1. Pathogenic REST variant causing Jones syndrome and a review of the literature

Abstract

Jones syndrome is a rare dominantly inherited syndrome characterized by gingival fibromatosis and progressive sensorineural hearing loss becoming symptomatic in the second decade of life. Here, we report a father and his two daughters presenting with a typical Jones syndrome (OMIM %135550) phenotype. Exome sequencing identified a repressor element 1-silencing transcription factor (REST, OMIM *600571) (NM_005612.5) c.2670_2673del p.(Glu891Profs*6) heterozygous variant segregating with Jones syndrome in the family. We review the clinical data from all previously published patients with Jones syndrome and previously published patients with pathogenic REST variants associated with gingival fibromatosis or sensorineural hearing loss. This study suggests that pathogenic REST variants cause Jones syndrome.

Abstract

HIDEA syndrome is caused by biallelic pathogenic variants in P4HTM. The phenotype is characterized by muscular and central hypotonia, hypoventilation including obstructive and central sleep apneas, intellectual disability, dysautonomia, epilepsy, eye abnormalities, and an increased tendency to develop respiratory distress during pneumonia. Here, we report six new patients with HIDEA syndrome caused by five different biallelic P4HTM variants, including three novel variants. We describe two Finnish enriched pathogenic P4HTM variants and demonstrate that these variants are embedded within founder haplotypes. We review the clinical data from all previously published patients with HIDEA and characterize all reported P4HTM pathogenic variants associated with HIDEA in silico. All known pathogenic variants in P4HTM result in either premature stop codons, an intragenic deletion, or amino acid changes that impact the active site or the overall stability of P4H-TM protein. In all cases, normal P4H-TM enzyme function is expected to be lost or severely decreased. This report expands knowledge of the genotypic and phenotypic spectrum of the disease.

3. Comportamiento meiótico y descripción morfológica del polen de pronto alivio

Abstract

Este estudio fue realizado en la Universidad Nacional de Colombia y el Centro Internacional de Agricultura Tropical CIAT, para evaluar la meiosis se utilizaron las metodologías adaptadas por Dempsey (1993). Se observaron irregularidades tales como ascensión precoz de cromosomas, termi­nación tardía de quiasmas, adherencia cromosómica, formación de tétradas con microcitos y poliadas que llevaron a desbalances cromosómicos y a la producción de gametos inviables. Se verificó relativa estabilidad meiótica, medida a través del porcentaje de tétradas de microsporas viables o índice meiótico (IM). Aunque el IM fue de 77.3 %, la viabilidad polínica fue 11.4%, lo que sugiere que pudieron actuar genes mutantes en la post-meiosis. El tamaño del polen fue mediano, con 28.0 μm de diámetro ecuatorial y 26.9 μm de eje polar en vista ecuatorial, oblato esferoidal, tipo de área polar, grande y abertura corta, triaperturado, tricolporado, y zonoaperturado, perforado y tectado (microscopia de luz y electrónica de barrido MEB). Ensayos preliminares mostraron baja viabilidad y germinación de semillas. Palabras claves: Lippia alba, Verbenaceae, meiosis, viabilidad polínica, morfología polínica.

Abstract

Background Laparoscopic sleeve gastrectomy (LSG) is a well established treatment for severe obesity and type 2 diabetes. Although the gut microbiota is linked to the efficacy of LSG, the underlying mechanisms remain elusive. The effect of LSG for morbid obesity on the gut microbiota and bile acids was assessed here. Methods Severely obese subjects who were candidates for LSG were included and followed until 6 months after surgery. The composition and abundance of the microbiota and bile acids in faeces were assessed by 16S ribosomal RNA sequencing, quantitative PCR and liquid chromatography–mass spectrometry. Results In total, 28 patients with a mean(s.d.) BMI of 44·2(6·6) kg/m2 were enrolled. These patients had achieved excess weight loss of 53·2(19·0) per cent and showed improvement in metabolic diseases by 6 months after LSG, accompanied by an alteration in the faecal microbial community. The increase in α-diversity and abundance of specific taxa, such as Rikenellaceae and Christensenellaceae, was strongly associated with reduced faecal bile acid levels. These changes had a significant positive association with excess weight loss and metabolic alterations. However, the total number of faecal bacteria was lower in patients before (mean(s.d.) 10·26(0·36) log10 cells per g faeces) and after (10·39(0·29) log10 cells per g faeces) operation than in healthy subjects (10·83(0·27) log10 cells per g faeces). Conclusion LSG is associated with a reduction in faecal bile acids and greater abundance of specific bacterial taxa and α-diversity that may contribute to the metabolic changes.

Abstract

Oil palm agricultural biomass has a great potential for biofuel production due to its abundance and less pretreatment needed. In this study, bioethanol production from oil palm trunks sap was carried out by using yeast Saccharomyces cerevisiae. The suitability of the oil palm trunk sap for bioethanol production was tested by investigating the relationship between the oil palm trunk sap concentration and ethanol produced. The fermentation was conducted in static condition and without pH adjustment using 60, 80, and 100% oil palm trunk sap concentration. Results showed that the highest bioethanol production rate was achieved for 100% oil palm trunk sap (1.033 g/L.h), followed by 80% (0.744 g/L.h), and 60% (0.645 g/L.h). These results were influenced by the presence of sugars in the initial sap, suggesting that high substrate concentration does not inhibit bioethanol production. Therefore, the oil palm trunks sap showed significant potential as a renewable feedstock for the production of bioethanol without dilution.

Abstract

Nomenclatural type definitions are one of the most important concepts in biological nomenclature. Being physical objects that can be re-studied by other researchers, types permanently link taxonomy (an artificial agreement to classify biological diversity) with nomenclature (an artificial agreement to name biological diversity). Two proposals to amend the International Code of Nomenclature for algae, fungi, and plants (ICN), allowing DNA sequences alone (of any region and extent) to serve as types of taxon names for voucherless fungi (mainly putative taxa from environmental DNA sequences), have been submitted to be voted on at the 11th International Mycological Congress (Puerto Rico, July 2018). We consider various genetic processes affecting the distribution of alleles among taxa and find that alleles may not consistently and uniquely represent the species within which they are contained. Should the proposals be accepted, the meaning of nomenclatural types would change in a fundamental way from physical objects as sources of data to the data themselves. Such changes are conducive to irreproducible science, the potential typification on artefactual data, and massive creation of names with low information content, ultimately causing nomenclatural instability and unnecessary work for future researchers that would stall future explorations of fungal diversity. We conclude that the acceptance of DNA sequences alone as types of names of taxa, under the terms used in the current proposals, is unnecessary and would not solve the problem of naming putative taxa known only from DNA sequences in a scientifically defensible way. Abstract

Gut microbiota affects the host’s metabolism, and it is suggested that there are differences in gut microbiota composition between patients with type 2 diabetes and healthy individuals. Additionally, dysbiosis may increase the concentration of lipopolysaccharides (LPS), causing metabolic endotoxemia, which induces impaired glucose tolerance. Several studies have reported relationships between metabolic diseases and the gut microbiota; and prebiotics, such as oligosaccharides, are commonly consumed to regulate gut microbiotas in healthy individuals. Galacto-oligosaccharides (GOS) are a major prebiotic, which specifically increase Bifidobacteriaceae abundance. Recent studies have reported that Bifidobacteriaceae improved metabolic endotoxemia or impaired glucose tolerance. However, there are few studies reporting the effects of GOS on patients with type 2 diabetes. In the current study, we compared clinical parameters, faecal gut microbiota, their associated metabolic products and their components such as LPS, and LPS-binding protein (LBP) produced by the host, between patients with diabetes and healthy controls. We then assessed the effects of GOS on glycaemic control, and gut microbiotas and metabolites in patients with type 2 diabetes in a double-blind controlled manner. LBP levels were significantly higher in patients with diabetes than those of healthy subjects, which was consistent with previous reports. The abundance of Bifidobacteriaceae and the diversity of intestinal microbiota were significantly lower in patients with diabetes than in healthy subjects. Interestingly, Bifidobacteriaceae was markedly restored in patients with diabetes after consumption of GOS, whereas LBP and glucose tolerance did not improve during this short-term trial period. In the present study, we demonstrated that GOS can ameliorate dysbiosis in patients with diabetes, and continuous intake of GOS may be a promising method for managing type 2 diabetes.

Abstract

Chitin is one of the most abundant biopolymers widely distributed in the marine and terrestrial environments. Chitinase enzyme has received increased attention due to its wide range of biotechnological applications. Aspergillus terrus was found to be a good chitinase producer among the five fungi isolated from different soil samples from Al-Jouf city, Saudi Arabia. Maximum production of chitinase was obtained when using 2% of Shrimp-shell powder as a sole carbon source in the fermentation medium. The high level of chitinase production was observed in the culture medium with pH 5 at 30°C for five days at shaking conditions. Some properties of the crude chitinase were studied. In the present study, the antifungal activity of crude A. terrus chitinase was investigated against Apergillus niger, Aspergillus oryzae, Pencillum oxysporium, Rhizoctonia solani, Fusarium oxysporium, Rhizopus sp. and Mucor sp. and also estimated. The chitinase was found to inhibit the growth of some phytopathogenic fungi tested. The present work provides a suitable medium composition for enhancement of chitinase production by A. terrus and some properties of crude enzyme. It occurs during transition from a high pressure environment to one of lower pressure, resulting in a range of conditions from itching to joint pain, convulsions, and death. A carcass of a stranded Sei Whale, Balaenoptera borealis, found on the shores of southern coast of Peninsular Malaysia was presented for postmortem. Investigation results showed that the male Sei Whale, named Si Corner showed pathologic lesions as seen from histology with the formation of fibrosis, emphysema and edema in the lungs and hepatic atrophy which indicated chronic starvation. It believed that he also suffering from “Barotrauma or decompression sickness” which affected the ear or lungs which lead to unbalanced movement due to changes in air pressure. This may have caused the whale to beach and consequent mortality. The pathologic lesions found give an insight into possible causes of death of beached whales in Malaysia.

Abstract

We performed measurements of carrier-phase-based two-way satellite time and frequency transfer (TWST-FT) with an A/D sampler and conventional TWSTFT system. We found that an instability resulting from a local signal at the satellite transponder was negligible. The short-term stability of 4 × 10(-13) at 1 s was achieved in a short-baseline measurement. The results showed good agreement with the GPS carrier phase.

Abstract

Reflected signals of the Global Navigation Satellite System (GNSS) from the sea or land surface can be utilized to deduce and monitor physical and geophysical parameters of the reflecting area. Unlike most other remote sensing techniques, GNSS-Reflectometry (GNSS-R) operates as a passive radar that takes advantage from the increasing number of navigation satellites that broadcast their L-band signals. Thereby, most of the GNSS-R receiver architectures are based on dedicated hardware solutions. Software-defined radio (SDR) technology has advanced in the recent years and enabled signal processing in real-time, which makes it an ideal candidate for the realization of a flexible GNSS-R system. Additionally, modern commodity graphic cards, which offer massive parallel computing performances, allow to handle the whole signal processing chain without interfering with the PC’s CPU. Thus, this paper describes a GNSS-R system which has been developed on the principles of software-defined radio supported by General Purpose Graphics Processing Units (GPGPUs), and presents results from initial field tests which confirm the anticipated capability of the system.

Abstract

Two-way satellite time and frequency transfer (TWSTFT) is one of the main techniques used to compare atomic time scales over long distances. To both improve the precision of TWSTFT and decrease the satellite link fee, a new software-defined modem with dual pseudo-random noise (DPN) codes has been developed. In this paper, we demonstrate the first international DPN-based TWSTFT experiment over a period of 6 months. The results of DPN exhibit excellent performance, which is competitive with the Global Positioning System (GPS) precise point positioning (PPP) technique in the short-term and consistent with the conventional TWSTFT in the long-term. Time deviations of less than 75 ps are achieved for averaging times from 1 s to 1 d. Moreover, the DPN data has less diurnal variation than that of the conventional TWSTFT. Because the DPN-based system has advantages of higher precision and lower bandwidth cost, it is one of the most promising methods to improve international time-transfer links.

Abstract

Delays in signal transmission and reception paths of an Earth station must be calibrated accurately, and their variation should be reduced in the two-way satellite time and frequency transfer (TWSTFT). We have developed a portable TWSTFT station using a modem built by the National Institute of Information and Communications Technology (NICT) and performed calibration between NICT and Telecommunication Laboratories. An uncertainty of about 1 ns in the differential calibration was achieved. In addition, we have monitored the delay variation in the Earth station using a measurement system installed at NICT to improve accuracy of the TWSTFT. The contributed instability was 50 ps/day

Abstract

In this paper we report that carrier-phase two-way satellite time and frequency transfer (TWSTFT) was successfully demonstrated over a very long baseline of 9,000 km, established between the National Institute of Information and Communications Technology (NICT) and the Physikalisch-Technische Bundesanstalt (PTB). We verified that the carrier-phase TWSTFT (TWCP) result agreed with those obtained by conventional TWSTFT and GPS carrier-phase (GPSCP) techniques. Moreover, a much improved short-term instability for frequency transfer of $2\times10^{-13}$ at 1 s was achieved, which is at the same level as previously confirmed over a shorter baseline within Japan. The precision achieved was so high that the effects of ionospheric delay became significant which are ignored in conventional TWSTFT even over a long link. We compensated for these effects using ionospheric delays computed from regional vertical total electron content maps. The agreement between the TWCP and GPSCP results was improved because of this compensation.

Abstract

In automotive application new point-to-point (P2P) wideband communication systems e.g. Automotive Ethernet 100BASE-T1 imply an increasing effort in safeguarding its electromagnetic compatibility (EMC). The state-of-the-art EMC safeguarding process proposes a pre-compliant system level test before the vehicle level test, which may have significant variation in test results due to the different electromagnetic environment. This work presents an approach to calculate the emission signal at the vehicle antenna from the ECU output signal given by the power spectral density (PSD) of the Automotive Ethernet ECUs. The determination is performed via a transfer function which describes the coupling between the Ethernet wire harness and the antenna. This approach considers the modelling of the complex vehicle environment by the transfer function which promises the determination of an accurate emission signal before performing the vehicle level test. Further, the transfer function for a specifically EMC critical cable harness routing along the cockpit area is analyzed in more detail with a 3D simulation, which validates the chosen measurement technique of the transfer function.

Abstract

A new method of frequency transfer service using a color subcarrier from a geostationary broadcasting satellite is proposed, in which the velocity vector of the satellite is determined by measuring the Doppler shifts of color subcarriers from the satellite at four stations whose frequencies are synchronized. Using the determined velocity vector, a user can correct the Doppler shift at his location with simple calculations. An error analysis and an experiment have demonstrated that the frequency transfer error of this method is less than 1/spl times/10/sup -11/ for the averaging time of 1000 s.

Abstract

We have applied microspot photoemission spectroscopy to the surface imaging of a polycrystalline Cu plate with a lateral resolution of 0.3 μm and an energy resolution of 30 meV. The images of the crystalline islands with the (1 1 1) surface were measured by detecting photoelectrons due to the Shockley surface state (SS) and the sp-band specific to the (1 1 1) surface. The dependence of the images on the photoelectron energies was related to the mean length of the (1 1 1) terrace. The (1 1 1) island measured with the photoelectron close to the work function cutoff was found to be shifted from that measured with the Shockley state. The result demonstrates that the very high-energy resolution is essential for precise understanding of the surface images.

Abstract

Censos visuales realizados en cuatro estados sucesionales en la zona del río Tatabro, revelaron la presencia de 15 especies en bosque primario. 19 especies en bosque secundario, 14 en borde de bosque y 25 para carretera. La mayor diversidad de especies fue para bosque secundario. El inventario realizado en la zona, durante el transcurso del año 1995, muestra una gran diversidad de mariposas aún existentes allí. Este inventario muestra 211 especies, 107 géneros agrupados en 14 subfamilias y tres familias Hesperidae, Papilionidae y Castniidae.

Abstract

We have applied photoemission microspectroscopy to copper phthalocyanine film grown on a polycrystalline copper with a lateral resolution of 0.3μm and an energy resolution of 30meV. The photoemission band due to the highest occupied molecular orbital peaked at the binding energy of either 1.6 or 1.2eV depending on the sample positions, while the work functions were 4.3 and 4.5eV for the respective positions. The band was intense when the binding energy was low. The results demonstrate that a large inhomogeneity exists in the interface electronic structure.

A measurement of electron energy has been carried out using a RFEA technique. The averaged electron energies in a RF discharge plasma were evaluated to be 20 ∼ 35 eV under 2 ∼ 8 Pa for D2 gas and the RF power of 100 W. It became very clear that the electron energies were large enough to dissociate the hydrogen molecules.

Abstract

Tritium permeation, particularly plasma driven permeation (PDP) through plasma facing materials has been identified as a critical safety issue for future fusion power devices. While PDP through metals has been reported earlier, this work presents the first measurements of permeation rates through SiC/SiC composite samples exposed to a modified RF-discharge plasma device. The response of the SiC/SiC composite samples to impinging plasma radiation was also investigated using SEM. In contrast to metals, SiC/SiC composite showed no evidence of PDP of deuterium. The SEMs revealed attack only of directly exposed fibers and fiber-matrix interfaces. In lieu of these measurements, SiC/SiC composites offer promising characteristics as plasma facing materials for future fusion devices.

National Institute of Information and Communications Technology, Japan (NICT) developed the time management system of the first Quasi-Zenith Satellite (QZS-1), which consisted of on-board time transfer subsystem (TTS) and related ground systems such as time management stations (TMS). TTS can measure the time differences between an on-board Rb clock and an atomic clock of the TMS. TTS can also measure the time differences between the on-board Rb clocks and the satellite reference clock, moreover between the L-band signals and the satellite reference clock. This paper describes about the time management system and shows results of a time comparison experiment between the QZS-1 and the ground TMSs using TTS.

Abstract

We have developed a new two-way time transfer modem to improve the time transfer precision of remote clock comparison. As a timing signal, we apply a binary offset carrier, which is similar to those signals used for the next-generation Global Navigation Satellite Systems. We took advantage of versatile A/D and D/A converters, and most of the digital signal processing stages were realized by software, running on an off-the-shelf PC. This enabled us to realize the complete system with cheaper equipment, leading to an affordable low-cost modem. For the real-time digital signal processing stages implemented in software, we relied on a graphics processing unit (GPU) developed for computer game enthusiast. The developed modem can receive four channels at the same time with a single GPU card. We performed two-way satellite time transfer experiments using these modems between Japan and Taiwan. The obtained results are consistent within 200 ps with respect to the results of GPS carrier phase time transfer. As a consequence, we improved the time transfer precision by nearly one order of magnitude as compared to a conventional two-way modem without increasing the connection fees caused by commercial communication satellites.

PDP is a very interesting phenomenon and presents a significant problem to the fuel control and the safety in Tokamak devices. Systematic experimental studies on the PDP mechanism using an ECR discharge plasma have been carried out to investigate the role of the neutrals and to estimate the isotope effects occurred in PDP for H, d, and T. The maximum isotope effects observed are 1.42 for H/D, and 1.1 for D/T mixed plasma in the case of SUS 304 test membrane. In this paper, the magnitudes of the isotope effects between the three hydrogen isotopes are satisfactorily explained by the role of neutrals in PDP.

Backgrounds: There is a high prevalence of obesity and hyperinsulinism among Chilean prepuberal children. Aim: To evaluate insulin sensitivity (IS) using fasting insulin, the Homeostasis Model Assessment (HOMA) and quantitative insulin-sensitivity check index (QUICKI) in Chilean children. Material and Methods: Body mass index (BMI), total body fat percentage (%TBF) using the sum of 4 skin folds, abdominal obesity determined through waist circumference (WC), pubertal maturation using five Tanner stages, fasting glucose (Glu) and insulin (Ins), were measured in 354 children aged 6 to 15 years (173 males). Abstract

Protein kinase C (PKC) is an important molecule involved in various cell function, and mediates induced secretion of vascular endothelial growth factor (VEGF). It is hypothesized that PKC and VEGF may be associated with tooth development. Using the laser microdissection method and real-time reverse-transcription-polymerase chain-reaction (RT-PCR), we investigated the expression of PKC betaI and betaII, VEGF, and amelogenin (used as a marker of differentiation to ameloblasts) in the inner and outer enamel epithelia, stellate reticulum, and dental papilla in each stage of the dental germ. We found that the expression levels of PKC betaI and betaII were increased in the inner enamel epithelium during the early bell stage. In addition, the increased expression levels of PKC betaI and betaII were accompanied by increased VEGF expression. These results indicate that PKC betaI, betaII, and VEGF are closely associated with the differentiation of the inner enamel epithelium to ameloblasts.

Abstract

This paper discuses how to design the capacity of address cache tables for large-scale computer communication networks. We show that destination addresses of packets can, be assumed to be characterized by two types of Zipf's law. Based on the complementary use of these laws, we derive the relation between the number of accesses and the number of destination addresses. Experimental results show that the relation gives a good approximation. Applying this relation, we derive the upper/lower bounds for cache hit probability. Using the probabilities, design issues including the capacity of the cache table and aging algorithm of cache entries are also discussed.

Abstract

Encephalocel is leff frequent than spina bifida and anencephalia. Its incidence is 0.25@1000 births. It concern as much female than male. This neural defect is frequency associated to others malformations face (20%) squelette (17%), kidney (13%). In our study the ultra sound is performant in 82% of the cases. Encephalocel is still a lethal and severe abnormality.
