Abstract

The RFX-mod2 device, the upgraded version of the previous RFX-mod with a modified magnetic boundary, is presently under realization and will start to be operated in 2025. Significant upgrades of the diagnostic capabilities have been proposed and are under development. These include a largely increased number of in-vessel magnetic and electrostatic sensors, a new fast reciprocating manipulator for the exploration of the edge plasma in a wide range of experimental conditions, the improved Thomson scattering and soft x-ray diagnostics system for a detailed determination of the behavior of the electron temperature profile, new dedicated systems for the space and time resolved analysis of x-ray spectra and neutron rate, a reflectometric diagnostic for real-time determination of plasma position, two diagnostics devoted to the imaging of light impurities and influxes behavior along with arrays of halo current sensors. These diagnostic upgrades will be accompanied by a significant effort to improve the control of the electron density and of the impurity influxes by means of proper treatment of plasma facing components with in-vessel fixed electrodes distributed over the first wall. The described advancements will allow a deeper understanding of physics phenomena in the wide variety of magnetic configurations, including the tokamak, the reversed-field pinch and the Ultra-low q, which can be produced in RFX-mod2 thanks to its flexibility and unique MHD control capabilities.

3. Development of a new toolbox for mouse PET–CT brain image analysis fully based on CT images and validation in a PD mouse model

Abstract

Abstract Automatic analysis toolboxes are popular in brain image analysis, both in clinical and in preclinical practices. In this regard, we proposed a new toolbox for mouse PET–CT brain image analysis including a new Statistical Parametric Mapping-based template and a pipeline for image registration of PET–CT images based on CT images. The new templates is compatible with the common coordinate framework (CCFv3) of the Allen Reference Atlas (ARA) while the CT based registration step allows to facilitate the analysis of mouse PET–CT brain images. From the ARA template, we identified 27 volumes of interest that are relevant for in vivo imaging studies and provided binary atlas to describe them. We acquired 20 C57BL/6 mice with [18F]FDG PET–CT, and 12 of them underwent 3D T2-weighted high-resolution MR scans. All images were elastically registered to the ARA atlas and then averaged. High-resolution MR images were used to validate a CT-based registration pipeline. The resulting method was applied to a mouse model of Parkinson’s disease subjected to a test–retest study (n = 6) with the TSPO-specific radioligand [18F]VC701. The identification of regions of microglia/macrophage activation was performed in comparison to the Ma and Mirrione template. The new toolbox identified 11 (6 after false discovery rate adjustment, FDR) brain sub-areas of significant [18F]VC701 uptake increase versus the 4 (3 after FDR) macro-regions identified by the Ma and Mirrione template. Moreover, these 11 areas are functionally connected as found by applying the Mouse Connectivity tool of ARA. In conclusion, we developed a mouse brain atlas tool optimized for PET–CT imaging analysis that does not require MR. This tool conforms to the CCFv3 of ARA and could be applied to the analysis of mouse brain disease models.

Abstract

Introduction The current standard of care for glioblastoma (GBM) is surgical resection, followed by adjuvant radiotherapy. Complete resection remains challenging because of unclearly defined tumor margins. Presurgery tumor segmentation is generally done by a neurosurgeon on the contrast-enhanced T1 (T1c) scan. A semi-automated tumor segmentation can ease this process, exploiting also information contained in other scans besides T1c. The aim of this study is to propose a settlement to qualitatively assess segmentations on a multicentric GBM cohort. The Brats-Toolkit [1] tool was considered. Methods Preoperative MRI studies of 72 GBM patients (41 males, 31 females, mean age 63 yr.) surgically treated at Fondazione IRCCS San Gerardo were considered. Images were acquired with heterogeneous protocols, all containing T1, T1c, T2 and T2 FLAIR scans. T1c images were manually segmented by a neurosurgeon, as to clinical practice (unique Region of Interest containing core + necrosis + core enhanced). BraTSPreprocessor, BraTS-Segmentor (8 Docker images from the repository), and BraTS-Fusionator (SIMPLE method) were applied, providing as output core + necrosis, core-enhanced and edema contours [2], [3]. Manual and BraTS segmentations were visually evaluated by a senior neuroradiologist. Results/Discussion MRI studies were classified in 6 groups based on scans characteristics (Group1: 3T - isotropic T1, T1c, FLAIR - anisotropic T2; Group2: 1.5T - isotropic T1, T1c, FLAIR - anisotropic T2; Group3: 3T - isotropic T1c, FLAIR - anisotropic T1, T2; Group4 1.5T - isotropic T1c, FLAIR - anisotropic T1, T2; Group5: 1.5T - isotropic T1, T1c - anisotropic FLAIR, T2; Group6: 1.5T - isotropic T1c - anisotropic T1, FLAIR, T2). Abstract

Cosmological constraints from key probes of the Euclid imaging survey rely critically on the accurate determination of the true redshift distributions, $n(z)$, of tomographic redshift bins. We determine whether the mean redshift, $$, of ten Euclid tomographic redshift bins can be calibrated to the Euclid target uncertainties of $\sigma()$ is measured in each tomographic redshift bin to an accuracy of order 0.01 or better. By measuring the clustering redshifts on subsets of the full Flagship area, we construct scaling relations that allow us to extrapolate the method performance to larger sky areas than are currently available in the mock. For the full expected Euclid, BOSS, and DESI overlap region of approximately 6000 deg$^{2}$, the uncertainties attainable by clustering redshifts exceeds the Euclid requirement by at least a factor of three for both $n(z)$ models considered, although systematic biases limit the accuracy. Clustering redshifts are an extremely effective method for redshift calibration for Euclid if the sources of systematic biases can be determined and removed, or calibrated-out with sufficiently realistic simulations. Abstract

Background: The foreseeable clinical availability in Europe of disease-modifying drugs effective in early phases of some neurodegenerative diseases (such as Alzheimer’s disease), shifts the focus to the etiological diagnosis of pre-dementia stages, such as Mild Cognitive Impairment (MCI) and Subjective Cognitive Decline (SCD). Current diagnostic approaches employ costly and limited methods such as PET-FDG. Pseudo-continuous Arterial Spin Labeling (pCASL) is emerging as a promising non-invasive alternative, offering rapid, radiation-free brain perfusion measurements through MRI. Recent research demonstrates pCASL's comparable performance to PET-FDG in neurodegenerative dementias, revealing aligned affected brain regions. However, pCASL's suitability for diagnosing MCI and SCD remains unexplored. The CAPE study (NCT05756270) aims to evaluate pCASL's diagnostic potential in MCI and SCD during initial assessments, by correlating cerebral blood flow patterns from pCASL and PET-FDG and comparing their accuracy. Methods: CAPE primary objective will be to compare brain cerebral blood flow and glucose metabolism patterns among MCI and SCD subjects both in general and across ATN classifications of CSF biomarkers. Secondary objectives will encompass evaluating pCASL's potential in distinguishing various subgroups of SCD and MCI, examining correlations between pCASL/PET-FDG and neuropsychological tests, predicting conversion to dementia using pCASL and PET-FDG, and exploring associations between pCASL and diverse fluid biomarkers. The study will recruit up to 150 MCI and SCD subjects, with CDR ≤ 0.5 and age ≤ 80. The research will span 36 months (24-month recruitment phase, and partially overlapping 24-month follow-up). Participants will undergo baseline brain MRI, PET-FDG, neuropsychological tests, lumbar puncture, and blood tests. 9. A combined VANDELS and LEGA-C study: the evolution of quiescent galaxy size, stellar mass, and age from z = 0.6 to z = 1.3

Abstract

We study the relationships between stellar mass, size, and age within the quiescent population, using two mass-complete spectroscopic samples with log10(M⋆/M⊙) > 10.3, taken from VANDELS at 1.0 < z < 1.3, and LEGA-C at 0.6 < z < 0.8. Using robust Dn4000 values, we demonstrate that the well-known ‘downsizing’ signature is already in place by z ≃ 1.1, with Dn4000 increasing by ≃ 0.1 across a ≃ 1 dex mass interval for both VANDELS and LEGA-C. We then proceed to investigate the evolution of the quiescent galaxy stellar mass–size relation from z ≃ 1.1 to z ≃ 0.7. We find the median size increases by a factor of 1.9 ± 0.1 at log10(M⋆/M⊙) = 10.5, and see tentative evidence for flattening of the relation, finding slopes of α = 0.72 ± 0.06 and α=0.56±0.04 for VANDELS and LEGA-C, respectively. We finally split our sample into galaxies above and below our fitted mass–size relations, to investigate how size and Dn4000 correlate. For LEGA-C, we see a clear difference, with larger galaxies found to have smaller Dn4000 at fixed stellar mass. Due to the faintness and smaller numbers of the VANDELS sample, we cannot confirm whether a similar relation exists at z ≃ 1.1. We consider whether differences in stellar age or metallicity are most likely to drive this size–Dn4000 relation, finding that any metallicity differences are unlikely to fully explain the observed offset, meaning smaller galaxies must be older than their larger counterparts. We find the observed evolution in size, mass, and Dn4000 across the ≃ 2 Gyr from z ∼ 1.1 to z ∼ 0.7 can be explained by a simple toy model in which VANDELS galaxies evolve passively whilst experiencing a series of minor mergers.

Loureiro et al., We present a tomographic weak lensing analysis of the Kilo Degree Survey Data Release 4 (KiDS-1000), using a new pseudo angular power spectrum estimator (pseudo-Cℓ) under development for the ESA Euclid mission. Over 21 million galaxies with shape information are divided into five tomographic redshift bins, ranging from 0.1 to 1.2 in photometric redshift. Abstract

The Euclid space telescope will survey a large dataset of cosmic voids traced by dense samples of galaxies. In this work we estimate its expected performance when exploiting angular photometric void clustering, galaxy weak lensing and their cross-correlation. To this aim, we implement a Fisher matrix approach tailored for voids from the Euclid photometric dataset and present the first forecasts on cosmological parameters that include the void-lensing correlation. We examine two different probe settings, pessimistic and optimistic, both for void clustering and galaxy lensing. We carry out forecast analyses in four model cosmologies, accounting for a varying total neutrino mass, $M_\nu$, and a dynamical dark energy (DE) equation of state, $w(z)$, described by the CPL parametrisation. We find that void clustering constraints on $h$ and $\Omega_b$ are competitive with galaxy lensing alone, while errors on $n_s$ decrease thanks to the orthogonality of the two probes in the 2D-projected parameter space. We also note that, as a whole, the inclusion of the void-lensing cross-correlation signal improves parameter constraints by $10-15\%$, and enhances the joint void clustering and galaxy lensing Figure of Merit (FoM) by $10\%$ and $25\%$, in the pessimistic and optimistic scenarios, respectively. Finally, when further combining with the spectroscopic galaxy clustering, assumed as an independent probe, we find that, in the most competitive case, the FoM increases by a factor of 4 with respect to the combination of weak lensing and spectroscopic galaxy clustering taken as independent probes. The forecasts presented in this work show that photometric void-clustering and its cross-correlation with galaxy lensing deserve to be exploited in the data analysis of the Euclid galaxy survey and promise to improve its constraining power, especially on $h$, $\Omega_b$, the neutrino mass, and the DE evolution., Comment: 22 pages, 9 figures - published in A&A

Abstract

The greatest diversity of anurans in the world is in Brazil and one of the major challenges is to reconcile the accelerated economic development with strategies that aim to maintain this diversity in forest fragments, often representing ESUs of some biomes. This study aimed to obtain data that will support conservation projects through the pioneering use of ISSR analysis in Neotropical anurans, estimating the intra- and interpopulation genetic diversity of four populations of P. cuvieri (Paraná and São Paulo regions). Of the 65 loci scored 58 were polymorphic, with 0.797 intrapopulation variation and 0.203 interpopulation variation. The index of interpopulation genetic differentiation (Fst) proved to be high among the population of Marmeleiro-PR and the three populations of SP (Fst > 0.288); genetic dissimilarity was related to the geographical distance. The ISSR proved to be efficient and useful molecular markers in comparison with other markers most widely used for preliminary diagnosis of genetic diversity in populations of amphibians, and could be applied as a tool for future conservation projects, since they could identify potential ESUs and influence decisions on the preservation of fragments.

Abstract

Euclid Consortium: M. S. Cagliari et al., [Context] The ESA Euclid mission will produce photometric galaxy samples over 15 000 square degrees of the sky that will be rich for clustering and weak lensing statistics. The accuracy of the cosmological constraints derived from these measurements will depend on the knowledge of the underlying redshift distributions based on photometric redshift calibrations., [Aims] A new approach is proposed to use the stacked spectra from Euclid slitless spectroscopy to augment broad-band photometric information to constrain the redshift distribution with spectral energy distribution fitting. The high spectral resolution available in the stacked spectra complements the photometry and helps to break the colour-redshift degeneracy and constrain the redshift distribution of galaxy samples., [Methods] We modelled the stacked spectra as a linear mixture of spectral templates. The mixture may be inverted to infer the underlying redshift distribution using constrained regression algorithms. We demonstrate the method on simulated Vera C. Rubin Observatory and Euclid mock survey data sets based on the Euclid Flagship mock galaxy catalogue. We assess the accuracy of the reconstruction by considering the inference of the baryon acoustic scale from angular two-point correlation function measurements., [Results] We selected mock photometric galaxy samples at redshift z > 1 using the self-organising map algorithm. Considering the idealised case without dust attenuation, we find that the redshift distributions of these samples can be recovered with 0.5% accuracy on the baryon acoustic scale. The estimates are not significantly degraded by the spectroscopic measurement noise due to the large sample size. However, the error degrades to 2% when the dust attenuation model is left free. An observational detection of a violation of these assumptions at any redshift would immediately indicate the presence of new physics., [Aims] We quantify the ability of the Euclid mission, together with contemporary surveys, to improve the current sensitivity of null tests of the canonical cosmological constant Λ and the cold dark matter (ΛCDM) model in the redshift range 0, [Methods] We considered both currently available data and simulated Euclid and external data products based on a ΛCDM fiducial model, an evolving dark energy model assuming the Chevallier-Polarski-Linder parameterization or an inhomogeneous Lemaître-Tolman-Bondi model with a cosmological constant Λ, and carried out two separate but complementary analyses: a machine learning reconstruction of the null tests based on genetic algorithms, and a theory-agnostic parametric approach based on Taylor expansion and binning of the data, in order to avoid assumptions about any particular model., [Results] We find that in combination with external probes, Euclid can improve current constraints on null tests of the ΛCDM by approximately a factor of three when using the machine learning approach and by a further factor of two in the case of the parametric approach. However, we also find that in certain cases, the parametric approach may be biased against or missing some features of models far from ΛCDM., [Conclusions] Our analysis highlights the importance of synergies between Euclid and other surveys. 15. Design of a new reflectometric system for real time plasma position control on the RFX-mod2 device

Abstract

RFX-mod2 (R = 2.0 m, a = 0.49 m), the upgraded version of the previous RFXmod fusion device, will be equipped with a new reflectometry system specifically designed for plasma position control purposes. Featuring a high temporal and spatial resolution and being suitable for harsh fusion reactor experimental conditions (long pulses, high neutron fluxes), reflectometry has been proposed as a good candidate for this task. On RFX-mod2 the diagnostic system will consist of four bistatic ultrafast independent reflectometric units working in the frequency range (16–26.5 GHz) and installed in four different poloidal locations at the same toroidal angle: two on the equatorial plane (high field side/low field side) and two at the vertical top/bottom ports. Standard pyramidal horns will be installed in the external midplane and in the vertical ports, while parabolic hoghorn reflectors have been designed for the internal midplane. Different technical solutions for the integration in the machine, as the additive manufacturing for the antenna production, are presented. Despite the differences with respect to the application on large Tokamaks like DEMO, the RFX-mod2 plasma position reflectometer can contribute to test on a simple machine some of the issues related to the development of a reflectometry based plasma position and shape control.

Abstract

Euclid Consortium: N. Hamaus et al., Euclid is poised to survey galaxies across a cosmological volume of unprecedented size, providing observations of more than a billion objects distributed over a third of the full sky. Approximately 20 million of these galaxies will have their spectroscopy available, allowing us to map the three-dimensional large-scale structure of the Universe in great detail. This paper investigates prospects for the detection of cosmic voids therein and the unique benefit they provide for cosmological studies. In particular, we study the imprints of dynamic (redshift-space) and geometric (Alcock–Paczynski) distortions of average void shapes and their constraining power on the growth of structure and cosmological distance ratios. To this end, we made use of the Flagship mock catalog, a state-of-the-art simulation of the data expected to be observed with Euclid. We arranged the data into four adjacent redshift bins, each of which contains about 11 000 voids and we estimated the stacked void-galaxy cross-correlation function in every bin. Fitting a linear-theory model to the data, we obtained constraints on f/b and DMH, where f is the linear growth rate of density fluctuations, b the galaxy bias, DM the comoving angular diameter distance, and H the Hubble rate. In addition, we marginalized over two nuisance parameters included in our model to account for unknown systematic effects in the analysis. With this approach, Euclid will be able to reach a relative precision of about 4% on measurements of f/b and 0.5% on DMH in each redshift bin. Better modeling or calibration of the nuisance parameters may further increase this precision to 1% and 0.4%, respectively. Our results show that the exploitation of cosmic voids in Euclid will provide competitive constraints on cosmology even as a stand-alone probe. Abstract

Pair-instability supernovae are theorized supernovae that have not yet been observationally confirmed. They are predicted to exist in low-metallicity environments. Because overall metallicity becomes lower at higher redshifts, deep near-infrared transient surveys probing high-redshift supernovae are suitable to discover pair-instability supernovae. The Euclid satellite, which is planned to be launched in 2023, has a near-infrared wide-field instrument that is suitable for a high-redshift supernova survey. The Euclid Deep Survey is planned to make regular observations of three Euclid Deep Fields (40 deg2 in total) spanning the Euclid's 6 year primary mission period. While the observations of the Euclid Deep Fields are not frequent, we show that the predicted long duration of pair-instability supernovae would allow us to search for high-redshift pair-instability supernovae with the Euclid Deep Survey. Based on the current observational plan of the Euclid mission, we conduct survey simulations in order to estimate the expected numbers of pair-instability supernova discoveries. We find that up to several hundred pair-instability supernovae at z < ~ 3.5 can be discovered within the Euclid Deep Survey. We also show that pair-instability supernova candidates can be efficiently identified by their duration and color that can be determined with the current Euclid Deep Survey plan. We conclude that the Euclid mission can lead to the first confirmation of pair-instability supernovae if their event rates are as high as those predicted by recent theoretical studies. Abstract

An accurate covariance matrix is essential for obtaining reliable cosmological results when using a Gaussian likelihood. In this paper we study the covariance of pseudo-$C_\ell$ estimates of tomographic cosmic shear power spectra. Using two existing publicly available codes in combination, we calculate the full covariance matrix, including mode-coupling contributions arising from both partial sky coverage and non-linear structure growth. For three different sky masks, we compare the theoretical covariance matrix to that estimated from publicly available N-body weak lensing simulations, finding good agreement. We find that as a more extreme sky cut is applied, a corresponding increase in both Gaussian off-diagonal covariance and non-Gaussian super-sample covariance is observed in both theory and simulations, in accordance with expectations. Studying the different contributions to the covariance in detail, we find that the Gaussian covariance dominates along the main diagonal and the closest off-diagonals, but further away from the main diagonal the super-sample covariance is dominant. Forming mock constraints in parameters describing matter clustering and dark energy, we find that neglecting non-Gaussian contributions to the covariance can lead to underestimating the true size of confidence regions by up to 70 per cent. The dominant non-Gaussian covariance component is the super-sample covariance, but neglecting the smaller connected non-Gaussian covariance can still lead to the underestimation of uncertainties by 10--20 per cent. A real cosmological analysis will require marginalisation over many nuisance parameters, which will decrease the relative importance of all cosmological contributions to the covariance, so these values should be taken as upper limits on the importance of each component., 15 pages, 8 figures; matches version accepted by A&A; code available at https://github.com/robinupham/shear_pcl_cov

Improving the photo-z quality is a major challenge but is needed to advance our understanding of cosmology. In this paper we explore how the synergies between narrow-band photometric data and large imaging surveys can be exploited to improve broadband photometric redshifts. We used a multi-task learning (MTL) network to improve broadband photo-z estimates by simultaneously predicting the broadband photo-z and the narrow-band photometry from the broadband photometry. The narrow-band photometry is only required in the training field, which also enables better photo-z predictions for the galaxies without narrow-band photometry in the wide field. This technique was tested with data from the Physics of the Accelerating Universe Survey (PAUS) in the COSMOS field. We find that the method predicts photo-zs that are 13% more precise down to magnitude iAB < 23; the outlier rate is also 40% lower when compared to the baseline network. Furthermore, MTL reduces the photo-z bias for high-redshift galaxies, improving the redshift distributions for tomographic bins with z > 1. Applying this technique to deeper samples is crucial for future surveys such as Euclid or LSST. For simulated data, training on a sample with iAB < 23, the method reduces the photo-z scatter by 16% for all galaxies with iAB < 25. Abstract

The partial element equivalent circuit (PEEC) method is applied to a frequency-sweep study for investigating the resonant modes of planar spiral resonators. A low-rank approximation algorithm, based on hierarchical matrices, is coupled to the method in order to reduce the computational effort due to the use of dense matrices. The partitioning of the matrices is carefully addressed in order to allow an efficient hierarchical off-diagonal low-rank structure of the system matrix.

Abstract

The ITER Neutral Beam Test Facility (NBTF), called PRIMA (Padova Research on ITER Megavolt Accelerator), is hosted in Padova, Italy and includes two experiments: MITICA, the full-scale prototype of the ITER heating neutral beam injector, and SPIDER, the full-size radio frequency negative-ions source. The NBTF realization and the exploitation of SPIDER and MITICA have been recognized as necessary to make the future operation of the ITER heating neutral beam injectors efficient and reliable, fundamental to the achievement of thermonuclear-relevant plasma parameters in ITER. This paper reports on design and R&D carried out to construct PRIMA, SPIDER and MITICA, and highlights the huge progress made in just a few years, from the signature of the agreement for the NBTF realization in 2011, up to now—when the buildings and relevant infrastructures have been completed, SPIDER is entering the integrated commissioning phase and the procurements of several MITICA components are at a well advanced stage.

Abstract

Simple Summary Glioblastoma is the most common and malignant primary brain tumor, with a median survival of around 14 months. The aggressiveness of glioblastoma is due to intense cell proliferation, angiogenesis, invasiveness, genetic instability, resistance to therapies and high frequency of relapses. These features render glioblastoma almost incurable, considered an extreme therapeutic challenge. In the last few decades, it has been observed a reduced cancer incidence in diabetic patients treated with metformin, an oral hypoglycemic drug. The reported ability of metformin to arrest cancer cell growth in in vitro and in vivo experimental tumor models, have suggested the possibility to reconsider metformin as an anti-cancer add-on therapy, but further investigations about molecular mechanisms and optimal therapeutic regimens are needed. Here, we tested the efficacy of metformin against primary glioblastoma endothelial cells, responsible for tumor angiogenesis, invasiveness and resistance to therapy, reporting promising results and advancing a novel target of metformin, the "sphingolipid rheostat". Background: Glioblastoma is the most aggressive primary brain malignancy in adults, with a poor prognosis of about 14 months. Recent evidence ascribed to metformin (MET), an antihyperglycemic drug, the potential to reduce cancer incidence and progression, but the molecular mechanisms underlying these effects need to be better investigated. Methods: Here, we tested the efficacy of MET on n = 10 primary glioblastoma endothelial cells (GECs), by viability and proliferation tests, as MTT and Live/Dead assays, apoptosis tests, as annexin V assay and caspase 3/7 activity, functional tests as tube-like structure formation and migration assay and by mRNA and protein expression performed by quantitative real-time PCR analysis (qRT-PCR) and Western Blot, respectively. Results: Data resulting revealed a time- and mu-dependent ability of MET to decrease cell viability and proliferation, increasing pro-apoptotic mechanisms mediated by caspases 3/7. Also, MET impacted GEC functionality with a significant decrease of angiogenesis and invasiveness potential. Mechanistically, MET was able to interfere with sphingolipid metabolism, weakening the oncopromoter signaling promoted by sphingosine-1-phosphate (S1P) and shifting the balance toward the production of the pro-apoptotic ceramide. Conclusions: These observations ascribed to MET the potential to serve as add-on therapy against glioblastoma, suggesting a repurposing of an old, totally safe and tolerable drug for novel oncology therapeutics.

Abstract

Brain energy metabolism actively regulates synaptic transmission and activity. We have previously shown that acute footshock (FS)-stress induces fast and long-lasting functional and morphological changes at excitatory synapses in prefrontal cortex (PFC). Here, we asked whether FS-stress increased energy metabolism in PFC, and modified related cognitive functions. Using positron emission tomography (PET), we found that FS-stress induced a redistribution of glucose metabolism in the brain, with relative decrease of [18F]FDG uptake in ventro-caudal regions and increase in dorso-rostral ones. Absolute [18F]FDG uptake was inversely correlated with serum corticosterone. Increased specific hexokinase activity was also measured in purified PFC synaptosomes (but not in total extract) of FS-stressed rats, which positively correlated with 2-Deoxy [3H] glucose uptake by synaptosomes. In line with increased synaptic energy demand, using an electron microscopy-based stereological approach, we found that acute stress induced a redistribution of mitochondria at excitatory synapses, together with an increase in their volume. The fast functional and metabolic activation of PFC induced by acute stress, was accompanied by rapid and sustained alterations of working memory performance in delayed response to T-maze test. Taken together, the present data suggest that acute stress increases energy consumption at PFC synaptic terminals and alters working memory.

Abstract

Heating neutral beam (HNB) injectors, necessary to achieve burning conditions and to control plasma instabilities in ITER, are characterized by such demanding parameters that a neutral beam test facility (NBTF) dedicated to their development and optimization is being realized in Padua (Italy) with direct contributions from the Italian government (through Consorzio RFX as the host entity) and the ITER international organization (with kind contributions from the ITER domestic agencies of Europe, Japan and India) and technical and scientific support from various European laboratories and universities. The NBTF hosts two experiments: SPIDER, devoted to ion source optimization for the required source performance, and MITICA, the full-size prototype of the ITER HNB, with an ion source identical to SPIDER. This paper gives an overview of the progress towards NBTF realization, with particular emphasis on issues discovered during this phase of activity and on solutions adopted to minimize the impact on the schedule and maintain the goals of the facilities. The realization of MITICA is well advanced; operation is expected to start in 2023 due to the long procurement time of the in-vessel mechanical components. The beam source power supplies, operating at 1 MV, are in an advanced phase of realization; all high-voltage components have been installed and the complex insulation test phase began in 2018. At the same time, construction and installation of SPIDER plant systems was successfully completed with their integration into the facility. The mechanical components of the SPIDER ion source were installed inside the vessel and connected to the plants. Integrated commissioning with the control, protection and safety systems ended positively and the first experimental phase has begun. The first results of the SPIDER experiment, with data from operational diagnostics, and the plans for the 1 MV insulation tests on the MITICA high-voltage components are presented.

Abstract

The first SiPM-based clinical SPECT ring for brain simultaneous imaging in standard MR scanners has been put into operation. Reliable operation of the instrument has been achieved by means of its optimization, such as the introduction of a dual-cooling system (liquid-gas) of the DAQ electronics. The static gantry is composed of 20 MRI-compatible γ-detection modules mounting 5 cm × 10 cm CsI(Tl) scintillators for a total of 1440 pixels (targeting a transaxial FoV of 20 cm). Here we present the results of a preliminary characterization showing 220 kcps max. count rate, 365cps/MBq sensitivity with the multislit-slat collimator, ~17% energy resolution and an average extrinsic spatial resolution of ~8 mm in the trans-axial plane.

Abstract

For the study of electrostatic and magnetic properties of filaments characterizing the edge region of the stellarator experiment W7-X, a specifically designed insertable probe head was constructed within the framework of EUROfusion WP.S1 work package in collaboration between Consorzio RFX, IPP Greifswald and Forschungszentrum Mich. The probe head, named High Resolution Probe (HRP), was conceived to be installed on the mid-plane multi-purpose fast reciprocating manipulator on W7-X. Electromagnetic filamentary turbulent structures are found to characterize the edge region of different magnetic configurations including Reversed Field Pinch, stellarator and tokamak, where strong currents are associated also to ELM filamentary structures. The study of those phenomena in W7-X stellarator is of particular interest as the electromagnetic features of filaments are expected to become more relevant with the increase of the local plasma beta. In particular the aim is to provide information about the presence and the features of parallel current density associated to filamentary turbulent or ELM-like structures. Furthermore, the possibility to measure the time evolution of the flow radial profiles using the Mach probe array was considered as a further interesting part of the study, given the strong interplay expected between the turbulent fluctuations and the average flows. Further important information provided is the radial propagation of turbulent flux. The contribution will present the design development, the R&D studies and the applied solutions for the sensors embedded in the probe head. In particular, the presence of 140 GHz ECRH plasma environment represents one of the main challenges for reliable magnetic fluctuation measurements. First measurements were performed during the W7-X experimental campaign OP1.2b, where electromagnetic features of turbulence were measured with the probe, at different magnetic configurations.

Abstract

Introduction Postoperative delirium is a disabling mental disorder in patients undergoing cardiac surgery. Post-cardiac surgery delirium is both a common and costly complication in the cardiac surgical ICU. Depending on the methodology, estimates of incidence have ranged from 3 to 70%. The potential consequences of delirium after cardiac surgery include increased morbidity, decreased functional status, cognitive decline and increased long-term mortality. Despite the impact of this condition, few studies exist in literature that help to identify risk factors associated with this condition in the specific setting of cardiac surgery. In this pilot, non-pharmacological, observational study we assessed the risk associated with the development of delirium in a cohort of patients who underwent cardiac surgery Methods We prospectively included 145 patients: women 42(28,9%) and 103men(71%) who underwent cardiac surgery between April and June 2018 at the cardiothoracic surgery department of the Tor Vergata University Hospital in Rome, Italy. Data about pre-intra and post-operatory period were collected. The presence of delirium was diagnosed if the CAM–ICU scale was positive when evaluated within the third post-operatory day. Univariate logistic regression was used to evaluate perioperative risk factors associated with delirium. Odds Ratio was used to estimate the importance of each factor in the genesis of delirium. Multivariate logistic regression was applied to the positive variables to assess the independent risk factors. A value p Results Among the 145 patients evaluated, 13 developed symptoms of delirium (5,09%). We found that the risk of Delirium was associated with pre-operatory EuroSCORE II (p = 0,028) and history of previous cardiac surgery (p = 0,042). Moreover, in the intra-operatory period the risk of Delirium was associated with red blood cell transfusion, intervention for Aortic Dissection (p = 0,013), HCA with ACP (p = 0,036). In the post-operatory period risk of Delirium was associated with levels of creatinine clearance (p = 0,035) and CRP (p = 0,029). Discussion Delirium is relatively frequent in the cardiac surgical ICU. High EuroScore correlates with the onset of delirium, suggesting that delirium is a multifactorial pathology linked with pre-operatory comorbidities. The complexity of surgery has a big influence on the development of delirium, especially in the cases of aortic dissection. Interestingly, the development of delirium was associated with intra-operatory blood transfusions in agreement with similar studies in literature. Finally, our data point to a bridge between post-operatory electrolytic disturbances, as well as inflammatory response to infections as factors potentially triggering delirium onset.

Abstract

In June 2018 the SPIDER device, which is the full-size prototype of the negative ion source and extractor for the ITER Heating Neutral Beam has entered the first operation phase at the Neutral Beam Test Facility (NBTF) in Padova, Italy. This paper describes the present status of the device, the experimental plans and the results obtained during the first experimental campaign of the radio-frequency driven plasma source.

Abstract

The results of a systematic and quantitative experimental characterization of simultaneous operation of a preclinical SPECT insert inside an unmodified standard MR scanner (Philips 3T Achieva) are presented. The static gantry (20 cm diameter, 15 mm field of view) is composed of 10 MRI-compatible gamma cameras based on 8-mm thick CsI(Tl) scintillator (50 mm × 50mm) coupled with arrays of SiPMs. Mutual compatibility is achieved by proper design of the SPECT system: shielding, choice of materials, design of the analog boards. The measured energy resolution is 13% at 122 keV after correction of SiPM gain variability and local crystal luminosity. The intrinsic spatial resolution, after planar image statistical reconstruction by means of maximum likelihood, is 1mm, while the extrinsic resolution is below 0.9 mm across the entire transaxial FoV, measured with capillaries of 290 pm diameter. With the custom-designed 3 × 3 multi-pinhole collimator (whose minimal perturbation of the magnetic field can be reduced below 1ppm thanks to shimming), the measured sensitivity with 99mTc is 1100 cps/MBq. The MR image is not distorted by the operation of the SPECT, as well as SPECT performances are no degraded by MRI (T2W and gradient sequences). Simultaneous imaging of different phantoms (capillaries, Derenzo hot rods of 0.85-1.7 mm diameter) as well as a mouse brain are reported.

Abstract

Molecular genetic assessments that consider ecological information, in addition to endogamy levels, genetic diversity, and the genetic differentiation among species and populations, are particularly important for the conservation of biological diversity. Prime candidates for conservation genetic review are those subject to human use, including harvests for the ornamental fish trade. Colorful South American tetra, such as Moenkhausia oligolepis and M. forestii, are good examples of fish species that are widely collected and exported worldwide. This study aimed to evaluate the population-specific characteristics of M. oligolepis and M. forestii by comparing morphometric and molecular analyses based on ISSR markers, to provide information that would facilitate the sustainable management of these 2 species. Seventy-two specimens were collected from the Araguaia-Tocantins and Paraguay River Basins in Brazil. All specimens were measured and analyzed using ISSR markers. Population-exclusive bands were found among the 86 detected bands, while morphometric clusters reflected the geographical distribution of individuals. Correlated genetic and morphological variation supported the presence of 3 distinct groups from tributaries of the Araguaia and Mortes Rivers. Using the same techniques, all M. oligolepis populations were isolated from M. forestii. This study on Moenkhausia presents an interesting example that could be used to construct a framework of South American ichthyodiversity, and reinforces the necessity of habitat conservation to prevent the loss of biological diversity.

Abstract

The realization of the ITER Neutral Beam Test Facility (NBTF) and the start the experimental phase are important tasks of the fusion roadmap, since the target requirements of injecting to the plasma a beam of Deuterium atoms with a power up to 16.5 MW, at 1MeV of energy and with a pulse length up to 3600s have never been reached together before. The ITER NBTF, called PRIMA (Padova Research on ITER Megavolt Accelerator), is hosted in Padova, Italy; it includes two experiments: MITICA, the full-scale prototype of the ITER injector and SPIDER, the full-size negative ion source. The realization promoted by the ITER organization is carried out with the contribution of the European Union, channeled through the Joint Undertaking for ITER (F4E), of the Consorzio RFX which hosts the Test Facility, the Japanese and the Indian ITER Domestic Agencies (JADA and INDA) and several European laboratories, such as IPP-Garching, KIT-Karlsruhe, CCFECulham, CEA-Cadarache. The early start of operation of PRIMA experiments is urgent because sufficient experimental time is necessary to face and solve the issues related to the achievement of the desired performance in time for the ITER operation, requiring NBI since the beginning. Substantial progresses have been recently achieved: the buildings construction, begun in October 2012, has been completed by the end of 2015 and the installation of some components has been started since the end of 2014. The SPIDER realization is well advanced: the installation phase is proceeding in good agreement with the general plan; it is expected to be almost completed by the end of 2016. In parallel, the commissioning of the SPIDER power supply (PS) and auxiliary plant systems is being proceeding. Tests at full power and remote control are planned, including also those addressed to reproduce the grid breakdowns and to test the relevant protections. The design of the MITICA injector components was completed in 2015their procurement is being made through a number of tenders, some of them already launched. The HV Power Supply system of the MITICA 1MV accelerator, provided by JADA, was delivered on site in December 2015. The challenging installation of these components, including the step-up transformers and the SF6 gas insulated transmission line, started soon after and will go on throughout 2016. The present phase, with the PRIMA buildings continuously filled with new components, with the installation activities progressing and with also the commissioning and testing phase starting represents a substantial step forward toward the main target. The paper will describe the main challenges the Project Team has dealt during this phase and the important feedback derived for the ITER HNB systems both from the technical and the organizational standpoints.

Abstract

A multi-purpose manipulator (MPM) system has been developed and installed on the W7-X vessel, aimed at investigating the edge plasmas of the stellarator. It is a flexible tool for integration of a variety of different diagnostics as e. g. electrical probes, probing magnetic coils, material collection, or material exposition probes, and gas injection. In the framework of the EUROfusion S1 work program for the preparation and exploitation of W7-X campaigns, a diagnostic insertable probe head called HRP (High Resolution Probe) has been developed by Consorzio RFX in collaboration with IPP Greifswald, to study the electrostatic and electromagnetic features of turbulence in the edge region of W7-X using the MPM.

Abstract

This paper reports the main results of the RFX-mod fusion science activity since the last 2014 IAEA Fusion Energy Conference. The RFX-mod device is characterized by a unique flexibility in terms of accessible magnetic configurations. Axisymmetric and helically shaped Reversed-field pinch equilibria have been studied, along with tokamak plasmas in a wide range of q(a) regimes (spanning from 4 down to 1.2 values). The full range of magnetic configurations in between the two, the so-called ultra low-q ones, has been explored, with the aim of studying specific physical issues common to all equilibria, such as, for example, the density limit phenomenon. The powerful RFX-mod feedback control system has been exploited for MHD control, which allowed to extend the range of experimental parameters, as well as to induce specific magnetic perturbations for the study of 3D effects. In particular, transport, edge and isotope effect in 3D equilibria have been investigated, along with runaway mitigations through induced magnetic perturbations. The first transitions to H-mode in circular and D-shaped tokamak plasmas have been obtained thanks to an active modification of the edge electric field through a polarized electrode. The experiments are supported by intense modelling with 3D MHD, gyrokinetic, guiding center and transport codes. Proposed modifications to the RFX-mod device, which will enable further contributions to the solution of key issues in the roadmap to ITER and DEMO, are also briefly presented.

Abstract

The modified RFX is a very flexible device used for a variety of control schemes for MHD instabilities and for advanced reverse field pinch scenarios. Relative to the previous machine, RFX-mod has a thin Cu shell with vertical field penetration time τS, lowered from 450 to 50 ms and shell/plasma proximity from b/a ≤ 1.24-1.1. Toroidal equilibrium is feedback-controlled and new power supplies provide better Bφ control. Newly designed graphite tiles protect the vessel from localized power deposition. A mesh of 192 external saddle coils, supervised by a digital feedback system, controls radial fields due to field errors and MHD modes. The paper presents an overview of the very encouraging results obtained using both new and 'standard' advanced operational modes in the current range 0.3-1 MA. A dramatic improvement of plasma performance was obtained by using the saddle coils to cancel all the radial field components, an operation mode dubbed virtual shell (VS). The toroidal voltage was lowered by more than 25% and the pulse length was tripled, up to 7 times the τS. Steady-state RFP pulses are now limited only by the applied volt-seconds. The improved magnetic boundary also has an effect on the tearing modes underlying the sustainment of the RFP, whose core amplitude is more than halved. The VS combined with new schemes for the active rotation of the MHD dynamo modes has allowed us to obtain reliable and well-controlled long RFP pulses in the MA current range. This results in a 100% increase in the particle and energy confinement time relative to the previous experiment and opens the possibility of exploring the machine performance in the 2 MA design range. © 2007 IAEA.

Abstract

We present the Galaxy Stellar Mass Function (MF) up to z~1 from the zCOSMOS-bright 10k spectroscopic sample. We investigate the total MF and the contribution of ETGs and LTGs, defined by different criteria (SED, morphology or star formation). We unveil a galaxy bimodality in the global MF, better represented by 2 Schechter functions dominated by ETGs and LTGs, respectively. For the global population we confirm that low-mass galaxies number density increases later and faster than for massive galaxies. We find that the MF evolution at intermediate-low values of Mstar (logM, Revised version, accepted for publication on Astronomy and Astrophysics

Abstract

The ITER project requires additional heating by two neutral beam injectors, each accelerating to 1 MV a 40 A beam of negative deuterium ions, to deliver to the plasma a power of about 17 MW for one hour. As these requirements have never been experimentally met, it was recognized as necessary to setup a test facility, PRIMA (Padova Research on ITER Megavolt Accelerator), in Italy, including a full-size negative ion source, SPIDER, and a prototype of the whole ITER injector, MITICA, aiming to develop the heating injectors to be installed in ITER. This realization is made with the main contribution of the European Union, through the Joint Undertaking for ITER (F4E), the ITER Organization and Consorzio RFX which hosts the Test Facility. The Japanese and the Indian ITER Domestic Agencies (JADA and INDA) participate in the PRIMA enterprise; European laboratories, such as IPP-Garching, KIT-Karlsruhe, CCFE-Culham, CEA-Cadarache and others are also cooperating. Presently, the assembly of SPIDER is on-going and the MITICA design is being completed. The paper gives a general overview of the test facility and of the status of development of the MITICA and SPIDER main components at this important stage of the overall development; then it focuses on the latest and most critical issues, regarding both physics and technology, describing the identified solutions.

Abstract

This paper reports the main recent results of the RFX-mod fusion science activity. The RFX-mod device is characterized by a unique flexibility in terms of accessible magnetic configurations. Axisymmetric and helically shaped reversed-field pinch equilibria have been studied, along with tokamak plasmas in a wide range of q(a) regimes (spanning from 4 down to 1.2 values). The full range of magnetic configurations in between the two, the so-called ultra-low q ones, has been explored, with the aim of studying specific physical issues common to all equilibria, such as, for example, the density limit phenomenon. The powerful RFX-mod feedback control system has been exploited for MHD control, which allowed us to extend the range of experimental parameters, as well as to induce specific magnetic perturbations for the study of 3D effects. In particular, transport, edge and isotope effects in 3D equilibria have been investigated, along with runaway mitigations through induced magnetic perturbations. The first transitions to an improved confinement scenario in circular and D-shaped tokamak plasmas have been obtained thanks to an active modification of the edge electric field through a polarized electrode. The experiments are supported by intense modeling with 3D MHD, gyrokinetic, guiding center and transport codes. Proposed modifications to the RFX-mod device, which will enable further contributions to the solution of key issues in the roadmap to ITER and DEMO, are also briefly presented. © 2017 IAEA, Vienna.

Abstract

The aim of this study was to assess, by positron emission tomography (PET), the effect on cerebral functional activity of a new lorazepam-γ-aminobutyric acid (GABA) conjugate [3-(4-acetamido)-butyrril lorazepam (DDS2700)]. Ten healthy volunteers were studied by PET and [ 18 F]fluoro-deoxy-D-glucose ([ 18 F]FDG) under baseline conditions and following the administration of DDS2700. Regional cerebral blood flow (rCBF) was measured by PET and 15 O-water in three additional participants while they performed attentive tasks, before and after drug administration. DDS2700 induced a decrease in the regional cerebral metabolic rate of glucose (rCMRglu) in the thalamus (-17%), cerebellum (-11%) and caudate nucleus (-8%). The observed effects on glucose metabolism were probably related to the subjective sedation and tiredness reported by the participants. During the attentive tasks, rCBF increased in frontal and temporal regions associated with attentional processing of auditory material. These circuits were no longer active after DDS2700 administration, while rCBF increased in cingulate cortex, occipitoparietal regions, pons and cerebellum. These drug-induced activations might be directly related to intervening sleepiness and to the consequent effort in keeping attention focused on the tasks. The effects of DDS2700 on glucose metabolism at rest, and on rCBF during activation conditions, indicate a drug action on cerebral networks involved in alertness, vigilance and attention maintenance. PET assessment by [ 18 F]FDG and water may provide complementary information in pharmacodynamic studies.

Abstract

This study investigates the effect of chronic treatment with Fluvoxamine, a potent and specific serotonin reuptake sites inhibitor (SSRI), on 5HT(2) serotonin and D(2) dopamine receptors in the brain of drug naive unipolar depressed patients. Drug effect was evaluated in different cortical areas and in the basal ganglia by positron emission tomography (PET) and fluoroethyl-spiperone ([(18)F]FESP), an high affinity 5HT(2) serotonin and D(2) dopamine receptors antagonist. Patients underwent a PET study at recruitment and after clinical response to Fluvoxamine treatment. Nine of the 15 patients recruited completed the study. Fluvoxamine treatment significantly improved clinical symptoms and modified [(18)F]FESP binding in the frontal and occipital cortex of all of the nine patients who completed the study; in these regions a mean 31% increase in the in vivo [(18)F]FESP binding was found (P < 0.01). On the contrary, no significant changes in the in vivo [(18)F]FESP binding were found in the basal ganglia where [(18)F]FESP binds mainly to D(2) dopamine receptors. Chronic treatment with Fluvoxamine significantly increases the in vivo binding of [(18)F]FESP in the frontal and occipital cortex of drug naive unipolar depressed patients. The increase of the in vivo binding of [(18)F]FESP may reflect a modification in 5HT(2) binding capacity secondary to changes in cortical serotonin activity. (C) 2000 Academic Press.

Abstract

We evaluated [ 11 C]fluvoxamine as a tracer for the serotonin re-uptake site. Studies of the distribution of the tracer in rat and primate brain showed adequate uptake of [ 11 C]fluvoxamine, but failed to reveal regions with known high density of serotoninergic re-uptake sites. Pretreatment with unlabeled fluvoxamine did not substantially change the distribution. In rat brain tissue, nearly all radioactivity represented intact [ 11 C]fluvoxamine. [ 11 C]Fluvoxamine does not function as a tracer for serotonin re-uptake sites, owing to high nonspecific binding in the brain.

Abstract

There have been a number of indications that sex hormones can affect the rate of growth of meningiomas during pregnancy. The presence of oestrogen or progesterone receptors in meningiomas and the influence of sex hormones upon cell cultures derived from human meningiomas have been extensively investigated. The results have been controversial, with most of the discussion centring upon the presence and possible role of oestrogen receptors. The aim of the present study was to assess oestrogen receptors in human meningiomas with 16α[ 18 F]fluoro-17β-oestradiol ([ 18 F]FES) and positron emission tomography (PET). With this purpose in mind, we measured the regional brain uptake of [ 18 F]FES in six patients with a neuroradiological and histological diagnosis of meningioma, comparing the in vivo PET data with the immunohistological analysis of oestrogen receptors performed on formalin-fixed, paraffin-embedded tissue obtained at surgery. Two analyses were used for the in vivo measurement of [ 18 F]FES binding to oestrogen receptors: the ratio of tumour activity to that of normal tissue (T/NT), calculated 90 min after tracer injection, and the ratio between the equilibrium distribution volume (DV) in normal and pathological tissues, calculated by means of a graphical kinetic analysis. PET studies demonstrated a marked uptake of [ 18 F]FES by the tumour in four of the six patients. Immunohistochemical assay using a manual staining method capable of detecting oestrogen receptors at a level of > 10 pmol mg -1 of protein detected only sparse immunostaining in one of the six meningiomas. Distinct albeit weak immunostaining was demonstrated in five of the six meningiomas when the sensitivity of the immunohistochemical assay was increased to < 10 pmol mg -1 of protein by use of an automated staining method (Bioteck 1000). Comparison of the in vivo and immunohistochemical results showed a correlation in five of the six patients, thus indicating the high sensitivity of [ 18 F]FES for the in vivo evaluation of oestrogen receptor expression.

Abstract

Both FLB 457 and FLB 463, two substituted benzamides with high affinity for the dopamine D-2 receptors, were labeled with bromine-76 for PET investigations. [76Br]FLB 457 was prepared by electrophilic substitution of the tributyltin precursor. The radiochemical yield was 80%. [76Br]FLB 463 was prepared by a direct electrophilic substitution enhanced by the hydroxyl group of the debromo analogue, with a total radiochemical yield of 50%. Radiochemical and chemical purity values of the radioligands as analyzed by radio-TLC and HPLC were > 99%, and the specific radioactivity was -40 GBq/mumol. During PET examinations of [76Br]FLB 457 and [76Br]FLB 463 binding in baboons there was a rapid and high uptake in the striatum. The striatal radioactivity concentration reached a plateau 1 h postinjection (p.i.). The striatum-to-cerebellum radioactivity concentration ratio increased from 11 at 1 h p.i., to 28 at 4 h p.i. for [76Br]FLB 457, owing to a continuous wash-out from the cerebellum. For [76Br]FLB 463 the corresponding value increased from 10 to 19.5. [76Br]FLB 457 has in contrast to [76Br]FLB 463 a high uptake in thalamic structures and has therefore an additional potential as a radioligand for PET examination of extrastriatal dopamine D-2 receptors in the living human brain.

Dal punto di vista della medicina, cio ha rappresentato la base per l’identificazione di approcci terapeutici innovativi fondati sul concetto di medicina molecolare e di terapia personalizzata. A loro volta, medicina molecolare e terapia personalizzata hanno determinato un notevole input e ampia diffusione delle metodiche di imaging biomedico sia in campo clinico sia, soprattutto negli ultimi anni, nel settore preclinico.
