1. Assessment of contamination and drug resistance profiles of non-typhoid Salmonella in pig slaughtering industry in Shandong Province

Abstract

ObjectiveTo understand the contamination and drug resistance characteristics of non-typhoidal Salmonella （NTS） during pig slaughter.MethodsA total of 170 carcasses and environmental samples were collected from two large-scale pig slaughterhouses in May and August 2021 in Shandong Province， China. Whole genome sequencing （WGS） combined with bioinformatics was used to analyze the data of the NTS isolates.ResultsNTS was detected in 29 of 170 pork samples， detection rate was 17.1%. Salmonella typhimurium was the predominant serotype （58.6%， 17/29） in the samples. The results of the drug sensitivity analysis showed that 29 NTS strains had 15 drug resistance spectra， with the highest rate of tetracycline resistance （100.0%， 29/29）， followed by ampicillin and chloramphenicol （79.3%， 23/29）. The multidrug resistance rate was 86.2% （25/29 patients）. Multilocus sequence typing （MLST） showed that ST19 was the main type， accounting for 48.3% （14/29） of cases. Analysis of drug resistance genes showed that they carried mainly tet（A）， tet（B）， tet（M）， blaTEM-1B， blaCMY-97， floR， cmlA1， etc.ConclusionThe phenomenon of NTS contamination exists in some pig slaughterhouses in Shandong Province. NTS has shown resistance to multiple antibiotics， suggesting that attention should be paid to the prevention of NTS in pig slaughterhouses.

Abstract

In this work, a series of red emitting Lu3Te2Li3O12: Eu3+ phosphors were synthesized for the first time by high temperature solid state reaction. The structure was characterized by X-ray diffraction. The excitation spectra, emission spectra and CIE chromaticity coordinate were studied. The phosphors show strong red emission at 611 nm due to the 5D0-7F2 transition of Eu3+ ions. The optimal doping concentration of Eu3+ in LTL host is x = 0.5 due to the concentration quenching mechanism of dipole-dipole interaction. The CIE chromaticity coordinates of the LTL: 0.5Eu3++ phosphor is (0.644,0.355), and the purity of the color is up to 93.61%. The phosphor can be considered as a potential red-emitting candidate for near UV WLEDs.

Abstract

Simple Summary: Rotavirus is an increasingly widespread zoonotic agent, the segmented feature of rotavirus genome is responsible for the emergence of new diverse viruses and inter-species transmission. Therefore, monitoring the genetic diversity of rotavirus is helpful to understand its evolution. In this study, a novel potential zoonotic G4P[6] rotavirus with a 344-nt duplication sequence in the non-encoding gene of NSP5 was for the first time detected in a diarrhoea pig sample collected from Guangdong Province, China. Further genome analyses revealed genome reassortment between human-origin and porcine-origin rotavirus. Rotavirus is a major causative agent of diarrhoea in children, infants, and young animals around the world. The associated zoonotic risk necessitates the serious consideration of the complete genetic information of rotavirus. A segmented genome makes rotavirus prone to rearrangement and the formation of a new viral strain. Monitoring the molecular epidemiology of rotavirus is essential for its prevention and control. The quantitative RT-PCR targeting the NSP5 gene was used to detect rotavirus group A (RVA) in pig faecal samples, and two pairs of universal primers and protocols were used for amplifying the G and P genotype. The genotyping and phylogenetic analysis of 11 genes were performed by RT-PCR and a basic bioinformatics method. A unique G4P[6] rotavirus strain, designated S2CF (RVA/Pig-tc/CHN/S2CF/2023/G4P[6]), was identified in one faecal sample from a piglet with severe diarrhoea in Guangdong, China. Whole genome sequencing and analysis suggested that the 11 segments of the S2CF strain showed a unique Wa-like genotype constellation and a typical porcine RVA genomic configuration of G4-P[6]-I1-R1-C1-M1-A8-N1-T1-E1-H1. Notably, 4 of the 11 gene segments (VP4, VP6, VP2, and NSP5) clustered consistently with human-like RVAs, suggesting independent human-to-porcine interspecies transmission. Moreover, a unique 344-nt duplicated sequence was identified for the first time in the untranslated region of NSP5.
