2. Tailoring microstructure and mechanical properties of an LPBF-processed beta Ti-Nb alloy through post-heat treatments

This study provides a comprehensive analysis of a Ti‑42Nb alloy produced via laser powder bed fusion (LPBF) with varying post-heat treatment durations within the α + β phase range at 723 K. Synchrotron XRD analysis revealed the formation of the metastable orthorhombic αiso'' phase during heat treatment, acting as an intermediate to the stable α phase. With prolonged heat treatment, the αiso'' phase fraction increased, reaching approximately 25 % after 108.0 ks. SEM analysis identified β grain boundaries as primary sites for early αiso'' precipitation, while intragranular αiso'' precipitation was delayed. Up to 28.8 ks, volume fraction and size of intragranular precipitates exhibited notable variations due to minor Nb content fluctuations from LPBF processing, resulting in an increased spread of hardness and Young's modulus on the micro scale. Tensile tests revealed significant strength enhancement through post-heat treatment for 108 ks compared to the as-built state, achieving a yield strength of around 1060 MPa (50 % increase) and ultimate tensile strength of 1125 MPa (55 % increase). Extended growth of the αiso'' phase led to an increased Young's modulus, reaching 87 GPa after 108.0 ks. These findings provide valuable insights for developing post-heat treatment strategies for LPBF-produced Ti‑42Nb implants, including both bulk materials and lattice structures.

3. Microbial methane oxidation efficiency and robustness during lake overturn

Abstract Many seasonally stratified lakes accumulate substantial amounts of the greenhouse gas methane in the anoxic zone. Methane oxidizing bacteria in the water column act as a converter, oxidizing methane into carbon dioxide and biomass before it reaches the atmosphere. Current observations and estimates of this methane oxidation efficiency are diverging, especially for the lake overturn period. Here, we combine a model of turbulent mixing, gas exchange, and microbial growth with a comprehensive data set for autumn mixing to quantify the relevant physical and microbial processes for a 16 m deep, wind‐sheltered Swiss lake. Scenario analysis suggests that the methane converter is efficient and robust under a large range of mixing velocities and only rare events of pronounced surface cooling can trigger substantial outgassing. This case study combines in situ observation and a deterministic physically based model and suggests that the frequency of storms may strongly impact methane emissions for similar temperate lakes.

5. Lake mixing regime selects apparent methane oxidation kinetics of the methanotroph assemblage

In lakes, large amounts of methane are produced in anoxic sediments. Methane-oxidizing bacteria effectively convert this potent greenhouse gas into biomass and carbon dioxide. These bacteria are present throughout the water column, where methane concentrations can range from nanomolar to millimolar. In this study, we tested the hypothesis that methanotroph assemblages in a seasonally stratified freshwater lake are adapted to the contrasting methane concentrations in the epi- and hypolimnion. We further hypothesized that lake overturn would change the apparent methane oxidation kinetics as more methane becomes available in the epilimnion. In addition to the change in the methane oxidation kinetics, we investigated changes in the transcription of genes encoding methane monooxygenase, the enzyme responsible for the first step of methane oxidation, with metatranscriptomics. Using laboratory incubations of the natural microbial communities, we show that the half-saturation constant (Km) for methane – the methane concentration at which half the maximum methane oxidation rate is reached – was 20 times higher in the hypolimnion than in the epilimnion during stable stratification. During lake overturn, however, the kinetic constants in the epi- and hypolimnion converged along with a change in the transcriptionally active methanotroph assemblage. Conventional particulate methane monooxygenase appeared to be responsible for methane oxidation under different methane concentrations. Our results suggest that methane availability is one important factor for creating niches for methanotroph assemblages with well-adapted methane oxidation kinetics. This rapid selection and succession of adapted lacustrine methanotroph assemblages allowed the previously reported high removal efficiency of methane transported to the epilimnion to be maintained – even under rapidly changing conditions during lake overturn. Consequently, only a small fraction of methane stored in the anoxic hypolimnion is emitted to the atmosphere.

Systemic sclerosis (SSc) is a rare chronic disease of unknown pathogenesis characterized by fibrosis of the skin and internal organs, vascular alteration, and dysregulation of the immune system. In order to better understand the immune system and its perturbations leading to diseases, the study of the mechanisms regulating cellular metabolism has gained a widespread interest. Here, we have assessed the metabolic status of plasma and dendritic cells (DCs) in patients with SSc. We identified a dysregulated metabolomic signature in carnitine in circulation (plasma) and intracellularly in DCs of SSc patients. In addition, we confirmed carnitine alteration in the circulation of SSc patients in three independent plasma measurements from two different cohorts and identified dysregulation of fatty acids. We hypothesized that fatty acid and carnitine alterations contribute to potentiation of inflammation in SSc. Incubation of healthy and SSc dendritic cells with etoposide, a carnitine transporter inhibitor, inhibited the production of pro-inflammatory cytokines such as IL-6 through inhibition of fatty acid oxidation. These findings shed light on the altered metabolic status of the immune system in SSc patients and opens up for potential novel avenues to reduce inflammation.

Nearly fully dense, glassy Zr52.5Cu17.9Ni14.6Al10Ti5 bulk specimens were fabricated by selective laser melting (SLM) and their behaviour during compressive loading, during wear testing and in a corrosive medium was investigated. Their performance was compared with as-cast material of the same composition. The additively manufactured samples exhibit a yield strength around 1700 MPa combined with a plastic strain of about 0.5% after yielding despite the residual porosity of 1.3%, which is distributed uniformly in the samples. The propagation of shear bands in the bulk metallic glass prepared by SLM was studied. The specific wear rate and the worn surfaces demonstrated that similar wear mechanisms are active in the SLM and the as-cast samples. Hence, manufacturing the glass in layers does not adversely affect the wear properties. The same holds for the corrosion tests, which were carried out in 0.01 M Na2SO4 and 0.1 M NaCl electrolyte. The anodic polarization curves of SLM samples and as-cast samples revealed a similar corrosion behaviour. However, the SLM samples have a slightly reduced susceptibility to pitting corrosion and exhibit an improved surface healing ability, which might be attributed to an improved homogeneity of the additively manufactured glass. 11. Five Steps to Teach Simple Sentence Writing to Students with Learning Disabilities

Many students with learning disabilities struggle with sentence writing fluency, the skill of quickly and accurately generating words that follow rules of semantics, spelling, syntax, and usage within sentence structures understandable to readers. Students who struggle with sentence writing fluency may face difficulty fully expressing their ideas while engaging in academic writing. In the present article, we describe how a combination of explicit instruction and fluency practice can improve the simple sentence writing fluency of students with learning disabilities. We detail how five design and delivery steps can help to create a supplemental writing intervention that addresses simple sentence structure, syntax, and usage.

NEST consiste en una estructura fija con una serie módulos individuales que son utilizados como oficinas o apartamentos donde poder vivir y trabajar. Además, cada uno de estos módulos actúa a modo de laboratorio donde se investigan diferentes aspectos como el uso de madera como material de construcción o la fabricación digital y donde, continuamente, se lleva a cabo el desarrollo de materiales innovadores, así como su evaluación. NEST es un edificio modular cuyas unidades están individualmente conectadas por medio de redes comunes de suministro de energía y gestión y tratamiento de aguas. Cuando finalizan los estudios asociados a un módulo y los nuevos productos han sido optimizados, éste es desinstalado y remplazado por una nueva unidad de cara a abordar nuevos retos e investigaciones.

13. Interdependencies between temperature and moisture sensitivities of CO2 emissions in European land ecosystems

Soil respiration is one of the largest terrestrial fluxes of carbon dioxide (CO2) to the atmosphere. Hence, small changes in soil respiration rates could have large effects on atmospheric CO2. In order to assess CO2 emissions from diverse European soils with different land-use types and climate (soil moisture and temperature), we conducted a laboratory incubation experiment. Emission measurements of CO2 under controlled conditions were conducted using soil monoliths of nine sites from a European flux network (ÉCLAIRE). The sites are located all over Europe – from the United Kingdom in the west to Ukraine in the east, and from Italy in the south to Finland in the north – and can be separated according to four land-use types (forests, grasslands, arable lands and one peatland). Intact soil cores were incubated in the laboratory in a two-way factorial design, with temperature (5, 10, 15, 20 and 25 °C) and water-filled pore space (WFPS; 5, 20, 40, 60 and 80 %) as the independent variables, while CO2 flux was the response variable. The latter was measured with an automated laboratory incubation measurement system. Land use generally had a substantial influence on carbon dioxide fluxes, with the order of CO2 emission rates of the different land-use types being grassland > peatland > forest/arable land (P < 0.001). CO2 efflux responded strongly to varying temperature and moisture content with optimum moisture contents for CO2 emissions between 40 and 70 % WFPS and a positive relationship between CO2 emissions and temperature. The relationship between temperature and CO2 emissions could be well described by a Gaussian model. Q10 values ranged between 0.86 and 10.85 and were negatively related to temperature for most of the moisture contents and sites investigated. At higher temperatures the effect of water and temperature on Q10 was very low. In addition, under cold temperatures Q10 varied with moisture contents, indicating a stronger prospective effect of rain events in cold areas on temperature sensitivity. At both coniferous forest sites we found a strong increase in the temperature sensitivity at a moisture range between 20 and 40 % WFPS. We developed a new approach to calculate moisture sensitivity (MS) of CO2 efflux. MS was calculated as the slope of a polynomial function of second degree. Moisture sensitivities were highest under dry and wet conditions. In addition we found a positive relationship between MS of CO2 efflux and temperature for both arable lands.

The development of susceptibility maps for debris flows is of primary importance due to population pressure in hazardous zones. However, hazard assessment by process-based modelling at a regional scale is difficult due to the complex nature of the phenomenon, the variability of local controlling factors, and the uncertainty in modelling parameters. A regional assessment must consider a simplified approach that is not highly parameter dependant and that can provide zonation with minimum data requirements. A distributed empirical model has thus been developed for regional susceptibility assessments using essentially a digital elevation model (DEM). The model is called Flow-R for Flow path assessment of gravitational hazards at a Regional scale (available free of charge under http://www.flow-r.org) and has been successfully applied to different case studies in various countries with variable data quality. It provides a substantial basis for a preliminary susceptibility assessment at a regional scale. The model was also found relevant to assess other natural hazards such as rockfall, snow avalanches and floods. The model allows for automatic source area delineation, given user criteria, and for the assessment of the propagation extent based on various spreading algorithms and simple frictional laws. We developed a new spreading algorithm, an improved version of Holmgren's direction algorithm, that is less sensitive to small variations of the DEM and that is avoiding over-channelization, and so produces more realistic extents. The choices of the datasets and the algorithms are open to the user, which makes it compliant for various applications and dataset availability. Amongst the possible datasets, the DEM is the only one that is really needed for both the source area delineation and the propagation assessment; its quality is of major importance for the results accuracy. We consider a 10 m DEM resolution as a good compromise between processing time and quality of results. However, valuable results have still been obtained on the basis of lower quality DEMs with 25 m resolution.

Estudaram-se os efeitos da infusão contínua da associação propofol e cetamina sobre variáveis fisiológicas e eletrocardiográficas e sua possível analgesia em 12 cadelas. Após indução com propofol, os animais receberam 0,4mg/kg/min de propofol + 0,2mg/kg/min de cetamina racêmica (n = 6, grupo PC) ou 0,4mg/kg/min de propofol + 0,1mg/kg/min de cetamina S+ (n = 6, grupo PCS). Avaliaram-se: teste álgico, freqüência cardíaca (FC), parâmetros eletrocardiográficos, freqüência respiratória (FR), pressão arterial sistólica, média e diastólica (PAS, PAM, PAD), saturação da oxiemoglobina (SpO2) e temperatura retal (TR). Houve elevação da FC sem alterações eletrocardiográficas, com exceção de aumento na amplitude da onda T em um animal de cada grupo. A FR diminuiu, e os valores de SpO2 ficaram abaixo de 90% em alguns momentos nos dois grupos. PAS, PAM e PAD diminuíram, mas não houve diferença entre os protocolos. Não se observou analgesia em sete animais, três cadelas apresentaram analgesia discreta, e apenas duas demonstraram analgesia favorável. Conclui-se que os protocolos são seguros em cadelas, contudo não há analgesia suficiente para procedimento cirúrgico. As alterações eletrocardiográficas foram relacionadas à FC e à amplitude de onda T, sendo esta sugestiva de hipóxia do miocárdio.The effects of propofol and ketamine on physiological parameters, electrocardiography, and analgesia were evaluated in twelve dogs that received propofol-ketamine (0.4mg/kg/min + 0.2mg/kg/min, n=6, PK group) or propofol-S+ketamine (0.4mg/kg/min + 0.1mg/kg/min, n=6, PKS group) after induction of anesthesia with propofol (8.0mg/kg). Assessments of pain; heart rate (HR); electrocardiography (ECG); respiratory rate (RR); systolic, medium, and diastolic arterial pressures (SAP, MAP, DAP); saturation of hemoglobin (SpO2); and rectal temperature (RT) were conducted. There was a rise in HR with no electrocardiographically changes, but an increase in amplitude of T wave in one animal of each group. RR decreased and SpO2 were lower than 90% in two moments of both groups. SAP, MAP, and DAP were reduced during the experimental period, but with no statistical difference between the protocols. There was no analgesia in seven bitches, mild analgesia was observed in three, and satisfactory analgesia in only two animals tested. It was concluded that the protocols are safe for dogs, but there is no analgesia for surgical procedures. The changes in ECG are related to HR and amplitude of T wave, which may be due to myocardial hypoxia.

Utilizaram-se membranas de látex para o reparo de defeitos diafragmáticos em 12 cães, distribuídos em três grupos: no G1 utilizou-se membrana comercial e no G2, membrana experimental. O G3 foi usado como controle. Foi feito um defeito retangular no músculo diafragma, com 4cm de comprimento por 3cm de largura, que nos grupos G1 e G2 foi substituído pelo implante da membrana de látex correspondente. Os animais foram avaliados por estudo radiográfico, hemograma, videocirurgia e análise histológica. Os resultados mostraram que a membrana de látex do grupo 2 foi eficiente na correção de defeito no diafragma, promovendo a reparação e neovascularização tecidual local, sem causar rejeição durante o período de avaliação.Latex membranes were experimentally used to repair diaphragmatic defects in 12 dogs, distributed in 3 groups. In group 1, a commercial membrane was used, and in group 2, an experimental membrane. Group 3, animals were used as control. A rectangular defect (4cm in length and 3cm in width) was surgically performed in the diaphragm muscle, which was substituted, in group 1 and 2, by the implantation of corresponding latex membrane. The animals were evaluated by radiography, blood count, video-surgery, and histologic study. Results showed that the latex membrane of group 2, were efficient in the correction of the defect in the diaphragm, promoting the repairing and local neovascularization, without causing rejection during the evaluated period.

Signals transmitted over a multi-path propagation channel exhibit Inter-Path Interference (IPI) and fading. The receiver has to employ measures to mitigate these effects or it will incur severe performance degradation. A classic approach in Code Division Multiple Access (CDMA) communications is the Rake receiver. Alternatively, a Linear Minimum Mean Square Error (LMMSE) equalizer can further improve performance. This paper compares performance and computational complexity of these two algorithms. A hybrid solution will be introduced in order to decrease computational complexity while retaining most of the interference suppression capabilities of the LMMSE equalizer.

18. Relative stability of soil carbon revealed by shifts in δ15N and C:N ratio

Life on earth drives a continuous exchange of carbon between soils and the atmosphere. Some forms of soil carbon, or organic matter, are more stable and have a longer residence time in soil than others. Relative differences in stability have often been derived from shifts in δ13C (which is bound to a vegetation change from C3 to C4 type) or through 14C-dating (which is bound to small sample numbers because of high measurement costs). Here, we propose a new concept based on the increase in δ15N and the decrease in C:N ratio with increasing stability. We tested the concept on grasslands at different elevations in the Swiss Alps. Depending on elevation and soil depth, it predicted mineral-associated organic carbon to be 3 to 73 times more stable than particulate organic carbon. Analysis of 14C-ages generally endorsed these predictions.

19. Membrana de látex natural de Hevea brasiliensis auxilia no processo de reparação tecidual em bovinos

RESUMO Feridas cutâneas em bovinos são um constante desafio clínico cirúrgico por desencadearem perdas econômicas bastante significativas. O látex proveniente da seiva da seringueira (Hevea brasiliensis) apresenta potencial terapêutico para incrementar o processo de reparação tecidual. Portanto, pretendeu-se com esse estudo avaliar o tipo de reação tecidual e os possíveis mecanismos de angiogênese desencadeados pelo implante de uma membrana de látex natural em bovinos. Para tal, foram utilizados seis bovinos da raça Nelore, submetidos ao implante subcutâneo experimental de três fragmentos de membranas de látex natural. Foram coletadas amostras de tecido e da membrana aos 15, 30 e 45 dias após a implantação, para avaliações histológicas, ultraestruturais por microscopia eletrônica de varredura e imunoistoquímicas com anticorpos antimarcador de macrófagos (MAC), CYR 61 e VEGF. O implante de látex proporcionou aumento da angiogênese e reparação tecidual em bovinos, não mediada pela expressão do VEGF e CYR 61.

A protocol for the mechanochemical synthesis of copper(I)/N-heterocyclic carbene complexes using cheap and readily available K3PO4 as base has been developed. This method employing a ball mill is amenable to typical simple copper(I)/NHC complexes but also to a sophisticated copper(I)/N-heterocyclic carbene complex bearing a guanidine moiety. In this way, the present approach circumvents commonly employed silver(I) complexes which are associated with significant and undesired waste formation and the excessive use of solvents. The resulting bifunctional catalyst has been shown to be active in a variety of reduction/hydrogenation transformations employing dihydrogen as terminal reducing agent.

Since its first proposal, the concept of on- and off-track lesions in anterior shoulder instability has gained clinical relevance as a tool to predict the failure rate of arthroscopic Bankart repair. Current literature only reports either short-term follow-up or long-term results of small sample sizes. The aim of this study was to provide a long-term evaluation of recurrent instability following arthroscopic Bankart repair in a large cohort using the on-track vs. off-track concept as a predictor for failure.We retrospectively analyzed 271 patients who underwent primary arthroscopic Bankart repair for anterior shoulder instability between 1998 and 2007. All patients with a minimum follow-up of 78 months and a preoperative computed tomographic (CT) or magnetic resonance imaging (MRI) scan were included into the study. Preoperative CT and/or MRI scans were used to determine the glenoid track and width of Hill-Sachs lesion. Recurrence of instability was defined as presence of instability symptoms (dislocation, subluxation, and/or apprehension) or revision surgery (stabilization procedure) and was assessed as the primary outcome parameter.The glenoid track of 163 shoulders was assessed (female n = 51, male n = 112) with a mean follow-up of 124 months (99.4-145.6, standard deviation = 2.5) and a mean age of 24 years (20-34.). An off-track Hill-Sachs lesion was found in 77 cases (47%), and in 86 cases (53 %) it was on-track. The rate of recurrent instability in the off-track group was 74% (n = 57) compared with 27% (n = 23) in the on-track group (P.001). The overall rate of revision surgery due to instability was 29% (n = 48) after a mean time of 50.9 months (±42.8) following Bankart repair. The rate of revision surgery in the off-track group was 48% (n = 37) after a mean of 53.5 months (±42.0) vs. 13% (n = 11) after 42.3 months (±46.3) in the on-track group (P.001).This study shows that the on- and off-track concept helps to distinguish patients for whom an isolated arthroscopic Bankart repair yields long-term benefits. Because of the high rate of recurrent instability in the off-track group, an off-track lesion should be treated surgically in such a way that the off-track lesion is converted into an on-track lesion.

Headache is a common symptom during pregnancy and in puerperium that requires careful consideration, as it may be caused by a life-threatening condition. Headaches in pregnant women and women in puerperium are classified as primary or secondary; acute, severe and newly diagnosed headaches should prompt further investigation. We aimed to further characterise the demographic features, symptoms, examination findings, and neuroimaging results of cases of headache during pregnancy and in puerperium. All pregnant women or women in postpartum conditions who attended neurological consultations at the emergency department of the clinic for Gynaecology, Obstetrics and Reproductive Medicine of Saarland University/Germany between 2001/2015 and 2012/2019 were enrolled in this retrospective chart review. Data collected from the charts included demographic/pregnancy characteristics, clinical features and imaging findings. Descriptive statistics as well as binary logistic regression were performed. More than 50% of 97 patients had abnormal findings in their neurological examination. Magnetic resonance imaging findings were pathological for almost 20% of patients—indicating conditions such as cerebral venous thrombosis, reversible posterior leukoencephalopathy, brain tumour and intracranial bleeding. The odds of abnormal neuroimaging results were 2.2-times greater among women with abnormal neurological examination findings than among those with normal examination results. In cases of headache during pregnancy and in puerperium, neuroimaging should be indicated early on. Further research is needed to determine which conditions indicate a need for immediate neuroimaging.

28. Finite element analysis of medial closing and lateral opening wedge osteotomies of the distal femur in relation to hinge fractures

PurposeIntraoperative hinge fractures in distal femur osteotomies represent a risk factor for loss of alignment and non-union. Using finite element analysis, the goal of this study was to investigate the influence of different hinge widths and osteotomy corrections on hinge fractures in medial closed-wedge and lateral open-wedge distal femur osteotomies.MethodsThe hinge was located at the proximal margin of adductor tubercle for biplanar lateral open-wedge and at the upper border of the lateral femoral condyle for biplanar medial closed-wedge distal femur osteotomies, corresponding to optimal hinge positions described in literature. Different hinge widths (5, 7.5, 10 mm) were created and the osteotomy correction was opened/closed by 5, 7.5 and 10 mm. Tensile and compressive strain of the hinge was determined in a finite element analysis and compared to the ultimate strain of cortical bone to assess the hinge fracture risk.ResultsDoubling the correction from 5 to 10 mm increased mean tensile and compressive strain by 50% for lateral open-wedge and 48% for medial closed-wedge osteotomies. A hinge width of 10 mm versus 5 mm showed increased strain in the hinge region of 61% for lateral open-wedge and 32% for medial closed-wedge osteotomies. Medial closed-wedge recorded a higher fracture risk compared to lateral open-wedge osteotomies due to a larger hinge cross-section area (60-67%) for all tested configurations. In case of a 5 mm hinge, medial closed-wedge recorded 71% higher strain in the hinge region compared to lateral open-wedge osteotomies.ConclusionDue to morphological features of the medial femoral condyle, finite element analysis suggests that lateral-open wedge osteotomies are the preferable option if larger corrections are intended, as a thicker hinge can remain without an increased hinge fracture risk., Journal of Experimental Orthopaedics, 10 (1), ISSN:2197-1153

Background: Previous studies have shown that quality of life improves after coronary revascularization more so after coronary artery bypass grafting (CABG) than after percutaneous coronary intervention (PCI). This study aimed to evaluate the effect of fractional flow reserve guidance and current generation, zotarolimus drug-eluting stents on quality of life after PCI compared with CABG. Methods: The FAME 3 trial (Fractional Flow Reserve Versus Angiography for Multivessel Evaluation) is a multicenter, international trial including 1500 patients with 3-vessel coronary artery disease who were randomly assigned to either CABG or fractional flow reserve–guided PCI. Quality of life was measured using the European Quality of Life–5 Dimensions (EQ-5D) questionnaire at baseline and 1 and 12 months. The Canadian Cardiovascular Class angina grade and working status were assessed at the same time points and at 6 months. The primary objective was to compare EQ-5D summary index at 12 months. Secondary end points included angina grade and work status. Results: The EQ-5D summary index at 12 months did not differ between the PCI and CABG groups (difference, 0.001 [95% CI, –0.016 to 0.017]; P =0.946). The trajectory of EQ-5D during the 12 months differed ( P Conclusions: In the FAME 3 trial, quality of life after fractional flow reserve–guided PCI with current generation drug-eluting stents compared with CABG was similar at 1 year. The rate of significant angina was low in both groups and not significantly different. Complexity arises from: combinatorics; many design variables interacting with each other; coupling of technical and economical perspectives. This paper presents an approach based on (1) building a complex system model of modular models; (2) identifying the potential for standardization from a technical perspective; (3) cost-optimizing the degree of standardization. A product family of electric vehicles was optimized.

- Abstract
Complexity in systems design can be reduced by computing permissible ranges for some crucial design variables that need to be defined in an early design phase. These ranges are calculated such that there is sufficient tolerance for the remaining design variables in later design phases, while still achieving the overall system design goals. A new algorithm for this approach is presented and applied to the design of a vehicle powertrain mount system. The results show large permissible ranges for mount positions while maintaining sufficient tolerance for mount stiffnesses.

- Abstract
A digital twin (DT) relies on a detailed, virtual representation of a physical product. Since uncertainties and deviations can lead to significant changes in the functionality and quality of products, they should be considered in the DT. However, valuable product properties are often hidden and thus difficult to integrate into a DT. In this work, a Bayesian inverse approach based on surrogate models is applied to infer hidden composite laminate ply angles from strain measurements. The approach is able to find the true values even for ill-posed problems and shows good results up to 6 plies.

Lactic acid (LA) fermentation of dog’s mercury (M. perennis L.) herbal parts was investigated in samples inoculated with either Lactobacteria (Lactobacillus plantarum and Pediococcus pentosaceus, LBF) or whey (WF). Depending on fermentation time, LA concentrations were monitored in a range of 3.4–15.6 g/L with a concomitant pH decline from 6.5 to 3.9. A broad spectrum of cinnamic acids depsides containing glucaric, malic and 2-hydroxyglutaric acids along with quercetin and kaempferol glycosides were detected by LC-DAD-ESI-MSn. Moreover, in this study novel constituents were also found both in unfermented and fermented extracts. Furthermore, amino acids and particular Lactobacteria metabolites such as biogenic amines (e.g., putrescine, 4-aminobutyric acid, cadaverine) and 5-oxoproline were assigned in WF extracts by GC-MS analysis after silylation. Enhanced NFκB and cytokine expression (IL-6, TNFα, IL-8 and IL-1β) was induced by all extracts, both non-fermented and fermented, in NFκB-THP-1 reporter cells, showing a concentration-dependent immunostimulatory effect. The WF extracts were tested for micronuclei formation in THP-1 cells and toxicity in luminescent bacteria (V. fischeri), whereby no mutagenic or toxic effects could be detected, which corroborates their safe use in pharmaceutical remedies.

A 58-year-old male with prior history of mechanical aortic valve replacement (AVR) in 2009 for severe symptomatic aortic regurgitation in a bicuspid aortic valve, and since 2013 a new-onset severe asymptomatic primary mitral regurgitation (MR) due to prolapse of the anterior mitral valve leaflet (AMVL) presented himself with acute heart failure. Based on current guidelines recommendations, this patient was not eligible for transcutaneous mitral valve edge-to-edge repair (TEER), as well he was found as too high risk for conventional mitral valve repair. However, as a last resort TEER was undertaken with an unconventional strategy, which resulted in resolution of the MR and improvement of clinical, biochemical findings.

The uptake of COVID-19 vaccines has varied considerably across European countries. This study investigates people’s decision-making process regarding vaccination by analyzing qualitative interviews (n = 214) with residents from five European countries: Austria, Germany, Italy, Portugal, and Switzerland. We identify three factors that shape vaccination decision-making: individual experiences and pre-existing attitudes towards vaccination, social environment, and socio-political context. Based on this analysis, we present a typology of decision-making regarding COVID-19 vaccines, where some types present stable stances towards vaccines and others change over time. Trust in government and relevant stakeholders, broader social factors, and people’s direct social environment were particularly relevant to these dynamics. We conclude that vaccination campaigns should be considered long-term projects (also outside of pandemics) in need of regular adjustment, communication and fine-tuning to ensure public trust. This is particularly pertinent for booster vaccinations, such as COVID-19 or influenza., The first and corresponding authors contributed to this manuscript in equal parts and share first authorship. Solidarity and personal responsibility have been repeatedly called upon during the COVID-19 pandemic. This study quantifies and contextualizes the use of these terms in newspaper coverage in Germany and German-speaking Switzerland based on n = 640 articles from six functionally equivalent newspapers. The term solidarity in the context of the COVID-19 pandemic was mentioned in 541/640 articles (84.5%) and was primarily used during phases with high death rates and comparatively stringent policies in place, supporting the idea that solidarity was used to explain restrictive measures to the population and motivate people to comply with these measures. German newspapers published more articles on solidarity than Swiss-German newspapers, consistent with more stringent COVID-19 policies in Germany. Personal responsibility was mentioned in 133/640 articles (20.8%), meaning that the term was less frequently discussed than solidarity. Articles covering personal responsibility included more negative evaluations during phases of high infection rates as compared to phases of low infection rates. Findings indicate that the two terms were, at least to some extent, used in newspaper reporting to contextualize and justify COVID-19 policy during phases of high infection rates. Moreover, the term solidarity was used in a high variety of different contexts and the inherent limits of solidarity were rarely mentioned. Policymakers and journalists need to take this into account for future crises to not jeopardize the positive effects of solidarity.

The natural product aurachin D is a farnesylated quinolone alkaloid, which is known to possess activity against the causative agent of malaria, Plasmodium spp. In this study, we show that aurachin D inhibits other parasitic protozoa as well. While aurachin D had only a modest effect on Trypanosoma brucei rhodesiense, two other trypanosomatids, T. cruzi and Leishmania donovani, were killed at low micromolar and nanomolar concentrations, respectively, in an in vitro assay. The determined IC50 values of aurachin D were even lower than those of the reference drugs benznidazole and miltefosine. Due to these promising results, we set out to explore the impact of structural modifications on the bioactivity of this natural product. In order to generate aurachin D derivatives with varying substituents at the C-2, C-6 and C-7 position of the quinolone ring system, we resorted to whole-cell biotransformation using a recombinant Escherichia coli strain capable of aurachin-type prenylations. Quinolone precursor molecules featuring methyl, methoxy and halogen groups were fed to this E. coli strain, which converted the substrates into the desired analogs. None of the generated derivatives exhibited improved antiprotozoal properties in comparison to aurachin D. Obviously, the naturally occurring aurachin D features already a privileged structure, especially for the inhibition of the causative agent of visceral leishmaniasis.

The human SEC62 gene is located on chromosome 3q, was characterized as a tumor driver gene and is found to be overexpressed in an ever-growing number of tumors, particularly those with 3q26 amplification. Where analyzed, SEC62 overexpression was associated with poor prognosis. Sec62 protein is a membrane protein of the endoplasmic reticulum (ER) and has functions in endoplasmic reticulum protein import, endoplasmic reticulum-phagy and -in cooperation with the cytosolic protein calmodulin- the maintenance of cellular calcium homeostasis. Various human tumors show SEC62 overexpression in immunohistochemistry and corresponding cell lines confirm this phenomenon in western blots and immunofluorescence. Furthermore, these tumor cells are characterized by increased stress tolerance and migratory as well as invasive potential, three hallmarks of cancer cells. Strikingly, plasmid-driven overexpression of SEC62 in non-SEC62 overexpressing cells introduces the same three hallmarks of cancer into the transfected cells. Depletion of Sec62 from either type of SEC62 overexpressing tumor cells by treatment with SEC62-targeting siRNAs leads to reduced stress tolerance and reduced migratory as well as invasive potential. Where tested, treatment of SEC62 overexpressing tumor cells with the small molecule/calmodulin antagonist trifluoperazine (TFP) phenocopied the effect of SEC62-targeting siRNAs. Recently, first phase II clinical trials with the prodrug mipsagargin/G202, which targets cellular calcium homeostasis in prostate cells as well as neovascular tissue in various tumors were started. According to experiments with tumor cell lines, however, SEC62 overexpressing tumor cells may be less responsive or resistant against such treatment. Therefore, murine tumor models for tumor growth or metastasis were evaluated with respect to their responsiveness to treatment with a mipsagargin analog (thapsigargin), or trifluoperazine, which had previously been in clinical use for the treatment of schizophrenia, or with the combination of both drugs. So far, no additive effect of the two drugs was observed but trifluoperazine had an inhibitory effect on tumor growth and metastatic potential in the models. Here, we review the state of affairs.

The sudden and dramatic advent of the COVID-19 pandemic led to urgent demands for timely, relevant, yet rigorous research. This paper discusses the origin, design, and execution of the SolPan research commons, a large-scale, international, comparative, qualitative research project that sought to respond to the need for knowledge among researchers and policymakers in times of crisis. The form of organization as a research commons is characterized by an underlying solidaristic attitude of its members and its intrinsic organizational features in which research data and knowledge in the study is shared and jointly owned. As such, the project is peer-governed, rooted in (idealist) social values of academia, and aims at providing tools and benefits for its members. Vaccine uptake is essential to managing the ongoing COVID-19 pandemic, and vaccine hesitancy is a persistent concern. At the same time, both decision-makers and the general population have high hopes for COVID-19 vaccination. Drawing from qualitative interview data collected in October 2020 as part of the pan-European SolPan study, this study explores early and anticipatory expectations, hopes and fears regarding COVID-19 vaccination across seven European countries. We find that stances towards COVID-19 vaccines were shaped by personal lived experiences, but participants also aligned personal and communal interests in their considerations. Trust, particularly in expert institutions, was an important prerequisite for vaccine acceptance, but participants also expressed doubts about the rapid vaccine development process. Employing a bifunctional catalyst based on a copper(I)/NHC complex and a guanidine organocatalyst, catalytic ester reductions to alcohols with H2 as terminal reducing agent are facilitated. The approach taken here enables the simultaneous activation of esters through hydrogen bonding and formation of nucleophilic copper(I) hydrides from H2, resulting in a catalytic hydride transfer to esters. The reduction step is further facilitated by a proton shuttle mediated by the guanidinium subunit. This bifunctional approach to ester reductions for the first time shifts the reactivity of generally considered "soft" copper(I) hydrides to previously unreactive "hard" ester electrophiles and paves the way for a replacement of stoichiometric reducing agents by a catalyst and H2.

Background The use of genome-wide sequencing in pediatric medicine and research is growing exponentially. While this has many potential benefits, the normative and empirical literature has highlighted various ethical issues. There have not been, however, any systematic reviews of these issues. The aim of this systematic review is to determine systematically the spectrum of ethical issues that is raised for stakeholders in in pediatric genome-wide sequencing. Methods A systematic review in PubMed and Google Books (publications in English or German between 2004 and 2021) was conducted. Further references were identified via reference screening. Data were analyzed and synthesized using qualitative content analysis. Ethical issues were defined as arising when a relevant normative principle is not adequately considered or when two principles come into conflict. Results Our literature search retrieved 3175 publications of which 143 were included in the analysis. Together these mentioned 106 ethical issues in pediatric genome-wide sequencing, categorized into five themes along the pediatric genome-wide sequencing lifecycle. Most ethical issues identified in relation to genome-wide sequencing typically reflect ethical issues that arise in general genetic testing, but they are often amplified by the increased quantity of data obtained, and associated uncertainties. The most frequently discussed ethical aspects concern the issue of unsolicited findings. Conclusion Concentration of the debate on unsolicited findings risks overlooking other ethical challenges. An overarching difficulty presents the terminological confusion: both with regard to both the test procedure/ the scope of analysis, as well as with the topic of unsolicited findings. It is important that the genetics and ethics communities together with other medical professions involved work jointly on specific case related guidelines to grant the maximum benefit for the care of the children, while preventing patient harm and disproportionate overload of clinicians and the healthcare system by the wealth of available options and economic incentives to increase testing.

46. Practical Benefits, Challenges, and Recommendations on Social Media Recruitment: Multi-Stakeholder Interview Study (Preprint)

- Abstract
BACKGROUND The increasing use of social media opens new opportunities for recruiting patients for research studies. However, systematic evaluations indicate that the success of social media recruitment in terms of cost-effectiveness and representativeness depends on the type of study and its purpose. OBJECTIVE This study aims to explore the practical benefits and challenges of recruiting study participants with social media in the context of clinical and nonclinical studies and provide a summary of expert advice on how to conduct social media–based recruitment. METHODS We conducted semistructured interviews with 6 patients with hepatitis B who use social media and 30 experts from the following disciplines: (1) social media researchers or social scientists, (2) practical experts for social media recruitment, (3) legal experts, (4) ethics committee members, and (5) clinical researchers. The interview transcripts were analyzed using thematic analysis. RESULTS We found diverging expert opinions regarding the challenges and benefits of social media recruitment for research studies in four domains: (1) resources needed, (2) representativeness, (3) web-based community building, and (4) privacy considerations. Moreover, the interviewed experts provided practical advice on how to promote a research study via social media. CONCLUSIONS Even though recruitment strategies should always be sensitive to individual study contexts, a multiplatform approach (recruiting via several different social media platforms) with mixed-methods recruitment (web-based and offline recruitment channels) is the most beneficial recruitment strategy for many research studies. The different recruitment methods complement each other and may contribute to improving the reach of the study, the recruitment accrual, and the representativeness of the sample. However, it is important to assess the context- and project-specific appropriateness and usefulness of social media recruitment before designing the recruitment strategy.

Background: The best treatment of degenerative hip abductor tendon lesions remains largely unknown, as the natural course of the disease has not yet been reported. The aim of the present study was to investigate the natural history of symptomatic degenerative hip abductor lesions. Hypothesis: Nonoperatively treated hip abductor lesions progress over time, resulting in refractory hip pain and low functional outcomes. Study Design: Case series (prognosis); Level of evidence, 4. Methods: Consecutive patients with greater trochanteric pain syndrome and degenerative changes on magnetic resonance imaging (MRI) of the symptomatic hip were included. Bilateral hip MRI scans and a clinical examination were performed at a minimum follow-up of 36 months to study the type and location of hip abductor lesion. Progression of a lesion was defined as a more severe lesion as compared with the initial MRI results or if the lesion extended to another, initially not involved, trochanteric facet. The muscle’s fatty infiltration (FI) was also described. Results: From 106 patients identified, 58 patients (64 hips) aged 66 ± 14 years (mean ± SD) agreed to return to the clinic for follow-up MRI and met the inclusion criteria. At a mean 71-month follow-up, an overall 34% (22/64) of lesions had progressed over time: from trochanteric bursitis to tendinopathy (9/64, 14%) or partial tear (5/64, 8%), from tendinopathy to partial tear (4/64, 6%), from a partial to complete tear (3/64, 4.5%), and with 1 complete tear (1/64, 1.5%) extending to another trochanteric facet. Interestingly, 90% of partial tears remained stable or transformed into a scar. Although patients with a progressive lesion experienced more trochanteric pain (visual analog scale, 4.6 vs 2.8; P = .001), the functional outcomes were comparable with patients with a stable lesion. The majority of hips with a partial tear (64%) demonstrated a progression of gluteus minimus FI from a median grade of 1 to 2, whereas only 1 hip (3%) progressed from grade 2 to 3. Only 3 hips (9%) with a partial tear had a progression of gluteus medius FI, which did not differ significantly from the contralateral unaffected side. Conclusion: Nonoperative treatment might be a valid long-term option for degenerative hip abductor lesions, especially for partial tears, which demonstrated a low risk of clinically relevant progression or muscle FI and similar clinical outcomes to those reported in the literature for operatively treated hip abductor tendon lesions.

Background: Absolute hyperaemic coronary blood flow (Q, in mL/min) and resistance (R, in Wood units [WU]) can be measured invasively by continuous thermodilution. Aims: The aim of this study was to assess normal reference values of Q and R. Methods: In 177 arteries (69 patients: 25 controls, i.e., without identifiable coronary atherosclerosis; 44 patients with mild, non-obstructive atherosclerosis), thermodilution-derived hyperaemic Q and total, epicardial, and microvascular absolute resistances (R tot, R epi, and R micro) were measured. In 20 controls and 29 patients, measurements were obtained in all three major coronary arteries, thus allowing calculations of Q and R for the whole heart. In 15 controls (41 vessels) and 25 patients (71 vessels), vessel-specific myocardial mass was derived from coronary computed tomography angiography. Results: Whole heart hyperaemic Q tended to be higher in controls compared to patients (668±185 vs 582±138 mL/min, p=0.068). In the left anterior descending coronary artery (LAD), hyperaemic Q was significantly higher (293±102 mL/min versus 228±71 mL/min, p=0.004) in controls than in patients. This was driven mainly by a difference in R epi (43±23 vs 83±41 WU, p=0.048), without significant differences in R micro. After adjustment for vessel-specific myocardial mass, hyperaemic Q was similar in the three vascular territories (5.9±1.9, 4.9±1.7, and 5.3±2.1 mL/min/g, p=0.44, in the LAD, left circumflex and right coronary artery, respectively). Conclusions: The present report provides reference values of absolute coronary hyperaemic Q and R. Q was homogeneously distributed in the three major myocardial territories but the large ranges of observed hyperaemic values of flow and of microvascular resistance preclude their clinical use for inter-patient comparison.

Assessment of the microcirculation of the heart has gained interest over recent years. This is partly due to the fact that up to 50% of patients with chest pain visiting the catherization laboratory do not present with significant epicardial stenosis (so-called Angina with Non Obstructive Coronary Artery disease (ANOCA).1Most knowledge regarding microvascular resistance came from non-invasive imaging, from invasive index of microvascular resistance2, or from doppler wires3, both being semi-quantitative and operator dependent.Recently, direct quantitative measurement of coronary blood flow and microvascular resistancehas become possible by thermodilution with saline infusion, using a pressure-temperature guidewire and a multisidehole infusion catheter. Such measurements have been validated versus Positron Emission Tomography (PET)4, have a high reproducibility and are operator independent.5 Procedural safety has been reported before5, but long term safety and absence of late complications have not been described yet. The present study evaluates the safety of absolute flow measurements, both periprocedural, at 30 days and to one year follow-up.
