Background and Objective: In recent years, management and disposal of municipal solid waste has become a global problem and the most important environmental concern. Anaerobic digestion is a cost-effective solution for treatment of both solid waste and wastewater. The aim of this study was to investigate the positive or negative effects of calcium chloride content in anaerobic digestion process of municipal solid waste and leachate on biogas production. Materials and Methods: Experiments with 8 levels of calcium chloride on co-digestion of municipal solid waste and leachate were investigated in 500 ml digesters under mesophilic conditions in a completely randomized design with three replications. Reactors with a ratio of substrate to inoculum of 2 (on VS basis) were operated and the volume of the biogas was measured daily. Volatile and total solids, carbon/nitrogen of waste, COD, BOD and heavy metals were measured by following APHA. Results: The results of the experiment showed that the pH was decreased with increasing calcium chloride concentration. The highest amount of cumulative biogas production was obtained in CaCl2 of 2 g/L with the highest VS and TS reduction. Higher concentrations of CaCl2 (≥3 g/L) caused a reduction in the degradability of volatile and total solids and, as a result, a decreased performance of the digester. Conclusion: The results clearly confirmed that the addition of calcium chloride was an effective solution to improve biodegradability in co-digestion of the MSW and leachate and consequently to reduce the total and volatile solids and to increase the amount of‌ biogas.

Background: Sustainable and commercial production of secondary metabolites is a critical issue when dealing with its clinical application. Efforts are still being made to look for biotic or abiotic elicitors with more efficient and universal effects on the improvement of secondary metabolites. Objective: In order to evaluate the suitability of different biotic elicitors on P. harmala L. cell suspension cultures was established to enhance the &beta-carboline alkaloids (harmaline and harmine) production. Methods: The elicitation of cell suspension cultures of Peganum harmala L. was done by adding various fungal mycelium homogenates (Aspergillus flavus, Alternaria alternate, Coriolus versicolor, Fusarium oxysporum, Mucor sp, Penicillium notatum, and Rhizopus stonifer), Casein hydrolysate and Saccharomyces cerevisiae at different concentrations. The cell cultures of P. harmala L. were subcultured on MS medium with optimal treatment of biotic elicitor. CAMAG analytical HPTLC system was used for estimation of harmaline and harmine after extraction of &beta-carboline alkaloids. Results: The maximum harmine production (91.2±1.8 µg g-1 DW) was observed at 1000 mg l-1 S. cerevisiae in cell suspension culture of P. harmala L. (1.68 fold over than the control). Also the results showed that supplement of 75-100 mg l-1 casein hydrolysate in cell cultures media increased biomass of cell culture and harmaline and harmine production (1.61 and 1.46 times over than the control, respectively). Conclusion: The conclusion of the research showed that by applying biotic elicitors, we can reach to higher secondary metabolites (harmaline and harmine) in cell suspension culture of P. harmala L. We suggest future investigation on using other elicitors like bacterial extract or signal transduction compounds in cell suspension culture of P. harmala L. in order to increase the production of different kind of secondary metabolites.

Background: Hypericum perforatum is one of the valuable medicinal plants in Iran that is used in treating human diseases likes mania, anxiety and depression. Objective : Iranian H. perforatum populations were gathered from deferent region of Iran and also their genetic diversity in company with the possible relationship between genetic diversity and geographical distribution were studied. Methodes: DNA was isolated by CTAB method from young leaves and double digested by EcoRI and Tru1I enzymes. Polymorphic DNA markers generated by DNA fingerprinting technique AFLP (Vos method) using 12 primers combinations. DNA fragments detected with silver nitrate staining according to Basam protocol. Results: Totally 235 bands were scored, that 97% of them were polymorphic. The PIC values ranged between 0.31 and 0.45 with mean of 0.38. The highest and the lowest levels of genetic similarity were 0.89 and 0.29, respectively. Cluster analysis using UPGMA method and DICE similarity coefficient indicated a high genetic diversity among H. perforatum populations. There was no relationship between genetic diversity and geographical distribution. Also for the all loci, the PC1 and PC2 explained 12.8% and 8.3% of the variation, respectively. Conclusion: Wide genetic diversity between Iranian H. perforatum pupulations provide applied information to performance of breeding programs and genetic resource management. Of course, investigation of amount of hypericin and hyperforin metabolites in these populations are requiring to selection paramount genotypes.

Flyrock represents a significant and fundamental challenge in surface mine blasting, carrying inherent risks to humans and the environment. Consequently, accurate prediction, minimization, and identification of the factors influencing flyrock distance are imperative for effective control and mitigation of its destructive consequences. Machine learning and artificial intelligence methodologies have emerged as viable means to predict and simulate in different scientific fields. This study employs Deep Neural Network in conjunction with three optimization algorithms including the JAYA Algorithm, Multi-Verse Optimization Algorithm, and Gravitational Search Algorithm to predict blasting flyrock distance. The developed model consists of a combination of seven input parameters, encompassing both blasting design parameters and rock geomechanical properties. The output of the Deep Neural Networks model is the flyrock distance. For the training and testing of the model, a dataset comprising of 245 blasting records, collected from Songun copper mine, Iran, was utilized. The DNN model yielded an R2 value of 0.96 and an MSE value of 34.11. These results demonstrate the high accuracy and predictive capability of the model. Aim: This study aimed to understand the current level of linezolid (LNZ) resistance in Streptococcus pneumoniae isolates reported over the past 10 years. Material & methods: An electronic search was conducted for the following keywords: (( Streptococcus pneumoniae [title/abstract]) OR ( Pneumococcus [title/abstract]) OR (Pneumococci [title/abstract]) AND (linezolid [title/abstract]) OR (Zyvox [title/abstract])) OR (Zyvoxid [title/abstract])). Result: Out of all the studies, 80 had a cross-sectional design, while 11 followed a cohort approach. The prevalence of LNZ resistance among S. pneumoniae isolates ranged from 0% to 4.86%. Discussion: Urgent, high-powered, randomized, controlled trials with participants from endemic regions are needed to gain a comprehensive understanding of the impact on and significance of LNZ treatment to patients.

The hydrangea ( Hydrangea macrophylla (Thunb). Ser.), an ornamental plant, has good marketing potential and is known for its capacity to change the colour of its inflorescence depending on the pH of the cultivation media. The molecular mechanisms causing these changes are still uncertain. In the present study, transcriptome and targeted metabolic profiling were used to identify molecular changes in the RNAome of hydrangea plants cultured at two different pH levels. De novo assembly yielded 186,477 unigenes. Transcriptomic datasets provided a comprehensive and systemic overview of the dynamic networks of the gene expression underlying flower colour formation in hydrangeas. Weighted analyses of gene co-expression network identified candidate genes and hub genes from the modules linked closely to the hyper accumulation of Al 3+ during different stages of flower development. F3'5'H , ANS , FLS , CHS , UA3GT , CHI , DFR , and F3H were enhanced significantly in the modules. In addition, MYB , bHLH , PAL6 , PAL9 , and WD40 were identified as hub genes. Thus, a hypothesis elucidating the colour change in the flowers of Al 3+ -treated plants was established. This study identified many potential key regulators of flower pigmentation, providing novel insights into the molecular networks in hydrangea flowers.

Purpose: Modern research revealed that plants belonging to the Sida rhombifolia family (Malvaceae) contain biologically active compounds that make them prone to discovering and developing anticancer drugs. This study aimed to evaluate the apoptosis effects of S. rhombifolia extracts in HepG2 Cell Line was performed., Methods: The extractions were prepared, and an MTT assay was applied to evaluate its role in decreasing the viability of HepG2 and HFF cells. Phenolic compounds were analyzed using High-performance liquid chromatography (HPLC). FlowCytometry and RT-qPCR evaluated apoptosis was performed to measure the mRNA expression of pro-and anti-apoptotic mediators., Results: The results can be summarized as EtOAc extract was more cytotoxic against the HepG2 cells (IC50= 364.3 µg/mL) compared to MeOH and HEX extracts (720.2 µg/mL) (560.4 µg/mL) with less cytotoxicity in HFF cells (353.2 µg/mL). The HPLC analysis results revealed most phenolic compounds, such as Epicatechin(1.3 mg/g). The EtOAc extract (300 μg/mL) induced 34% apoptosis in HepG2 cells. RT-qPCR data showed upregulation of the proapoptotic gene (Bax) and increased Bax/BCL-2 ratio by S. rhombifolia EtOAc extract (300 μg/mL)., Conclusion: In conclusion, the EtOAc extract of S. rhombifolia is capable of inducing apoptosis in HepG2 cells through modulation of the mitochondrial pathway, which explains their antitumor activity.

Background and Objectives: Probiotics are added into the food or feed systems and provide beneficial effects to the human or animal health. This study aimed to isolate the gastrointestinal native Lactobacillus strains with high probiotic, cholesterol-assimilation and aflatoxin-degradation capabilities from native chickens., Materials and Methods: About 70 Lactobacillus isolates were isolated from ileum of the Fars province native chickens and were investigated for their probiotic properties., Results: Of 70 Lactobacillus isolates, 10 showed high probiotic capabilities, including survival at acidic conditions (pH up to 2.5), tolerance of 0.5% bile and 6-10% NaCl salts, growth in a wide range of temperature from 15 to 45°C, antagonistic effects against different important bacterial pathogens ( Pseudomonas aeruginosa, Escherichia coli, Streptococcus mutans, Clostridium defficile, Enterococcus hirae, Salmonella enterica and Staphylococcus aureus ) and sensitivity to some important antibiotics. The selected strains had an aggregation time less than 120 min. The 16S-rDNA sequencing showed that the selected strains were highly related to Lactobacillus reuteri and L. casei. Finally, the selected strains in this study along with 10 other probiotic strains isolated and characterized in our pervious study were used to evaluate their cholesterol assimilation and aflatoxin B1 degradation capabilities. The potentials of cholesterol assimilation of the selected strains were significantly different ( P <0.05) and ranged from 2.3% to 99%. The highest content of cholesterol assimilation was obtained in isolates M20 and M4 with more than 98% absorption. Moreover, four strains 43, OR7, M21 and OR9 were able to absorb AFB1 with 58.6%, 52.33%, 47% and 31.6% efficiency respectively., Conclusion: It could be concluded that the strains 43, M21 and OR7 showed high probiotic potentials for application in the poultry industry., (Copyright © 2022 The Authors. Calcineurin-B-like proteins (CBLs) are essential components of the calcium signaling network and act during plant's response to stress and normal conditions. A combined research strategy of in-silico analysis and gene silencing experiment was employed to investigate the role of different classes of CBLs in tomato (Solanum lycopersicum L.) during the response to drought stress. Two different classes of CBL genes, including SlCBL3-1, and SlCBL3-2, with the minimum and a maximum number of drought-responsive cis-elements, were selected and were targeted for transient gene silencing in tomato followed by the drought treatment. The effect of silencing events was evaluated by determining of further growth and physiological traits in plants under both control and drought stress conditions. The results showed that silencing of SlCBL3-1 significantly reduced shoot and root growth, relative water content (RWC), and the concentration of pigments while increased free radical accumulation, lipid peroxidation, and leakage from the cells. On the other hand, no antioxidant enzyme activity or proline induction was triggered in plants after SlCBL3-1 silencing. Some of these adverse events were more significantly enhanced when the silenced plants were exposed to drought stress. Overall, a significant role for SlCBL3-1 in the life cycle of plant suggested under both normal and stress conditions. The SlCBL3-2 silencing showed more efficient plants recovery from silencing or drought stress conditions. Therefore, SlCBL3-2 gene may act as a negative regulator under stress conditions. Background: One of the most important causative agents of visceral leishmaniasis (VL) is Leishmania infantum , which is mainly spread by Phlebotomus and Lutzomyia sandflies in the Old and New World, respectively. Novel and effective drugs to manage this neglected vector-borne disease are urgently required. In this study, we evaluated the toxicity of carvacrol, thymol and linalool, three common essential oil constituents, on amastigotes and promastigotes of L. infantum . Methods : in vitro experiments were performed by 24 h MTT assay. Carvacrol, thymol and linalool at concentrations ranging from 1.3 to 10 μg/mL were tested on promastigotes of L. infantum . For in vivo test, two groups of hamsters ( Mesocricetus auratus ) received 100 mg/kg of body weight/day of carvacrol and thymol as intraperitoneal injection on day 7 post-infection, followed by a 48 h later injection. The third group was treated with the glucantime as standard drug (500 mg/kg) and the last group (control) just received normal saline. On the 16th day, the number of parasites and histopathological changes in liver and spleen were investigated., Results: 24 h MTT assay showed promising antileishmanial activity of thymol and carvacrol, with IC 50 values of 7.2 (48 μM) and 9.8 μg/mL (65 μM), respectively. Linalool at all concentrations did not affect L. infantum promastigote viability. In vivo toxicity data of carvacrol and thymol showed that the former at 100 mg/kg was the safest and most effective treatment with little side effects on the liver., Conclusions: Overall, thymol and carvacrol are highly promising candidates for the development of effective and safe drugs in the fight against VL.

Study Design: This is an animal study., Objectives: Metformin is a safe drug for controlling blood sugar in diabetes. It has been shown that metformin improves locomotor recovery after spinal cord injury (SCI). Neuropathic pain is also a disturbing component of SCI. It is indicated that metformin has neuroprotective and anti-inflammatory effects, which attenuate neuropathic pain and hyperalgesia in injured nerves. Thus, we evaluated metformin's therapeutic effects on SCI neuroinflammation and its sensory and locomotor complications. Meanwhile, results were compared to minocycline, an anti-neuroinflammation therapy in SCI., Setting: Experimental Medicine Research Center, Tehran University of Medical Sciences, Iran METHODS: In an animal model of SCI, 48 male rats were subjected to T9 vertebra laminectomy. Animals were divided into a SHAM-operated group and five treatment groups. The treatments included normal saline as a vehicle control group, minocycline 90 mg/kg and metformin at the doses of 10, 50 and 100 mg/kg. Locomotor scaling, behavioral tests for neuropathic pain and weight changes were evaluated and compared through a 28-days period. At the end of the study, tissue samples were taken to assess neuroinflammatory changes., Results: Metformin 50 mg/kg improved the locomotors ability (p < 0.001) and decreased sensitivity to mechanical and thermal allodynia (p < 0.01). These results were compatible with minocycline effect on SCI (p > 0.05). While metformin led to weight loss, both metformin and minocycline significantly decreased neuroinflammation in the assessment of cord tissue histopathology, and levels of TNF-α and interleukin-1β (p < 0.001)., Conclusions: Metformin could be considered as an alternative therapeutic agent for SCI, as it potentially attenuates neuroinflammation, sensory and locomotor complications of cord injury.

MicroRNAs (miRNAs) are small endogenous regulatory RNAs that are involved in a variety of biological processes related to proliferation, development, and response to biotic and abiotic stresses. miRNA profiles of rice (Oryza sativa L. cv. IR64.) leaves in a partial root zone drying (PRD) system were analysed using a high-throughput sequencing approach to identify miRNAs associated with drought signalling. The treatments performed in this study were as follows: well-watered ("wet" roots, WW), wherein both halves of the pot were watered daily; drought ("dry" roots, DD), wherein water was withheld from both halves of the pot; and well-watered/drought ("wet" and "dry" roots, WD), wherein one half of each pot was watered daily, the same as in WW, and water was withheld from the other part, the same as in DD. High-throughput sequencing enabled us to detect novel miRNAs and study the differential expression of known miRNAs. A total of 209 novel miRNAs were detected in this study. Differential miRNA profiling of the DD, WD and WW conditions showed differential expression of 159 miRNAs, among which 83, 44 and 32 miRNAs showed differential expression under both DD and WD conditions. The detection of putative targets of the differentially expressed miRNAs and investigation of their functions showed that most of these genes encode transcription factors involved in growth and development, leaf morphology, regulation of hormonal homeostasis, and stress response. The most important differences between the DD and WD conditions involved regulation of the levels of hormones such as auxin, cytokinin, abscisic acid, and jasmonic acid and also regulation of phosphor homeostasis. Overall, differentially expressed miRNAs under WD conditions were found to differ from those under DD conditions, with such differences playing a role in adaptation and inducing the normal condition. The mechanisms involved in regulating hormonal homeostasis and involved in energy production and consumption were found to be the most important regulatory pathways distinguishing the DD and WD conditions.

Background: Drought stress is one of the most important abiotic stresses and the main constraint to rice agriculture. MicroRNA-mediated post-transcriptional gene regulation is one of the ways to establish drought stress tolerance in plants. MiRNAs are 20-24-nt regulatory RNAs that play an important role in regulating plant gene expression upon exposure to biotic and abiotic stresses., Methodology/principal Findings: In this study, we applied a partial root drying system as well as a complete root drying system to identify miRNAs involved in conditions of drought stress, drought signaling and wet signaling using high-throughput sequencing. To this end, we produced four small RNA libraries: (1) fully-watered (WW), (2) fully-droughted (WD), and split-root systems where (3) one-half was well watered (SpWW) and (4) the other half was water-deprived (SpWD). Our analysis revealed 10,671 and 783 unique known and novel miRNA reads in all libraries, respectively. We identified, 65 (52 known + 13 novel), 72 (61 known + 11 novel) and 51 (38 known + 13 novel) miRNAs that showed differential expression under conditions of drought stress, drought signaling and wet signaling, respectively. The results of quantitative real-time PCR showed expression patterns similar to the high-throughput sequencing results. Furthermore, our target prediction led to the identification of 244, 341 and 239 unique target genes for drought-stress-, drought-signaling- and wet-signaling-responsive miRNAs, respectively., Conclusions/significance: Our results suggest that miRNAs that are responsive under different conditions could play different roles in the regulation of abscisic acid signaling, calcium signaling, detoxification and lateral root formation.

In the present study, the lenses and ciliary bodies of 20 ostrich eyes were studied macroscopically and microscopically. The histological slides were studied after staining by hematoxylin and eosin, Verhoeff, Van Gieson, and periodic acid-Schiff (PAS). Posterior surface of lens was more convex than its anterior surface. The average lens diameter and thickness were respectively measured as 1.43 ± 0.00 and 0.85 ± 0.00 cm. The average ciliary body thickness was measured as 1.48 ± 0.01 cm. In addition, the ciliary body was seen annular with mean horizontal and vertical external diameters as 4.80 ± 0.07 and 4.36 ± 0.06 cm, respectively. The retina is extended on ciliary body in this bird. The number of ciliary body processes was about 120. The epithelium of lens was cuboidal and the lens capsule had intense positive PAS reaction. Also, the anterior surface of capsule was thicker than its posterior surface. The lens fibers in the central part were thicker than other parts. Elastic and collagen fibers were not observed in the lens. The epithelium of ciliary processes had two layers; superficial cuboidal non-pigmented layer, and deep heavily pigmented layer. The ciliary body was supported by a hyaline cartilage. In addition to the smooth muscle fibers, many isolated skeletal muscle fibers were also seen in ciliary body. In conclusion, the lens and ciliary body of ostrich were similar to other birds, although there were little differences in anatomical dimensions and histological characteristics.

Epidemiological and clinical studies indicate a close relationship between obesity and asthma. Here, we determined the impact of diet-induced obesity on the expression levels of IL-1β, IRAK-1 and TRAF-6 mRNA as well as IL-1β protein level and pathological changes in male Wistar rat's lung after sensitization with ovalbumin (OVA). Twenty male Wistar rats divided into four groups, control with normal diet (C+ND), OVA-sensitized with normal diet (S+ND), control with high-fat diet (C+HFD), and OVA-sensitized with high-fat diet (S+HFD). All rats fed for 12 weeks with standard pellets or high-fat diet while sensitization and challenging with OVA or saline were done for groups in the last month. In the end of intervention, lung was isolated and tested for the expression levels of IL-1β, IRAK-1 and TRAF-6 mRNA with real time-PCR method, and pathological changes were determined. Diet-induced obesity groups showed increased weight, obesity indexes and lipid profiles The expression levels of IL-1β mRNA in OVA-sensitization groups (S+ND and S+HFD) showed a significant increase compared with other groups. Also in S+HFD group, expression level of IRAK-1 and TRAF-6 mRNA were markedly higher than other groups (p<0.001). The pathological changes were marked in sensitized groups compared to non-sensitized groups; with marked increase in obese sensitized rat. The results showed that high fat diet caused overexpression of IL-1β, IRAK-1 and TRAF-6 mRNA as well as IL-1β protein in an experimental model of asthma. Our results suggest that obese-asthmatic conditions may lead to the local production and activation of pro-inflammatory agents.

α-hederin, a saponin that is a major constituent of English Ivy (Hedera helix) is effective in the treatment of asthma. In the present study, the effect of α-hederin on lung tissue pathology and the levels of the inflammatory mediators; IL-2 mRNA, IL-17 mRNA, and MicroRNAs (miRNA)-133a was evaluated in a rat ovalbumin (OVA)-sensitized model of asthma. Rats were divided randomly into control (C), OVA-sensitized (S), OVA-sensitized pretreated with the antioxidant, thymoquinone (3 mg/kg, S + TQ) or OVA-sensitized pretreated with α-hederin (0.02 mg/kg, S + AH) groups. Levels of IL-2 and IL-17 mRNA were higher in the OVA-sensitized group than controls while the level of miRNA-133a gene expression was lower. IL-2 mRNA and miRNA-133a gene expression in the S + TQ group was higher than in the control and OVA-sensitized groups while the level of IL-17 mRNA in the S + TQ group was lower than in the OVA-sensitized group. Pretreatment with α-hederin decreased IL-17 mRNA levels and increased miRNA-133a gene expression compared with OVA-sensitized animals. All pathological changes in pretreated groups were lower than the OVA-sensitized group. These results showed a beneficial effect of α-hederin in OVA-sensitized rats, suggesting that α-hederin affects the IL-2 and IL-17 secretion pathways, altering miRNA-133a expression., (© 2016 Wiley Periodicals, Inc.)

Objective: In previous studies the therapeutic effects of Nigella sativa have been demonstrated on asthmatic animals. In the present study, the preventive effect of single dose of alpha-hederin, its active constituent, has been evaluated on lung inflammation and some inflammatory mediators in lungs of ovalbumin sensitized rat in order to elicit its mechanism., Materials and Methods: Forty rats were randomly grouped in 4 groups; control (C), sensitized (S), sensitized pretreated groups with thymoquinone (3 mg/kg i.p., S+TQ) and alpha-hederin (0.02 mg/kg i.p., S+AH). Levels of IL-13 mRNA and miRNA-126 in lung tissue and its pathological changes in each group were assessed., Results: Elevated levels of miRNA-126, IL-13 mRNA and pathological changes were observed in the sensitized group compared to the control group (p<0.001 to p<0.05). All of these factors were significantly reduced in S+TQ and S+AH groups in comparison to S group (p<0.001 to p<0.05). Although alpha-hederin decreased the levels of miRNA-126, IL-13 mRNA and pathological changes in comparison with thymoquinone, the results were statistically not significant., Conclusion: The results suggested that alpha-hederin had preventive effect on sensitized rats like thymoquinone. It may intervene in miRNA-126 expression, which consequently could interfere with IL-13 secretion pathway leading to a reduction in inflammatory responses.

In the present study, 193 Aspergillus strains were isolated from a total of 100 soil samples of pistachio orchards, which all of them were identified as Aspergillus flavus as the most abundant species of Aspergillus section Flavi existing in the environment. Approximately 59%, 81%, and 61% of the isolates were capable of producing aflatoxins (AFs), cyclopiazonic acid (CPA), and sclerotia, respectively. The isolates were classified into four chemotypes (I to IV) based on the ability to produce AFs and CPA. The resulting dendrogram of random amplified polymorphic DNA (RAPD) analysis of 24 selected A. flavus isolates demonstrated the formation of two separate clusters. Cluster 1 contained both aflatoxigenic and non-aflatoxigenic isolates (17 isolates), whereas cluster 2 comprised only aflatoxigenic isolates (7 isolates). All the isolates of cluster 2 produced significantly higher levels of AFs than those of cluster 1 and the isolates that produced both AFB(1) and AFB(2) were found only in cluster 2. RAPD genotyping allowed the differentiation of A. flavus from Aspergillus parasiticus as a closely related species within section Flavi. The present study has provided for the first time the relevant information on distribution and genetic diversity of different A. flavus populations from nontoxigenic to highly toxigenic enable to produce hazardous amounts of AFB(1) and CPA in soils of pistachio orchards. These fungi, either toxigenic or not-toxigenic, should be considered as potential threats for agriculture and public health.

Objective: To examine the preventive effect of a hydro-ethanolic extract of Nigella sativa on the tracheal responsiveness and white blood cell count in the lung lavage fluid of sensitized guinea pigs., Methods: Three groups of guinea pigs sensitized to intraperitoneally injected and inhaled ovalbumin were given drinking water alone (group S), drinking water containing a low concentration of N. sativa extract (group S+LNS) or drinking water containing a high concentration of N. sativa extract (group S+HNS). The tracheal responses of control animals (group C) and the three groups of sensitized guinea pigs (n = 7 for all groups) to methacholine were measured by the assessment of the tracheal smooth muscle response to increasing concentrations of methacholine, and the effective concentration causing 50% of the maximum response (EC50) was determined. Tracheal responses to 0.1% ovalbumin and white blood cell counts in the lung lavage fluid were also examined., Results: The tracheal response of the group S guinea pigs to both methacholine and ovalbumin was significantly higher than the response of the controls (p<0.01 for both cases). The tracheal responses of the S+LNS and S+HNS groups to both methacholine and ovalbumin were significantly decreased compared to those of the S group (p<0.05 to p<0.01). The total white blood cell and eosinophil counts in the lung lavage fluid of group S were significantly higher than those of group C (p<0.01). The white blood cell counts in both treated groups showed significant improvements (p<0.01 for both cases)., Conclusions: These results demonstrate the preventive effect of the N. sativa extract on the tracheal response and lung inflammation in sensitized guinea pigs.

In previous studies, the relaxant, anticholinergic (functional antagonism) and antihistaminic, effects of Nigella sativa have been demonstrated on guinea pig tracheal chains. In the present study, the prophylactic effect of thymoquinone (one of the constituents of Nigella sativa) on tracheal responsiveness and white blood cell (WBC) count in lung lavage of sensitized guinea pigs was examined. Four groups of sensitized guinea pigs to ovalbumin (OA) were given drinking water alone (group S), drinking water containing low or high concentrations of thymoquinone (S + LTQ and S + HTQ groups) or inhaled fluticasone propionate (FP 250 microg) twice a day (positive control group) (n = 7, for all groups). Tracheal responses of control and four groups of sensitized animals to methacholine at an effective concentration causing 50 % of maximum response (EC(50) M) were measured. Tracheal responses to 0.1 % OA, relative to contraction induced by 10 microM methacholine were also examined. Total WBC and its differential count in lung lavage were also measured. The tracheal responsiveness to methacholine, OA and WBC of S guinea pigs was significantly higher than those of controls (p < 0.001 for all cases). Tracheal responsiveness in S + LTQ, S + HTQ, and FP groups to both methacholine (p < 0.05 to p < 0.001) and OA (p < 0.001 for all cases) was significantly decreased compared to that of the S group. Total WBC was also decreased in all experimental groups compared to that of the S group (p < 0.001 for all groups). There was an increase in eosinophils and a decrease in neutrophils, lymphocytes and monocytes in the S animals compared to the controls (p < 0.001 for all cases). Treatment with both concentrations of thymoquinone and FP variably improved differential WBC count changes compared to the S animals (nonsignificant to p < 0.001). The improvement in tracheal responsiveness, total WBC, eosinophils and lymphocytes changes in the S animals treated with both concentrations of TQ were significantly greater than those of FP (p < 0.05 to p < 0.001). These results showed a preventive effect of thymoquinone, one constituent of N. sativa, on tracheal responsiveness and inflammatory cells of lung lavage of sensitized guinea pigs which was comparable or even greater than that of the inhaled steroid., (Georg Thieme Verlag KG Stuttgart-New York.)

Anthrax is a zoonotic disease caused by the gram-positive spore-forming bacterium Bacillus anthracis. Human infection occurs after the ingestion, inhalation, or cutaneous inoculation of B. anthracis spores. The subsequent progression of the disease is largely mediated by two native virulence plasmids, pXO1 and pXO2, and is characterized by septicemia, toxemia, and meningitis. In order to produce meningitis, blood-borne bacteria must interact with and breach the blood-brain barrier (BBB) that is composed of a specialized layer of brain microvascular endothelial cells (BMEC). We have recently shown that B. anthracis Sterne is capable of penetrating the BBB in vitro and in vivo, establishing the classic signs of meningitis; however, the molecular mechanisms underlying the central nervous system (CNS) tropism are not known. Here, we show that attachment to and invasion of human BMEC by B. anthracis Sterne is mediated by the pXO1 plasmid and an encoded envelope factor, BslA. The results of studies using complementation analysis, recombinant BslA protein, and heterologous expression demonstrate that BslA is both necessary and sufficient to promote adherence to brain endothelium. Furthermore, mice injected with the BslA-deficient strain exhibited a significant decrease in the frequency of brain infection compared to mice injected with the parental strain. In addition, BslA contributed to BBB breakdown by disrupting tight junction protein ZO-1. Our results identify the pXO1-encoded BslA adhesin as a critical mediator of CNS entry and offer new insights into the pathogenesis of anthrax meningitis.

Study Design: Retrospective audit and interview-based study of a traumatic spinal cord injured cohort, assessing the incidence and risk of epididymo-orchitis (E-O)., Objectives: Assess the potential risk factors for E-O in this cohort (spinal cord injured patients)., Setting: Janbazan Clinic for Spinal Cord Injuries, Mashad, Iran., Methods: A retrospective notes audit of 169 male traumatic spinal cord injured (SCI) patients was performed. In addition, interviews were performed to confirm any equivocal data. The following risk factors were assessed: history of recurrent urinary tract infections (UTIs), urethral stricture, urethral diverticuli, urinary fistula, urinary calculi, spinal injury type, neurogenic bladder type, autonomic dysreflexia, vesico-ureteral reflux, sphincterotomy, vasectomy, marriage status, bladder residual and emptying method, ejaculation, spinal injury level, micturation control, and muscular spasm, which included detrusor, external sphincter or lower limb spasm., Results: A total of 65 patients from our group (38.5%) had suffered E-O at least once. E-O presented on average, 3.9 years after the SCI. Patients with a history of muscular spasm appeared less likely to develop E-O (P<0.05). None of the vasectomised patients developed E-O. The relation between all the other factors and E-O were not significant., Conclusions: Our study has shown that the presence of muscular spasm decreases the risk of E-O, although the mechanism remains unclear. Surprisingly, the other historical risk factors showed no clear relation with E-O occurrence.
