Abstract

The next generation of experiments for rare-event searches based on skipper Charge Coupled Devices (skipper-CCDs) presents new challenges for the sensor packaging and readout. Scaling the active mass and simultaneously reducing the experimental backgrounds in orders of magnitude requires a novel high-density silicon-based package that must be massively produced and tested. In this work, we present the design, fabrication, testing, and empirical signal model of a multi-channel silicon package. In addition, we outline the chosen specifications for the ongoing production of 1500 wafers that will add up to a 10 kg skipper-CCD array with 24000 readout channels., Comment: 14 pages, 13 figures, 2 tables

Abstract

Charge Coupled Devices (CCD) are used for reactor neutrino experiments and already shown their potential in constraining new physics models. The prospect of a Skipper-CCD experiment looking for standard and beyond standard model physics (BSM) in a nuclear reactor has been recently evaluated for different benchmark scenarios. Here we report the installation of the first 2 g Skipper-CCD inside the containment building of a 2 GW$_{th}$ nuclear power plant, positioned 12 meters from the center of the reactor core. We discuss the challenges involved in the commissioning of the detector and present data acquired during reactor ON and reactor OFF periods, with the detector operating with a sub-electron readout noise of 0.17 e-. The ongoing efforts to improve sensitivities to CEvNS and BSM interaction are also discussed., Comment: 12 pages, 7 figures, 1 table

Abstract

The Fermilab Proton-Improvement-Plan-II (PIP-II) is being implemented in order to support the precision neutrino oscillation measurements at the Deep Underground Neutrino Experiment, the U.S. flagship neutrino experiment. The PIP-II LINAC is presently under construction and is expected to provide 800~MeV protons with 2~mA current. This white paper summarizes the outcome of the first workshop on May 10 through 13, 2023, to exploit this capability for new physics opportunities in the kinematic regime that are unavailable to other facilities, in particular a potential beam dump facility implemented at the end of the LINAC. Various new physics opportunities have been discussed in a wide range of kinematic regime, from eV scale to keV and MeV. We also emphasize that the timely establishment of the beam dump facility at Fermilab is essential to exploit these new physics opportunities.

Abstract

We present results from a 3.25 kg-day target exposure of two silicon charge-coupled devices (CCDs), each with 24 megapixels and skipper readout, deployed in the DAMIC setup at SNOLAB. With a reduction in pixel readout noise of a factor of 10 relative to the previous detector, we investigate the excess population of low-energy events in the CCD bulk previously observed above expected backgrounds. We address the dominant systematic uncertainty of the previous analysis through a depth fiducialization designed to reject surface backgrounds on the CCDs. The measured bulk ionization spectrum confirms the presence of an excess population of low-energy events in the CCD target with characteristic rate of ${\sim}7$ events per kg-day and electron-equivalent energies of ${\sim}80~$eV, whose origin remains unknown., Comment: 8 pages, 4 figures

Abstract

We present a new instrument composed of a large number of sub-electron noise Skipper-CCDs operated with a two stage analog multiplexed readout scheme suitable for scaling to thousands of channels. New, thick, $1.35$ Mpix sensors, from a new foundry, are glued into a Multi-Chip Module (MCM) printed circuit board on a ceramic substrate which has 16 sensors each. The instrument, that can hold up-to 16 MCMs, a total of 256 Skipper-CCD sensors (called a Super-Module with $\approx 130$ grams of active mass and $346$ Mpix), is part of the R$\&$D effort of the OSCURA experiment which will have $\approx 94$ super-modules. Experimental results with $10$ MCMs and $160$ Skipper-CCDs sensors are presented in this paper. Abstract

Nuclear reactors are uniquely powerful, abundant, and flavor-pure sources of antineutrinos that continue to play a vital role in the US neutrino physics program. The US reactor antineutrino physics community is a diverse interest group encompassing many detection technologies and many particle physics topics, including Standard Model and short-baseline oscillations, BSM physics searches, and reactor flux and spectrum modeling. The community's aims offer strong complimentary with numerous aspects of the wider US neutrino program and have direct relevance to most of the topical sub-groups composing the Snowmass 2021 Neutrino Frontier. Reactor neutrino experiments also have a direct societal impact and have become a strong workforce and technology development pipeline for DOE National Laboratories and universities. This white paper, prepared as a submission to the Snowmass 2021 community organizing exercise, will survey the state of the reactor antineutrino physics field and summarize the ways in which current and future reactor antineutrino experiments can play a critical role in advancing the field of particle physics in the next decade., Comment: Contribution to Snowmass 2021

Abstract

Many low-threshold experiments observe sharply rising event rates of yet unknown origins below a few hundred eV, and larger than expected from known backgrounds. Due to the significant impact of this excess on the dark matter or neutrino sensitivity of these experiments, a collective effort has been started to share the knowledge about the individual observations. For this, the EXCESS Workshop was initiated. In its first iteration in June 2021, ten rare event search collaborations contributed to this initiative via talks and discussions. The contributing collaborations were CONNIE, CRESST, DAMIC, EDELWEISS, MINER, NEWS-G, NUCLEUS, RICOCHET, SENSEI and SuperCDMS. They presented data about their observed energy spectra and known backgrounds together with details about the respective measurements. In this paper, we summarize the presented information and give a comprehensive overview of the similarities and differences between the distinct measurements. Abstract

Fully-depleted thick silicon Skipper-charge-coupled devices (Skipper-CCDs) are an important technology to probe neutrino and light-dark-matter interactions due to their sub-electron read-out noise. However, the successful search for rare neutrino or dark-matter events requires the signal and all backgrounds to be fully characterized. In particular, a measurement of the electron-hole pair creation energy below 150 eV and the Fano factor are necessary for characterizing the dark matter and neutrino signals. Moreover, photons from background radiation may Compton scatter in the silicon bulk, producing events that can mimic a dark matter or neutrino signal. We present a measurement of the Compton spectrum using a Skipper-CCD and a $^{241}$Am source. With these data, we estimate the electron-hole pair-creation energy to be $\left(3.71 \pm 0.08\right)$ eV at 130 K in the energy range between 99.3 eV and 150 eV. By measuring the widths of the steps at 99.3 eV and 150 eV in the Compton spectrum, we introduce a novel technique to measure the Fano factor, setting an upper limit of 0.31 at 90% C.L. These results prove the potential of Skipper-CCDs to characterize the Compton spectrum and to measure precisely the Fano factor and electron-hole pair creation energy below 150 eV.

Abstract

We explore the sensitivity to new physics of the recently proposed vIOLETA experiment: a 10 kg Skipper Charged Coupled Device detector deployed 12 meters away from a commercial nuclear reactor core. We investigate two broad classes of models which benefit from the very low energy recoil threshold of these detectors, namely neutrino magnetic moments and light mediators coupled to neutrinos and quarks or electrons. We find that this experimental setup is very sensitive to light, weakly coupled new physics, and in particular that it could probe potential explanations of the event excess observed in XENON1T. We also provide a detailed study on the dependence of the sensitivity on the experimental setup assumptions and on the neutrino flux systematic uncertainties., Comment: 17 pages, 7 figures

Abstract

Introduction: This retrospective study on patients with biopsy-proven lupus nephritis (LN) aimed to assess the probability of sustained clinical remission (sCR) and to investigate sCR effects on disease flares and impaired kidney function (IKF). Methods: sCR was defined as clinical-Systemic Lupus Erythematosus Disease Activity Index 2000 (SLEDAI-2K) = 0 and estimated glomerular filtration rate (eGFR) >60 ml/min per 1.73 m2 lasting ≥1 year; IKF: eGFR <60 ml/min per 1.73 m2 for >3 months. We analyzed the probability of achieving and maintaining sCR, and the yearly risk of flare. Cox models were used to identify predictors of sCR and IKF with variables analyzed as time-dependent covariates when appropriate. Results: Of 303 patients followed-up with for 14.8 (interquartile range: 9.8–22) years, 257 (84.8%) achieved sCR. The probability of achieving sCR progressively increased over time reaching 90% at 15 years. Baseline age (hazard ratio [HR]: 1.017; 95% confidence interval [CI]: 0.005–1.029; P = 0.004), hydroxychloroquine intake (HR: 1.385; 95% CI: 1.051–1.825; P = 0.021), and absence of arterial hypertension (HR: 0.699; 95% CI: 0.532–0.921; P = 0.011) were independent predictors of sCR. Among patients who achieved sCR, 142 (55.3%) developed a lupus flare after a median time of 3.6 (2.3–5.9) years. In the remaining 115 patients, sCR persisted for 9.5 (5.8–14.5) years. The probability of sCR to persist at 15 years was 38%. SLE flare risk decreased to 10%, 5%, and 2% in patients with sCR lasting <5, 5 to 10, and >10 years, respectively. At the last observation, 57 patients (18.81%) had IKF. sCR achievement (HR: 0.18, P < 0.001) and its duration (HR: 0.83, P < 0.001) were protective against IKF. Conclusion: sCR is an achievable target in LN management and protects against IKF. The longer the sCR, the higher the chance of its persistence and the lower the risk of SLE flares.

Abstract

This thesis contributes to the economics debate on human capital development. It focuses on the role of the family and investigates the consequences and determinants of parental inputs on child development. It consists of three chapters, of which the first two delve into the consequences of parental inputs in early childhood, whereas the third explores the determinants. Parental inputs broadly indicate the family environment influencing child development and refer specifically to the choice of family structure - parental divorce in Chapter 1 and parental cohabitation in Chapter 3 - and parental investments and parental skills in Chapter 2.

Abstract

The DUNE IDR describes the proposed physics program and technical designs of the DUNE far detector modules in preparation for the full TDR to be published in 2019. It is intended as an intermediate milestone on the path to a full TDR, justifying the technical choices that flow down from the high-level physics goals through requirements at all levels of the Project. These design choices will enable the DUNE experiment to make the ground-breaking discoveries that will help to answer fundamental physics questions. Abstract

The DUNE IDR describes the proposed physics program and technical designs of the DUNE Far Detector modules in preparation for the full TDR to be published in 2019. It is intended as an intermediate milestone on the path to a full TDR, justifying the technical choices that flow down from the high-level physics goals through requirements at all levels of the Project. These design choices will enable the DUNE experiment to make the ground-breaking discoveries that will help to answer fundamental physics questions. Volume 1 contains an executive summary that describes the general aims of this document. The remainder of this first volume provides a more detailed description of the DUNE physics program that drives the choice of detector technologies. It also includes concise outlines of two overarching systems that have not yet evolved to consortium structures: computing and calibration. Volumes 2 and 3 of this IDR describe, for the single-phase and dual-phase technologies, respectively, each detector module's subsystems, the technical coordination required for its design, construction, installation, and integration, and its organizational structure., Comment: 83 pages, 11 figures

Abstract

We have measured the ionization efficiency of silicon nuclear recoils with kinetic energy between 1.8 and 20 keV. We bombarded a silicon-drift diode with a neutron beam to perform an elastic-scattering experiment. A broad-energy neutron spectrum was used and the nuclear recoil energy was reconstructed using a measurement of the time of flight and scattering angle of the scattered neutron. The overall trend of the results of this work is well described by the theory of Lindhard et al. above 4 keV of recoil energy. Below this energy, the presented data shows a deviation from the model. The data indicates a faster drop than the theory prediction at low energies.

Abstract

We present direct detection constraints on the absorption of hidden-photon dark matter with particle masses in the range 1.2-30 eV$c^{-2}$ with the DAMIC experiment at SNOLAB. Under the assumption that the local dark matter is entirely constituted of hidden photons, the sensitivity to the kinetic mixing parameter $\kappa$ is competitive with constraints from solar emission, reaching a minimum value of 2.2$\times$$10^{-14}$ at 17 eV$c^{-2}$. These results are the most stringent direct detection constraints on hidden-photon dark matter in the galactic halo with masses 3-12 eV$c^{-2}$ and the first demonstration of direct experimental sensitivity to ionization signals $<$12 eV from dark matter interactions., Comment: 5 pages, 5 figures

Abstract

We present results of a dark matter search performed with a 0.6 kg day exposure of the DAMIC experiment at the SNOLAB underground laboratory. We measure the energy spectrum of ionization events in the bulk silicon of charge-coupled devices down to a signal of 60 eV electron equivalent. The data are consistent with radiogenic backgrounds, and constraints on the spin-independent WIMP-nucleon elastic-scattering cross section are accordingly placed. A region of parameter space relevant to the potential signal from the CDMS-II Si experiment is excluded using the same target for the first time. This result obtained with a limited exposure demonstrates the potential to explore the low-mass WIMP region (<10 GeV/$c^{2}$) of the upcoming DAMIC100, a 100 g detector currently being installed in SNOLAB., Comment: 11 pages, 11 figures

Abstract

The CONNIE detector prototype is operating at a distance of 30 m from the core of a 3.8 GW$_{\rm th}$ nuclear reactor with the goal of establishing Charge-Coupled Devices (CCD) as a new technology for the detection of coherent elastic neutrino-nucleus scattering. We report on the results of the engineering run with an active mass of 4 g of silicon. The CCD array is described, and the performance observed during the first year is discussed. A compact passive shield was deployed for the detector, producing an order of magnitude reduction in the background rate. The remaining background observed during the run was stable, and dominated by internal contamination in the detector packaging materials. The {\it in-situ} calibration of the detector using X-ray lines from fluorescence demonstrates good stability of the readout system. The event rates with the reactor on and off are compared, and no excess is observed coming from nuclear fission at the power plant. The upper limit for the neutrino event rate is set two orders of magnitude above the expectations for the standard model. The results demonstrate the cryogenic CCD-based detector can be remotely operated at the reactor site with stable noise below 2 e$^-$ RMS and stable background rates. The success of the engineering test provides a clear path for the upgraded 100 g detector to be deployed during 2016.

Abstract

Background. Three different histological scores—histopathologic classification (Berden), Renal Risk Score (RRS) and the Mayo Clinic Chronicity Score (MCCS)—for anti-neutrophil cytoplasmic antibody (ANCA)-associated glomerulonephritis (ANCA-GN) were compared to evaluate their association with patient and kidney prognosis of ANCA-GN. Methods. Patients aged >18 years with at least 1 year of follow-up and biopsy-proven ANCA-GN entered this retrospective study. Renal biopsies were classified according to Berden’s classification, RRS and MCCS. The first endpoint was end-stage kidney disease (ESKD), defined as chronic dialysis or estimated glomerular filtration rate <15 mL/min/1.73 m2. The second endpoint was ESKD or death. Results. Of 152 patients 84 were males, with median age of 63.8 years and followed for 46.9 (interquartile range 12.8–119) months, 59 (38.8%) reached the first endpoint and 20 died. The Kaplan–Meier curves showed that Berden and RRS were associated with first (Berden: P = .004, RRS: P < .001) and second (Berden: P = .001, RRS: P < .001) endpoint, MCCS with the first endpoint only when minimal + mild vs moderate + severe groups were compared (P = .017), and with the second endpoint (P < .001). Among the clinical/histological presentation features, arterial hypertension [odds ratio (OR) = 2.75, confidence interval (95% CI) 1.50–5.06; P = .0011], serum creatinine (OR = 1.17, 95% CI 1.09–1.25; P < .0001), and the percentage of normal glomeruli (OR = 0.97, 95% CI 0.96–0.99; P = .009) were the independent predictors of ESKD at multivariate analysis. When the three scores were included in multivariate analysis, RRS (OR = 2.21, 95% CI 1.15–4.24; P = .017) and MCCS (OR = 2.03, 95% CI 1.04–3.95; P = .037) remained predictive of ESKD, but Berden (OR = 1.17, 95% CI 0.62–2.22; P = .691) did not. Conclusion. RRS and MCCS scores were independent predictors of kidney survival together with high serum creatinine and arterial hypertension at diagnosis

Abstract

Nella tesi mostriamo che lo spazio delle orbite di B_2 può essere dotato di due struttre di varietà Frobenius associate rispettivamente alle equazione di Shrodinger non lineare defocusing e focusing. Motiviati da questo esempio, generalizziamo questa costruzione per ogni B_n, con n>2. Tale costruzione è basata su l'esistenza di un fascio piatto e omogeneo di cometriche definito sullo spazio delle orbite di B_n. La dimostrazione dell'esistenza di un fascio piatto e omogeneo si basa sulla procedura di Dubrovin-Saito modificata in modo opportuno. Partendo da questo fascio, si ricostruisce un'unica struttura di varietà di Frobenius M sullo spazio delle orbite di B_n, per ogni n>2, seguendo una procedura alternativa rispetto a quella standarda proposta da Dubrovin; questa complicazione tecnica è dovuta alla non regolarità del fascio di cometriche. Sorprendentemente, M è isomorfo a una struttura di varietà di Hurwitz-Frobenius. Questa è associata alla gerarchia constrained KP. Tale identificazione rende possibile calcolare esplicitamente le costanti di struttura del prodotto duale di M., In the thesis, we show that the orbit space of B_2 can be equipped with two Frobenius manifolds structures related to the defocusing and focusing NLS (nonlinear Shrodinger) equation respectively. Motivated by this example, we generalize this construction for any B_n, with n>2. Such a construction is based on the existence of a homogeneous flat pencil of cometrics defined on the orbit space of B_n. The proof of the existence of a homogeneous flat pencil relies on the Dubrovin-Saito procedure suitably modified. Starting from this pencil, one can reconstruct a unique Frobenius manifold structure M on the orbit space of B_n, for any n > 2, by following an alternative procedure with respect to the standard one presented by Dubrovin; this technical obstacle is due to the non-regularity of the pencil of cometrics. Remarkably, M is isomorphic to a Hurwitz-Frobenius manifold structure. This is related to the constrained KP hierarchy. Such an identification makes it possible to explicitly compute the structure constants corresponding to the dual product of M.

Abstract

The DAMIC (Dark Matter in CCDs) experiment uses high resistivity, scientific grade CCDs to search for dark matter. The CCD's low electronic noise allows an unprecedently low energy threshold of a few tens of eV that make it possible to detect silicon recoils resulting from interactions of low mass WIMPs. In addition the CCD's high spatial resolution and the excellent energy response results in very effective background identification techniques. The experiment has a unique sensitivity to dark matter particles with masses below 10 GeV/c$^2$. Previous results have demonstrated the potential of this technology, motivating the construction of DAMIC100, a 100 grams silicon target detector currently being installed at SNOLAB. In this contribution, the mode of operation and unique imaging capabilities of the CCDs, and how they may be exploited to characterize and suppress backgrounds will be discussed, as well as physics results after one year of data taking., Comment: Presented at the 34th International Cosmic Ray Conference (ICRC2015), The Hague, The Netherlands

Abstract

The DAMIC experiment uses fully depleted, high resistivity CCDs to search for dark matter particles. With an energy threshold $\sim$50 eV$_{ee}$, and excellent energy and spatial resolutions, the DAMIC CCDs are well-suited to identify and suppress radioactive backgrounds, having an unrivaled sensitivity to WIMPs with masses $<$6 GeV/$c^2$. Early results motivated the construction of a 100 g detector, DAMIC100, currently being installed at SNOLAB. Abstract

We present measurements of radioactive contamination in the high-resistivity silicon charge-coupled devices (CCDs) used by the DAMIC experiment to search for dark matter particles. Novel analysis methods, which exploit the unique spatial resolution of CCDs, were developed to identify $\alpha$ and $\beta$ particles. Uranium and thorium contamination in the CCD bulk was measured through $\alpha$ spectroscopy, with an upper limit on the $^{238}$U ($^{232}$Th) decay rate of 5 (15) kg$^{-1}$ d$^{-1}$ at 95% CL. We also searched for pairs of spatially correlated electron tracks separated in time by up to tens of days, as expected from $^{32}$Si-$^{32}$P or $^{210}$Pb-$^{210}$Bi sequences of $\beta$ decays. The decay rate of $^{32}$Si was found to be $80^{+110}_{-65}$ kg$^{-1}$ d$^{-1}$ (95% CI). An upper limit of $\sim$35 kg$^{-1}$ d$^{-1}$ (95% CL) on the $^{210}$Pb decay rate was obtained independently by $\alpha$ spectroscopy and the $\beta$ decay sequence search. These levels of radioactive contamination are sufficiently low for the successful operation of CCDs in the forthcoming 100 g DAMIC detector., Comment: 18 pages, 20 figures

Abstract

Introduction: Diagnosis and management of microscopic polyangiitis (MPA) have evolved considerably over the past decades, but it is unknown whether clinical and histological presentation and patient and renal outcomes have changed accordingly. Methods: We compared clinical and histopathological characteristic at diagnosis, risk of death, end-stage kidney disease (ESKD), and relapse rate in patients diagnosed with MPA between 1980 and 2022, after grouping them in 2 periods (p): p1980-2001 and p2002-2022. We compared the mortality rate between the 2 periods using Kaplan-Meier estimator and Cox-regression, and competing risks of ESKD and death using the Aalen-Johansen estimator, Fine-Gray multiple regression, and multistate models. Results: Out of 187 patients, 77 were in p1980-2001 and 110 in p2002 to 2022. Patients in p2002 to 2022 were older (66.2 ± 14.0 SD vs. 57.7 ± 15.8; P < 0.001), had a better kidney function (estimated glomerular filtration rate [eGFR] 25.9 ± 24.8 vs. 21.5 ± 28.2 ml/min per 1.73 m2; P = 0.011) and a lower prevalence of the Berden sclerotic class (5.9 vs. 20.9%; P = 0.011). Despite a similar crude and adjusted patient survival, the risk of ESKD decreased during p2002 to 2022 (subdistribution hazard ratio [HR] 0.30, 95% confidence interval [CI]: 0.16-0.57; P < 0.001). The results remained significant after accounting for death after ESKD and after adjusting for potential confounders (HR 0.33 [95% CI: 0.18-0.63; P < 0.001]). The risk of relapse was numerically higher during p2002 to 2022 (subdistribution-HR 1.64 [95% CI: 0.95-2.83; P = 0.075]). Conclusion: MPA kidney involvement has become less severe over the past decades, leading to a reduced risk of ESKD and a higher relapse rate, despite a comparable risk of death.

01 februari 2023, Item does not contain fulltext, Our understanding of the various aspects of pregnancy in women with kidney diseases has significantly improved in the last decades. Nevertheless, little is known about specific kidney diseases. Glomerular diseases are not only a frequent cause of chronic kidney disease in young women, but combine many challenges in pregnancy: immunologic diseases, hypertension, proteinuria, and kidney tissue damage. An international working group undertook the review of available current literature and elicited expert opinions on glomerular diseases in pregnancy with the aim to provide pragmatic information for nephrologists according to the present state-of-the-art knowledge. This work also highlights areas of clinical uncertainty and emphasizes the need for further collaborative studies to improve maternal and fetal health.

36. A Dubrovin-Frobenius manifold structure of NLS type on the orbit space of B-n

Abstract

Generalizing a construction presented in Arsie and Lorenzoni (Lett Math Phys 107:1919–1961, 2017), we show that the orbit space of B2 less the image of the coordinate lines under the quotient map is equipped with two Dubrovin-Frobenius manifold structures which are related respectively to the defocusing and the focusing nonlinear Schrödinger (NLS) equations. Motivated by this example, we study the case of Bn and we show that the defocusing case can be generalized to arbitrary n leading to a Dubrovin-Frobenius manifold structure on the orbit space of the group. The construction is based on the existence of a non-degenerate and non-constant invariant bilinear form that plays the role of the Euclidean metric in the Dubrovin–Saito standard setting. Up to n= 4 the prepotentials we get coincide with those associated with the constrained KP equations discussed in Liu et al. (J Geom Phys 97:177–189, 2015).

Abstract

Nuclear reactors are uniquely powerful, abundant, and flavor-pure sources of antineutrinos that continue to play a vital role in the US neutrino physics program. The US reactor antineutrino physics community is a diverse interest group encompassing many detection technologies and many particle physics topics, including Standard Model and short-baseline oscillations, BSM physics searches, and reactor flux and spectrum modeling. The community's aims offer strong complimentary with numerous aspects of the wider US neutrino program and have direct relevance to most of the topical sub-groups composing the Snowmass 2021 Neutrino Frontier. Reactor neutrino experiments also have a direct societal impact and have become a strong workforce and technology development pipeline for DOE National Laboratories and universities. This white paper, prepared as a submission to the Snowmass 2021 community organizing exercise, will survey the state of the reactor antineutrino physics field and summarize the ways in which current and future reactor antineutrino experiments can play a critical role in advancing the field of particle physics in the next decade., Contribution to Snowmass 2021

Abstract

We use a science-grade skipper charge-coupled device (skipper CCD) operating in a low-radiation background environment to develop a semiempirical model that characterizes the origin of single-electron events in CCDs. We identify, separate, and quantify three independent contributions to the single-electron events, which were previously bundled together and classified as "dark counts": dark current, amplifier light, and spurious charge. We measure a dark current, which depends on exposure, of (5.89±0.77)×10-4e-/pix/day, and an unprecedentedly low spurious charge contribution of (1.52±0.07)×10-4e-/pix, which is exposure independent. In addition, we provide a technique to study events produced by light emitted from the amplifier, which allows the detector's operation to be optimized to minimize this effect to a level below the dark-current contribution. Our accurate characterization of the single-electron events allows one to greatly extend the sensitivity of experiments searching for dark matter or coherent neutrino scattering. Moreover, an accurate understanding of the origin of single-electron events is critical to further progress in ongoing research and development efforts of skipper and conventional CCDs.

Abstract

Mechanisms for the generation of anti-dsDNA autoantibodies are still not completely elucidated. One theory states that dsDNA interacts for mimicry with antibodies raised versus other antigens but molecular features for mimicry are unknown. Here we show that, at physiological acid-base balance, anti-Annexin A1 binds IgG2 dsDNA in a competitive and dose-dependent way with Annexin A1 and that the competition between the two molecules is null at pH 9. On the other hand, these findings also show that dsDNA and Annexin A1 interact with their respective antibodies on a strictly pH-dependent basis: in both cases, the binding was minimal at pH 4 and maximal at pH9-10. The anionic charge of dsDNA is mainly conferred by the numerous phosphatidic residues. The epitope binding site of Annexin A1 for anti-Annexin A1 IgG2 was here characterized as a string of 34 amino acids at the NH2 terminus, 10 of which are anionic. Circulating levels of anti-dsDNA and anti-Annexin A1 IgG2 antibodies were strongly correlated in patients with systemic lupus erythematosus (n 496) and lupus nephritis (n 425) stratified for age, sex, etc. These results show that dsDNA competes with Annexin A1 for the binding with anti-Annexin A1 IgG2 on a dose and charged mediated base, being able to display an inhibition up to 75%. This study provides the first demonstration that dsDNA may interact with antibodies raised versus other anionic molecules (anti-Annexin A1 IgG2) because of charge mimicry and this interaction may contribute to anti-dsDNA antibodies generation.

Abstract

Objectives: The impact of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in patients with SLE remains unclear and data on clinical manifestations after infection are lacking. The aim of this multicentre study is to describe the effect of SARS-CoV-2 in SLE patients. Methods: SLE patients referring to four Italian centres were monitored between February 2020 and March 2021. All patients with SARS-CoV-2 infection were included. Disease characteristics, treatment, disease activity and SARS-CoV-2-related symptoms were recorded before and after the infection. Results: Fifty-one (6.14%) SLE patients were included among 830 who were regularly followed up. Nine (17.6%) had an asymptomatic infection and 5 (9.8%) out of 42 (82.6%) symptomatic patients developed interstitial pneumonia (no identified risk factor). The presence of SLE major organ involvement (particularly renal involvement) was associated with asymptomatic SARS-CoV-2 infection (P = 0.02). Chronic corticosteroid therapy was found to be associated with asymptomatic infection (P = 0.018). Three SLE flares (5.9%) were developed after SARS-CoV-2 infection: one of them was characterized by MPO-ANCA-positive pauci-immune crescentic necrotizing glomerulonephritis and granulomatous pneumonia. Conclusions: SARS-CoV-2 infection determined autoimmune flares in a small number of patients. Our data seem to confirm that there was not an increased risk of SARS-CoV-2 in SLE. Patients with asymptomatic SARS-CoV-2 infections were those having major SLE organ involvement. This may be explained by the high doses of corticosteroids and immunosuppressive agents used for SLE treatment.

Abstract

Vulnerable subjects, including systemic lupus erythematosus (SLE) patients, have been prioritised to receive anti-SARS-CoV-2 vaccines. Few data about the safety of these vaccines in SLE are available. The aim of our study is to investigate the safety of anti-SARS-CoV-2 vaccines in SLE. We included 452 SLE patients, referring to seven tertiary centres, who were immunised. A total of 119 (26%) reported side effects (SE) after the first and/or the second shot (the most frequent SE were fever, local reaction, fatigue, and arthralgia). Patients with constitutional symptoms and those on an immunosuppressive regimen (especially belimumab) showed more SE. In addition, 19 (4%) had a flare after the immunisation (flares classified by organ involvement: six musculoskeletal with constitutional symptoms, four renal, three cardio-respiratory, three haematological, two mucocutaneous). None of the patients needed hospitalisation and none died. Moreover, 15 required a transient increase in corticosteroids and four were treated with steroid pulses. One patient required an additional rituximab course. Anti-dsDNA, moderate/high DAS before vaccine, and belimumab were found more frequently in patients with disease flare. Anti-SARS-CoV-2 vaccines are safe in SLE patients, and they should be recommended in these patients, as the potential benefits widely outweigh the risk of SE. Treatment adjustment might be considered with the aim of minimising SE risk and flare.
