in vivo tumor growth insensitive to AZD1152. Finally, the upregulation of MDR1 or BCRP is predictive of tumor cell sensitivity to this agent, both in vitro and in vivo. The data provide a genetic basis for resistance to Aurora kinase inhibitors, which could be utilized to predict clinical response to therapy.

Small-molecule inhibitors of the Aurora A and B kinases interfere with mitotic centrosome function and disrupt the mitotic spindle assembly checkpoint resulting in polyploidization and apoptosis of proliferating cells. As such, several Aurora kinase inhibitors are at various stages of clinical development as anticancer agents. To identify candidate apoptosis-sensitizing genes that could be exploited in combination with Aurora kinase inhibitors in malignant glioma, we have carried out global gene expression analysis in a D54MG glioma cell derivative treated with three Aurora kinase inhibitors chosen for their distinctive selectivities: MLN8054 (Aurora A-selective), AZD1152 (Aurora B-selective), and VX-680 (Aurora A/B). The modulation of apoptotic gene expression by p53 under these conditions was ascertained, as p53 expression can be toggled on and off in this D54MG derivative by virtue of a stable, inducible, p53-targeting short hairpin RNA (D54MG(shp53)). This analysis identified the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) death receptor, TRAIL receptor 2 (TRAIL-R2), as an apoptosis-sensitizing gene induced selectively following inhibition of Aurora B. In glioma cell lines where TRAIL-R2 was induced following polyploidization, the sensitivity, kinetics, and magnitude of TRAIL-mediated apoptosis were enhanced. Our data shed light on the apoptotic program induced during polyploidization and suggest that TRAIL-R2 activation is a putative point of therapeutic intervention in combination with inhibitors of Aurora B.

Adaptive mesh methods are reliable and efficient approaches in hydrological and hydraulic modelling. In some cases, however, big challenges are faced when constructing the adaptive meshes that not only have a good quality but also maintain the integrity of the geological structures. In this paper, a local refinement technique using Delaunay triangulation method is developed for these purposes. Specific treatments are proposed to deal with the soil interfaces and boundaries usually encountered in geological structures. The application and quantitative evaluation show that a satisfactory result has been obtained for a complicated geological slope.

Geodiversity has been used as a surrogate for biodiversity when species locations are unknown, and this utility can be extended to situations where species locations are in flux. Recently, scientists have designed conservation networks that aim to explicitly represent the range of geophysical environments, identifying a network of physical stages that could sustain biodiversity while allowing for change in species composition in response to climate change. Because there is no standard approach to designing such networks, we compiled 8 case studies illustrating a variety of ways scientists have approached the challenge. These studies show how geodiversity has been partitioned and used to develop site portfolios and connectivity designs; how geodiversity-based portfolios compare with those derived from species and communities; and how the selection and combination of variables influences the results. Collectively, they suggest 4 key steps when using geodiversity to augment traditional biodiversity-based conservation planning: create land units from species-relevant variables combined in an ecologically meaningful way; represent land units in a logical spatial configuration and integrate with species locations when possible; apply selection criteria to individual sites to ensure they are appropriate for conservation; and develop connectivity among sites to maintain movements and processes. With these considerations, conservationists can design more effective site portfolios to ensure the lasting conservation of biodiversity under a changing climate.

Management of Meloidogyne incognita (root-knot nematode) in cotton in the United States was substantially affected by the decision to stop production of aldicarb by its principle manufacturer in 2011. The remaining commercially available tools to manage M. incognita included soil fumigation, nematicide seed treatments, postemergence nematicide application, and cultivars partially resistant to M. incognita. Small plot field studies were conducted on a total of nine sites from 2011–2013 to examine the effects of each of these tools alone or in combinations, on early season galling, late-season nematode density in soil, yield, and value ($/ha = lint value minus chemical costs/ha). The use of a partially resistant cultivar resulted in fewer galls/root system at 35 d after planting in eight of nine tests, lower root-knot nematode density late in the growing season for all test sites, higher lint yield in eight of nine sites, and higher value/ha in six of nine sites. Galls per root were reduced by aldicarb in three of nine sites and by 1,3-dichloropropene (1,3-D) in two of eight sites, relative to the nontreated control (no insecticide or nematicide treatment). Soil fumigation reduced M. incognita density late in the season in three of nine sites. Value/ha was not affected by chemical treatment in four of nine sites, but there was a cultivar × chemical interaction in four of nine sites. When value/ha was affected by chemical treatment, the nontreated control had a similar value to the treatment with the highest value/ha in seven of eight cultivar-site combinations. The next “best” value/ha were associated with seed treatment insecticide (STI) + oxamyl and aldicarb (similar value to the highest value/ha in six of eight cultivar-site combinations). The lowest valued treatment was STI + 1,3-D. In a semi-arid region, where rainfall was low during the spring for all three years, cultivars with partial resistance to M. incognita was the most profitable method of managing root-knot nematode in cotton.

In landslide-prone areas the magnitude of events is related to recurring rainfall intensity. In a large sector of the Sannio Apennines (Southern Italy), predictive mapping of recurrent shallow landslides was undertaken by combining deterministic and probabilistic predictive approaches. This, with the aim to minimize the negative influence of the uniform distribution of the initial water table depth in steady condition that usually influence the theoretical instability resulting from the application of methods for large-scale estimation. The deterministic approach was performed by means of the Transient Rainfall Infiltration and Grid-based Regional Slope-stability model to obtain triggering maps in multi-temporal transient pore-water pressures. The optimized physical modeling was validated by back-analysis on large-magnitude landslide events which occurred in 2003 by means of the introduction of two cross-mapping correlation indexes. Subsequently, different predictive scenarios were proposed for different probabilistic return periods of the rainstorm events. The output data permitted the definition of a linear log regression curve to estimate the theoretical instability of the study area. This curve is defined as a function of cumulative precipitation, duration and return periods of the possible rainfall events. © 2013 Springer-Verlag Berlin Heidelberg.

β-CuI nanocrystallites (NCs) are synthesized on the atomically smooth graphite basal plane surface using a new hybrid electrochemical/chemical (E/C) method. This method involves the following steps: (1) electrochemical deposition of copper NCs onto an electrode surface, (2) electrochemical oxidation of these copper NCs to yield Cu2O, and (3) displacement of oxygen in Cu2O by iodide in an aqueous KI solution. Dispersions of CuI NCs having mean diameters ranging from 10 to 180 A were prepared using the E/C method. Selected-area electron diffraction analysis reveals that β-CuI NCs are obtained; these NCs are epitaxially aligned with the hexagonal periodicity of the graphite surface, as are the Cu2O precursor particles. Intraspecific variation in nutrient reserves is believed to affect reproductive success of female waterfowl during egg laying and incubation (the nutrient-limitation hy- pothesis). We tested the nutrient-limitation hypothesis in Canvasbacks (Aythya vahsineria) and Redheads (A. americana) by relating the body masses of incubating females with sub- sequent nest success, brood survival, and adult survival. Our analyses were based on 392 Canvasbacks and 16 Redheads captured and weighed during late incubation in southwestern Manitoba during 1983-1993. For Canvasbacks, there was a significant interaction between body mass and age; failed adults (ASY) weighed 3 g more than successful adults (P = 0.65) but failed yearlings (SY) weighed 37 g less than successful yearlings (P = 0.03). For Redheads, nest success was unrelated to body mass. For both species, brood survival and adult survival (i.e., return rates) were unrelated to female body mass. Thus, future reproductive success was correlated with body mass in only one out of seven test comparisons (i.e., nest success of yearling Canvasbacks). Although we had large sample sizes and used similar methods, our results were notably different from a previous study of prairie dabbling ducks (Anas spp.), where light-weight females were more likely to experience nest failure. For both studies, we believe that investigator-induced nest abandonment was an important cause ofcondition- dependent nest failure. We conclude that nutrient reserves were of minor importance in determining future reproductive success of adult Canvasbacks and Redheads, but may be important for yearlings.

Purpose To develop a method for generating high spatial resolution (10 µm) matrix-assisted laser desorption ionization (MALDI) images of lipids in rodent optic nerve tissue. Methods Ice-embedded optic nerve tissue from rats and mice were cryosectioned across the coronal and sagittal axes of the nerve fiber. Sections were thaw mounted on gold-coated MALDI plates and were washed with ammonium acetate to remove biologic salts before being coated in 2,5-dihydroxybenzoic acid by sublimation. MALDI images were generated in positive and negative ion modes at 10 µm spatial resolution. Lipid identification was performed with a high mass resolution Fourier transform ion cyclotron resonance mass spectrometer. Results Several lipid species were observed with high signal intensity in MALDI images of optic nerve tissue. Several lipids were localized to specific structures including in the meninges surrounding the optic nerve and in the central neuronal tissue. Specifically, phosphatidylcholine species were observed throughout the nerve tissue in positive ion mode while sulfatide species were observed in high abundance in the meninges surrounding the optic nerve in negative ion mode. Accurate mass measurements and fragmentation using sustained off-resonance irradiation with a high mass resolution Fourier transform ion cyclotron resonance mass spectrometer instrument allowed for identification of lipid species present in the small structure of the optic nerve directly from tissue sections. Conclusions An optimized sample preparation method provides excellent sensitivity for lipid species present within optic nerve tissue. This allowed the laser spot size and fluence to be reduced to obtain a high spatial resolution of 10 µm. This new imaging modality can now be applied to determine spatial and molecular changes in optic nerve tissue with disease.

A high temperature field reversed configuration (FRC) has been produced in the newly built, world’s largest compact toroid (CT) facility, C-2, by colliding and merging two high-β CTs produced using the advanced field-reversed θ-pinch technology. This long-lived, stable merged state exhibits the following key properties: (1) apparent increase in the poloidal flux from the first pass to the final merged state, (2) significantly improved confinement compared to conventional θ-pinch FRCs with flux decay rates approaching classical values in some cases, (3) strong conversion from kinetic energy into thermal energy with total temperature (Te + Ti) exceeding 0.5 keV, predominantly into the ion channel. Detailed modeling using a new 2-D resistive magnetohydrodynamic (MHD) code, LamyRidge, has demonstrated, for the first time, the formation, translation, and merging/reconnection dynamics of such extremely high-β plasmas.

Eight rhesus macaques inoculated with molecularly cloned SIV mac 239 developed viremia and virus-binding antibod-ies, but only one (macaque 2D) developed neutralizing antibodies to the virus. Viremia persisted in macaque 2D even in the presence of neutralizing antibodies. Neutralizing antibodies in the plasma collected from macaque 2D late in infection neutralized virus isolated early in infection. In contrast, these antibodies failed to neutralize the plasma viruses isolated after the appearance of neutralizing antibodies. Only antigenic variants were isolated from blood, spleen, and lymph nodes. Viruses isolated from other macaques that did not develop neutralizing antibodies were neutralized by 2D serum and were of the parental (SIV mac 239) phenotype. The variant viruses maintained their strict tropism for lymphocytes, similar to the parental virus.

A hot stable field-reversed configuration (FRC) has been produced in the C-2 experiment by colliding and merging two high-β plasmoids preformed by the dynamic version of field-reversed θ-pinch technology. The merging process exhibits the highest poloidal flux amplification obtained in a magnetic confinement system (over tenfold increase). Most of the kinetic energy is converted into thermal energy with total temperature (T{i}+T{e}) exceeding 0.5 keV. The final FRC state exhibits a record FRC lifetime with flux confinement approaching classical values. These findings should have significant implications for fusion research and the physics of magnetic reconnection.

A commercial hybrid quadrupole time-of-flight mass spectrometer has been modified for high-speed matrix-assisted laser desorption ionisation (MALDI) imaging using a short-pulse optical technology Nd:YVO(4) laser. The laser operating in frequency-tripled mode (lambda = 355 nm) is capable of delivering 1.5-ns pulses of energy at up to 8 microJ at 5-10 kHz and 3 microJ at 20 kHz. Experiments to improve beam homogeneity and reduce laser speckle by mechanical vibration of the fibre-optic laser delivery system are reported along with data from trial and tissue imaging experiments using the modified instrument. The laser appeared to yield best results for MALDI-MS imaging experiments when operating at repetition rates 5-10 kHz. Combining this with raster imaging allowed images of rat brain sections to be recorded in 37 min. Similarly, images of the distribution of peptides in "on-tissue" digest experiments from tumour tissues were recorded in 1 h and 30 min rather than the 8-h acquisition time previously used. A brief investigation of targeted protein analysis/imaging by multiple reaction monitoring experiments "on-tissue" is reported. A total of 26 transitions were recorded over a 3-s cycle time and images of abundant proteins were successfully recorded.

Comparison of neutralization of SIVmac251 in primary macrophage cultures with neutralization in lymphocytes (CEM174 cells) showed that neutralizing antibodies induced by SIV251 in infected rhesus macaques protected both macrophages and T lymphocytes against infection when the virus was preincubated with the antibodies. In macrophages, the neutralizing antibodies also protected against infection when added 1 hour after the virus. Addition of antisera to macrophages between 24 and 48 hours after virus inoculation resulted in infection with continuous release of small amounts of p24 into the supernatant fluids but these antibody-treated cultures failed to exhibit cytopathic virus replication. In contrast, the same neutralizing antisera did not protect lymphocytes against infection and subsequent cytopathic replication of the virus when added only 1 hour after virus inoculation. This distinction in the effect that neutralizing antibodies had on the development of cytopathic infection in lymphocytes and macrophages when added after virus inoculation, suggests that they could alter the dynamics of virus replication and therefore the pathogenesis of disease.

Matrix-assisted laser desorption/ionisation mass spectrometry imaging (MALDI-MSI) has been used to image the distribution of the pesticide nicosulfuron (2-[[(4,6-dimethoxypyrimidin-2-yl)aminocarbonyl]aminosulfonyl]-N,N-dimethyl-3-pyridinecarboxamide) in plant tissue using direct tissue imaging following root and foliar uptake. Sunflower plants inoculated with nicosulfuron were horizontally sectioned at varying distances along the stem in order to asses the extent of translocation; uptake via the leaves following foliar application to the leaves and uptake via the roots from a hydroponics system were compared. An improved sample preparation methodology, encasing samples in ice, allowed sections from along the whole of the plant stem from the root bundle to the growing tip to be taken. Images of fragment ions and alkali metal adducts have been generated that show the distribution of the parent compound and a phase 1 metabolite in the plant. Positive and negative controls have been included in the images to confirm ion origin and prevent false-positive results which could originate from endogenous compounds present within the plant tissue.

Rapidity distributions of protons from central 197Au+197Au collisions measured by the E895 Collaboration in the energy range from (2-8)A GeV at the Brookhaven AGS are presented. Longitudinal flow parameters derived using a thermal model including collective longitudinal expansion are extracted from these distributions. The results show an approximately linear increase in the longitudinal flow velocity,betagamma(L), as a function of the logarithm of beam energy.

Limit equilibrium methods are widely used for slope design applications, dynamically coupled hydrological-slope stability models and geomorphological investigations of long-term stability. Typically, computer codes generate a single minimum factor of safety and associated slip surface. Although this is attractive in that a single statement of stability is generated, such an approach ignores the many other slip surfaces that have been analysed, some of which may be capable of providing important additional information on the stability of the slope. In this paper the authors show that it is necessary to retain and visualise all slip surface information to determine the full extent of potential slope instability.

The PERIMOVE model is capable of simulating moisture and heat transfers within soil profiles. In addition, the simulation of these processes formed the basis for the prediction of the volume changes that moisture redistribution and freezing create within soil profiles and the resulting movement of material and loss of soil strength during freezing and thawing cycles. In order to aid the development and validation of the model a suite of data sources have been identified and assembled. These have enabled model testing at each stage of development. Areas of further work are suggested.

The simian immunodeficiency virus, SIVmac, causes disease affecting multiple organ systems in macaques similar to human immunodeficiency virus infection in humans. Molecularly cloned SIVmac with a strong lymphocyte tropism was used in pathogenesis experiments to correlate viral cell tropism with disease. In 5 animals, exhaustive analyses on viruses from tissues and identification of infected precursor cells were done at multiple times during infection to ensure the virus had not mutated into a macrophage-tropic variant. Viral replication was measured by infectivity, infectious center assays, and in situ hybridization. Lymphocytes produced most virus in tissues, indicating the virus maintained its cell tropism in vivo. Lymphocytes in bone marrow were latently infected and those in the spleen and lymph nodes were productively infected. The virus failed to replicate in the brain after intracerebral inoculation. SIVmac that maintained a strong tropism for lymphocytes and a corresponding poor tropism for macrophages can cause persistent infection and AIDS but not other diseases such as primary pneumonia and encephalitis in rhesus macaques.

The overall objective of this contract is to improve the predictive capabilities of MILHY2 whilst retaining its parsimonious data requirements, portability and simplicity of application. This objective has been achieved through the investigation of the following specific objectives: to develop the modular structure of MILHY to allow not only flexibility in catchment representation but also choice in the process resolution of the solution; to develop the channel routing components of MILHY2 to improve prediction of out- of-bank flood events; to investigate and implement several alternative validation schemes for the revised MILHY scheme, using both field data and hydrodynamic simulation of the River Fulda catchment in West Germany; to explore the suitability of a finite-element hydrodynamic model (RMA-2V) for large scale floodplain modelling; and to investigate the linking of MILHY with a hydrodynamic model (RMA-2V) to provide enhanced inundation modelling within a catchment scheme.

Foscarnet was administered to eight AIDS patients for suspected cytomegalovirus (CMV) pneumonitis as a continuous intravenous infusion for a minimum of 8 days. All the patients improved, three showing complete resolution of symptoms. Evidence of CMV infection from bronchoalveolar lavage samples was lacking in two patients. Adverse drug experiences consisted of thrombophlebitis, transient decreases in haemoglobin concentration, and reversible rises in serum creatinine levels.
